ABSTRACT. The stingless bee Tetragonisca angustula is an important pollinator of different agricultural and native crops. This study evaluated changes in the relative activity of esterases and critical electrolyte concentration in brain cells after exposure to pesticides malathion and thiamethoxam. Lethal concentration 50% showed greater toxicity of thiamethoxam in relation to malathion. Esterases EST-3 and EST-4 (carboxylesterase) were partially inhibited after contamination by contact and ingestion of malathion and contamination by contact with thiamethoxam, suggesting participation of these esterases in the metabolization of these compounds. The lowest critical electrolyte concentration (CEC) was found after contamination by malathion ingestion (0.15 M), indicating changes in gene expression. The alterations observed in the intensity of EST-3 and EST-4 and the chromatin structure indicate that pesticides can act in gene expression and be used as biomarkers of contaminant residues. Furthermore, knowing the susceptibility of T. angustula bees to pesticides, it would be possible to use this species for biomonitoring environmental quality in preserved areas and agroecosystems.

Diatraea saccharalis são consideradas as principais pragas da cana-de-açúcar, estando distribuídas em todas as regiões canavieiras do Brasil. As informações sobre a genética de populações em D. saccharalis são raras e sua análise é muito importante para o sucesso de programas de controle da praga. O controle de praga necessita de uma constante avaliação da variabilidade genética para que estratégias adequadas sejam empregadas. A estrutura de populações de D. saccharalis presentes em culturas de cana-de-açúcar foi analisada por meio do marcador PCRRAPD. As amostras foram coletadas em canaviais de quatro regiões do noroeste do estado do Paraná e uma do estado de São Paulo. Foram empregados 12 primers RAPD, os quais produziram 216 fragmentos, com 99,54% de polimorfismo. O índice de Shannon foi em média 0,3797 (± 0,1729) e o valor médio encontrado do GST nas cinco populações foi de 0,0909, mostrando que as populações estão diferenciadas. Análise molecular de variância mostrou que 92% do polimorfismo ocorre dentro das populações. O amplo polimorfismo intrapopulacional mostra que esses insetos têm potencial para desenvolver resistência a inseticidas. O controle e o manejo da D. saccharalis deve ser realizado com cautela e integrado, ou seja, controle químico e biológico.

Diatraea saccharalis is the principal pest of sugarcane in Brazil and is found throughout the sugarcane crop. Information about its population genetics is scarce, but population genetic analysis is of particular importance as a basis for a successful pest control program. Pest control requires a constant evaluation of genetic variability so that appropriate strategies can be employed. In this study, the structure of D. saccharalis populations in sugarcane crops was analyzed with PCR-RAPD (Polymerase Chain Reaction-Random Amplified Polymorphic DNA) markers. Samples were collected from four sugarcane fields in ParanáState and one in São PauloState. Twelve PCR-RAPD primers were used. The analysis produced 216 fragments, with a 99.54% polymorphism rate. The Shannon index averaged 0.3797 (± 0.1729), and the average value of GST for the five populations was 0.0909. These results showed that the populations are differentiated. An analysis of molecular variance showed that 92% of the polymorphism occurs within populations. The high polymorphism rate within these populations shows that these insects have the potential to develop resistance to insecticides. The control and management of D. saccharalis should be conducted cautiously and integrated using chemical and biological control.

A pesar de la condición de plaga de las hormigas cortadoras en agroecosistemas, no se pueden negar los beneficios que ellas pueden traer en determinadas situaciones o en determinados ambientes. Las hormigas cortadoras del género Acromyrmex atacan principalmente las hojas de hortalizas y hojas de plantas fructíferas, exponiéndose no solo a los agroquímicos utilizados para su control sino también a aquellos utilizados en el control de otras plagas. Debido al potencial bioindica-dor de la calidad ambiental de las hormigas y su frecuente exposición a los agrotóxicos como organofosforados, neonicotinoides y reguladores de crecimiento, utilizados en el control de plagas, es necesario el estudio de los efectos subletales que estos agrotóxicos pueden causar. La técnica de electroforesis fue utilizada para evaluar la actividad de las isoenzimas esterasas involucradas en el metabolismo de xenobióticos de A. niger , asociada a las alteraciones en la expresión de las isoenzimas después de la contaminación con pesticidas. Acromyrmex Niger demostró ocho regiones de actividad esterasa, las cuales fueron denominadas EST-1, EST-2, EST-3, EST-4, EST-5, EST-6, EST-7 e EST-8 de acuerdo con la movilidad electroforética. En cuanto a la especificación de los substratos ± y b-nafitl acetato, las EST-7 e EST-8 son clasificadas como ±- esterasa y las demás ±b esterasas. EST-5 es considerada una enzima del tipo colinesterasa II y las demás son carboxilesterasas. El análisis electroforético presentó inhibición parcial para todas las esterasas sometidas al contacto con malathion en las concentraciones 1 x 10-3 % e 5 x 10-3 %, que puede ser considerado un biomarcador para la presencia de residuos de este insecticida en el ambiente. El análisis de regresión para el efecto subletal de los pesticidas evaluados demostró correlación entre la dosis y la causa de muerte solo para el pesticida neonicotinoide thiametoxam

Despite the condition of leaf-cutting ant pests in agroecosystems, it is undeniable the benefits they can bring in certain situations or environments. The leaf-cutting ants of the genus Acromyrmex attack mainly leaves of vegetables and fruit trees exposing not only to the agrochemicals used for their control as well as to those used for the control of other pests. Due to the bioindicator potential of environmental quality of the ants and their frequent exposure to agrochemicals such as organophosphates, neonicotinoids and growth regulators insecticide used for pest control, it is necessary to study the sublethal effects that these pesticides may cause. The electrophoresis technique was used to study the activity of esterase isozymes involved in the metabolism of xenobiotics of A. niger , combined with changes in the expression of isozymes after contamination by pesticides. A. niger showed eight regions of esterase activity, which were called EST-1, EST-2, EST-3, EST-4, EST-5, EST-6, EST-7 and EST-8 according to the electrophoretic mobility. As the specificity to ± and b-naphthyl acetate substrates, the Est- 7 and Est-8 may be ified as ±-esterase and the others as ±b esterases. EST-5 is considered an enzyme of the type cholinesterase II and the others are of the type carboxilesterase. The electrophoretic analysis showed partial inhibition to all esterases subjected to the contact with Malathion organophosforate at the concentrations 1 x 10-3 % and 5 x 10-3 %, which may be considered as a biomarker for the presence of residues of this insecticide in the environment. The regression analysis for sublethal effects of the tested pesticides demonstrated correlation between dose and mortality only for Thiametoxam neonicotinoid pesticide.

La variabilidad genética de las hormigas Atta sexdens rubropilosa colectadas en cinco lugares distintos de Brasil fueron evaluados por la técnica PCR-RAPD. Un total de 15 primers produjeron 148 fragmentos, de los cuales 123 fueron polimórficos, lo que corresponden al 83,11 %. La estimación de la diversidad genética por el índice de Shannon fue 0,3836 y el desviación estándar fue de ± 0,2335. Estos valores demuestran una alta diversidad genética. El valor de G ST fue 0,2372 y ΦPT = 0,184 lo que indica que las poblaciones están moderadamente diferenciadas y que el 82 % de la variación obtenida se produce dentro de las poblaciones. Aunque la prueba de Mantel ha demostrado una correlación entre la distancia genética y geográfica se observó que Ivatuba e Itambé (33,8 km) tiene una pequeña distancia geográfica y la mayor distancia genética. La distancia inferior genética fue estimada para Maringá e Ivatuba pero estas localidades cuentan con una distancia geográfica pequeña (42,3 km), lo que indica que no hay barreras para el apareamiento entre los reproductores en estas poblaciones. El alto valor de polimorfismo (83,11 %) y la capacidad de emparejamiento entre las poblaciones presentes en las regiones estudiadas, indican que esta especie de hormiga cortadora está bien adaptada a la región, y deben ser desarrollados programas integrados de control si se convierten en plagas.

The genetic variability of Atta sexdens rubropilosa leaf-cutting ants collected from five brazilian localities was evaluated with PCR-RAPD technique. We used 15 primers producing 148 fragments of which 123 (83.11 %) contained polymorphisms. The estimated Shannon index was 0.3836 ± 0.2335 showing that these ants possess high genetic diversity. The G ST value was 0.2372 and ΦPT = 0.184, indicating that the analyzed populations are moderately differentiated and 82 % of the variation obtained occur within populations. Although Mantel's test had shown correlation between genetic distances and geographic was observed that Ivatuba and Itambé (33.8 km) have the small geographical distance and the largest genetic distance. The lower genetic distance was estimated for Maringá and Ivatuba but this localities have a small geographic distance (42.3 km), indicating that there are no barriers for mating among reproducers in these populations. The high degree of polymorphism (83.11 %) and the ability to cross among the populations in the studied regions indicate that this species of leaf-cutting ant is well adapted to the region; therefore, integrated control programs can be developed.

Este experimento teve como objetivos avaliar a polinização realizada por abelhas Apis mellifera, estudar a biologia floral e observar o comportamento de coleta nas flores de soja (Glycine max L. Merril), variedade BRS-133 plantadas na região de Maringá-PR. Os tratamentos constituíram de áreas demarcadas de livre visitação por insetos, áreas cobertas por gaiolas, com uma colônia de abelhas (A. mellifera) no seu interior e plantas também cobertas por gaiola que impedia a visitação por insetos. Todas as áreas possuíam 24 m² (4 m x 6 m). As flores de soja permaneceram abertas por um tempo maior (82,82 ± 3,48 horas) no tratamento coberto sem abelhas. O estigma das flores também se mostrou mais receptivo (P=0,0021) no tratamento coberto sem abelhas (87,3 ± 33%) e, às 10h42min, foi o horário de maior receptividade. O pólen se manteve viável em todos tratamentos, a média foi de 99,60 ± 0,02%, não apresentado diferenças entre tratamentos. O teste de fidelidade demonstrou que as abelhas A. mellifera foram 100% fiéis às flores de soja. A porcentagem de aborto das flores foi de 82,91% no tratamento sem abelhas e este resultado foi superior (P=0,0002) aos 52,66% e 53,95% dos tratamentos livre e coberto com abelhas, respectivamente. As quantidades médias de açúcar total e de glicose medidas no néctar das flores foram, respectivamente, de 14,33 ± 0,96 µg/flor e de 3,61 ± 0,36 µg/flor, não apresentando diferenças (P>0,05) entre os tratamentos. Os sólidos totais, medidos através do refratômetro manual foram de 21,33 ± 0,22% no tratamento livre e de 22,33 ± 0,38% no tratamento coberto com abelhas e estes resultados diferiram entre si (P=0,0001). A quantidade de néctar nas flores foram similares entre os tratamentos, sendo que o volume médio de néctar/flor foi 0,072 ± 0,04 µL/flor. As abelhas A. mellifera foram os insetos mais freqüentes (95,18%). Outros insetos observados foram os lepidópteros (3,51%) e outras abelhas (1,32%), no tratamento livre. Neste tratamento, o tempo de coleta para néctar foi de 2,55 ± 0,07 segundos que foi menor (P=0,0039) que o tempo de 2,87 ± 0,08 segundos, observado no tratamento coberto com abelhas. O comportamento para o tipo de coleta observado nas abelhas A. mellifera mostrou que as coletoras de néctar não apresentaram um padrão, mas para pólen, o horário de pico das coletoras foi às 11h36min. As flores de soja se mostraram sensíveis aos tratamentos, revelando alterações na sua biologia quando foram expostas ou não às abelhas.

This research was carried out to evaluate the pollination by Africanized honeybees Apis mellifera, the floral biology and to observe the hoarding behavior in the soybean flowers (Glycine max Merril), var. BRS-133. The treatments were constituted of demarcated areas for free visitation of insects, covered areas by cages with a honeybee colony (A. mellifera) and also covered areas by cage without insects visitation. All areas had 24 m² (4m x 6m). The soybean flowers stayed open for a larger time (82.82 ± 3.48 hours) in covered area without honeybees. The stigma of the flowers was also more receptive (P=0.0021) in covered area without honeybees (87.3 ± 33.0%) and at 10:42 o'clock was the schedule of greater receptivity. The pollen stayed viable in all treatments, the average was 99.60 ± 0.02%, which did not present differences among treatments. The percentage of abortion of the flowers was 82.91% in covered area without honeybees, this result was superior (P=0.0002) to the 52.66% and 53.95% of the treatments uncovered and covered with honeybees, respectively. Honeybees were responsible for 87.7% of the pollination accomplished by the insects. The medium amounts of total sugar and glucose measured in the nectar of the flowers were, 14.33 ± 0.96 mg/flower and 3.61 ± 0.36 mg/ flower, respectively, not showing differences (P<0.05) among the treatments. The total solids, measured through the manual refratometer were 21.33 ± 0.22% in uncovered area and 22.33 ± 0.38% in covered with honeybees and differed to each other (P=0.0001). The honeybees were the most frequent insect (95.18%). Other observed insects were the Lepidoptera with 3.51% and other bees with 1.32%, in uncovered area. Honeybees visited 2.24 flowers on average in uncovered area and 1.58 in covered with honeybees and presented behavior for nectar hoarding and hoarding nectar/pollen in this period. In uncovered area the time of nectar hoarding was 2.55 ± 0.07 seconds, this time was smaller (P=0.0039) than 2.87 ± 0.08 seconds observed in covered area with honeybees. The behavior for the hoarding type observed in honeybees foraging the soybean flowers, through the content of its honey stomach and pollen loads of its pollen basket showed that the nectar forager did not show a pattern for the nectar collection, but for the pollen collection the schedule of pollen peak of the forager was at 11:36 o'clock. The soybean flowers showed alteration in floral biology when exposed to honeybees.

Este experimento teve como objetivo avaliar a polinização realizada pelas abelhas na produção e qualidade das sementes da soja (Glycine max L. Merril) na região de Maringá-PR. Os tratamentos constituíram de áreas demarcadas de livre visitação por insetos, áreas cobertas por gaiolas com uma colônia de abelhas (Apis mellifera) e plantas também cobertas por gaiolas que impediam a visitação por insetos. Todas as áreas possuíam 24 m² (4 m x 6 m), com cinco repetições cada. A produção de sementes foi maior (P=0,0001) nas áreas cobertas com abelhas e de livre visitação com um incremento na produtividade de 50,64% e 57,73%, respectivamente, em relação à área coberta sem abelhas. Pode-se considerar que as abelhas A. mellifera foram responsáveis por 95,5% da polinização realizada pelos insetos no tratamento livre. O número de vagens no tratamento coberto com abelhas foi 61,38% maior (P=0,0002) do que no coberto sem abelhas. Onde as abelhas A. mellifera foram responsáveis pela polinização cruzada, houve um aumento de 58,86% no número de sementes em relação ao tratamento onde não foi permitida a polinização realizada por insetos. Entretanto, o peso médio de 100 sementes foi maior (P=0,0001) na área coberta sem abelhas, atingiu um peso médio de 17,80 g, mostrando que plantas com menor produção formaram sementes maiores. No tratamento livre, o peso médio de 100 sementes foi de 15,26 g e no coberto com abelhas foi de 15,37 g. O teor médio de proteína bruta no grão foi de 36,69% e a média do teor de óleo foi de 20,24%. O teste de germinação não mostrou diferenças entre as sementes nos diferentes tratamentos. Pode-se concluir que as abelhas A. mellifera foram eficientes no trabalho de polinização na soja, proporcionando um aumento considerável na produção de grãos e estes resultados reforçam a necessidade do uso das abelhas A. mellifera para elevar a produtividade da soja.

This experiment was carried out to evaluate the effect of the honeybee pollination in the production and quality of soybean seeds (Glycine max L. Merril). Seed production was higher (P=0.0001) in covered areas with honeybee colonies (50.64%) and uncovered areas (57.73%) than in covered areas without honeybee colonies. It could be concluded that honeybees were responsible for 95.5% of the pollination accomplished by insects. The pod number in covered treatment with honeybees was 61.38% higher (P=0.0002) than in the covered treatment without honeybees. The average weight of 100 seeds was larger (P=0.0001) in the area covered without honeybees, and reached 17.8 g. The medium content of crude protein in grains was 36.7% and the average oil content was 20.2%. The germination test did not show differences (P>0.05) among the seeds in different treatments. It was concluded that the honeybee pollination in the soybean increased the seeds production.

As abelhas sem ferrão são responsáveis por cerca de 30-90% da reprodução dos vegetais nas florestas tropicais, através do mecanismo de polinização, na fecundação cruzada (Kerr et al. , 1994). Esse trabalho teve o objetivo de realizar um levantamento de angiospermas e os insetos que as polinizam na região de Maringá, PR, com o intuito de contribuir com o entendimento da biodiversidade dessa região. As abelhas foram coletadas entre agosto de 1999 a janeiro de 2000, com o auxílio de uma rede entomológica, durante suas visitas às flores. As coletas foram semanais. Foram coletados 331 insetos, dos quais 39,88% eram Trigona spinipes, 38,37% Apis mellifera, 8,16% Tetragonisca angustula, 3,93% Halictidae, 1,21% Megachilidae, 2,42% Anthophoridae e 3,32% de outros Hymenoptera. Foram observadas 11 espécies vegetais pertencentes a nove famílias. As quatro famílias mais visitadas pelas abelhas A. mellifera foram Pontederiaceae - 93,55%, Lamiaceae - 55,56%, Sterculiaceae e Polygoniaceae - 47,22%; por T. spinipes foram Lamiaceae - 63,95%, Apocynaceae - 57,14%, Sterculiaceae - 51,85% e Anacardiaceae - 48,39% e por T. Angustula, Lamiaceae - 60,87%, Apiaceae - 28,57%, Anacardiaceae - 16,13% e Asteraceae - 12,50%. Três espécies vegetais foram predominantes em número de visitas pelas abelhas, concentrando 61,7% das visitas, destacando-se: Dombeya wallichii (32,63%), Ocimum americanum (16,31%) e Antigonon leptopus (10,88%). T. angustula foi o visitante mais freqüente (60,87%) em O. gratissimum.

The present study was carried out to survey the bees as visitors to melliferous flora in the region of Maringá, state of Paraná, Brazil. A total of 331 insects were captured, and the fauna comprised 39.88% Trigona spinipes, 38.37% Apis mellifera, 8.16% Tetragonisca angustula, 3.93% Halictidae, 1.21% Megachilidae, 2.42% Anthophoridae, and 3.32% other Hymenoptera. Eleven plant species from nine families were observed. The four families most frequently visited by A. mellifera were Pontederiaceae (93.53%), Sterculiaceae and Polygoniaceae (47.22%), Apocynaceae and Apiaceae (42.86%). The families most visited by T. spinipes were Lamiaceae (64.70%), Apocynaceae (57.14%), Sterculiaceae (51.85%) and Anacardiaceae (48.39%), and the families most visited by T. angustula were (28.57%), Asteraceae (22.22%) and Labiatae (16.47%). Three species predominated in number of bee visits Dombeya wallichii (32.63%), Ocimum americanum (15.5%) and Antigonon leptopus (15.2%). T. angustula was the most frequent visitor of O. gratissimum flowers (60.87%).
