Abstract Hepatitis C virus (HCV) genotypes vary greatly in different regions. The aim of this study is to investigate the distribution of HCV genotypes in HCV infected patients, in Ningxia Hui Autonomous Region. Nucleic acid extraction and amplification were performed with test kits on 153 HCV infected patients serum samples. The HCV viral load was measured using reverse transcriptase PCR (RT-PCR) and HCV genotypes were determined. Among the 153 HCV-infected patients, 56 had genotype (GT)1b (36.60%), 45 had GT2a (29.40%), 23 had GT3a (15.00%), 14 had GT3b (9.20%),13 had GT6a (8.50%), 1 had GT1g (0.70%), 1 had GT6xa (0.70%). In GT1b, 21.40% were female and 78.60% were male; in GT2a, 42.20% were female and 57.80% were male;Males were most prevalent in genotypes 1b(39.30%), while female were most prevalent in genotype 2a(46.30%). Rare GT1g and GT6xa were also detected in males. The 41-50 year age group had the highest HCV prevalence of 32.00%. HCV GT1b is the predominant HCV genotype in Ningxia Hui Autonomous Region.

Resumo Os genótipos do vírus da hepatite C (HCV) variam muito de acordo com a região. O objetivo deste estudo é investigar a distribuição do genótipo do vírus da hepatite C em pessoas infectadas pelo vírus na região autônoma de Ningxia Hui. A extração de ácido nucleico e a expansão de amostras séricas em 153 pacientes infectados com HCV foram realizadas utilizando kits de ensaio. A capacidade do vírus HCV foi medida pela reação em cadeia da polimerase retrotranscrição (RT-PCR) e o genótipo HCV foi determinado. Os genótipos (Gt) tiveram a seguinte distribuição entre os 153 casos de infecção HCV: 56 Gt 1-B (36, 60%), 45 Gt 2-A (29, 40%), 23 Gt 3-A (15, 00%), 14 Gt 3-B (9, 20%), 13 Gt 6-A (8, 50%), 1 Gt 1-G (0, 70%) e 1 Gt 6-Xa (0, 70%). Já o sexo dos indivíduos teve a seguinte distribuição: Gt 1-B, 21, 40% mulheres e 78, 60% homens; Gt 2-A, 42, 20% mulheres e 57, 80% homens. A presença de homens é mais comum no genótipo 1-B (39, 30%), enquanto as mulheres ocorrem mais comumente no genótipo 2-A (46, 30%). Gt 1-G e Gt 6-Xa raros também foram detectados em homens. A taxa de infecção com HCV para grupos etários de 41 a 50 anos é a mais alta, com 32,00%. HCV Gt 1-B é o genótipo dominante do HCV na região autônoma de Ningxia Hui.

Yeast cultures (YCs) are defined as promising feed additives that maintain the health of birds and improve growth performance by modulating gut microbiota. YCs contain effective metabolites such as glycine, fructose, inositol, galactose, and sucrose. This study investigated the effects of YCs and their effective metabolites on carcass traits and cecal microflora in broilers. A total of 280 one-day-old mixed-sex Arbor Acres broilers were randomly allocated to seven groups. The basal diet (control DZ) was supplemented with various proportions of glycine, fructose, inositol, galactose, and sucrose (Groups A, B, and C), 24-hour grown Saccharomyces cerevisiae cultures (Group D) (YC), and a commercial yeast culture product (SZ) at concentrations of 0.1% and 1% (Groups E and F). Bodyweight of broilers was correlated positively with proportions of Proteobacteria in Group C and Lactobacillus and Roseburia in Group B (P <0.05). Broilers fed diets supplemented with YC or its active metabolites had the highest proportions of bacteria involved in nucleotide metabolism, and amino acid and carbohydrate metabolism. These results suggested that the dietary addition of YC could alter the proliferation of beneficial bacteria in broilers.

ABSTRACT Infestation of Bemisia tabaci MEAM1 (Gennadius) causes significant phenotypic changes in a variety of plants. However, plants damaged by Trialeurodes vaporariorum (Westwood) show no similar changes. In this study, to explore the effects of MEAM1 infestation on tobacco growth and the mechanism underlying these effects, growth index, and photosynthesis of infested tobacco (Nicotiana tabacum L.), as well as content of hormones, including auxin, gibberellin, cytokinin, abscisic acid, methyl jasmonate, and ethylene were measured. The results demonstrated that infestation of MEAM1 significantly inhibited tobacco growth and the inhibition effect varied with MEAM1 feeding densities, 50 MEAM1 per infested tobacco plant decreased plant height, internode length, and dry weight cm-2, and this inhibition peaked at 200 MEAM1 per plant, which reduced by 52.17%, 43.83%, and 45.61% that of controls, respectively. Additionally, MEAM1 infestation inhibited photosynthesis of tobacco, causing significant decrease in chlorophyll content, photosynthetic rate and stomatal conductance. However, the effects of T. vaporariorum infestation on tobacco were significantly lighter than that of MEAM1. Hormones assays indicated that infestation with 200 MEAM1 per plant for 30 d significantly affected hormones content, among which gibberellin content was specifically reduced compared with that of uninfested control plants and T. vaporariorum infested plants. Furthermore, exogenous application of 1 and 2 μM gibberellin alleviated the reduction in plant height mediated by MEAM1. Therefore, infestation of MEAM1 significantly inhibited tobacco growth and photosynthesis, and the reduction of gibberellin content may contribute to this inhibition.

Abstract Neurosyphilis (NS) is a neurological disorder caused by Treponema pallidum subspecies pallidum (T. pallidum), but how T. pallidum attach to and cross the blood−brain barrier (BBB) and how BBB response to this bacteria remain unclear. To explore how the human brain microvascular endothelial cells (HBMECs) response to T. pallidum, the Agilent SurePrint G3 Human Gene Expression 8×60K microarray was used. The results revealed that 249 genes were differentially expressed in HBMECs infected with T. pallidum. In particular, genes encoding proteins involved in bacterial adhesion, endothelial cell activation and immune response were regulated by T. pallidum. Furthermore, Gene Ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis were performed to determine the biological functions of differentially expressed genes. In summary, T. pallidum changes the gene expression profile in HBMECs, and differentially expressed genes are associated with widespread biological and pathophysiological functions. Above all, this is the first paper reporting the effects of T. pallidum on HBMECs. These data develop a new platform for further molecular experiments on the pathogenesis of NS.

The objective of the study was to determine the effects of antibiotics, yeast culture (YC), and Lactobacillus culture (LC) on the gut microbiome and metabolites in the serum and caecum of weaning piglets. Twenty-four weaning piglets were divided into four treatment groups: control, antibiotic (1% chlortetracycline), 1.8% yeast culture (YC), and 1.6% Lactobacillus culture groups (LC). Each group had six replicated pens with one pig per pen. Feed and water were available ad libitum. Dietary supplementation with antibiotics, YC and LC increased the abundance of phylum, Firmicutes, and decreased the abundance of phylum, Proteobacteria. Beneficial bacteria such as Lactobacillus and Megasphaera in YC and LC groups increased, whereas the proportion of Shigella was decreased. Genera Alloprevotella and Lachnospira were biomarkers in the control and antibiotic groups, respectively. Phylum, Bacteroidetes, and genus, Collinsella, were biomarkers in the YC group, and Mitsuokella, Anaerotruncus, Syntrophococcus and Sharpea were biomarkers in the LC group. Dietary supplementation with different probiotics changed the serum and caecum metabolite profiles too. Antibiotic supplementation increased the levels of D-mannose, D-glucose, and hexadecanoic acid in the serum, and the levels of myo-inositol, D-mannose and benzenepropanoic acid in the caecum. LC increased the concentrations of D-mannose, cis-9-hexadecenoic acid and heptadecanoic acid in caecum compared with the control group. YC and LC supplementation in the weaning diet could improve the abundance of beneficial bacteria by changing the concentrations of some metabolites in the serum and caecum. Therefore, dietary supplementation with YC or LC could be used as additives instead of antibiotics in weaning piglets.

Multiple growth factors can be administered to mimic the natural process of bone healing in bone tissue engineering. We investigated the effects of sequential release of bone morphogenetic protein-2 (BMP-2) and vascular endothelial growth factor (VEGF) from polylactide-poly (ethylene glycol)-polylactide (PELA) microcapsule-based scaffolds on bone regeneration. To improve the double emulsion/solvent evaporation technique, VEGF was encapsulated in PELA microcapsules, to which BMP-2 was attached. The scaffold (BMP-2/PELA/VEGF) was then fused to these microcapsules using the dichloromethane vapor method. The bioactivity of the released BMP-2 and VEGF was then quantified in rat mesenchymal stem cells (rMSCs). Immunoblotting analysis showed that BMP-2/PELA/VEG promoted the differentiation of rMSCs into osteoblasts via the MAPK and Wnt pathways. Osteoblast differentiation was assessed through alkaline phosphatase expression. When compared with simple BMP-2 plus VEGF group and pure PELA group, osteoblast differentiation in BMP-2/PELA/VEGF group significantly increased. An MTT assay indicated that BMP-2-loaded PELA scaffolds had no adverse effects on cell activity. BMP-2/PELA/VEG promoted the differentiation of rMSCs into osteoblast via the ERK1/2 and Wnt pathways. Our findings indicate that the sequential release of BMP-2 and VEGF from PELA microcapsule-based scaffolds is a promising approach for the treatment of bone defects.

Abstract Objective This study aimed to reveal the mitochondrial genomes (mtgenomes) of Tetrix japonica and Alulatettix yunnanensis, and the phylogenetics of Orthoptera species. Methods The mtgenomes of A. yunnanensis and T. japonica were firstly sequenced and assembled through partial sequences amplification, and then the genome organization and gene arrangement were analyzed. Based on nucleotide/amino acid sequences of 13 protein-coding genes and whole mtgenomes, phylogenetic trees were established on 37 Orthoptera species and 5 outgroups, respectively. Results Except for a regulation region (A+T rich region), a total of 37 genes were found in mtgenomes of T. japonicaand A. yunnanensis, including 13 protein-coding genes, 2 ribosomal RNA genes, and 22 transfer RNA genes, which exhibited similar characters with other Orthoptera species. Phylogenetic tree based on 13 concatenated protein-coding nucleotide sequences were considered to be more suitable for phylogenetic reconstruction of Orthoptera species than amino acid sequences and mtgenomes. The phylogenetic relationships of Caelifera species were Acridoidea and Pamphagoidea > Pyrgomorphoidea > Pneumoroidea > Eumastacoidea > Tetrigoidea > Tridactyloidea. Besides, a sister-group relationship between Tettigonioidea and Rhaphidophoroidea was revealed in Ensifera. Conclusion Concatenated protein-coding nucleotide sequences of 13 genes were suitable for reconstruction of phylogenetic relationship in orthopteroid species. Tridactyloidea was a sister group of Tetrigoidea in Caelifera, and Rhaphidophoroidea was a sister group of Tettigonioidea in Ensifera.

Levels of hydrogen sulfide (H2S), a gaseous signaling molecule, are reduced in the serum of individuals who smoke. We hypothesized that tobacco smoke influenced smooth muscle relaxation by decreasing H2S levels and this effect could also influence expression of cystathionine γ-lyase (CSE) and sulfonylurea receptor-2 (SUR-2). The aim of this study was to explore the effect of tobacco smoke on H2S-mediated rat thoracic aorta relaxation and its possible mechanism. Thirty-two Sprague-Dawley rats were divided into four groups: control (C) group, short-term smoker (SS) group, mid-term smoker (MS) group, and long-term smoker (LS) group. H2S concentrations in serum, action of H2S on rat aortic vascular relaxation, and expression of CSE and SUR-2 in thoracic aortic smooth muscle were measured. Although there was no significant difference in H2S between the C and the SS groups, concentration of H2S was significantly reduced in both the LS and MS groups compared to control (P<0.01). Furthermore, H2S was significantly lower in the LS than in the MS group (P<0.05). Rat aortic vascular relaxation was lower in all three treatment groups compared to the control, with the most significant decrease observed in the LS group (P<0.05 compared to the MS group). Expression of CSE and SUR-2 was reduced in the LS and MS groups compared to control (P<0.05), with the lowest levels observed in the LS group (P<0.05). Therefore, tobacco smoke reduced expression of CSE and SUR-2 in rat thoracic aorta, which may inhibit H2S production and vascular dilation.

We conducted pot experiments on the cypress Thujopsis dolabrata (Linn. f.) Sieb. et Zucc. in order to study the interaction of silicon (Si) and root exudates on cadmium (Cd) bioavailability in the rhizosphere,. Each variety was planted with 100 mg/kg Cd and/or 400 mg/kg Si for 210 days. The results showed that adding Si increased Cd tolerance in T. dolabrata, but that the mechanism was specifical. In T. dolabrata, Si did not prevent Cd translocation from roots to shoots, and it significantly enhanced Cd accumulation without inhibiting growth. Moreover, Si mobilized Cd from the rhizospheric soil by stimulating phenolic exudation from the roots, suggesting that Cd-chelation combined with Si-induced phenolics were involved in Cd detoxification.

Se condujeron experimentos en potes con el ciprés Thujopsis dolabrata (Linn. f.) Sieb. et Zucc. para estudiar la interacción del silicio (Si) y exudados radicales sobre la biodisponibilidad de cadmio (Cd) en la rizósfera. Cada variedad se plantó con 100 mg/kg de Cd y/o 400 mg/kg de Si durante 210 días. Los resultados mostraron que el agregado de Si incrementó la tolerancia al Cd en T. dolabrata, pero que el mecanismo fue específico. En T. dolabrata, el Si no previno la movilización de Cd desde las raíces a los tallos, y mejoró significativamente la acumulación de Cd pero sin inhibir el crecimiento. Más aún, el Si movilizó el Cd desde el suelo rizosférico estimulando la exudación de compuestos fenólicos desde las raíces, sugiriendo que el ligamento de Cd combinado con los compuestos fenólicos exudados desde las raíces estuvieron implicados en eliminar la toxicidad del Cd.

The Gd55Al20Co25 amorphous wires exhibit a relatively strong magnetocaloric effect (MCE). These melt-extracted amorphous wires display second-order magnetic transition (SOMT) and the value of maximal magnetic entropy (–ΔSm) for the melt-extracted wires is calculated to be ~9.7 J·kg–1·K–1 around the Curie point (TC) of ~110 K with applied field of 5 T. Moreover, the melt-extracted amorphous wires show a high refrigerating efficiency with a relatively large cooling power (RCP, ~804 J·kg–1) and refrigeration capacity (RC, ~580 J·kg–1) under an applied magnetic field of 5T due to the broad paramagnetic-ferromagnetic (PM-FM) region associated with amorphous alloys. These favorable properties make melt-extracted Gd-based amorphous wires to be the potential refrigerant for magnetic refrigeration (MR) of liquid oxygen.

The current study was designed to evaluate the aqueous extract of Terminalia chebula activity, and the main pathway was detected on lung cancer by extracts of T. chebula. Aqueous extract of T. chebula was separated using a zeolite, and five fractions of T. chebula extract were obtained and analyzed by ultraviolet (UV) and infrared (IR) spectroscopy. Antiproliferative activity was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) methods against human lung cancer (A549) and mouse lung cancer cell line LLC. T. chebula acts by regulating the Bcl-2 family protein-mediated mitochondrial pathway detected by western blot. Fraction 4 of the T. chebula extract showed much function and was thus studied further. Fraction 4 increased the activation of caspase-3, induced PARP cleavage, and promoted cytochrome c release into the cytoplasm. These data suggest that T. chebula acts by regulating the Bcl-2 family protein-mediated mitochondrial pathway and provide evidence that T. chebula deserves further investigation as a natural agent for treating and preventing cancer.

The effects of ultra-high pressure (UHP) treatment on the chemical properties, colour and sensory quality of young red wine were studied. UHP did not significantly affect the alcohol content, and the methanol content was higher than that of the control, with the latter first increasing and then decreasing with the increase in the pressure or period of treatment. The glycerol content was also markedly changed by UHP, although without a regular pattern. The pH value was not markedly changed by the treatments, and the contents of total acids and volatile acids were scarcely affected. The fructose and glucose contents were clearly changed according to the different treatment conditions. The tartaric acid, citric acid and lactic acid levels showed evident changes, whereas the malic acid level was not changed by UHP treatment. The trend of these changes was similar to what occurs during natural ageing. As the pressure of the treatment was increased to 200 MPa, the chroma value increased, followed by a decline upon higher pressure treatments. The hue of the wine was significantly changed by treatment at different pressures, reaching the highest value after treatment at 400 MPa and then decreasing with increasing pressure. The chroma and hue values of the wine were changed significantly according to the duration of the UHP. After UHP, the appearance, aroma and taste of the wine was improved, with the score for appearance obviously increased. When the wine was treated at 500 MPa for 30 min, its sensory quality received the highest score.
