Avaliaram-se as alterações histológicas do tecido laminar, obtido por biopsia, em 20 equinos portadores de laminite induzida por sobrecarga de carboidratos e tratados com ketoprofeno, fenilbutazona ou flunixin meglumine. A biopsia foi colhida dos dígitos torácicos 72 horas após a indução. Os achados histológicos foram comparados com os achados de amostras de equinos isentos de laminite. Infiltrado inflamatório neutrofílico foi observado em 80%, congestão em 50%, hemorragia em 35% e hiperplasia na túnica íntima das arteríolas das lâminas dérmicas primárias em 15% das amostras. As taxas de microtrombos e coágulos foram 15% e 20%, respectivamente. Estes achados parecem decorrer dos distúrbios circulatórios que ocasionaram edema, congestão e hiperemia, seguidos de degeneração. Em 70% das análises realizadas nos animais tratados, as lesões histológicas foram inferiores aos graus de claudicação observados. Conclui-se que a biopsia de tecido laminar digital de equinos é viável, os artefatos decorrentes da técnica de biopsia não prejudicam a análise histológica das amostras e os anti-inflamatórios não esteroidais não são capazes de evitar as lesões laminares quando administrados após o início da sintomatologia clínica de laminite.

Experimental laminitis caused by carbohydrate overload was induced in 20 healthy horses. Seventy two hours after induction, samples of the laminar tissue were obtained by biopsy from the thoracic limbs digits for histopathology. The histological findings were compared to samples from horses without laminitis. Neutrophilic infiltrate was observed in 80% of the samples, congestion in 50%, hemorrhage in 35%, and hyperplasia of the arteriolar intima layer of the primary dermal lamina in 15%. Thrombi and intravascular blood clots were observed in 15% and 20% of the samples, respectively. Apparently, these findings were due to circulatory changes that resulted in edema, congestion, and hyperemia, followed by degeneration. In 70% of analyses performed on treated horses, the histological lesions were less severe than the clinical signs of lameness. It is concluded that: (i) the biopsy technique of laminar digital tissue from horses is viable; (ii) the artifacts generated by the biopsy technique do not compromise the histological analyses; and (iii) non-steroidal anti-inflammatory drugs do not avoid laminar lesions when administered after the beginning of clinical signs of laminitis.

RESUMO A pesquisa objetivou analisar, morfológica e quantitativamente, os tipos de hemócitos em lagartas do terceiro ínstar de Diatraea saccharalis F., desafiadas imunologicamente por Metarhizium anisopliae (Metsch.) Sorok. e Beauveria bassiana (Bals.) Vuill., nas concentrações 103, 105 e 107 conídios mL-1, em comparação a lagartas não tratadas. Foram coletados 10 µL de hemolinfa de cinco lagartas por tratamento, nos intervalos de 24, 36, 48, e 60 horas pós-inoculação, sendo contadas 300 células em campos aleatórios da lâmina, utilizando-se a objetiva de imersão (100X). Os hemócitos mais freqüentes foram os esferulócitos (34,8%), plasmatócitos (29,7%) e granulócitos (22,2%), enquanto os menos freqüentes foram os prohemócitos (8,3%), adipohemócitos (2,6%) e oenocitóides (2,4%). B. bassiana promoveu interferência efetiva na população de granulócitos na concentração de 107 conídios mL-1 (24 e 36h) e plasmatócitos nas concentrações 103 conídios mL-1 (60h) e 107 conídios mL-1 (24 e 36h), onde ocorreu aumento do número de granulócitos e redução do número de plasmatócitos na maior concentração e, ainda, o aumento do número de plasmatócitos na concentração 103 conídios mL-1. M. anisopliae não interferiu quantitativamente nos granulócitos e plasmatócitos. No que se refere aos esferulócitos, verificou-se maior tendência de redução do número dessas células no intervalo de 36h para ambos os fungos, porém, diferença significativa só ocorreu na concentração de 105 conídios mL-1 no intervalo de 60h, apresentando a menor média. Conclui-se que B. bassiana tem ação mais efetiva sobre a dinâmica populacional dos granulócitos e plasmatócitos em lagartas de D. saccharalis em relação ao M. anisopliae.

ABSTRACT This study was aimed to morphologically and quantitatively analyze the types of hemocytes in Diatraea saccharalis F. thirdinstar caterpillar immunologicaly challenged by Metarhizium anisopliae (Metsch.) Sorok. and Beauveria bassiana (Bals.) Vuill. in concentrations of 103, 105 and 107 conidia mL-1, compared to nontreated specimens. A 10µL-sample of hemolymphs from five caterpillars for each treatment, in the intervals of 24, 36, 48 and 60h; 300 cells were counted in random fields of the lamina, using the immersion objective (100X). The most frequent hemocytes were the spherulocytes (34.8%), plasmatocytes (29.7%) and granulocytes (22.2%), while the least frequent were the prohemocytes (8.3%), the adipohemocytes (2.6%) and the oenocytoids (2.4%). B. bassiana promoted effective interference in the population of granulocytes at the concentration of 107 conidia mL-1 (24 and 36h) and of plasmatocytes at the concentrations of 103 conidia mL-1 (60h) and 107 conidia mL-1 (24 and 36h); there occurred an increase of the number of the granulocytes and a decrease of the number of plasmotocytes at the highest concentration, and an increase in the number of plasmatocytes at the concentration of 103 conidia mL-1. M. anisopliae do not interfere quantitatively in the granulocytes and plasmatocytes. Regarding the spherulocytes, there was observed a higher tendency of reduction in the number of these cells in the interval of 24 and 36h for both fungi, but a meaningful difference only occurred at the concentration of 105 conidia mL-1 in the 60h interval, presenting the smallest average. It was possible to conclude that B. bassiana has a more effective action over the populational dynamics of the granulocytes and plasmatocytes of D. saccharalis caterpillars in association with M. anisopliae.

RESUMO A pesquisa teve o objetivo de descrever a histologia dos ovários, ovidutos laterais, oviduto comum e espermateca, e realizar a morfometria dos ovários de Tropidacris collaris (Stoll, 1813) (Orthoptera: Romaleidae), submetido aos fotoperíodos de 10L:14E, 12L:12E e 14L:10E. Para morfometria utilizou-se uma lupa binocular adaptada com uma ocular milimétrica, sendo registrado o comprimento e as larguras dos ovários. Posteriormente, os ovaríolos foram separados e contados. Para análise histológica, os órgãos foram fixados em Boüin alcoólico, incluídos em “paraplast” e corados pela H.E., tricrômico de Mallory e P.A.S (Ácido Periódico de Schiff). Os resultados mostraram que não houve influência dos fotoperíodos sobre a morfometria dos ovários, número de ovaríolos e histologia dos órgãos. Foram encontrados ovários préreprodutivos e reprodutivos. As médias do número de ovaríolos foram de 195,62, 202,62 e 208,25 para os fotoperíodos de 10L:14E, 12L:12E e 14L:10E, respectivamente, sendo mais numerosos no ovário esquerdo. Cada ovaríolo apresentou morfologia tubular com regiões bem distintas (filamento terminal, germário e vitelário). O oviduto lateral é revestido internamente por tecido epitelial simples cúbico com numerosas dobras, apoiado no tecido conjuntivo e externamente por uma camada de tecido muscular estriado. O oviduto comum apresenta a mesma constituição histológica do oviduto lateral, exceto pela presença de epitélio com íntima cuticular e uma camada muscular bem desenvolvida. A espermateca é constituída por tecido epitelial pseudo-estratificado colunar com íntima cuticular e tecido muscular estriado associado a tecido conjuntivo.

ABSTRACT The research aimed to study the histology of ovaries, lateral oviducts, common oviducts and spermatheca, and morphometry of the ovaries of Tropidacris collaris (Stoll, 1813) (Orthoptera: Romaleidae), reared under the photoperiods 10:14, 12:12 and 14:10 (L:D). Morphometry was conducted using a binocular magnifying glass adapted with a milimetric ocular, being registered the length and the width of the ovaries. Subsequently the ovarioles were separated and counted. For the histological analysis, the organs were fixated in alcoholic Boüin, included in “paraplast” and stained by H.E., Mallory’s trichromic and P.A.S. Photoperiods studied did not influence the morphometry of the ovaries, number of ovarioles and the histology of the organs. The averages of the ovarioles number were of 195,62, 202,62 and 208,25 for the fotoperíodos of 10L:14D, 12L:12D and 14L:10D, respectively, being more numerous in the left-side ovary. Each ovarioles presented tubular morphology with distinct regions (terminal filament, germarium and vitellarium). The lateral oviducts are internally covered by simple cubic epithelial tissue with numerous folds, sustained on the connective tissue and, externally, by a striated muscular tissue layer. The common oviduct presents the same histological constitution of the lateral oviduct, except by the presence of the epithelium with cuticular intima and a well developed muscular layer. The spermatheca is constituted by columnar pseudo-stratified epithelial tissue with cuticular intima and striated muscular tissue associated to connective tissue.
