ABSTRACT: Rhododendron fortune belongs to a scented Rhododendron species native to China, which produces fragrant flowers of great ornamental and environmental values for landscaping or indoor beautification. However, the scents in Rhododendron fortuneihave not yet been investigated, let alone the mechanism of the formation of these fragrance in the flowers. In this study, we measured the scents in terms of its volatile components and contents (VOC) in Rhododendron fortuneiat four different flowering stages and in different tissues of the plant by headspace solid-phase micro-extraction combined (HS-SPME) with gas chromatography-mass spectrometry (GC-MS). Then the characteristic aromatic values, which reflects the degree of scent perception by human, of each VOC in the plant was calculated according to its respective aromatic thresholds. Results showed that three main VOCs measured from highest to lowest are methyl benzoates, terpenes and fatty acid derivatives. Their content increased after the flower bud opening and reached the highest at half to full blossom. In a flower most VOC contents were measured in petals and only trace amount in other tissues such as stamen, pistil. A small amount of VOCs was determined in leaves as well.All aromatic values were almost corresponded to the contents of three main VOCs, indicating that the flower fragrance arises truly from these VOC components. S-adenosyl-L-methionine: benzoic acid carboxyl methyl transferase (BAMT) catalyzes the final step to form methyl benzoates. To understand the mechanism of the formation of this main type fragrance and its regulation, we firstly isolate a gene of RfBAMT from petal of Rhododendron fortuneiby using homologous cloning and RACE technology. The full length of its cDNA was 1383 bp，with an open reading frame of 1104 bp, encoding a total of 368 amino acids. The phylogenetic tree analysis showed that RfBAMT was the closest to the BSMT of Camellia japonica, belonging to methyltransferases family. Then we measured the expression level of RfBAMT again at four flower developmental stages and in different flower tissues and leaves. The results showed that the expression level of this gene was highly positively correlated with the emitted content of methyl benzoates in the flowering, implying that RfBAMT plays a pivotal role in the formation and regulation of methyl benzoates in Rhododendron fortune.Thisresearchshowed that the RfBAMT was cloned and identified in our study and its expression level was highly positively correlated with the emitted content of methyl benzoates in the flowers and leaves, which indicated this gene may play an important role on regulation of methyl benzoate synthesis in Rhododendron fortunei.

RESUMO: Rhododendron fortunei pertence a uma espécie de rododendro perfumada nativa da China, que produz flores perfumadas de grande valor ornamental e ambiental para paisagismo ou embelezamento de interiores. No entanto, os aromas em Rhododendron fortunei ainda não foram investigados, muito menos o mecanismo de formação dessas fragrâncias nas flores. Neste estudo, medimos os aromas em termos de seus componentes e conteúdos voláteis (VOC) em Rhododendron fortunei em quatro diferentes estágios de floração e em diferentes tecidos da planta por microextração em fase sólida headspace combinada com cromatografia gasosa-espectrometria de massa. Em seguida, foram calculados os valores aromáticos característicos, que refletem o grau de percepção olfativa pelo ser humano, de cada VOC na planta de acordo com seus respectivos limiares aromáticos. Os resultados mostraram que três COVs principais medidos do mais alto ao mais baixo são benzoatos de metila, terpenos e derivados de ácidos graxos. Seu conteúdo aumentou após a abertura do botão floral e atingiu o máximo na metade da floração total. Em uma flor, a maioria dos teores de COV foram medidos em pétalas e apenas traços em outros tecidos, como estame, pistilo. Uma pequena quantidade de COVs foi determinada nas folhas também. Todos os valores aromáticos foram quase correspondentes aos teores de três COVs principais, indicando que a fragrância da flor surge verdadeiramente desses componentes de COV. Para entender o mecanismo de formação deste tipo principal de fragrância e sua regulação, primeiramente isolamos um gene de RfBAMT da pétala de Rhododendron fortunei usando clonagem homóloga e tecnologia RACE. O comprimento total de seu cDNA era de 1383 bp, com um quadro de leitura aberto de 1104 bp, codificando um total de 368 aminoácidos. A análise da árvore filogenética mostrou que RfBAMT foi o mais próximo do BSMT de Camellia japonica, pertencente à família das metiltransferases. Em seguida, medimos o nível de expressão de RfBAMT novamente em quatro estágios de desenvolvimento da flor e em diferentes tecidos e folhas de flores. Os resultados mostraram que o nível de expressão deste gene foi altamente correlacionado positivamente com o conteúdo emitido de benzoatos de metila na floração, implicando que RfBAMT desempenha um papel fundamental na formação e regulação de benzoatos de metila em Rhododendron fortune foi clonado e identificado em nosso estudo e seu nível de expressão foi altamente correlacionado positivamente com o conteúdo emitido de benzoatos de metila nas flores e folhas, o que indicou que este gene pode desempenhar um papel importante na regulação da síntese de benzoato de metila em Rhododendron fortunei.

Abstract Background Significant association between bullous pemphigoid (BP) and stroke has been reported. This study aimed to evaluate the level of anti-BP180 antibody in stroke patients to explore the relationship between BP and stroke in their pathogenesis. Methods We collected serum samples from stroke patients and matched controls between February 2019 and June 2020. The anti-BP180 antibody levels were measured by enzyme-linked immunosorbent assay (ELISA). Results A total of 1183 stroke patients including 970 with cerebral infarction (CI), 192 with intracerebral hemorrhage (ICH), 21 with CI and ICH, and 855 controls were enrolled in this study. Anti-BP180 autoantibody values were significantly higher in stroke patients than in controls (p < 0.001). Anti-BP180 autoantibody-positive rates were 12.51% (148) in stroke patients and 4.68% (40) in controls (p < 0.001, OR = 2.65). In anti-BP180 autoantibody-positive subjects, the values were significantly higher in stroke patients than in controls (p < 0.001). However, only 10 (6.76%) stroke patients and 3 (7.5%) controls had high values (> 100 RU/mL) (p = 0.87). Stratified analysis showed that anti-BP180 antibody positive rates were independent of age, sex, and stroke subtypes in the stroke group. Positive rates in patients with both CI and ICH were nearly two times higher than those in patients with either CI or ICH alone (p = 0.11, OR = 1.94). Study limitations This study had a limited sample size and lacked quantitative criteria for stroke severity. Conclusions Anti-BP180 antibody values and positive rates were higher in stroke patients than in controls, suggesting that stroke patients may have higher of developing BP.

ABSTRACT Introduction: Remote ischemic preconditioning (RIPC) is a new noninvasive myocardial protection strategy that uses blood pressure cuf inflation to simulate transient non-fatal ischemia to protect the myocardium and reduce ischemia-reperfusion injury. Sulfonylureas may mask the effects of RIPC due to their cardioprotec-tive effect. This meta-analysis aimed to evaluate whether RIPC, in the absence of sulfonylureas, reduces troponin release in patients undergoing cardiac surgery. Methods: We conducted a meta-analysis of randomized controlled clinical trials to determine whether RIPC can reduce postoperative troponin release in cardiac surgery patients undergoing cardiopulmonary bypass without treatment with sulfonylureas. The data were normalized to equivalent units prior to the analysis. A random-effects model was used to provide more conservative estimate of the effects in the presence of known or unknown heterogeneity. Results: Six studies with a total of 570 participants were included. The analysis showed that troponin release was lower in the RIPC group than in the control group at six hours (test of standardized mean differences = 0, Z=3.64, P<0.001) and 48 hours (Z=2.72, P=0.007) postoperatively. When the mean of cross-clamping time was > 60 minutes, RIPC reduced troponin release at six hours (Z=2.84, P=0.005), 24 hours (Z=2.64, P=0.008), and 48 hours (Z=2.87, P=0.004) postoperatively. Conclusion: In cardiac surgery patients who are not taking sulfonylureas, RIPC can reduce troponin release at six and 48 hours postoperatively; hence, RIPC may serve significant benefits in certain cardiac surgery patients.

Abstract Background Alpha-kinase 1 (ALPK1) is a master regulator in inflammation and has been proved to promote renal fibrosis by promoting the production of IL-1β in diabetic nephropathy (DN) mice. Pyroptosis is involved in high glucose (HG)-induced tubular cells injury, characterized by activation of Gasdermin D (GSDMD) and the release of IL-1β and IL-18, resulting in inflammatory injury in DN. It is reasonable to assume that ALPK1 is involved in pyroptosis-related tubular injury in DN. However, the mechanism remains poorly defined. Methods Immunohistochemistry (IHC) staining was performed to detect the expression of pyroptosis- and fibrosis-related proteins in renal sections of DN patients and DN mice. DN models were induced through injection of streptozotocin combined with a high-fat diet. Protein levels of ALPK1, NF-κB, Caspase-1, GSDMD, IL-1β, IL-18 and α-SMA were detected by Western blot. HK-2 cells treated with high-glucose (HG) served as an in vitro model. ALPK1 small interfering RNA (siRNA) was transfected into HK-2 cells to down-regulate ALPK1. The pyroptosis rates were determined by flow cytometry. The concentrations of IL-1β and IL-18 were evaluated by ELISA kits. Immunofluorescence staining was used to observe translocation of NF-κB and GSDMD. Results The heat map of differentially expressed genes showed that ALPK1, Caspase-1 and GSDMD were upregulated in the DN group. The expression levels of ALPK1, Caspase-1, GSDMD and CD68 were increased in renal biopsy tissues of DN patients by IHC. ALPK1expression and CD68+ macrophages were positively correlated with tubular injury in DN patients. Western blot analysis showed increased expressions of ALPK1, phospho-NF-κB P65, GSDMD-NT, and IL-1β in renal tissues of DN mice and HK-2 cells, accompanied with increased renal fibrosis-related proteins (FN, α-SMA) and macrophages infiltration in interstitial areas. Inhibition of ALPK1 attenuated HG-induced upregulation expressions of NF-κB, pyroptosis-related proteins Caspase-1, GSDMD-NT, IL-1β, IL-18, α-SMA, and pyroptosis level in HK-2 cells. Also, the intensity and nuclear translocation of NF-κB and membranous translocation of GSDMD were ameliorated in HG-treated HK-2 cells after treatment with ALPK1 siRNA. Conclusions Our data suggest that ALPK1/NF-κB pathway initiated canonical caspase-1-GSDMD pyroptosis pathway, resulting in tubular injury and interstitial inflammation of DN.

Abstract The study aimed to improve polysaccharide-based packaging to extend the shelf life of fresh produce; a composite film with antimicrobial function was developed and tested. The packaging film is a blend of agar, a film-forming substrate; carrageenan, as a reinforcing agent; glycerol, as a plasticizer; and Nisin, an antimicrobial agent. The film was evaluated for its antibacterial, mechanical, and barrier properties at different humidity levels, using fresh-cut watermelon as the test food material. The film effectively inhibited the growth of Staphylococcus aureus and Listeria monocytogenes. Under relative humidity of 20%, 50%, and 90%, the tensile strength of the antibacterial film containing 0.28% Nisin was 23.08 ± 0.65, 16.09 ± 1.73, and 6.52 ± 0.56 MPa, respectively, and the film also had excellent barrier and heat-sealing properties. The packaging test using fresh-cut watermelon sealed in the antibacterial film containing 0.28% Nisin, under controlled atmosphere storage at either 4 °C or 20 °C, effectively inhibited microbial colonization in the melon and slowed the deterioration of the fruit, as indicated by measures of hardness, weight loss, and soluble solids. This method can extend the shelf life of fresh-cut fruit and provide a reference for further research on polysaccharide-based protective film for fresh produce.

ABSTRACT Due to its high nutritional value, broccoli (Brassica oleracea var. italica Plenck) is one of the most popular vegetables worldwide. This study assessed 36 phenotypic characteristics of 111 broccoli varieties to understand the phenotypic diversity of new broccoli varieties and improve their breeding speed with advantages and characteristics in China, including 108 new varieties and three varieties of common knowledge. The genetic diversity, the principal component, and the cluster of phenotypic characteristics of broccoli varieties were further investigated. The results showed that the coefficients of variation of 36 characteristics ranged between 11.18 % and 94.99 %, with their diversity index between 0.26 and 1.82. The 111 broccoli varieties were further classified into eight groups, primarily attributed to the differences in phenotypic characteristics, including curd weight, main stem thickness, plant development degree, plant height, and anthocyanin coloration. The cumulative contribution rate of the first five principal components reached 81.186 %, corresponding to 12 representative phenotypic traits. The analysis indicated that the phenotypic characteristics of broccoli were rich in diversity, especially for several characteristics appreciated by the market, such as weight, curd firmness, and anthocyanin coloration. This study revealed the basic information on the genetic diversity of new broccoli varieties in China from 2017 to 2019 and provided potential breeding strategies for broccoli to meet diverse market demands.

Pulmonary fibrosis (PF) is a major public health issue with limited treatment options. As the active ingredient of the n-butanol extract of Amygdalus mongolica (BUT), amygdalin inhibits PF. However, its mechanisms of action are unclear and need further verification. Therefore, the purpose of the present studies was to investigate the anti-fibrotic effects of BUT on PF by serum metabolomics and the transforming growth factor β (TGF-β) pathway. Sixty male Sprague-Dawley rats were randomly divided into control, untreated PF, prednisone-treated (5 mg/kg), and BUT-treated (1.75, 1.25, 0.75 g/kg) groups, and the respective drugs were administered intragastrically for 21 days. The serum metabolomics profiles were determined by ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry (UPLC-QTOF/MS) and metabolism network analysis. The expression of TGF-β1, Smad-3, Smad-7, and α-smooth muscle actin (α-SMA) was measured using a real-time polymerase chain reaction in the lung tissue. BUT significantly alleviated fibrosis by reducing the mRNA expressions of TGF-β1 (from 1.73 to 1.13), Smad-3 (from 2.01 to 1.19), and α-SMA (from 2.14 to 1.19) and increasing that of Smad7 (from 0.17 to 0.62). Twenty-eight potential biomarkers associated with PF were identified. In addition, four key biomarkers were restored to baseline levels following BUT treatment, with the lowest dose showing optimal effect. Furthermore, A. mongolica BUT was found to improve PF by the pentose phosphate pathway and by taurine, hypotaurine, and arachidonic acid metabolism. These findings revealed the mechanism of A. mongolica BUT antifibrotic effects and metabolic activity in PF rats and provided the experimental basis for its clinical application.

RESUMO Introdução O surfe depende principalmente da força motriz resultante das oscilações nas ondas. Os atletas precisam remar na zona de ondas e completar as ações da competição na seção mais forte nas paredes das ondas. Isso exige extrema habilidade de equilíbrio, que pode ser fortalecida com treinos específicos. Dentre eles, acredita-se que o treino de core poderia ser benéfico. Objetivo Estudar a influência do treino em plataforma estável e instável na regulação da postura ortostática do surfista sob perturbação do equilíbrio. Métodos Membros da Equipe Nacional de Treinamento de Surf de Hainan foram voluntários do experimento, foram analisados os seus métodos de treino físico, treino de memória e avaliação dos efeitos. Resultados 90% dos atletas conseguem completar o controle de equilíbrio em uma posição autônoma de curto prazo na prancha de equilíbrio em 1 semana, a velocidade de seu progresso também depende da duração do treino, tempos e intervalos, a regra da progressão encontra-se de acordo com a teoria literária do treino esportivo. Conclusão Com a extensão adicional do ciclo de treino da prancha de equilíbrio, acredita-se que a capacidade dos atletas no controle do equilíbrio e capacidades abrangentes de competição reais continuarão a aumentar, o nível técnico geral e a estabilidade de seu desempenho serão ainda mais aprimorados. ABSTRACT Introduction Surfing depends mainly on the driving force resulting from wave oscillations. Athletes need to paddle in the wave zone and complete the competition actions in the most robust section on the wave walls. This requires extreme balance skills, which can be strengthened with specific training. Among them, it is believed that core training could be beneficial. Objective To study the influence of training on a stable and unstable platform on the regulation of the surfer’s orthostatic posture under balance disturbance. Methods Members of the Hainan National Surf Training Team were volunteers for the experiment; their physical training methods, memory training, and effects evaluation were analyzed. Results 90% of athletes manage to complete the balance control in a short-term autonomous position on the balance board in 1 week; their progress also depends on the training duration, times, and intervals; the progression rule lies according to the literary theory of sports training. Conclusion With the further extension of the balance board training cycle, it is believed that athletes’ ability to control balance and real competition comprehensive capabilities will continue to increase, the overall technical level and stability of their performance will be even greater. RESUMEN Introducción El surf depende principalmente de la fuerza motriz resultante de las oscilaciones de las olas. Los deportistas necesitan remar en la zona de la ola y completar las acciones de la competición en la sección más robusta de las paredes de la ola. Para ello, es necesario contar con habilidades de equilibrio extremas, que pueden reforzarse con un entrenamiento específico. Entre ellas, se cree que el entrenamiento del núcleo podría ser beneficioso. Objetivo Estudiar la influencia del entrenamiento en una plataforma estable e inestable sobre la regulación de la postura ortostática del surfista bajo una perturbación del equilibrio. Métodos Los miembros del Equipo Nacional de Entrenamiento de Surf de Hainan fueron voluntarios para el experimento; se analizaron sus métodos de entrenamiento físico, el entrenamiento de la memoria y la evaluación de los efectos. Resultados El 90% de los atletas consiguen completar el control del equilibrio en una posición autónoma a corto plazo en la tabla de equilibrio en 1 semana; su progreso también depende de la duración del entrenamiento, los tiempos y los intervalos; la regla de progresión se encuentra según la teoría literaria del entrenamiento deportivo. Conclusión Con la ampliación del ciclo de entrenamiento en la tabla de equilibrio, se cree que la capacidad de los atletas para controlar el equilibrio y las capacidades integrales de la competición real seguirán aumentando, el nivel técnico general y la estabilidad de su rendimiento serán aún mayores. Abstract Objectives: To investigate the clinical value of using Head-Shaking Test (HST) + Head-Shaking Tilt Suppression Test (HSTST) to distinguish between peripheral and central vertigo as well as to analyze the consistency of findings between tests at the bedside vs. in the examination room. Methods: We retrospectively analyzed patients who presented for central or peripheral vertigo from July 2019 to July 2021. The results were compared between HST and HST+HSTST. The concordance between bedside and examination room outcomes was analyzed. Results: Forty-seven (58.8%) patients in the peripheral vertigo group and 33 (41.2%) patients in the central vertigo group were included. In the peripheral group, 44 (both examination room and bedside: 93.6%) patients had horizontal Head-Shaking Nystagmus (hHSN), most of which were suppressed in HSTST. However, in the central group, most cases had perverted HSN (pHSN; examination room: 72.7%; bedside: 66.7%), which was seldomly suppressed in HSTST. The HST+HSTST showed a >20% higher specificity in identifying peripheral vertigo than HST alone. The bedside results were consistent with the examination room results using the kappa test (p< 0.001). Conclusions: Suppressed hHSN was a strong indicator of peripheral vertigo. Conversely, pHSN was more often seen in central vertigo, which was not readily suppressed in HSTST. The bedside results of HST+ HSTST yielded qualitative agreement with the tests in the examination room. HST+ HSTST could be used as reliable methods in the clinic to distinguish between peripheral and central vestibular disorders. Level of evidence: Level 3.

Abstract This study aimed to prepare the total saponins from Panax japonicus (TSPJ), and investigate their protective effects on learning and memory ability of aging mice induced by D-galactose. The TSPJ product with content of 81.03% was obtained. Fifty mice were randomly divided into control, model, and low-, middle- and high-dose TSPJ groups. The D-galactose-induced aging model was established in model, and low-, middle- and high-dose TSPJ groups. Then, the low-, middle- and high-dose TSPJ groups were treated with 20, 40 and 80 mg/kg TSPJ by gavage, respectively, for 8 weeks. At the end of treatment, compared with model group, in 40 and 80 mg/kg TSPJ groups the brain aging symptoms were obviously moderated, the step-down latency was increased, the frequency of mistake was decreased, the brain index was increased, the brain tissue SOD and GSH-Px levels were increased, the brain tissue MDA level was decreased, and the brain tissue Na+-K+-ATPase and Ca2+-Mg2+-ATPase activities were increased. In conclusion, TSPJ can reduce the brain tissue oxidative stress injury and protect the cell membrane transport capacity, thus improving the learning and memory ability in aging mice.

The fluorescent microspheres are prepared by the swelling-evaporation method. The optimal swelling conditions are obtained by researching related factors, such as types and dosages of swelling agent, dosages of fluorescent substance, swelling time and swelling temperature. Considering that the structure of microspheres can influence the swelling properties, the paper mainly focuses on the effect of molecular weight and surface carboxyl group content of polystyrene microspheres on swelling properties. When the molecular weight of microspheres is 75276 g mol-1, the density of carboxyl group on their surface is 6.94×10-4 mol g-1, and the doping amount of europium complex reaches the maximum of 24.9 mg g-1 with the strongest fluorescence intensity. Meanwhile, the as-prepared fluorescent microspheres have high monodispersity, uniform particles size and high thermal stability (the decomposition temperature is 385oC), which sets up the good foundation for biomolecule detection. So, this work provides a significant experience for the preparation of fluorescent microspheres.

Abstract Background: The internal NAD(P)H dehydrogenase (NDA) gene family was a member of the NAD(P)H dehydrogenase (ND) gene family, mainly involved in the non-phosphorylated respiratory pathways in mitochondria and played crucial roles in response to abiotic stress. Methods: The whole genome identification, structure analysis and expression pattern of NDA gene family were conducted to analyze the NDA gene family. Results: There were 51, 52, 26, and 24 NDA genes identified in G. hirsutum, G. barbadense, G. arboreum and G. raimondii, respectively. According to the structural characteristics of genes and traits of phylogenetic tree, we divided the NDA gene family into 8 clades. Gene structure analysis showed that the NDA gene family was relatively conservative. The four Gossypium species had good collinearity, and segmental duplication played an important role in the evolution of the NDA gene family. Analysis of cis-elements showed that most GhNDA genes contained cis-elements related to light response and plant hormones (ABA, MeJA and GA). The analysis of the expression patterns of GhNDA genes under different alkaline stress showed that GhNDA genes were actively involved in the response to alkaline stress, possibly through different molecular mechanisms. By analyzing the existing RNA-Seq data after alkaline stress, it was found that an NDA family gene GhNDA32 was expressed, and then theGhNDA32 was silenced by virus-induced gene silencing (VIGS). By observing the phenotype, we found that the wilting degree of silenced plants was much higher than that of the control plant after alkaline treatment, suggesting that GhNDA32 gene was involved in the response to alkaline stress. Conclusions: In this study, GhNDAs participated in response to alkaline stress, especially NaHCO3 stress. It was of great significance for the future research on the molecular mechanism of NDA gene family in responding to abiotic stresses.

Abstract Present study aims to establish a reliable infant digestion model using pH-STAT method and the model is also applied to static in-vitro digestion of different infant formulas. Model-3 which consisted of NaCl, KCl and CaCl2 as gastrointestinal digestive fluids demonstrated better repeatability than model-1 was selected for the evaluation of static in-vitro digestion of different infant formulas (different fatty acids, protein composition and with/without milk fat globule membrane) using pH-STAT method. Three infant formulas (IF) with different fatty acids composition were evaluated and IF2 which contained the highest amounts of medium-chain saturated fatty acids (8.02 ± 0.47%) demonstrated the highest in-vitro gastrointestinal lipolysis rate (65.88 ± 0.24%). Infant formulas containing peptides and amino acids were more easily hydrolyzed by protease, which probably enhanced the release of FFA (IF1, 50.60 ± 0.87%; IF4, 60.17 ± 2.97%; IF5, 62.21 ± 2.27%; IF6, 66.51 ± 2.38%). In addition, the presence of milk fat globule membrane (MFGM, IF7) was found to accelerate in-vitro gastrointestinal lipolysis (IF1, 50.60 ± 0.87%; IF7, 58.05 ± 1.77%). In summary, the new developed model is suitable for investigating in-vitro digestion of different infant formulas.
