Isolation and characterization of the aerobic bacterial microbiota of the esophagus and its probable association with obstructive caseous lesions in green turtles (Chelonia mydas)

RESUMO: A ocorrência de lesão caseosa no esôfago de tartarugas-verdes (Chelonia mydas) da costa do Brasil tem sido descrita como de caráter obstrutivo e pode causar a morte dos animais. No entanto, sua etiologia permanece pouco esclarecida. Objetivou-se isolar e caracterizar a microbiota aeróbica esofágica das tartarugas-verdes (C. mydas) da costa brasileira e verificar sua possível participação na etiologia das lesões caseosas. Foram utilizados 42 animais, 33 vivos e hígidos e nove mortos naturalmente que apresentavam lesão esofágica confirmada pela necropsia, provenientes de Anchieta e Piúma, Espírito Santo, nos quais foram feitos testes microbiológicos e avaliação morfológica do esôfago. Foram isolados 14 agentes bacterianos diferentes nas amostras de animais saudáveis, com prevalência de Pseudomonas aeruginosa (36,36%), Staphylococcus aureus (33,33%), Aeromonas hydrophila (27,27%) e Vibrio alginolyticus (24,24%). Nos animais mortos, foram isolados apenas três agentes distintos: S. aureus (50,00%), A. hydrophila (25,00%) e V. alginolyticus (25,00%). A avaliação morfológica revelou predominância da lesão em junção gastroesofágica, com distribuição multifocal a coalescente, intensidade discreta e ausência de obstrução. Observou-se ainda ulceração e exsudato caseoso, infiltrado inflamatório, ovos de parasitos e células gigantes do tipo corpo estranho, além de grumos bacterianos e de alterações glandulares, como necrose, adenite e fragmentos de parasitos adultos. Houve correlação positiva dos grumos bacterianos com cultivo microbiológico e negativa dos grumos bacterianos e cultivo microbiológico com parasitos. Assim, nota-se que a microbiota esofágica aeróbica de C. mydas é constituída predominantemente por bactérias Gram-negativas como P. aeruginosa, A. hydrophila e V. alginolyticus, além de diversas enterobatérias e por Gram-positivas, como S. aureus. Esses agentes são oportunistas e podem estar envolvidos na etiologia da esofagite caseosa em associação a outros patógenos como co-fatores agindo em associação, ou mesmo, por via de infecção secundária.
ABSTRACT: Caseous lesions in the esophagus of green turtles (Chelonia mydas) from the coast of Brazil have been described as obstructive lesions and can lead to the death of these animals. However, their etiology remains unclear. The aim of this study was to isolate and characterize the aerobic bacterial microbiota of the esophagus of green turtles (C. mydas) from the Brazilian coast and to verify its possible participation in the etiology of caseous lesions. For this, 42 animals were used, 33 alive and healthy and 9 naturally dead that had esophageal lesions confirmed by necropsy, from Anchieta and Piúma beaches, Espírito Santo. Microbiological tests and morphological evaluation of the esophagus were performed. We isolated 14 different bacterial agents from healthy animal samples, with the prevalence of Pseudomonas aeruginosa being (36.36%), Staphylococcus aureus (33.33%), Aeromonas hydrophila (27.27%), and Vibrio alginolyticus (24.24%). In dead animals, only three distinct agents were isolated: S. aureus (50.00%), A. hydrophila (25.00%), and V. alginolyticus (25.00%). Morphological evaluation revealed a predominance of the lesions at the gastroesophageal junction, with multifocal-to-coalescent distribution, discrete intensity, and absence of obstruction. Ulcerations and caseous exudates, inflammatory infiltrates, parasitic eggs, and giant foreign body cells were also observed as well as bacterial lumps and glandular alterations, such as necrosis, adenitis, and fragments of adult parasites. There was a positive correlation between bacterial lumps and microbiological culture and a negative correlation between bacterial lumps and microbiological culture with parasites. Thus, it was noted that the esophageal aerobic microbiota of C. mydas was predominantly composed of Gram-negative bacteria such as P. aeruginosa, A. hydrophila, and V. alginolyticus, in addition to several enterobacteria and Gram-positive bacteria, such as S. aureus. These agents are opportunists and may be involved in the etiology of caseous esophagitis in association with other pathogens as co-factors working in association or, even in a secondary way.

Differences in Total Brain Volume between Sexes in a Cognitively Unimpaired Elderly Population

OBJECTIVES: Although a large number of studies have shown brain volumetric differences between men and women, only a few investigations have analyzed brain tissue volumes in representative samples of the general elderly population. We investigated differences in gray matter (GM) volumes, white matter (WM) volumes, and intracranial volumes (ICVs) between the sexes in individuals older than 66 years using structural magnetic resonance imaging (MRI). METHODS: Using FreeSurfer version 5.3, we obtained the ICVs and GM and WM volumes from the MRI datasets of 84 men and 92 women. To correct for interindividual variations in ICV, GM and WM volumes were adjusted with a method using the residuals of a least-square-derived linear regression between raw volumes and ICVs. We then performed an analysis of covariance comparing men and women, including age and years of schooling as confounding factors. RESULTS: Women had a lower socioeconomic status overall and fewer years of schooling than men. The comparison of unadjusted brain volumes showed larger GM and WM volumes in men. After the ICV correction, the adjusted volumes of GM and WM were larger in women. CONCLUSION: After the ICV correction and taking into account differences in socioeconomic status and years of schooling, our results confirm previous findings of proportionally larger GM in women, as well as larger WM volumes. These results in an elderly population indicate that brain volumetric differences between sexes persist throughout the aging process. Additional studies combining MRI and other biomarkers to identify the hormonal and molecular bases influencing such differences are warranted.

STENOSTOMUM LEUCOPS DUGÈS, 1828 (PLATYHELMINTHES, CATENULIDA): A PUTATIVE SPECIES COMPLEX WITH PHENOTYPIC PLASTICITY

RESUMO Stenostomum são pequenos vermes que vivem em água doce e normalmente se reproduzem assexualmente por paratomia. Eles estão na base da filogenia dos platelmintes. Por mais de um século, espécies desse gênero, especialmente S. leucops, vêm sendo empregadas em estudos biológicos, principalmente sobre regeneração. Entretanto, alguns aspectos básicos da biologia destes vermes são ainda pobremente conhecidos. Neste estudo, caracterizamos uma linhagem que vem sendo mantida no laboratório por cinco anos. O tempo necessário para reprodução assexuada e completa formação de zoóides, a 28°C, é de aproximadamente 42,5 horas. O número de células nos zoóides, logo após a paratomia, é de aproximadamente 2.500. O número de zoóides presentes nos vermes é uma característica variável e depende das condições de cultivo. Em alguns procedimentos de cultivo de S. leucops, apenas cadeias com dois zoóides são formadas. No entanto, em outras condições de cultivo, cadeias de até cinco zoóides podem ser observadas. Análise filogenética empregando sequência do gene de Citocromo C Oxidase (COI) mostrou que S. leucops e S. grande constituem um complexo de espécies cujas linhagens mostram altas divergências intraespecíficas.
ABSTRACT Species of Stenostomum are small flatworms that live in freshwater and normally reproduce asexually by paratomy. They are basal in the phylogeny of Platyhelminthes. For more than a century, species of this genus, especially S. leucops, have been used in regeneration and other biological studies. However, some basic aspects of their biology are poorly known. Here, we characterized a strain of S. leucops that has been maintained in the laboratory for five years and a recent strain of S. grande. The time required for complete formation of zooids of S. leucops by asexual reproduction is approximately 42.5 hours at 28°C. The number of cells in the zooids, soon after paratomy, is approximately 2,500. The number of zooids formed in the chain is a plastic characteristic and is dependent on the conditions for cultivation. In some cultivation conditions of S. leucops, only worms with two zooids are formed. However, in other conditions, worms with up to five zooids are observed. Phylogenetic analyses of a fragment of the Cytochrome C Oxidase I (COI) sequence showed S. leucops and S. grande species constitute a species complex, the lineages of which having high intraspecific divergences.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The aim of this work was to evaluate different transport times for pacama juvenile (Lophiosilurus alexandri) previously trained to feed formulated diets. Four hundred and eighty-six pacama juveniles (2.52±0.71g) were submitted to 24 hours of fasting before transport. The animals were divided into nine plastic bags, with density of 54 juveniles in 5L of clean water for each bag after that inflated with pure oxygen and then sealed. Fish were transported for three hours and bags were open at 5, 6 and 7.5 hours after closing. For each time three replications were used. Survival was monitored for 96 hours post-transport. The longest time tested showed the lowest values of dissolved oxygen and pH, while the total ammonia was similar in the different treatments. Survival was 100% for different transport times after 96 hours. Some juveniles returned to feed 48 hours and the majority 72 hours after opening the bags, suggesting the efficiency of transport made at different times.

Introduction: Trigeminal neuralgia is a facial pain syndrome known and characterized by severe, intermittent, electric shock-like, shooting pain in the face for which a number of surgical therapies have been used. To date no ideal, minimally invasive, patient-acceptable treatment is available, hence both patients and specialists face substantial uncertainty when making decisions regarding therapeutic action. Objective: To identify which surgical treatment for TN offers the best opportunity in terms of cost-effectiveness. Material and method: A study was performed using a decision analysis/cost-effectiveness analysis design considering five surgical alternatives in the management of TN: radiofrequency thermocoagulation, glycerol microcompression, balloon microcompression, radiosurgery, and microvascular decompression. Data were obtained from the literature. A review was carried out of the MEDLINE database from 2000 to 2010 via PubMed Central. Studies were selected that addressed primary trigeminal neuralgia and demonstrated pain relief and reduced mortality and complication rates. In all 196 studies were identified but only 22 (11.22 %) were eligible for the study. The software package used was the DATA 3.5 for Health Care, version 3.5.5, program by TreeAge Software Inc. Results: Microvascular decompression and thermocoagulation are the techniques more likely to provide effective pain relief, with expected values of 0.8946 and 0.8863, respectively. For the relationship between cost and outcome pain-free days were considered; the best choice in terms of cost-effectiveness is surgery and thermocoagulation, with a lower value in the marginal cost-effectiveness ratio: $171.58 for an effectiveness of 89 % per treated patient. Conclusion: Considering that both radiofrequency thermocoagulation and microvascular decompression are the most commonly used surgical procedures because of their high effectiveness, we conclude that radiofrequency thermocoagulation is more beneficial, has few complications, and is less costly.
Introducción: La neuralgia trigeminal es un síndrome de dolor facial conocido y caracterizado por dolores severos, intermitentes, eléctricos y como de sacudidas en la cara, para el cual han sido aplicados diversos tratamientos quirúrgicos, hasta la fecha, no hay un tratamiento ideal que sea invasivo en grado mínimo y aceptable para el paciente, como consecuencia, los pacientes y los especialistas se enfrentan a una incertidumbre considerable al hacer decisiones sobre la conducta terapéutica. Objetivo: Identificar cuál de los tratamientos quirúrgicos para la NT brinda la mejor oportunidad en términos de coste-efectividad. Material y método: Se realizó un estudio con un diseño de Análisis de Decisión: coste-efectividad considerando cinco alternativas quirúrgicas en el tratamiento de la NT: termocoagulación por radiofrecuencia, microcompresión con glicerol, microcompresión con balón, radiocirugía, y microdescompresión vascular. Los datos se obtuvieron de la literatura. Se revisó la base de datos MEDLINE desde el año 2000 hasta el 2010, a través de PubMed Central. Se seleccionaron los estudios que abordaran la neuralgia trigeminal primaria y mostraran resultados relacionados con el alivio del dolor, la tasa de mortalidad o la presencia de complicaciones. Se identificaron 196 estudios pero solo 22 (11,22 %) fueron elegibles para el estudio. Se utilizo el programa DATA 3.5 for Health Care, versión 3.5.5 de TreeAge Software Inc. Resultados: La microdescompresión vascular y la termocoagulación son las técnicas que ofrecen mayores probabilidades de efectividad para el alivio del dolor con valores esperados de 0,8946 y 0,8863. Para la relación que se establece entre coste y resultado se consideró los días libres de dolor. La mejor elección coste-efectividad es el tratamiento quirúrgico con termocoagulación con menos valor en la razón coste-efectividad marginal, $171.58 para una efectividad de la cirugía de 89 % por cada paciente tratado. Conclusión: Considerando que la termocoagulación por radiofrecuencia y la microdescompresión vascular constituyen las variantes quirúrgicas más utilizadas por la alta efectividad, concluimos que la termocoagulación por radiofrecuencia es más beneficiosa, ofrece escasas complicaciones y es menos costosa.

Estudou-se a dinâmica das populações de Staphylococcus spp. durante a maturação do queijo Canastra, em três fazendas localizadas no estado de Minas Gerais. A presença dos genes que codificam para a produção das enzimas coagulase (coa), termonuclease (nuc) e produção de enterotoxinas (sea, seb, sec e sed), em linhagens de Staphylococcus isoladas durante os 60 dias de maturação do queijo foi analisada. Também foi investigada a presença de enterotoxina estafilocócica A, C e D nas amostras de queijo. As amostras de queijo com 0, 7, 15, 30 e 45 dias de maturação apresentaram contagens de Staphylococcus spp. entre 10³ e 10(8)ufc / g. Todos os isolados coagulase positivo nos testes fisiológicos apresentaram o gene coa. Não foi observada associação entre os resultados obtidos com os testes bioquímicos e aqueles obtidos com a PCR usando iniciadores gene-específicos para linhagens coagulase negativa. Os genes da coagulase e termonuclease ocorreram simultaneamente em 41,3% dos Staphylococcus spp. testados. Nenhum dos isolados de Staphylococcus apresentou os genes que codificam para a produção das enterotoxinas SEA, SEB, SEC ou SED. As enterotoxinas A, C ou D não foram detectadas em nenhuma das amostras de queijo analisadas.
The population dynamics of Staphylococcus spp. was studied during the ripening of Canastra Minas cheese at three farms located in the State of Minas Gerais, Brazil. The presence of coagulase (coa), thermonuclease (nuc), and enterotoxin (sea, seb, sec, and sed) genes was investigated in Staphylococcus strains isolated during the 60-day cheese-ripening period. The presence of the staphylococcal enterotoxins A, C, and D was also investigated in the cheese samples. Cheese samples that were matured for 0, 7, 15, 30, and 45 days presented staphylococci counts from 10³ to 10(8)cfu/g. All isolates considered coagulase-positive by physiological tests had the coa gene. However, no association was observed between the results obtained with biochemical tests and those obtained by PCR using gene-specific primers for coagulase-negative strains. Coagulase and thermonuclease genes occurred simultaneously in 41.3% of Staphylococcus spp. tested. None of the investigated Staphylococcus strains expressed enterotoxins SEA, SEB, SEC, and SED. Enterotoxins A, C, and D were not detected in any of the cheese samples.

RESUMO O estudo da ixodofauna de fragmentos de mata em área urbana é fundamental para o conhecimento das espécies de carrapatos e para a dinâmica de transmissão entre hospedeiros silvestres e domésticos, gerando subsídios para propostas de conservação da fauna e controle de zoonoses como a Febre Maculosa Brasileira. Para o levantamento, quatro armadilhas de CO2 foram montadas em seis localidades na mata do Morro do Imperador, Juiz de Fora, MG, durante um ano, totalizando 12 coletas. Os ixodídeos foram coletados por monitoramento das armadilhas e vegetação adjacente durante o período de duas horas e acondicionados em etanol 70° GL. No laboratório, os carrapatos foram identificados sob esterioscopia. Ao final de um ano de estudo, foram coletados 2.122 ixodídeos, sendo 793 larvas (37,3%) e 1.277 ninfas (57, 8%) do gênero Amblyomma, além de 102 adultos (4,9%) do gênero Amblyomma. A espécie mais constante foiAmblyomma cajennense, presente em 83,33% das coletas (71 espécimes), seguido de Amblyomma dubitatum presente em 50% (30 espécimes) e Amblyomma calcaratum em 8,33% (um espécime). As observações mostram que, no ambiente estudado, os ixodídeos têm um padrão de distribuição agregado, possivelmente em função da distribuição agregada do recurso, no caso hospedeiros silvestres.
ABSTRACT The study of the ixodofauna in fragments of woods in an urban area is important to the knowledge of species of tick and the dynamic among wild and domestic hosts, allowing to subsidize proposals for the conservation of the fauna and the control of zoonoses such as Brazilian spotted fever. To accomplish the survey, CO2 traps were built up in six different sites of the forest, during a oneyear period, totalizing 12 collections. The ixodids were collected by monitoring the traps and the adjacent vegetation during two hours and put in ethanol 70° GL. At the laboratory they were identified under the stereoscopic optics. At the end of a one-year period of study 2.122 ixodids were colleted, being 793 larvae (37,3%) and 1.277 nymphs (57,8%) of the genus Amblyomma and 102 were adults (4,9%) of the Amblyomma species. The most constant species was Amblyomma cajennense, present in 83,33% of the collections (71 specimen) followed by Amblyomma dubitatum, present in 50% (30 specimen), and Amblyomma calcaratum, 8,33% (one specimen). The observations showed that in the observed environment the populations of Ixodidae have an aggregate distribution, possibly because of the aggregate distribution of the resource, in the case wild hosts.

Os mecanismos de virulência das amostras de Escherichia coli potencialmente patogênicas para aves (APEC) têm sido continuamente estudados e acredita-se ser multifatorial. Certas propriedades são associadas primariamente a amostras virulentas e vêm sendo identificadas em amostras de E. coli isoladas de aves. Neste estudo um total de 61 amostras de E. coli, isoladas de frangos de corte com problemas respiratórios, foram testadas através da Reação em Cadeia da Polimerase (PCR), para a presença dos genes responsáveis pela capacidade de adesão, fimbria P (papC) e fimbria F11 (felA), produção de colicinas (cvaC), presença de aerobactina (iutA), resistência sérica (iss), hemaglutinina temperatura sensível (tsh) e presença de dos antígenos capsulares K1 e K5 (kpsII). O gene iss foi detectado em 73,8%, tsh em 55,7%, iutA em 45,9%, felA em 39,3%, papC em 24,3%, cvaC em 23% e kpsII em 18%.
The virulence mechanisms of avian pathogenic Escherichia coli (APEC) have been continually studied and are believed to be multi-factorial. Certain properties are primarily associated with virulent samples and have been identified in avian isolates. In this study a total of 61 E. coli, isolates from chicken flocks with respiratory symptomatology, were probed by Polimerase Chain Reation (PCR) for the presence of genes responsible for the adhesion capacity, P fimbria (papC) e F11 fimbria (felA), colicin production (cvaC), aerobactin presence (iutA), serum resistance (iss), temperature-sensitive hemagglutinin (tsh), and presence of K1 and K5 capsular antigens (kpsII). The iss gene was detected in 73,8%, tsh in 55,7%, iutA in 45,9%, felA in 39,3%, papC in 24,3%, cvaC in 23% and kpsII in18%.

The syndrome of class II division I is considered as a clinical picture composed of various signs, morphological and functional syndromes. In most of the cases, it is caused by functional alterations of the stomatognathic apparatus leading to the appearance of mandibular retrognathia. To solve this problem, it is necessary the use of a functional orthopedic apparatus capable of restoring the functions and of transmitting adequate propioceptive stimuli to the muscles and tooth to attain an appropriate relation of the maxillae in vertical and sagittal sense. In order to give information to students and specialists, it is described an apparatus of functional orthopedics of the maxillae: the combined method L.S. 78 designed by the Uruguayan professor Indalecio Buño Loureiro, who combines the use of the active plaque and the universal activator. It is specifically indicated for the treatment of class II division I or class I with characteristics of the above.
El síndrome de clase II división 1 es considerado como un cuadro clínico integrado por varios signos, síntomas morfológicos y funcionales; en la mayoría de los casos se produce por alteraciones funcionales del aparato estomatognático, que conducen a la aparición de un retrognatismo mandibular. Para la solución de este problema, se hace necesario el uso de un aparato de ortopedia funcional, que sea capaz de reestablecer las funciones y transmitir estímulos propioceptivos adecuados a la musculatura y los dientes, logrando así una relación adecuada de los maxilares, tanto en sentido vertical como sagital. Con el objetivo de brindar información a estudiantes y especialistas, se realiza la descripción de un aparato de ortopedia funcional de los maxilares: el método combinado L.S. 78 diseñado por el profesor uruguayo Indalecio Buño Loureiro, el cual combina la utilización de la placa activa y el activador universal, y que está indicado específicamente para el tratamiento de la clase II división 1 o clase I con características de la anterior.
