Green Synthesis of Cellulose-Based Biofilms Doped with AgNPs with Antibacterial Activity

In this work, cellulose-based composite biofilms doped with silver nanoparticles (AgNPs) have been produced by eco-friendly process. The AgNPs were synthesized from Ilex paraguariensis with an average size of 45 nm, a characteristic plasmon band at 450 nm and zeta potential of -23 mV. The biofilm has been synthesized by casting method, using different volumes of AgNPs suspension with glycerol plasticizing agent (T6-T10) or not (T1-T5). The AgNPs were successfully incorporated into the biofilms, which was confirmed by scanning electron microscopy. The mechanical and optical structural and the antimicrobial properties of the biofilms have been determined. In the presence of AgNPs, the mechanical properties, thermal stability and water vapor barrier of the biofilms have not been altered. However, the addition of glycerol decreased the tensile strength and the modulus of elasticity, in addition to increasing the elongation of the biofilms and the water vapor barrier. The biofilm T10 showed a transmittance of ca. 10%, with high potential to be used as a UV light blocker. Finally, the biofilms doped with AgNPs showed activity against the bacterium Pseudomonas fluorescens. Given the above, this work presented a new biofilm with activity against the bacterium Pseudomonus fluorescens obtained by eco-friendly process.

Lupeol, a triterpene frequently found in Asteraceae plant species, showed moderate to low activity in different strains of Plasmodium falciparum, the most virulent malaria etiological agents. In this work, lupeol was isolated from Parahancornia fasciculata, a plant that is used to treat malaria in the Amazonia region. In the search of more activity lupeol derivatives, five new 1,2,3-triazole hybrid molecules were synthetized by copper-catalyzed azide-alkyne cycloaddition. The antiplasmodial activity of the semi-synthetic compounds were evaluated by the lactate dehydrogenase assay; the lupeol propargyl ether was the only one to disclosing increased activity (half maximal inhibitory concentration-IC50-62.0 ± 1.92 µmol L-1) in relation to lupeol (IC50 117.00 µmol L-1). Therefore, this work revealed a new class of interesting lupeol derivatives that can be obtained by linking electron donors to the hydroxy group at C-3.

We present here our results on the temperature controlled, selective hydrochalcogenation of 1,4-diorganyl-1,3-butadiynes with nucleophilic species of selenium, tellurium and sulfur generated in situ from the respective diaryl dichalcogenide and NaBH4. Using polyethylene glycol (PEG)-400 at 30 ºC the (Z)-chalcogenoenynes are obtained and at 90 ºC the (Z,Z)-bis-chalcogen-1,3-butadienes are produced in good to excellent yields. Alternatively to conventional oil bath heating, the use of microwave irradiation is also presented as an alternative energy source that provides the expected products in few minutes.

We herein report the synthesis of hybrid kauranoid molecules of type 1,2,3-triazole-1,4-disubstituted aiming to improve the antimalarial activity of kaurenoic and xylopic acids. The CuI-catalyzed cycloaddition of azides and kauranoid terminal alkynes was explored as a hybridization strategy. Kauranoid terminal alkynes were prepared from kaurenoic and xylopic acids that were isolated from Wedelia paludosa D. C. (Asteraceae) and Xylopia frutescens Aubl. (Annonaceae). A total of 15 kauranoid derivatives, including nine new triazoles, were obtained and five out of these were more active than the original diterpenes. Interestingly, an increased activity was observed for a kauranoid propargyl ether. Interaction between ent-kaurane diterpene derivatives and Ca2+-ATPase (PfATP6) was investigated. Synthesis of diterpene derivatives emerges as a possible route to be explored in the quest of potentially new inhibitors of PfATP6.

Resumo Até 2010, o conhecimento sobre a diversidade de fungos do Brasil estava registrado em publicações esparsas de taxonomia e ecologia e em algumas poucas listas de espécies. Com a publicação do Catálogo de Plantas e Fungos do Brasil, e a disponibilização da lista online, tem sido possível agregar o conhecimento disperso. A versão ora apresentada acrescenta 2.111 nomes de espécies aos 3.608 listados em 2010. São citadas 5.719 espécies de fungos distribuídas em 1.246 gêneros, 102 ordens e 13 divisões, consistindo em considerável aumento em relação a 2010, quando estavam registrados 924 gêneros e 78 ordens. Predominam os Basidiomycota (2.741 espécies, em 22 ordens) e Ascomycota (1.881 espécies, em 41 ordens). A Mata Atlântica possui a maior quantidade de registros, com 3.017 espécies, seguido pela Amazonia (1.050), Caatinga (999), Cerrado (638) e Pampa e Pantanal com 84 e 35 espécies, respectivamente. A região Nordeste tem a maior riqueza (2.617 especies), seguida pelo Sudeste (2.252), Sul (1.995), Norte (1.301) e Centro Oeste (488 espécies). Em relação aos Estados da Federação, São Paulo (1.846 espécies), Pernambuco (1.611) e Rio Grande do Sul (1.377) são os mais diversos.
Abstract Knowledge about the Brazilian fungal diversity was, until 2010, recorded in few taxonomy and ecology publications, as well as in a handful of species lists. With the publication of the Catálogo de Plantas e Fungos do Brasil and the continued availability of an online list, it has been possible to aggregate this dispersed knowledge. The version presented here adds 2,111 species names to the 3,608 listed in 2010. A total of 5,719 species of fungi distributed in 1,246 genera, 102 orders and 13 phyla represents a considerable increase over the last five years, when only 924 genera and 78 orders were registered. Basidiomycota (2,741 species in 22 orders) and Ascomycota (1,881 species in 41 orders) predominate over other groups. The Atlantic Rainforest has the largest number of records, with 3,017 species, followed by Amazon Rainforest (1,050), Caatinga (999), Cerrado (638) and Pampa and Pantanal with 84 and 35 species, respectively. The Northeast region has the greatest richness (2,617 species), followed by Southeast (2,252), South (1,995), North (1,301) and Central-West (488 species). Regarding the States of the Federation, São Paulo with 1,846 species, Pernambuco with 1,611 and Rio Grande do Sul with 1,377 species are the most diverse.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Cromatografia gasosa bidimensional abrangente acoplada à espectrometria de massas por tempo de voo (GC×GC-TOFMS) é uma técnica apropriada para a elucidação da composição molecular de amostras petroquímicas, como óleos biodegradados. Biomarcadores foram separados, identificados e razões de biomarcadores convencionais foram determinados usando cromatografia gasosa acoplada à espectrometria de massas (GC-MS) e GC×GC-TOFMS. No cromatograma de íons extraídos m/z 123 + 177 + 191, coeluições entre terpanos tricíclicos, hopanos e 25-nor-hopanos com secohopanos foram resolvidas usando GC×GC-TOFMS. GC×GC-TOFMS permitiu a identificação da série completa dos 25-nor-hopanos, nor-gamacerano, C29 28-nor-espergulano e oleanano, que não foram identificados por GC-MS. A avaliação dos parâmetros geoquímicos dos óleos estudados indicou uma origem marinha e um ambiente deposicional sob condições anóxicas. A alta resolução cromatográfica e sensibilidade alcançada usando GC×GC-TOFMS permitiram sugerir três novos parâmetros geoquímicos para a caracterização de óleos altamente biodegradados. Estes resultados demonstram a superioridade da técnica GC×GC-TOFMS na separação e identificação de compostos individuais e não-alvos em óleos severamente biodegradados
Comprehensive two-dimensional gas chromatography with time of flight mass spectrometry (GC×GC-TOFMS) is an appropriate technique for the elucidation of molecular composition of petrochemical samples, such as biodegraded oils. Biomarkers were separated and identified, and conventional biomarker ratios were determined via gas chromatography-mass spectrometry (GC-MS) and GC×GC-TOFMS. In the extracted ion chromatogram m/z 123 + 177 + 191, coelutions between tricyclic terpanes, hopanes and 25-nor-hopanes with secohopanes were resolved by GC×GC-TOFMS. GC×GC-TOFMS allowed the identification of complete series of 25-nor-hopanes, nor-gammacerane, C29 28-nor-spergulanes and oleanane not identified by using GC-MS. The biomarker ratios from the studied oils indicated that they derived from marine source rock deposited under anoxic conditions. The higher chromatographic resolution and sensitivity achieved by using GC×GC-TOFMS allowed for three new parameters to characterize biodegraded oils. These results indicated the superiority of GC×GC-TOFMS for separation and identification of individual and non-target compounds in severely biodegraded oils.

O objetivo deste trabalho foi avaliar critérios de seleção em progênies de cruzamento entre a cultivar de tomateiro Santa Clara (Solanum lycopersicum) e a espécie silvestre S. habrochaites f. glabratum, quanto a atributos de qualidade dos frutos e de resistência à requeima (Phytophthora infestans). As famílias foram avaliadas em delineamento de blocos ao acaso, em dois ensaios, com duas repetições e seis testemunhas comuns a ambos os ensaios. Ganhos diretos e indiretos foram estimados entre famílias F2:3 para seleção simultânea quanto à resistência à requeima, determinada pela quantificação da área abaixo da curva de progresso da doença (AACPD), e quanto à acidez titulável e aos teores de sólidos solúveis dos frutos. Os critérios de seleção proporcionaram ganhos genéticos satisfatórios, adequados ao ideótipo proposto de decréscimo na AACPD e de incremento nos valores médios de sólidos solúveis e acidez titulável. A seleção direta e indireta e o índice de Mulamba & Mock resultam em ganhos individuais mais equilibrados e em maiores ganhos totais.
The objective of this work was to evaluate selection criteria in progenies from the crossing of tomato cultivar Santa Clara (Solanum lycopersicum) and the wild species S. habrochaites f. glabratum as to attributes of fruit quality and of resistance to late blight (Phytophthora infestans). The families were evaluated in a randomized block design, in two trials, with two replicates and six controls, common to both trials. Direct and indirect gains were estimated between F2:3 families for simultaneous selection regarding resistance to late blight, determined by the quantification of the area under the disease progress curve (AUDPC), and regarding titratable acidity and soluble solid contents of fruit. The selection criteria provided satisfactory genetic gains, suitable for the proposed ideotype of decreases in the AUDPC and increases in the average values of soluble solids and titratable acidity. The direct and indirect selection and the Mulamba & Mock index result in more balanced individual gains and higher total gains.

Degradation of non-vulcanized natural rubber - renewable resource for fine chemicals used in polymer synthesis

In the current scenario, there is growing interest in the products of degradation of rubber (natural and synthetic) for specific applications in different industry sectors, whose benefits in replacing conventionally used products are mainly related to sustainability. Since the degradation products of rubber can be used in different areas, several research groups may have the interest aroused by these products, but are not familiar with the aspects related to the chemical behavior of rubber. This review aims to bring together the key information in the published literature on the degradation of natural rubber, emphasizing metatheses reactions, oxidative damage and splitting of the double bond, in order to serve as a reference source for researchers from different fields interested in obtaining such kind of products. The structures and properties as well as additional chemical transformations resulting in oligomers of isoprene, functionalised oligomers and polymers based on both are also described.

Glicocorticosteroides estão incluídos na lista de substâncias proibidas da Agência Mundial Antidoping como analitos que dispensam quantificação. Entretanto, abordagens semiquantitativas são necessárias antes de relatar um resultado analítico adverso para essa classe de substâncias. Cromatografia líquida acoplada à espectrometria de massa (LC-MS) tem sido empregada com excelente seletividade e especificidade na análise de xenobiontes em misturas complexas. Foram analisadas amostras contendo glicocorticosteroides exógenos, a partir de diferentes vias de administração, utilizando o método proposto. Amostras geradas após administração sistêmica (via oral) apresentaram níveis de prednisolona ou prednisona superiores a 30 ng mL-1, limite mínimo de desempenho requerido usado como parâmetro de diagnóstico. Por outro lado, vias não-sistêmicas parecem não apresentar biodisponibilidade para gerar um caso adverso em análises de controle de dopagem. Considerando o número reduzido de estudos similares na literatura, mais estudos de excreção podem ser realizados usando o método relatado com o objetivo de fornecer mais informações sobre o perfil de excreção de glicocorticosteroides administrados por vias sistêmicas e não sistêmicas.
Glucocorticosteroids are included in the World Anti-Doping Agency (WADA) prohibited list as non-threshold analytes. Nevertheless, at least semi-quantitative approaches are necessary before reporting an adverse analytical finding for this class of substances. The liquid chromatography coupled to mass spectrometry (LC-MS) has been used with excellent selectivity and specificity in xenobiotic analysis in complex matrices. Real samples containing exogenous glucocorticosteroids from different administration routes were analyzed using the method proposed. Samples from systemic administration (oral route) presented levels of prednisolone or prednisone higher than 30 ng mL-1, the minimum requirement performance level used as diagnostic parameter. On the other hand, non-systemic routes seem to not have bioavailability to generate an adverse case in doping control analysis. Considering the low number of similar studies available in the literature, more urinary studies could be done using the method reported aiming to provide more information about the excretion profile of glucocorticosteroids administered by systemic and non-systemic routes.

Dengue is a tropical disease caused by an arbovirus transmitted by Aedes aegypti. Since no effective vaccine is available for treating dengue, the present study focused on population vector control through investigating the use of the lignan grandisin, isolated from Piper solmsianum C. DC., Piperaceae, against the larvae of A. aegypti. Grandisin caused larval (L3) mortality at LC50 150 µg/mL. Histological analysis on A. aegypti larvae treated with grandisin (LC50 50 µg/mL) showed changes in the anterior-middle midgut, with intense tissue destruction and cell disorganization.

Muitos métodos para estimativa de idade de mamíferos têm sido utilizados ultimamente. Entretanto, a maioria deles requerem procedimentos laboratoriais, que geralmente, não estão disponíveis em campo. Neste artigo nós definimos categorias de idade para seis espécies de mamíferos através da análise de desgaste dentário, erupção dentária e massa corporal. Nós analisamos 632 crânios, incluindo Mazama nemorivaga (veado cinza), Mazama americana (veado vermelhor), Tayassu pecari (queixada), Pecari tajacu (cateto), Cuniculus paca (paca) and Dasyprocta leporina (cotia). Usando critérios biológicos e morfológicos nós definimos quatro categorias de idade para cada espécie: jovem, sub-adulto, adulto e adulto senescente. Foram definidos padrões de desgaste e erupção dos molares para cada categoria de idade para cada uma das espécies analisadas. Nós encontramos diferenças significativas das médias de peso entre as categorias de idade para as espécies D. leporina (F = 81.1; df = 3; p < 0.05), C. paca (F = 39.0; df = 3; p < 0.05), P. tajacu (F = 24.6; df = 3; p < 0.05) and M. americana (F = 30.6; df = 3; p < 0.05), sugerindo que a massa corporal pode ser usada como um critério complementar de estimativa de idade. Embora seja um método menos quantitativo do que aqueles citados com mais frequência na literatura, como deposição de bandas de cementum, o método descrito neste artigo mostrou ser uma ferramenta eficiente para pesquisadores e manejadores em trabalhos de campo na Amazônia brasileira. Embora métodos similares já tenham sido utilizados em estudos de caça, é a primeira vez que esta metodologia é descrita em detalhes, podendo ser replicada em outros estudos semelhantes.
Many methods of age determination have been used to date. However, most of them require laboratory procedures which are, in general, not available for wildlife managers in the field. In this paper we attempt to standardize the age categories for six mammal species through the analysis of dental wear, dental eruption and body mass. We analyzed 632 skulls, including Mazama nemorivaga (brown brocket deer), Mazama americana (red brocket deer), Tayassu pecari (white-lipped peccary), Pecari tajacu (collared peccary), Cuniculus paca (paca) and Dasyprocta leporina (agouti). Using biological and morphological criteria we define four age categories: young, sub adult, adult and senescent adult. Scores and variables of molar cusp wear and eruption were defined for each age category within each species. We found significant differences in mean weights among age categories for D. leporina (F = 81.1; df = 3; p < 0.05), C. paca (F = 39.0; df = 3; p < 0.05), P. tajacu (F = 24.6; df = 3; p < 0.05) and M. americana (F = 30.6; df = 3; p < 0.05), suggesting that body mass can be a complementary criterion for estimating age. Despite being less quantitative than methods such as those which use deposition of cementum layers, the method described here proved to be an efficient tool for field researchers and managers in the Brazilian Amazon. Although similar methods have already been applied in the study of hunted mammals in the Peruvian and Brazilian Amazon, it is the first time such a method is described in detail and therefore it can be easily used in future works.

A expressão do sistema enzimático produzido por fungos basidiomicetos envolvido na degradação de xenobióticos é bastante dependente das condições de cultivo, principalmente da composição do meio de cultivo. Trametes villosa CCB176 é uma linhagem com comprovado potencial biotecnológico para degradação de compostos organoclorados e descoloração de corantes têxteis. Foi avaliada a influência da concentração de glicose, adição de emulsão de óleo vegetal e surfactante, natureza do surfactante e os metais manganês e cobre no crescimento, pH e na produção das atividades de lacase, de peroxidases totais e de peroxidase dependente de manganês. No geral, ocorreu acidificação do meio com pH atingindo valor próximo a 3,5, nos primeiros dias de crescimento. Lacase foi a principal atividade detectada nas diferentes condições e sua produção se deu durante todo o período de cultivo do fungo, independente da fase de crescimento. Suplementação do meio com óleo vegetal emulsificado com surfactante resultou em indução da atividade de peroxidase dependente de manganês produzida por T. villosa. Maiores valores de rendimento específico da atividade de lacase foram propiciados pelo cobre.
The expression of the enzymatic system produced by basidiomycetous fungi, which is involved in the degradation of xenobiotics, mainly depends on culture conditions, especially of the culture medium composition. Trametes villosa is a strain with a proven biotechnological potential for the degradation of organochlorine compounds and for the decolorization of textile dyes. The influence of glucose concentration, addition of a vegetable oil-surfactant emulsion, nature of the surfactant and the presence of manganese and copper on the growth, pH and production of laccase, total peroxidase and manganese-dependent peroxidase activities were evaluated. In general, acidification of the medium was observed, with the pH reaching a value close to 3.5 within the first days of growth. Laccase was the main activity detected under the different conditions and was produced throughout the culture period of the fungus, irrespective of the growth phase. Supplementation of the medium with vegetable oil emulsified with a surfactant induced manganese-dependent peroxidase activity in T. villosa. Higher specific yields of laccase activity were obtained with the addition of copper.
