Germination response of palm seeds on a two-way thermogradient plate

ABSTRACT Palm trees are propagated almost exclusively by seeds and each species germinates under a certain temperature range. In this sense, the two-way thermogradient plate may be used to determine temperature limits for germination and seed response to temperature. The objective was to define the alternating temperature regime promoting higher and faster seed germination of Carpentaria acuminata and Phoenix canariensis palms using a two-way thermogradient plate. This equipment allowed 64 combinations of alternating and constant temperatures, ranging from 6.97 to 36.42 ºC for C. acuminata, and 7.96 to 35.94 ºC for P. canariensis. Seeds were sown in Petri dishes (25 x 9 cm) containing 1% water agar. Linear regressions were estimated to determine cardinal temperatures. After 50 days, non-germinated seeds were transferred from the two-way thermogradient plate to a germination chamber at 30 °C. The temperature regime promoting highest seed germination percentage of C. acuminata was 30.45/33.00 °C (day/night), with minimum, optimum, and maximum temperatures of 9.13, 28.53, and 36.33 °C, respectively. For seed germination of P. canariensis, the most appropriate temperature regime was 29.77/17.93 °C (day/night), with minimum, optimum, and maximum temperatures of 9.53, 28.03, and 35.43 °C, respectively.

Abstract The reproduction of monogamous wild birds in captivity it`s difficult and the apparent low fertility in males requires more investigations. The objective of the study was to test the hypothesis that wild bird species in captivity would present low reproductive potential, through the analysis of the morphological characteristics of Ara ararauna testicles, maintained in captivity, correlating them with the climate variations in the Cerrado Biome. For that, testicles were captured in April (dry) and October (rainy). The right and left testicles showed mean weight, gonadosomatic index, longer axis, and volume similar between the dry and rainy season. Only the shorter axis demonstrated higher values during the rainy season. The morphometric variables of the seminiferous tubules have also higher values during the rainy season. By these histological and morphometric characteristics of the seminiferous epithelium we can conclude that, during the rainy season, the testicles were in gonadal recrudescence, which precedes the reproduction phase. During the dry season, the testicles were in the rest phase of the seminiferous epithelium. Therefore, we concluded that the species in captivity, under Cerrado environmental conditions, have kept their reproductive potential, presenting a complete spermatogenic cycle during the rainy season, which can guarantee the species perpetuation.

The lizard that never sleeps: activity of the pampa marked gecko Homonota uruguayensis

ABSTRACT It is generally assumed that lizards are active whenever climatic conditions are favorable. Homonota uruguayensis (Vaz-Ferreira & Sierra de Soriano, 1961) is the only native gecko - and nocturnal lizard - living in the northern Pampa biome, and its ecology is poorly known. This study aimed at describing this species’ pattern of daily and annual activity and its relation with environmental temperatures. The study was conducted in the extreme south of Brazil (Rosário do Sul, State of Rio Grande do Sul), between May 2010 and January 2011 at a rocky outcrop located in the Pampa biome. The study was carried out in a total of four seasonal field trips, totalizing 1185 hours of field work. The data were collected, both during the day and the night in 6-hour shifts (duration of the sampling period). The area was randomly covered at each shift to record activity and microhabitat use by the lizards. In total 1541 specimens were recorded throughout the study. Homonota uruguayensis showed diurnal and nocturnal activity in the four seasons, with periods of daily activity varying significantly between all seasons in a cyclic and multimodal pattern, with no significant relation with environmental temperatures. There was no difference in activity related to sex and age classes. Most active lizards were found when air temperatures ranged from 14 °C to 32.9 °C (82% of active lizards) and substrate temperatures ranged from 10 °C to 32.9 °C (87% of active lizards).
RESUMEN La suposición general ha sido que los lagartos son activos cuando las condiciones climáticas son favorables. Homonota uruguayensis (Vaz-Ferreira & Sierra de Soriano, 1961) es la única lagartija nativa - y la lagartija nocturna - que vive en el norte de bioma Pampa. La ecología de esta especie es poco conocida y este estudio tuvo como objetivo describir su patrón de actividad diaria y anual y su relación con las temperaturas ambientales. El estudio se llevó a cabo en el extremo sur de Brasil (Rosário do Sul, Rio Grande do Sul), a partir de mayo 2010 - enero 2011, en un afloramiento rocoso situado en el bioma Pampa. El estudio totalizó cuatro excursiones de campo estacionales y 1185 horas. Los datos fueron recogidos a lo largo de los turnos de 6 horas durante el día y la noche. El área fue recorrida aleatoriamente en cada turno siempre registrandose la actividad y el microhabitat utilizado por los lagartos. A lo largo del estudio, 1541 especímenes se registraron. Homonota uruguayensis presentó hábito diurno y nocturno en las cuatro estaciones, con períodos de actividad diaria variando significativamente entre todas las estaciones en forma cíclica y multimodal, no se observó relación significativa con las temperaturas ambientales. No fue constatada diferencia entre los sexos con relación a la actividad y las clases de edad. La mayoría de los lagartos activos fue encontrada a temperaturas del aire que variaron de 14 °C a 32,9 °C (82% de lagartos activos) y a temperaturas del sustrato entre 10 °C y 32,9 °C (87% de lagartos activos).

OBJECTIVES: We designed a cohort study to describe characteristics and outcomes of patients with coronavirus disease (COVID-19) admitted to the intensive care unit (ICU) in the largest public hospital in Sao Paulo, Brazil, as Latin America becomes the epicenter of the pandemic. METHODS: This is the protocol for a study being conducted at an academic hospital in Brazil with 300 adult ICU beds dedicated to COVID-19 patients. We will include adult patients admitted to the ICU with suspected or confirmed COVID-19 during the study period. The main outcome is ICU survival at 28 days. Data will be collected prospectively and retrospectively by trained investigators from the hospital’s electronic medical records, using an electronic data capture tool. We will collect data on demographics, comorbidities, severity of disease, and laboratorial test results at admission. Information on the need for advanced life support and ventilator parameters will be collected during ICU stay. Patients will be followed up for 28 days in the ICU and 60 days in the hospital. We will plot Kaplan-Meier curves to estimate ICU and hospital survival and perform survival analysis using the Cox proportional hazards model to identify the main risk factors for mortality. RESULTS: We expect to include a large sample of patients with COVID-19 admitted to the ICU and to be able to provide data on admission characteristics, use of advanced life support, ICU survival at 28 days, and hospital survival at 60 days. CONCLUSIONS: This study will provide epidemiological data about critically ill patients with COVID-19 in Brazil, which could inform health policy and resource allocation in low- and middle-income countries.

ABSTRACT During the Brazilian slavery period, many African migrants were brought to the American continent. Historically, some of these migrants escaped from the Brazilian gold mines and farms to which they had been brought and settled in remote valleys and this was the main mode of resistance to the slavery system. These runaway-slave descendant communities are called quilombos, a group with distinct ethnic identity, specific behavioral habits, including geographic isolation and conservative practices. The objective of this study was to investigate the prevalence of rodent-borne viruses in two Afro-descendent communities from Mato Grosso do Sul State, Midwestern Brazil. A total of 319 individuals from rural and urban quilombola communities were enrolled. Twelve (3.76%) had anti-rodent-borne virus IgG antibodies. Seven (2.19%) were anti-mammarenavirus reactive and nine (2.82%) had anti-orthohantavirus antibodies. The literature includes limited data on the health status of quilombola communities, but all the studies emphasize the disparity of attention of local healthcare personnel to these communities compared to the general population. The findings of this study highlight the vulnerability and the precarious health conditions of quilombola groups, especially those living in rural areas and thus, point to the need of preventive measures to improve access to healthcare for this ethnic group.

Resumo Objetivo Comparar concentrações séricas de IgE específica e diâmetros médios das pápulas induzidas no teste cutâneo de leitura imediata com leite de vaca e suas frações com resultados do teste de provocação oral e estabelecer pontos de corte, capazes de predizer reatividade clínica ao leite de vaca em pacientes atendidos em um serviço de referência. Métodos Cento e vinte e duas crianças (mediana 17 meses), com história de reações imediatas ao leite de vaca e presença de IgE específicas para leite de vaca e/ou frações (testes cutâneos e/ou IgE sérica positivos) foram submetidas ao teste de provocação oral aberto com leite de vaca. Resultados O teste de provocação oral foi positivo em 59,8% das crianças, 49% eram do sexo masculino. Os níveis séricos de IgE específica, assim como os diâmetros médios das pápulas para leite de vaca, foram significantemente maiores nos alérgicos (medianas: 3,39kUA/L vs. 1,16 kUA/L; 2,5 mm vs. 0 mm). Os "pontos de corte ótimos" (Índice de Youden) das IgE séricas específicas para o leite de vaca e suas frações capazes de predizer a reatividade ao leite de vaca (teste de provocação oral positivo) foram: 5,17kUA/L para o leite de vaca, 0,95 kUA/L para α-lactoalbumina, 0,82kUA/L para β-lactoglobulina e 0,72kUA/L para caseína e para os diâmetros de pápulas foram 3,5 mm para leite de vaca e 6,5 mm, 9,0 mm e 3,0 mm para as frações α-lactoalbumina, β-lactoglobulina e caseína, respectivamente. Conclusões Os níveis de corte capazes de predizer reatividade clínica ao leite de vaca foram: 5,17kUA/L para IgE sérica específica e 3,5 mm para a medida do diâmetro da pápula, valores considerados discriminatórios para o diagnóstico da alergia ao leite de vaca.
Abstract Objective To compare serum concentrations of specific IgE and mean papule diameters induced in the immediate skin reactivity test with cow's milk and its fractions with results of the oral challenge test, and to establish cutoff points capable of predicting clinical reactivity to cow's milk in patients treated at a referral service. Methods One hundred and twenty-two children (median of 17 months) with a history of immediate reactions to cow's milk and presence of specific IgE for cow's milk and/or its fractions (positive skin and/or IgE serum tests) were submitted to open oral challenge test with cow's milk. Results The oral challenge test was positive in 59.8% of the children, 49% of whom were males. Serum levels of specific IgE, as well as mean cow's milk papule diameters, were significantly higher in allergic patients (medians: 3.39 kUA/L vs. 1.16 kUA/L, 2.5 mm vs. 0 mm). The optimal cutoff points (Youden's index) of serum IgE specific for cow's milk and its fractions capable of predicting cow's milk reactivity (positive oral challenge test) were: 5.17 kUA/L for cow's milk, 0.95 kUA/L for α-lactalbumin, 0.82 kUA/L for β-lactoglobulin, and 0.72 kUA/L for casein, whereas for papule diameters the cutoff points were 3.5 mm for cow's milk and 6.5 mm, 9.0 mm, and 3.0 mm for the α-lactalbumin, β-lactoglobulin, and casein fractions, respectively. Conclusions The cutoff points capable of predicting clinical reactivity to cow's milk were: 5.17 kUA/L for serum-specific IgE and 3.5 mm for papule diameter measurement, values considered discriminatory for the diagnosis of cow's milk allergy.

RESUMO Introdução: A cirrose gera alterações nas trocas gasosas e a desnutrição proteico-calórica em pacientes hepatopatas. Objetivo: Avaliar e comparar as variáveis cardiopulmonares, a força do aperto de mão (FAM) e a composição corporal entre pacientes cirróticos pelo vírus da hepatite C e indivíduos saudáveis, e correlacionar o consumo máximo de oxigênio (VO2MAX) com a FAM. Métodos: Esta pesquisa caracteriza-se como estudo de caso-controle composto por 36 participantes (18 pacientes cirróticos com HCV e 18 indivíduos hígidos) de ambos os sexos, maiores de 18 anos. A força de preensão palmar foi mensurada por dinamometria com dinamômetro mecânico de empunhadura com alça ajustável. As variáveis ventilatórias foram avaliadas por ergoespirometria com teste de carga progressiva em cicloergômetro. A composição corporal foi mensurada por um técnico em cineantropometria nível II. Foram utilizados os testes t independente e Mann-Whitney para comparação entre os grupos e a correlação de Spearman para associação entre as variáveis. Resultados: Foram encontradas diferenças no consumo máximo de oxigênio 16,20 (11,60-18,55), mediana e intervalo interquartil x 19,90 (16,27-26,85), ventilação 45,40 (36,45-54,20) x 63,40 (50,40-78,00), produção de dióxido de carbono 785,88 (655,81-963,14) x 988,04 (826,93-1546,21), frequência cardíaca máxima (127,66 ± 23,26 média e ± DP) x (146,29 ± 23,31), primeiro limiar ventilatório (10,700 ± 3,19) x (14,912 ± 4,45) e segundo limiar ventilatório (14,16 ± 4,48) x (18,25 ± 5,54) entre cirróticos e controles, respectivamente. Encontramos correlação positiva moderada entre o consumo máximo de oxigênio e a força do aperto de mão (r = 0,474, p = 0,047). Conclusão: Existem alterações nas variáveis cardiopulmonares e há associação entre o VO2MAX e a FAM em pacientes cirróticos pelo vírus da hepatite C.
ABSTRACT Introduction: Cirrhosis causes changes in gas exchange and protein-calorie malnutrition in patients with liver disease. Objective: To evaluate and compare cardiopulmonary variables, handgrip strength (HGS) and body composition between cirrhotic patients with hepatitis C virus and healthy individuals, and to correlate maximal oxygen uptake (VO2MAX) with HGS. Methods: This survey is characterized as a case-control study composed of 36 participants (18 cirrhotic patients with HCV and 18 healthy individuals) of both sexes, older than 18 years. The palmar grip strength was measured by dynamometry using a mechanical handle dynamometer with adjustable handle. The ventilatory variables were evaluated by ergospirometry with a progressive load test on a cycloergometer. The body composition was measured by a level II cineanthropometry technician. Independent t test and Mann-Whitney test were used for comparison between groups and Spearman correlation for association between variables. Results: There were differences in maximal oxygen uptake 16.20 (11.60-18.55), median and interquartile range x 19.90 (16.27-26.85), ventilation 45.40 (36.45-54.20) x 63.40 (50.40-78.00), carbon dioxide production 785.88 (655.81-963.14) x 988.04 (826.93-1546.21), maximum heart rate (127.66 ± 23.26, mean and ± SD) x (146.29 ± 23.31), first ventilatory threshold (10.700 ± 3.19) x (14.912 ± 4.45) and second ventilatory threshold (14.16 ± 4.48) x (18.25 ± 5.54) between cirrhotic patients and controls, respectively. We found a moderate positive correlation between maximal oxygen uptake and handgrip strength (r=0.474, p=0.047) Conclusion: There are changes in cardiopulmonary variables and there is an association between VO2MAX and HGS in cirrhotic patients with hepatitis C virus.
RESUMEN Introducción: La cirrosis genera cambios en los intercambios gaseosos y la desnutrición proteico-calórica en pacientes con enfermedad hepática. Objetivo: Evaluar y comparar las variables cardiopulmonares, la fuerza de prensión manual (FPM) y la composición corporal entre pacientes cirróticos por el virus de la hepatitis C y sujetos sanos, y correlacionar el consumo máximo de oxígeno (VO2MAX) con la FPM. Métodos: Esta investigación se caracteriza como estudio caso-control compuesto por 36 participantes (18 pacientes cirróticos con VHC y 18 individuos sanos) de ambos sexos, mayores de 18 años. La fuerza de agarre palmar se midió mediante dinamometría con dinamómetro mecánico con mango ajustable. Las variables ventilatorias se evaluaron mediante ergoespirometría con prueba de carga progresiva en un cicloergómetro. La composición corporal fue medida por un técnico en cineantropometría de nivel II. Se utilizaron las pruebas t independiente y Mann-Whitney para la comparación entre los grupos y la correlación de Spearman para la asociación entre las variables. RESUMO Objetivo: Avaliar a dispersão de luz intraocular antes e depois da ceratectomia fotorrefrativa (PRK) para baixa miopia com e sem a aplicação tópica de mitomicina C. Métodos: Pacientes submetidos à PRK para baixa miopia foram selecionados para o estudo. PRK sem MMC foi realizado em 21 olhos (12 pacientes) e PRK com MMC tópica a 0,02% foi realizado em 25 olhos (25 pacientes). Ambos os grupos foram tratados com o excimer laser da Nidek EC5000. Avaliações foram realizadas usando o medidor de dispersão de luz C-Quant no pré-operatório e com 2 e 4 meses de pós-operatório. Resultados: A média de idade dos pacientes foi 30 ± 4 anos e a média do equivalente esférico foi -2,2 ± 0,75 D. As médias da dispersão de luz intraocular no pré-operatório foram 1,07 ± 0,10 no grupo PRK sem MMC e 1,07 ± 0,11 log(s) no grupo PRK com MMC tópica. Após 2 meses da cirurgia houve uma diminuição na média da dispersão de luz intraocular em ambos os grupos. Entretanto uma diferença estatisticamente significante, comparado com os valores pré-operatórios, foi observada apenas no grupo PRK com MMC (0,98 ± 0,09 log(s), p=0,002), provavelmente devido as medidas com maior espalhamento de luz no grupo sem MMC (1,03 ± 0,13 log(s), p=0,082). Após 4 meses de pós-operatório, os valores de dispersão de luz não apresentavam diferença estatisticamente significantes quando comparados com os valores iniciais, tanto no grupo sem MMC (1,02 ± 0,14 log(s), p=0,26) quanto no grupo com MMC tópica (1,02 ± 0,11 log(s), p=0,13). Conclusão: PRK para baixa miopia diminui a dispersão de luz ocular e a aplicação de MMC contribui para uma ainda menor dispersão de luz no período pós-operatório inicial. Entretanto, quatro meses após a cirurgia a dispersão de luz intraocular não é significantemente diferente das medidas pré-operatórias.
ABSTRACT Purpose: To evaluate ocular straylight before and after photorefractive keratectomy (PRK) for low myopia with and without topical mitomycin (MMC) treatment. Methods: Patients who underwent PRK for low myopia were enrolled into the study. PRK without MMC was performed in 21 eyes (12 patients), whereas PRK with topical 0.02% MMC was performed in 25 eyes (14 patients). Both groups were treated using the NIDEK EC5000 excimer laser. Measurements were performed using the C-Quant straylight meter preoperatively and at two and four months postoperatively. Results: The mean patient age was 30 ± 4 years, and the mean spherical equivalent refractive error was -2.2 ± 0.75 D. The mean preoperative intraocular straylight values were 1.07 ± 0.10 in the PRK without MMC group and 1.07 ± 0.11 log(s) in the PRK with topical MMC group. At two months after surgery, there was a decrease in mean intraocular straylight values in both groups. However, a significant difference was only reached in the PRK with MMC group [0.98 ± 0.09 log(s), p=0.002] compared with preoperative values, which was likely due to a greater scatter of measurements in the PRK without MMC group [1.03 ± 0.13 log(s), p=0.082]. At four months postoperatively, ocular straylight values were not significantly different compared with those at baseline in either the PRK without MMC group [1.02 ± 0.14 log(s), p=0.26] or in the PRK with topical MMC group [1.02 ± 0.11 log(s), p=0.13]. Conclusion: PRK for low myopia decreases ocular straylight, and MMC application further reduces straylight in the early postoperative period. However, ocular straylight values do not significantly differ at four months after surgery compared with those at baseline.

RESUMO: Palma forrageira foi usada como fonte de nutrientes para a produção de enzimas celulolíticas durante a fermentação em estado sólido com auxílio dos fungos filamentosos Aspergillus niger e Rhizopus sp. Os efeitos da ativiade de água, do tempo de fermentação e da temperatura sobre a produção enzimática foram determinados segundo o protocolo padrão de Delineamento Box-Behnken. A condição ideal para produção das enzimas endoglucanase pelo fungo Aspergillus niger foi 70,35 h de fermentação, atividade de água de 0,875 e temperatura de 29,56 ° C enquanto que para o fungo Rhizopus sp. foi 68,12 h e 30,41 ° C. Entretanto, a celulase total produzida pelo fungo Rhizopus sp. foi otimizada em 74,27 h de fermentação e temperatura de 31,22 ° C e para o fungo Aspergillus niger foi 72,48 h e 27,86 ° C. A atividade de água só tinha uma influência significativa sobre a endoglucanase para Aspergillus niger. A alta estabilidade dos extratos enzimáticos em diferentes tepenraturas e pH's foram confirmadas após caracterização enzimática, indicando viabilidade em aplicações industriais. Os ensaios de desativação cinética indicaram que essas enzimas permaneceram ativas após o congelamento do extrato bruto. Com base nos resultados, utilizar a palma forrageira com matéria prima em bioprocessos é uma alternativa excelente para a produção de enzimas termoestaveis.
ABSTRACT: Prickly palm cactus husk was used as a solid-state fermentation support substrate for the production of cellulolytic enzymes using Aspergillus niger and Rhizopus sp. A Box-Behnken design was used to evaluate the effects of water activity, fermentation time and temperature on endoglucanase and total cellulase production. Response Surface Methodology showed that optimum conditions for endoglucanase production were achieved at after 70.35 h of fermentation at 29.56°C and a water activity of 0.875 for Aspergillus niger and after 68.12 h at 30.41°C for Rhizopus sp. Optimum conditions for total cellulase production were achieved after 74.27 h of fermentation at 31.22°C for Aspergillus niger and after 72.48 h and 27.86°C for Rhizopus sp. Water activity had a significant effect on Aspergillus niger endoglucanase production only. In industrial applications, enzymatic characterization is important for optimizing variables such as temperature and pH. In this study we showed that endoglucanase and total cellulase had a high level of thermostability and pH stability in all the enzymatic extracts. Enzymatic deactivation kinetic experiments indicated that the enzymes remained active after the freezing of the crude extract. Based on the results, bioconversion of cactus is an excellent alternative for the production of thermostable enzymes.

Approaches to capturing the Black and White Tegu Salvator merianae (Squamata: Teiidae)

ABSTRACT The use of traps is extremely important in several types of ecological studies, and may assist in the capture of individuals in areas that are difficult to access. In the present study, we compared the effectiveness of wooden (Schramm) versus "Tomahawk" traps to capture Salvator merianae (Duméril & Bibron, 1839) lizards. The study was conducted in Eldorado do Sul, Rio Grande do Sul, Brazil. Field data were collected from August 2013 to March 2015, during the reproductive period of the species. The study involved two types of baited traps: i) "Tomahawk", made of galvanized steel; and ii) Schramm, a wooden trap. The capture rate of the Schramm wooden traps was 1.63 individuals/day, and of the "Tomahawk" was 0.36 individuals/day. These results are important for researchers working with large lizards and may help to increase sampling efficiency for these organisms.

OBJECTIVE To assess adherence to clinical appointments by health care workers (HCW) and students who suffered accidents with potentially infectious biological material. METHOD A retrospective cross-sectional study that assessed clinical records of accidents involving biological material between 2005 and 2010 in a specialized unit. RESULTS A total of 461 individuals exposed to biological material were treated, of which 389 (84.4%) were HCWs and 72 (15.6%) students. Of the 461 exposed individuals, 307 (66.6%) attended a follow-up appointment. Individuals who had suffered an accident with a known source patient were 29 times more likely to show up to their scheduled follow-up appointments (OR: 29.98; CI95%: 16.09-55.83). CONCLUSION The predictor in both univariate and multivariate analyses for adherence to clinical follow-up appointment was having a known source patient with nonreactive serology for the human immunodeficiency virus and/or hepatitis B and C.

MÉTODO Estudio de corte transversal, de carácter retrospectivo, que evaluó las fichas de atenciones referentes a los accidentes con material biológico ocurridos de 2005 a 2010, en una unidad especializada. RESULTADO Fueron atendidas 461 personas, siendo 389 (84,4%) profesionales y 72 (15,6%) estudiantes del área de la salud que sufrieron exposición a material biológico. La asistencia al seguimiento clínico fue realizada por 307 (66,6%) de los accidentados. Los sujetos víctimas de accidente con paciente fuente identificado tuvieron 29 veces más probabilidad de acudir a los retornos citados (OR: 29,98; IC95%: 16,09-55,83). CONCLUSIÓN Tanto en el análisis univariado como en el multivariado el pronosticador para la asistencia al seguimiento clínico fue ser el paciente fuente conocido y con serología no reactiva para los virus de la inmunodeficiencia humana y/o las hepatitis B y C.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Resumo: Para determinar as concentrações plasmáticas de proteínas e metabólitos de cascavéis em cativeiro, foram utilizadas 60 serpentes adultas, sendo 30 machos e 30 fêmeas. O sangue foi coletado através de punção do seio venoso paravertebral cervical e armazenado em tubos com heparina. As análises bioquímicas foram processadas colorimetricamente em Analisador Automático de Bioquímica Chemwell (Awareness Technology®, Inc). Foram calculadas as médias e desvios padrão dos seguintes constituintes: proteínas totais, albumina, globulinas, relação albumina/globulinas, ácido úrico, creatinina, ureia, colesterol, colesterol HDL e triglicérides. Os valores obtidos foram semelhantes aos descritos na literatura para repteis e serpentes, sendo as diferenças observadas provavelmente decorrentes da diferença entre espécies, clima, estação do ano e metodologia utilizada. Não houve diferenças significativas entre machos e fêmeas para os parâmetros estudados. Estes resultados podem ser úteis no estabelecimento de valores de referência para planos de conservação destes ofídios em cativeiro.
Abstract: For determining plasma concentrations of proteins and metabolites of rattlesnakes in captivity, 60 adult snakes, 30 males and 30 females were used. Blood was collected by puncture of the cervical paravertebral venous sinus and stored in tubes with heparin. Biochemical analyzes were colorimetrically processed using an Automatic Biochemistry Analyzer Chemwell (Awareness Technology®, Inc). The mean and standard deviation were calculated for the following constituents: total protein, albumin, globulin, albumin/globulin ratio, uric acid, creatinine, urea, cholesterol, HDL cholesterol and triglycerides. The values were similar to those previously reported for reptiles and snakes, with the differences observed probably due to the difference between species, climate, season and the methodology used. There were no significant differences between males and females for the parameters studied. These results may be useful in establishing normal biochemical values for conservation plans for these snakes in captivity.

Neste trabalho desenvolveu-se um processo alternativo para separação cromatográfica dos enantiômeros (+)-(3R,4R) e ( - )-(3S,4S) do β-aminoácido quiral trans-2,2,5,5-tetrametilpirrolidina-3-amino-4-carboxílico (POAC), que estava protegido no grupo amínico para posterior ligação a um peptídeo, polímero ou outra macromolécula. A enantioseparação foi obtida por HPLC usando uma fase estacionária normal à base de celulose quiral e eluição isocrática. O sistema n-hexano:isopropanol, sempre com maior quantidade do primeiro solvente, foi usado como fase móvel, pois forneceu os melhores resultados na separação dos dois componentes, constatado pelos valores mais elevados de fator de separação e de índice de resolução cromatográfica. Estes parâmetros apresentaram valores de 3,7 e 18,4 e de 2,0 e 6,7 nas soluções com proporção 90:10 (v/v) e 80:20 (v/v) de n-hexano:isopropanol, respectivamente. Estes dados indicam que a estratégia de purificação cromatográfica em uma única etapa usando fase normal é viável, abrindo assim a perspectiva de uma produção rápida e em grande escala desta sonda paramagnética.
In this work an alternative chromatographic process was developed for fractionating the (+)-(3R,4R) and ( - )-(3S,4S) enantiomers of the chiral trans β-amino acid trans-2,2,5,5-tetramethylpyrrolidine-3-amino-4-carboxylic acid (POAC), which was protected at its amine group for further coupling to a peptide, polymer or other macromolecule. The HPLC enantioseparation was achieved using a chiral cellulose-based normal stationary phase and isocratic elution. The n-hexane:isopropanol system, always with greater amount of the former component, was used as mobile phase as revealed by improved fractionation property of both components, demonstrated by the separation factor and resolution index values. These parameters presented values of 3.7 and 18.4 and of 2.0 and 6.7 when in 90:10 (v/v) and 80:20 (v/v) n-hexane:isopropanol solutions, respectively. These findings indicate that the one-step chromatographic purification strategy using normal-phase is feasible, thus opening the perspective of a fast large-scale production this paramagnetic spin probe.
