Abstract Stylosanthes (Papilionoideae, Leguminosae) is a predominantly Neotropical genus with ~48 species that include worldwide important forage species. This study presents the chromosome number and morphology of eight species of the genus Stylosanthes (S. acuminata, S. gracilis, S. grandifolia, S. guianensis, S. hippocampoides, S. pilosa, S. macrocephala, and S. ruellioides). In addition, staining with CMA and DAPI, in situ hybridization with 5S and 35S rDNA probes, and estimation of DNA content were performed. The interpretation of Stylosanthes chromosome diversification was anchored by a comparison with the sister genus Arachis and a dated molecular phylogeny based on nuclear and plastid loci. Stylosanthes species showed 2n = 20, with low cytomolecular diversification regarding 5S rDNA, 35S rDNA, and genome size. Arachis has a more ancient diversification (~7 Mya in the Pliocene) than the relatively recent Stylosanthes (~2 Mya in the Pleistocene), and it seems more diverse than its sister lineage. Our data support the idea that the cytomolecular stability of Stylosanthes in relation to Arachis could be a result of its recent origin. The recent diversification of Stylosanthes could also be related to the low morphological differentiation among species, and to the recurrent formation of allopolyploid complexes.

RESUMO A citometria de fluxo e os marcadores microssatélites foram utilizados para avaliar a fidelidade genética de plantas micropropagadas de dois clones de Eucalyptus urophylla x E. globulus e um clone de Eucalyptus grandis x E. globulus, provenientes de material adulto. Os clones foram multiplicados in vitro em 25 subcultivos sucessivos em meio de cultura MS suplementado com BA (2,22 µM) e ANA (0,05 µM). O alongamento das brotações foi obtido no meio MS acrescido de AIB (2,46 µM) e BA (0,22 µM). O enraizamento ex vitro e a aclimatização foram realizados conjuntamente. Os clones micropropagados demonstraram estabilidade genética através das análises de citometria de fluxo e de marcadores microssatélites. Os resultados revelaram que a micropropagação com fins de rejuvenescimento clonal, corresponde a uma técnica viável quanto à obtenção de clones híbridos de E. globulus, geneticamente estáveis.

ABSTRACT Flow cytometry and microsatellite markers were used to determine a genetic fidelity of micropropagated plants from the two Eucalyptus urophylla x E. globulus clones and a Eucalyptus grandis x E. globulus clone derived from adult material. Clones were repeatedly subcultured for 25 subcultures on MS medium supplemented with BA (2.22 µM) and ANA (0.05 µM) for in vitro shoot multiplication. The elongation was performed in MS culture medium supplemented with AIB (2.46 µM) and BA(0.22 µM). The ex vitro rooting and acclimatization phases were lead at the same time. The micropropagated clones showed genetic stability by flow cytometry and microsatellite markers. The results proved that micropropagation, for purposes of rejuvenation, can be a viable technique to generate genetically stable or identical E. globulus hybrid clones.

O gênero Aloysia reúne aproximadamente 30 espécies, porém sua circunscrição tem sido motivo de controvérsia. São poucos os estudos citogenéticos para o gênero, relatando apenas o número e o comportamento cromossômico. O presente trabalho teve como objetivo, identificar os caracteres citogenéticos de Aloysia virgata da flora brasileira que possam ser utilizados para um melhor entendimento e caracterização dos aspectos citogenéticos da família Verbenaceae, que possam contribuir para a organização taxonômica do grupo. Aloysia virgata apresentou cariótipo com 2n = 36 pequenos cromossomos, todos com o mesmo tamanho. Grande quantidade de heterocromatina foi observada com Giemsa e confirmada pela técnica de banda C. Foram detectadas quatro regiões organizadoras de nucléolo (NORs) e dois sítios de rDNA 5S pela técnica de hibridação in situ fluorescente (FISH). O núcleo interfásico foi classificado como semireticulado. Foi realizada a caracterização meiótica, na qual os cromossomos apresentaram comportamento normal, com a presença de 18 bivalentes na prófase I e na metáfase I. O número de cromossomos, de NORs e de segmentos de DNAr 5S não excluem uma possível origem poliplóide.

Since previous cytogenetic reports of Aloysia have only described the meiotic behavior and chromosomal number of some species, the aim of this work was to provide detailed cytogenetic description of Aloysia virgata that would contribute to the understanding of the taxonomical organization of the Verbenaceae. Aloysia virgata had a karyotype with 2n = 36 metacentric chromosomes, all with similar size. The large amount of heterochromatin seen after Giemsa staining was confirmed by C-banding. Four nucleolar organizer regions (NORs) were detected with an rDNA 45S probe in two homologous pairs and two sites of 5S rDNA located on one chromosomal pair were detected by fluorescence in situ hybridization. The interphase nucleus was classified as semi-reticulate. Meiotic analysis showed a normal chromosomal behavior, with 18 bivalents in some parts of prophase I and in metaphase I. The number of chromosomes, NORs and 5S rDNA segments did not exclude a possible polyploid origin.

Os objetivos deste trabalho foram estabelecer um protocolo eficiente de embriogênese somática, em híbridos triploides entre capim elefante (Pennisetum purpureum Schumach.) e milheto (P. glaucum (L.) R. Br.), e avaliar por citometria de fluxo a estabilidade genômica das plantas obtidas in vitro. A embriogênese somática e a regeneração das plantas foram estabelecidas a partir de embriões zigóticos maduros de híbridos entre capim elefante e milheto. Foram testados quatro tratamentos com 2,4 ácido diclorofenoxiacético (2,4 D), nas concentrações 0, 1, 2 e 3 mg L-1, para indução de calos embriogênicos, e dois tratamentos com inositol a 1 e 2 g L-1, para regeneração das plantas. Os tratamentos foram dispostos em delineamento inteiramente ao acaso. A combinação ótima de hormônios foi de 2 mg L-1 de 2,4 D, para indução de calos embriogênicos, e de 1 g L-1 de inositol, para conversão de embriões e regeneração de plantas. A análise de quantidade de DNA, por citometria de fluxo das plantas regeneradas, indicou a não ocorrência de alterações em ploidia durante a embriogênese somática e a regeneração das plantas. A quantidade de DNA nuclear e a ploidia das plantas regeneradas foram estáveis e homogêneas em comparação às das plantas controle. Não ocorreu instabilidade cariotípica no sistema de regeneração usado para híbridos de Pennisetum.

The objectives of this study were to establish an efficient protocol for somatic embryogenesis in triploid hybrids between napiergrass (Pennisetum purpureum Schumach.) and pearl millet (P. glaucum (L.) R. Br.), and to assess the genomic stability by flow cytometry of the plants obtained in vitro. Somatic embryogenesis and plant regeneration were successfully established from mature zygotic embryos of napiergrass and pearl millet hybrids. Four treatments with 2,4 dichlorophenoxyacetic acid (2,4-D) at 0, 1, 2 e 3 mg L-1 were tested for embryogenic calli induction and two treatments with inositol at 1 e 2 g L-1 were tested for plant regeneration. The treatments were arranged in a completely randomized design. The optimum hormone combinations were 2 mg L-1 of 2,4 D for embryogenic callus induction, and 1 g L-1 of inositol for embryos conversion and plant regeneration. The analysis of DNA content by flow cytometry of the regenerated plantlets indicated that no ploidy changes had been induced during somatic embryogenesis and plant regeneration. The nuclear DNA content and ploidy levels of the regenerated plants were stable and homogeneous in comparison to those of the control plants. There was no occurrence of karyological instability in the regeneration system utilized for Pennisetum hybrid.

The cytotoxicity effect of aqueous extracts of the lichens species Myelochroa lindmanii and Canoparmelia texana (Lecanorales, Parmeliaceae) were evaluated using meristematic cells of lettuce (Lactuca sativa) and maize (Zea mays). The seasonal effect was also evaluated. Extracts of M. lindmanii and C. texana inhibited root growth and/or percentage germination, possibly due to alterations in the cell cycle. The M. lindmanii extract showed anti-mitotic effects and blocked the cell cycle in metaphase so that c-mitosis and cells with chromosome duplication were produced. The C. texana extract appeared to hinder cell division, increasing the number of interphase cells. In addition, both extracts caused an increase in percentage of cell death. Clastogenic effects were also observed, such as sticky chromosomes, bridges, fragments and later segregation. Both lichen species are thus potential sources of biologically active substances with possible applications in biology, medicine and agronomy.

ABSTRACT This work aimed to verify the existence of genetic variability among three Solanum lycocarpum populations from two Brazilian ecosystems: Cerrado and Mata Atlântica. The evaluation of germination and some morphological characters were performed twenty days after seedlings emergency. The analysis of variance revealed the existence of genetic variability among the populations for germination, average length of cotyledons and average leaves number.

RESUMO O presente trabalho teve como objetivo verificar a existência de variabilidade genética entre três populações de Solanum lycocarpum estabelecidas em dois ecossistemas brasileiros: Cerrado e da Mata Atlântica. Avaliaram-se a porcentagem de germinação e seis caracteres morfológicos aos 20 dias após a emergência das plântulas. A análise de variância revelou a existência de variabilidade genética entre as populações para a porcentagem de germinação, comprimento médio do cotilédone e número médio de folhas.

O estudo dos fatores que afetam a eficiência da radiação é importante, pois facilita a obtenção de mutantes que possam servir como fonte de variabilidade em programas de melhoramento e de aberrações cromossômicas estruturais para estudos citogenéticos. O presente trabalho foi realizado com o objetivo de investigar a influência do teor de água na resposta de sementes de milho à radiação gama. Verificou-se, nos experimentos realizados, o efeito radiossensibilizador da água, uma vez que, nas sementes com maior teor de umidade, intensificaram-se os danos causados pela radiação. Verificou-se, também, que o aumento dos danos foi diretamente proporcional às doses de radiação.

The study of the factors that affect the radiation efficiency is important, because it makes easier to get mutants that may be used as source of variability on improvement programs or as structural chromossomic aberrations for cytogenetics studies. The main aim of this research was to investigate the influence of corn seed water content on gamma radiation response. As a rule, the damage caused by radiation was more evident on seeds with higher water content. Also, increased damages were observed with higher radiation doses.

A identificação de substâncias químicas que aumentam a eficiência da radiação é importante para facilitar a obtenção de plantas portadoras de aberrações cromossômicas estruturais que possam ser usadas em um programa alternativo de produção de milho híbrido (Zea mays L.). Realizou-se o presente trabalho com o objetivo de investigar o efeito da substância química metronidazol na resposta de plântulas de milho à radiação gama. Diversos tratamentos foram feitos, embebendo as sementes em soluções com variadas concentrações da substância ativa, combinadas com filtragem e radiação gama. No terceiro dia do experimento, foram avaliados a porcentagem de germinação e os comprimentos das raízes e dos caules. Em alta concentração (1.250 mg/50 mL), o metronidazol, em presença de radiação, comportou-se como radiossensibilizador e, na sua ausência, como substância tóxica, mesmo em baixa concentração (250 mg/50 mL e 750 mg/50 mL).

The identification of chemical substances which increase the efficiency of radiation is important to make easier the obtention of plants with structural chromosome aberrations which may be used in an alternative program for hybrid maize production. The present work was carried out to investigate the effect of the chemical substance metronidazole in maize seedlings submitted to gamma radiation. Several treatments were done, soaking the seeds in solutions with varied concentrations of the active substance combined with solution filtration and gamma radiation. On the third day of the experiment, germination percentage, root and stem lengths were evaluated. At a high concentration (1,250 mg/50 mL) metronidazole behaved as a radiosensibilizer in the presence of radiation. Even at a low concentration (250 mg/50 mL; 750 mg/50 mL) and in the absence of radiation, metronidazole behaved as toxic substance.
