ABSTRACT The expression ‘you need to know to conserve’ is a well-known cliche among biologists. Documenting the richness of a group of organisms is the first step towards understanding biodiversity and preparing efficient conservation plans. In this context, many efforts have been made to quantify the number of species on Earth and estimate the number of species still unknown to science. A few countries have complete and integrated databases estimating the approximate number of species recorded for their territory, particularly in the Global South. In Brazil, a country of continental dimensions, revealing the richness of the second most diverse clade of invertebrates (=Mollusca) has been a goal of taxonomists. Recently, in an unprecedented, collective, and integrated effort among Brazilian malacologists, it was possible to estimate how many valid species of molluscs are there in Brazil. In this effort, more than 30 mollusc experts joined together to update the Taxonomic Catalogue of the Brazilian Fauna (TCBF), a governmental website that allows a quick and real-time updating of all Metazoan. So far, more than 5,000 updates have been made in TCBF, indicating the presence of 3,552 valid species of molluscs in Brazil, distributed among the main clades as follows: Caudofoveata (10 spp.), Solenogastres (6 spp.), Polyplacophora (35 spp.), Scaphopoda (43 spp.), Cephalopoda (92 spp.), Bivalvia (629 spp.) and Gastropoda (2,737 spp.). The present study, in addition to demonstrating for the first time the richness of Brazilian molluscs, also presents the state of the art of this important phylum of invertebrates highlighting its most representative and neglected groups.

The need to fully exploit fishing resources due to increasing production and consequent waste generation requires research to promote the sustainability of the fishing industry. Fish waste from the industry is responsible for relevant environmental contamination. However, these raw materials contain high amounts of collagen and other biomolecules, being attractive due to their industrial and biotechnological applicability. Thus, to reduce the waste from pirarucu (Arapaima gigas) processing, this study aimed to obtain collagen from pirarucu skin tissue. The extraction process used 0.05 M sodium hydroxide, 10% butyl alcohol, and 0.5 M acetic acid, with extraction temperature of 20°C. The obtained yield was 27.8%, and through sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), it was determined that the collagen obtained was type I. This study showed that collagen solubility was highest at pH 3 and the lowest solubility was at concentrations of 3% sodium chloride. The denaturation temperature of collagen was 38.1°C, and its intact molecular structure was observed using the Fourier transform infrared spectrophotometry technique with an absorption radius of 1. The results showed that it was possible to obtain collagen from pirarucu skin at 20°C, which has the typical characteristics of commercial type I collagen. In conclusion, the procedures used may be considered to be an interesting alternative for collagen extraction, a new product obtained from the processing of fish waste.

Neste trabalho utilizamos elementos de geometria diferencial, tais como vetores unitários, tangente e normal, e raio de curvatura, para estudar a compatibilidade de modelos mecânicos em dois problemas de perseguição. No primeiro estudo referente ao problema da curva tractriz, mostramos como a escolha de um modelo de duas partículas conectadas por uma haste rígida, em um primeiro momento, uma escolha aparentemente natural e intuitiva, é incompatível com os demais vínculos do problema. Logo em seguida ampliamos a discussão e consideramos o problema no qual as duas partículas obedecem à cinemática do problema de perseguição de Pierre Bouguer. Neste caso mostramos como a escolha de um modelo de haste hookeana extensível conectando ambas as partículas é compatível com os vínculos do referido problema de perseguição. O software gratuito Modellus é utilizado para simular o comportamento das forças necessárias para justificar a cinemática do sistema. In this work, we use elements of differential geometry, such as unit vectors, tangent and normal, and radius of curvature, to study the compatibility of mechanical models in two pursuit problems. In the first study concerning the problem of the tractrix curve, we show how the choice of a model of two particles connected by a rigid stem, at first a natural and intuitive choice, is incompatible with the other constraints of the problem. Soon after, we expand the discussion and consider the situation in which the two particles obey the kinematics of Pierre Bouguer’s pursuit problem. In this case, we show how the choice of an extensible hookean stem model connecting both particles is compatible with the other constraints of the problem. The free software Modellus is used to simulate the behavior of the forces necessary to justify the system’s kinematics. Resumen Marco contextual: El enfoque de los enfermeros debe abarcar los factores de vulnerabilidad para comprender la dimensión de la intensidad de sus intervenciones, para facilitar el proceso de transición a los cuidados paliativos. Objetivo: Determinar la percepción de los expertos en enfermería sobre los determinantes de la transición para Cuidados Paliativos. Metodología: Estudio cualitativo, con enfoque fenomenológico descriptivo, utilizando el método de análisis de contenido de Bardin. Entrevistas semiestructuradas con 9 expertos de una Unidad de Cuidados Paliativos. Los datos se interpretaron a la luz de la Teoría de las Transiciones, de Afaf Meleis y se organizaron con el software ATLAS.ti®. Resultados: Se identificaron factores facilitadores para la transición, como actitudes y significados positivos sobre Cuidados Paliativos, espiritualidad y religiosidad; y otros identificados como obstáculos, como apoyo familiar insuficiente, nivel socioeconómico bajo, expectativas poco realistas y desconocimiento sobre Cuidados Paliativos. Conclusión: Los enfermeros deben convertirse en agentes facilitadores de la transición a los Cuidados Paliativos, identificando barreras y enfatizando los factores facilitadores para una transición saludable.

Resumo: Enquadramento: O foco de atenção dos enfermeiros deve abarcar os fatores de vulnerabilidade para compreender a dimensão da intensidade das suas intervenções, de forma a facilitar o processo de transição para cuidados paliativos. Objetivo: Determinar a perceção dos peritos em enfermagem sobre as condicionantes da transição para cuidados paliativos. Metodologia: Estudo qualitativo, com abordagem fenomenológica descritiva, com recurso ao método de análise de conteúdo, de Bardin. Entrevistas semiestruturadas a 9 peritos de uma Unidade de Cuidados Paliativos. Os dados foram interpretados à luz da Teoria das Transições, de Afaf Meleis e organizados com o software ATLAS.ti®. Resultados: Identificaram-se fatores considerados facilitadores, tais como atitudes e significados positivos sobre os cuidados paliativos, a espiritualidade e religiosidade; e outros identificados como inibidores, tais como suporte familiar insuficiente, expectativas irrealistas e falta de conhecimento sobre cuidados paliativos. Conclusão: Os enfermeiros devem tornar-se agentes facilitadores da transição para cuidados paliativos, identificando as barreiras e enfatizando os fatores facilitadores com vista a uma transição saudável.

Abstract Background: Nurses’ focus should include vulnerability factors to understand the intensity range of their interventions, in order to facilitate the transition process to palliative care. Objective: To determine the conditioning factors for the transition to Palliative Care from the Clinical Nurse Specialist’ perception. Methodology: Qualitative study, with a descriptive phenomenological approach, using Bardin's method of content analysis. Semi-structured interviews with 9 experts from a Palliative Care Unit. Data were interpreted in the light of Afaf Meleis' Theory of Transitions and organized with the ATLAS.ti®. Results: Facilitating factors were identified, such as positive attitudes and meanings about palliative care, spirituality and religiosity; and others identified as inhibiting conditions, such as insufficient family support, unrealistic expectations and lack of knowledge about palliative care. Conclusion: Nurses must become the facilitators of the patient’s transition process to the palliative care identifying and stepping in the obstacles to their development according to the individual, their family and the caregivers’ needs.

Abstract Currently, antimicrobial resistance has become a global public health problem, which has made the need for new antimicrobial compounds to deal with resistant infections an emergency. However, environments that once offered so many innovative molecules, now already exhaustively exploited, do not meet this need. In this context, a geographically isolated, under-explored and extreme environment, such as Antarctica, which holds organisms with unique physiological and biochemical characteristics, assumes great importance as a potential source of new compounds with antimicrobial activity. In this patent review, we investigate the state of technological development in the field of antimicrobial compounds obtained from Antarctic organisms, highlighting the main countries and researchers active in the field, the species utilized, the compounds obtained, and their possible therapeutic applications. As results, few patent documents were found, however they encompass a wide diversity of compounds and species, indicating a great antimicrobial potential present in Antarctic biota, including compounds active against the most important human pathogenic microorganisms, such as including methicillin-resistant Staphylococcus aureus, vancomycin-resistant Enterococcus spp. and multi-resistant Mycobacterium tuberculosis. Furthermore, due to the increasing trend in patent applications, a significant rise in the number of patents in this area is expected in the coming years.

Cisplatin is an effective antineoplastic agent, but its use is limited by its nephrotoxicity caused by the oxidative stress in tubular epithelium of nephrons. On the other hand, regular exercise provides beneficial adaptations in different tissues and organs. As with many drugs, dosing is extremely important to get the beneficial effects of exercise. Thus, we aimed to investigate the influence of exercise intensity and frequency on cisplatin-induced (20 mg/kg) renal damage in mice. Forty male Swiss mice were divided into five experimental groups (n=8 per group): 1) sedentary; 2) low-intensity forced swimming, three times per week; 3) high-intensity forced swimming, three times per week; 4) low-intensity forced swimming, five times per week; and 5) high-intensity forced swimming, five times per week. Body composition, renal structure, functional indicators (plasma urea), lipid peroxidation, antioxidant enzyme activity, expression of genes related to antioxidant defense, and inflammatory and apoptotic pathways were evaluated. Comparisons considered exercise intensity and frequency. High lipid peroxidation was observed in the sedentary group compared with trained mice, regardless of exercise intensity and frequency. Groups that trained three times per week showed more benefits, as reduced tubular necrosis, plasma urea, expression of CASP3 and Rela (NFkB subunit-p65) genes, and increased total glutathione peroxidase activity. No significant difference in Nfe2l2 (Nrf2) gene expression was observed between groups. Eight weeks of regular exercise training promoted nephroprotection against cisplatin-mediated oxidative injury. Exercise frequency was critical for nephroprotection.

Abstract The objective of this study was to evaluate the effects of different levels of digestible lysine, at fixed amino acid ratios, in performance, blood parameters and diet digestibility of barrows from 15 to 30 kg. Fifty barrows of the Duroc breed with an average initial body weight of 14.95±3.22 kg were assigned to five treatments in a completely randomized experimental design with five replicates and of two animals per experimental unit. Treatments consisted of crude protein and lysine levels, maintaining a fixed ratio with the other digestible amino acids (8.4, 9.4, 10.4, 11.4, and 12.4 g kg–1lysine/kg of diet). The feed conversion decreased linearly according to lysine levels. The apparent digestibility coefficients of dry matter and crude protein increased linearly, and the same response was observed for serum total protein and urea. The results indicate that digestible lysine level for Duroc barrows in the starter phase is 12.4 g kg–1 of diet, as it improved feed conversion ratio and apparent nutrient digestibility.

We evaluated the effects of exercise training (ET) on the profile of mood states (POMS), heart rate variability, spontaneous baroreflex sensitivity (BRS), and sleep disturbance severity in patients with obstructive sleep apnea (OSA). Forty-four patients were randomized into 2 groups, 18 patients completed the untrained period and 16 patients completed the exercise training (ET). Beat-to-beat heart rate and blood pressure were simultaneously collected for 5 min at rest. Heart rate variability (RR interval) was assessed in time domain and frequency domain (FFT spectral analysis). BRS was analyzed with the sequence method, and POMS was analyzed across the 6 categories (tension, depression, hostility, vigor, fatigue, and confusion). ET consisted of 3 weekly sessions of aerobic exercise, local strengthening, and stretching exercises (72 sessions, achieved in 40±3.9 weeks). Baseline parameters were similar between groups. The comparisons between groups showed that the changes in apnea-hypopnea index, arousal index, and O2 desaturation in the exercise group were significantly greater than in the untrained group (P<0.05). The heart rate variability and BRS were significantly higher in the exercise group compared with the untrained group (P<0.05). ET increased peak oxygen uptake (P<0.05) and reduced POMS fatigue (P<0.05). A positive correlation (r=0.60, P<0.02) occurred between changes in the fatigue item and OSA severity. ET improved heart rate variability, BRS, fatigue, and sleep parameters in patients with OSA. These effects were associated with improved sleep parameters, fatigue, and cardiac autonomic modulation, with ET being a possible protective factor against the deleterious effects of hypoxia on these components in patients with OSA.

Physical performance is a multifactorial and complex trait influenced by environmental and hereditary factors. Environmental factors alone have been insufficient to characterize all outstanding phenotypes. Recent advances in genomic technologies have enabled the investigation of whole nuclear and mitochondrial genome sequences, increasing our ability to understand interindividual variability in physical performance. Our objective was to evaluate the association of mitochondrial polymorphic loci with physical performance in Brazilian elite military personnel. Eighty-eight male military personnel who participated in the Command Actions Course of the Army were selected. Total DNA was obtained from blood samples and a complete mitochondrial genome (mtDNA) was sequenced using Illumina MiSeq platform. Twenty-nine subjects completed the training program (FINISHED, ‘F'), and fifty-nine failed to complete (NOT_FINISHED, ‘NF'). The mtDNA from NF was slightly more similar to genomes from African countries frequently related to endurance level. Twenty-two distinct mtDNA haplogroups were identified corroborating the intense genetic admixture of the Brazilian population, but their distribution was similar between the two groups (FST=0.0009). Of 745 polymorphisms detected in the mtDNA, the position G11914A within the NADPH gene component of the electron transport chain, was statistically different between F and NF groups (P=0.011; OR: 4.286; 95%CI: 1.198-16.719), with a higher frequency of the G allele in group F individuals). The high performance of military personnel may be mediated by performance-related genomic traits. Thus, mitochondrial genetic markers such as the ND4 gene may play an important role on physical performance variability.

Abstract Synthetic antioxidants (e.g.butylhydroxytoluene, BHT) are routinely used for to restrict oxidative processes of meat products, but they are implicated as harmful to the health of humans. Therefore natural alternatives, such as plant antioxidants, have been sought as replacements. Plant antioxidants when added to the diet can be incorporated into meat and reduce the need for the addition of synthetic antioxidants during processing. The objective of this study was to evaluate the effects of ethanol extracts of mango seeds (EEMS) in the diet of pigs on qualitative parameters and total antioxidant capacity of mortadella produced from these animals. Thirty-two pigs with an average 60 days of age were distributed among four treatments: control=no antioxidant; BHT=200ppm BHT; EEMS200=200ppm of EEMS and EEMS400=400ppm of EEMS. At 145 days of age the animals were slaughtered and loin was removed for the preparation of mortadella, which was analyzed during 90 days of storage at 4°C. A higher content of polyphenolic compounds and, total antioxidant capacity in mortadellas processed with meat of animals which consumed the EEMS400 ration after 60 and 90 days of storage was observed. EEMS polyphenolic antioxidants incorporated into pork through the diet results in an increase of total antioxidant capacity in the processed product.

It was previously demonstrated that the methanol fraction of Sideroxylon obtusifolium (MFSOL) promoted anti-inflammatory and healing activity in excisional wounds. Thus, the present work investigated the healing effects of MFSOL on human keratinocyte cells (HaCaT) and experimental burn model injuries. HaCaT cells were used to study MFSOL's effect on cell migration and proliferation rates. Female Swiss mice were subjected to a second-degree superficial burn protocol and divided into four treatment groups: Vehicle, 1.0% silver sulfadiazine, and 0.5 or 1.0% MFSOL Cream (CrMFSOL). Samples were collected to quantify the inflammatory mediators, and histological analyses were performed after 3, 7, and 14 days. The results showed that MFSOL (50 μg/mL) stimulated HaCaT cells by increasing proliferation and migration rates. Moreover, 0.5% CrMFSOL attenuated myeloperoxidase (MPO) activity and also stimulated the release of interleukin (IL)-1β and IL-10 after 3 days of treatment. CrMFSOL (0.5%) also enhanced wound contraction, promoted improvement of tissue remodeling, and increased collagen production after 7 days and VEGF release after 14 days. Therefore, MFSOL stimulated human keratinocyte (HaCaT) cells and improved wound healing via modulation of inflammatory mediators of burn injuries.

Exercise intolerance is the hallmark consequence of advanced chronic heart failure (HF). The six-minute step test (6MST) has been considered an option for the six-minute walk test because it is safe, inexpensive, and can be applied in small places. However, its reliability and concurrent validity has still not been investigated in participants with HF with reduced ejection fraction (HFrEF). Clinically stable HFrEF participants were included. Reliability and error measurement were calculated by comparing the first with the second 6MST result. Forty-eight hours after participants underwent the 6MST, they were invited to perform a cardiopulmonary exercise test (CPET) on a cycle ergometer. Concurrent validity was assessed by correlation between number of steps and peak oxygen uptake (V̇O2 peak) at CPET. Twenty-seven participants with HFrEF (60±8 years old and left ventricle ejection fraction of 41±6%) undertook a mean of 94±30 steps in the 6MST. Intra-rater reliability was excellent for 6MST (ICC=0.9), with mean error of 4.85 steps and superior and inferior limits of agreement of 30.6 and -20.9 steps, respectively. In addition, strong correlations between number of steps and CPET workload (r=0.76, P<0.01) and peak V̇O2 (r=0.71, P<0.01) were observed. From simple linear regression the following predictive equations were obtained with 6MST results: V̇O2 peak (mL/min) = 350.22 + (7.333 × number of steps), with R2=0.51, and peak workload (W) = 4.044 + (0.772 × number of steps), with R2=0.58. The 6MST was a reliable and valid tool to assess functional capacity in HFrEF participants and may moderately predict peak workload and oxygen uptake of a CPET.

RESUMO: O conhecimento das alterações fisiológicas que ocorrem quando os animais são submetidos a alterações climatológicas consideradas estressantes é fundamental para manter o bem-estar animal, e poder explorar o seu potencial reprodutivo de forma eficiente e racional. O presente estudo foi realizado com o objetivo de avaliar a influência de variáveis climáticas sobre parâmetros fisiológicos e de ejaculados de búfalos, da raça Murrah, criados em clima tropical úmido da Amazônia. As análises imediatas pertinentes às características físicas e morfológicas dos ejaculados foram realizadas e corresponderam no período chuvoso (PCh) o volume de 3,4±2,0mL, turbilhonamento de 4,4±0,5; motilidade de 80,4±5,6%; vigor de 4,4±0,4; concentração de 657.300±237.865,1 x 106sptz/mL; defeitos maiores de 9,0±2,6%; defeitos menores de 11,2±3,9%; defeitos totais de 20,2±5,3% e integridade da membrana plasmática (IMP) de 84,8±5,6%, enquanto que no período não chuvoso (PnCh), os resultados foram de 4,0±2,1mL; turbilhonamento de 3,0±1,0; motilidade de 56,2±13,4%; vigor de 3,0±1,0; concentração de 586.000±291.925,9 x 106sptz/mL; defeitos maiores de 20,8±9,9%; defeitos menores de 27,5±6,3%; defeitos totais de 48,3±9,3% e IMP de 57,9±12,4%. Observou-se diferença estatística (P<0,05) para os parâmetros movimento de massa, motilidade, vigor, defeitos maiores, defeitos menores, defeitos totais e integridade da membrana plasmática entre os dois períodos. Dados de frequência cardíaca, temperatura superficial (cabeça, dorso, virilha e bolsa escrotal) diferiram estatisticamente entre os períodos (P<0,05). Conclui-se que se faz necessário usar de um manejo específico no período não chuvoso para que o estresse térmico não seja um fator determinante na redução da qualidade reprodutiva dos búfalos, para isto se faz necessário utilizar de meios para melhorar o bem-estar animal, sendo uma das alternativas fazer uso de banhos regularmente para estes animais, ou disponibilizar acesso constante destes a áreas de rios ou lagos, assim como sombreamentos, evitando que os búfalos fiquem expostos diretamente ao ambiente térmico desfavorável.

ABSTRACT: Awareness of the physiological changes that occur when animals are subjected to climatic changes that are considered stressful is essential to maintain animal welfare and to be able to exploit their reproductive potential efficiently and rationally. The present study was carried out to evaluate climatic variables’ influence on physiological parameters, and Murrah buffalo ejaculates reared in a humid tropical climate in the Amazon. The immediate analyzes pertinent to the physical and morphological characteristics of the ejaculates were carried out and corresponded in the rainy season (RS) volume of 3.4±2.0mL; the mass activity of 4.4±0.5; motility of 80.4±5.6%; vigor of 4.4±0.4; concentration of 657,300±237,865.1 x 106sptz/mL; major defects of 9.0±2.6%; minor defects of 11.2±3.9%; total defects 20.2±5.3% and sperm plasma membrane integrity (SPMI) 84.8±5.6%, whereas in the non-rainy season (nRS), the results were 4.0±2.1mL; the mass activity of 3.0±1.0; motility of 56.2±13.4%; vigor of 3.0±1.0; concentration of 586,000±291,925.9 x 106sptz/mL; major defects of 20.8±9.9%; minor defects of 27.5±6.3%; total defects 48.3±9.3% and SPMI of 57.9±12.4%. Furthermore, a statistical difference (P<0.05) was observed for the parameters mass activity, motility, vigor, major defects, minor defects, total defects, and sperm plasma membrane integrity between both periods. The data on heart frequency, superficial temperature (head, back, groin, and scrotal pouch) showed a statistical difference between both periods (P<0.05). To conclude is necessary specific management in the non-rainy season that thermal stress is not a determining factor in reducing the reproductive quality of buffaloes; it is necessary to use means to improve animal welfare; one alternative is to use baths regularly for these animals or provide constant access to areas of rivers or lakes, as well as shading, preventing the buffaloes from being directly exposed to the unfavorable thermal environment.

RESUMO A fim de avaliar o efeito de diferentes doses da rbST sobre a dinâmica folicular, a produção e a maturação in vitro de oócitos, 20 vacas Sindi, divididas em três grupos, receberam um dispositivo de progesterona intravaginal, estradiol e PGF2α, além de 2mL de solução salina (grupo controle), 250 (grupo rbST 250) ou 500mg de rbST (grupo rbST 500). Cinco dias depois, realizou-se a ovum pick up, e os complexos cumulus-oócitos (CCOs) recuperados foram selecionados, classificados e maturados in vitro. Os dados de contagem foram comparados pelo procedimento glht (General Linear Hypothesis Test), e os dados em porcentagem foram submetidos ao qui-quadrado, no programa estatístico R, onde as diferenças foram consideradas significativas quando P<0,05. Não houve diferença (P>0,05) entre os grupos quanto à quantidade de folículos e à taxa de maturação. Os grupos rbST 250 e rbST 500 foram significativamente superiores (P<0,05) ao grupo controle em relação ao número de folículos grandes (0,42±0,20 vs. 0). O grupo rbST 500 apresentou maior (P<0,05) porcentagem de oócitos viáveis (91,52%) do que os grupos controle (67,85%) e rbST 250 (53,33%). A rbST aumenta o número de folículos grandes, e 500mg de rbST aumentam a porcentagem de oócitos viáveis em vacas Sindi.

ABSTRACT In order to evaluate the effect of different doses of rbST on the follicular dynamics, production, and in vitro maturation of oocytes, 20 Sindhi cows were divided into three groups, receiving an intravaginal progesterone device, estradiol and PGF2α, and 2mL of solution saline (Control Group), 250 (rbST 250 Group) or 500mg rbST (rbST 500 Group). Five days later, the ovum pick up was performed, and the cumulus-oocyte (CCO) complexes recovered were selected, classified, and matured in vitro. The counting data were compared by the glht (General Linear Hypothesis Test) procedure, and the percentage data were submitted to Qui- square, in the statistical program R, where differences were considered significant when P< 0.05. There was no difference (P> 0.05) between the groups regarding follicle quantity and maturation rate. The rbST 250 and rbST 500 groups were significantly higher (P< 0.05) than the Control group in relation to the number of large follicles (0.42±0.20 versus 0). The rbST 500 group presented higher (P< 0.05) percentage of viable oocytes (91.52%) than the Control (67.85%) and rbST 250 (53.33%) groups. rbST increases the number of large follicles and 500mg rbST increases the percentage of viable oocytes in Sindhi cows.
