RESUMO A mancha-alvo da soja (Corynespora cassiicola) tornou-se uma importante doença na maioria das regiões produtoras de soja no Brasil. A sensibilidade de 34 isolados de Corynespora cassiicola a 11 fungicidas foi avaliada pela inibição do crescimento micelial (boscalida, carbendazim, ciproconazol, fluopiram, fluxapiroxada, protioconazol e tiofanato-metílico) ou inibição da germinação de esporos (azoxistrobina, picoxistrobina, piraclostrobina e trifloxistrobina). Além disso, a eficácia de cinco fungicidas para controlar mancha-alvo foi testada em quatro ensaios no campo, conduzidos durante três safras: 2011/2012, 2012/2013 e 2013/2014. Os isolados do fungo foram coletados de plantas de soja em diversas regiões produtoras de soja do Brasil. A concentração efetiva para inibir o crescimento micelial ou a germinação dos esporos em 50% (EC50), para cada fungicida, foi calculada para todos os isolados. Fluxapiroxada e protioconazol proporcionaram as maiores inibições de crescimento micelial do patógeno e piraclostrobina a menor porcentagem de germinação de esporos, com os menores valores de EC50. Nos ensaios de campo, ciproconazol e carbendazim apresentaram percentuais de controle de mancha-alvo variando de 26% a 29%. Por outro lado, fluxapiroxada e protioconazol preveniram a epidemia da doença na ordem de 45% e 55%, respectivamente. No geral, a eficácia dos fungicidas no campo refletiu as médias de sensibilidade in vitro. Foi detectada ampla redução da sensibilidade aos benzimidazóis (MBC) indicando que esse grupo de fungicidas não deve ser mais usado no controle de mancha-alvo. Houve uma correlação negativa e significativa (-0,265) entre severidade da doença e produção da soja. O patógeno mostrou variabilidade na sensibilidade aos grupos fungicidas carboxamidas (SDHI), triazóis (DMI) e estrobilurinas (QoI), o que demonstra alto risco de seleção para resistência.

ABSTRACT Soybean target spot (Corynespora cassiicola) has become an important disease in most soybean growing regions in Brazil. The sensitivity of 34 isolates of C. cassiicola to 11 fungicides was evaluated based on mycelial growth inhibition (boscalid, carbendazim, cyproconazole, fluopyram, fluxapyroxad, prothioconazole and thiophanate-methyl) or spore germination inhibition (azoxystrobin, picoxystrobin, pyraclostrobin and trifloxystrobin). In addition, the efficacy of five fungicides to control target spot was tested in four field trials carried out during three crop seasons: 2011/2012, 2012/2013 and 2013/2014. Fungal isolates were collected from soybean plants in several soybean growing areas in Brazil. The effective concentration of each fungicide to inhibit fungal growth or spore germination by 50% (EC50) was calculated for all isolates. Fluxapyroxad and prothioconazole provided the greatest mycelial growth inhibition and pyraclostrobin led to the lowest spore germination percentage, with the lowest EC50 values. At field experiments, cyproconazole and carbendazim showed target spot control ranging from 26% to 29%. On the other hand, fluxapyroxad and prothioconazole prevented an epidemic of the disease by 45% to 55%, respectively. In general, the efficacy of fungicides in the field reflected the in vitro sensitivity averages. Large sensitivity reduction was detected to benzimidazoles (MBC), indicating that this group of fungicides should no longer be used for target spot control. There was a negative and significant correlation (-0.265) between target spot severity and soybean yield. The pathogen showed variability in sensitivity to the fungicide groups carboxamides (SDHI), triazoles (DMI) and strobilurins (QoI), which denotes a high risk of selection for resistance.

Corynespora cassiicola, agente causal da mancha alvo, causa prejuízos econômicos em diversas culturas. Como a disponibilidade de protocolos padronizados para a produção de inóculo facilitam a condução de testes de patogenicidade, os objetivos deste trabalho foram analisar o crescimento micelial em diferentes temperaturas; avaliar o crescimento micelial e a esporulação de isolados obtidos de diferentes hospedeiros em diferentes regimes de luz; avaliar o efeito da duração de umidade contínua para a máxima germinação dos esporos in vitro e a patogenicidade de sete isolados de C. cassiicola em diversos hospedeiros. As temperaturas para o maior crescimento micelial variaram de 23,3 a 29,5ºC; o crescimento de oito isolados foi indiferente aos regimes de luz, enquanto os outros seis foram mais influenciados pelo escuro contínuo; a maioria dos isolados esporulou mais sob luz contínua; cinco a vinte horas de umidade contínua foi a variação de tempo para que 85% dos esporos germinassem; quanto à patogenicidade, foi observado que o algodoeiro, o meloeiro e o pepineiro foram os hospedeiros mais suscetíveis, e menos suscetíveis, o tomateiro, aceroleira e o cafeeiro. Para produção massal de inóculo de C. cassiicola recomenda-se utilizar a temperatura de 25ºC sob luz contínua.

Corynespora cassiicola is the causal agent of target spot on several crops of economic importance. Since the availability of standardized protocols for inoculum production makes the conduction of patogenicity tests easier, the objectives of this work were to evaluate the mycelial growth under different temperatures, to evaluate the mycelial growth and spore production under different photoperiods for isolates obtained from different hosts, to evaluate the effect of continuous humidity on spore germination, and the pathogenicity of seven isolates of C. cassiicola on several hosts. Temperatures between 23,3 and 29,5ºC allowed the best growth of the isolates. The growth of eight isolates did not respond to the photoperiods used whereas six isolates were sensitive to the dark. The best sporulation for the majority of isolates was under continuous light. Five to twenty hours of continuous humidity were enough for 85% of the spores to germinate. As for pathogenicity, cotton, melon and cucumber were the most susceptible hosts, while tomato, coffee and Antilles cherry were the least susceptible. For massal production of C. cassiicola inoculum, a temperature of 25ºC under continual light is recommended.

O fungo Corynespora cassiicola, agente causal da mancha-alvo em soja, tem ocorrido com frequência na região Centro-Oeste do Brasil e pode, sob condições de alta temperatura e alta umidade, causar sérios danos à cultura. O uso de cultivares resistentes ao patógeno é recomendado, porém são poucas as disponíveis no mercado. Este trabalho foi realizado visando avaliar a reação de doze cultivares comerciais de soja inoculados com C. cassiicola, em casa de vegetação e no campo. A severidade foi avaliada utilizando escala diagramática de severidade da mancha-alvo. As cultivares menos suscetíveis ao patógeno na casa de vegetação foram BRSGO 7960 e BRS Sambaíba e as mais suscetíveis foram BMX Potência RR e M-SOY 7908 RR. No campo, as cultivares menos suscetíveis foram M-SOY 8866, M-SOY 7908 RR e BMX Potência RR e as mais suscetíveis foram BRSGO 8360 e BRS Tracajá.

The fungus Corynespora cassiicola, the causal agent of soybean target spot, frequently occurs in the Brazilian midwestern region. Under favorable climate conditions it can cause serious damage to this crop. The use of resistant cultivars is recommended although there are only a few available in the market. This investigation was carried out to evaluate the reaction of twelve soybean commercial cultivars to C. cassiicola. Severity was evaluated using a diagrammatic scale of target spot. The less susceptible cultivars in greenhouse were BRSGO 7960 and BRS Sambaíba and the most susceptible were BMX Potencia RR and M-SOY 7908 RR. In the field, the less susceptible cultivars were M-SOY 8866, M-SOY 7908 RR and BMX Potência RR and the most susceptible were BRSGO 8360 and BRS Tracajá.

O fungo Corynespora cassiicola, agente causal da mancha alvo em pepino, pode, sob condições de alta temperatura e alta umidade, causar sérios danos à cultura. No Brasil, não se tem conhecimento suficiente sobre um manejo adequado dessa doença. Este trabalho foi realizado visando avaliar: a) a reação de nove híbridos de pepino desafiados por C. cassiicola; b) a sensibilidade in vitro de seis isolados de C. cassiicola a fungicidas (captan, clorotalonil, mancozeb, azoxystrobin, difenoconazole, carbendazin, tebuconazole e tiofanato-metílico), utilizados nas concentrações de 0, 1, 10, 100 e 1.000 µg mL-1 de ingrediente ativo e c) o tratamento químico preventivo e curativo com os mesmos fungicidas testados in vitro, em plantas de pepino, inoculadas com C. cassiicola. A severidade foi avaliada utilizando escalas de notas e diagramática de severidade da mancha alvo. Os híbridos mais resistentes ao patógeno foram Taisho, Nikkey, Yoshinari e Safira. O fungicida difenoconazole proporcionou as maiores inibições de crescimento micelial (ICM) do patógeno in vitro e a menor dose efetiva capaz de inibir o crescimento micelial em 50% (DE50); já tiofanato-metílico foi o pior, sendo incapaz de inibir o crescimento micelial do fungo. Quanto à aplicação dos fungicidas de forma preventiva e curativa, em plantas de pepino, azoxystrobin foi o mais eficiente no controle da doença, porém para sua utilização há necessidade de seu registro junto ao Ministério da Agricultura, Pecuária e Abastecimento.

The fungus Corynespora cassiicola is the causal agent of target spot in cucumber. Under favorable climatic conditions it can cause serious damage in this horticultural crop. In Brazil, there exists not enough knowledge to determine efficient control measures to the disease. This investigation was carried out to evaluate: a) the reactions of nine cucumber hybrids to C. cassiicola; b) the sensibility of six isolates of C. cassiicola to fungicides in vitro (captan, chlorothalonil, mancozeb, azoxystrobin, difenoconazole, carbendazin, tebuconazole and thiophanate-methyl), used in concentrations of 0, 1, 10, 100 and 1,000 µg mL-1 of active ingredient and c) protective and curative chemical treatments with the same fungicides used in vitro in cucumber plants inoculated with C. cassiicola. The cucumber hybrids were evaluated using the scale of notes and diagrammatic of target spot severity. The more resistant hybrids to the pathogen were Taisho, Nikkey, Yoshinari and Safira. The difenoconazole fungicide caused the most mycelial growth inhibition (MGI) and showed the lowest ED50. Thiophanate-methyl was the worst fungicide, it did not inhibit the mycelial growth of the fungus. Azoxystrobin was the most efficient in controlling the disease, although it has to be registered in Ministry of Agriculture, Livestock and Food Supply in Brazil before its recommendation.

Devido à ausência de escala diagramática para a mancha alvo do pepineiro, causada por Corynespora cassiicola, verificou-se a necessidade de um sistema de quantificação da doença, que proporcionasse maior rapidez e precisão na obtenção dos resultados. Para tanto, uma escala diagramática foi elaborada, considerando-se o limite máximo e mínimo de severidade da doença observados no campo. Os valores intermediários da escala seguiram incrementos logarítmicos para os sintomas da mancha alvo (0,3%; 0,8%; 2%; 5%; 11,5%; 25%; e 46%). Para a validação da escala, oito avaliadores quantificaram a severidade da doença, em cinquenta folhas com diferentes níveis de doença. Inicialmente, a estimativa da severidade foi feita sem auxílio da escala. A seguir, os mesmos avaliadores, utilizando a escala diagramática proposta, estimaram a severidade nas mesmas folhas avaliadas anteriormente. As avaliações com a escala diagramática foram mais precisas e acuradas, nas estimativas da maioria dos avaliadores. A escala diagramática proposta foi considerada adequada para estimar a severidade da mancha alvo, em folhas de pepineiro, e poderá ser utilizada em estudos epidemiológicos e de avaliação de estratégias de controle desta doença.

The lack of a diagrammatic scale to evaluate the cucumber target spot, caused by Corynespora cassiicola, led to the development of an assessment system for a faster and more reliable way to quantify this disease. So, a diagrammatic scale was elaborated considering the maximum and minimum severity limits of the disease observed in the field. The intermediate values followed logarithmic increments for the target spot symptoms (0.3%, 0.8%, 2%, 5%, 11.5%, 25%, and 46%). For validating the scale, eight raters quantified the disease severity on fifty leaves infected with different levels of the disease. Initially, the severity estimation was carried out without the aid of the scale. Then, the same raters used it to estimate the severity on the same leaves previously assessed. Assessments with the aid of the diagrammatic scale were more precise and accurate, according to most of the raters. The proposed diagrammatic scale was considered adequate to estimate the target spot severity in cucumber leaves, and it can be used in epidemiologic studies and in evaluation strategies for controlling this disease.

O fungo Corynespora cassiicola (Berk. & M.A. Curtis) C.T. Wei é o agente causal da mancha alvo, em diversas culturas de importância econômica. Assim, estudos de preservação de isolados de C. cassiicola, que possam manter viáveis isolados do patógeno, durante experimentos, são necessários. O objetivo deste trabalho foi testar a eficiência de três métodos de preservação de isolados: Castellani, repicagens periódicas e óleo mineral, durante seis meses de armazenamento. As avaliações foram efetuadas por meio de análise do desenvolvimento do fungo, em meio de cultivo contendo batata-dextrose-ágar. Constatou-se que o crescimento micelial foi maior para o método de repicagens periódicas; a esporulação, no método Castellani, foi um pouco superior à dos demais métodos; e todos os métodos propiciaram a preservação da patogenicidade dos isolados estudados. Desta forma, recomenda-se, para este fitopatógeno, o método de preservação Castellani.

Corynespora cassiicola (Berk. & M.A. Curtis) C.T. Wei is the causal agent for target spot on several economically important crops. So, studies on the C. cassiicola isolates preservation are necessary to maintain mycelium growth and sporulation, during experiments. The objective of this study was to evaluate the efficiency of three isolates preserving methods: Castellani, periodical transference, and mineral oil, during six months of storage. The evaluations were carried out by analyzing mycelial growth on potato-dextrose-agar medium. It was observed that mycelial growth was higher in the periodical transference method, sporulation was a little higher in the Castellani method, and all methods preserved isolates pathogenicity. Thus, for that phythopatogen, the Castellani method is the most recommended one.
