ABSTRACT Nitric oxide application has been seen as a promising technique to extend the postharvest life of various fresh fruits and vegetables. This is the first work involving the spray application of nitric oxide donor S-nitrosoglutathione (GSNO) on Golden papaya. Considering that results are very distinct depending on the type of the nitric oxide donor, the form of application, the concentration used and the species studied, the application must be adapted to each necessity. The aim of this study was to relate the application of GSNO, spray applied at 10, 100, 1,000 ?M, with the physiological, physical-chemical, and biochemical changes of Golden papaya, in the first 72 h of ripening. Control fruit was sprayed with distilled water. GSNO application did not interfere on color and chlorophyll fluorescence of the peel, on soluble solids, titratable acidity, lipid peroxidation, and in the level of S-nitrosothiols. Control fruit and 10-?M GSNO sprayed showed lower respiration. After 72 h of ripening at 25°C, all fruits showed an increase in ethylene biosynthesis, except for those treated with 10 ?M GSNO. Papaya sprayed with 10-?M GSNO showed the highest pulp firmness and 52% less weight loss when compared to control fruit. GSNO was also responsible for increasing the ascorbic acid in papayas, besides showing an increase in total antioxidant activity production. The results indicated that the application of 10 ?M of GSNO by spray can potentially preserve the quality characteristics of Golden papaya, mainly due to the lower ethylene production, the delay in the firmness loss, and the less weight loss.

ABSTRACT The root system is essential for sugarcane regrowth and the vigor of ratoon cycles as it represents the unique source of carbon skeletons and energy for the initial plant development. However, root system dynamics after shoot harvesting and its role in sugarcane regrowth remains poorly known. Here, it was hypothesized that sugarcane plants with small volume of root system will accumulate less biomass after shoot harvesting than plants with larger volume and that such regrowth is dependent on root reserves. In sugarcane plants grown in nutrient solution, shoots were cut, and two root treatments were established: reference plants with the entire root system (100%); and plants with half of the root system (50%), randomly removing half of root system. After 37 days of shoot harvesting, plants with the entire root system showed higher shoot, root and total dry mass, root length, root diameter, root area and root volume, when compared with those with 50% of the root system. Sugarcane plants with the entire root system had higher root content of starch, soluble sugar and nonstructural carbohydrates as compared to plants with 50% of the root system. A significant positive correlation was found between the variation of shoot dry mass and the variation of root nonstructural carbohydrates. Interestingly, this data revealed a disproportionate effect of root system size on sugarcane regrowth, with plants with the entire root system accumulating almost three times more biomass than plants with half of the root system during regrowth.

ResumoUm importante índice para a indústria sucroalcooleira é a produtividade de sacarose em cana-de-açúcar, que pode ser relacionada à capacidade da planta em acumular sacarose durante a maturação. Apesar de se conhecer a eficiência de tratamentos como a restrição hídrica e a aplicação de reguladores para estimular o acúmulo de sacarose nos colmos, pouco se conhece sobre as respostas fisiológicas da planta que levam à maturação. Nesse contexto, o objetivo deste estudo foi avaliar as respostas fisiológicas da cana-de-açúcar a diferentes tratamentos indutores de maturação e, assim, compreender melhor a fisiologia desse processo. Duas variedades, a IACSP95-5000, de alta produtividade, e a IACSP94-2094, de produtividade moderada, foram submetidas ao déficit hídrico, à aplicação de regulador químico (etefom 480 g ha–1) e aos dois tratamentos associados. Foram medidos o crescimento, a concentração de carboidratos em folhas e colmos e as trocas gasosas. Concluiu-se que o efeito do etefom em cana-de-açúcar é genótipo-dependente, estimulando o acúmulo de sacarose no colmo e o suprimento de fotoassimilados pela fonte na variedade responsiva (IACSP95-5000). Tais efeitos não são associados à restrição do crescimento. Em relação à aplicação dos tratamentos de déficit hídrico e etefom associados, a variedade responsiva apresenta aumento no teor de sacarose no colmo nos mesmos níveis apresentados quando apenas etefom é aplicado, de modo que os tratamentos não apresentam efeitos aditivos na maturação da cana-de-açúcar.

AbstractAn important index for the ethanol industry is the sucrose yield in sugarcane, which is affected by plant ability to accumulate sucrose during ripening. Despite the known efficiency of treatments such as water restriction and the application of regulators to stimulate the sucrose storage in culms, little is known about the physiological responses of the plant that lead to ripening. In this context, the aim of this study was to evaluate the physiological responses of sugarcane to different ripening treatments. Two varieties, IACSP95-5000, with high yield, and IACSP94-2094, with moderate yield were subjected to water deficit or application of chemical regulator (ethephon 480 g ha–1) and both treatments associated. Growth, accumulation of carbohydrates in leaves and culms were measured. It can be concluded that the effects of ethephon on sugarcane are genotype-dependent. Ethephon stimulates sucrose accumulation in the culm and the photosynthate supply by the source in the responsive variety (IACSP95-5000). Such effects are not associated with growth restriction. In relation to the drought combined with the application of ethephon, the responsive variety shows increased sucrose content in culm at the same level as when ethephon is applied alone, hence treatments have no additive effects on sugarcane ripening.

Neste trabalho, avaliou-se o crescimento e a partição de carboidratos em plantas jovens de Sebastiania membranifolia em resposta ao alagamento. Sementes foram germinadas em câmara tipo BOD a 25 °C, sobre papel germitest umedecido. As plântulas foram aclimatadas durante 20 dias em sala de crescimento e, em seguida, transplantadas para sacos de polipropileno, contendo substrato constituído de areia, esterco bovino e solo (1:1:1), e transferidas para casa de vegetação coberta com sombrite 50% de sombreamento, onde permaneceram durante 40 dias. Ao final desse período, foram estabelecidos três tratamentos: plantas não alagadas, com as raízes alagadas e totalmente alagadas. O alagamento foi realizado, colocando os sacos contendo as plantas, individualmente, em baldes com maior capacidade. As avaliações ocorreram com intervalo de 12 dias por um período total de 36 dias, sendo utilizadas quatro repetições para cada tratamento e período de avaliação. O número de folhas, ramificações, altura do caule e massa seca das partes, aumentou linearmente nas plantas não alagadas ao longo do período analisado. Nas plantas com as raízes alagadas, o mesmo incremento ocorreu somente até o 24º dia e, para as totalmente alagadas, de maneira geral, não houve crescimento significativo. Os teores de açúcares solúveis totais, redutores e amido foram significativamente inferiores nas plantas totalmente alagadas, tanto nas folhas quanto nas raízes, sendo que nas raízes também foi observada significativa redução nos teores de amido para as plantas com as raízes alagadas, igualando-se, na ultima avaliação, aos valores das plantas totalmente alagadas. Esses resultados sugerem que S. membranifolia pode ser uma espécie promissora para a revegetação de áreas de depleção sujeitas à inundação intermitente.

The aim of this study was to evaluate the growth and carbohydrate partitioning in seedlings of Sebastiania membranifolia in response to flooding. Seeds were germinated in BOD type chamber at 25 ºC, on moistened germitest paper. The seedlings were acclimatized for 20 days in a growth chamber and then transplanted into polypropylene bags containing substrate consisting of sand , manure and soil (1:1:1), and transferred to a greenhouse protected with 50% shading for 40 days. At the end of this period three treatments were established: control (well watered), root flooded and completely flooded. The flooding was accomplished by placing the bags containing the plants individually in buckets with greater capacity. The evaluations were conducted at intervals of 12 days for a total period of 36 days, with four replicates for each treatment and evaluation period. The number of leaves, branches, stem height and dry weight of these parts, increased linearly in control plants over the period analyzed. For the flooded roots treatment, these parameters increased only until the 24th day, and in completely flooded plants, in general, this stress did not allow the growth of plants. The content of total soluble sugars, reducing sugars and starch were significantly lower in completely flooded plants, both in the leaves and in the roots. Root flooded plants had significant starch reduction in the roots, with values of the last measurement equal to those observed in completely flooded plants. These results suggest that S. membranifolia may be a promising species for revegetation of depleted areas subject to intermittent flooding.

Sebastiania membranifolia, popularmente conhecida como sarandi, é uma espécie arbórea nativa do Brasil, geralmente encontrada em terrenos de várzeas aluviais e beira de rios, indicada para reflorestamentos destinados à recuperação de áreas degradadas. Apesar da importância comercial dessa espécie, pouco se sabe sobre os seus mecanismos de propagação. Nesse contexto, objetivou-se avaliar o comportamento germinativo das sementes em diferentes condições de temperatura, substrato e luz, e determinar métodos de superação de dormência, de forma a estabelecer um protocolo adequado para propagação sexuada. Este estudo foi conduzido em câmaras BOD com controle térmico e de fotoperíodo, testando os seguintes tratamentos: temperaturas de 25; 30 e alternada 20/30ºC; substratos (areia-EA; sobre papel-SP e entre papel-EP); quatro soluções de giberelina (50; 100; 200; 400 mgL-1 ); nitrato de potássio (0,1; 0,2; 0,3 e 0,4%), na presença (12 h de fotoperíodo) e ausência de luz. O delineamento empregado foi em DIC com quatro repetições de 50 sementes cada. Observamos que o grau médio de umidade das sementes foi de 10,4% e o peso de mil sementes (PMS) de 3,16 g. O maior percentual de germinação foi obtido com fotoperíodo, a 25ºC, sobre papel (SP) e entre papel (EP). As concentrações de 100 mg L-1 de giberelina e 0,1% de nitrato de potássio proporcionaram maior germinação e vigor. O tratamento das sementes com solução de giberelina a 100 mgL-1 favoreceu o crescimento das plântulas, ao contrário do nitrato de potássio que inibiu o crescimento mesmo em baixa concentração.

Sebastiania membranifolia, popularly known as sarandi, is a tree species native to Brazil, usually found in land of alluvial floodplains and river banks, suitable for restoration of degraded areas. Despite the commercial importance of this species, little is known about its mechanisms of propagation. This research evaluated the germinating seeds under different conditions of temperature, substrate and light, and determined methods of scarification in order to establish a suitable protocol for sexual propagation. The study was conducted in chambers with BOD and thermal control photoperiod, testing the following treatments: temperatures of 25, 30 and alternating 20/30º C; substrates (sand-EA, on paper-SP and from paper -EP), four solutions gibberellin (50, 100, 200, 400 mgL-1); potassium nitrate (0.1, 0.2, 0.3 and 0.4%) in the presence (12 h photoperiod) and absence of light. The experimental design was completely randomized with four replications of 50 seeds each. We found that the average degree of seed moisture was 10.4% and the weight of a thousand seeds (PMS) of 3.16 g. The highest percentage of germination was obtained with photoperiod at 25 º C, on paper (SP) and between role (EP). The concentrations of 100 mg L-1 gibberellin and 0.1% potassium nitrate showed higher germination and vigor. Seed treatment with a gibberellin solution of 100 mgL-1 favored the growth of seedlings, unlike potassium nitrate that inhibit the growth even at low concentration.

Para o cafeeiro (Coffea arabica) existe uma comprovada relação positiva entre fechamento estomático e formação e acúmulo de H2O2. Entretanto, tal relação para a cultura sob restrição hídrica ainda não foi estudada. Avaliou-se o movimento estomático e a capacidade antioxidante em mudas de cafeeiro sob diferentes regimes hídricos. Mudas de cafeeiro cv. Catuaí IAC 99, com oito meses de idade, foram submetidas à capacidade de campo, suspensão gradativa e suspensão total da irrigação por um período de 21 dias. Foram realizadas avaliações do potencial hídrico (Ψw) foliar na antemanhã e resistência estomática, taxa transpiratória e déficit de pressão de vapor foram avaliados as 10h00 e 17h00. As determinações bioquímicas e enzimáticas foram realizadas em folhas coletadas às 17h00. Todas as avaliações e coletas foram realizadas em intervalos de três dias. Nas plantas em capacidade de campo não houve variação no Ψw durante o período de avaliação. Para a suspensão gradativa da irrigação, houve queda expressiva a partir dos 12 dias, chegando próximo a -2,5 Mpa, ao final do experimento. Já nas plantas em suspensão total da irrigação observou-se queda no Ψw a partir do sexto dia, chegando a -2,5 MPa aos 15 dias. A queda no Ψw para as plantas em suspensão gradual e total da irrigação refletiu em aumentos na resistência estomática e diminuição da taxa transpiratória, ocasionando aumento na formação de peróxido de hidrogênio e nos períodos finais, aumentos na peroxidação de lipídios. Em conseqüência obervaram-se aumentos na atividade das enzimas antioxidantes, bem como nos teores de ascorbato e dehidroascorbato, atuando na detoxificação dos radicais livres formados em função do estresse.

Coffee (Coffea arabica) plants show a positive relationship between stomatal closure and formation and accumulation of H2O2. However, for coffee plants under water restriction such relationship has never been studied. The objective of the present study was evaluate the stomatal movement and the antioxidant capacity of coffee seedlings under different water regimes. Eight months old coffee seedlings of cv. Catuaí IAC 99 were submitted to field capacity, gradual and total suspension of irrigation during a period of 21 days. Evaluations of leaf water potential (Ψw) were performed in the beginning of the morning, and stomatal resistance, transpiration rate and vapor pressure deficit were determined at 10 am and 5 pm. All biochemical and enzymatic determinations were performed in leaves collected at 5 pm. Evaluations and samplings were performed at three days intervals. There was no variation in Ψw during the evaluated period for plants in field capacity. However, an expressive decrease of Ψw following day 12, reaching values near -2.5 MPa at the end of the experiment was observed for plants submitted to gradual suspension of irrigation. For plants submitted to total suspension of irrigation, Ψw decreases after the sixth day, reaching -2.5 MPa at day 15. The decay of Ψw in plants submitted to gradual and total suspension of irrigation reflected in increased stomatal resistance and in a decreased transpiration rate leading to an increase in hydrogen peroxide formation and, on final stages, increase in lipid peroxidation. As a conclusion, an increase in the activity of antioxidant enzymes as well as in the levels of ascorbate and dehydroascorbate was observed, which act in the detoxification of free radicals formed as result of the water stress.

O pinhão-manso (Jatropha curcas L.) vem ganhando importância por seu potencial como insumo ao biodiesel. Estudos visam à adequação agronômica e a cultura de tecidos contribui indiretamente com este aspecto, auxiliando em pesquisas aplicadas como em biotecnologia, em situações que visam à regeneração de plantas. Nesse intuito, é fundamental avaliar, durante os períodos de cultivo dos calos, o comportamento dos metabólitos primários. A massa calogênica, por ocasião da inoculação, não apresentou reserva na forma de carboidratos. Aos 42 dias iniciou a absorção da sacarose do meio, enquanto o acúmulo no explante ocorreu a partir dos 98 dias de cultivo. O teor de aminoácidos foi alto no dia da inoculação e, por volta dos 98 dias, tanto os níveis de aminoácidos quanto os de proteínas decresceram.

The physic nut (Jatropha curcas L.) is gaining importance because of its potential as raw material for biodiesel. Studies aiming the adequacy of agronomic techniques and tissue culture contribute, indirectly, to this aspect, helping applied research as biotechnology, in situations that aim plant regeneration. Therefore, it is essential to evaluate, the behavior of primary metabolites during the callus growing season. The callus mass at the inoculation time showed no carbohydrates reserve. At the 42nd day the sucrose absorption started in the medium, while the accumulation in the explant occurred from the 98th day of cultivation. Amino acid content was high at the inoculation's day and around the 98th day, both amino acids and protein levels decreased.

Plantas em seu ambiente natural estão sujeitas a condições adversas, passando por períodos de estresse breves ou duradouros. Algumas espécies desenvolvem mecanismos que as fazem suportar as condições adversas, permitindo sua sobrevivência. O objetivo deste estudo foi avaliar as respostas fisiológicas e anatômicas no desenvolvimento inicial de plantas de sibipiruna submetidas à hipoxia. Plantas com três a quatro folhas completamente expandidas foram submetidas a três tratamentos: plantas sem alagamento, alagadas nas raízes e totalmente alagadas. Foram realizadas avaliações dos teores de amido, açúcares solúveis totais e redutores em folhas e raízes coletadas a cada cinco dias, durante 30 dias. Características anatômicas foram avaliadas 15 dias após o início de cada tratamento. A maior redução nos teores de amido nas folhas e raízes foi observada nas plantas totalmente alagadas. Para açúcares solúveis totais e redutores, foi observado comportamento semelhante ao obtido em amido; porém, as plantas alagadas nas raízes apresentaram, em alguns períodos de avaliação, teores, principalmente nas raízes, superiores aos observados nas plantas sem alagamento. Essas plantas também apresentaram maior densidade estomática em relação aos demais tratamentos. Para a espessura do córtex e do cilindro vascular das raízes, foi observada uma redução para os tratamentos alagados, cujas plantas totalmente alagadas apresentaram uma maior barreira apoplástica imposta pelo espessamento em fi caracterizada pela formação de septos nas células próximas à endoderme.

Plants in their natural environment are subject to adverse conditions, and the stress periods can be short or long. Some species develop mechanisms that make them bear the adverse conditions, allowing their survival. The aim of this study was to evaluate the anatomic and physiological responses in the initial development of Sibipiruna plants submitted to hypoxia. Plants with three to four leaves fully expanded were subjected to three treatments: plants without flooding, flooded on roots and totally flooded. Evaluations were carried out in the starch, total soluble sugars and reducing sugars levels in the leaves and roots collected every five days, for 30 days. Anatomical characteristics were evaluated 15 days after the beginning of each treatment. The largest reduction in starch level in leaves and in roots was observed in plants completely flooded. For total soluble sugars and reducing sugars were observed behavior similar to that achieved in starch However, the plants which had flooded roots, in some periods of assessment, had levels, mainly in the roots, higher than the plants without flooding. These plants also showed higher stomatal density than the other treatments. For the thickness of the cortex and the cylinder vascular roots, a reduction was observed for the flooded treatment, which completely flooded plants had a higher apoplastic barrier imposed by thickening in phi, which is characterized by the formation of septa in cells near the endodermis.

A soja é um dos produtos agrícolas de maior importância no Brasil, e segundo maior produtor mundial. A tolerância ao excesso de umidade do solo e da rápida recuperação com o término do alagamento são características desta cultura, porém, a deficiência de oxigênio, causada pelo excesso de água, é um dos fatores determinantes que pode reduzir sua produtividade. O objetivo deste trabalho foi verificar o comportamento de três cultivares de soja ao alagamento em diferentes estádios de desenvolvimento, por meio de análise da produção de massa seca, metabolismo de carboidratos e atividade das enzimas do metabolismo anaeróbio. O experimento foi desenvolvido na Universidade Federal de Lavras (MG), em 2007. As cultivares BRS 267, BRS 257 e BRS 213 foram submetidas a 15 dias de alagamento, nos estádios V6 e V8 e 11 dias no estádio R4. Os resultados revelaram redução na produção de massa seca nos tratamentos alagados no estádio V8 para a cultivar BRS 267 e no estádio V6 para BRS 257 e BRS 213; esta última foi a mais tolerante ao alagamento, pois obteve o menor decréscimo de massa seca em relação ao controle. O alagamento também induziu aumento nos teores de amido e açúcares solúveis totais nas folhas, assim como maior atividade das enzimas álcool desidrogenase e piruvato descarboxilase para as três cultivares, quando comparadas a seus respectivos controles. Já para a enzima lactato desidrogenase, essa diferença foi menos expressiva. Esses resultados revelam comportamentos distintos comparando as três cultivares.

The soybean is one of the most important agricultural products in Brazil, the second in world production. The soil high tolerance to excess of moisture and the rapid recovery after flooding are important characteristics of this culture. However, the oxygen, deficiency resulting from excess of water is one factor that may cause reduction in soybean productivity. This study aimed to evaluate the behavior of three soybeans cultivars upon flooding at various stages of development, based on dry matter production, carbohydrate metabolism and some anaerobic enzymes activities. The experiment was conducted at the Federal University of Lavras, Brazil, in 2007. Cultivars BRS 267, BRS 257 and BRS 213 were submitted to 15 days of flooding, for stages V6 and V8 and 11 days for stage R4. Results indicated a reduction in the production of dry matter for the treatments flooded BRS 267 in the stage V8 and for varieties BRS 257 and BRS 213, V6 stage the latter being the most tolerant to flooding, once exhibited a lower reduction in dry matter compaired to control. The flooding also lead to an increase of total soluble sugars and starch levels in the leaves, as well as to an increase in the activity of the enzymes alcohol dehydrogenase and pyruvate decarboxylase, in all three evaluated cultivars. Nom significant differences were observed for the lactate dehydrogenase enzyme activity. These results indicate a different behavior when comparing the three cultivars.
