Abstract Background: Forests of Pinus pinceana in the Eastern Sierra Madre of Tamaulipas grow in dry environmental conditions. It is important to know the composition and floristic diversity, as well as the useful structural parameters for future conservation programs. Questions: What is the structure, composition and floristic diversity of the vegetation associated with the Pinus pinceana forest in Tamaulipas? Studied species / data description /Mathematical model: Pinus pinceana, structural parameters, diversity, and linear regression models. Study site and dates: State of Tamaulipas, Mexico. March 2016 to March 2017. Methods: At each site (A and B) three strata were analyzed, ten 100 m2 plots for high stratum, 25 m2 for medium stratum and 1 m2 for low stratum, for a total of 30 plots per site. Results: A total of 48 species, 39 genera and 24 families of vascular plants were recorded. Fabaceae, Euphorbiaceae and Asparagaceae were the most representative families. Pinus pinceana had the highest Importance value index (IVI), whereas Flourensia laurifolia and Acacia roemeriana had the highest IVI values for the medium stratum. For the low stratum Croton ciliatoglandulifer and Hechtia hernandez-sandovalii had the highest IVI value. Significant differences were also found in the structure of the middle and upper stratum between the two study sites. Conclusions: The vegetation of the Pinus pinceana forests in Tamaulipas are similar in composition and diversity of species between sites, but the structure of the middle and upper stratum between the two study sites is different.

Resumen Antecedentes: Los bosques de Pinus pinceana en Tamaulipas crecen en condiciones de aridez en la Sierra Madre Oriental. Es importante conocer la composición y diversidad florística, además de los parámetros estructurales útiles para futuros programas de conservación. Preguntas: ¿Cómo es la estructura, composición y diversidad florística de la vegetación asociada al bosque de Pinus pinceana en Tamaulipas? Especies de estudio / Descripción de datos / Modelo matemático: Pinus pinceana, parámetros estructurales, diversidad y regresiones lineales. Sitio y años de estudio: Tamaulipas, México. Marzo de 2016 a Marzo de 2017. Métodos: En cada sitio (A y B) se establecieron 10 parcelas de 100 m2 para el estrato alto; 10 de 25 m2 para el medio y 10 de 1 m2 para el bajo. Un total de 30 parcelas por sitio. Resultados: Un total de 48 especies, 39 géneros y 24 familias de plantas vasculares fueron registradas. Fabaceae, Euphorbiaceae y Asparagaceae fueron las familias dominantes. Pinus pinceana mostró el mayor Índice de Valor de Importancia (IVI) en el estrato alto, Flourensia laurifolia y Acacia roemeriana en el estrato medio y Croton ciliatoglandulifer y Hechtia hernandez-sandovalii en el estrato bajo. También, se encontraron diferencias significativas en la estructura del estrato medio y superior entre los dos sitios de estudio. Conclusiones: La vegetación de los bosques de Pinus pinceana en Tamaulipas son semejantes en composición y diversidad de especies entre sitios, pero la estructura del estrato medio y superior entre los dos sitios de estudio es diferente.

Abstract In recent years, there has been considerable interest in bioactive peptides derived from food proteins, which might benefit human health. Pitaya and jiotilla are fruits with demonstrated biological functions in its stem, peel, and pulp. However, no studies have been conducted to evaluate the peptides. The aim of this study was to determine biofunctional properties (antioxidant, antihypertensive, and antidiabetic activities) of peptides from cacti seed protein. Antioxidant activity by ABTS, ACE inhibition activity, and α-amylase inhibition activity was carried out in the peptides. Based on the obtained results, hydrolyzed total protein isolate of pitaya showed high antioxidant activity (1328.40 ± 63.50 μM Trolox equivalents/mg protein). Moreover, hydrolyzed glutelin fraction of pitaya has a high α-amylase inhibition activity (76.74 ± 2.15%). On the other hand, hydrolyzed total protein isolate of jiotilla had a good ACE inhibition activity (47.64 ± 3.16). These results demonstrated that pitaya and jiotilla seeds protein hydrolysates had excellent in vitro nutraceutical potential.

Abstract Purslane (Portulaca oleracea L.) is a species recognized for its high levels of bioactive compounds, among which the compounds with antioxidant properties stand out. The objective of the work was to determine the variation of phenolic metabolites and their antiradical capabilities in wild accessions of P. oleracea. The research was conducted with samples from five municipalities in Tamaulipas, Mexico collected in October 2018. The contents of total phenolic compounds (CFT) and free radical scavenging capacity against ABTS (2,2’-azino-bis- (3-ethylbenzothiazoline-6-sulfonic) acid) and DPPH (2,2-diphenyl-1-picrylhydracyl) were determined. Extractions were made from vegetative parts using three solvents: acetone, water and ethanol. The accession of the localities of Abasolo and Padilla were the ones that had the highest amount of CFT with 5.8938 ±0.03 and 5.3742 ±0.11 mEAG g-1 PS, respectively, using water in the extraction. Regarding the free radical scavenging capacity against ABTS, the accession of Abasolo and Jiménez recorded the highest values, with 3.27 ±0.06 and 3.2226 ±0.04 mM ET g-1 PS. Regarding the level against DPPH radicals, the accession of Abasolo was the highest with 2.0204 ±0.05 mM ET g-1 PS, using water in the extraction. Water was the best solvent for the extraction of the determined contents. Heterogeneity was observed in the composition and levels of the parameters evaluated among the accessions. Wild P. oleracea accessions represent reservoirs of phenolic compounds and free radical scavenging capacity, including cultivated and wild varieties.

Resumen La verdolaga (Portulaca oleracea L.) es reconocida por sus altos niveles de compuestos bioactivos con propiedades antioxidantes. El objetivo del trabajo fue determinar la variación de metabolitos fenólicos y sus capacidades antirradicalarias en accesiones silvestres de P. oleracea. La investigación se realizó con muestras de cinco municipios de Tamaulipas, recolectadas en octubre 2018. Se determinaron los contenidos de compuestos fenólicos totales (CFT) y capacidad colectora de radicales libres contra ABTS (ácido 2,2’-azino-bis- (3-etilbenzotiazolina-6-sulfónico)) y DPPH (2,2-difenil-1-picrilhidracilo). Las extracciones se realizaron de partes vegetativas usando tres solventes: acetona, agua y etanol. La accesión de las localidades de Abasolo y Padilla fueron la que presentaron mayor cantidad de CFT con 5.8938 ±0.03 y 5.3742 ±0.11 mEAG g-1 PS, respectivamente, usando agua en la extracción. Respecto a la capacidad colectora de radicales libres contra ABTS la accesión de Abasolo y Jiménez registraron los valores más altos, con 3.27 ±0.06 y 3.2226 ±0.04 mM ET g-1 PS. En cuanto a nivel contra radicales DPPH, la accesión de Abasolo fue la más alta con 2.0204 0.05 mM ET g-1 PS usando agua en la extracción. El agua fue el mejor solvente para la extracción de los contenidos determinados. Se observó una heterogeneidad en la composición y niveles de los parámetros evaluados entre las accesiones. Las accesiones silvestres de P. oleracea representan reservorios de compuestos fenólicos y capacidad colectora de radicales libres, incluyendo las variedades cultivadas y silvestres.

Abstract Capsicum annuum var. glabriusculum presents local adaptations to different anthropocentric and ecological dynamics. Therefore, expanding its food uses based on its antioxidant potential will contribute to knowledge about ways to strengthen value chains, enhance their use and encourage the consumption of local wild edible plants. The aim of this work was to evaluate the antioxidant capacity of commercial edible safflower oil supplemented with piquín chili by determining the content of phenolic compounds and by assays of free radical scavenging in ABTS+ and DPPH·. The evaluation included: 1) the piquín chili sample through two extraction solvents and 2) the supplemented and unsupplemented oil at 0 d,7 d, 14 d, 21 d and 28 d after preparation (DPP). The piquín chili presented high antioxidant values. The analysis of variance (ANOVA) indicated that the aqueous extract surpassed the hydroalcoholic extract. The factorial ANOVA showed significant differences in the three antioxidant parameters evaluated. These dissimilarities were associated with supplementation, time (DPP) and the combination of supplementation and DPP. Safflower oil supplementation with C. annuum var. glabriusculum enriched 66 % more its antioxidant value. The comparison of means showed significant differences in the combination of treatments and DPP. The combination of supplemented oil and DPP showed variability in total polyphenol data and ability against ABTS +. An inverse relationship was observed between DPP and total phenolic compounds, and capacity against ABTS +, contrary to the test for DPPH·. The results obtained validate the argument that the addition of piquín chili increases the biofunctionality of safflower oil and can be an alternative source of natural antioxidants.

Resumen Capsicum annuum var. glabriusculum presenta adaptaciones locales a distintas dinámicas antropocéntricas y ecológicas, por lo que ampliar sus usos alimenticios a partir de su potencial antioxidante permitirá contribuir al conocimiento para fortalecer cadenas de valor, robustecer su aprovechamiento y el consumo de plantas comestibles silvestres locales. El objetivo de este trabajo fue suplementar con chile piquín un aceite comestible comercial de cártamo y evaluar su capacidad antioxidante, a través de la determinación del contenido de compuestos fenólicos y mediante ensayos de captación de radicales libres ABTS+ y DPPH·, evaluados en: 1) la muestra de chile piquín a través de dos solventes de extracción y 2) en el aceite suplementado y sin suplementar a los 0 d, 7 d, 14 d, 21 d y 28 d posteriores a la preparación (DPP). El chile piquín presentó altos valores antioxidantes, el análisis de varianza (ANOVA) indicó que el extracto acuoso superó al hidroalcohólico. El ANOVA factorial mostró diferencias significativas en los tres parámetros antioxidantes evaluados. Estas disimilitudes se asociaron a la suplementación, tiempo (DPP) y la combinación de la suplementación y DPP. La suplementación del aceite de cártamo con C. annuum var. glabriusculum enriqueció 66 % su valor antioxidante. La comparación de medias mostró diferencias significativas en la combinación de tratamientos y DPP. La combinación de aceite suplementado y los DPP presentó variabilidad en los datos de polifenoles totales y habilidad contra ABTS+. Se observó una relación inversa entre los DPP y compuestos fenólicos totales y capacidad contra ABTS+, contrario a la prueba para DPPH·. Los resultados obtenidos validan que la adición de chile piquín incrementa la biofuncionalidad del aceite de cártamo y puede ser alternativa de fuente de antioxidantes naturales.

ABSTRACT The article is part of the research project «Integrative conception of the promotion of health and natural sciences in the initial and permanent teacher training», from the University of Pinar del Río «Hermanos Saíz Montes de Oca». It is an experimental research, in its pre-experimental variant and aimed to develop a pedagogical strategy that contributes to the comprehensive education of sexuality with a pedagogical professional approach, in the students of pedagogical careers. The methodological base was assumed as the dialectical-materialist method, which supported the use of methods of the theoretical level, used in the determination of the theoretical referents of the research and empirical level methods were used, which allowed to verify the current state of integral education of sexuality, in the students of the sample. The designed strategy is based on a system of principles, in which the principle of double intentionality of the integral education of sexuality in the training of teaching staff is emphasized. Its implementation in practice allowed the appropriation of content related to sexuality as a personality dimension, placing the student at the center of the process, such as being sexed, both individually and professionally, with the mission of contributing to education of the sexuality of others, a task that is part of his role as a teacher, which proved the validity of the proposal.

RESUMEN El artículo es resultado del proyecto de investigación «Concepción integradora de la promoción de salud y las ciencias naturales en la formación inicial y permanente de profesores», de la Universidad de Pinar del Río «Hermanos Saíz Montes de Oca». Es una investigación experimental, en su variante pre-experimento y tuvo como objetivo elaborar una estrategia pedagógica que contribuya a la educación integral de la sexualidad con enfoque profesional pedagógico, en los estudiantes de las carreras pedagógicas. Se asumió como base metodológica el método dialéctico-materialista, que sustentó el empleo de métodos del nivel teórico, utilizados en la determinación de los referentes teóricos de la investigación y se emplearon métodos del nivel empírico, que permitieron constatar el estado actual de la educación integral de la sexualidad en los estudiantes de la muestra. La estrategia elaborada se sustenta en un sistema de principios, en el que se destaca el principio de la doble intencionalidad de la educación integral de la sexualidad, en la formación del personal docente. Su implementación en la práctica posibilitó la apropiación de los contenidos relacionados con la sexualidad como dimensión de la personalidad, ubicando al estudiante en el centro del proceso, como ser sexuado, tanto en el plano individual como profesional, con la misión de contribuir a la educación de la sexualidad de los otros, tarea que forma parte de su rol como docente, lo que demostró la validez de la propuesta.

ABSTRACT In this study, Differential Expressed Sequence Tags (ESTs) of immature fruits of Persea americana Mill var. drymifolia of the state of Nuevo León, Mexico were developed and identified. Ten genotypes with fruits of different size and shape were selected to generate ESTs by the differential display technique. In total, 393 amplified differential fragments were obtained, 82 fragments were sequenced and edited for identification and comparison with NCBI nucleotide and protein databases. Forty sequences showed significant similarity to mRNA sequences and / or sequences of hypothetical or predicted proteins belonging to P. americana and / or other genera. Some sequences were related to enzymes such as flavanone-3-hydroxylase (F3H), lecithin-cholesterol acyltransferase, glutathione-S-transferase microsomal and pleiotropic drug resistance protein. With the information of the nucleotide composition of ESTs, primers could be designed to quantify expression levels by real-time RT-PCR of genes in different phenological stages of the fruit and to make comparisons among genotypes that allow determining alternative uses of their fruits.

RESUMEN En este estudio se desarrollaron e identificaron etiquetas de secuencias expresadas diferencialmente (ESTs) de frutos inmaduros de Persea americana Mill. var. drymifolia del estado de Nuevo León, México. Diez genotipos con frutos de forma y tamaño diferentes fueron seleccionados para generar ESTs por la técnica de despliegue diferencial.Se obtuvieron en total 393 fragmentos diferenciales amplificados, 82 fragmentos diferenciales fueron secuenciados y editados para identificación y comparación en las bases de datos para nucleótidos y proteínas del NCBI. Cuarenta secuencias mostraron similitud significativa con secuencias de ARNm y/o secuencias de proteínas hipotéticas o predichas pertenecientes a P. americana y/o a otros géneros. Algunas secuencias estuvieron relacionadas a enzimas como flavanona-3-hidroxilasa (F3H), lecitina-colesterol aciltransferasa, glutatión-S-transferasamicrosomal y proteína pleiotrópica de resistencia a drogas. Con la información de la composición nucleotídica de las ESTs se podrán diseñar iniciadores para cuantificar niveles de expresión por RT-PCR en tiempo real de los genes, en diferentes etapas fenológicas del fruto y hacer comparaciones entre los genotipos que permitan determinar usos alternos de sus frutos.

Este trabajo provee una visión general de la actividad proteolítica en el intestino de adultos de Moneilema armatum (Coleoptera: Cerambycidae) y la inhibición de dichas enzimas por proteínas obtenidas de los cladodios de Opuntia ficus-indica . Las proteasas intestinales activas fueron detectadas de pH neutral a ligeramente alcalino a través de zimografía y ensayos espectrofotométricos usando sustratos sintéticos. Estas enzimas fueron susceptibles a un inhibidor específico para serin proteasas (PMSF) y también se demostró que las actividades proteolíticas presentes correspondieron a actividades tipo tripsina, quimotripsina y elastasa. Aunque los cladodios de O. ficus-indica poseen inhibidores de serin proteasas endógenos, parece ser que no todas las proteasas de M. armatum fueron afectadas, particularmente la actividad de tipo elastasa. Estos resultados indican que M. armatum exhibe una variedad de actividades de serin proteasas y que algunas de estas actividades podrían haberse vuelto insensibles a la presencia de dichos inhibidores endógenos.

This work provides an overview of proteolytic activity in the intestine of Moneilema armatum (Coleoptera: Cerambycidae) adults and inhibition of such enzymes by proteins obtained from the cladodes of Opuntia ficus-indica. Active intestinal proteases were detected at a neutral and slightly alkaline pH through zymography and spectrophotometric assays using synthetic substrates. These enzymes were susceptible to a specific serine protease inhibitor (PMSF). It was also demonstrated that trypsin-, chymotrypsin-, and elastase-like activities were present. Although the cladodes of O. ficus-indica possess endogenous inhibitors of serine proteases, it appears that not all proteases of M. armatum were affected, particularly those with elastase-like activity. The results indicated that M. armatum exhibits a variety of serine protease activities, and some of these proteolytic activities were insensitive to endogenous O. ficus-indica protease inhibitors.

Background The high capacity of chloroplast genome response to integrate and express transgenes at high levels makes this technology a good option to produce proteins of interest. This report presents the stable expression of Pectin lyase (PelA gene) and the first stable expression of manganese peroxidase (MnP-2 gene) from the chloroplast genome. Results pES4 and pES5 vectors were derived from pPV111A plasmid and contain the PelA and MnP-2 synthetic genes, respectively. Both genes are flanked by a synthetic rrn16S promoter and the 3'UTR from rbcL gene. Efficient gene integration into both inverted repeats of the intergenic region between rrn16S and 3'rps'12 was confirmed by Southern blot. Stable processing and expression of the RNA were confirmed by Northern blot analysis. Enzymatic activity was evaluated to detect expression and functionality of both enzymes. In general, mature plants showed more activity than young transplastomic plants. Compared to wild type plants, transplastomic events expressing pectin lyase exhibited enzymatic activity above 58.5% of total soluble protein at neutral pH and 60°C. In contrast, MnP-2 showed high activity at pH 6 with optimum temperature at 65°C. Neither transplastomic plant exhibited an abnormal phenotype. Conclusion This study demonstrated that hydrolytic genes PelA and MnP-2 could be integrated and expressed correctly from the chloroplast genome of tobacco plants. A whole plant, having ~ 470 g of biomass could feasibly yield 66,676.25 units of pectin or 21,715.46 units of manganese peroxidase. Also, this study provides new information about methods and strategies for the expression of enzymes with industrial value.

Leaf explants of Persea americana Mill. var. drymifolia were established on DCR medium (Gupta and Durzan, 1985) supplemented with vitamins and growth regulators to produce callus, two sub cultures were made in intervals of 30 to 45 days between them. In order to determine the genetic variability 341 AFLP (amplified fragment lenght polymorphism) polymorphism with 10 primer combinations were generated, 94 samples were analyzed: the mother plant, 31 calluses, 31 calluses from the first sub culture and 31 calluses from the second sub culture. Polymorphic information content (PIC) was determined by primer combination and the genetic distance between samples was calculated for the cluster analysis. Genetic diversity between the mother plant, calluses and calluses of the two sub cultures were obtained and the analysis of molecular variance was performed. The PIC values indicated that the primer combinations were adequate to estimate the genetic variability of calluses. In the cluster analysis, the mother plant showed distances of 0.63 to 0.7 in regard to their calluses, distances of 0.78 - 0.99 were obtained between calluses and calluses of the two sub cultures. Genetic diversity showed significant differences, analysis of molecular variance demonstrated genetic variability between the mother plant, calluses and calluses of the two sub cultures as well as within the calluses, calluses of the first sub culture and calluses of the second sub culture. The results indicated that genetic variability can occur in any sub culture. Due to genetic variability presented in callus clones were not considered.

Explantes foliares de Persea americana Mill. var. drymifolia, se establecieron en medio DCR (Gupta y Durzan, 1985) suplementado con vitaminas y reguladores del crecimiento para producir callos, dos sub cultivos se realizaron en intervalos de 30 a 45 días entre ellos. Con el fin de determinar la variabilidad genética se generaron 341 marcadores AFLP (amplified fragment lenght polymorphism) polimórficos con 10 combinaciones de iniciadores, se analizaron 94 muestras: la planta madre, 31 callos, 31 callos del primer sub cultivo y 31 callos del segundo sub cultivo. Se determinó el contenido de información polimórfica (PIC) por combinación de iniciadores y se calculó la distancia genética entre muestras para hacer el análisis de agrupamiento. Se obtuvo la diversidad genética entre la planta madre, callos y callos de los dos sub cultivos y se realizó ei análisis de varianza molecular. Los valores PIC indicaron que las combinaciones de iniciadores fueron adecuadas para estimar la variabilidad genética de los callos. En el análisis de agrupamiento, la planta madre mostró distancias de 0.63 a 0.7 con respecto a sus callos, distancias de 0.78-0.99 se obtuvieron entre los callos y los callos de los dos sub cultivos. La diversidad genética presentó diferencias significativas, el análisis de varianza molecular demostró variabilidad genética entre la planta madre, callos y callos de los dos sub cultivos, así como dentro de los callos, callos del primer sub cultivo y callos del segundo sub cultivo. Los resultados indicaron que la variabilidad genética puede ocurrir en cualquier sub cultivo. Debido a la variabilidad genética presentada los callos no se consideraron clones.
