This study investigated the acute blockade of endogenous melatonin (MLT) using Luzindole with or without systemic lipopolysaccharide (LPS) challenge and evaluated changes in inflammatory and oxidative stress markers in the mouse jejunum. Luzindole is an MT1/MT2 MLT receptor antagonist. Both receptors occur in the small intestine. Swiss mice were treated with either saline (0.35 mg/kg, ip), Luzindole (0.35 mg/kg, ip), LPS (1.25 mg/kg, ip), or Luzindole+LPS (0.35 and 1.25 mg/kg, ip, respectively). Jejunum samples were evaluated regarding intestinal morphometry, histopathological crypt scoring, and PAS-positive villus goblet cell counting. Inflammatory Iba-1, interleukin (IL)-1β, tumor necrosis factor (TNF)-α, nuclear factor (NF)-kB, myeloperoxidase (MPO), and oxidative stress (NP-SHs, catalase, MDA, nitrate/nitrite) markers were assessed. Mice treated with Luzindole, LPS, and Luzindole+LPS showed villus height shortening. Crypt damage was worse in the LPS group. Luzindole, LPS, and Luzindole+LPS reduced the PAS-goblet cell labeling and increased Iba-1-immunolabelled cells compared to the saline group. Immunoblotting for IL-1β, TNF-α, and NF-kB was greater in the Luzindole group. The LPS-challenged group showed higher MPO activity than the saline and Luzindole groups. Catalase was reduced in the Luzindole and Luzindole+LPS groups compared to saline. The Luzindole group showed an increase in NP-SHs, an effect related to compensatory GSH activity. The acute blockade of endogenous MLT with Luzindole induced early changes in inflammatory markers with altered intestinal morphology. The other non-detectable deleterious effects of Luzindole may be balanced by the unopposed direct action of MLT in immune cells bypassing the MT1/MT2 receptors.

RESUMO O uso de soluções eletrolíticas hipotônicas na hidratação enteral ainda é pouco estudado em bezerros. O objetivo do presente estudo foi avaliar os efeitos de soluções eletrolíticas enterais de manutenção com diferentes concentrações de acetato de sódio e diferentes osmolaridades em bezerros. Para isso, foram utilizados 18 bezerros, seis machos e 12 fêmeas, holandeses, com 20 dias de nascidos e pesando por volta dos 52kg. Os animais foram divididos aleatoriamente em três grupos e cada grupo recebeu um dos tratamentos. As três soluções eletrolíticas continham os mesmos componentes, mas em diferentes concentrações, resultando em uma solução hiposmótica, uma isosmótica e uma hiperosmótica. Cada animal foi mantido em hidratação enteral durante 12 horas com taxa de infusão de 15mL kg-1h-1. Foram aferidos perímetro abdominal, peso corporal, consistência das fezes, glicose e lactato plasmático, pH, pCO2, HCO- 3 e excesso de base nos seguintes tempos: T0h, T6h, T12h e T24h. A solução hiposmótica não gerou aparecimento de diarreia, enquanto a isosmótica e a hiperosmótica geraram. Independentemente da dose utilizada, o acetato não causou alcalose metabólica nos animais avaliados. Os resultados sugerem que o uso da solução hiposmótica em bezerros diarreicos, desidratados e sem acidose metabólica, pode ser clinicamente importante.

ABSTRACT The use of hypotonic electrolytic solutions in enteral fluid therapy is still understudied in calves. The objective of the present study was to evaluate the effects of maintenance enteral electrolytic solutions with different concentrations of sodium acetate and different osmolarities in calves. For this, 18 Holstein calves, six male and 12 female, 20 days old and weighing around 52kg, were used. The animals were randomly divided into three groups and each group received one of the treatments. The three electrolytic solutions contained the same components in different concentrations, resulting in a hyposmotic, an isosmotic and a hyperosmotic solution. Each animal was maintained in enteral fluid therapy for 12 hours with infusion rate of 15mL kg-1 h-1. Abdominal circumference, body weight, feces consistency, glucose and plasma lactate, pH, pCO2, HCO- 3 and BE were measured at the following times: T0h, T6h, T12h and T24h. The hyposmotic solution did not generate the onset of diarrhea, while the isosmotic and the hyperosmotic did. Regardless of the dose used, acetate did not cause metabolic alkalosis in the evaluated animals. The results suggest that the use of hyposmotic solution in diarrheic calves, dehydrated and without metabolic acidosis, may be clinically important.

RESUMO Avaliou-se a influência da somatotropina recombinante bovina (rbST) sobre os metabolismos energético e mineral de búfalas entre 63e 154 dias em lactação. Foram utilizadas 22 búfalas, distribuídas em dois grupos experimentais: grupo rbST - aplicação de 500mg de rbST a cada 14 dias; grupo Controle - sem aplicação de rbST. A cada sete dias, foram coletadas amostras de sangue para a determinação do perfil bioquímico e mensuraram-se a produção de leite e o escore de condição corporal dos animais. As médias dos parâmetros estudados para os grupos rbST e Controle foram, respectivamente: produção de leite (PL): 6,44kg vs. 6,68kg; escore de condição corporal-ECC (1-5): 3,51 vs. 3,57; glicose: 70,58 vs. 64,81mg/dL (P = 0,0003); colesterol: 132,38 vs. 133,40mg/dL; triglicérides: 29,18 vs. 28,32mg/dL; proteína total: 8,57 vs. 8,75g/dL; albumina: 3,47 vs. 3,60g/dL; ureia: 32,46 vs. 33,86mg/dL; creatinina: 1,27 vs. 1,39mg/dL; cálcio:10,25 vs. 10,73mg/dL; fósforo:5,76 vs. 5,62mg/dL; e magnésio:3,70 vs. 3,70mg/dL. O uso de 500mg de rbSTinfluenciou o metabolismo da glicose, porém não modificou a PL, o ECC e os níveis dos demais parâmetros metabólicos estudados.

ABSTRACT The aim was to evaluate the influence of recombinant bovine somatotropin (rbST) on the energy and mineral metabolism of buffaloes between 63 - 154 days in milk. Twenty-two buffaloes distributed in two experimental groups were used: Group rbST (n= 11) - application of 500mg of rbST every 14 days; Control Group (n= 11) - no rbST. Every seven days, blood samples were taken to determine the biochemical profile, and milk production and body condition score were measured. The averages of the variables for rbST and Control groups were, respectively: milk yield (MY) - 6.44kg vs. 6.68kg; body condition score (BCS) - 3.51 vs 3.57 (1-5); glucose - 70.58 vs. 64.81mg/dL (P = 0.0003); cholesterol - 132.38 vs. 133.40mg/dL; triglycerides -29.18 vs. 28.32mg/dL; total protein - 8.57 vs. 8.75g/dL; albumin - 3.47 vs 3.60g/dL; urea - 32.46 vs 33.86mg/dL; creatinine - 1.27 vs 1.39mg/dL; calcium - 10.25 vs. 10.73mg/dL; phosphorus - 5.76 vs 5.62mg/dL; and magnesium - 3.70 vs 3.70mg/dL. Use of 500mg rbST influenced glucose metabolism, but did not modify the MY, BCS and the levels of the other metabolic parameters studied.

RESUMO Amostras de sangue de 50 bezerros de búfalo Murrah foram examinados nesse estudo. Os animais foram distribuídos em três grupos de acordo com a paridade de suas genitoras: G1 (n=15) bezerros de búfalas primíparas, G2 (n=19) bezerros de búfalas com 2 a 4 gestações, e G3 (n=16) bezerros de búfalas com cinco ou mais gestações. Amostras de sangue foram colhidas ao nascimento, antes da ingestão de colostro e 24h, 48h, e 72h após o nascimento e 7, 14, 21 e 30 dias após nascimento para determinar níveis de gammaglutamil transferase (GGT), fosfatase alcalina (ALP), aspartato aminotrasferase (AST), creatina quinase, proteínas totais, albumina, globulina (inclusive imunoglobulina G), ferro, cálcio total, cálcio ionizado, fósforo, sódio e potássio. A idade dos bezerros influenciou todos os parâmetros bioquímicos, exceto cálcio ionizado e potássio nos bezerros dos grupos G1 e G3. A ordem de nascimento influenciou AST, GGT, proteínas totais, albumina e globulinas, inclusive IgG. Intensa atividade ALP no soro nos primeiros dois dias após nascimento indica que medidas dos níveis dessa enzima podem ser utilizados como método indireto de avaliar transferência passiva de imunidade.

ABSTRACT Serum blood samples from 50 Murrah buffalo calves were examined in this study. The animals were allocated into three groups according to the number of parturitions of their mothers: G1 (n= 15) calves from primiparous buffaloes, G2 (n= 19) calves from buffaloes with two to four parturitions, and G3 (n= 16) calves from buffaloes with five or more parturitions. Blood samples were taken at birth, before colostrum ingestion, at 24h, 48h, and 72h after birth, and at 7, 14, 21, and 30 days after birth for determination of levels of gammaglutamyl transferase (GGT), alkaline phosphatase (ALP), aspartate aminotransferase (AST), creatine kinase, total protein, albumin, globulins (including immunoglobulin G), iron, total calcium, ionized calcium, phosphorus, sodium, and potassium. The age of the calves was found to influence all of the biochemical parameters, with the exception of ionized calcium and potassium in the calves in groups G1 and G3. The calving order was found to influence AST, GGT, total protein, albumin, and globulins, including IgG. The high serum ALP activity in the first two days after birth indicates that measurement of the levels of this enzyme may be used as an indirect method of assessing passive immunity transfer.

RESUMO Objetivou-se avaliar a influência da somatotropina recombinante bovina (rbST) sobre a produção e os constituintes do leite de búfalas entre 63 e 154 dias em lactação. Foram utilizadas 22 búfalas, distribuídas em dois grupos experimentais: grupo rbST - aplicação de 500 mg de rbST a cada 14 dias; grupo controle - sem aplicação de rbST. A cada sete dias, foi aferida a produção de leite de todas as búfalas e coletada uma amostra para análise físico-química. As variáveis produtivas e as oriundas de análises laboratoriais foram avaliadas como medidas repetidas no tempo, utilizando-se o comando Repeated gerado pelo procedimento GLM do SAS. A média dos parâmetros estudados para os grupos rbST e controle foram, respectivamente: produção de leite - 6,54 vs. 6,68 kg; gordura - 6,31 vs. 6,34%; proteína 3,86 vs. 3,81%; lactose - 4,96 vs. 5,02%; sólidos totais - 16,05 vs. 16,03%; extrato seco desengordurado - 9,75 vs. 9,74%; contagem de células somáticas - 329,90 vs. 171,68 (x 1000/mL); e elecondutividade - 2,87 vs. 2,81mS/cm. A utilização de 500mg de rbST administrados quinzenalmente, entre 63 e 154 dias em lactação não alterou a produção de leite, a proporção dos constituintes e a CCS do leite de búfalas leiteiras.

ABSTRACT This study aimed to evaluate the effect of recombinant bovine somatotropin (rbST) on milk yield and the proportion of buffalo milk components during lactation. Twenty-two buffaloes randomly distributed in two experimental groups were used: Group rbST - application of 500mg rbST every 14 days, between 63 and 154 days in milk (DIM); Control Group - without treatment. Weekly, the milk yield of buffaloes was measured and a sample was collected for physicochemical analysis. The response variables were evaluated as repeated measures, using the Repeated procedure through the GLM procedure of SAS. Means of each variable after rbST and Control were: Milk yield - 6,54 vs. 6,68 kg; Fat - 6,31 vs. 6,34%; Protein - 3,86 vs. 3,81%; Lactose - 4,96 vs. 5,02%; Milk solids - 16,05 vs. 16,03%; Defatted dry matter - 9,75 vs. 9,74%; Somatic Cells Count - 329,90 vs. 171,68 (x 1000/mL); and electrical conductivity- 2,87 vs. 2,81mS/cm. The use of 500mg of rbST administered every two weeks, between 63 and 154 DIM did not affect milk yield, proportion of milk constituents and SCC of dairy buffaloes.

Com base na Teoria da Autodeterminação (TAD; Deci & Ryan, 1985), o presente estudo objetivou investigar as relações entre a prática de exercícios físicos e suas regulações motivacionais, bem como os fatores associados a estas em estudantes adolescentes. Realizou-se um estudo transversal com a participação de 471 adolescentes de 14 a 18 anos de idade, estudantes de uma escola pública estadual no município de Florianópolis/SC, Brasil. Questionários foram utilizados para a realização da pesquisa e os dados foram tratados com estatística descritiva e inferencial. Meninos foram mais ativos e mais autodeterminados para a prática de exercícios físicos do que as meninas. Para ambos os sexos as regulações motivacionais mais internas, bem como o índice de autodeterminação, estiveram associados positivamente com a prática de exercícios físicos. Meninas com maior idade e com presença de sobrepeso apresentaram maior regulação identificada. Entre os meninos, os estudantes com sobrepeso mostraram-se mais amotivados e menos motivados intrinsecamente. De maneira geral, adolescentes que estudam no período vespertino apresentaram um maior índice de autodeterminação. Conclui-se que adolescentes mais autodeterminados praticam mais exercícios físicos regularmente. Destacam-se ainda os fatores associados à motivação e a existência de diferenças entre meninas e meninos nas relações entre as variáveis investigadas.

Based on Self-Determination Theory (SDT; Deci & Ryan, 1985), this study investigated the relationship between physical exercise, motivational regulations and associated factors in adolescents. We conducted a cross-sectional study involving 471 adolescents aged 14 to 18 years old, students at a public school in Florianópolis/SC, Brazil. Questionnaires were used to conduct the survey and the data was analyzed by descriptive and inferential statistics. Boys were more active and more self-determined for physical exercise than girls. For both genders the internal motivations and the self-determination were positively associated with physical exercise. The older and overweight girls had higher identified regulation. Among boys, students who are overweight were more amotivated and less intrinsically motivated. All students, adolescents who are studying in the afternoon period showed a higher baseline of self-determination. The study concluded that adolescents more self-determined did more physical exercise regularly. Also the study showed that there are factors associated with the motivational regulations and that there are differences between girls and boys.

RESUMO Este trabalho teve como objetivo estabelecer a ocorrência de anticorpos contra neosporose, brucelose e leptospirose em alguns rebanhos bubalinos da Amazônia. Foram colhidas 212 amostras de sangue de búfalas, na Ilha do Marajó (n = 135) e na região nordeste do Estado do Pará (n = 77). Examinaram-se 212 amostras submetidas à prova de ELISA para pesquisa de anticorpos anti-Neospora caninum utilizando o kit (HerdeChek IDEXX Laboratories, USA); 160 amostras de soro utilizando a prova de fixação de complemento (FC) para pesquisa de anticorpos anti-Brucella sp.; e 205 amostras através do método de soroaglutinação microscópica (SAM) para pesquisar anticorpos contraLeptospira spp., utilizando uma bateria de 27 antígenos. A frequência de búfalas com anticorpos contra N. caninum, Brucella sp. e Leptospira spp. observada foi, respectivamente, 88,21%, 3,75% e 80,00%. As análises estatísticas foram realizadas pela prova de qui-quadrado (x2, p < 0,05). Concluiu-se que não houve uma variação da prevalência de anticorpos contra essas doenças em função da idade do animal e que a elevada presença de anticorpos contraN. caninun e Leptospira spp. demonstrou claramente a necessidade de um controle sanitário mais efetivo para essas enfermidades. Constatou-se ainda que a reduzida prevalência de anticorpos anti-Brucella sp. indicou que o controle aplicado nas fazendas para essa doença tem sido eficaz.

ABSTRACT This study was aimed at establishing the occurrence of antibodies against neosporosis, brucellosis and leptospirosis in some herds in the Amazônia region. A total of 212 blood samples from buffaloes from the Island of Marajó (n = 135) and in Pará State (n = 77) were collected. A total of 212 serum samples were submitted to the ELISA test for antibodies to Neospora caninum using the kit (HerdeChek – IDEXX Laboratories, USA); 160 serum samples were examined forBrucella sp. using the complement fixation test (CF); 205 samples were examined for Leptospira spp., using the microscopic agglutination test (MAT) with a battery of 27 antigens. The frequency of buffaloes with antibodies against Neospora caninum, Brucella sp. and Leptospira spp. was 88.21%, 3.75% and 80.00% respectively. Statistical analyses were performed by the chi-square test (p < 0.05). It was concluded that there was no variation in the prevalence of antibodies against Neospora caninun, Brucella sp and Leptospiraspp related to the age of the animals, and that the high presence of antibodies against Neospora caninun and Leptospira spp. clearly demonstrates the need for a more effective control of these two diseases. The low brucellosis prevalence might indicate efficient control measures for this disease.

Utilizaram-se 10 cães neonatos (cinco machos e cinco fêmeas), obtidos a partir de cinco ninhadas (um casal de cada) com o objetivo de estabelecer o perfil ecodopplercardiográfico normal durante a fase neonatal (de um até 30 dias). Por meio do Doppler pulsado foram mensurados os picos de velocidade dos fluxos sangüíneos através das valvas mitral, tricúspide, aórtica e pulmonar. Observou-se correlação positiva de todas as mensurações com o peso corporal e a idade dos neonatos de ambos os sexos. Não houve diferenças significativas no padrão de velocidade dos fluxos sangüíneos estudados entre machos e fêmeas durante a fase neonatal.

Ten healthy puppies (five males and five females) were used to establish the regular echodopplercardiography profile during the neonatal period (from one to 30 days) utilizing Doppler (pulsed-wave and color flow). Mitral, tricuspid, aortic and pulmonary blood flows were measured. Positive correlations of all the measures, with body weight and age of the puppies for both sexes were observed. No differences in the cardiac development between male and female during the neonatal phase were observed.
