Abstract Fruit ripening promoted by the exogenous application of ethylene analogs, such as calcium carbide, has commercial advantages. Thus, the knowledge of the responses of fruits treated with ethylene-inducing agents is essential to optimize the use of these substances. This work aimed to evaluate the influence of exposure to calcium carbide on the anticipation and standardization of postharvest ripening of cajá-manga fruits. Physiologically mature fruits were exposed to calcium carbide for 24 hours at concentrations 0, 20, 40, 80, and 110 g m-3. The fruits were stored at a temperature of 28±2 °C and evaluated at 0, 2, 4, and 6 days for the loss of fresh mass, color of the epidermis and pulp given by the CIELAB color space, titratable acidity, soluble solids content, the ratio between soluble solids content and titratable acidity, and vitamin C content. Cajá-manga fruits treated with different concentrations of calcium carbide had their ripening anticipated without compromising their characteristics. The concentrations of 20, 40, and 80 g m-3 of calcium carbide allowed the anticipation and standardization of fruit ripening within four days during storage, while for the highest concentration (110 g m-3), complete maturation was accelerated, occurring between two and four days of storage.

Resumo O amadurecimento de frutos promovido pela aplicação exógena de análogos de etileno, como o carbureto de cálcio, tem vantagens comerciais. Assim, o conhecimento das respostas de frutos tratados com agentes indutores de etileno é essencial para otimizar o uso dessas substâncias. Este trabalho teve como objetivo avaliar a influência da exposição ao carbureto de cálcio na antecipação e na padronização do amadurecimento pós-colheita de frutos de cajá-manga. Frutos fisiologicamente maduros foram expostos ao carbureto de cálcio nas concentrações de 0; 20; 40; 80 e 110 g m-3 por 24 horas. Os frutos foram armazenados a uma temperatura de 28±2 °C e avaliados aos 0; 2; 4 e 6 dias quanto à perda de massa fresca, cor da epiderme e polpa dada pelo espaço de cor CIELAB, acidez titulável, teor de sólidos solúveis, relação entre o teor de sólidos solúveis e a acidez titulável, e teor de vitamina C. Frutos de cajá-manga tratados com diferentes concentrações de carbureto de cálcio tiveram seu amadurecimento antecipado sem comprometer suas características. As concentrações de 20; 40 e 80 g m-3 de carbureto de cálcio permitiram a antecipação e a padronização do amadurecimento dos frutos em quatro dias, durante o armazenamento, Enquanto para a maior concentração (110 g m-3), a maturação completa foi acelerada, ocorrendo entre dois e quatro dias de armazenamento.

ABSTRACT Background: The treatment of multiple myeloma (MM) has evolved significantly in the past decade, and new drug combinations have improved the response rates and prolonged survival. Studies comparing different induction chemotherapy regimens have shown that triple combinations have better results than double combinations. However, comparisons among different triple combinations are rare in the literature. Methods: We retrospectively compared two triple combinations comprising bortezomib, cyclophosphamide and dexamethasone (VCD) versus thalidomide, cyclophosphamide and dexamethasone (CTD), and aimed at identifying which of the two combinations would yield better response rates following four induction cycles prior to hematopoietic cell transplantation in patients with untreated multiple myeloma. Results: We retrospectively reviewed the medical records of 311 patients from 24 different centers.The VCD regimen was used as induction therapy by 117 (37.6%) patients, whereas 194 (62.4%) patients received the CTD regimen. After four cycles of induction on an intention-to-treat basis, 54% of the patients in the VCD group achieved at least very good partial response versus 42.8% in the CTD group (p = 0.05). We observed no difference in neuropathy or thrombotic events rates among the two regimens. Conclusion: Our results corroborate the superiority of the triple combination regimes containing bortezomib over the triple combination with thalidomide as pre ASCT induction therapy in MM.

ABSTRACT Clavulanic acid is a β-lactam compound with potent inhibitory activity against β-lactamases. Studies have shown that certain amino acids play essential roles in CA biosynthesis. However, quantitative evaluations of the effects of these amino acids are still needed in order to improve CA production. Here, we report a study of the nutritional requirements of Streptomyces clavuligerus for CA production. Firstly, the influence of the primary nitrogen source and the salts composition was investigated. Subsequently, soybean protein isolate was supplemented with arginine (0.0–3.20 g L-1), threonine (0.0–1.44 g L-1), ornithine (0.0–4.08 g L-1), and glutamate (0.0–8.16 g L-1), according to a two-level central composite rotatable design. A medium containing ferrous sulfate yielded CA production of 437 mg L-1, while a formulation without this salt produced only 41 mg L-1 of CA. This substantial difference suggested that Fe2+ is important for CA biosynthesis. The experimental design showed that glutamate and ornithine negatively influenced CA production while arginine and threonine had no influence. The soybean protein isolate provided sufficient C5 precursor for CA biosynthesis, so that supplementation was unnecessary. Screening of medium components, together with experimental design tools, could be a valuable way of enhancing CA titers and reducing the process costs.

Resumo: O objetivo deste trabalho foi avaliar o desempenho reprodutivo de jundiá (Rhamdia quelen) alimentado com dietas contendo sorgo em substituição ao milho, suplementadas ou não com fitase. O experimento foi realizado com 360 jundiás, com peso médio inicial de 16,02±0,58 g, alimentados com dietas compostas por 0, 50 e 100% de sorgo de baixo tanino, em substituição ao milho, com ou sem a suplementação de fitase (1.500 UFA por quilograma da dieta). Dietas com suplementação de fitase ou com 100% de sorgo reduziram a deposição de gordura visceral e a massa hepática nos machos. Nas fêmeas, o mesmo efeito foi observado quanto às dietas com suplementação de fitase. As fêmeas alimentadas com dietas apenas com sorgo, suplementadas ou não com fitase, apresentaram ovócitos maiores. Foram encontrados machos e fêmeas nas fases finais de desenvolvimento e maturação gonadal, em todos os tratamentos. A substituição do milho pelo sorgo não afeta a qualidade seminal nem a taxa de fertilização dos gametas. Portanto, a substituição completa do milho pelo sorgo de baixo tanino pode ser efetuada sem afetar os parâmetros reprodutivos e os índices somáticos do jundiá.

Abstract: The objective of this work was to evaluate the reproductive performance of silver catfish (Rhamdia quelen) fed diets containing sorghum, as a replacement for corn, supplemented or not with phytase. The experiment was carried out with 360 fish, with 16.02±0.58 g mean initial weight, fed diets in which low-tannin sorghum replaced corn at 0, 50, and 100%, with or without phytase supplementation (1,500 FTU per kilogram of feed). Diets supplemented with phytase or with 100% sorghum led to a reduction of visceral fat deposition and hepatic mass in males. In females, similar results were observed for diets containing phytase. Females fed sorghum-only diets, supplemented or not with phytase, had bigger oocytes. Males and females at the developing and final stages of gonadal maturation were found in all treatments. The replacement of corn by sorghum affects neither semen quality nor the fertilization rate of gametes. Therefore, complete replacement of corn with low-tannin sorghum in the diet can be performed without compromising the reproductive performance or the somatic indexes of silver catfish.

Resumo A produção de farinhas a partir de frutas, hortaliças e de tubérculos não convencionais para esse processo vem sendo estudada como uma alternativa de produção de alimentos. Dentre os tubérculos, tem-se o taro, o qual pode ser consumido de várias maneiras: cozido, em forma de purê, como ingrediente para sopas e ensopados e em forma de farinha. Desta forma, objetivou-se com este trabalho estudar o processo de secagem por atomização do taro, em leito de jorro, sob diferentes temperaturas e avaliar a influência de condições operacionais no rendimento e na qualidade do produto após sua secagem. As análises físico-químicas realizadas no produto in natura e nas farinhas foram: teor de água, pH, acidez total titulável e ácido ascórbico total, atividade de água (25°C) e cor, além de compostos fenólicos e flavonoides. As farinhas obtidas apresentaram vitamina C variando de 0,05 a 0,17 mg/100g, compostos fenólicos variando de 4,42 a 5,60 mg/100 g e quantidade de flavonoides de 1,05 a 4,30 µg/100 g, tornando-as alimentos fonte de compostos bioativos, os quais podem ser adicionados na elaboração de outros produtos ou inseridos na dieta humana. Conclui-se que a farinha obtida sob a temperatura de 90°C apresentou os maiores conteúdos de vitamina C, porém as farinhas obtidas sob as temperaturas de 70 °C e 80 °C possuem pH próximo da neutralidade, além do teor de água em porcentagem reduzida prolongar a vida de prateleira do produto.

Summary The production of flours from fruits, vegetables and not conventionally processed tubers, has been studied as alternative food production. Taro is one of these tubers, which can be consumed in various ways; cooked, pureed, as ingredients for soups and stews, and in the form of flours. Thus, the aim of this work was to study the drying process of taro by atomization in a spouted bed at different temperatures, and assess the influence of the operating conditions on the yield and quality of the product after drying. The following physicochemical analyzes were carried out on the fresh product and the meal: water content, pH, titratable acidity, total ascorbic acid, water activity (25 °C) and colour, as well as the phenolic compound and flavonoid contents. The flours obtained showed amounts of vitamin C ranging from 0.05 to 0.17 mg/100 g, phenolic compounds varying from 4.42 to 5.60 mg/100 g, and flavonoids from 1.05 to 4.30 µg/100 g making them sources of bioactive compounds, which could be added in the preparation of other products or inserted into the human diet. It was concluded that the flour obtained at 90 °C showed the highest vitamin C content, but the flours obtained at temperatures of 70 °C and 80 °C showed pH values near neutrality, besides a reduced water content favouring a longer storage life of the product.

O objetivo deste estudo foi determinar a composição centesimal e o teor de colesterol da carne do camarão branco do Pacífico (Litopenaeus vannamei), oriundo de uma fazenda localizada no estado do Rio Grande do Norte, Brasil. As análises na porção muscular do camarão foram realizadas após a retirada do cefalotórax e exoesqueleto. De acordo com os resultados obtidos, os teores de umidade, proteínas, cinzas, lipídios e colesterol apresentaram valores médios de 74,1g 100g-1, 21,9g 100g-1, 1,5g 100g-1, 0,3g 100 g-1 e 26,1mg 100g-1, respectivamente. Comparando-se os resultados da presente pesquisa com os encontrados na literatura para outras espécies, observou-se que a carne de L. vannamei apresentou baixo teor de lipídeos e colesterol e maiores concentrações de proteínas, representando grande valor nutricional para o produto.

This study aimed to determine the proximate composition and cholesterol content in muscular portion of Pacific white shrimp (Litopenaeus vannamei) collected from a shrimp farm located in the state of Rio Grande do Norte, Brazil. The analysis in the muscular portion were performed after cephalothorax and exoskeleton removal. According to the results, the moisture, protein, ash, lipids and cholesterol had an average value of 74.1g 100 g-1, 21.9 g 100 g-1, 1.5g 100g-1, 0.3g 100g-1 and 26.1mg 100g-1, respectively. Comparing the results found on this research to data reported in the literature for other species, it can be observed that the flesh of the shrimp L. vannamei showed a low content of lipids and cholesterol and higher concentrations of protein, representing a wide nutritional value to the product.

It has been shown previously that the laticifer fluid of Calotropis procera (Ait.) R.Br. is highly toxic to the egg hatching and larval development of Aedes aegypti L. In the present study, the larvicidal potential of other laticifer fluids obtained from Cryptostegia grandiflora R.Br., Plumeria rubra L. and Euphorbia tirucalli L. was evaluated. We attempted to correlate larvicidal activity with the presence of endogenous proteolytic activity in the protein fraction of the fluids. After collection, the fluids were processed by centrifugation and dialysis to obtain the soluble laticifer protein (LP) fractions and eliminate water insoluble and low molecular mass molecules. LP did not visibly affect egg hatching at the doses assayed. LP from Cr. grandiflora exhibited the highest larval toxicity, while P. rubra was almost inactive. E. tirucalli was slightly active, but its activity could not be correlated to proteins since no protein was detected in the fluid. The larvicidal effects of LP from C. procera and Cr. grandiflora showed a significant relationship with the proteolytic activity of cysteine proteinases, which are present in both materials. A purified cysteine proteinase (papain) from the latex of Carica papaya (obtained from Sigma) was similarly effective, whereas trypsin and chymotrypsin (both serine proteinases) were ineffective. The results provide evidence for the involvement of cysteine proteinase activity in the larvicidal action of some laticifer fluids. C. procera is an invasive species found in areas infested with Ae. aegypti and thus could prove useful for combating mosquito proliferation. This is the first report to present evidence for the use of proteolytic enzymes as chemical agents to destroy Ae. aegypti larvae.

O vírus respiratório sincicial humano (HRSV), isolado em 1955, é a principal causa da hospitalização de bebês e crianças pequenas com sintomas de doença respiratória. No mundo inteiro, vários estudos para o desenvolvimento de uma vacina segura e eficiente contra o HRSV têm tido alta prioridade. A região G2 da glicoproteína G é usada como padrão para genotipagem do HRSV. Neste estudo, foi realizada a análise filogenética da glicoproteína G e o estudo comparativo entre a região G2 e o ectodomínio dessa glicoproteína. Cinquenta e três amostras de swab nasal de crianças com menos de cinco anos de idade, apresentando doença respiratória aguda, atendidas no Hospital Universitário (HU) da Universidade de São Paulo durante o ano de 2004, foram submetidas a sequenciamento por PCR e comparadas com seqüências do GenBank. A região G2 mostrou ser adequada para a genotipagem do HRSV.

The Human Respiratory Syncytial Virus (HRSV), isolated in 1955, is the main cause of hospitalization of babies and infants with respiratory illness. Several studies have been conducted worldwide aiming the development of a safe and effective vaccine against HRSV. The G2 region of glycoprotein G is used as genotyping default. In the present study, we performed a phylogenetic analysis of G protein and a comparative study between G2 region and ectodomain of attachment glycoprotein. Fifty-three nasal swab samples from children less than 5 years old and presenting symptoms of acute respiratory illness, assisted at the University Hospital (UH) of University of Sao Paulo (USP) in 2004, were submitted to sequencing by PCR and compared with GenBank sequences. We concluded that the G2 region is adequate for HRSV genotyping.

A major goal in the treatment of acute ischemia of a vascular territory is to restore blood flow to normal values, i.e. to "reperfuse" the ischemic vascular bed. However, reperfusion of ischemic tissues is associated with local and systemic leukocyte activation and trafficking, endothelial barrier dysfunction in postcapillary venules, enhanced production of inflammatory mediators and great lethality. This phenomenon has been referred to as "reperfusion injury" and several studies demonstrated that injury is dependent on neutrophil recruitment. Furthermore, ischemia and reperfusion injury is associated with the coordinated activation of a series of cytokines and adhesion molecules. Among the mediators of the inflammatory cascade released, TNF-alpha appears to play an essential role for the reperfusion-associated injury. On the other hand, the release of IL-10 modulates pro-inflammatory cytokine production and reperfusion-associated tissue injury. IL-1beta, PAF and bradykinin are mediators involved in ischemia and reperfusion injury by regulating the balance between TNF-alpha and IL-10 production. Strategies that enhance IL-10 and/or prevent TNF-alpha concentration may be useful as therapeutic adjuvants in the treatment of the tissue injury that follows ischemia and reperfusion.

Amastigotes of Leishmania major have a great ability to evade destruction in host cells. This study investigated the activation in resident, inflammatory macrophages and J774 cells in vitro treated with lipopolysaccharide (LPS), soluble Leishmania antigen (SLA), calcium ionophore (CaI) and magnesium (Mg2+) alone or combined. An increase in nitric oxide (NO) production was observed in J774 or inflammatory macrophages treated with LPS alone or in combination with SLA and CaI. The same treatments did not affect the NO release by resident macrophages. There was no interference in uptake of L. major but CaI decreased intracellular proliferation of the parasite. This study demonstrated the importance of CaI in decreasing L. major proliferation inside murine macrophages while Mg2+ seemed to increase parasite proliferation. These finding may help to understand the events involved in host cells' clearance of this pathogen
