RESUMO Introdução: Em função da urgência e demanda de uma resposta à pandemia do novo coronavírus (Covid-19), vários testes de detecção de anticorpos para a síndrome respiratória aguda grave do coronavírus 2 (Sars-CoV-2) têm sido desenvolvidos. Objetivo: Este estudo teve como objetivo avaliar o desempenho do teste rápido utilizado em um inquérito epidemiológico para Sars-CoV-2 em comparação com outros ensaios sorológicos. Métodos: Foram avaliadas 86 amostras de soro em três ensaios sorológicos: um imunoensaio de fluxo lateral - Wondfo Sars-CoV-2 Antibody Test (TRW) - e dois imunoensaios de quimioluminescência: Elecsys anti-Sars-CoV-2 (ECLIA) e Sars-CoV-2 IgG (CMIA-IgG). Resultados: As sensibilidades diagnósticas estimadas dos testes sorológicos na avaliação dessas amostras foram: TRW 59% [95% intervalo de confiança (IC) 43,4%-72,9%], ECLIA 66,7% (51%-79,4%) e CMIA-IgG 61,5% (47,1%-73%). Enquanto isso, a especificidade diagnóstica estimada para TRW foi 78,7% (95% CI 65,1%-88%), ECLIA 72,3% (58,2%-83,1%) e CMIA-IgG 76,6% (74%-95,5%). Os valores de sensibilidade e especificidade foram inferiores aos afirmados pelos fabricantes. Embora 16,2% (14/86) dos resultados tenham sido discordantes entre os três ensaios serológicos para Sars-CoV-2, o grau de concordância pelo índice Kappa foi adequado: TRW/CMIA-IgG [0,757 (95% IC 0,615-0,899)], TRW/ECLIA [0,715 (0,565-0,864)] e ECLIA/CMIA-IgG [0,858 (0,748-0,968)]. Conclusão: O teste sorológico pode ser uma ferramenta diagnóstica útil, o que reforça sua avaliação criteriosa, bem como o momento correto de sua utilização.

ABSTRACT Introduction: Due to urgency and demand of a response to the Covid-19 pandemic, numerous Sars-CoV-2 immunoassays have been rapidly developed. Objective: This study aimed at assessing the performance of rapid Sars-CoV-2 antibody test in comparison to high-throughput serological assays. Methods: A total of 86 serum samples were evaluated in the three assays: a lateral flow immunoassay - Wondfo Sars-CoV-2 Antibody Test (WRT) - and two chemiluminescence immunoassays: Elecsys Anti-Sars-CoV-2 (ECLIA), and Sars-CoV-2 IgG (CMIA-IgG). Results: The estimated diagnostic sensitivities of serological tests in the evaluation of serum samples from the epidemiological survey were: WRT 59% [95% confidence interval (CI) 43.4%-72.9%], ECLIA 66.7% (51%-79.4%), and CMIA-IgG 61.5% (47.1%-73%). Meanwhile, the estimated diagnostic specificity was for WRT 78.7% (95% CI 65.1%-88%), ECLIA 72.3% (58.2%-83.1%), and CMIA-IgG 76.6% (74%-95.5%). The sensitivity and specificity values were lower than manufacturers’ claimed. Although 16.2% (14/86) of serological results were discordant among the three Sars-CoV-2 serological assays, the degree of agreement by the kappa index was adequate: WRT/CMIA-IgG [0.757 (95% CI 0.615-0.899)], WRT/ECLIA [0.715 (0.565-0.864)], and ECLIA/CMIA-IgG [0.858 (0.748-0.968)]. Conclusion: The serological testing may be a useful diagnostic tool, which reinforces its careful evaluation, and, as well as the correct time to use it.

RESUMEN Introducción: Debido a la urgencia y la demanda de una respuesta a la pandemia de Covid-19, se han desarrollado rápidamente numerosos inmunoensayos del Sars-CoV-2. Objetivo: Este estudio tuvo como objetivo evaluar el rendimiento de la prueba rápida de anticuerpos contra el Sars-CoV-2 en comparación con los ensayos serológicos de alto rendimiento. Métodos: Se evaluaron un total de 86 muestras de suero en los tres ensayos: un inmunoensayo de flujo lateral - Wondfo Sars-CoV-2 Antibody Test (TRW) - y dos inmunoensayos de quimioluminiscencia: Elecsys Anti-Sars-CoV-2 (ECLIA) y Sars-CoV-2 IgG (CMIA-IgG). Resultados: Las sensibilidades diagnósticas estimadas de las pruebas serológicas en la evaluación de muestras de suero de la encuesta epidemiológica fueron: WRT 59% [intervalo de confianza (IC) del 95%: 43,4%-72,9%], ECLIA 66,7% (51%-79,4%) y CMIA-IgG 61,5% (47,1%-73%). Mientras tanto, la especificidad diagnóstica estimada fue para WRT 78,7% (95% CI 65,1%-88%), ECLIA 72,3% (58,2%-83,1%) y CMIA-IgG 76,6% (74%-95,5%). Los valores de sensibilidad y especificidad fueron más bajos que los declarados por los fabricantes. Conclusión: La prueba serológica puede ser una herramienta diagnóstica útil, lo que refuerza su evaluación cuidadosa, así como el momento adecuado para usarla.

The present work aims to prepare and characterize very thin films of poly(fullerene)s in order to investigate their electrical properties and the influence of xylene and chloroform solvents on these materials. The fullerenes studied were phenyl-C61-butyric acid methyl ester (PCBM), oligo{(phenyl-C61-butyric acid methyl ester)-alt-[1,4-bis(bromomethyl)-2,5-bis(octyloxy)benzene]} (OPCBMMB) and poly{[bispyrrolidino(phenyl-C61-butyric acid methyl ester)]-alt-[2,5-bis(octyloxy) benzene]} (PPCBMB), along with poly(3-hexylthiophene) (P3HT). The Langmuir-Schaeffer technique was used to prepare films, which were deposited on interdigitated gold substrates, and electrically characterized, with emphasis on the study of transport, conductivity and mobility mechanisms with respect to the solvents used. We found that the addition of P3HT significantly increased the conductivity of these materials. The xylene cast PPCBM, in both pure and mixed forms under dark conditions, presented the best conductivity results with respect to the other materials. However, when chloroform was used, it was found that OPCBMMB in both pure and mixed forms under light, exhibited the best conductivities. This is the first treatment, to our knowledge, of the impact of solvents on the electronic properties of poly(fullerene)s.

Ferulic acid (FERH) is known to exert leishmanicidal action against Leishmania (Leishmania) amazonensis. In this study, the ferulate anion (FER–) was intercalated into a layered double hydroxide (LDH) for FER– delivery during an antileishmania in vitro test. The LDH-FER– intercalation compound was prepared by coprecipitation and was characterized by X-ray powder diffraction, thermogravimetry, UV-Vis, infrared, and Raman spectroscopy analyses. Leishmania (L.) amazonensisin vitro assays were conducted to assess the LDH-FER– potential. LDH-FER– was found to promote a dose-dependent reduction of L. (L.) amazonensis amastigotes (intracellular form, half maximal inhibitory concentration (IC50) = 1.78 µg mL-1) without exerting a deleterious effect on the host macrophages (medium cytotoxic concentration (CC50) = 212.61 µg mL-1 and selectivity index = 119.4). LDH-FER– was found to be an effective inhibitor of the amastigote form of L. (L.) amazonensis, suggesting that LDH-FER– was able to deliver FER– inside the parasitophorous vacuoles where the pH is more acidic.

The challenge of the present work concerned the development and validation of high performance liquid chromatography (HPLC) and UV-Vis spectrophotometric methods for quantitation of methotrexate (MTX) loaded on biodegradable microparticles, composed of copolymers with different solubilities such as chitosan and poly (lactic-co-glycolic acid) (PLGA). The linearity of the analytical curves for MTX were > 0.999 (r2) and the limits of detection (LOD) were 0.014 µg mL-1 and 0.060 µg mL-1 for HPLC and UV-Vis spectrophotometric method, respectively. In addition, both methods were specific, robust, and accurate with a recovery between 89.5% and 105.5%. The method showed precision with relative standard deviations lower than 3.39% for HPLC and 2.90% for UV-Vis spectrophotometry. Statistical analysis revealed that both methods provide equivalent results, and also can be used for quality control of MTX-loaded in drug delivery systems.

A espectrometria de absorção atômica em forno de grafite de alta resolução com fonte contínua (HR-CS GF AAS) é proposta para determinar Pb em sombra de olho e blush empregando a análise direta de sólidos. O emprego da mistura de modificadores químicos Pd(NO3)2 + Mg(NO3)2 possibilitou a calibração aquosa de 0,25 a 2,5 ng Pb (r = 0,998). O desempenho do método proposto foi avaliado por meio da análise de digeridos de sombra e de blush por espectrometria de absorção atômica em forno de grafite com fonte de linhas (LS GF AAS) como técnica comparativa. Os resultados obtidos por HR-CS GF AAS e LS GF AAS foram concordantes entre si a um nível de confiança de 95% (teste t pareado). O limite de quantificação (massa seca) foi de 0,020 ng mg–1. A concentração de Pb nas amostras de sombra de olho e de blush variou de 1,222 a 9,632 ng mg–1 e 0,362 a 28,091 ng mg–1, respectivamente.

Direct solid sampling is proposed for Pb determination in eye shadow and blush samples by high-resolution continuum source graphite furnace atomic absorption spectrometry (HR-CS GF AAS). A mixture of Pd(NO3)2 + Mg(NO3)2 was employed as chemical modifier, and aqueous calibration (0.25-2.5 ng Pb, r = 0.998) was obtained. Accuracy of the determination of Pb in eye shadow and blush samples by the proposed method was verified by line source graphite furnace AAS as a comparative technique employing digested samples. The results obtained by the two methods were in agreement at a 95% confidence level (paired t-test). The limit of quantification (dry mass) was 0.020 ng mg–1. The Pb content in the eye shadow and blush samples varied between 1.222 and 9.632 ng mg–1 and 0.362 and 28.091 ng mg–1, respectively.

Estudos neurossonológicos, especialmente o Doppler transcraniano (DTC) e o duplex transcraniano codificado a cores (DTCC), apresentam elevados níveis de especificidade e sensibilidade quando utilizados como exames complementares no diagnóstico de morte encefálica (ME). Um grupo de peritos do Departamento Científico de Doppler transcraniano da Academia Brasileira de Neurologia criou uma força-tarefa de forma a determinar os critérios neurossonológicos para os seguintes aspectos no diagnóstico de ME no Brasil: metodologia do DTC; metodologia do DTCC; treinamento e habilidades em Neurossonologia; diagnóstico de parada circulatória encefálica e documentação do exame para a ME. Os resultados deste encontro foram apresentados neste artigo.

Neurosonological studies, specifically transcranial Doppler (TCD) and transcranial color-coded duplex (TCCD), have high level of specificity and sensitivity and they are used as complementary tests for the diagnosis of brain death (BD). A group of experts, from the Neurosonology Department of the Brazilian Academy of Neurology, created a task force to determine the criteria for the following aspects of diagnosing BD in Brazil: the reliability of TCD methodology; the reliability of TCCD methodology; neurosonology training and skills; the diagnosis of encephalic circulatory arrest; and exam documentation for BD. The results of this meeting are presented in the current paper.
