Resumen La evaluación de la expresión del ligando 1 de muerte programada (PD-L1) es un criterio de elegibilidad para la inmunoterapia anti-PD-L1 en el carcinoma oral de células escamosas (COCE). También deben evaluarse otros componentes del microambiente tisular. Aquí se informaron cuatro casos de carcinoma de células escamosas orales. Se realizaron análisis histopatológicos del grado tumoral y TIL (infiltrado inflamatorio asociado a tumor) e inmunohistoquímica para CD4, CD8 y PD-L1. Los casos se clasificaron según el microambiente tisular en resistencia inmunitaria adaptativa, ignorancia inmunológica, tolerancia o inducción intrínseca. Este análisis destaca la importancia de comprender el microambiente inmunológico que puede presentar el COCE para la identificación de dianas terapéuticas más eficaces contra la enfermedad.
Abstract The evaluation of the expression of programmed cell death protein (PD-L1) is an eligibility criterion for anti-PD-L1 immunotherapy in oral squamous cell carcinoma (OSCC). Other components of tissue microenvironment should also be evaluated. Here, four cases of oral squamous cells carcinoma were reported. Histopathological analysis of tumor grade and TILs and immunohistochemistry for CD4, CD8, and PD-L1 was done. The cases were classified according to the tissue microenvironment in adaptive immune resistance, immunological ignorance, tolerance, or intrinsic induction. This analysis highlights the importance of understanding the immunological microenvironment that the OSCC can present for the identification of more effective therapeutic targets against the disease.

The benzophenones are synthetic and natural compounds presenting a variety of activities, including photoprotective, cytotoxic and antiviral. It is herein described the preparation of a series of twenty-seven benzophenone derivatives bearing 1,2,3-triazole functionalities and the evaluation of their photoprotective, cytotoxic and antiviral on Zika Virus (ZIKV) activities. The compounds were prepared in three steps, namely reduction of benzophenone, alkylation of diphenylmethanol and CuAAC reactions. The in vitro evaluation of the photoprotective activity revealed that the most active derivative 4-((benzhydryloxy)methyl)-1-(4-nitrobenzyl)-1H-1,2,3-triazole (4k) displayed UVB sun protection factor equal to 6,9±0,53, which make this compound a possible candidate to be used in formulations for photoprotective applications. In terms of cytotoxicity, the compounds were evaluated against MDA-MB-231 and B16F10 cell lines. It was observed that the compounds were more active against MDA-MB-231 cells and three of them were capable of reducing cell viability by approximately 55% at 100 µmol L-1. In the antiviral screening against ZIKV, compound 4-(3-benzhydryloxy)propyl)-1-(3-methylbenzyl)-1H-1,2,3-triazole (5j) was the most effective in mantaining Vero cell viability.

Infestation of two shrimp species of the genus Palaemon Fabricius, 1798 (Decapoda, Palaemonidae) by an isopod of the genus Probopyrus Giard & Bonnier, 1888 (Bopyridae) from the Brazilian southeast coast

Abstract We determined the infestation rate of Probopyrus sp. in populations of Palaemon pandaliformis (Stimpson, 1871) and P. northropi (Rankin, 1898) in the Ubatumirim River, localized in a mangrove ecosystem on Ubatumirim Beach, northern coast of the state of São Paulo, Brazil. Samplings were carried out monthly from April 2003 to March 2004. Monthly prevalence varied from 0 to 4.94 % for P. pandaliformis, and from 0 to 4.54 % for P. northropi. This is the first record of Probopyrus sp. infesting the studied species in this region. Species of Probopyrus (Giard and Bonnier, 1888) seem to have a high plasticity with regard to palaemonid hosts, as they can be parasites of shrimps in both Palaemon (Fabricius, 1798) and Macrobrachium (Spence Bate, 1868). The linear relationships between the parasite and host sizes suggest that the parasite infests both hosts early in their development. We concluded that the infestation of Probopyrus sp. has little impact on Palaemon populations, mostly due to the low prevalence of infestation.

Urochloa decumbens growth and P uptake as affected by long-term phosphate fertilization, mycorrhizal inoculation and historical land use in contrasting Oxisols of the Brazilian Cerrado

RESUMO No manejo da fertilidade de Latossolos brasileiros altamente intemperizados e lixiviados, o P é o macronutriente mais limitante. Assim, conduziu-se um experimento em casa de vegetação, objetivando-se avaliar a influência da interação entre doses de P, inoculação micorrízica e histórico de uso do solo no crescimento de Urochloa decumbens e absorção de P, em quatro Latossolos com atributos químicos, físicos e mineralógicos contrastantes. As plantas foram cultivadas em vasos contendo 4 kg de solo, dispostos em um delineamento inteiramente casualizado, com quatro repetições e esquema fatorial 2x2x2: duas doses de P; com e sem inoculação micorrízica; solos cultivados por longos períodos e não cultivados (sob vegetação nativa). Realizaram-se dois cultivos com duração de 10 semanas cada. Após cada cultivo avaliaram-se a matéria seca da parte aérea, teor e acúmulo de P na parte aérea. No primeiro cultivo, maior resposta ao P aconteceu nos solos sob vegetação nativa em associação à inoculação e adição de P. No segundo cultivo, a inoculação teve maior efeito em todos os solos, associada à menor dose de P. O aumento do P no solo aumenta a concentração desse nutriente na parte aérea. A inoculação não teve efeito na concentração e no acúmulo de P na parte aérea de Urochloa. O crescimento de Urochloa decumbens foi fortemente influenciado pela interação entre classe de solo x histórico de uso da terra x dose de P x inoculação.
ABSTRACT In the fertility management of highly weathered-leached Brazilian Oxisols, P is the most limiting macronutrient. A greenhouse experiment was conducted with the objective to evaluate the influence of the interaction between P doses, mycorrhizal inoculation and historical land use on Urochloa decumbens growth and P uptake in four Oxisols with contrasting chemical, physical and mineralogical properties. The plants were cultivated in plastic pots containing 4 kg of soil in a completely randomized design, four replications and 2x2x2 factorial scheme: with two P doses; with and without mycorrhizal inoculation; soils cultivated for long periods and non-cultivated (under native vegetation). There were two plantings of ten weeks each. Shoot dry mater, concentration and accumulation of P in the shoot were evaluated. In the first planting, the Urochloa response was greater in non-cultivated soils associated with inoculation and P addition. However, in the second planting, the inoculation had a greater effect in all soils compared to the first planting associated with the lowest P dose. As the P concentration in the soil increased, P in the shoot dry matter increases. The inoculation did not affect the P concentration and accumulation in the shoot of Urochloa. The growth of Urochloa decumbens was strongly influenced by the interaction among soil class x history of land use x dose of P x inoculation.

Chlorophyll index for real-time prediction of nutritional status of 'Prata' banana

RESUMO Objetivou-se, com o presente trabalho, selecionar equações de regressão com base na correlação entre o índice de clorofila e os teores foliares de nutrientes para predição em tempo real do estado nutricional de bananeiras tipo Prata. Utilizaram-se, como tratamentos, seis cultivares de bananeira tipo Prata com cinco repetições e quatro plantas úteis por parcela, dispostas em delineamento experimental inteiramente casualizado. Avaliaram-se os teores de nutrientes com base em análises laboratoriais e os índices de clorofila com um medidor portátil na terceira folha do ápice para a base. Os dados foram submetidos à análise de variância; calcularam-se as correlações entre teores foliares de nutrientes e índices de clorofila e se ajustaram equações de regressões para as associações significativas e com maior magnitude. Os modelos selecionados estimam os teores foliares de nutrientes e permitem predizer com segurança em tempo real e a baixo custo, o estado nutricional de bananeiras tipo Prata. Constitui uma técnica auxiliar que, aliada à diagnose foliar e à análise de solos, contribui para uma avaliação nutricional mais criteriosa e segura.
ABSTRACT This study aimed to select regression equations based on the correlation between chlorophyll index and leaf nutrient contents, for real-time prediction of the nutritional status of 'Prata' banana. Six cultivars of 'Prata' banana were used as treatments, with five replicates and four plants per plot, arranged in a completely randomized design. Nutrient levels were evaluated based on laboratorial analysis and chlorophyll indices using a portable chlorophyll meter, in the third leaf from the apex to the base. Data were subjected to analysis of variance, calculating the correlations between leaf nutrient contents and chlorophyll indices, and the regression equations were adjusted to the associations that were significant and with greater magnitude. The selected models estimate leaf nutrient contents and allow a real-time, low-cost reliable prediction of the nutritional status of 'Prata' banana. It is an auxiliary technique that, combined with leaf and soil analysis, contributes to a more precise and secure nutritional evaluation.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Control of papaya fruits anthracnose by essential oil of Ricinus communis

The aim of this work was to investigate the potential of castor oil for the control of papaya diseases caused by the fungus Colletotrichum gloeosporioides and the bacterium Pseudomonas caricapapayae. The treatment with 1% castor oil did not significantly affect the fungal growth. The effectiveness of castor oil for the control of anthracnose was shown when 5% and 10% (v/v) were used in the assays resulting in reduced mycelial growth. Fungal sporulation was strongly inhibited at 10% (v/v) concentration of essential oil. The studies with the fresh fruits treated with 5% (v/v) castor oil in aqueous emulsions resulted in effective reduction of pathogen spread in these fruits. No lesion was found in the fruits treated with oil, when compared to the control fruits. Castor oil showed no effect against the P. caricapapayae when tested in vitro. These results suggested the potential use of the castor bean essential oil and its fatty acids constituents for the control of anthracnose in papaya fruits.

A recent substantial rearrangement of the 882 described eleutherodactyline frog species has considerably improved the understanding of their systematics. Nevertheless, many taxonomic aspects of the South American eleutherodactyline species remain unknown and require further investigation using morphological, cytogenetic and molecular approaches. In this work, the karyotypes of the Brazilian species Ischnocnema juipoca (Atibaia and Campos do Jordão, SP), Barycholos cf. ternetzi (Uberlândia, MG, and Porto Nacional, TO), and Pristimantis crepitans (Chapada dos Guimarães and São Vicente, MT) were analyzed using Giemsa staining, Ag-NOR labeling, and C-banding techniques. All individuals had a diploid number of 22 chromosomes, but the Fundamental Numbers were different among species. The herein described low chromosome number of Pristimantis crepitans is unique within this genus, suggesting that cytogenetically this species is not closely related either to its congeneric species or to Ischnocnema. In addition, karyotype differences, mainly in the NOR position, clearly distinguished the two Barycholos populations, besides indicating the existence of a so far undescribed species in this genus. A taxonomic review could clarify the systematic position of P. crepitans and verify the hypothetic new Barycholos species.

O número de sessões necessárias para tratar um canal radicular infectado é um dos assuntos mais controversos da endodontia. O objetivo deste estudo foi analisar, em cães, a resposta dos tecidos perirradiculares ao tratamento endodôntico de canais infectados em uma ou duas consultas, usando diferentes medicamentos entre as sessões. Lesões perirradiculares foram induzidas pela inoculação de Enterococcus faecalis nos canais. Após a confirmação do desenvolvimento de uma lesão perirradicular, os canais foram tratados em uma ou duas sessões, usando óleo ozonizado ou hidróxido de cálcio associado ao paramonoclorofenol canforado (PMCC) como medicação intracanal. Após 6 meses, os animais foram sacrificados e os espécimes processados para análise histológica e histobacteriológica. Os canais tratados em sessão única apresentaram uma taxa de sucesso de 46% dos casos. Quando a medicação usada entre as sessões foi o hidróxido de cálcio associado com o PMCC, 74% dos casos resultaram em sucesso. Nos casos em que o óleo ozonizado foi usado, uma taxa de sucesso de 77% foi observada. Esses achados demonstraram que o tratamento em duas sessões oferece uma taxa de sucesso mais alta quando comparado à terapia em uma sessão. Além disso, o óleo ozonizado mostrou potencial para ser usado como medicação intracanal.
The number of appointments necessary to treat infected root canals is one of the most controversial issues in endodontics. This study evaluated, in dogs, the response of the periradicular tissues to the endodontic treatment of infected root canals performed in a single visit or in two visits, using different interappointment dressings. Periradicular lesions were induced by inoculating Enterococcus faecalis in the root canals. After confirming that a periradicular lesion developed, the root canals were treated within one or two visits, using either ozonized oil or calcium hydroxide in camphorated paramonochlorophenol (CMCP) as an intracanal medication. After 6 months, the animals were sacrificed and the specimens were processed for histological and histobacteriological analysis. The root canals treated in a single visit showed a success rate of 46%. When a calcium hydroxide/CMCP-based interappointment intracanal medication was used, 74% of the cases were categorized as success. In cases where ozonized oil was used as the intracanal medication, a success rate of 77% was observed. These results of the present study demonstrated that the two-visit treatment offered a higher success rate compared to one-visit therapy. In addition, ozonized oil may potentially be used as an intracanal medication.

O objetivo do presente estudo foi avaliar as alterações promovidas, por intervenção multidisciplinar, nas concentrações plasmáticas de grelina e leptina, adiposidade visceral e prevalência de esteatose hepática não alcoólica (NAFLD), em adolescentes obesos. Foram avaliados 28 adolescentes obesos, 16 meninas (IMC 34,58 ± 3,86kg/m²) e 12 meninos (IMC 37,08 ± 3,17kg/m²), com idade entre 15 e 19 anos, quanto à concentração de leptina, grelina, insulina, assim como a adiposidade visceral e o diagnóstico de NAFLD pelo método de ultra-sonografia. Os resultados demonstraram redução significante na concentração circulante de grelina e leptina e na adiposidade visceral (p < 0,01). Houve ainda redução percentual na prevalência de NAFLD, sendo este um resultado relevante, visto que esta doença pode progredir para cirrose, tanto em crianças quanto em adolescentes obesos. Este tipo de tratamento demonstrou ser eficiente na melhora do perfil metabólico e hormonal, contribuindo para o controle da obesidade e suas co-morbidades em adolescentes obesos.
The aim of this study was to assess the changes promoted by a multidisciplinary therapy in ghrelin and leptin concentrations, visceral adiposity and non-alcoholic fat liver disease-NAFLD, in obese adolescents. A total of 28 obese adolescents, 16 girls (BMI 34.58 ± 3,86 wt/ht²) and 12 boys (BMI 37.08 ± 3.17 wt/ht²), aged between 15 and 19 years old, was evaluated to leptin, ghrelin and insulin concentrations, visceral adiposity and NAFLD through ultrasonography. The results showed a significant decrease in ghrelin, leptin concentrations and visceral adiposity (p < 0.01). Moreover, a decrease in the NAFLD prevalence was observed. It is an important result, since this disease can progress to cirrhosis, not only in children but also in obese adolescents. This kind of treatment can be efficient to improve metabolic and hormonal profile, as well as, to control obesity and related co-morbidities in obese adolescents.
El objetivo del presente trabajo ha sido evaluar las alteraciones promovidas por la intervención multidisciplinar, en las concentra- ciones plasmáticas de grelina y leptina, adiposidad visceral y prevalencia de esteatosis hepática no alcohólica - NAFLD, en adolescentes obesos. 28 adolescentes obesos, 16 chicas (IMC 34,58 ± 3,86 kg/m²) y 12 chicos (IMC 37,08 ± 3,17 kg/m²), con edades entre 15 y 19 años, fueron evaluados respecto a la concentración de leptina, grelina, insulina, así como a la adiposidad visceral y el diagnóstico de NAFLD por el método de ultrasonografía. Los resultados demostraron una reducción significante en la concentra- ción circulante de grelina y leptina y en la adiposidad visceral (p < 0,01). Hubo aún una reducción porcentual en la prevalencia de NAFLD, siendo este un resultado relevante, ya que esta enfermedad puede progresar hasta la cirrosis, tanto en niños como en adolescentes obesos. Este tipo de tratamiento demostró ser eficiente en la mejora del perfil metabólico y hormonal, contribuyendo para el control de la obesidad y su comorbilidad en adolescentes obesos.

Micetoma por Actinomadura madurae: relato de dois casos

São relatados dois casos de micetoma por Actinomadura madurae, atendidos no Hospital Universitário Clementino Fraga Filho (UFRJ), em 1990, e no Hospital Universitário Antonio Pedro (UFF), em 1984. Caso 1: paciente masculino, pardo, de 27 anos, iniciou o quadro em 1988, após traumatismo no pé esquerdo, com aumento de volume com nódulos apresentando fistulas drenando secreção e grãos branco-amarelados. A radiografia mostrou lesões líticas nos ossos do tarso e 2º e 3º metatarsianos do pé esquerdo. O exame histopatológico evidenciou grãos basofílicos recobertos por franja eosinofílica, arredondados, medindo até 1mm. Ao exame micológico foi isolado em cultivo A. madurae. Diante do fracasso de outras tentativas, foi instituida terapêutica com tetraciclina oral por 6 meses; contudo, como não houve resolução do quadro clínico-radiológico, foi indicada a amputação do membro. Caso 2: paciente masculino, branco, de 70 anos, iniciou quadro em 1974, após traumatismo no pé direito, que evoluiu com aumento de volume e fístulas. Ao exame histopatológico, grãos basofílicos com franjas eosinofílicas ao redor, tamanho grande, de 1 a 2mm de diâmetro, características de A. madurae. Discreta melhora com tetraciclina e sulfamídicos. Sem controle posterior. O primeiro caso adquiriu a infecção no Rio de Janeiro e o segundo caso a adquiriu em Pernambuco. Além de aspectos clínicos, diagnósticos e terapêuticos, é comentada a ocorrência dos micetomas nas Américas, destacando a freqüência dos actinomicetomas por A. madurae.
Actinomadura madurae mycetoma was diagnosed in two patients by the dermatologic outpatient clinic of the Clementino Fraga Filho University Hospital (UFRJ) and the Antonio Pedro University Hospital (UFF). The first case was a 27-year-old negro male from the out-skirsts of Rio de Janeiro, who was injured prior to the onset of the disease in 1988. The affected left foot showed swelling, nodules, sinus tracts, purulent discharge containing grains, and serious bone involvement with lytic lesions. Histo-logical examination showed deeply basophylic stained grains with typical widefringed borders. The mycological examination revealed an actinomycete identified as A. madurae by culture. Oral tetracycline for six months did not improve the clinical-radiological picture and the patient was submitted to amputation. The second case was a 70-year-old white male, with previous injury en 1974, which occurred in Pernambuco State. Enlargement of the right foot draining sinuses formation with discharge of pus and grains. In tissue sections the grains were large and surrounded by amorphous eos-inophylic clubs radially oriented. There was discrete improvement by tetracycline and sulfonamide. No follow-up.
