Abstract This work describes the circulation over the continental slope and the São Paulo Plateau in the Santos Basin during the SANSED winter 2019 survey. The cruise consisted of four legs in the period between June, 11 and August, 03 2019. The observed circulation is dominated by the Atlantic southwestern boundary current regime and remotely-generated anticyclones and cyclones. The former is composed by the Brazil Current, the Intermediate Western Boundary Current and their mesoscale meanders; the latter are 300km vortical rings with origin in the eastern side of the South Atlantic Basin. A Lagrangian scheme applied over satellite altimeter maps indicate that the origin of these rings is primarily the Cape Basin of South Africa. The interaction between the boundary currents, their cyclonic meanders, and the anticyclonic rings is complex, and varies widely. During the SANSED winter 2019 survey period, three anticyclones interacted with the Brazil Current, instabilizing it, forming dipoles with the current cyclonic meanders, leading to their downstream propagation. Ancienter cyclonic eddies within Santos Basin may interfere with the propagation of the large anticyclones further south. In addition, the continuous arrival of remotely-originated anticyclones, the larger portion over the São Paulo Plateau presented a tendency of counter-clockwise circulation during the whole cruise period.

Abstract Background Major advances have been seen in techniques and devices for performing percutaneous coronary interventions (PCIs) for chronic total occlusions (CTOs), but there are limited real-world practice data from developing countries. Objectives To report clinical and angiographic characteristics, procedural aspects, and clinical outcomes of CTO PCI performed at dedicated centers in Brazil. Methods Included patients underwent CTO PCI at centers participating in the LATAM CTO Registry, a Latin American multicenter registry dedicated to prospective collection of these data. Inclusion criteria were procedures performed in Brazil, age 18 years or over, and presence of CTO with PCI attempt. CTO was defined as a 100% lesion in an epicardial coronary artery, known or estimated to have lasted at least 3 months. Results Data on 1196 CTO PCIs were included. Procedures were performed primarily for angina control (85%) and/or treatment of moderate/severe ischemia (24%). Technical success rate was 84%, being achieved with antegrade wire approaches in 81% of procedures, antegrade dissection and re-entry in 9%, and retrograde approaches in 10%. In-hospital adverse cardiovascular events occurred in 2.3% of cases, with a mortality rate of 0.75%. Conclusions CTOs can be treated effectively in Brazil by using PCI, with low complication rates. The scientific and technological development observed in this area in the past decade is reflected in the clinical practice of dedicated Brazilian centers.
Resumo Fundamento Tem sido observado um grande avanço nas técnicas e nos dispositivos para a realização de intervenções coronárias percutâneas (ICP) em oclusões totais coronarianas crônicas (OTC), mas existem poucos dados da prática do mundo real em países em desenvolvimento. Objetivos Relatar as características clínicas e angiográficas, os aspectos dos procedimentos e os resultados clínicos da ICP de OTC em centros dedicados a esse procedimento no Brasil. Métodos Os pacientes incluídos foram submetidos à ICP de OTC em centros participantes do LATAM CTO Registry, um registro multicêntrico latino-americano dedicado à coleta prospectiva desses dados. Os critérios de inclusão foram procedimentos realizados no Brasil, idade acima de 18 anos e presença de OTC com tentativa de ICP. A definição de OTC foi lesão de 100% em uma artéria coronária epicárdica, conhecida ou estimada como tendo pelo menos 3 meses de evolução. Resultados Foram incluídos dados de 1.196 ICPs de OTC. Os procedimentos foram realizados principalmente para controle da angina (85%) e/ou tratamento de uma grande área isquêmica (24%). A taxa de sucesso técnico foi de 84% e foi alcançada com técnicas de fios anterógrados em 81%, dissecção/reentrada anterógrada em 9% e retrógrada em 10% dos procedimentos. Os eventos cardiovasculares adversos intra-hospitalares ocorreram em 2,3% dos casos, sendo a mortalidade de 0,75%. Conclusões As OTC podem ser tratadas no Brasil por intervenção coronária percutânea de forma efetiva e com baixas taxas de complicações. O desenvolvimento científico e tecnológico observado nessa área na última década reflete-se na prática clínica de centros brasileiros dedicados a essa técnica.

Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development.

RESUMO Neste trabalho, foi realizada a previsão da distribuição de M. urundeuva Fr. All., com base na região de ocorrência natural da espécie. Suas coordenadas geográficas foram obtidas nas bases de dados online CRIA e SpecialLinks, a partir de artigos científicos e trabalhos de campo realizados pela Universidade Estadual Paulista (UNESP) em Ilha Solteira, São Paulo, Brasil. M. urundeuva é uma espécie arbórea nativa com grande potencial para uso comercial no Brasil. Para tanto, foi utilizada a modelagem de nicho ecológico, com camadas atuais de variáveis climáticas e camadas preparadas para cenários climáticos futuros, de acordo com o 4º Relatório do Painel Intergovernamental sobre Mudanças Climáticas (AR4/IPCC), utilizando dados do Worldclim no Brasil. Com os programas Open Modeller e ArcGIS, foram gerados mapas para prever sua ocorrência para o período atual e para cenários climáticos futuros, feitos de acordo com as projeções de mudanças climáticas globais. Com a projeção de aumentos de temperatura e precipitação na região onde ocorre a espécie, ela tende a migrar para áreas do Brasil onde o clima é atualmente mais ameno, nas regiões Sul e Sudeste. Devido às mudanças climáticas, a espécie tende a sofrer mudanças na distribuição e tamanho de área até 2080. Foi projetado para a Caatinga e Pantanal, nos dois períodos, um aumento de área, enquanto para o Cerrado, no primeiro período, a área aumentou e para o segundo, diminuiu. Portanto, de acordo com os resultados dos mapas de projeções futuras para as próximas décadas, conclui-se que haverá mudanças na distribuição de M. urundeuva, com redução significativa da área potencial de ocorrência na região.
ABSTRACT In this work, the prediction of the distribution of M. urundeuva Fr. All. was performed based on the region of natural occurrence of the species. Its geographic coordinates were obtained from online databases CRIA and SpecialLinks, from scientific articles and fieldwork carried out by Universidade Estadual Paulista (UNESP) in Ilha Solteira, São Paulo, Brazil. M. urundeuva is a native tree species with great potential for commercial use in Brazil. For this purpose, ecological niche modeling was used, with current layers of climate variables and layers prepared for future climate scenarios, according to the 4th Report of the Intergovernmental Panel on Climate Change (AR4/IPCC), using Worldclim data on Brazil. With the Open Modeller and ArcGIS programs, maps were generated to predict its occurrence for the current period and future climate scenarios, made according to the projections of global climate changes. With the projection of increases in temperature and precipitation in the area where the species occurs, it tends to migrate to areas of Brazil where the climate is currently milder, in the south and southeast regions. Due to climatic changes, the species tends to undergo changes in distribution and area size until 2080. It was projected for Caatinga and Pantanal, in both periods, an increase in area, while for the Cerrado, in the first period, the area increased, and, for the second, it decreased. Therefore, according to the results of the maps of future projections for the next decades, it is concluded that there will be changes in the distribution of M. urundeuva, with a significant reduction of the potential area of occurrence in the region.

Initial performance of 'Rebo' wine grapes on different rootstocks and at various planting densities

Resumo O objetivo deste trabalho foi avaliar a influência de diferentes porta-enxertos e densidades de plantio sobre o desempenho inicial da produtividade e da qualidade de uvas da cultivar Rebo, cultivada em região de alta altitude do estado de Santa Catarina, Brasil. Realizou-se um delineamento experimental de blocos ao acaso, em arranjo fatorial 5×3. Os tratamentos consistiram da combinação de cinco porta-enxertos ('101-14 Mgt', 'IAC 572', 'Paulsen 1103', 'Harmony' e 'VR 043-43') com a uva Rebo e de três espaçamentos entre plantas (1,0, 1,2 e 1,5 m). As avaliações ocorreram durante as safras 2018/2019 e 2019/2020. Avaliaram-se as variáveis produtivas iniciais e as características físico-químicas das uvas. O porta-enxerto '101-14 Mgt' apresentou a brotação mais precoce, e os porta-enxertos 'VR043-43' e 'IAC 572', as mais tardias. Os porta-enxertos '101-14 Mgt' e 'VR043-43' proporcionaram maior produtividade. Os espaçamentos de 1,0 e 1,2 m entre plantas aumentaram a produtividade inicial da vinha. Os porta-enxertos '101-14 Mgt' e 'Harmony' contribuíram para diminuir a acidez total do mosto. O desempenho inicial da produtividade e da qualidade da uva vinífera 'Rebo' é favorecido pelo porta-enxerto '101-14 Mgt' às densidades de plantio de 1,0 a 1,2 m.
Abstract The objective of this work was to evaluate the influence of different rootstocks and planting densities on the initial performance of the yield and quality of the Rebo grapes cultivars grown in a high altitude region of the state of Santa Catarina, Brazil. An experimental design with randomized complete blocks was carried out in a 5×3 factorial arrangement. The treatments consisted of the combination of five rootstocks ('101-14 Mgt', 'IAC 572', 'Paulsen 1103', 'Harmony', and 'VR 043-43') with the 'Rebo' grape, and of three spacing between plants (1.0, 1.2, and 1.5 m). The evaluations took place during the 2018/2019 and 2019/2020 crop seasons. The initial yield variables and the physicochemical characteristics of the grapes were evaluated. The '101-14 Mgt' rootstock provided early shoots, and the 'VR043-43' and 'IAC 572' rootstocks the latest shoots. The '101-14 Mgt' and 'VR043-43' rootstocks provided a higher yield. Plant spacing at 1.0 and 1.2 m increased the initial yield of the vineyard. The '101-14 Mgt' and 'Harmony' rootstocks contributed to the lower total acidity of must. The initial performance of the yield and quality of 'Rebo' wine grape is favored by the '101-14 Mgt' rootstock at the planting densities of 1.0 to 1.2 m.

A vertical-mode extrapolation scheme applied to the Brazil Current domain: quasi-synoptic scenarios

Abstract The Brazil Current (BC) is a western boundary current, with an average vertical extension of 500 m, which occupies the southwestern margin of Atlantic Ocean. Recently, the South Brazil Bight - SBB (23º S - 28º S) has been intensively explored by the oil and gas industries, which brought a high demand in operational oceanography and ocean state monitoring. In this study we present a new technique based on the vertical dynamic modes of oscillation, to extrapolate velocity profiles in the SBB, with potential applications to data acquired by Vessel Mounted Acoustic Doppler Current Profilers (VMADCP). Using four quasi-synoptic crosssectional velocity data, acquired by Lowered Acoustic Doppler Current Profilers (LADCP), we observed that the mean circulation of this region can be characterized as a 2-dynamical mode system. This mean circulation is mainly dominated at surface by the BC and at intermediate levels by the Intermediate Western Boundary Current (IWBC), which flows northward. Furthermore, observed mesoscale vortical features in the SBB, such as eddies and meanders, are shown to be primarily barotropic-mode dominated. In our extrapolation methodology, the use of an independent set of velocity time series obtained by four moorings has shown to be a valid way for obtaining the relative contribution of each dynamical mode in the velocity field in the SBB. Normalized mean squared errors below 10% and radar diagrams show that the presented methodology was able to reproduce the main dynamic features observed by LADCP transects. Also, with this technique, we extrapolated three VMADCP sections and the results agreed with previous observations.

Treatment compliance and risk and protective factors for suicide ideation to completed suicide in adolescents: a systematic review

Objective: To review the literature regarding adolescent suicide risk and explore the associations between treatment compliance (expressed as a concept including measured adherence to treatment and/or mental health service utilization) and risk and protective factors for suicidal behavior (SB), as well as the association between treatment compliance and reattempts. Methods: PubMed, LILACS, and Google Scholar were searched using the following terms: (adolescent*) AND (suicide*) AND (risk factor OR protective factors) AND (treatment compliance OR treatment attrition OR treatment adherence OR treatment drop out OR treatment retention OR mental health utilization). We retrieved studies that focused on the relation of treatment compliance to risk and protective factors for SB and that had only adolescent samples. Results: Of 4,841 articles, 30 original articles were selected for review. Most studies indicated high mental health service (MHS) utilization and poor treatment adherence by SB patients. Social minority status and conduct disorder were associated with less treatment adherence, while female sex, parental perceived need for treatment, and major depression were associated with greater treatment adherence. Inpatient and intensive emergency care after SA and family interventions improved MHS utilization and treatment compliance. However, we found no substantial protective effect of treatment compliance against reattempts. Conclusion: Effective treatment planning for compliance requires considering psychopathology, treatment planning, and social, familial, and individual factors.

RESUMO Objetivo: Descrever as práticas de ressuscitação volêmica em unidades de terapia intensiva brasileiras e compará-las com as de outros países participantes do estudo Fluid-TRIPS. Métodos: Este foi um estudo observacional transversal, prospectivo e internacional, de uma amostra de conveniência de unidades de terapia intensiva de 27 países (inclusive o Brasil), com utilização da base de dados Fluid-TRIPS compilada em 2014. Descrevemos os padrões de ressuscitação volêmica utilizados no Brasil em comparação com os de outros países e identificamos os fatores associados com a escolha dos fluidos. Resultados: No dia do estudo, foram incluídos 3.214 pacientes do Brasil e 3.493 pacientes de outros países, dos quais, respectivamente, 16,1% e 26,8% (p < 0,001) receberam fluidos. A principal indicação para ressuscitação volêmica foi comprometimento da perfusão e/ou baixo débito cardíaco (Brasil 71,7% versus outros países 56,4%; p < 0,001). No Brasil, a percentagem de pacientes que receberam soluções cristaloides foi mais elevada (97,7% versus 76,8%; p < 0,001), e solução de cloreto de sódio a 0,9% foi o cristaloide mais comumente utilizado (62,5% versus 27,1%; p < 0,001). A análise multivariada sugeriu que os níveis de albumina se associaram com o uso tanto de cristaloides quanto de coloides, enquanto o tipo de prescritor dos fluidos se associou apenas com o uso de cristaloides. Conclusão: Nossos resultados sugerem que cristaloides são usados mais frequentemente do que coloides para ressuscitação no Brasil, e essa discrepância, em termos de frequências, é mais elevada do que em outros países. A solução de cloreto de sódio 0,9% foi o cristaloide mais frequentemente prescrito. Os níveis de albumina sérica e o tipo de prescritor de fluidos foram os fatores associados com a escolha de cristaloides ou coloides para a prescrição de fluidos.
Abstract Objective: To describe fluid resuscitation practices in Brazilian intensive care units and to compare them with those of other countries participating in the Fluid-TRIPS. Methods: This was a prospective, international, cross-sectional, observational study in a convenience sample of intensive care units in 27 countries (including Brazil) using the Fluid-TRIPS database compiled in 2014. We described the patterns of fluid resuscitation use in Brazil compared with those in other countries and identified the factors associated with fluid choice. Results: On the study day, 3,214 patients in Brazil and 3,493 patients in other countries were included, of whom 16.1% and 26.8% (p < 0.001) received fluids, respectively. The main indication for fluid resuscitation was impaired perfusion and/or low cardiac output (Brazil: 71.7% versus other countries: 56.4%, p < 0.001). In Brazil, the percentage of patients receiving crystalloid solutions was higher (97.7% versus 76.8%, p < 0.001), and 0.9% sodium chloride was the most commonly used crystalloid (62.5% versus 27.1%, p < 0.001). The multivariable analysis suggested that the albumin levels were associated with the use of both crystalloids and colloids, whereas the type of fluid prescriber was associated with crystalloid use only. Conclusion: Our results suggest that crystalloids are more frequently used than colloids for fluid resuscitation in Brazil, and this discrepancy in frequencies is higher than that in other countries. Sodium chloride (0.9%) was the crystalloid most commonly prescribed. Serum albumin levels and the type of fluid prescriber were the factors associated with the choice of crystalloids or colloids for fluid resuscitation.

RESUMO: Este trabalho teve como objetivo identificar a variabilidade espacial da resistência do solo à penetração e na produtividade da cultura da soja, em área de várzea. Foram realizadas determinações de resistência do solo à penetração, em quatro profundidades (0 a 0,10 m; 0,11 a 0,20 m; 0,21 a 0,30 m e 0,31 a 0,40 m); umidade volumétrica do solo, em duas profundidades (0 a 0,20 m e 0,21 a 0,40 m); e produtividade da soja em uma área de 1,13 hectares, utilizando-se malha amostral de 10 x 10 m. Os dados foram submetidos à análise estatística descritiva. Realizou-se análise de correlação linear simples de Pearson (p≤0,05) e a dependência espacial foi avaliada pela análise de semivariogramas isotrópicos, utilizando os modelos: esférico, exponencial, linear e gaussiano. Os resultados indicaram que, a resistência do solo à penetração aumentou em profundidade, com valores restritivos ao crescimento radicular entre 0,05 e 0,35 m. Não se obteve correlação entre produtividade e resistência do solo à penetração sendo que, para os dados de resistência do solo à penetração, os semivariogramas não apresentaram uma região ascendente definida. Para as condições em que o experimento foi realizado, a malha amostral de 10 x 10 m utilizada foi adequada para avaliar a variabilidade espacial da resistência do solo à penetração em profundidades superiores a 0,10 m.
ABSTRACT: This study determined the spatial variability of soil penetration resistance and yield of the soybean crop in lowland areas. The soil resistance to penetration at four different depths (0 to 0.10 m; 0.11 to 0.20 m; 0.21 to 0.30 m and 0.31 to 0.40 m), volumetric humidity of the soil at two depths (0 to 0.20 m and 0.21 to 0.40 m) and soybean yield were determined in an area of 1.13 hectares, using a sample mesh of 10 x 10 m. The corresponding data were subjected to descriptive statistical analysis. Pearson’s simple linear correlation analysis (p≤0.05) was conducted and the spatial dependence was assessed by analyzing the isotropic semivariograms using spherical, exponential, linear, and Gaussian models. The results showed that the soil penetration resistance increased with depth, with restrictive values to root growth between 0.05 and 0.35 m. There was no correlation between yield and soil penetration resistance, and the semivariograms did not show a defined ascending region regarding the soil penetration resistance data. For the conditions under which the experiment was conducted, the sample 10 x 10 m mesh was suitable for assessing the spatial variability of soil resistance to penetration in depths exceeding 0.10 m.

Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

RESUMO Introdução: Embora a reação em cadeia da polimerase de transcrição reversa (rRT-PCR) seja o método padrão-ouro para detecção de coronavírus da síndrome respiratória aguda grave 2 (SARS-CoV-2), alguns fatores como a presença de inibidores de amplificação levam a resultados falso negativos. Objetivo: Descrevemos as diferenças entre os resultados de rRT-PCR para infecção por SARS-CoV-2 em amostras normais e diluídas, simulando a necessidade de diluição devido à presença de inibidores de amplificação. Material e método: A extração de ácido ribonucleico (RNA) viral de amostras de suabes nasofaríngeos de 20 pacientes previamente detectados como “negativos” e 21 pacientes detectados como “positivos” para SARS-CoV-2 foi realizada com kit o EasyExtract DNA-RNA (Interprise®). A rRT-PCR foi realizada com o kit OneStep/COVID-19 (IBMP), com amostras normais e diluídas (80 µl de H2O RNAse-free), totalizando 82 testes. Resultados: Os resultados indicam que existe uma variação média (a < 0,05) atrasando o Cq entre os resultados de amplificação do controle interno (CI), gene N (GN) e ORF1ab (OF) de 1,811 Cq, 3,840 Cq e 3,842 Cq, respectivamente. Discussão: O kit de extração não purifica completamente os compostos inibidores, portanto, pode ocorrer não amplificação. Obtivemos um diagnóstico falso negativo de 19,04% após a diluição da amostra; esse processo reduz a eficiência da rRT-PCR para 29,8% na detecção de SARS-CoV-2. Conclusão: Conhecer os padrões da rRT-PCR de amostras diluídas pode auxiliar na identificação de casos falso negativos e, consequentemente, evitar um diagnóstico incorreto.
ABSTRACT Introduction: Although reverse transcription-polymerase chain reaction (rRT-PCR) is the gold standard method for detecting severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), some factors, such as the presence of amplification inhibitors, lead to false-negative results. Objective: Here we describe the differences between rRT-PCR results for SARS-CoV-2 infection in normal and diluted samples, simulating the need for dilution due to the presence of amplification inhibitors. Material and method: Viral ribonucleic acid (RNA) from samples of nasopharyngeal swabs from 20 patients previously detected as “Negative” and 21 patients detected as “Positive” for SARS-CoV-2 was performed with the EasyExtract DNA-RNA kit (Interprise®). The rRT-PCR was performed with the OneStep/COVID-19 kit (IBMP), with normal and diluted (80 µl of H2O RNAse free) samples, totaling 82 tests. Results: The results indicate that there is an average variation (a < 0.05) delaying the Cq between the results of amplification of the internal control (IC), N gene (NG), and ORF1ab (OF), 1.811 Cq, 3.840 Cq, and 3.842 Cq, respectively. Discussion: The extraction kit does not completely purify the inhibitor compounds; therefore, no amplified product result may occur. In this study, we obtained a 19.04% false-negative diagnosis after sample dilution; this process reduces the efficiency of rRT-PCR to 29.8% in detecting SARS-CoV-2. Conclusion: Knowing the rRT-PCR standards of diluted samples can assist in the identification of false-negative cases and, consequently, avoid incorrect diagnosis.

Objetivo: Describimos las diferencias entre los resultados de rRT-PCR para infección por Sars-CoV-2 en muestras normales y diluidas, simulando la necesidad de dilución debido a la presencia de inhibidores de amplificación. Material y método: La extracción de ácido ribonucleico (ARN) viral de muestras de hisopos nasofaríngeos de 20 pacientes previamente detectados como “negativos” y 21 pacientes detectados como “positivos” para Sars-CoV-2 se realizó con el kit Easy Extract DNA-RNA (Interprise®). La rRT-PCR se realizó con el kit OneStep/Covid-19 (IBMP), con muestras normales y diluidas (80 µl de H2O libre de ARNasa), totalizando 82 pruebas. Resultados: Los resultados indican que hay una variación media (a < 0,05) retrasando el ciclo de cuantificación (Cq) entre los resultados de amplificación del control interno (CI), gen N (GN) y ORF1ab (OF) de 1,811 Cq, 3,840 Cq y 3,842 Cq. Discusión: El kit de extracción no purifica completamente los compuestos inhibidores; por lo tanto, puede ocurrir no amplificación. Obtuvimos un diagnóstico falso negativo de 19,04% después de la dilución de la muestra; ese proceso reduce la eficiencia de la rRT-PCR hacia 29,8% en la detección de Sars-CoV-2. Conclusión: Conocer los patrones de la rRT-PCR de muestras diluidas puede ayudar en la identificación de casos falsos negativos y, por consiguiente, evitar un diagnóstico equivocado.

Abstract Pereskia grandifolia Haworth (PGH) and Pereskia aculeata Miller (PAM) are recognized sources of proteins; dietary fiber; vitamins and minerals make this plant leaves, raw, cooked, and braised, an important ally against protein and micronutrient deficiencies. One of the main problems is the presence of antinutritional factors that may interfere in the digestibility and bioavailability of some nutrients. The objective was to evaluate the amino acid profile and the chemical score of the raw leaves and the effects of heating media and time on the total dietary fiber, minerals, trypsin inhibition, oxalic acid and tannins of leaves of PGH and PAM. The samples had similar amino acid profiles and total dietary fiber. With regard to antinutritional compounds, heating the leaves of PGH led to a decrease in trypsin inhibition, primarily after the first minutes of wet cooking. Oxalic acid and tannins predominated in both species. Considering the interaction with time, the variables related to iron and zinc minimized the tannin responses in PGH and PAM, respectively. Heating media and times interfered with the chemical components present in the leaves of Pereskia species and led to high antinutrient retention after heat treatment.

OBJECTIVES: We designed a cohort study to describe characteristics and outcomes of patients with coronavirus disease (COVID-19) admitted to the intensive care unit (ICU) in the largest public hospital in Sao Paulo, Brazil, as Latin America becomes the epicenter of the pandemic. METHODS: This is the protocol for a study being conducted at an academic hospital in Brazil with 300 adult ICU beds dedicated to COVID-19 patients. We will include adult patients admitted to the ICU with suspected or confirmed COVID-19 during the study period. The main outcome is ICU survival at 28 days. Data will be collected prospectively and retrospectively by trained investigators from the hospital’s electronic medical records, using an electronic data capture tool. We will collect data on demographics, comorbidities, severity of disease, and laboratorial test results at admission. Information on the need for advanced life support and ventilator parameters will be collected during ICU stay. Patients will be followed up for 28 days in the ICU and 60 days in the hospital. We will plot Kaplan-Meier curves to estimate ICU and hospital survival and perform survival analysis using the Cox proportional hazards model to identify the main risk factors for mortality. RESULTS: We expect to include a large sample of patients with COVID-19 admitted to the ICU and to be able to provide data on admission characteristics, use of advanced life support, ICU survival at 28 days, and hospital survival at 60 days. CONCLUSIONS: This study will provide epidemiological data about critically ill patients with COVID-19 in Brazil, which could inform health policy and resource allocation in low- and middle-income countries.

In vitro and in vivo efficacy of tea tree essential oil for bacterial and yeast ear infections in dogs

RESUMO: Otite externa é queixa frequente em cães. Bactérias e leveduras estão comumente envolvidas e podem perpetuar as reações inflamatórias dentro do canal auditivo. Dentre as formas de diagnóstico, encontram-se a otoscopia, o exame citológico da secreção e a cultura microbiológica. Citologia também tem grande utilidade no acesso à evolução do tratamento. A terapia consiste de limpeza dos canais auditivos e posterior utilização de antibióticos ou produtos antifúngicos. Como alguns antimicrobianos utilizados no tratamento podem causar hipersensibilidade e até mesmo ototoxicidade, a busca por novas bases farmacológicas justifica a existência deste estudo, que teve como objetivo avaliar in vitro e in vivo a eficiência do óleo de Melaleuca alternifolia em otites bacterianas e fúngicas de cães. Vinte e oito cães, de um abrigo particular, apresentando sinais clínicos de otite externa, foram incluídos neste estudo clínico. Todos passaram por avaliação clínica, citologia e cultura de material das orelhas afetadas. De cada animal, uma orelha foi tratada com óleo de Melaleuca 5% e a outra com formulação ótica padrão, de acordo com a afecção (bacteriana, fúngica ou mista). As culturas também foram submetidas a testes de susceptibilidade in vitro aos mesmos agentes utilizados no tratamento in vivo. Os resultados da cultura mostraram 62,5% de infecção mista (bacteriana e fúngica), 33,9% de infecção bacteriana e 3,6%, de infecção fúngica a partir das 56 orelhas. Os micro-organismos mais isolados foram Staphylococcus intermedius, Staphylococcus aureus, Proteus mirabilis e Malassezia pachydermatis. As bactérias GRAM-positivas foram sensíveis à gentamicina em 60,5% e resistentes em 16,3% das amostras. A formulação com 5% de óleo essencial de Melaleuca produziu uma zona de inibição de 5mm em torno dos discos em uma das 63 amostras avaliadas. A formulação pura (100%) do mesmo produto produziu uma zona de 10mm de inibição em quatro das 63 amostras analisadas, uma zona de 9 mm em três amostras, uma zona de 8mm em 16 amostras, uma zona de 7mm em sete amostras, uma zona de 6mm em duas amostras e não havia nenhuma zona clara em 31 amostras. Zonas de inibição foram produzidas por estirpes de Staphylococcus intermedius, Staphylococcus hyicus, Corynebacterium sp., Proteus mirabilis e Enterobacter sp. Clinicamente, a formulação com o óleo essencial de melaleuca induziu significativamente uma melhora nas manifestações, tanto nas otites bacterianas quanto nas causadas por leveduras, sendo estatisticamente semelhante ao tratamento com nistatina (nas otites por levedura), mas menos eficaz que a solução de gentamicina nas otites bacterianas. Mais estudos devem ser realizados para avaliar as propriedades de difusão in vitro do óleo essencial de melaleuca. O bom espectro antimicrobiano, a boa resposta clínica e a ausência de reações adversas confirmam a possibilidade de desenvolvimento de formulação ótica com o óleo essencial de melaleuca, como uma alternativa para a terapia de infecções do ouvido em cães.
ABSTRACT: Otitis externa is a common complaint in dogs. Bacteria and yeasts are commonly involved and may perpetuate inflammatory reactions inside the ear canal. Otoscopy, cytological examination of secretion and microbiological culture embody forms of diagnosis. Cytology also has great use in accessing treatment evolution. Therapy usually consists of cleaning ear canals and subsequent use of antibiotics or antifungal products. As some of them may cause hypersensitivity and even ototoxicity, searching for new pharmacological bases is currently necessary and justifies this study, which aimed to evaluate in vitro and in vivo efficacy of tea tree essential oil for bacterial and yeast ear infections in dogs. Twenty-eight dogs from a particular shelter in Cuiabá (Mato Grosso, Brazil), presenting clinical signs of otitis externa, were enrolled in this clinical trial. In all of them, clinical and cytological evaluations, as well as culture and susceptibility testing of the affected ears were carried out. From each dog, one ear was treated with 5% tea tree essential oil lotion and the other with standard otic formulation, according to the type of infection (bacterial, yeast or both). In vitro susceptibility testings of all ear cultures, to the same drugs used in treatment, were also carried out. Culture results showed 62.5% bacterial and fungal infection, 33.9% bacterial infection and 3.6% fungal infection, from the 56 ear samples collected. The most common microorganisms isolated were Staphylococcus intermedius, Staphylococcus aureus, Proteus mirabilis and Malassezia pachydermatis. Gram-positive bacteria were susceptible to gentamycin in 60.5% and resistant in 16.3% of the samples. Five percent tea tree essential oil formulation produced a 5mm clear zone of inhibition around the disks in one of the 63 samples evaluated. Pure (100%) tea tree essential oil formulation produced a 10mm clear zone of inhibition around the disks in four of the 63 samples evaluated, a 9mm zone in three samples, an 8mm zone in 16 samples, a 7mm zone in seven samples, a 6mm zone in two samples and there was no clear zone in 31 samples. Inhibition zones were produced by strains of Staphylococcus intermedius, Staphylococcus hyicus, Corynebacterium sp., Proteus mirabilis and Enterobacter sp. tea tree essential oil ear solution significantly induced remission of clinical signs both in bacterial and yeast ear infections. It also reduced as much Malassezia pachydermatis ear infection as the nystatin solution used in this study, while gentamycin solution showed better antibacterial effect. More studies should be conducted to evaluate in vitro diffusion properties of tea tree essential oil. Good antimicrobial spectrum and the absence of adverse reactions confirm the importance of developing a tea tree formulation as an alternative therapy for ear infections in dogs.

Susceptibilidade antibiótica de isolados bacterianos em diferentes tipos de ceratites ulcerativas de cães na cidade de Cuiabá

RESUMO: Objetivou-se identificar microrganismos isolados de diferentes tipos de ceratite ulcerativa em cães, juntamente com a sua susceptibilidade a antimicrobianos. O resultado do tratamento médico e cirúrgico também foi correlacionado com o tipo de isolado. Amostras para microbiologia foram obtidas com auxílio de swab estéril em 104 olhos de 72 pacientes sem histórico prévio de tratamento com antibióticos tópicos, atendidos no período de maio de 2012 a março de 2015. Os antibióticos testados foram: neomicina, gentamicina, tobramicina, cloranfenicol, polimixina B, ciprofloxacino, ofloxacino e moxifloxacina. No total, 131 bactérias foram isoladas de 96/104 olhos estudados, sendo o gênero Staphylococcus (48,09%) predominante, seguido por Pseudomonas aeruginosa (16,01%). O Shih Tzu foi a raça mais prevalente (33,33%) e o número de isolados gram-negativos foi significativamente maior nessa raça, comparativamente aos Pinschers (p=0,003), aos Filas, aos Poodles e aos sem raça definida (p=0,046). As bactérias isoladas neste estudo apresentaram maior susceptibilidade ao ofloxacino (84,55%), que foi significativamente mais eficaz em relação a neomicina e a polimixina B (p<0,0001), ao cloranfenicol (p=0,0001), a tobramicina (p=0,0007), a gentamicina (p=0,0021) e as outras fluorquinolonas, ciprofloxacino (p=0,0004) e moxifloxacino (p<0,0001). Os organismos gram-positivos foram isolados de um número significativamente maior de olhos que apresentavam ceratite ulcerativa não complicada, comparativamente àqueles com olhos acometidos por ceratite ulcerativa complicada (p=0,011). Igualmente, o número de bactérias gram-positivas foi maior que o de gram-negativas, tanto nos casos que receberam tratamento médico, como nos que foram operados, sem significativa estatística (p=0,745). Na presente pesquisa, Staphylococcus sp. foi a bactéria mais encontrada nas ceratites ulcerativas não complicadas. Já nos olhos com ceratites complicadas, embora a Pseudomonas aeruginosa tenha sido a bactéria mais predominante, o tratamento clínico foi suficiente para cura da afecção corneal na maior parte dos casos. O ofloxacino e a gentamicina foram os agentes mais eficazes contra a maioria dos isolados, com exceção do Streptococcus sp., onde o cloranfenicol se mostrou o mais eficaz. Ceratites ulcerativas sem complicações que apresentem culturas negativas podem evoluir para ceratites ulcerativas complicadas, salientando a necessidade de tratamento anti-colagenolítico em todos os casos.
ABSTRAT: The purpose of the present study was to analyze antibiotic susceptibility of bacteria associated with different types of ulcerative keratitis in dogs. The outcome of medical or surgical treatment was also correlated with the type of isolate. Samples for microbiology were obtained by means of sterile swab from 104 eyes of 72 canine patients with ulcerative keratitis without previous history of antibiotic treatment, seen from May 2012 to March 2015. Only patients with no previous treatment with antibiotics were included in the study. Bacterial isolates were identified and the antibiotic susceptibility was tested to neomycin, gentamicin, tobramycin, chloramphenicol, polymyxin B, ciprofloxacin, ofloxacin and moxifloxacin. In total, 131 species of bacteria were isolated from 96/104 eyes, and Staphylococcus sp. predominated (48.09%), followed by Pseudomonas aeruginosa (16.01%). Shih Tzus were over represented (33.33%) and the number of gram-negative isolates were significantly higher in this breed, in comparison to Pinchers (P=0.003), Filas, Poodles, and other mixed-breeds (P=0.046). All species isolated in this study were more sensitive to ofloxacin (84.55%), that was significantly most efficient than neomycin and polymyxin B (P<0.0001), chloramphenicol (P=0.0001), tobramycin (P=0.0007), gentamicin (P=0.0021) and the other fluoroquinolones, ciprofloxacin (P=0.0004) and moxifloxacin (P<0.0001). Gram-positive organisms were isolated in a significant larger number of eyes with uncomplicated ulcerative keratitis, in comparison to those eyes with complicated ulcerative keratitis (P=0.011). Likewise, gram-positive were isolated in a larger number than gram-negatives microorganisms in cases that received either medically or surgical treatment, without statistical significance (P=0.745). In the present research, Staphylococcus sp. was the bacteria most commonly isolated in the eyes with uncomplicated ulcerative keratitis. Although Pseudomonas aeruginosa was the most common isolate in the eyes with complicated ulcerative keratitis, the majority of cases managed clinically had a successful outcome. Ofloxacin and gentamicin were found to be effective against the majority of isolates, with the exception of Streptococcus. sp, in which chloramphenicol was the most effective antibiotic. Uncomplicated ulcerative keratitis presenting negative culture may evolve to complicated ulcerative keratitis, warring the necessity of anti-collagenolytic treatment in all cases.
