Targeted massively parallel sequencing for congenital generalized lipodystrophy

ABSTRACT Objective: Our aim is to establish genetic diagnosis of congenital generalized lipodystrophy (CGL) using targeted massively parallel sequencing (MPS), also known as next-generation sequencing (NGS). Subjects and methods: Nine unrelated individuals with a clinical diagnosis of CGL were recruited. We used a customized panel to capture genes related to genetic lipodystrophies. DNA libraries were generated, sequenced using the Illumina MiSeq, and bioinformatics analysis was performed. Results: An accurate genetic diagnosis was stated for all nine patients. Four had pathogenic variants in AGPAT2 and three in BSCL2. Three large homozygous deletions in AGPAT2 were identified by copy-number variant analysis. Conclusions: Although we have found allelic variants in only 2 genes related to CGL, the panel was able to identify different variants including deletions that would have been missed by Sanger sequencing. We believe that MPS is a valuable tool for the genetic diagnosis of multi-genes related diseases, including CGL.

RESUMO A água é um elemento essencial à vida e empregada em diversas atividades humanas, todavia, em função do montante utilizado na agricultura e do aumento de área irrigada, o uso sustentável deste recurso tem se tornado uma preocupação mundial. Neste estudo foram avaliados os dados dos Censos Agropecuários publicados pelo Instituto Brasileiro de Geografia e Estatística (IBGE) de 1960 a 2017, com relação às regiões e aos métodos de irrigação, sendo realizada uma análise detalhada entre as informações dos Censos de 2006 e 2017. Houve aumento na área irrigada no Brasil em todas as regiões geográficas, atingindo 6,9 milhões de hectares em 2017. Os métodos de irrigação por aspersão são os mais utilizados, com 48% da área irrigada, seguido pela irrigação localizada (24,4%) e superfície (22,3%). O gotejamento é o sistema mais empregado em estabelecimentos de até 50 ha (617.423 ha), seguido da aspersão convencional (514.893 ha), enquanto o pivô central é o que se destaca em áreas acima de 50 ha (1.362.828 ha). A região Sudeste (38,6%) e os Estados do Rio Grande do Sul (20,4%), Minas Gerais (16,6%) e São Paulo (16,0%) se destacam como os de maior área irrigada do país. O Estado de Minas Gerais apresentou o maior crescimento de área irrigada no período 2006-2017 (116,1%), devido à expansão das áreas irrigadas com pivô central.
ABSTRACT Water is an essential element to life and used in various human activities. However, because of the amount used in agriculture and the increase in irrigated area, the sustainable use of this resource has become a worldwide concern. In this study, data from the Censuses of Agriculture published by the Instituto Brasileiro de Geografia e Estatística (IBGE) from 1960 to 2017 were evaluated in relation to the regions and irrigation methods, performing a detailed analysis between the data from the 2006 and 2017 Censuses. There was an increase in irrigated area in Brazil in all geographic regions, reaching 6.9 million hectares in 2017. Sprinkler irrigation methods are the most used, with 48% of the irrigated area, followed by localized irrigation (24.4%) and surface irrigation (22.3%). Drip irrigation is the most used system in establishments up to 50 ha (617,423 ha), followed by conventional sprinkler (514,893 ha), while the center pivot stands out in areas larger than 50 ha (1,362,828 ha). The Southeast region (38.6%) and the states of Rio Grande do Sul (20.4%), Minas Gerais (16.6%) and São Paulo (16.0%) stand out as the ones with the largest irrigated area in the country. The State of Minas Gerais showed the highest growth of irrigated area in the 2006-2017 period (116.1%), due to the expansion of areas irrigated with center pivot.

A phytochemical investigation from aerial parts of Casearia arborea (Rich.) Urb. (Salicaceae) led to isolation and identification of: sitosterol, 4-en-stigmast-3-one, 13-hydroxy-trans-ent-cleroda-3,14-diene,3-hydroxy-2-oxo-trans-ent-cleroda-3,14-diene (kolavelone), a mixture of 13-hydroxy-trans-ent-cleroda-3,14-diene and an ester ethyl hexadecanoate, kaempferol-3-O-a-L-arabinofuranoside and 4',5,7-trihydroxy-3',5'-dimethoxyflavone (tricin). The compounds have been described for the first time in this species. Five trihydroxy-flavone-hexoside derivatives have been identified by LC-ESI-HR-MS. The antimicrobial activity of kolavelone was evaluated against strains of Staphylococcus, Bacillus, Pseudomonas, Shigella and Candida spores, and its minimum inhibitory concentration (MIC) was determined. The results showed the antimicrobial activity of kolavelone against several bacteria and Candida tropicalis indicating its potential use as antimicrobial agent.

Resumo Foram analisados os nematóides em Iguana iguana, um grande lagarto Iguanidae que possui ampla distribuição pelas Américas. Os objetivos do estudo foram investigar os helmintos associados ao lagarto I. iguana e seus índices parasitológicos (prevalência e intensidade média de infecção). Foram examinados 18 espécimes (oito machos e 10 fêmeas). A prevalência total foi de 66,6% (12/18), nos machos foi de 75% (6/8) e nas fêmeas 60% (6/10). Iguana iguana foi identificado como um novo hospedeiro para Physaloptera sp., Atractis sp., Piratuba sp., e Subulura sp. Este registro contribui para o conhecimento da diversidade de helmintos associados a este hospedeiro.
Abstract Nematodes were analyzed in Iguana iguana, a large lizard Iguanidae that is widely distributed throughout the Americas. The aims of the study were investigate the helminths associated with the lizard, I. iguana in semi-arid areas of northeastern Brazil and analyze the parasitological indices (prevalence, and mean intensity of infection). A total of 18 specimens of I. iguana were examined (8 males and 10 females). The overall prevalence of infection was 66.6% (12/18), while in males, it was 75% (6/8) and in females, 60% (6/10). Iguana iguana was identified as a new host for Physaloptera sp., Atractis sp., Piratuba sp. and Subulura sp. This registered contributes to the knowledge of helminth diversity associated with this host.

Resumo O lagarto Salvator merianae é uma espécie bem distribuída na América do Sul, ocorrendo do sul da Amazônia até o leste dos Andes e norte da Patagônia. Estudos com a fauna parasitária desse lagarto revelaram que ele hospeda helmintos em vários biomas brasileiros. O presente trabalho fornece novo dado parasitológico sobre nematoda gastrointestinal associado ao lagarto S. merianae. Dezesseis amostras foram analisadas a partir de nove localidades diferentes na região semiárida do nordeste do Brasil. Foram identificadas cinco espécies de nematóide. Oswaldofilaria petersi foi registrado pela primeira vez parasitando S. merianae, aumentando assim o conhecimento da fauna de parasitas que infectam grandes lagartos neotropicais.
Abstract The lizard Salvator merianae is a widely distributed species in South America, occurring from southern Amazonia to the eastern Andes and northern Patagonia. Studies on the parasitic fauna of this lizard have revealed that it is a host for helminths in various Brazilian biomes. The present work provides new parasitological data on the gastrointestinal nematodes associated with the lizard S. merianae. Sixteen specimens were analyzed from nine different locations in a semi-arid region in northeastern Brazil. Five species of nematodes were identified. Oswaldofilaria petersi was first recorded as a parasite of the S. merianae, thus increasing the knowledge of the fauna of parasites that infect large Neotropical lizards.

Vernonanthura polyanthes leaves aqueous extract enhances doxorubicin genotoxicity in somatic cells of Drosophila melanogaster and presents no antifungal activity against Candida spp.

Resumo Vernonanthura polyanthes (Spreng.) A.J. Vega & Dematt. (Asteraceae), conhecida como “assa-peixe”, tem sido utilizada na medicina popular para o tratamento de várias doenças, como bronquite, pneumonia, hemoptise, tosse persistente, abcessos internos, afecções gástricas e cálculo renal. Além disso, alguns estudos já demonstraram que espécies do Gênero Vernonia apresentam atividade antifúngica. Devido à relevância biológica dessa espécie, o objetivo deste estudo foi investigar os efeitos citotóxico, genotóxico, antigenotóxico e antifúngico do extrato aquoso das folhas de V. polyanthes em células somáticas de Drosophila melanogaster ou contra Candida spp. O extrato aquoso da planta não apresentou atividade citotóxica, genotóxica e antigenotóxica nas condições experimentais testadas usando o teste de recombinação e mutação somática em asa (SMART-asa). No entanto, quando o extrato foi associado com a doxorrubicina, utilizada neste trabalho como controle positivo, o potencial mutagênico da doxorrubicina foi potencializado, aumentando o número de mutações em células somáticas de D. melanogaster. Por outro lado, nenhuma atividade inibitória contra Candida spp. foi observada utilizando o extrato aquoso das folhas de V. polyanthes por meio do método de difusão em ágar. Mais estudos são necessários para desvendar os componentes presentes no extrato aquoso das folhas de V. polyanthes que possam contribuir para potencializar a genotoxicidade da doxorrubicina.
Abstract Vernonanthura polyanthes (Spreng.) A.J. Vega & Dematt. (Asteraceae), known as “assa-peixe”, has been used in ethnomedicine for the treatment of various diseases such as bronchitis, pneumonia, hemoptysis, persistent cough, internal abscesses, gastric and kidney stone pain. Moreover, some studies demonstrated that species of Genus Vernonia present antifungal activity. Due to the biological relevance of this species, the aim of this study was to investigate the toxic, genotoxic, antigenotoxic and antifungal potential of V. polyanthes leaves aqueous extract in somatic cells of Drosophila melanogaster or against Candida spp. The aqueous extract of the plant showed no toxic, genotoxic and antigenotoxic activity in the experimental conditions tested using the wing somatic mutation and recombination test (SMART/wing). However, when the extract was associated with doxorubicin, used in this work as a positive control, the mutagenic potential of doxorubicin was enhanced, increasing the number of mutations in D. melanogaster somatic cells. In the other hand, no inhibitory activity against Candida spp. was observed for V. polyanthes leaves aqueous extract using agar-well diffusion assay. More studies are necessary to reveal the components present in the V. polyanthes leaves aqueous extract that could contribute to potentiate the doxorubicin genotoxicity.

Hypertension is associated with greater heat exchange during exercise recovery in a hot environment

Individuals with systemic arterial hypertension have a higher risk of heat-related complications. Thus, the aim of this study was to examine the thermoregulatory responses of hypertensive subjects during recovery from moderate-intensity exercise performed in the heat. A total of eight essential hypertensive (H) and eight normotensive (N) male subjects (age=46.5±1.3 and 45.6±1.4 years, body mass index=25.8±0.8 and 25.6±0.6 kg/m2, mean arterial pressure=98.0±2.8 and 86.0±2.3 mmHg, respectively) rested for 30 min, performed 1 h of treadmill exercise at 50% of maximal oxygen consumption, and rested for 1 h after exercise in an environmental chamber at 38°C and 60% relative humidity. Skin and core temperatures were measured to calculate heat exchange parameters. Mean arterial pressure was higher in the hypertensive than in the normotensive subjects throughout the experiment (P<0.05, unpaired t-test). The hypertensive subjects stored less heat (H=-24.23±3.99 W·m−2 vs N=-13.63±2.24 W·m−2, P=0.03, unpaired t-test), experienced greater variations in body temperature (H=-0.62±0.05°C vs N=-0.35±0.12°C, P=0.03, unpaired t-test), and had more evaporated sweat (H=-106.1±4.59 W·m−2 vs N=-91.15±3.24 W·m−2, P=0.01, unpaired t-test) than the normotensive subjects during the period of recovery from exercise. In conclusion, essential hypertensive subjects showed greater sweat evaporation and increased heat dissipation and body cooling relative to normotensive subjects during recovery from moderate-intensity exercise performed in hot conditions.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Flaxseed and its contribution to body growth and brain of Wistar rats during chidhood and adolescence

Objectives: To evaluate the effect of flaxseed upon body growth and brain of rats. Methods: Experimental phase lasted 52 days, using 42 Wistar rats which were divided into four groups: Control (CG, n = 12), 10% casein diet; Flaxseed (GL, n = 12), 10% flaxseed diet plus casein; Modified Control (GCM, n = 12), 10% casein diet with changes in lipid and fiber comparable to GL; Non-protein (GA, n = 6), diet without protein. Considering food intake, protein intake and weight variation, the Protein Efficiency Ratio (PER) was calculated. Net Protein Retention (NPR) and the Food Efficiency Ratio (CEA) were also determined relative to brain weight at 30 days of life (M30) and at 52 days (M52). Analysis of Variance (ANOVA), Test of Sheffe and post-test of Bonferroni were used, p < 0.05. Results: GL had lower food intake, protein and weight variation than GC, but had higher values than GCM. Concerning PER, GL was lower than GC and similar to the GCM, as well as CEA. As for NPR, GL had lower values than the CG and GCM. At M30, GL was superior to GC in relation to brain weight. Likewise, the same was observed at M52. Conclusion: Flaxseed promoted adequate growth and better brain development in animals, which might be explained by increased incorporation of omega-3 into these tissues.
Objetivos: Evaluar el efecto de la linaza en el crecimiento corporal y del cerebro de ratones. Métodos: El estudio experimental duró 52 días, utilizando 42 ratones Wistar, que fueron divididos en cuatro grupos: Control (GC, n = 12), dieta con 10% de caseína; Linaza (GL, n = 12), 10% de la dieta de linaza adicionada de caseína; Control Modificado (GCM n = 12), 10% de caseína con los cambios en los lípidos y fibra comparable a GL, Sin proteína (GA, n = 6), dieta sin proteínas. Fueron controlados la ingesta de alimentos, la ingesta de proteínas y la variación de peso, fueron calculados el Índice de Eficiencia Proteica (PER), la retención proteica neta (NPR) y el Índice de Eficiencia de Alimentación (CEA). Se determinó también el peso cerebral relativo a los 30 días de vida (M30) y 52 días (M52). Fue utilizado ANOVA, prueba de Sheffé y coeficiente de Bonferroni, con p< 0,05. Resultados: GL tuvo menor consumo de alimentos, proteínas y variación del peso sobre GC, pero tuvo valores más altos que GCM. Con relación a PER, GL fue inferior a GC y similar a GCM, así como en el CEA. GL tuvo valores menores valores de NPR que GC y GCM. En M30, GL fue superior a GC en relación al peso relativo del cerebro. Lo mismo se observó en M52. Conclusiones: La linaza promueve un crecimiento adecuado y un mejor desarrollo cerebral en los animales, lo que podría explicarse por una mayor incorporación de ácidos grasos omega-3 en estos tejidos.

Nutritional assessment of adult patients admitted to a hospital of the Amazon region

Changes in nutritional status are important in clinical practice because they relate to an increase in morbidity and mortality. Studies about nutritional problems in hospitalized adults have been reported since the 1970s. The prevalence of malnutrition has varied from 10 to 70%, depending on the diagnostic criteria used. The hospital studied and the duration of admission. Aim: To assess, in the first day of hospital stay, the nutritional status of adults admitted to undergo elective surgery in a public hospital of the State of Acre, Amazon Region, Brazil. Study design: Sectional study from April 7 to May 22, 2002. Patients and methods: 155 consecutive nutritional evaluations were performed using several parameters - global subjective assessment (GSA), anthropometric measurement and some laboratory tests. For the diagnosis of malnutrition the Index Suggestive of Malnutrition (ISM), as proposed by Waitzberg, was adopted. In the evaluation of obesity the BMI (Body Mass Index), with cutoffs suggested by WHO was used. Results: 75.2% were women. Average age was 34.4 ± 10.1 years. Mulatto (63.1%) was the prevailing racial group. Gynecological (43.6%) was the most frequent surgery. ISM disclosed a 12.1% prevalence of malnutrition. BMI classified 2.0% of the patients as grade I overweight and 15.4% as being obese. Among 76 patients classified as normal by BMI, 15 (19.7%) were considered malnourished by ISM criteria. Concordance between ISM and BMI was weak (k = 0.07). GSA classified 100% of the cases as well nourished. Conclusion: Lack of a golden standard to make the diagnosis of changes in nutritional status has been one of the determinants of the wide variations observed in the pertinent literature and it has hindred valid comparisons. The present study suggests that BMI should be used as an indicator of proportion and not of nutritional status. GSA underestimates the diagnosis of malnutrition. This paper concludes that establishing uniform standards for diagnostic criteria for malnutrition is urgently required and suggests preferring IMS (Index Suggestive of Malnutrition) because of its easy use, low coast and high sensitivity.
Los cambios del estado nutricional son importantes en la práctica clínica porque se relacionan con un aumento de la morbilidad y de la mortalidad. Desde los años 1970 se han comunicado estudios sobre los problemas nutricionales de los adultos hospitalizados. La prevalencia de la malnutrición varía del 10 al 70% según los criterios diagnósticos empleados, el hospital en cuestión y la duración del ingreso. Objetivo: Evaluar en el primer día de estancia hospitalaria el estado nutricional de adultos ingresados para cirugía programada en un hospital público del estado de Acre, región amazónica, Brasil. Diseño del estudio: Estudio transversal desde el 7 de abril hasta el 22 de mayo de 2002. Pacientes y métodos: Se efectuaron 155 evaluaciones consecutivas del estado nutricional mediante varios parámetros, a saber, la evaluación subjetiva global (ESG), medidas antropométricas y algunos datos de laboratorio. Para el diagnóstico de la malnutrición se adoptó el índice de malnutrición (ISM) propuesto por Waitzberg. Para evaluar la obesidad se empleó el IMC (índice de masa corporal) con los valores discriminatorios recomendados por la OMS. Resultados: El 75,2% pertenecía al sexo femenino. El promedio de edad ascendía a 34,4 ± 10,1 años. El grupo racial predominante era el mulato (63,1%). La cirugía más común fue la ginecológica (43,6%). El ISM reveló una prevalencia de malnutrición del 12,1%. Entre los 76 pacientes con un IMC normal, 15 (19,7%) se hallaban mal nutridos según los criterios de ISM. La concordancia entre el ISM y el IMC resultó débil (k = 0,07). La ESG reveló que todos los pacientes se hallaban bien nutridos. Conclusión: La ausencia de un patrón oro para el diagnóstico de las variaciones del estado nutricional es uno de los determinantes del margen tan grande de variación descrito en la bibliografía pertinente, que ha dificultado cualquier comparación válida. En este estudio se sugiere que el IMC debiera emplearse como índice de proporción y no del estado nutricional. La ESG subestima el diagnóstico de la malnutrición. En este trabajo se concluye que se necesitan, de forma urgente, normas uniformes para establecer el diagnóstico de la malnutrición y se recomienda el IMS (índice de malnutrición) debido a su sencillez, bajo coste y elevada sensibilidad.

HEPATITIS B VIRUS INFECTION PROFILE IN CENTRAL BRAZILIAN HEMODIALYSIS POPULATION

A hepatite B tem sido uma grande ameaça aos pacientes de hemodiálise. Para investigar o perfil da infecção pelo vírus da hepatite B (VHB) na população de hemodiálise de Goiânia _ Brasil Central, 282 pacientes foram estudados. A prevalência de marcadores do VHB (AgHBs, anti-HBc e anti-HBs) foi de 56,7% (IC 95%: 51,1 _ 62,7) variando de 33,3% a 77,7% entre as unidades de diálise. O VHB-DNA foi detectado nas amostras AgHBs positivas em 67,6% e 88,2%, nas AgHBs e AgHBe em 91,3% e 100%, e nas AgHBe e anti-HBe soro reativas em 18,2% e 63,6% por hibridização e PCR, respectivamente. O tempo de tratamento hemodialítico mostrou-se estatisticamente associado à soropositividade ao VHB. Somente 10% dos pacientes relataram vacinação para a hepatite B. Assim, uma prevalência elevada para infecção pelo VHB nesta população e o risco aumentado do tempo de tratamento hemodialítico sugerem a transmissão ambiental deste vírus, enfatizando a necessidade urgente de reavaliação das medidas de controle e prevenção adotadas nestas unidades.
Hepatitis B has proved to be a major health hazard in hemodialysis patients. In order to investigate the hepatitis B virus (HBV) infection profile in the hemodialysis population of Goiânia city - Central Brazil, all dialysis patients (N=282) were studied. The prevalence of any HBV marker (HBsAg, anti-HBs, and anti-HBc) was 56.7% (95% CI: 51.1-62.7), ranging from 33.3% to 77.7% depending on dialysis unit. HBV-DNA was detected in 67.6% and 88.2% of the HBsAg-positive serum samples, in 91.3% and 100% of the HBsAg/HBeAg-positive samples, and in 18.2% and 63.6% of the HBsAg/anti-HBe-reactive sera by hybridization and PCR, respectively. The length of time on hemodialysis was significantly associated with HBV seropositivity. Only 10% of the patients reported received hepatitis B vaccination. The findings of a high HBV infection prevalence in this population and the increased risk for HBV infection on long-term hemodialysis suggest the environmental transmission, emphasizing the urgent need to evaluate strategies of control and prevention followed in these units.
