Foliar structural differences between glyphosate-resistant and glyphosate-susceptible biotypes of Digitaria insularis (L.) Fedde

ABSTRACT Digitaria insularis (L.) Fedde stands out for its resistance to glyphosate and this characteristic may have a relationship with structural alterations. In such context, this research aimed at the characterization of the foliar anatomical structure of two populations of D. insularis (glyphosate-resistant and glyphosate-susceptible biotypes) collected in agricultural areas of Paraná, a Brazilian state. The experiment was conducted at the Laboratory of Botany of the Universidade Estadual do Oeste do Paraná – Unioeste, Brazil. The resistant biotypes of D. insularis differ from the susceptible ones in several structural parameters. Among them, the Mesophyll Thickness in the interveinal region was 7.3% thicker in the resistant biotype, which was also observed in the thickness of the keel, in the percentage of 11.3%, and in the thickness of cuticles in the adaxial surface (TCad), which was 53.8% thicker in the resistant biotype. In this way, we concluded that the resistant biotypes of D. insularis differ from the susceptible ones in several anatomical foliar characteristics, therefore, they present possible mechanisms of resistance to glyphosate.

We had, as an objective, characterizing, anatomically, the leaves of two populations of D. insularis (resistant biotype and susceptible biotype), which were collected in cropping areas in Paraná. In order to do this, a trial design in random sets was employed, which were repeated ten times. To obtain the data, it was evaluated the structural and quantitative variables of foliar blades of anatomical parts, which belonged to different leaves in the mature stage that were removed from distinct resistant and susceptible biotypes of D. insularis. The structural variables of the cross-sectional parts evaluated from the foliar blades were: the thickness of the foliar blade in the intercostal region (TBI); thickness of the keel (TK); stomata number (SN) and thickness of cuticle (TC) in the adaxial and abaxial faces. The resistant biotypes of D. insularis differ from the susceptible ones in several structural parameters, among them, there is the foliar blade in the interveinal region that was 7.3 ticker in the resistant biotype, which was also observed in the thickness of the keel, in the percentage of 11.3%, and in the thickness of the cuticle in the adaxial face (TCD), which was 53.8% thicker in the resistant biotype. In this way, we concluded that the anatomical foliar characteristics observed in the resistant biotypes, which differ from the susceptible ones, can be related to the reduction in the absorption and the herbicides translocation speed, constituting, therefore, possible mechanisms of resistance to glyphosate
Tivemos como objetivo caracterizar anatomicamente as folhas de duas populações de D. insularis (biótipo resistente e biótipo suscetível) coletadas em áreas agrícolas no Paraná. Para isso, utilizou-se delineamento experimental em blocos ao acaso, com dez repetições. Para a obtenção dos dados, foram avaliadas variáveis estruturais quantitativas de secções anatômicas da lâmina foliar, pertencentes a diferentes folhas no estágio maduro retiradas de distintos biótipos resistentes e suscetíveis de D. insularis. Entre as variáveis estruturais das secções transversais da lâmina foliar avaliadas foram: espessura da lâmina foliar na região intercostal (ELI); espessura da quilha (EQ); número de estômatos (NE); espessura da cutícula (EC) nas faces adaxial e abaxial. Os biótipos resistentes de D. insularis diferem dos susceptíveis em vários parâmetros estruturais, entre eles, quanto a lâmina foliar na região internervural que foi 7,3% mais espessa no biótipo resistente, o que também foi observado na espessura da quilha com 11,3% e na espessura da cutícula na face adaxial (ECD) que foi 53,8% mais espessa no resistente. Dessa forma, concluímos que as características anatômicas foliares observadas nos biótipos resistentes que diferem dos susceptíveis, podem estar relacionadas com a redução na absorção e na velocidade de translocação de herbicidas, constituindo assim possíveis mecanismos de resistência ao glyphosate.

Resumo O estudo objetivou avaliar os aspectos anatômicos e o comportamento fisiológico de sementes de Peltophorum dubium (Spreng.) Taub. durante o armazenamento, submetidas à ação de tratamentos para superação da dormência. Os frutos maduros foram beneficiados e as sementes armazenadas à temperatura de 5ºC por um período de 360 dias e, posteriormente, submetidas a tratamentos para superação da dormência: (T1 - sem escarificação, T2 - escarificação mecânica e T3 - escarificação química com ácido sulfúrico por 10 minutos). Em intervalos pré-estabelecidos, as sementes foram avaliadas por meio da análise anatômica do tegumento e pelo teste de germinação. As variáveis analisadas foram anatomia do tegumento, porcentagem de germinação, índice de velocidade de germinação e correlação de Pearson entre a espessura do tegumento e as variáveis fisiológicas. Para as análises anatômicas, as sementes foram fixadas, inclusas e seccionadas em micrótomo. A espessura do tegumento das sementes foi mensurada por meio de imagens das secções transversais. O teste de germinação foi conduzido com quatro repetições de 25 sementes, realizado em papel germitest à temperatura constante de 25ºC com fotoperíodo de 12 horas. O delineamento experimental foi inteiramente casualizado em esquema fatorial 5x3 (lotes armazenamento e tratamentos para superação da dormência) e as médias comparadas pelo teste de Skott-Knott a 5% de probabilidade. As análises anatômicas demonstraram perda de resistência física das estruturas do tegumento ao longo do armazenamento. A porcentagem de germinação durante o armazenamento não diferiu significativamente entre os tratamentos, entretanto o índice de velocidade de germinação aumentou e forma significativa quando associado a tratamentos de superação da dormência (T2. 7,08 - sementes não armazenadas e 9,29-armazenadas 360 dias; T3. 6,01-sementes não armazenadas e 9,42-armazenadas 360 dias). Foram obtidas correlações significativas entre a espessura do tegumento e as variáveis fisiológicas analisadas. O armazenamento, por até 360 dias, promoveu modificações anatômicas tegumentares, manteve o alto percentual germinativo das sementes de Peltophorum dubium e, quando associado aos tratamentos de superação da dormência, facilitou a ação destes a partir de 180 dias de armazenagem, aumentando o índice de velocidade de germinação.
Abstract The objective of this study was to evaluate the anatomical aspects and the physiological behavior of seeds of Peltophorum dubium (Spreng.) Taub. during storage and while undergoing treatments to overcome dormancy. Mature fruits were collected and seeds stored at 5ºC for a period of 360 days and later submitted to treatments to overcome dormancy: T1 - without scarification, T2 - mechanical scarification and T3 - chemical scarification with sulfuric acid for 10 minutes. At pre-established intervals, the seeds were evaluated through an anatomical analysis of the tegument and a germination test. The variables analyzed were tegument anatomy, germination percentage, germination speed index and Pearson correlation between tegument thickness and physiological variables. For the anatomical analyses, the seeds were fixed, embedded, and sectioned on a microtome. The thickness of the tegument was measured from the images of the cross sections. The germination test was conducted with four replicates of 25 seeds, carried out on germitest paper at 25ºC with photoperiod of 12 hours. The experimental design was completely randomized in a 5×3 factorial scheme (storage and overcoming dormancy treatment groups), and the means were compared by the Skott-Knott test at 5% probability. The anatomical analysis showed loss of physical resistance of the tegumentary structures during storage. The percentage of germination for the treatments during storage did not differ significantly, however, the rate of germination showed a significant increase when associated with treatments for overcoming dormancy (T2: 7.08 - seeds not stored and 9.29- stored 360 days; T3: 6.01-seeds not stored and 9.42-stored 360 days). Significant correlations were obtained between the thickness of the tegument and the physiological variables analyzed. Storage for up to 360 days promoted tegumentary anatomical modifications, maintained a high germination percentage of the seeds of Peltophorum dubium, and, when associated with treatments for overcoming dormancy, facilitated their action from 180 days of storage, increasing the rate of germination.

RESUMO Mimosa flocculosa Burkart é uma espécie nativa, utilizada para recuperação de áreas degradadas. Porém, a produção de mudas em larga escala é dificultada pela dormência tegumentar presente na semente. Esse trabalho avaliou a germinação e a anatomia do tegumento de sementes submetidas à tratamentos de superação da dormência. Sementes de M. flocculosa foram submetidas aos tratamentos: controle, água aquecida a 50, 80 e 100 ºC e H2SO4 durante 1, 3 e 5 minutos. A germinação contou com quatro repetições de 25 sementes em delineamento inteiramente casualizado. As variáveis analisadas foram: germinação, tempo médio e índice de velocidade de germinação. Foram realizadas ainda análises anatômicas do tegumento das sementes. Os tratamentos usando água a 80 ºC, e ácido sulfúrico por 3 e 5 minutos promoveram mudanças anatômicas benéficas no tegumento da semente, refletindo no aumento da porcentagem e velocidade de germinação. Recomendamos a água aquecida a 80 ºC pela eficiência e maior segurança na manipulação.
ABSTRACT Mimosa flocculosa Burkart is a native species used for the recovery of degraded areas. Large scale seedling production in nurseries is hindered due to seed physical dormancy. This study evaluated the germination and seed coat anatomy of M. flocculosa seeds subjected to dormancy overcome treatments. Seeds of M. flocculosa were subjected to: control with distilled water, heated water at 50, 80 and 100 °C, and H2SO4 for 1, 3 and 5 minutes. Germination tests were conducted with four replicates of 25 seeds in an entirely casualized delineation, analyzing germination percentage, mean germination time and germination speed index. Moreover, anatomical analysis of the seed coat was conducted. The treatments with heated water at 80 ºC, and sulfuric acid for 3 and 5 minutes promoted benefic anatomical alterations in the seed integument, reflecting in higher and faster in germination rates. Heated water at 80 ºC is recommended because of its efficiency and safety.

Synopsis of Cyperaceae in the grasslands of Guartelá State Park, Paraná, Brazil

Resumo Cyperaceae é a terceira maior família dentre as monocotiledôneas, com grande representatividade em ambientes campestres. O presente estudo teve como objetivo realizar o levantamento florístico de Cyperaceae nas formações campestres do Parque Estadual do Guartelá (PEG) e elaborar chaves de identificação para as espécies, sendo o primeiro para a família nestes tipos de formações para o estado do Paraná. O PEG apresenta uma vegetação relictual do Paraná, onde predominam fisionomias campestres, destacando-se áreas abertas e afloramentos rochosos. O levantamento foi realizado com base em coletas em campo e buscas em banco de dados. A família Cyperaceae no PEG está representada por 32 táxons pertencentes a sete gêneros, sendo Rhynchospora (11 spp.), Bulbostylis (8 spp.) e Cyperus (7 spp.) os que apresentaram a maior riqueza. Ressalta-se que todas as espécies são nativas, sendo duas endêmicas do Brasil. Registrou-se a ocorrência das espécies tanto em áreas drenadas quanto úmidas, destacando a importância da conservação dos microambientes do parque. São fornecidas principais literaturas para cada gênero, informações acerca da distribuição e habitat.
Abstract Cyperaceae is the third largest family among the monocotyledons and is highly represented in grassland environments. The aim of the present study was to conduct a floristic inventory of Cyperaceae in the grasslands of Guartelá State Park (GSP) and to elaborate keys to identify the species, the first for this family in this vegetation type in Paraná state. The GSP is home to relictual vegetation in Paraná, where grassland phytophysiognomies predominate, especially in open areas and rocky outcrops. The inventory was based on field collections and database searches. The family Cyperaceae is represented in the GSP by 32 taxa belonging to seven genera, with Rhynchospora (11 spp.), Bulbostylis (8 spp.), and Cyperus (7 spp.) being the genera with the greatest richness. It should be noted that all the species are native and two are endemic to Brazil. Species occurrence was recorded both in drylands and wetlands environments, emphasizing the importance of microenvironmental conservation in the park. The main literature sources for each genus, as well as information regarding distribution and habitat, are here provided.

RESUMO Poaceae é uma família cosmopolita que inclui espécies encontradas em formações florestais e campestres. O objetivo deste estudo foi realizar o primeiro levantamento florístico de Poaceae no Parque Estadual do Guartelá (PEG), um dos últimos remanescentes de vegetação nativa de formação campestre no Estado do Paraná, Brasil, e fornecer recursos para auxiliar na identificação dos gêneros e espécies. A pesquisa foi realizada com base em coletas a campo e busca na base de dados online Species Link. No PEG, Poaceae é representada por 29 gêneros e 54 espécies, sendo Paspalum L. (oito spp.), Andropogon L. (cinco spp.) e Eragrostis Wolf (quatro spp.) os gêneros mais ricos. O número de espécies de Poaceae no PEG foi incrementado em 29 novos registros. Além disso, cinco espécies foram classificadas como exóticas, cinco como endêmicas para o Brasil e duas foram consideradas menos preocupantes em relação à sua conservação. Também são apresentadas informações sobre distribuição, hábitat e status de conservação de cada espécie.
ABSTRACT Poaceae is a cosmopolitan family that includes species found in both forest and grassland formations. The objective of this study was to carry out the first floristic survey of Poaceae in the Parque Estadual do Guartelá (PEG), one of the last remnants of native grassland vegetation in the State of Paraná, Brazil, and provide resources to assist the identification of genera and species. The survey was conducted based on field collections and searches in the Species Link online database. In the PEG, Poaceae is represented by 29 genera and 54 species, being Paspalum L. (eight spp.), Andropogon L. (five spp.) and Eragrostis Wolf (four spp.) the richest genera. The number of species in the PEG was incremented by 29 new records. In addition, five species were classified as exotic, five as endemic to Brazil, and two were considered least concern in relation to their conservation. We also present information regarding the distribution, habitat and conservation status for each species.

PURPOSE: To evaluate wound healing in rats by using low-level laser therapy (LLLT) associated with hydrocolloid occlusive dressing . METHODS: Forty male, adult, Wistar rats were used, distributed into four groups: LG (received 2 J/cm² of laser therapy); HG (treated with hydrocolloid); LHG (treated with 2 J/cm² of laser therapy and hydrocolloid); and the CG (treated with 1 mL of 0.9% saline). The wound was evaluated at pre-determined periods 3rd and 7th days, considering the macroscopic and histological parameters (inflammatory cells, capillary neoformation, fibroblasts, collagen formation and reepithelialization). RESULTS: The LG group at seven days showed increased collagen formation, the LHG group at 3 days showed mild collagen formation. The HG group and the CG at 7 days showed complete reepithelialization. CONCLUSION: Low-level laser therapy as well as the hydrocolloid dressing have favored the wound-healing process in rats.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

INTRODUÇÃO: As meningites bacterianas apresentam grande relevância social, devido à sua capacidade de produzir sequelas e mortes. Ocorrem mais frequentemente nos países em desenvolvimento, especialmente entre crianças. Meningite meningocócica ocorre com maior intensidade em populações de baixas condições de vida. Este estudo descreve a evolução temporal das meningites bacterianas em Salvador, 1995-2009, e verifica a associação entre sua variação espacial e condições de vida da população. Métodos: Realizou-se um estudo ecológico no qual as zonas de informação foram classificadas por um índice de condições de vida. Foram examinadas flutuações nas curvas de tendência, e a relação entre este índice e a distribuição espacial das meningites foi verificada por meio de regressão linear simples. RESULTADOS: De 1995 a 2009 foram confirmados, em Salvador, 3.456 casos de meningites bacterianas. Observou-se no período uma tendência descendente, incidência anual de 9,1/100.000 e letalidade de 16,7%. Crianças menores de cinco anos de idade e do sexo masculino foram mais afetadas. Não houve autocorrelação espacial significativa ou padrão na distribuição espacial da doença. Áreas com piores condições de vida apresentaram maior letalidade da doença meningocócica (β = 0,0078117, p <0,005). CONCLUSÕES: As meningites bacterianas atingem todos os estratos sociais, no entanto em áreas onde a população é menos favorecida encontra-se maior proporção de casos que evoluem para a morte. Este achado reflete as dificuldades para pronto acesso e a má qualidade dos cuidados médicos enfrentadas por essas populações.
INTRODUCTION: Bacterial meningitis has great social relevance due to its ability to produce sequelae and cause death. It is most frequently found in developing countries, especially among children. Meningococcal meningitis occurs at a high frequency in populations with poor living conditions. This study describes the temporal evolution of bacterial meningitis in Salvador, Brazil, 1995-2009, and verifies the association between its spatial variation and the living conditions of the population. METHODS: This was an ecological study in which the areas of information were classified by an index of living conditions. It examined fluctuations using a trend curve, and the relationship between this index and the spatial distribution of meningitis was verified using simple linear regression. RESULTS: From 1995-2009, there were 3,456 confirmed cases of bacterial meningitis in Salvador. We observed a downward trend during this period, with a yearly incidence of 9.1 cases/100,000 population and fatality of 16.7%. Children aged <5 years old and male were more affected. There was no significant spatial autocorrelation or pattern in the spatial distribution of the disease. The areas with the worst living conditions had higher fatality from meningococcal disease (β = 0.0078117, p < 0.005). CONCLUSIONS: Bacterial meningitis reaches all social strata; however, areas with poor living conditions have a greater proportion of cases that progress to death. This finding reflects the difficulties for ready access and poor quality of medical care faced by these populations.
