Resumo O protocolo de imersão floral mediado por Agrobacterium é o método de transformação mais amplamente utilizado para uma planta-modelo Arabidopsis thaliana. Existem vários métodos úteis para transformações de Arabidopsis mediados por Agrobacterium tumefaciens, mas são demorados e com baixa eficiência de transformação. Aqui, desenvolvemos uma linha transgênica de Arabidopsis TET12p::TET12-RFP em um curto período de tempo e eficiência de transformação aprimorada usando um método de transformação modificado por meio da aplicação de estresse hídrico após o mergulho floral. Neste protocolo, células de Agrobacterium transportando vetores recombinantes TET12p::TET12-RFP foram ressuspensas em uma solução de 5% de sacarose, 0,05% (v/v) silwet L-77 para transformar gametas femininos de inflorescências de Arabidopsis em desenvolvimento. Arabidopsis tratadas foram então aplicadas com diferentes níveis de estresse hídrico para estimular as plantas a utilizar o máximo de energia da planta no processo de maturação das sementes. Os estresses aplicados alcançaram a rápida maturação das inflorescências já tratadas enquanto paravam o crescimento de inflorescências não tratadas recém-surgidas, diminuindo, assim, as chances de coleta errada de sementes não transformadas. Consequentemente, as sementes coletadas eram principalmente transgênicas, com uma frequência de transformação de pelo menos 10%; portanto, a triagem para seleção dos transformantes positivos foi mais vantajosa em um meio seletivo em comparação com um método clássico de imersão floral. Dentro de 2-3 meses, 200 plantas transgênicas individuais foram produzidas a partir de apenas 10 plantas infiltradas. Este estudo conclui que a aplicação de estresse hídrico em um estágio específico da planta é uma estratégia benéfica para alcançar a Arabidopsis transgênica em um curto período de tempo com alta eficiência de transformação.

Abstract The Agrobacterium-mediated floral dip protocol is the most extensively used transformation method for a model plant Arabidopsis thaliana. Several useful methods for Agrobacterium tumefaciens–mediated transformations of Arabidopsis are existing, but they are time consuming and with low transformation efficiency. Here, we developed a transgenic Arabidopsis lines TET12p::TET12-RFP in a short period of time and enhanced transformation efficiency by using a modified transformation method by applying drought stress after floral dip. In this protocol, Agrobacterium cells carrying TET12p::TET12-RFP recombinant vectors were resuspended in a solution of 5% sucrose, 0.05% (v/v) silwet L-77 to transform female gametes of developing Arabidopsis inflorescences. Treated Arabidopsis were then applied with different levels of drought stresses to stimulate plants for the utilization of maximum plant energy in seed maturation process. The applied stresses achieved the fast maturation of already treated inflorescences while stopped the growing of newly arising untreated inflorescence, thus decreased the chances of wrong collection of untransformed seeds. Consequently, the collected seeds were mostly transgenic with a transformation frequency of at least 10%, thus the screening for positive transformants selection was more advantageous on a selective medium as compared to a classical floral dip method. Within 2-3 months, two hundred of individual transgenic plants were produced from just 10 infiltrated plants. This study concludes that application of drought stresses in a specific stage of plant is a beneficial strategy for achieving the transgenic Arabidopsis in a short period of time with high transformation efficiency.

Resumo Usando dados de inventário, este estudo avaliou a composição de espécies, volume de estoque crescente (GSV) e carbono de biomassa (BMC) das cinco principais espécies madeireiras nas regiões subtropicais e temperadas/subalpinas do Paquistão. Constatou-se que a densidade do caule variou entre 50 e 221 árvores ha-1, com média de 142 árvores ha-1 (13,68 milhões de árvores para toda a área florestal). Entre as espécies, Pinus wallichiana apresentou alta composição de espécies (27,80%), seguida de Picea smithiana (24,64%). O GSV foi encontrado na faixa de 67,81 a 425,94 m3 ha-1, com um valor total de 20,68 milhões de m3 para toda a região. Da mesma forma, o BMC variou de 27,04 a 169,86 mg ha-1, com valor médio de 86,80 mg ha-1. A quantidade total de carbono armazenado foi de 8,69 milhões de toneladas para um total de 95.842 ha de floresta manejada comercialmente. Além disso, a análise de correlação entre área basal (BA), GSV e BMC mostrou que BA é o melhor preditor de GSV e BMC. As descobertas fornecem insights para os formuladores de políticas e gestores florestais sobre o manejo florestal comercial sustentável, bem como o manejo florestal de carbono no recente gerenciamento global de carbono para a mitigação das mudanças climáticas.

Abstract Using inventory data, this study evaluates the species composition, growing stock volume (GSV), and biomass carbon (BMC) of the five major timber species in the sub-tropical, and temperate/sub-alpine regions of Pakistan. It was found that the stem density varies between 50 and 221 trees ha -1, with a mean of 142 trees ha-1 (13.68 million trees for entire forest area). Among the species, Pinus wallichiana showed a high species composition (27.80%) followed by Picea smithiana (24.64%). The GSV was found in the range of 67.81 to 425.94 m3 ha-1, with a total GSV value of 20.68 million m3 for the entire region. Similarly, The BMC ranged from 27.04 to 169.86 Mg ha-1, with a mean BMC value of 86.80 Mg ha-1. The total amount of stored carbon was found at 8.69 million tons for a total of 95842 ha of commercially managed forest. Furthermore, the correlation analysis between the basal area (BA) and GSV and BMC showed that BA is the best predictor of GSV and BMC. The findings provide insights to the policy makers and forest managers regarding the sustainable commercial forest management as well as forest carbon management in the recent global carbon management for climate change mitigation.

Resumo O objetivo básico deste estudo foi determinar a diversidade ictiofaunística do rio Panjkora nos distritos de Lower e Upper de Dir, na província de Khyber Pakhtunkhwa, no Paquistão. Amostras de peixes foram coletadas com redes de arrasto de março a setembro de 2020. Foram coletados 724 espécimes, classificados em 5 famílias, 14 gêneros e 18 espécies. Os resultados gerais revelaram que a maioria da ictiofauna do rio Panjkora contém 8 espécies da família Cyprinidae (56,49%), seguidas por 4 espécies de Nemacheilidae (24,44%), 2 espécies de Channidae (10,63%) e Sisoridae (7,04%) e 1 espécie de Mastacembelidae (1,38%). Entre todas as espécies de peixes, Schizothorax plagiostomus (16,57%) foi altamente predominante, seguida por Carassius auratus (11,87%) e Racoma labiata (9,66%), e elas foram relatadas como altamente abundantes, especialmente nos meses de abril, maio e junho. As espécies menos abundantes foram Glyptothorax punjabensis, Glyptothorax sufii e Mastacembelus armatus, que constituíram 2,48%, 2,20% e 1,38%, respectivamente, do total de peixes amostrados. O índice de diversidade de Simpson (1-D = 0,919), o índice recíproco de Simpson (1/D = 12,3876) e o índice de Shannon (H = 2,68) indicaram que o rio Panjkora contém um grupo bastante rico e diversificado de espécies de peixes. Os microplásticos mais altos foram observados no local 7 em comparação com outra área de estudo. Medidas de conservação devem ser tomadas como prioridade máxima para proteger e conservar o ambiente marinho e o patrimônio natural de novas perdas e extinção e para parar ou minimizar as perdas ocorridas por práticas de pesca irresponsáveis.

Abstract The basic aim of this study was aimed to determine the ichthyofaunal diversity of River Panjkora in both upper and lower Dir districts in Khyber Pakhtunkhwa province of Pakistan.Fish samples were collected by using fishnets from March to September 2020. A total of 724 specimens were collected and classified into 5 families, 14 genera, and 18 species. The overall results revealed that most fish fauna of river Panjkora contains 8 species of family Cyprinidae (56.49%) followed by 4 species of Nemacheilidae (24.44%), 2 species of Channidae (10.63%), and Sisoridae (7.04%), and 1 species of Mastacembelidae (1.38%), respectively. Among all kinds of fish species, Schizothorax plagiostomus (16.57%) was highly dominated and followed by Carassius auratus (11.87%) and Racoma labiata (9.66%) and were reported as highly abundant, especially during April, May, and June. The least abundant species were Glyptothorax punjabensis, Glyptothorax sufii, and Mastacembelus armatus, that constituting 2.48%, 2.20%, and 1.38% of the total fish samples. The Overall Simpson’s diversity (1-D= 0.919) and Simpson’s Reciprocal index values (1/D= 12.3876), and Shannon’s index (H= 2.68) were indicating that river Panjkora contains a quite rich and diverse group of fish species. The highest microplastics observed in site 7 compared to other study area. Conservation steps should be taken as a top priority to protect and conserve the marine environment and natural heritage from further loss, extinction and stop or minimize losses incurred through irresponsible fishery practices

Abstract The presence of microplastics in aquatic environments has raised concerns about their abundance and potential hazards to aquatic organisms. This review provides insight into the problem that may be of alarm for freshwater fish. Plastic pollution is not confined to marine ecosystems; freshwater also comprises plastic bits, as the most of plastic fragments enter oceans via rivers. Microplastics (MPs) can be consumed by fish and accumulated due to their size and poor biodegradability. Furthermore, it has the potential to enter the food chain and cause health problems. Evidence of MPs s ingestion has been reported in >150 fish species from both freshwater and marine systems. However, microplastic quantification and toxicity in freshwater ecosystems have been underestimated, ignored, and not reported as much as compared to the marine ecosystem. However, their abundance, influence, and toxicity in freshwater biota are not less than in marine ecosystems. The interaction of MPs with freshwater fish, as well as the risk of human consumption, remains a mystery. Nevertheless, our knowledge of the impacts of MPs on freshwater fish is still very limited. This study detailed the status of the toxicity of MPs in freshwater fish. This review will add to our understanding of the ecotoxicology of microplastics on freshwater fish and give subsequent research directions.

Resumo A presença de microplásticos em ambientes aquáticos levantou preocupações sobre sua abundância e perigos potenciais para os organismos que vivem nesse meio. Esta revisão fornece informações sobre o problema que pode ser alarmante para os peixes de água doce. A poluição plástica não se limita aos ecossistemas marinhos; a água doce também contém pedaços de plástico, já que a maioria dos fragmentos de plástico entra nos oceanos por meio dos rios. Os microplásticos (MPs) podem ser consumidos pelos peixes e acumulados devido ao seu tamanho e baixa biodegradabilidade. Além disso, tem o potencial de entrar na cadeia alimentar e causar problemas de saúde. Evidências de ingestão de MPs foram relatadas em mais de 150 espécies de peixes de sistemas de água doce e marinhos. No entanto, a quantificação e a toxicidade de microplásticos em ecossistemas de água doce foram subestimadas, ignoradas e não relatadas tanto quanto em comparação com o ecossistema marinho. No entanto, sua abundância, influência e toxicidade na biota de água doce não são menores que nos ecossistemas marinhos. A interação de MPs com peixes de água doce, bem como o risco de consumo humano, permanece um mistério. Resumo O diabetes mellitus (DM) é uma doença não transmissível em todo o mundo, na qual existe nível glicêmico persistentemente alto em relação à normalidade. O diabetes e a resistência à insulina são os principais responsáveis pelas morbidades e mortalidades de humanos no mundo. Essa doença é regulada principalmente por várias enzimas e hormônios, entre os quais a glicogênio sintase quinase-3 (GSK-3) é uma enzima principal e a insulina é o principal hormônio que a regula. A GSK-3, que é a enzima-chave, normalmente mostra suas ações por vários mecanismos que incluem sua fosforilação, formação de complexos de proteínas e outras distribuições celulares e, portanto, controla e afeta diretamente a morfologia celular, seu crescimento, mobilidade e apoptose do célula. Perturbações na ação da enzima GSK-3 podem levar a várias condições de doença que incluem resistência à insulina que leva ao diabetes, doenças neurológicas como a doença de Alzheimer e câncer. As fluoroquinolonas são a classe mais comum de drogas que apresentam efeitos disglicêmicos por meio da interação com a enzima GSK-3. Portanto, é necessário hoje em dia compreender adequadamente as funções e mecanismos da GSK-3, principalmente seu papel na homeostase da glicose via efeitos na glicogênio sintase.

Abstract Diabetes mellitus (DM) is a non-communicable disease throughout the world in which there is persistently high blood glucose level from the normal range. The diabetes and insulin resistance are mainly responsible for the morbidities and mortalities of humans in the world. This disease is mainly regulated by various enzymes and hormones among which Glycogen synthase kinase-3 (GSK-3) is a principle enzyme and insulin is the key hormone regulating it. The GSK-3, that is the key enzyme is normally showing its actions by various mechanisms that include its phosphorylation, formation of protein complexes, and other cellular distribution and thus it control and directly affects cellular morphology, its growth, mobility and apoptosis of the cell. Disturbances in the action of GSK-3 enzyme may leads to various disease conditions that include insulin resistance leading to diabetes, neurological disease like Alzheimer’s disease and cancer. Fluoroquinolones are the most common class of drugs that shows dysglycemic effects via interacting with GSK-3 enzyme. Therefore, it is the need of the day to properly understand functions and mechanisms of GSK-3, especially its role in glucose homeostasis via effects on glycogen synthase.

Resumo O urso-negro asiático está presente em uma variedade de habitats, como folhas largas e florestas de coníferas, estendendo-se do nível do mar até 4.300 m de altitude e mudando seu habitat para fins alimentares sazonalmente. O presente estudo foi conduzido nos vales Kaghan e Siran, distrito de Mansehra, Khyber Pakhtunkhwa, Paquistão, para avaliar o habitat do urso preto. O método de transecto linear foi usado para a observação de sinais do urso. Parcelas de raio circular de 12 m foram selecionadas para a vegetação de preocupação, ou seja, árvores, arbustos e ervas, e 3 a 6 parcelas foram colocadas em cada transecto. No resultado da pesquisa de sinais, 13 categorias diferentes de sinais de urso foram registradas e a taxa de encontro foi calculada para cada sinal. Um total de 1858 sinais foi observado durante os levantamentos de campo. O total (81%) de espécies de coníferas foi recodificado entre as árvores, com maior aparecimento de Pinus wallichiana (34,22%) e Spruce spp (27,76%); árvores de folhas largas (18,56%) também foram recodificadas em parcelas de habitat. A maioria dos sinais foi encontrada em áreas com arbustos, enquanto um alto número de Viburnum spp (60,29%) estava presente. É indicado que o urso-negro prefere a mistura de árvores coníferas, espécies de viburnos e samambaias; provavelmente porque essas plantas fornecem alimento suficiente, proteção e abrigo meticuloso porque mais de 80% do habitat é composto por essas três espécies. Atualmente, a destruição do habitat e o aumento da população humana são questões crescentes para os animais selvagens, especialmente o urso-negro asiático, que é altamente sensível a esses problemas. Altos níveis de esforços de conservação são recomendados para a proteção do habitat do urso-negro e para evitar a interferência humana em seu território.

Abstract Asiatic black bear is present in variety of habitats like broad-leaves and coniferous forests, extending form sea level to 4300m elevation and change their habitat for food purpose seasonally. The present study was conducted at Kaghan and Siran Valleys, District Mansehra, Khyber Pakhtunkhwa, Pakistan to assess habitat of black bear. Line transect method was used for observation of bear signs. Twelve meter circular radius plots were selected for the concern vegetation’s i.e. (trees, shrubs and herbs) and three to six plots were placed in each transect. At the result of sign survey, thirteen different categories of bear signs were recorded and encounter rate was calculated for each sign. A total of 1858 signs were observed during field surveys. Total (81%) coniferous species were recoded among trees, with the highest appearance of Pinus wallichiana (34.22%) and Spruce spp (27.76%), similarly broad leaves trees (18.56%) were also recoded from habitat plots. Most of the signs were encountered in bushy areas, whereas high number of Viburnum Spp (60.29%) was present. It is indicated that black bear prefers blend of Coniferous Trees, Viburnum and Ferns Species; probably because these plants provide enough food, protection, and meticulous shelter because more than 80% of habitat composed of these three species. Currently habitat destruction and increase in human population are the up-growing issues for wild animals (especially Asiatic black bear), which is highly sensitive to such problems. High levels of conservation efforts are recommended for the protection of black bear habitat and to avoid human interference in their territory.

Resumo O objetivo deste estudo foi avaliar a eficácia dos antibióticos comuns contra diferentes microrganismos em bovinos aparentemente saudáveis na província de Shandong e seus subúrbios. Um total de 220 amostras de esfregaço nasal foi coletado e cultivado para avaliação bacteriológica. Todos os isolados de bactérias após identificação preliminar foram submetidos a estudos de antibiograma seguindo o método de difusão em disco. Verificou-se no estudo que E. coli é o isolado mais comumente associado (21%), seguido por Klebsiella spp. (18%), Pseudomonas aeruginosa (13%), Salmonella spp. (15%), Shigella spp (12%) e Proteus spp (11%). Enquanto os estudos de antibiograma revelaram que o maior número de isolados bacterianos apresentou resistência à Ampicilina (95%), seguido por Augmentin (91%), Cefuroxima (85%) e Tetraciclina (95%) de (Escherichia coli e Klebsiella spp). No caso de Pseudomonas spp. e Salmonella, a maior resistência foi apresentada pela Ampicilina (90%) seguida pela Amoxicilina + Ácido Clavulânico (80%), Cefixima (90%) e Eritromicina (80%). Em Shigella spp e Salmonella spp, a maior resistência foi demonstrada por Amoxicilina, Ceftazidima, Augmentina (60%) e Amoxicilina + Ácido Clavulânico (50%). Conclui-se que estudos de antibiograma in vitro de isolados bacterianos revelaram maior resistência para Ampicilina, Augmentina, Cefuroxima, Cefixima, Tetraciclina, Eritromicina e Amoxicilina + Ácido Clavulânico. Os altos índices de resistência a antibióticos múltiplos (MARI) observados em todos os isolados neste estudo variaram de 0,6 a 0,9. O valor MARI de > 0,2 sugere várias bactérias resistentes a antibióticos e indica a presença de bactérias altamente resistentes.

Abstract The objective of this study was to evaluate the effectiveness of common antibiotics against different microorganisms in apparently healthy cattle in Shandong province and its suburb. A total of 220 nasal swab samples were collected and cultured for bacteriological evaluation. All the bacteria isolates after preliminary identification were subjected to antibiogram studies following disc diffusion method. It was found in the study that E. coli is the most commonly associated isolate (21%), followed by Klebsiella spp. (18%), Pseudomonas aeruginosa (13%), Salmonella spp. (15%), Shigella spp (12%), and Proteus spp (11%). While the antibiogram studies reveled that highest number of bacterial isolates showed resistance to Ampicillin (95%), followed by Augmentin (91%), Cefuroxime (85%) and Tetracycline (95%) of (Escherichia coli and Klebsiella spp). In the case of pseudomonas spp. and Salmonella the highest resistance was showed by Ampicillin (90%) followed by Amoxicillin + Clavulanic Acid (80%), Cefixime (90%), and Erythromycin (80%). In Shigella spp and Salmonella spp highest resistance was showed by Amoxicillin, Ceftazidime, Augmentin (60%), and Amoxicillin + Clavulanic Acid (50%). It is concluded that in vitro antibiogram studies of bacterial isolates revealed higher resistance for Ampicillin, Augmentin, Cefuroxime, Cefixime, Tetracycline, Erythromycin, and Amoxicillin + Clavulanic Acid. The high multiple Antibiotics resistance indexes (MARI) observed in all the isolates in this study ranging from 0.6 to 0.9. MARI value of >0.2 is suggests multiple antibiotic resistant bacteria and indicate presence of highly resistant bacteria.

RESUMO Fatores climáticos desempenham papel essencial no crescimento da largura dos anéis das árvores. Neste estudo, objetivou-se avaliar a correlação entre variáveis climáticas e características de crescimento de anéis de árvores de Pinus sibirica nas montanhas de Altai, noroeste da China. Este estudo é o primeiro desse tipo na análise do crescimento climático de Pinus sibirica no noroeste da China. O estudo mostrou grande potencial para entender as espécies que crescem sob as condições climáticas específicas. Total de 70 testemunhos coletados em três locais na área de amostragem, dos quais 63 testemunhos considerados para este estudo. O efeito das variáveis climáticas estudadas incluem precipitação, temperatura e PDSI. Nossos resultados mostraram que a cronologia da Largura do Anel da Árvore tem uma correlação significativamente positiva com a temperatura do final do inverno (março) e uma correlação negativa significativa com as temperaturas de julho. Uma correlação significativa foi observada com a precipitação do final do verão, enquanto nenhuma relação significativa foi encontrada com o Índice de Severidade de Seca de Palmer. Essas correlações significativas com a temperatura e precipitação sugeriram que esta espécie de árvore tinha o potencial para a reconstrução do clima passado na área.

Abstract Climatic factors play an essential role in the growth of tree ring width. In this study, we aimed to evaluate the correlation between climatic variables and tree-ring growth characteristics of Pinus sibirica in Altai mountains, northwestern China. This study being is first of its kind on climate growth analysis of Pinus sibirica in northwestern China. The study showed great potential to understand the species growing under the specific climatic conditions. Total of 70 tree cores collected from three sites in the sampling area, out of which 63 tree cores considered for this study. The effect of climatic variables which was studied include precipitation, temperature and PDSI. Our results showed that Tree Ring Width chronology has a significantly positive correlation with the late winter (March) temperature and significant negative correlation with the July temperatures. A significant correlation was observed with the late summer precipitation whereas no significant relation found with the Palmer Drought Severity Index. These significant correlations with temperature and precipitation suggested that this tree species had the potential for the reconstruction of the past climate in the area.

Bovine leukosis is an economically important disease of dairy cattle caused by the bovine leukaemia virus (BLV). The study aimed to determine the seroprevalence, haemato-biochemical effects, and risk factors pertinent to the prevalence of bovine leukosis in Holstein-Friesian purebred dairy cattle in the D.I. Khan region of Pakistan. A total of 192 sera were assayed using an enzyme-linked immunosorbent assay. Overall, 31.3% (60/192) cattle were detected as seropositive. There was a marked increase in total leukocyte count (11.29 ± 0.48χ10³/μl), lymphocytes (5.73 ± 0.42%), monocytes (0.81 ± 0.06%), haemoglobin (11.09 ± 0.46 g/dl), red blood cells (7.23 ± 0.37 χ10(6)/μl), and packed cell volume (31.75 ± 1.48%) in seropositive cattle. Serum biochemical parameters in seropositive cattle showed a marked increase in the liver enzymes, alanine transaminase (24.25 ± 1.03 U/l) and aspartate aminotransferase (49.33 ± 3.31 U/l), with a marked decrease in glutathione peroxidase (1365.63 ± 12.03 (U/l) and superoxide dismutase (2.14 ± 0.13 U/ml) activity. A significant association of age, pregnancy, breeding method, milk yield, and health status of seropositive animals with bovine leukosis was also recorded. The prevalence was higher in animals which were older, pregnant, artificially inseminated, low milk producers, and had a history of ailments. The current study found that bovine leukosis virus could cause changes in internal homeostasis, oxidative stress, and liver dysfunction, all of which should be considered during a control regimen. It was concluded that bovine leukosis was moderately prevalent in the D.I. Khan region in Pakistan.

BACKGROUND: In the current era, innovation has become the basis for the success of all industries. In reality, fast innovation facilitated by rapidly changing technological discoveries is critical to global economic progress. AIM: The primary goal of this article is to examine the effect of knowledge exchange and development of supervisory support, trust, training, information technology, and industrial cluster resources on innovation capabilities in the dairy sector of Pakistan. SETTING: From a total of 520 small and medium enterprise (SMEs) dairy farms, 227 owners and managers were carefully chosen to participate in the survey. METHOD: The current study's research framework was based on the resources and diffusion of innovation perspective theories. The data were gathered from dairy farm owners and managers in Punjab, Pakistan. SmartPLS-SEM was used to examine the multivariate connection among the variables. RESULTS: The current research finds that training and development, supervisory assistance, and industrial cluster resources strongly influence knowledge sharing. Furthermore, trust has a favourable influence on innovative capabilities. However, the mediation effect of knowledge sharing (KS) did not support information technology (IT) training and development (T) and innovative capabilities (IC CONCLUSION: According to findings in the study, T as a form of learning connect employees through the sharing of new ideas, allowing the business to improve and the concept to be modified. This study found that supervisory assistance significantly impacts innovative capabilities and knowledge sharing.

Brucellosis is a major threat to public health especially in developing countries including Pakistan. This study reveals the characterisation of Brucella species affecting humans and goats in the Swat region of Khyber Pakhtunkhwa, Pakistan. Blood samples were collected from shepherds and goats and analysed by Rose Bengal precipitation test (RBPT), standard plate agglutination test (SPAT), polymerase chain reaction (PCR) and Sanger sequencing of 16S rRNA gene. The findings of the study indicated 24% (36/150) and 11.3% (17/150) positivity for Brucella abortus and Brucella melitensis, respectively, in human samples. In samples of goats, 26.66% (40/150) were positive for B. abortus and 16.66% (25/150) samples were positive B. melitensis by SPAT. The species-specific PCR confirmed B. abortus in 24% (36/150) of human samples and 26.66% (17/150) of goat samples by targeting the IS711 locus. The remaining seropositive samples were confirmed as B. melitensis using IS711 M species-specific primer. The sequences of the amplified fragments of the 16S rRNA gene were blasted, and phylogenetic analysis revealed that Brucella species circulating in the Swat district were closely related to B. melitensis and B. abortus reported from India, China, Philippines, and the United States (US) showing the existence of the possible epidemiological linkage among the Brucella species. This study concluded that there was a higher prevalence of B. abortus (26.6%) in humans and goats compared to B. melitensis (16.6%). These results revealed that the Brucella species were circulating in both humans and goats in the study areas. The findings of the study concluded that B. abortus and B. melitensis were circulating in goats and shepherds with a higher prevalence of B. abortus than B. melitensis. Furthermore, the Brucella species identified in Swat were phylogenetically related to the Brucella species reported from India, China, Philippines and the US. CONTRIBUTION: The proposed study covers the scope of the journal. The species of the genus Brucella affect both animals and shepherds. This study investigates the seroprevalence of brucellosis in shepherds and goats in different geographical areas in the Swat district. The phylogenetic analysis of the Brucella spp. identified in Swat showed close relationships to the Brucella species reported in India, China, Philippines and the US, which shows the possible epidemiological linkages between the Brucella spp.

Abstract A new series of N-Mannich bases of 2-Phenyl-5-benzimidazole sulfonic acid have been synthesized through amino methylation reaction with secondary amines. The two moieties were held together through a methylene bridge, which comes from formaldehyde (Formalin Solution 37%) used in the reaction. Chemical structures of the newly synthesized compounds have been confirmed using FT-IR, 1HNMR and 13CNMR. Different in vitro assays including Anti-oxidant, Enzyme inhibition, Anti-microbial and Cytotoxicity assay were performed to evaluate the biological potential with reference to the standard drug. Among the synthesized library, compound 3a shows maximum alpha-glucosidase inhibition with an IC50 value of 66.66 μg/ml, compound 3d was found most toxic with LC50 value of 10.17 μg/ml. ADME evaluation studies were performed with the help of Molinspiration online software. Docking calculations were also performed. Given the importance of the nucleus involved, the synthesized compound might find extensive medicinal applications as reported in the literature.

Abstract Morphometric measurement and roosting ecology of Pteropus medius were aimed to find out in Mansehra district of KP, Pakistan. Total 3149 numbers of bats were found in eight biological spots visited; Baffa Doraha, Darband, Dadar, Jallu, Hazara University, Garhi Habibullah Chattar Plain and Jabori, in total 299 numbers of different species of trees including; Morus alba, Pinus raxburghi, Eucalyptus camaldulensis, Morus nigra, Grevillea robusta, Brousonetia papyrifera,Platanus orientalis, Ailanthus altissima, Hevea brasiliensis and Populus nigra. Morphometric features were measured and found vary according to sex of the bats. The average wing span, wing`s length from tip of wing to neck, from thumb to tip of wing and the body`s length from head and claws were recorded to be 102.98 cm, 49.07cm, 28.7 cm and 22.78 cm respectively in males while 93.67 cm, 44.83cm, 24.78cm and 22.78 cm respectively in female bats. Mean circumference of the body including wings and without wing were measured as 22.78 cm and 17.29 cm in males and that of female were 20.07 cm and 16.9 cm. Average length of thumb 3.64 cm, ear`s length 3.1 cm, snout 5.62cm, eye length were 1.07 cm for both sexes and length between the feet in extended position were16.3 cm. Generally different measurement of males bodies were found to be greater than female such as mean body surface area, mass, volume and pressure were found to be 2691.79 cm2, 855.7gm,1236.4 ml and 295.77 dyne/ c m 3for male and 2576.46 cm2, 852.71gm,1207 ml and 290.2 dyne/ c m 3 respectively for female. While weight and density for both males and females bats were same with mean of 8.59 newton and 0.701 g/m3. Findings of current reports can add valued information in literature about bats, which can be used for species identification and conservation.

Resumo A medição morfométrica e a ecologia de poleiro de Pteropus medius foram realizadas no distrito de Mansehra de KP, Paquistão. Foram encontrados 3.149 morcegos em 8 pontos biológicos visitados; Baffa Doraha, Darband, Dadar, Jallu, Universidade Hazara, Garhi Habibullah Chattar Plain e Jabori, no total 299 números de diferentes espécies de árvores, incluindo: Morus alba, Pinus raxburghi, Eucalyptus camaldulensis, Morus nigra, Grevillea robusta, Brousonetia papyrifera, Platanus orientalis, Ailanthus altissima, Hevea brasiliensis e Populus nigra. As características morfométricas foram medidas e variam de acordo com o sexo dos morcegos. A envergadura média da asa, o comprimento da asa da ponta da asa ao pescoço, do polegar à ponta da asa e o comprimento do corpo da cabeça e garras foram registrados em 102,98 cm, 49,07 cm, 28,7 cm e 22,78 cm, respectivamente, nos machos. Enquanto 93,67 cm, 44,83 cm, 24,78 cm e 22,78 cm respectivamente, nas fêmeas. As circunferências médias do corpo incluindo asas e sem asas foram medidas de 22,78 cm e 17,29 cm nos machos e as das fêmeas foram 20,07 cm e 16,9 cm. O comprimento médio do polegar 3,64 cm, comprimento da orelha 3,1 cm, focinho 5,62 cm, comprimento dos olhos 1,07 cm para ambos os sexos e comprimento entre os pés em posição estendida foi de 16,3 cm. Geralmente, as medidas diferentes dos corpos masculinos foram maiores que as femininas, como a área de superfície corporal média, massa, volume e pressão foram encontrados em 2691,79 cm2, 855,7 gm,1236,4 ml e 295,77 dines para masculino e 2576,46 cm2, 852,71 gm, 1207 ml e 290,2 dines respectivamente para fêmeas. Enquanto o peso e a densidade para machos e fêmeas de morcegos foram os mesmos com média de 8,59 N e 0,701 g/m3. Os achados de relatórios atuais podem agregar informações valiosas na literatura sobre morcegos, que podem ser utilizadas para identificação e conservação de espécies.

Abstract Paclitaxel (PTX) is a potent anticancer drug. In the present study, PTX was loaded in poly-3-hydroxybutyrate-co-3-hydroxyvalarate (PHBV) to fabricate the PTX/PHBV (drug-loaded) nanoparticles via the nanoprecipitation method. Blank PHBV nanoparticles were also prepared. The drug-encapsulation efficiency of PTX/PHBV nanoparticles was 45±0.4%. The PTX/PHBV nanoparticles exhibited a pH-sensitive release profile and followed a quasi-Fickian diffusion mechanism. Cytotoxic properties of PHBV and PTX/PHBV nanoparticles were checked against the MCF-7 and Caco-2 cell lines. The PHBV nanoparticle did not inhibit the proliferation of MCF-7 and Caco-2 cell lines, thus depicting their non-toxic and biocompatible nature. On the other hand, the PTX/PHBV nanoparticles demonstrated 1.03-fold higher cytotoxicity and 1.61-fold enhanced apoptosis after treatment with the PTX/PHBV nanoparticles versus free PTX. In summary, the PHBV nanoparticles could be a potential candidate for the delivery of PTX for cancer treatment.

Resumo Paclitaxel (PTX) é um potente medicamento anticancerígeno. No presente estudo, o PTX foi carregado em poli-3-hidroxibutirato-co-3-hidroxivalarato (PHBV) para fabricar as nanopartículas de PTX / PHBV (carregadas com drogas) através do método de nanoprecipitação. Nanopartículas de PHBV em branco também foram preparadas. A eficiência de encapsulamento do fármaco das nanopartículas de PTX/PHBV foi de 45±0,4%. As nanopartículas de PTX/PHBV exibiram um perfil de liberação sensível ao pH e seguiram um mecanismo de difusão quase Fickiano. As propriedades citotóxicas das nanopartículas de PHBV e PTX/PHBV foram verificadas em relação às linhagens celulares MCF-7 e Caco-2. A nanopartícula de PHBV não inibiu a proliferação das linhagens celulares MCF-7 e Caco-2, retratando assim sua natureza atóxica e biocompatível. Por outro lado, as nanopartículas de PTX/PHBV demonstraram citotoxicidade 1,03 vezes maior e apoptose 1,61 vezes maior após tratamento com as nanopartículas de PTX/PHBV versus PTX livre. Em resumo, as nanopartículas de PHBV podem ser candidatas potenciais para a entrega de PTX no tratamento do câncer.

Resumo Esta pesquisa teve como objetivo explorar as doenças parasitárias por helmintos em Schizothorax plagiostomus (a truta das neves) do rio Swat e do rio Panjkora, Paquistão. A coleta de 360 espécimes de peixes foi feita nos trechos inferior, médio e superior de ambos os rios por meio de redes de emalhar, de lançamento, de dragão e anzóis. Todas as amostras foram examinadas na Universidade de Malakand, Departamento de Zoologia, para helmintos parasitas durante os meses de janeiro de 2015 a dezembro de 2016. Do total de amostras de peixes examinadas, 21,9% (n = 79) estavam infectados com Rhabdochona spp, incluindo 17,7% (n = 32/180) no rio Swat e 26,6% (n = 47/180) no rio Panjkora. A maior prevalência no mês (p = 0,09878, p <0,05) foi relatada em maio (30%), depois em fevereiro e outubro cada (26,6%), enquanto a menor em agosto (13,3%). A maior prevalência (p = 0,9723, p <0,05) foi relatada no verão (26,6%), enquanto a menor no inverno (20%). Os adultos foram altamente infectados (p = <0,0001) na prevalência (63,7%), seguidos por subadultos (13,2%), enquanto nenhuma infecção foi encontrada em espécimes juvenis. As amostras de peixes do sexo feminino tiveram maior (p = 0,0277, p> 0,05) prevalência (28,8%) do que o masculino (16,6%). Os peixes do curso inferior tiveram maior (p = 0,0029, P> 0,05) prevalência (31,7%) seguida do curso médio (16,5%), enquanto a menor prevalência foi observada em amostras de peixes coletados do curso superior (9,87%). O presente estudo aborda que Rhabdochona spp no intestino da truta das neves tem uma relação de longo prazo e pode ser considerada uma infecção natural em ciprinídeos e risco zoonótico para humanos.

ABSTRACT This research was aimed to explore the helminth parasitic diseases in Schizothorax plagiostomus (the snow trout) from river Swat and river Panjkora, Pakistan. Collection of 360 fish specimens have done from the lower, middle and upper reaches of both the rivers through gill nets, cast nets, dragon nets and hooks. All the samples were examined in the University of Malakand, Zoology Department for helminth parasites during the months from January 2015 to December 2016. Of the total examined fish samples 21.9% (n=79) were infected with Rhabdochona spp including 17.7% (n=32/180) in river Swat and 26.6% (n=47/180) in river Panjkora. Highest month-wise prevalence (p=0.09878,p<0.05) was reported in May (30%), then by February and October each (26.6%) while the lowest during August (13.3%). Highest prevalence (p=0.9723, p<0.05) was reported in summer season (26.6%), while lowest in the winter season (20%). Adults were highly infected (p=< 0.0001) in prevalence (63.7%) followed by sub-adults (13.2%) while no infection was found in juvenile specimens. Females fish samples had higher (p=0.0277, p>0.05) prevalence (28.8%) than males (16.6%). Fishes of the lower reaches had highest (p=0.0029, P>0.05) prevalence (31.7%) followed by middle reaches (16.5%) while the lowest prevalence was observed in samples of fish collected from upper reaches (9.87%). Present study address that Rhabdochona spp in the intestine of snow trout has a long term relationship and call as a natural infection in cyprinids and zoonotic risk to human.
