Resumo Um dos textos da série “Biodiversidade do Estado de São Paulo”, dentro do Programa FAPESP-Biota, foi dedicado à Infraestrutura para Conservação da Biodiversidade, com foco nas coleções biológicas e nas unidades de conservação. Do início dos anos 60, quando a FAPESP foi criada, até os dias atuais muito foi investido em pesquisa nas mais diversas áreas, incluindo a preservação da biodiversidade do patrimônio genético nacional. Uma história de longos anos de avanços no conhecimento científico foi construída, a qual pode ser retratada através dos projetos que resultaram em dados de alta qualidade com impacto nacional e internacional. As coleções microbiológicas são centros geradores de tecnologia e recursos humanos especializados, que atuam (dentre outros) como repositórios vivos, preservando material de referência, e como testemunho da história da biodiversidade microbiana, preservando o que pode não mais existir. Possuem enorme potencial para alavancar a bioeconomia global e tratar de problemas que resultaram do mau uso dos recursos naturais. Essa leitura traz a todos o histórico, os avanços e as perspectivas futuras das coleções de microrganismos, dentro dos esforços de 60 anos de atividades científicas no Brasil.

Abstract One of the texts in the “Biodiversity in the State of São Paulo” series, within the FAPESP-Biota Program, was dedicated to the Infrastructure for Biodiversity Conservation, with a focus on Biological Collections and Conservation Units. From the early 1960s, when FAPESP was established, to the present day, financial resources have been invested in the preservation of the biodiversity of the national genetic heritage, besides other fields. History of years of advances in scientific knowledge was built, which can be portrayed through the projects that resulted in high-quality data of national and international impact. Microbiological collections are centers that generate technology and specialized human resources, and act (among other things) as living repositories preserving reference material and as witnesses to the history of microbial biodiversity because they preserve what may no longer exist. They have enormous potential to promote the global bioeconomy and address problems that have resulted from the misuse of natural resources. This reading brings everyone the history, advances, and future perspectives of culture collections, within the efforts of 60-year scientific activities in Brazil.

Os índices séricos de glicose e lipídios, a microbiota intestinal e a produção de ácidos graxos voláteis de cadeias curtas (AGV) foram determinados em ratos Wistar submetidos às dietas: padrão (AIN-P), padrão modificada (AIN-M) e às dietas contendo frações de parede celular de levedura: glicana insolúvel (GI), manana (M) e glicana mais manana (G+M), como única fonte de fibra alimentar. O fracionamento da parede celular (PC) foi realizado por processos físicos e químicos de extração, centrifugação e secagem em "spray dryer". Os índices séricos foram dosados através de "kits" comerciais. A microbiota e a produção de AGV foram determinadas nos conteúdos intestinais, incluindo cólon, ceco e reto. Considerando os níveis de colesterol no tempo (T0) e no tempo 28 (T28), as dietas AIN-P, AIN-M e M apresentaram efeito hipocolesterolêmico, tendo em vista que a composição das dietas eram de natureza hipercolesterolêmica. Em relação à glicose sérica, no tempo (T0) observou-se uma elevação geral da glicemia, sugerindo um efeito hiperglicêmico das dietas estudadas. A dieta G+M foi a que apresentou valores significantemente mais elevados de lipídios séricos no tempo T14, e os níveis mais baixos foram observados na dieta M e na dieta GI no T14 e nas dietas AIN-M e AIN-P. A dieta AIN-P foi a que apresentou valor significantemente mais elevado de triacilgliceróis nos tempos T14 e T28. Os níveis mais baixos nos tempos T14 foram constatados para as dietas G+M e GI e no tempo T28 para as dietas AIN-M e M. De um modo geral, não houve modificações significativas na microbiota intestinal dos animais em nenhuma das dietas. Dentre os AGV, o ácido acético foi o predominante, seguido do propiônico e do butírico, em todas as dietas estudadas.

The blood serum indexes of glucose and lipids, the intestinal microbiota and the production of volatile fatty acids (VFA) were determined in Wistar rats which were fed a standard (AIN-P) diet, a modified standard (AIN-M) diet, and diets containing the following yeast cell wall fractions: insoluble glycan (IG), mannan (M), and a mixture of glycan + mannan (G+M) as the only source of dietary fiber. Fractionation of the cell wall was carried out by means of physicochemical extraction processes, centrifugation, and drying in a "spray dryer". Serum indexes were dosed by means of commercial kits and the influence of the fractions on the intestinal microbiota and the production of short-chain volatile fatty acids (VFA) were carried out by collecting intestinal contents (colon, cecum, and rectum). Considering the cholesterol levels at time zero (T0) and at time 28 (T28), diets AIN-P, AIN-M, and M presented a hypocholesterolemic effect considering the hypercholesterolemic nature of the diets. Regarding serum glucose, at the onset of the experiment (T0), a general increase in glycemia was observed which suggests a hyperglycemic effect of the examined diets. The G+M diet yielded the highest significant values of serum lipids at T14 and the lowest values were found in the M diet and the IG diet at T14 and in the AIN-M and AIN-P diets. The AIN-P diet yielded the highest significant values of triacylglycerols at T14 and T28. The lowest levels at T14 were found in the G+M and IG diets and at T28 for the AIN-M and M diets. In general, there were no significant modifications in the intestinal microbiota of the rats in any diet. Regarding the VFA, the acetic acid was predominant, followed by the propionic and butyric acids in all the tested diets.

Foi feito um estudo da ocorrência de E.coli O157:H7 em vegetais que são normalmente consumidos crus no Brasil e uma avaliação da sua resistência aos sanitizantes disponíveis no mercado para desinfecção de verduras, equipamentos e utensílios, incluindo compostos clorados e compostos de amônio quaternário. Na avaliação da ocorrência em vegetais foram analisadas 869 amostras, não sendo detectada a presença do patógeno. Os imunoensaios enzimáticos (ELISA) utilizados nas análises (Reveal E.coli O157 Neogem e EHEC Test Kit 3M Company) apresentaram uma taxa de falsos resultados presuntivos de 13,6 e 11,8%, respectivamente, não confirmados como E.coli O157 nos testes bioquímicos posteriores. Na avaliação da resistência aos sanitizantes pelo método 960.9 da AOAC, observou-se que os tratamentos com 100 e 200ppm de hipoclorito de sódio, dicloroisocianurato de sódio e cloreto de benzalcônio/30s se mostraram eficazes contra E.coli ATCC 11229 e E.coli O157:H7 ATCC 43890, promovendo mais de 5 reduções decimais nas populações alvo.

A study of E.coli O157:H7 occurrence in vegetables that are usually consumed raw in Brazil was carried out as well as an evaluation of its resistance to the sanitizers available on the market for disinfection of green vegetables, equipment and utensils, including chlorine and quaternary ammonium compounds. In the evaluation of the occurrence in vegetables, 869 samples were analyzed, without the presence of the pathogen being detected. The enzymatic immunoassay (ELISA) used in the analyses (Reveal E.coli O157 Neogem e EHEC Test Kit 3M Company) presented a false presumtive rate of 13,6 and 11,8%, respectively, not confirmed as E.coli O157 in the subsequent stages of identification. In the evaluation of the resistance to the sanitizers using the AOAC method 960.9 it was observed that the treatment with 100ppm and 200ppm of sodium hypochlorite, sodium dichloroisocyanurate and benzalconium chloride/30s showed effectiveness against E.coli ATCC 11229 e E.coli O157:H7 ATCC 43890, with more than 5 decimal reductions over the population.

Foi verificada a ocorrência de Escherichia coli O157:H7 em 340 amostras de produtos cárneos e ambiente industrial, provenientes de frigoríficos do Sul e Sudeste do Brasil, no período de abril/98 a abril/99. A presença de E.coli O157:H7 não foi detectada em nenhuma das amostras analisadas e os resultados da avaliação da sensibilidade dos métodos de detecção evidenciaram que tanto o método cultural quanto o imunoensaio da Neogem foram capazes de detectar a presença de E.coli O157:H7 em cultura pura em concentrações iniciais de menos de 0,5Log UFC/mL do caldo de enriquecimento.

The occurrence of E.coli O157:H7 was evaluated in 340 samples of meat products and industrial environment of meat manufacturers from the South and Southeast regions of Brazil, from April, 1998 to April, 1999. Pathogen was not detected in any of the samples analysed, and the evaluation of the sensibility of the studied detection methods showed that both, culture and immuno-assay methods detected E.coli O157:H7 in pure culture in initial population levels of 0.5Log CFU/mL of enrichment broth.
