Resumo O presente estudo foi desenhado para verificar as propriedades anti-helmínticas de algumas plantas locais. Sementes de Amomum (A.) subulatum e Vitex (V.) negundo em diferentes solventes foram submetidas à análise de atividade anti-helmíntica in vitro (ensaio de motilidade de adultos; AMA e teste de eclosão de ovos; EHA) e in vivo (teste de redução da contagem de ovos nas fezes; TRCOF), usando o Haemonchus (H.) contortus como modelo experimental no protocolo de teste. Os resultados dos testes AMA, EHA e TRCOF foram analisados estatisticamente por meio de regressão linear e teste de Duncan. No teste AMA, na concentração de 50 mg mL-1, o percentual de mortalidade de H. contortus foi maior com o uso de A. subulatum do que com V. negundo, enquanto, no teste EHA, A. subulatum apresentou maior ação ovicida (LC50=14,2 µg mL- 1) do que V. negundo (LC50= 65,7405 µg mL-1). O TRCOF também indicou a melhor eficácia do uso de A. subulatum do que de V. negundo contra a infecção natural de parasitas gastrointestinais (GI). O extrato bruto seco das plantas utilizadas neste estudo apresentou 29,6% a 57,7% de atividade anti-helmíntica. A taxa de redução observada com o uso de A. subulatum (3 g kg-1) foi maior que com o uso de V. negundo (7 g kg-1). Em relação à análise da eficácia dos solventes utilizados para o extrato de plantas, o acetato de etila e o clorofórmio apresentaram maior ação ovicida em vermes adultos (testes in vitro), enquanto o extrato bruto metanólico aquoso apresentou maior eficácia do que os extratos brutos secos em testes in vivo. Consideramos vantajoso documentar o conhecimento indígena relativos aos procedimentos científicos padronizados, para sua validação. Este estudo irá servir de motivação para que os agricultores façam escolhas melhores referentes ao cultivo das plantas indígenas devido às suas diferentes eficácias comprovadas, servindo como alternativa para a abordagem preventiva contra as infecções parasitárias GI.

Abstract The current study was designed to check the anthelmintic activities of some local plants. Seeds of Amomum (A.) subulatum and Vitex (V.) negundo in different solvents were subjected to in vitro (adult motility assay; AMA and egg hatch assay; EHA) and in vivo (faecal egg count reduction test; FECRT) anthelmintic activity testing protocols using Haemonchus (H.) contortus as an experimental model. The results of AMA, EHA, and FECRT were statistically analysed through linear regression and Duncan multiple range test. In AMA test, at 50 mg mL-1 concentration, the percent mortality of H. contortus was higher in A. subulatum than V. negundo, whereas, in EHA test, A. subulatum was proven better ovicidal (LC50=14.2 µg mL-1) than V. negundo (LC50= 65.7405 µg mL-1). The FECRT also indicated the better efficacy of A. subulatum than V. negundo against natural infection of gastrointestinal (GI) parasites. The crude powder of plants used in this study showed 29.6% to 57.7% anthelmintic. The reduction rate was found higher for A. subulatum (3 g kg-1) as compared to V. negundo (7 g kg-1). Reagrding efficacy analysis of solvents used for plants extract, ethyl acetate and chloroform were found better in increasing ovicidal activity in adult worms (in vitro testing), whereas, the crude aqueous methanol was found better than the crude powders in in vivo testing. It will be beneficial to document the indigenous knowledge to standard scientific procedures for their validation. This study will help to motivate the farmers to make a better choice of cultivation of the indigenous plants because of their varying efficacies as an alternative preventive approach against the GI parasitic infections.

Resumo O objetivo do presente trabalho de pesquisa é fornecer conhecimento sobre a diversidade da fauna de aranhas e sua ocorrência ao longo do ano no distrito Charsadda Khyber Pakhtunkhwa, Paquistão. Os dados da pesquisa foram coletados no período de março de 2015 a janeiro de 2017 em sete diferentes localidades do distrito de Charsadda por meio de câmera, garrafa, sacos plásticos, filmes de parafina, livro de campo, e 70% de álcool etílico e 20% de glicerina foram utilizados como produtos químicos. Usando chaves de identificação especiais, as aranhas foram diferenciadas em família, gênero e espécie. Durante o período de estudo foram coletados 2.734 espécimes de aranhas pertencentes a 35 gêneros, 15 famílias e 44 espécies. Salticidae foi a família dominante de acordo com os gêneros estudados mais o número de amostras de aranhas coletadas com 10 gêneros e contagem de 616 espécies. Altas ocorrências de aranhas foram estudadas durante o mês de julho. O resultado do estudo atual também mostra redução das espécies de aranhas no mês de dezembro devido à diminuição da temperatura. O estudo atual mostra que os Salticidae foram a família dominante como alcaparras para outras espécies. A ocorrência de espécies de aranhas depende muito da mudança das condições climáticas. O presente estudo também mostra grande flutuação na ocorrência de aranhas com a mudança do clima quente para mais frio durante a duração do estudo. Além disso, a estação chuvosa desempenha um grande papel no aumento e crescimento da população de aranhas.

Abstract The current research work aims to provide knowledge about the diversity of spiders’ fauna and their occurrence throughout the year from District Charsadda Khyber Pakhtunkhwa, Pakistan. Research data were collected from March-2015 to January-2017 from seven different localities of Charsadda District by using the camera, bottle, plastic bags, paraffin films, field book and 70% of ethylene alcohol and 20% of glycerine were used as chemicals. By using special identification keys, spiders were differentiated into families, genera and species. During the study time, a total of 2734 specimens of spiders were collected belonging from 35 genera, 15 families and 44 species were identified. Salticidae was the dominant family according to genera studied plus spiders samples numbers collected with 10 genera and 616 species specimens count. The high occurrence of spiders was studied during July. The result of the current study also shows a reduction of spider’s species in December due to lowering the temperature. The current study shows that Salticidae were the dominant family as capered to other species. The occurrence of spiders species greatly depends on changing the weather condition. The present study also shows great fluctuation in spider’s occurrence with changing of hot climate to colder during the study duration. Moreover, the wet season plays a great role in spiders’ population increase and growth.

Resumo A utilização de técnicas modernas de melhoramento para o desenvolvimento de tipos de plantas uniformes e de alto rendimento, em última análise, estreitando a composição genética da maioria das culturas. A composição genética restrita dessas plantações tornou-as vulneráveis a doenças e epidemias de insetos. Para uma produção agrícola sustentável, a variabilidade genética dessas culturas deve ser ampliada contra vários estresses bióticos e abióticos. Uma das maneiras de ampliar a configuração genética dessas culturas é identificar novas fontes adicionais de resistência durável. A esse respeito, os parentes selvagens das culturas estão fornecendo fontes valiosas de diversidade alélica para vários componentes de qualidade e tolerância ao estresse abiótico e biótico. Para incorporar a nova variabilidade da ampla técnica de hibridização de parente selvagem tornou-se um método de reprodução promissor. Para esse efeito, trigo-Th. As linhas anfiploides, de adição e translocação de bessarabicum foram selecionadas em condições de campo e de casa de tela para obter novas fontes de ferrugem amarela e resistência ao bunt de Karnal. A triagem de ferrugem em faixas em condições de campo revelou as linhas de adição 4JJ e 6JJ como resistentes a moderadamente resistentes, enquanto as linhas de adição 3JJ, 5JJ, 7JJ e as linhas de translocação Tr-3, Tr-6 como estoque genético de trigo-Thinopyrum bessarabicum moderadamente resistente. A triagem Karnal bunt descreveu as linhas de adição 5JJ e 4JJ como estoque genético altamente resistente. Esses estoques genéticos podem ser usados para introgressão da nova ferrugem e resistência ao bunt de Karnal do pool genético terciário em origens de trigo suscetíveis.

Abstract Utilization of modern breeding techniques for developing high yielding and uniform plant types ultimately narrowing the genetic makeup of most crops. Narrowed genetic makeup of these crops has made them vulnerable towards disease and insect epidemics. For sustainable crop production, genetic variability of these crops must be broadened against various biotic and abiotic stresses. One of the ways to widen genetic configuration of these crops is to identify novel additional sources of durable resistance. In this regard crops wild relatives are providing valuable sources of allelic diversity towards various biotic, abiotic stress tolerance and quality components. For incorporating novel variability from wild relative’s wide hybridization technique has become a promising breeding method. For this purpose, wheat-Th. bessarabicum amphiploid, addition and translocation lines have been screened in field and screen house conditions to get novel sources of yellow rust and Karnal bunt resistant. Stripe rust screening under field conditions has revealed addition lines 4JJ and 6JJ as resistant to moderately resistant while addition lines 3JJ, 5JJ, 7JJ and translocation lines Tr-3, Tr-6 as moderately resistant wheat-Thinopyrum-bessarabicum genetic stock. Karnal bunt screening depicted addition lines 5JJ and 4JJ as highly resistant genetic stock. These genetic stocks may be used to introgression novel stripe rust and Karnal bunt resistance from the tertiary gene pool into susceptible wheat backgrounds.

Resumo Fatores de transcrição (FT) são uma ampla classe de genes em plantas e podem regular a expressão de outros genes em resposta a vários estresses ambientais (estresses bióticos e abióticos). No presente estudo, a atividade do fator de transcrição na cana-de-açúcar foi examinada durante o estresse pelo frio. Inicialmente, as leituras de transcrição de RNA de duas cultivares de cana-de-açúcar (ROC22 e GT08-1108) sob estresse frio foram baixadas do banco de dados SRA NCBI. As leituras foram alinhadas em um genoma de referência e as análises de expressão diferencial foram realizadas com o pacote R / Bioconductor edgeR. Com base em nossas análises no cultivar ROC22, 963 genes TF foram significativamente regulados positivamente sob estresse pelo frio entre um total de 5.649 genes regulados positivamente, enquanto 293 genes TF foram regulados negativamente entre um total de 3.289 genes regulados negativamente. No cultivar GT08-1108, 974 genes TF foram identificados entre 5.649 genes regulados positivamente e 283 genes TF foram encontrados entre 3.289 genes regulados negativamente. Os fatores de transcrição, em sua maioria, foram anotados com categorias GO relacionadas à ligação de proteína, ligação de fator de transcrição, ligação específica de sequência de DNA, complexo de fator de transcrição, atividade de fator de transcrição em RNA polimerase II, atividade de fator de transcrição de ligação de ácido nucleico, atividade de corepressor de transcrição, sequência específica da região reguladora, atividade do fator de transcrição da RNA polimerase II, atividade do cofator do fator de transcrição, atividade do fator de transcrição do promotor do plastídio, atividade do fator de transcrição do promotor da RNA polimerase I, polimerase II e RNA polimerase III. As descobertas dos resultados acima ajudarão a identificar fatores de transcrição expressos diferencialmente durante o estresse pelo frio. Ele também fornece uma análise abrangente da regulação da atividade de transcrição de muitos genes. Portanto, este estudo fornece base molecular para melhorar a tolerância ao frio em cana-de-açúcar e outras gramíneas economicamente importantes.

Abstract Transcription factors (TF) are a wide class of genes in plants, and these can regulate the expression of other genes in response to various environmental stresses (biotic and abiotic). In the current study, transcription factor activity in sugarcane was examined during cold stress. Initially, RNA transcript reads of two sugarcane cultivars (ROC22 and GT08-1108) under cold stress were downloaded from SRA NCBI database. The reads were aligned into a reference genome and the differential expression analyses were performed with the R/Bioconductor edgeR package. Based on our analyses in the ROC22 cultivar, 963 TF genes were significantly upregulated under cold stress among a total of 5649 upregulated genes, while 293 TF genes were downregulated among a total of 3,289 downregulated genes. In the GT08-1108 cultivar, 974 TF genes were identified among 5,649 upregulated genes and 283 TF genes were found among 3,289 downregulated genes. Most transcription factors were annotated with GO categories related to protein binding, transcription factor binding, DNA-sequence-specific binding, transcription factor complex, transcription factor activity in RNA polymerase II, the activity of nucleic acid binding transcription factor, transcription corepressor activity, sequence-specific regulatory region, the activity of transcription factor of RNA polymerase II, transcription factor cofactor activity, transcription factor activity from plastid promoter, transcription factor activity from RNA polymerase I promoter, polymerase II and RNA polymerase III. The findings of above results will help to identify differentially expressed transcription factors during cold stress. It also provides a comprehensive analysis of the regulation of the transcription activity of many genes. Therefore, this study provides the molecular basis for improving cold tolerance in sugarcane and other economically important grasses.

Bovine leukosis is an economically important disease of dairy cattle caused by the bovine leukaemia virus (BLV). The study aimed to determine the seroprevalence, haemato-biochemical effects, and risk factors pertinent to the prevalence of bovine leukosis in Holstein-Friesian purebred dairy cattle in the D.I. Khan region of Pakistan. A total of 192 sera were assayed using an enzyme-linked immunosorbent assay. Overall, 31.3% (60/192) cattle were detected as seropositive. There was a marked increase in total leukocyte count (11.29 ± 0.48χ10³/μl), lymphocytes (5.73 ± 0.42%), monocytes (0.81 ± 0.06%), haemoglobin (11.09 ± 0.46 g/dl), red blood cells (7.23 ± 0.37 χ10(6)/μl), and packed cell volume (31.75 ± 1.48%) in seropositive cattle. Serum biochemical parameters in seropositive cattle showed a marked increase in the liver enzymes, alanine transaminase (24.25 ± 1.03 U/l) and aspartate aminotransferase (49.33 ± 3.31 U/l), with a marked decrease in glutathione peroxidase (1365.63 ± 12.03 (U/l) and superoxide dismutase (2.14 ± 0.13 U/ml) activity. A significant association of age, pregnancy, breeding method, milk yield, and health status of seropositive animals with bovine leukosis was also recorded. The prevalence was higher in animals which were older, pregnant, artificially inseminated, low milk producers, and had a history of ailments. The current study found that bovine leukosis virus could cause changes in internal homeostasis, oxidative stress, and liver dysfunction, all of which should be considered during a control regimen. It was concluded that bovine leukosis was moderately prevalent in the D.I. Khan region in Pakistan.

This paper describes the trace and ultra-trace mineral concentration in different forages utilized by sheep (Kajli and Fat tail) in the rangelands of Chakwal district, Punjab, Pakistan. The concentrations of Se, Mo, Zn, Mn, Cu, and Co were assessed using spectrophotometric methods after the digestion of forage samples. The dominant forage species consumed by sheep were Buxus pappillosa, Prosopis juliflora, Ficus palmata, Acacia modesta, Trichodesma indica, Olea permiginea, Lantana camara, Justicia adhatoda, Morus nigra, and Ipomoea carnea. The overall mean concentration of Co (1.27±0.24 mg/Kg of DM), Zn (35.14±1.28 mg/Kg of DM), and Cu (28.06±1.29 mg/Kg of DM) in forages were higher than the requirements for lactating ewes, while the concentration of Se was within the normal range. Molybdenum (0.030±0.004 mg/Kg of DM) and Mn (29.69±1.61 mg/Kg of DM) concentrations were lower than dietary requirements for sheep. In different administrative divisions (tehsils) like Chakwal, Talagang, Choa Saidan Shah, and Kalar Kahar of Chakwal district, the trace minerals Zn, Mn, and Co, showed non-significant variation, whereas Se, Mo, and Cu showed substantial variation. In conclusion, rangeland forages contained a sufficient concentration of most trace minerals (Co, Cu, Se, Zn) to meet the requirements of grazing sheep in the study district. However, Mn and Mo supplementation seems to be essential to maintain the optimal production capabilities of sheep in this area.

Resumo Mutações são alterações genéticas nas sequências do genoma e têm papel significativo na biotecnologia, genética e biologia molecular, até mesmo para descobrir as sequências do genoma de um DNA celular junto com o sequenciamento do RNA viral. As mutações são alterações no DNA que podem ser naturais ou espontâneas e induzidas devido a reações bioquímicas ou radiações que danificam o DNA celular. Há outra causa de mutações, conhecida como transposons ou genes saltadores, que podem mudar sua posição no genoma durante a meiose ou a replicação do DNA. Os elementos transponíveis podem induzir por si próprios no genoma devido a mecanismos celulares e moleculares, incluindo hipermutação que causou a localização dos elementos transponíveis para se moverem dentro do genoma. O uso de mutações induzidas para estudar a mutagênese em plantas cultivadas é muito comum, bem como um método promissor para a triagem de plantas cultivadas com características novas e aprimoradas para a melhoria da produtividade e da produção. A utilização de mutações de inserção por meio de transposons ou genes saltadores geralmente gera alelos mutantes estáveis que são marcados quanto à presença ou ausência de genes saltadores ou elementos transponíveis. Os elementos transponíveis podem ser usados para a identificação de genes mutados em plantas de cultivo e até mesmo para a inserção estável de elementos transponíveis em plantas de cultivo mutadas. As proteínas de ligação ao nucleotídeo guanina (GTP) têm papel importante na indução de tolerância em plantas de arroz para combater as condições de estresse abiótico.

Abstract The mutations are genetic changes in the genome sequences and have a significant role in biotechnology, genetics, and molecular biology even to find out the genome sequences of a cell DNA along with the viral RNA sequencing. The mutations are the alterations in DNA that may be natural or spontaneous and induced due to biochemical reactions or radiations which damage cell DNA. There is another cause of mutations which is known as transposons or jumping genes which can change their position in the genome during meiosis or DNA replication. The transposable elements can induce by self in the genome due to cellular and molecular mechanisms including hypermutation which caused the localization of transposable elements to move within the genome. The use of induced mutations for studying the mutagenesis in crop plants is very common as well as a promising method for screening crop plants with new and enhanced traits for the improvement of yield and production. The utilization of insertional mutations through transposons or jumping genes usually generates stable mutant alleles which are mostly tagged for the presence or absence of jumping genes or transposable elements. The transposable elements may be used for the identification of mutated genes in crop plants and even for the stable insertion of transposable elements in mutated crop plants. The guanine nucleotide-binding (GTP) proteins have an important role in inducing tolerance in rice plants to combat abiotic stress conditions.

N-Phthalimido β-amino acid derivatives, 3-phthalimido-3(2-hydroxyphenyl) propanoic acid (P2HPA) and 3-phthalimido-3(2-nitrophenyl) propanoic acid (P2NPA) with new series of di- and triorganotin(IV) complexes (1-12) have been designed and synthesized. All the ligands and organotin(IV) complexes were characterized by elemental analysis, Fourier transform infrared spectroscopy (FTIR), nuclear magnetic resonance (1H, 13C, 119Sn) spectroscopy and electron ionization mass spectrometry (EI-MS). Synthesized ligands and complexes were screened to determine the antibacterial activity and results showed that the triorganotin(IV) complexes have better activity compared to diorganotin(IV) complexes and ligands. In addition, molecular docking analysis of ligands on the catalytic pocket of sortase A (PDB ID 1T2W) showed that the ligands can bind the active amino acid residues in the pocket. The antioxidant activity was also performed by the DPPH (1,1-diphenyl-2-picrylhydrazyl radical) method and complexes showed better results than ligands. The compounds were also tested in vivo to determine the hypoglycemic activities on different groups of alloxan induced diabetic rabbits. The complexes (1-6) were found better hypoglycemic agents as they stabilized the glucose level to about 175-105 mg dL-1 as compared to ligand P2HPA.

RESUMO: Na fruticultura sustentável, é muito importante manter a fertilidade do solo adotando-se o controle adequado de plantas daninhas e adicionando-se matéria orgânica. Assim, esta pesquisa foi realizada para identificar o modo de controle de plantas daninhas mais adequado para pomares de tangerina Kinnow. Foram selecionadas plantas de Kinnow com idade de 6 anos, cultivadas sob irrigação por gotejamento. As plantas daninhas do pomar foram submetidas a controle por meio de cobertura (com cavacos de madeira ou com polietileno preto), controle mecânico (cultivador e enxada rotativa) e métodos químicos (glifosato). O controle mecânico de plantas daninhas foi adotado como tratamento de controle. As plantas onde foi utilizada a cobertura com cavacos de madeira para controle das plantas daninhas mostraram aumento máximo em comprimento da parte aérea (24,00 cm), teores de clorofila (valor do índice SPAD de 84,00) e rendimento (11,88 ton ha-1), seguidas pelas plantas nas quais o controle de plantas daninhas foi realizado quimicamente, em comparação com as plantas com controle por meio do cultivador e da enxada rotativa, nas quais houve menor comprimento da parte aérea (12,00 e 12,33 cm), menores teores de clorofila (valores de SPAD de 41,00 e 42,67) e menor rendimento (3,80 e 4,70 t), respectivamente. A cobertura com cavacos de madeira também melhorou as propriedades físicas do solo, reduzindo a densidade do solo (1,49 Mg m-3) e a sua resistência à penetração (785,33 kPa); em contrapartida, houve aumento do teor de matéria orgânica do solo (1,17%), da condutividade hidráulica (34,50 mm/hr-1) e do carbono da biomassa microbiana do solo (159,68 g kg-1). O segundo melhor resultado foi obtido pelo controle com glifosato, enquanto os menores valores das propriedades físicas do solo foram observados no controle de plantas daninhas com o uso do cultivador e da enxada rotativa. A cobertura com cavacos de madeira também reduziu a massa seca das plantas daninhas (45,25 g) e exerceu controle eficiente delas (95,66%), em comparação com outros métodos de controle. Nesse caso, a cobertura com cavacos de madeira obteve melhor desempenho no controle de plantas daninhas, em todos os parâmetros estudados, do que a cobertura plástica, a enxada rotativa, o cultivador e o glifosato. Assim, recomenda-se a cobertura com cavacos de madeira para que os citricultores consigam melhorar as propriedades físicas do solo e o rendimento.

ABSTRACT: Going towards sustainable fruit culture, it is very important that soil fertility should maintain by adopting the proper weeds control and adding the organic matter. This research was carried out to identify the most suitable weed control method for Kinnow orchard. The plants of Kinnow at the age of 6 years grown under drip irrigation were selected. Weeds of orchard were managed by mulching (wood chip and black polyethylene), mechanical (cultivator and rotavator) and chemical (glyphosate) methods. Mechanical weed control was taken as control treatment. The plants where wood chip mulching was used for weed control showed maximum increase in shoot length (24.00 cm), chlorophyll contents (84.00 SPAD value) and yield (11.88 ton ha-1) followed by the plants where weeds were controlled with chemical control while lowest shoot length (12.00 and 12.33 cm), chlorophyll contents (41.00 and 42.67 SPAD value) and yield (3.80 and 4.70 ton) was achieved in cultivator and rotavator treatment respectively. Wood chip mulch also improved soil physical properties by reducing bulk density (1.49 Mg m-3), soil penetration resistance (785.33 kPa) and by enhancing soil organic matter (1.17%), hydraulic conductivity (34.50 mm/hr-1) and soil microbial biomass carbon (159.68 g kg-1) followed by glyphosate control while lowest values of soil physical properties were obtained in cultivator and rotavator weed control. Wood chip mulch also reduced weed dry weight (45.25 g) and efficiently controlled weeds (95.66%) as compared to other weed control methods. In this case, wood chip mulch performed better in all studied parameters than plastic mulch, rotavator, cultivator and glyphosate weed control. So, wood chip mulch should be recommended to citrus grower for improving soil physical properties and yield.

Abstract: The transfer of chemically reactive species in stagnation point flow of a laminar micropolar viscoelastic fluid immersed in a porous medium over a stretching/shrinking surface is considered. The reactive species diffused into the fluid from the surface undergo a one stage isothermal and homogenous reaction. A similarity transformation is employed to transform the developed partial differential equations into a system of coupled ordinary differential equations. A convergent series solution is developed using homotopy analysis method in the whole spatial region (0 ≤ η ≤ ∞). The obtained solutions for velocity, microrotation and concentration of species are analysed for various emerging parameters through graphs and tables.
