ABSTRACT Luis Tavares revolutionized cardiac surgery, always bringing the most modern instruments and equipment from his travels to England - surgical forceps, scissors, scalpels, etc. He always insisted that he was not just a thoracic surgeon, for his work extended over a wide field and created three important cardiac surgery centers which promoted a great development of cardiology. He carried out the first open heart surgery (atrial septal defect) employing extracorporeal circulation and closure of a ventricular septal defect with deep surface hypothermia of north and northeast Brazil. He promoted an intense scientific exchange program between Recife and England, resulting in significant advances in medicine, and participated directly in the creation of HEMOPE), leading to radical changes and improvements in blood therapy in the whole country. The PROCAPE, inaugurated in 2006, was the result of the cardiac center created by him in early 1970 at Hospital Oswaldo Cruz and can be considered the second largest public-university cardiology center in Brazil. He is thus widely regarded as an outstanding name in medicine in the 20th century and one of the fathers of modern cardiac surgery in Brazil.

Resumo O presente estudo analisa os limites do controle judicial sobre os atos políticos. De modo delimitado, examina, com fundamento na political questions doctrine, as decisões judiciais do Supremo Tribunal Federal que suspenderam nomeações de Ministro de Estado, no período compreendido entre 1988 e 2020. Assim, busca-se responder ao seguinte problema: sob a perspectiva da doutrina das questões políticas, em que ponto se situam os limites às decisões judiciais do Supremo Tribunal Federal que suspendem nomeações de Ministro de Estado? Esta pesquisa é dividida em duas partes. Na primeira, sintetizam-se as principais compreensões jurisprudenciais e doutrinárias sobre a doutrina das questões políticas tanto no ordenamento jurídico norte-americano quanto no brasileiro. Na segunda, aborda-se a nomeação de Ministro de Estado na ordem constitucional brasileira de 1988, com ênfase para a dogmática do Direito Administrativo brasileiro e para a jurisprudência do STF. A hipótese levantada, que foi confirmada como conclusão, é de que a nomeação de Ministro de Estado, sendo de natureza estritamente política, configura ato judicialmente insindicável, salvo quanto aos requisitos formais previstos no ordenamento jurídico. Utiliza-se o método dialético acrescido de pesquisa bibliográfica e jurisprudencial realizadas com suporte no ordenamento jurídico brasileiro e em ordenamentos jurídicos estrangeiros, em um diálogo interdisciplinar.

Abstract This study analyzes the limits of judicial control over political acts. In a limited way, it examines, based on the political questions doctrine, the judicial decisions of the Federal Supreme Court that suspended appointments of Minister of State, in the period between 1988 and 2020. Thus, it seeks to answer the following problem: from the perspective of the political questions doctrine, at what point are the limits of the Supreme Court's judicial decisions that suspend Minister of State appointments? This research is divided into two parts. The first part summarizes the main jurisprudential and doctrinal understandings about the doctrine of political questions in both the American and Brazilian legal systems. In the second part, the appointment of Minister of State in the Brazilian constitutional order of 1988 is addressed, with emphasis on the dogmatics of Brazilian Administrative Law and the jurisprudence of the STF. The hypothesis raised, which was confirmed as a conclusion, is that appointments of Ministers of State, being of a strictly political nature, are judicially unreviewable acts, except with regard to the formal requirements provided for in the legal system. The dialectical method is used plus bibliographical and jurisprudential research carried out with support in the Brazilian legal system and in foreign legal systems, in an interdisciplinary dialogue.

Abstract Constant growth in the concrete floor sector claims new techniques to improve concrete performance and avoid undesirable issues. In this way, materials used in concrete floors are essential, and it is critical to investigate them. Therefore, this study evaluated the influence of surface hardeners on the surface hardness determined by the rebound hammer test of concrete floors prepared with distinct water/cement ratios, curing ages, and cement types. The control parameters of the concrete’s production, including the compressive strength, flexural tensile strength, and bleeding tests were analyzed. In addition, data were assessed by multifactorial analysis of variance (ANOVA). Thus, hardeners increased the surface resistance of the composites, therefore reducing the chances of pathological manifestations occurring. Taken together, we demonstrated that hardeners improved the concrete surface prepared with all mixture proportions, but it was more significant when using the cementitious hardener and higher w/c content (0.6).

Abstract Radiation-induced secondary tumors constitute a rare complication of radiation therapy and typically occur in or near the irradiated area. Women who undergo thoracic radiotherapy before 30 years of age have a significantly greater lifetime risk of developing breast cancer than do those in the general population. It is recommended that a patient who has previously received thoracic radiotherapy with a radiation dose ≥ 10 Gy subsequently undergo annual screening with mammography and magnetic resonance imaging, beginning eight years after the initial treatment or when the patient has reached 25 years of age (whichever comes later). The treatment of secondary breast cancer in this population should be individualized and should be discussed with a multidisciplinary team to avoid toxicity related to the treatment of the primary cancer.

Resumo Os cânceres secundários induzidos por radiação são complicações raras da radioterapia e geralmente ocorrem na área tratada ou próximo a ela. Mulheres com menos de 30 anos de idade tratadas com radioterapia torácica têm risco significativamente aumentado de desenvolver câncer de mama ao longo da vida, em comparação com a população em geral. Para as pacientes tratadas com irradiação torácica com dose igual ou superior a 10 Gy é recomendado rastreamento anual com mamografia e ressonância magnética iniciando oito anos após o tratamento, desde que este início seja após 25 anos de idade. O tratamento do câncer de mama secundário nessa população deve ser individualizado e discutido com equipe multidisciplinar, para evitar toxicidade relacionada ao tratamento do câncer primário.

Abstract Background People with multiple sclerosis (PwMS) show an increased risk of sexual dysfunction (SD), both in women and men. Objective The aim of the present study was to apply the Multiple Sclerosis Intimacy and Sexuality Questionnaire-19 (MSISQ-19) and evaluate our results by comparing them with those in in the literature, as well as to assess the ease of applying the scale and the engagement of the patients in discussing the topic of sexuality. Methods We developed and applied a web-based Google form questionnaire that the respondents completed online, which included the MSISQ-19, for the assessment of sexual function. Baseline characteristics were reported as proportions and mean ± standard deviation (SD) or median ± interquartile range (IQR) as appropriate according to data distribution. Categorical variables were stratified by sex and compared with chisquared tests. Statistical analyses were performed using STATA v. 16 (StataCorp., College Station, TX, USA). Results Of the 621 respondents, 541 were included in the analysis. Among the patients with MS, a total of 347 (64.14%) exhibited SD. When stratified by gender, the frequencies of SD were not significantly different. Conclusion There is a high incidence of sexual dysfunction among PwMS and we need to identify the reasons for this and implement strategies to treat and counsel our patients. The MSISQ-19 can be used to help clinicians to assess sexual functioning in a quick and easy way and give patients the possibility to address this topic and receive appropriate help and support.

Resumo Antecedentes Pacientes com esclerose múltipla apresentam altas taxas de disfunção sexual em diversos estudos. Objetivo Avaliar a aplicação e resultados da ferramenta para avaliação de disfunção sexual em pacientes com Esclerose Multipla, Multiple Sclerosis Intimacy and Sexuality Questionnaire-19 (MSISQ-19), bem como comparar os resultados encontrados com a literatura já existente e ter a percepção do engajamento dos pacientes sobre o tema. Métodos Realizado através de questionário on-line na plataforma Google Forms, composto por questões sobre perfil demográfico e relacionadas à doença e o questionário MSISQ-19. Os dados demográficos e sobre a doença foram descritos como proporções e média ± desvio padrão (DP) ou mediana ± intervalo interquartil (IIQ). As variáveis categóricas foram estratificadas por sexo e comparadas com testes de qui-quadrado. As análises estatísticas foram realizadas no programa STATA v. 16 (StataCorp., College Station, TX, EUA). Resultados Um total de 621 pacientes responderam ao questionário, sendo inclusos na análise 541 respostas, após a aplicação dos critérios de elegibilidade. Um total de 347 (64,14%) pacientes apresentaram disfunção sexual, sem diferença entre os gêneros. Conclusão Disfunção sexual apresenta alta prevalência no grupo estudado, sendo necessário identificar as causas e implementar estratégias de tratamento. O MSISQ-19 é uma ferramenta fácil e rápida de ser aplicada, podendo ser utilizada para facilitar a comunicação com os pacientes sobre o tema, possibilitando assim o tratamento das disfunções sexuais nessa população.

Abstract Objective Research and identification of Cutibacterium acnes (C. acnes) and other microorganisms in deeptissue samples collected in clean shoulder surgeries of patients who did not undergo any previous invasive joint procedure and who had no clinical history of infection. Methods We analyzed the results of cultures of intraoperative deep tissue samples from 84 patients submitted to primary clean shoulder surgery. Tubes containing culture medium were used for storage and transport of anaerobic agents, prolonged incubation time, and mass spectrometer for diagnosis of bacterial agents. Results Bacteria growth was evidenced in 34 patients (40.4%) of the 84 included in the study. Of these, 23 had growth of C. acnes in at least one sample of deep tissue collected, corresponding to 27.3% of the total patients. The second most common agent was Staphylococcus epidermidis, present in 7.2% of the total individuals included. We showed a higher relationship between sample positivity and males, a lower mean age, absence of diabetes mellitus, ASA I score, and antibiotic prophylaxis in anesthetic induction with cefuroxime. Conclusions A high percentage of isolates of different bacteria was found in shoulder tissue samples of patients undergoing clean and primary surgeries, who had no history of previous infection. Identification of C. acnes was high (27.6%), and Staphylococcus epidermidis was the second most frequent agent (7.2%).

Resumo Objetivo Pesquisa e identificação de Cutibacterium acnes (C. acnes) e de outros microrganismos em amostras de tecidos profundos coletados em cirurgias limpas de ombro em pacientes que não foram submetidos a nenhum procedimento invasivo articular prévio e que não possuíam antecedentes clínicos de infecção. Métodos Foram analisados os resultados das culturas de amostras de tecidos profundos intraoperatórias de 84 pacientes submetidos à cirurgia limpa primária do ombro. Foram utilizados tubos contendo meio de cultivo para armazenamento e transporte de agentes anaeróbicos, tempo prolongado de incubação e espectrômetro de massa para diagnósticos de agentes bacterianos. Resultados Foi evidenciado o crescimento de bactérias em 34 pacientes (40,4%) dos 84 incluídos no estudo. Desses, 23 apresentavam crescimento de C. acnes em pelo menos uma amostra de tecido profundo coletada, correspondendo a 27,3% do total de pacientes. O segundo agente mais encontrado foi o Staphylococcus epidermidis, presente em 7,2% do total de indivíduos incluídos. Evidenciamos maior relação da positividade de amostras com o gênero masculino, uma média de idade inferior, a ausência de diabetes mellitus, o escore ASA I e a profilaxia antibiótica na indução anestésica com cefuroxima. Conclusões Verificou-se um elevado percentual de isolados de diferentes bactérias em amostras de tecidos de ombros de pacientes submetidos a cirurgias limpas e primárias e sem histórico de infecção anterior. A identificação de C. acnes foi elevada (27,6%) e o Staphylococcus epidermidis foi o segundo agente mais frequente (7,2%).

Abstract The expansion of residential areas has increased the demand for exotic and increasingly compact landscape plants. In this context, this work aimed to evaluate the effects of paclobutrazol (PBZ) and the timing of ethylene application on growth reduction and flowering anticipation in ornamental pineapples grown in pots. The randomized block design was used with factorial arrangement (2 x 5) and 4 replications and 4 plants per plot. The primary treatments are the presence and absence of PBZ. The secondary treatments were five times of floral induction with ethylene: 90; 120; 150; 180; 210 days after transplanting (DAT) the seedlings into the pots. It was evaluated the variables: height ratio between the heights of the pot and the plant; rosette diameter; leaves length ‘D’ and flowering index. At 255 DAT, although the plants did not respond to floral induction with ethylene, only those treated with PBZ were more compact and had different characteristics such as smaller size and a ratio below 1/3 of the height of the vase in relation to the plant that to favor its commercialization, in pot.

Resumo A diminuição das áreas residenciais tem aumentado a demanda por plantas paisagísticas exóticas e cada vez mais compactas. Neste contexto, o objetivo deste trabalho foi avaliar os efeitos do paclobutrazol (PBZ) e das épocas de aplicação de etileno na redução do crescimento e na antecipação da floração em abacaxizeiro ornamental cultivado em vaso. O delineamento experimental utilizado foi o de blocos ao acaso, em arranjo fatorial (2 x 5), com 4 repetições e 4 plantas por parcela. Os tratamentos primários foram a presença e a ausência de PBZ. Os tratamentos secundários foram cinco épocas de indução floral com etileno: 90; 120; 150; 180; 210 dias após o transplantio (DAT) das mudas, para os vasos. Foram avaliadas as variáveis: altura da planta; relação entre as alturas do vaso e da planta; diâmetro da roseta; comprimento da folha ‘D’ e índice de florescimento. Aos 255 DAT, apesar das plantas não responderem à indução floral com o etileno, apenas aquelas tratadas com PBZ apresentaram-se mais compactas e com características diferenciadas como menor porte e relação abaixo de 1/3 da altura do vaso em relação a da planta que favorecer sua comercialização, em vaso.

Abstract In this work, measuring capabilities of a Sagnac Interferometer-based Distributed Optical Fiber Sensor (DOFS) system are demonstrated by comparing its results to a conventional amplitude-based Distributed Acoustic Sensing (DAS) system. Through a set of three different experiments, it was demonstrated that both sensing systems can detect and locate dynamical deformation along the fiber, in addition to determining its frequency components. The coherent amplitude-based DAS presented a lower background noise. In contrast, even though the Sagnac interferometry-based system presented low-frequency noise in the measurements, it is less susceptible to harmonic distortion for higher amplitude deformations since its sensing principles rely on the measurement of the accumulated phase rather than the amplitude of the backscattered signal purely. Abstract This comprehensive review compares clinical protocols of important entities regarding the management of fetal growth restriction (FGR), published since 2015. Five protocols were chosen for data extraction. There were no relevant differences regarding the diagnosis and classification of FGR between the protocols. In general, all protocols suggest that the assessment of fetal vitality must be performed in a multimodally, associating biophysical parameters (such as cardiotocography and fetal biophysical profile) with the Doppler velocimetry parameters of the umbilical artery, middle cerebral artery, and ductus venosus. All protocols reinforce that the more severe the fetal condition, the more frequent this assessment should be made. The timely gestational age and mode of delivery to terminate the pregnancy in these cases can vary much between the protocols. Therefore, this paper presents, in a didactic way, the particularities of different protocols for monitoring FGR, in order to help obstetricians to better manage the cases.

Resumo Esta revisão compreensiva compara protocolos clínicos de entidades importantes em relação ao manejo da restrição de crescimento fetal (RCF), publicados desde 2015. Cinco protocolos foram escolhidos para a extração de dados. Não houve diferenças relevantes quanto ao diagnóstico e classificação da RCF entre os protocolos. Em geral, todos os protocolos sugerem que a avaliação da vitalidade fetal deve ser realizada de forma multimodal, associando parâmetros biofísicos (como cardiotocografia e perfil biofísico fetal) aos parâmetros dopplervelocimétricos da artéria umbilical, artéria cerebral média e ducto venoso. Todos os protocolos reforçam que quanto mais grave a condição fetal, mais frequente essa avaliação deve ser feita. A idade gestacional oportuna e o modo de parto para interromper a gravidez nesses casos podem variar muito entre os protocolos. Portanto, este trabalho apresenta, de forma didática, as particularidades de diferentes protocolos de acompanhamento de RCF, a fim de auxiliar os obstetras no melhor manejo dos casos.

RESUMO OBJETIVO Estimar a prevalência da exposição ao vírus SARS-CoV-2 entre indivíduos vivendo em restrição de liberdade. MÉTODOS Foi realizado inquérito de soroprevalência com a população da penitenciária feminina do Centro de Progressão Penitenciária (CPP) do Butantan (município de São Paulo), entre 24 de junho e 20 de agosto de 2020. Nesse período, segundo a Secretaria de Administração Penitenciária (SAP), a positividade dos testes rápidos entre detentos variou de 65 a 78%. O método de avaliação utilizado no estudo foi o teste rápido “One Step COVID-19” (cromatografia), da empresa Wondfo, empregando-se também o método RT-PCR em participantes sintomáticos para confirmação do quadro viral. A população do estudo foi constituída por 879 reeducandas e 170 funcionários da instituição. RESULTADOS A prevalência de anticorpos totais (IgG/IgM) contra o vírus SARS-CoV-2 na população total de 1.049 participantes do estudo foi de 6,1%; entre a população de 879 reeducandas foi observada prevalência de 5,8% e entre os servidores da instituição, 7,5%. CONCLUSÃO Houve baixa prevalência de covid-19 no CPP do Butantan, o que se deve à implementação de medidas de prevenção simples na instituição, como o uso de máscaras (com trocas adequadas), ênfase na higiene, lavagem das mãos e distanciamento social, além de outras estratégias, como suspensão de visitas de familiares e amigos das reeducandas, cortes de consultas médicas eletivas e do trabalho externo.

ABSTRACT OBJECTIVE To estimate the prevalence of exposure to the SARS-CoV-2 virus among individuals living in restricted freedom. METHODS A seroprevalence survey was carried out with the population of the female penitentiary of the Centro de Progressão Penitenciária (CPP) in Butantan (municipality of São Paulo), between June 24 and August 20, 2020. During this period, according to the Secretariat of Penitentiary Administration (SAP), the positivity of rapid tests among inmates ranged from 65% to 78%. The evaluation method used in the study was the “One Step COVID-19” rapid test (chromatography), from the company Wondfo, also using the RT-PCR method in symptomatic participants to confirm the viral condition. The study population consisted of 879 female inmates and 170 employees of the institution. RESULTS The prevalence of total antibodies (IgG/IgM) against the SARS-CoV-2 virus in the total population of 1049 study participants was 6.1%; among the population of 879 inmates,a prevalence of 5.8% was observed, and among the institution’s employees, 7.5%. CONCLUSIONS The prevalence of covid-19 at the Butantan CPP was low, which is due to the implementation of simple prevention measures at the institution, such as the use of masks (with appropriate changes), emphasis on hygiene, hand washing and social distancing, in addition to other strategies, such as suspending inmates’ visits from relatives and friends and cutting back on elective medical appointments and outside work.

RESUMO: Objetivou-se mensurar o efeito da associação entre caracteres agronômicos relacionados à produtividade de grãos de canola cultivada em diferentes épocas de semeadura, através da análise de trilha. Assim como também objetivou-se obter um método para predizer o teor de óleo nos grãos, ajustando um modelo multivariado através da análise por espectroscopia na região do infravermelho próximo. O experimento foi conduzido em campo, utilizando-se o delineamento de blocos ao acaso, em parcelas subdivididas no tempo, sendo quatro parcelas (épocas de semeadura) e seis subparcelas (híbridos de canola), com quatro repetições. Em cada híbrido foram realizadas observações fenológicas e determinada a produtividade de grãos. Os dados foram submetidos à análise de variância em ambiente R pelo teste F, a cinco de probabilidade. O teor de óleo nos grãos foi determinado pelo método químico tradicional, e com base na assinatura espectral no infravermelho próximo de amostras dos grãos foi estabelecida regressão dos mínimos quadrados parciais (PLS-R) para estimar o teor de óleo nos grãos de canola. As épocas de semeadura influenciaram os componentes de produção e o teor de óleo dos grãos de todos híbridos. Os caracteres número de grãos em cinco plantas (0,6857) e altura (0,4943) apresentaram maiores estimativas de correlação positiva com a produtividade de grãos, assim como os maiores valores de efeito direto positivo sobre a produtividade, 0,2494 e 0,1595 respectivamente. Entretanto, o ciclo total (-0,7848), juntamente com dias em florescimento (-0,4520) apresentou correlação significativa negativa com a produtividade. A técnica NIR associada à PLS-R foi capaz de predizer o teor de óleo nos grãos, resultando em bons modelos preditivos (R2 de 0,86 e RMSE de 1,56 na validação externa) que podem ser usados com sucesso na análise da qualidade das amostras após colheita e nos programas de melhoramento genético.

ABSTRACT: This study measured the effect of the association between agronomic traits related to the yield of canola grains grown at different sowing dates through path analysis. Another objective was to obtain a method to predict the oil content in the grains, fitting a multivariate model through near-infrared (NIR) spectroscopy analysis. The experiment was conducted in the field using a randomized block design in plots subdivided by time, with four plots (sowing dates), six subplots (canola hybrids), and four replicates. In each hybrid, phenological observations were performed, and the grain yield was determined. The data were subjected to analysis of variance in the R environment using the F test at 5% probability. The oil content in the grains was determined by the traditional chemical method, and based on the NIR spectral signature of the grain samples, partial least squares regression (PLS-R) was established to estimate the oil content in the canola grains. The sowing dates influenced the production components and oil content of the grains of all hybrids. The trait number of grains in five plants (0.6857) and their height (0.4943) had greater estimates of positive correlations with grain yield, as well as higher values of positive direct effects on yield (0.2494 and 0.1595, respectively). The NIR technique combined with PLS-R was able to predict the oil content in the grains, resulting in good predictive models (R2 of 0.86 and root mean square error (RMSE) of 1.56 in external validation).

Abstract The TP53 3’UTR variant rs78378222 A>C has been detected in different tumor types as a somatic alteration that reduces p53 expression through modification of polyadenylation and miRNA regulation. Its prevalence is not yet known in all tumors. Herein, we examine tumor tissue prevalence of rs7837822 in Brazilian cohorts of patients from south and southeast regions diagnosed with lung adenocarcinoma (LUAD, n=586), sarcoma (SARC, n=188) and uterine leiomyoma (ULM, n=41). The minor allele (C) was identified in heterozygosity in 6/586 LUAD tumors (prevalence = 1.02 %) and none of the SARC and ULM samples. Additionally, next generation sequencing analysis revealed that all variant-positive tumors (n=4) with sample availability had additional pathogenic or likely pathogenic somatic variants in the TP53 coding regions. Among them, 3/4 (75 %) had the same pathogenic or likely pathogenic sequence variant (allele frequency <0.05 in tumor DNA) namely c.751A>C (p.Ile251Leu). Our results indicate a low somatic prevalence of rs78378222 in LUAD, ULM and SARC tumors from Brazilian patients, which suggests that no further analysis of this variant in the specific studied regions of Brazil is warranted. However, these findings should not exclude tumor molecular testing of this TP53 3’UTR functional variant for different populations.

ABSTRACT Background: Egg collection traps have been studied to assist in the integrated control of arbovirus vectors. Many enhancements have been made over the years. Methods: This study evaluated the use of a hydrated superabsorbent polymer (SAP) in the egg collection of Aedes aegypti and Aedes albopictus in ovitraps. An experiment was conducted in the laboratory to determine the minimum concentration of the product to be used in traps in the field to prevent the development of larvae into adults. In the field, the use of polymers has been evaluated using a traditional model of ovitraps. The positive ovitrap index and mean number of eggs per trap indicator were calculated. Results: In the laboratory, the larvae did not successfully develop to the adult stage, even at the lowest SAP concentration. In the field, the results showed that ovitraps with SAP proved to be effective for egg collection from both species. It was possible to identify sites with the highest concentration of species and expose the ovitraps for a longer period without larval development. Conclusions: There is a need for studies on the adequacy of this technology for control programs. However, the results showed that ovitraps with hydrogel were potentiated to capture eggs, configuring themselves as another tool for vector control.

Abstract Objectives Evidence suggests that ketamine’s influence on brain-derived neurotrophic factor (BDNF) might be involved in its mechanism of rapid antidepressant action. We aimed to evaluate the differential impact of ketamine and esketamine on serum BDNF levels and its association with response patterns in treatment-resistant depression (TRD). Methods Participants (n = 53) are from a randomized, double-blind clinical trial comparing the efficacy of single-dose ketamine (0.5mg/kg, n = 27) and esketamine (0.25mg/kg, n = 26) in TRD. Depression severity was assessed before and 24 hours, 72 hours, and 7 days after the intervention, using the Montgomery-Åsberg Depression Rating Scale (MADRS). Blood samples were collected before infusion, 24 hours, and 7 days afterwards. Results There were no significant changes in BDNF levels at post-infusion evaluation points, and no difference in BDNF levels comparing ketamine and esketamine. Both drugs exhibited similar therapeutic effect. There was no association between BDNF levels and response to treatment or severity of depressive symptoms. Conclusion There was no significant treatment impact on BDNF serum levels – neither with ketamine nor esketamine – despite therapeutic response. These results suggest that ketamine or esketamine intervention for TRD has no impact on BDNF levels measured at 24 hours and 7 days after the infusion. Abstract This study aimed to develop a protocol for the cryopreservation of Pseudoplatystoma corruscans semen. For this, mature males were hormonally induced with a single dose of carp pituitary extract (5 mg/kg body weight). Semen was collected and evaluated. Two cryoprotectants were tested to compose the diluents: dimethyl acetamide (DMA) and dimethyl sulfoxide (Me2SO), in two concentrations (8% and 10%), + 5.0% glucose + 10% egg yolk. The semen was diluted in a 1: 4 ratio (semen: extender), packed in 0.5 mL straws and frozen in a dry shipper container in liquid nitrogen vapors. After thawing, sperm kinetics, sperm morphology and DNA integrity of cryopreserved sperm were evaluated. Pseudoplatystoma corruscans males produced semen with sperm motility > 80%. After thawing, all treatments provided semen with total sperm motility > 40%, with no significant difference (P < 0.05) between them, as well as between the other sperm kinetic parameters evaluated. The treatments with DMA provided a smaller fragmentation of the DNA of the gametes. Sperm malformations were identified in both fresh and cryopreserved semen, with a slight increase in these malformations being identified in sperm from thawed P. corruscans semen samples.

Resumo Este estudo teve como objetivo desenvolver um protocolo para a criopreservação do sêmen de Pseudoplatystoma corruscans. Para tal, machos maduros foram induzidos hormonalmente com uma dose única de extrato de hipófise de carpa (5 mg/kg de peso vivo). O sêmen foi coletado e avaliado. Sendo testados para compor os diluentes, dois crioprotetores: dimetil acetamida (DMA) e dimetil sulfóxido (Me2SO), em duas concentrações (8% e 10%), + 5,0% glicose + 10% gema de ovo. O sêmen foi diluído na proporção 1: 4 (sêmen: extensor), embalado em palhetas de 0,5 mL e congelado em container dryshipper em vapores de nitrogênio líquido. Após o descongelamento, foram avaliados os aspectos cinéticos espermáticos, a morfologia espermática e a integridade do DNA dos espermatozoides criopreservados. Os machos de P. corruscans produziram sêmen com motilidade espermática > 80%. Todos os tratamentos proporcionaram após o descongelamento sêmen com motilidade espermática total > 40%, sem diferença significativa (P < 0,05) entre eles, como também entre os demais parâmetros cinéticos espermáticos avaliados. Os tratamentos com DMA proporcionaram uma menor fragmentação do DNA dos gametas. Malformações espermáticas foram identificadas, tanto no sêmen fresco, como no criopreservado, sendo identificado um aumento discreto dessas malformações nos espermatozoides das amostras de sêmen descongeladas de P. corruscans.
