In vivo embryo development in bitches inseminated laparoscopically after ovulation time estimated based on a single progesterone determination

Abstract Logistic and economical limitations are often the causes of dog owners not accurately monitoring the estrous cycle and the optimal insemination time. The aim of this study was to evaluate in vivo early embryonic development in bitches, after the analysis of sequential vaginal cytologies associated to single progesterone measurement and single laparoscopic insemination with high quality semen (fresh and with high spermatozoa concentration) or low-quality semen (frozen/thawed and with low spermatozoa concentration) at 48 h post- ovulation time predicted on a single progesterone measurement. Ten bitches were inseminated with 250 x 106 fresh spermatozoa (80% motility), and ten with 80 x 106 frozen/thawed spermatozoa (60% motility) in the cranial part of each uterine horn. Seven days later, ovariohysterectomy was performed and the oviducts and uterine horns and body were flushed to recover embryos and unfertilized oocytes. In 80% of the bitches inseminated with fresh and 50% of bitches inseminated with frozen/thawed semen, embryos at 2 to 8 cells stage were recovered mostly from the, oviducts. This study indicates that pregnancies can be obtained with a single laparoscopic intrauterine insemination after single serum progesterone measurement, although with a low number of embryos. This result should be taken into account in case economic or logistic restrictions that affect the possibility of owners to plan an accurate monitoring of the optimal breeding time using fresh and frozen semen.

Effect of assisted calving on uterine puerperal disorders occurrence, and on reproductive efficiency in crossbred dairy cows

Abstract The aim of this study was to evaluate the effect of the type of calving (normal or assisted) on the occurrence of puerperal uterine disorders and on the number of artificial inseminations (AI) per conception and pregnancy rate at 150 days postpartum (PPD). Cows were observed during parturition and the type of calving was classified as normal or assisted. Normal calving required no human interference, while assisted calving required a calf removal aid via vaginal access. There were 825 births, 7 stillbirths (0.85%) and 17 twins (2.06%). We analyzed 801 single births, from which 766 (95.63%) were normal and 35 (4.36%) assisted. Were evaluated the uterine disorders occurrence such as retained placenta combined with metritis (PR/ME) and clinical endometritis (CE). The overall occurrences were 10.24% (82/801) of RP/ME, 12.86% for CE and 5.12% for RP/ME and CE. Most of normal calving cows had healthy puerperium (73.89%), whereas a majority of assisted calving cows had uterine disorders (74.29%). The number of AI per conception was similar for cows that had either normal or assisted calving (2.39±0.08 and 3.00±0.43, P = 0.16). There was no evidence of negative influence of calving type on the 150 PPD pregnancy rate of lactating crossbreed dairy cows (P = 0.44). Healthy cows had higher 150 PPD pregnancy rate than cows affected by puerperium uterine disorders (51.65 vs. 42.92%). Normal calving crossbred dairy cows had a healthier puerperium, compared to cows with assisted calving which were more susceptible to puerperium uterine disorders. And cows with a health puerperium have a higher pregnancy rate at 150 days postpartum.
Resumo Objetivou-se avaliar em vacas leiteiras mestiças que pariram um bezerro vivo, o efeito do tipo de parto (normal ou assistido) na ocorrência de desordens uterinas puerperais e no número de inseminações artificiais (IA) por concepção e taxa de prenhez 150 dias pós-parto (DPP). As vacas foram monitoradas durante o parto e o tipo de parto foi classificado como normal ou assistido. O parto normal não precisou de interferência humana, enquanto o parto assistido precisou de auxílio para retirada do bezerro por via vaginal. Foram registrados 825 partos, sendo 7 natimortos (0,85%) e 17 gemelares (2,06%). Foram analisados 801 partos simples, dos quais 766 (95,63%) foram normais e 35 (4,36%) foram assistidos. Foram avaliadas a ocorrência de desordens uterinas como a retenção de placenta associada com metrite (RP/ME) e endometrite clínica (EC). A ocorrência das doenças foi de 10,24% (82/801) para RP/ME, 12,86% para EC e 5.12% para RP/ME com CE. A maioria das vacas com parto normal tiveram puerpério saudável (73,89%), enquanto a maioria das vacas com parto assistido apresentaram desordens uterinas (74,29%). O número de IA por concepção foi semelhante nas vacas que tiveram parto normal ou assistido (2,39±0,08 e 3,00±0,43, P = 0,16). Não houve evidência de influência negativa do tipo de parto na taxa de prenhez 150 DPP de vacas leiteiras mestiças em lactação (P = 0,44). Vacas saudáveis apresentaram maior taxa de prenhez 150 DPP do que vacas afetadas por desordens uterinas no puerpério (51,65 vs. 42,92%). Vacas leiteiras mestiças com parto normal tiveram um puerpério saudável, comparadas às que tiveram parto assistido, por sua vez foram mais susceptíveis as desordens uterinas no puerpério. E vacas com puerpério saudável resultaram em maiores taxas de prenhez aos 150 dias pós parto.

Application of PGF2α at the moment of fixed-time artificial insemination in crossbred beef cows

Abstract Although studies have shown positive effects of gonadotropin releasing hormone (GnRH) or prostaglandin F2α (PGF2α) at the moment of fixed-time artificial insemination (FTAI) in the conception rate (CR) of cattle, its effects on treatments based on progesterone (P4) and estradiol benzoate (EB) is still not conclusive. The objective of this study was (1) to evaluate the effect of a PGF2α analogue at FTAI in the CR of crossbred beef cows submitted to a 11d FTAI protocol based on P4 and EB; and (2) to describe the CR between PGF2α-treated and control cows in different body condition scores (BCS) and parity categories. Crossbred (½ Nellore and ½ Angus) beef cows were submitted to a synchronization protocol and randomly assigned into 2 groups: Control (n = 163), at FTAI cows received 2 mL of saline solution as a placebo, and PGF2α (n = 163), at FTAI cows were treated with PGF2α analogue (10 mg of dinoprost tromethamine). Pregnancy diagnosis was performed 33d post-FTAI. Binary logistic regression model was used to analyze the effect of PGF2α treatment on CR. There was no difference in CR between PGF2α and control groups (P > 0.05; odds ratio (OR) = 0.92; confidence interval (CI) = 0.59-1.4). A greater CR was found in heifers (P = 0.0006, OR = 2.65, CI = 1.61 - 4.38) and multiparous (P = 0.0006, OR = 2.12, CI = 1.04 - 4.3) when compared to primiparous cows. Cows with low BCS (4; 9-point scale) showed lower CR when compared with moderate BCS (5-6; 9-point scale) (P < 0.05; OR = 0.10; CI = 0.06 - 0.18). There was no numerical difference on CR between PGF2α-treated and control cows in different BCS and parity categories. The results suggested that the CR in this study was not influenced by 10 mg PGF2α analogue at FTAI.

Influence of gestation length, seasonality, and calf sex on birth weight and placental retention in crossbred dairy cows

Resumo Objetivou-se avaliar a influência da duração da gestação, da estação do ano ao parto (outono-inverno vs. primavera-verão) e do sexo do bezerro sobre o peso ao nascimento, bem como os efeitos destas variáveis sobre a incidência de retenção de placenta (RP) em vacas leiteiras mestiças. Foram avaliados 187 partos em uma fazenda comercial leiteira na região do Alto Paranaíba, Minas Gerais, Brasil. Vacas que não liberaram os anexos placentários nas primeiras 12 horas após a expulsão do feto foram diagnosticadas com RP. Ao parto, o sexo da cria foi registrado e o peso ao nascer foi obtido por meio da fita de pesagem. A incidência de RP foi de 36,89% (69/187). O peso médio ao nascer dos bezerros (41,73 kg) não diferiu entre sexo e estação do ano ao nascimento (P > 0,05), porém vacas cuja gestação teve duração abaixo da média (274,4 dias) pariram bezerros mais leves. A estação do ano ao parto e o sexo da cria também não influenciaram a incidência de RP (P > 0,05), no entanto uma maior incidência de RP foi observada em vacas que apresentaram gestações de menor duração (49,44%) e cujas crias tiveram peso ao nascer abaixo da média (51,76%). Conclui-se que vacas com período de gestação curto comprometem o peso ao nascer dos bezerros e são mais susceptíveis à desenvolver retenção de placenta.
Abstract The objective of this study was to evaluate the influence of gestation length, season at calving (autumn-winter vs. spring-summer) and calf sex on birth weight and incidence of retained placenta (RP) in crossbred lactating dairy cows. A total of 187 parturitions were evaluated in a commercial dairy farm at Alto Paranaiba region, Minas Gerais, Brazil. Cows that did not release the placental membranes within the first 12 hours after calf expulsion were diagnosed with RP. At birth, calf sex was registered, and birth weight was estimated by using a weighing tape. The incidence of RP was 36.89% (69/187). The mean birth weight of the calves (41.73 kg) did not differ among the calf sexes and seasons of the year at birth (P > 0.05), but the cows with below-average gestation lengths (274.4 days) calved lighter calves. Furthermore, the season of the year at calving and calf sex had no influence on RP incidence (P > 0.05), although a higher incidence of RP was observed in cows that had shorter gestation lengths (49.44%) and calves below-average birth weights (51.76%). In conclusion, cows with shorter gestation lengths produce lighter offspring and have a high predisposition to RP.

Abstract Genomic evaluations have revolutionized dairy cattle breeding, and the demand for embryos produced from very young heifers with high genetic merit has increased over time. The combination of low oocyte recovery, young age of donors, and milk production status can make the in vitro embryo production (IVP) of Holstein cattle incredibly challenging. Several factors need to be coordinated to obtain a live calf from an IVP embryo, but the quality of the oocyte at the start of the process is one of the key factors. Aspects related to oocyte quality, laboratory quality control, embryo quality and recipient selection are addressed here, based on the measures that the RuAnn Genetics Laboratory (Riverdale, California, USA) adopted in the last 12 years, with the goal of improving production of live, healthy calves from Holstein embryos. Follicular wave synchronization and stimulation with follicular stimulating hormone (FSH) is necessary to improve oocyte quality and consequently embryo production. Laboratory quality control and the use of high-quality supplies are essential to reduce variability in production and facilitate identification of other factors that might interfere with embryo production. High pregnancy rates can be achieved with good quality embryos selected at optimal time and stage of development, transferred by an experienced embryo transfer (ET) technician, to well managed recipients 7 or 8 days after estrus. Attention to detail at every step of the process is crucial to success.

Croton sapiifolius Müll.Arg. In: a new occurrence for the State of Espírito Santo, Brazil

RESUMO Durante visita aos herbários do Estado do Espírito Santo, foram encontrados espécimes de C. sapiifolius para este Estado, uma espécie conhecida até o momento apenas para o Estado da Bahia. Neste trabalho são apresentados dados atuais sobre a taxonomia e distribuição geográfica de C. sapiifolius. Essa nova ocorrência eleva para 20 o número de espécies conhecidas de Croton no Estado do Espírito Santo. O presente trabalho contribui para enriquecer o conhecimento sobre as espécies de Croton na Floresta Atlântica e no Brasil.
ABSTRACT While studying Croton material collected in Espírito Santo, we found several specimens of Croton sapiifolius, a species previously restricted to the State of Bahia. The present paper updates our knowledge about the taxonomy, habitat and geographic distribution of C. sapiifolius. This finding increases the number of known Croton species occurring in Espírito Santo State to 20 and contributes to the knowledge of Croton of the Brazilian Atlantic Forest.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Inseminação artificial em tempo fixo e diagnóstico precoce de gestação em vacas leiteiras mestiças

Avaliou-se, durante um ano, o desempenho reprodutivo de 94 vacas leiteiras mestiças Bos taurus x Bos indicus submetidas a um programa de reprodução assistida. Um protocolo de inseminação artificial em tempo fixo (IATF) foi executado por meio de dispositivo intravaginal contendo progesterona e das injeções de prostaglandina F2α e de cipionato de estradiol. Por meio de ultrassonografia, entre 7 e 14 dias após as inseminações ou montas controladas, realizou-se a detecção de corpo lúteo nos ovários a fim de determinar a taxa de ovulação e, no 28º dia, fez-se o diagnóstico de gestação para cálculo da taxa de concepção. Respeitou-se um período mínimo de 34 dias após o parto antes do tratamento. Não houve influência do escore de condição corporal e da presença de corpo lúteo no início do protocolo, nem da reutilização do dispositivo intravaginal e da monta controlada ou inseminação artificial, sobre as taxas de ovulação, concepção e concepção das vacas ovuladas. As taxas de concepção e de concepção das vacas ovuladas foram afetadas negativamente pelo elevado número de dias pós-parto (DPP), ou dias em lactação e pela época quente do ano, primavera/verão. A resposta ao protocolo de inseminação artificial em tempo fixo baseado no uso de progesterona, PGF2α e cipionato de estradiol é prejudicada pelo aumento dos dias em lactação e pela época quente do ano. A condição corporal não afeta a resposta ao protocolo de inseminação artificial, desde que as vacas tratadas apresentem escore acima de 2,25 pontos.
It was evaluated, during a period of one year, the reproductive performance of 94 Bos taurus x Bos indicus crossbred dairy cows submitted to an assisted reproduction program. A timed artificial insemination (TAI) protocol was carried out by using an intra-vaginal progesterone device containing progesterone and through injections with Prostaglandin F2α and estradiol cypionate. By using ultrasound, from 7 to 14 days after inseminations or controlled mattings, detection of corpus luteum in the ovaries was done to determine ovulation rate and, on the 28th day, diagnosis of pregnancy was carried out for calculation of conception rate. Treatment only started after a 34-day post-partum minimal period. Body condition score, presence of corpus luteum in the beginning of the protocol, reutilization of intra-vaginal device and controlled mating or artificial insemination did not influence rates of ovulation, conception and conception of ovulated cows. Rates of conception and rates of conception of ovulated cows were negatively affected by the higher number of days post partum (DPP), or days in milk and by the warmer seasons of the year, spring/summer. Response to protocol of timed artificial insemination based on the use of progesterone, PGF2α and estradiol cypionate is negatively affected by lactation days and by the warm season of the year. Body condition does not affect the response to protocol of artificial insemination provided that treated cows show body condition scores higher than 2.5 points.
