Abstract Introduction: Propofol is a widely used anesthetic and its dose is closely related to aging. Telomere length (TL) is a unique heritable trait, and emerging as a biomarker of aging, health and disease. Telomerase RNA component (TERC) plays an important role in maintaining TL. We proposed a hypothesis that propofol dose in general anesthesia can be predicted by measuring TL before operation, which greatly reduced the risk of anesthesia, especially the elderly. Methods: The association between the propofol dose in anesthesia induction and: TL in the DNA of peripheral blood leukocytes; body weight; sex; difference of the Bispectral Index (BIS) before and after anesthesia induction in patients was evaluated by multivariable linear regression analyses. The mutation at the 5’end or 3’end of TERC was detected. We recruited 100 patients of elective surgery. Results: We found that propofol dose in anesthesia induction was clearly correlated significantly with TL (r = 0.78, p < 0.001), body weight (r = 0.84, p = 0.004), sex (r = 0.83, p= 0.84, p = 0.004), sex (r = 0.83, p = 0.004), and difference of BIS before and after anesthesia induction (r = 0.85, p = 0.029). By comparing the absolute values of standardized regression coefficients (0.58, 0.21, 0.19, and 0.12) of the four variables, it can be seen that TL contributes the most to the propofol dose in anesthesia induction. However, the mutation at the 5’ end or 3’ end of TERC was not found. Conclusions: These findings provide preliminary evidence that the propofol dose in anesthesia induction was clearly correlated with genetically determined TL. TL may be a promising predictor of the propofol dose, which is beneficial to improve the safety of anesthesia and reduce perioperative complications.

Abstract: Barley (Hordeum vulgare L.) is a globally significant crop and serves as a pioneer crop for improving saline-alkaline soils due to its salt-alkali tolerant properties. However, the response mechanism of barley to alkali stress remains unclear. In this study, two barley genotypes with contrasting performance under alkali stress were selected: the alkali-tolerant SCMS and the alkali-sensitive QT9919. The morphological and physiological mechanisms of these two barley cultivars in response to alkali stress were elucidated. Results showed that alkali stress inhibited germination in QT9919, leading to significant decreases in germination vigor, plant height and root length. On the other hand, SCMS presented stronger water absorbing capacity than QT9919 under all circumstances. Furthermore, starch degradation and sugar metabolism were suppressed by alkali stress in both barley cultivars through down-regulation of genes expression and reduction of enzymes activities involved in the processes of EMP (Embden-Meyerhof-Parnas), tricarboxylic acid cycle (TCA), and pentose phosphate pathway (PPP). Notably, enzyme activity of hexokinase (HK) and malic dehydrogenase (MDH) showed significant changes under alkali stress between the two barley genotypes. This study provides insights into screening barley germplasm with strong alkali-tolerance and holds significant implications for improving and utilizing of saline-alkaline soils.

Resumo: A cevada (Hordeum vulgare L.) é uma cultura globalmente significativa e serve como uma cultura pioneira para melhorar solos salino-alcalinos devido às suas propriedades de tolerância aos sais e à alcalinidade. No entanto, o mecanismo de resposta da cevada ao estresse alcalino permanece desconhecido. Neste estudo, foram selecionados dois genótipos de cevada com desempenho contrastante sob estresse alcalino: o SCMS tolerante e o QT9919 sensível à alcalinidade. Os mecanismos morfológicos e fisiológicos destas duas cultivares de cevada em resposta ao estresse alcalino foram elucidados. Os resultados mostraram que o estresse alcalino inibiu a germinação em QT9919, levando a reduções significativas no vigor da germinação, na altura da planta e no comprimento da raiz. Por outro lado, o SCMS apresentou maior capacidade de absorção de água que o QT9919 em todas as circunstâncias. Além disso, a degradação do amido e o metabolismo do açúcar foram suprimidos pelo estresse alcalino em ambas as cultivares de cevada através da regulação negativa da expressão genética e da redução das atividades enzimáticas envolvidas nas vias de EMP (Embden-Meyerhof-Parnas), ciclo do ácido tricarboxílico (TCA) e via das pentoses fosfato (PPP). Claramente, as atividades enzimáticas de hexoquinase (HK) e desidrogenase málica (MDH) mostraram alterações significativas sob estresse alcalino entre os dois genótipos de cevada. Este estudo fornece informações sobre a triagem de germoplasma de cevada com forte tolerância à alcalinidade e tem implicações significativas para a melhoria e utilização de solos salino-alcalinos.

ABSTRACT Abdominal cocoon syndrome is a rare cause of intestinal obstruction, which is difficult to diag- nose preoperatively. We here report a case of abdominal cocoon. A 47-year-old male patient was referred to the general surgery department with complaints of abdominal pain, distension, nausea and vomiting for 1 day. An abdominal computed tomography examination detected the dilated small intestinal loops clustered in the abdomen and surrounded by a sac-like membrane. During the exploratory surgery, a capsular structure was identified in the lower quadrant with a regular surface that was solid fibrous in nature. The combination of physical examination, imaging signs and medical history may be helpful in the diagnosis.

RESUMO O presente trabalho visa investigar os efeitos do extrato de semente de uva Proanthocyanidin (GSPE) sobre o desempenho da produção, o metabolismo e o status antioxidante de gado leiteiro Holstein em lactação precoce. Quarenta e oito vacas leiteiras multíparas Holstein foram divididas em quatro grupos (CON, G20, G40 e G80) e receberam 0, 20, 40 e 80mg de GSPE/kg de peso corporal/dia, respectivamente. O G20 aumentou significativamente o rendimento do leite em comparação com os outros grupos. A proteína e os sólidos não gordurosos do leite foram aumentados nos grupos G20, G40 e G80 somente no sétimo dia durante a experiência. Não foi observada diferença significativa na gordura do leite e na contagem de células somáticas, bem como nos parâmetros de metabolismo energético no sangue, na função hepática e na função renal entre os grupos em relação ao grupo controle. Não houve diferença significativa na glutationa peroxidase, na dimutase de superóxido, na capacidade antioxidante total e no peróxido de hidrogênio entre os grupos, mas o conteúdo malondialdeído do G20 aumentou significativamente no dia 14 em comparação com o CON, e tendia a aumentar no dia 28. Em conclusão, a alimentação de 20mg de GSPE/kg de peso corporal/dia foi associada a um aumento significativo no rendimento do leite, sem efeitos nocivos sobre a função hepática ou a renal, com o metabolismo de energia substancial e a melhoria dos parâmetros antioxidantes de gado leiteiro no início da lactação.

ABSTRACT Aims to investigate the effects of grape seed proanthocyanidin extract (GSPE) on production performance, metabolism, and anti-oxidative status of Holstein dairy cattle in early lactation. Forty-eight multiparous Holstein dairy cattle were assigned to four groups (CON, G20, G40 and G80) and supplied with 0, 20, 40, and 80mg GSPE/kg of body weight/day. G20 significantly increased milk yield compared with other groups. Milk protein and non-fat-solids were increased in G20, G40 and G80 groups compared with the control group only at the 7th day during the experiment. No significant difference was observed in milk fat and somatic cell count, nor on parameters of energy metabolism in blood, liver function and kidney function between the four groups. There was no significant difference in glutathione peroxidase, superoxide dismutase, total antioxidant capacity, and hydrogen peroxide between the groups; but the malondialdehyde content of G20 significantly increased at day 14 in comparison with CON, and tended to increase at the 28th day. In conclusion, feeding 20mg GSPE/kg of body weight/day was associated with a significant increase in milk yield without detrimental effects on liver or kidney function and with substantial energy metabolism and antioxidant parameters improvement in early lactation dairy cattle.

This paper describes an efficient bacterial transformation system that was established for the preparation of competent cells, plasmid preparation, and for the storage in bacterial stocks in our laboratory. Using this method, a number of different plasmids have been amplified for further experiments. Competent cells for bacterial transformation were prepared by the calcium chloride method with an optimum concentration of 75 mM. Three different strains of Escherichia coli that were tested are DH5α, TG1 and XL1 blue, and the most efficient strain being XL1 blue. The optimal optical density (OD600) range for competent cell preparation varied for each of the strains investigated, and for XL1 blue it was 0.15-0.45; for TG1 it was 0.2-0.5; and for DH5α it was 0.145-0.45. The storage time of competent cells and its correlation to transformation efficiency has been studied, and the result showed that competent cells can be stored at -20ºC for 7 days and at -70ºC for 15 days. Three critical alterations to previous methods have been made, which are the changing of the normal CaCl2 solution to TB solution, the changing of the medium from LB to S.O.C., and addition of DMSO or PEG8000 during transformation of competent cells with plasmids. Changing the medium from LB to S.O.C., resulted in much faster growth of transformants, and the transformation efficiency was increased. Addition of DMSO or PEG8000 raised transformation efficiencies by 100-300 fold. Our improved bacterial transformation system can raise the transformation efficiency about 10³ times, making it becoming a highly efficient bacterial transformation system.
