Benzophenone Derivatives Showed Dual Anti-Inflammatory and Antiproliferative Activities by Inhibiting COX Enzymes and Promote Cyclin E Downregulation

Considering the promising antitumor effects of compounds with dual anti-inflammatory and antiproliferative activities, thus benzophenones analogs (2-7) were evaluated on in vivo anti-inflammatory assay and molecular docking analysis. Those with the best molecular docking results were in vitro evaluated on cyclooxygenase (COX) enzymes and tested regarding antiproliferative activity. All derivatives displayed in vivo anti-inflammatory activity. Among them, the substances 2’-hydroxy-4’-benzoylphenyl-β-D-glucopyranoside (4), 4-hydroxy-4’-methoxybenzophenone (5) and 4’-(4’’-methoxybenzoyl)phenyl-β-D-glucopyranoside (7) showed the best values of Glide Score in COX-2 docking evaluation and 4 and 5 selectively inhibited COX-2 and COX-1 in vitro enzymatic assay, respectively. Thus, 4 and 5 were tested against breast cancer (MCF-7, MDA MB-231, Hs578T) and non-small-cell-lung cancer (A549) cell lines. The estrogen-positive MCF-7 cell line was more responsive compared to other tested cell lines. They induced cell cycle arrest at G1/S transition in MCF-7 cell line once there was an increase in G0/G1 population with concomitant reduction of S population. The antiproliferative activity of these substances on MCF-7 was associated with their ability to inhibit cyclin E expression, a critical regulator of G1/S transition. Taken together, the data indicate that 4 and 5 have dual anti-inflammatory and antiproliferative activities and support further studies to evaluate their antitumor potential.

OBJECTIVE: To assess the incidence of intra-operative immediate hypersensitivity reactions and anaphylaxis. METHODS: A cross-sectional observational study was conducted at the Department of Anesthesiology, University of São Paulo School of Medicine, Hospital das Clínicas, São Paulo, Brazil, from January to December 2010. We developed a specific questionnaire to be completed by anesthesiologists. This tool included questions about hypersensitivity reactions during anesthesia and provided treatments. We included patients with clinical signs compatible with immediate hypersensitivity reactions. Hhypersensitivity reactions were categorized according to severity (grades I-V). American Society of Anesthesiologists physical status classification (ASA 1-6) was analyzed and associated with the severity of hypersensitivity reactions. RESULTS: In 2010, 21,464 surgeries were performed under general anesthesia. Anesthesiologists answered questionnaires on 5,414 procedures (25.2%). Sixty cases of intra-operative hypersensitivity reactions were reported. The majority patients (45, 75%) had hypersensitivity reactions grade I reactions (incidence of 27.9:10,000). Fifteen patients (25%) had grade II, III or IV reactions (intra-operative anaphylaxis) (incidence of 7:10,000). No patients had grade V reactions. Thirty patients (50%) were classified as ASA 1. The frequency of cardiovascular shock was higher in patients classified as ASA 3 than in patients classified as ASA 1 or ASA 2. Epinephrine was administered in 20% of patients with grade III hypersensitivity reactions and in 50% of patients with grade II hypersensitivity reactions. CONCLUSIONS: The majority of patients had hypersensitivity reactions grade I reactions; however, the incidence of intra-operative anaphylaxis was higher than that previously reported in the literature. Patients with ASA 3 had more severe anaphylaxis; however, the use of epinephrine was not prescribed in all of these cases. Allergists and anesthesiologists should implement preventive measures to reduce the occurrence of anaphylaxis.

RESUMO O objetivo foi avaliar a biologia reprodutiva do camarão- sossego Macrobrachium jelskii (Miers, 1877) no Rio São Francisco, com ênfase na razão sexual, período reprodutivo, maturidade sexual e fecundidade. As coletas foram realizadas mensalmente entre março de 2005 e fevereiro de 2006 exceto nos meses de abril e outubro, nos dois pontos de coleta do Rio São Francisco, Represa de Três Marias (Estação Ecológica de Pirapitinga), a montante da barragem da usina hidroelétrica de Três Marias e a jusante da barragem, localizados no município de Três Marias, região central de Minas Gerais, Brasil. O puçá de 1 m² de boca e malha de 1,42 mm foi passado sob a vegetação marginal e na água à margem dos dois pontos de coleta, por um coletor em um período de até 30 minutos. Em laboratório, os espécimes foram identificados, mensurados e sexados. Um total de 2.945 espécimes foi analisado a montante, no qual correspondem a 1.064 machos e 1.857 fêmeas (1.653 não ovígeras e 204 ovígeras) e 1.973 a jusante, onde 1.120 machos e 841 fêmeas (643 não ovígeras e 198 ovígeras). A razão sexual diferiu significativamente a favor de fêmeas de M. jelskii a montante (0,57; χ2= 215,28; p < 0.01) e a favor dos machos a jusante (1,33; χ2= 39,69; p < 0.01). Ambas as populações possuem um período reprodutivo contínuo-sazonal com pico em janeiro e novembro para os camarões a montante e a jusante, respectivamente. A fecundidade média de M. jelskii foi de 37 ± 14 ovos (5 a 78 ovos) a montante da barragem e de 35 ± 14 ovos (5 a 69 ovos) a jusante. A razão sexual desviada para os machos de M. jelskii à jusante sugere que as fêmeas apresentaram o comportamento de se abrigarem entre as vegetações marginais. A razão sexual desviada para as fêmeas a montante demonstra a real proporção de machos e fêmeas, que pode ser considerada para a espécie no Rio São Francisco. O tamanho dos ovos carregados pelas fêmeas a jusante foi influenciado pelas baixas temperaturas da água proveniente da represa.
ABSTRACT The objective was evaluate the reproductive biology of Macrobrachium jelskii (Miers, 1877) in São Francisco River, with emphasis on sex ratio, reproductive period, sexual maturity and fertility. The samples were collected monthly from March 2005 to February 2006 except April and October 2005, in two sites in São Fransico River, upstream of the Três Marias Dam (Pirapitinga Ecological Station) and downstream of the dam, located in Três Marias, central region of Minas Gerais state, Brazil. The netting mouth had 1 m² and the mesh 1.42 mm that it was passed under the marginal vegetation and the water next to the bank during 30 minutes. In the laboratory, specimens were identified, measured and sexed. A total of 2,945 specimens were captured in the upstream, 1,064 males and 1,857 females (1,653 non-ovigerous and 204 ovigerous) and 1,973 individuals in the downstream, 1,120 males and 841 females (643 non-ovigerous and 198 ovigerous). The sex ratio differed significantly for females of M. jelskii in upstream (0.57, χ2 = 215.28, p < 0.01), and males in the downstream (1.33, χ2 = 39.69, p < 0.01). In both sites the reproductive period was a continuous - seasonal with the peak occurring in January and November for shrimps of upstream and downstream, respectively. The mean fecundity of M. jelskii was 37 ± 14 eggs (5-78 eggs) in the upstream of the dam and 35 ± 14 eggs (5-69 eggs) in the downstream of the dam. The sex ratio showed deviation to males of M. jelskii in the downstream suggesting that females presenting behavior of hiding in the marginal vegetation. And in the upstream the sex was deviate to females, demonstrating the actual proportion of males and females of the population, probably because females do not find places to hide. The eggs size carried by females downstream was influenced by the low water temperatures from the dam.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Antimicrobial resistance and detection of mecA and blaZ genes in coagulase-negative Staphylococcus isolated from bovine mastitis

O presente estudo avaliou o perfil fenogenotípico de resistência aos antimicrobianos em espécies de Staphylococcus coagulase-negativo (ECN) isoladas do leite de vacas com mastite, em especial considerando a oxacilina. Dos 100 isolados de ECN, S.xylosus foi a espécie predominante, seguida por S. cohnii, S. hominis, S. capitis and S. haemolyticus Apenas 6% dos isolados foram fenotipicamente suscetíveis aos agentes antimicrobianos testados no ensaio de difusão em disco. O percentual de resistência à penicilina e ampicilina foi significativamente maior que aos outros antimicrobianos. Quatro isolados foram positivos para o gene mecA (4%), sendo todos representados pela espécie S.xylosus. O gene blaZ foi detectado em 16% dos isolados (16/100), sendo todos mecA positivos e a presença de ambos os genes foi correlacionada com a resistência fenotípica aos beta-lactâmicos. Foi possível concluir que as espécies de ECN provenientes de leite bovino apresentaram distintos perfis de resistência antimicrobiana, avaliados por testes fenotípicos e genotípicos, podendo dificultar a adoção de medidas de tratamento e manejo dos animais.
The present study evaluated the pheno- and genotypical antimicrobial resistance profile of coagulase-negative Staphylococcus (CNS) species isolated from dairy cows milk, specially concerning to oxacillin. Of 100 CNS isolates, the S. xylosus was the prevalent species, followed by S. cohnii, S. hominis, S. capitis and S. haemolyticus. Only 6% were phenotypically susceptible to the antimicrobial agents tested in disk diffusion assay. Penicillin and ampicillin resistance rates were significantly higher than others antimicrobials. Four isolates were positive to mecA gene (4%), all represented by the S. xylosus species. The blaZ gene was detected in 16% of the isolates (16/100). It was noticed that all mecA + were also positive to this gene and the presence of both genes was correlated to phenotypic beta-lactamic resistance. We conclude that CNS species from bovine milk presented significantly distinct antimicrobial resistance profiles, evaluated by phenotypic and genotypic tests, which has implications for treatment and management decisions.

Reproductive biology of the sesarmid crab Armases rubripes (Decapoda, Brachyura) from an estuarine area of the Sahy River, Sepetiba Bay, Rio de Janeiro, Brazil

O objetivo desse trabalho foi obter informações sobre a biologia reprodutiva do caranguejo Armases rubripes (Rathbun, 1897) numa área estuarina da Baía de Sepetiba. Exemplares foram obtidos mensalmente de fevereiro 2003 a janeiro 2004 no estuário do rio Sahy (22º56'S; 44º01'W), Rio de Janeiro, Brasil. Os caranguejos foram coletados manualmente durante 15 minutos por estação de coleta e por dois coletores. Em laboratório, os indivíduos foram separados por sexos, mensurados em relação à largura da carapaça e os estágios gonadais observados macroscopicamente. Um total de 830 indivíduos foram obtidos - 304 machos, 373 fêmeas (60 fêmeas ovígeras) e 153 juvenis. As fêmeas ovígeras foram encontradas durante todo o período de estudo, exceto em novembro e abril, demonstrando um período reprodutivo contínuo. O tamanho das fêmeas ovígeras variou entre 8,2 e 15,0 mm de largura da carapaça (12,1 ± 1,7 mm). As gônadas observadas apresentaram cinco estágios de desenvolvimento, de acordo com a coloração e o aspecto macroscópico (imatura, rudimentar, intermediária, desenvolvida e desovada). A maturidade sexual foi estimada aos 6,5 mm de largura da carapaça entre os machos e 8,1 mm entre as fêmeas. A fecundidade individual variou entre 200 e 11.460 ovos (4.458 ± 2.739 ovos). O tamanho médio dos ovos foi de 0,248 ± 0,026 mm, variando entre 0,213 e 0,333 mm, enquanto o volume variou entre 0,0051 e 0,0188 mm³ (0,0082 ± 0,0029 mm³).
The aim of this work was to gain knowledge about reproductive biology of the crab Armases rubripes (Rathbun, 1897) from an estuarine area of the Sepetiba Bay. Samples were taken monthly from February 2003 to January 2004 in the Sahy River estuary (22º56'S; 44º01'W), Rio de Janeiro, Brazil. The crabs were collected by hand during 15-minute catch-effort sessions conducted by two people. In the laboratory, the specimens were separated by sex, carapace width was measured and gonadal stage was checked macroscopically. A total of 830 individuals were caught - 304 males, 373 females (60 ovigerous females) and 153 juveniles. The ovigerous females were found almost year-round, except in November and April, showing a continuous reproductive period. They presented a size range from 8.2 to 15.0 mm carapace width (12.1 ± 1.7 mm). Color and macroscopical aspects determined five gonadal stages for males and females (immature, rudimentary, intermediary, developed and resting). First sexual maturity was estimated at 6.5 mm of carapace width for males and 8.1 mm for females. Individual fecundity varied from 200 to 11,460 eggs (4,458 ± 2,739 eggs). Mean egg size was 0.248 ± 0.026 mm, varying from 0.213 to 0.333 mm, while the volume ranged from 0.0051 to 0.0188 mm³ (0.0082 ± 0.0029 mm³).
