Abstract Long-term-ecological-research (LTER) faces many challenges, including the difficulty of obtaining long-term funding, changes in research questions and sampling designs, keeping researchers collecting standardized data for many years, impediments to interactions with local people, and the difficulty of integrating the needs of local decision makers with “big science”. These issues result in a lack of universally accepted guidelines as to how research should be done and integrated among LTER sites. Here we discuss how the RAPELD (standardized field infrastructure system), can help deal with these issues as a complementary technique in LTER studies, allowing comparisons across landscapes and ecosystems and reducing sampling costs. RAPELD uses local surveys to understand broad spatial and temporal patterns while enhancing decision-making and training of researchers, local indigenous groups and traditional communities. Sampling of ecological data can be carried out by different researchers through standardized protocols, resulting in spatial data that can be used to answer temporal questions, and allow new questions to be investigated. Results can also be integrated into existing biodiversity networks. Integrated systems are the most efficient way to save resources, maximize results, and accumulate information that can be used in the face of the unknown unknowns upon which our future depends.

Abstract For human/SARS-CoV-2 interactome genes ACE2, TMPRSS2 and BSG, there is a convincing evidence of association in Asians with influenza-induced SARS for TMPRSS2-rs2070788, tag-SNP of the eQTL rs383510. This case illustrates the importance of population genetics and of sequencing data in the design of genetic association studies in different human populations: the high linkage disequilibrium (LD) between rs2070788 and rs383510 is Asian-specific. Leveraging on a combination of genotyping and sequencing data for Native Americans (neglected in genetic studies), we show that while their frequencies of the Asian tag-SNP rs2070788 is, surprisingly, the highest worldwide, it is not in LD with the eQTL rs383510, that therefore, should be directly genotyped in genetic association studies of SARS in populations with Native American ancestry.

RESUMO: O objetivo do presente estudo foi obter valores ecobiométricos para os tecidos materno-fetais caninos, usando a ultrassonografia de alta definição (HDUS), desenvolvendo fórmulas para prever a idade gestacional das cadelas. Doze cadelas saudáveis de raças braquicefálicas pesando 7-13kg e com idades entre um e quatro anos foram incluídas. Os fetos foram analisados diariamente com ultrassonografia convencional e de alta definição, do oitavo dia após a inseminação artificial até o dia do parto, utilizando o equipamento de ultrassom ACUSON S2000/SIEMENS e software específico. Estruturas embrionárias e fetais; diâmetro da vesícula embrionária; comprimento do embrião; diâmetro biparietal, comprimento abdominal e altura renal; comprimento do fêmur, úmero, escápula, rádio e tíbia; espessamento da parede do estômago; comprimento adrenal; e tamanho cardíaco foram medidos. Essas variáveis foram correlacionadas com o teste gestacional e ajustado de acordo com os modelos de regressão. Os resultados obtidos (P<0,001) para: diâmetro lateral do saco gestacional (R2=81,8%); comprimento do embrião (R2=85,7%); diâmetro biparietal (R2=99,1%) e abdominal (R2=97,2%); espessura da parede gástrica (R2=86,9%); comprimento do fêmur (R2=96,6%), rádio (R2=97,5%), úmero (R2=96,5%), escápula (R2=95,8%) e tíbia (R2=97,3%); comprimento do rim (R2=95,8%) e altura (R2=96,0%); comprimento adrenal (R2=89,6%); o comprimento do coração (R2=93,0%) e a altura (R2=91,5%) dos fetos caninos apresentaram correlação significativa com os dias gestacionais. Isso permitiu monitorar o crescimento fetal e estimar a idade com alta precisão em diferentes períodos gestacionais.

ABSTRACT: The objective of the present study was to obtain echobiometric values for the most important canine maternal-fetal tissues using high-definition ultrasonography (HDUS) trying to update the formulas for predicting gestational age. Twelve healthy bitches of brachycephalic breeds weighing 7-13kg and aged one to four years were included. The fetuses were analyzed every day using conventional and high-definition ultrasonography from the eighth day after artificial insemination until the day of delivery, using the ACUSON S2000/SIEMENS ultrasound equipment and specific software. Embryonic and fetal structures; diameter of the embryonic vesicle; length of the embryo; biparietal diameter, abdominal length and kidney height; length of the femur, humerus, scapula, radius, and tibia; thickening of the stomach wall; adrenal length; and cardiac size were measured. These variables were correlated with gestational and adjusted tested according to regression models. The results obtained (P<0.001) for: lateral diameter of the gestational sac (R2=81.8%); length of the embryo (R2=85.7%); biparietal (R2=99.1%) and abdominal diameter (R2=97.2%); thickness of the gastric wall (R2=86.9%); length of the femur (R2=96.6%), radius (R2=97.5%), humerus (R2=96.5%), scapula (R2=95.8%) and tibia (R2=97.3%); kidney length (R2=95.8%) and height (R2=96.0%); adrenal length (R2=89.6%); heart length (R2=93.0%) and height (R2=91.5%) of the canine fetuses showed significant correlation with gestational days. This allowed monitoring fetal growth and estimation of age with high accuracy in different gestational periods.

RESUMO Objetivou-se avaliar a suplementação com diferentes fontes de carboidratos não fibrosos (CNF) em vacas manejadas a pasto em dietas com alta ou baixa concentração de proteína não degradável no rúmen (PNDR). Doze vacas multíparas com média de 30kg leite/ dia e 45±23 dias em lactação foram utilizadas em delineamento quadrado latino 4x4 com arranjo fatorial 2 × 2. As fontes de CNF foram milho moído (MM) e polpa cítrica seca (PC). A PNDR da dieta foi alterada pela substituição parcial do farelo de soja da dieta de baixa PNDR (BPNDR) por grão de soja tostado e farelo de glúten de milho dieta de alta PNDR (APNDR). Foi observado maior consumo de matéria seca (CMS) nos tratamentos com PC quando comparado aos tratamentos com MM (19,2 e 17,7kg / dia de MS, respectivamente). O teor de gordura do leite foi menor para os tratamentos com BPNDR (3,4%) em comparação aos tratamentos com APNDR (3,6%). O teor de proteína do leite foi maior nos tratamentos com MM (2,9%) em relação aos tratamentos com PC (2,8%). Foram observados menores teores de nitrogênio ureico no leite dos animais alimentados com PC em relação aos com MM (17,8 e 21,2mg / dL, respectivamente). O uso de PC como fonte de CNF aumentou o consumo de matéria seca, bem como o teor de gordura do leite, embora não tenham sido observadas diferenças na produção de leite relacionada a alteração nos teores de PNDR.

ABSTRACT The objective for this study was to evaluate the supplementation with different non-fibrous carbohydrate (NFC) sources for grazing dairy cows in diets with high or low concentrations of rumen undegradable protein (RUP). Twelve multiparous cows averaging 30kg/d of milk production and 45±23 days in milk in a 4x4 Latin square design with 2 × 2 factorial arrangement were used. The difference in NFC content of the diets was achieved using ground corn (GC) or dried citrus pulp (DCP). Dietary RUP was altered by the partial replacement of soybean meal (LRUP) by roasted whole soybean and corn gluten meal (HRUP). Greater dry matter intake (DMI) was observed in DCP treatments compared to GC treatments (19.2 and 17.7kg/day of DM, respectively). Milk fat concentration was lower for LRUP treatments (3.4%) when compared to HRUP (3.6%). The milk protein concentration was greater for GC treatments (2.9%) in relation to DCP treatments (2.8%). Lower milk ureic nitrogen was observed when DCP diets were fed compared to those with GC (17.8 and 21.2mg/dL, respectively). The use of DCP as CNF source increased dry matter intake as well as milk fat concentration, although no differences in milk production were observed for RUP level.

Abstract This study aimed to analyze the possible effects of diflubenzuron on the associated insect fauna under laboratory, semi-field and field conditions. Laboratory bioassays were performed in aquariums with mortality readings (%) every 24h until 96h, and in semi-fields, insects were kept in basins with readings every 24h for up to 12 days, in triplicates and a control. In the field experiment, a collection was performed before the application of diflubenzuron in ten brick factory pits (25m²) and 15 post-application. The values of LC50 and LC90 for Chironomus were 2.77x10-3g/L and 0.86g/L, respectively, and for Buenoa, they were 0.019g/L and 0.92g/L, a strong relationship was observed between mortality and exposure time (r²>0.8) in all concentrations used. In semi-field, similar mean values of emergency inhibition were observed, except for Euthyplociidae, which was more sensitive. There was no significant difference between the data of richness and diversity of aquatic insects in the field experiment, considering an interval of 15 days (p=0.32). Finally, the data suggest that diflubenzuron may have a negative effect on the associated insect fauna, but in the field experiment the environmental conditions of Anopheles breeding sites may have affected its efficiency.

RESUMO: No Brasil, segundo o Programa Nacional de Controle e Erradicação da Brucelose e Tuberculose Animal (PNCEBT), do Ministério da Agricultura, Pecuária e Abastecimento (MAPA), os testes de rotina para o diagnóstico de tuberculose bovina são o teste cervical simples (TCC), o teste da prega caudal (TPC) e o teste cervical comparativo (TCC), sendo que o último também é utilizado como teste confirmatório. Um grupo de 53 animais oriundos de três rebanhos leiteiros de área de foco para tuberculose bovina que foram submetidos a vazio sanitário no Rio Grande do Sul foi submetido ao TCC. Os tecidos destes animais foram cultivados e as colônias resultantes confirmadas por PCR e sequenciamento de DNA. Dos 53 animais analisados no TCC, 32 (60,4%) foram negativos, 14 (26,4%) positivos e sete (13,2%) inconclusivos, com base no PNCEBT. O TCC detectou como positivos 11 dos 39 animais com infecção por M. bovis confirmada por cultivo. Do total de 14 animais não infectados, baseado na cultura, o TCC detectou oito como negativos. Assim, o TCC apresentou, para a população amostrada, sensibilidade de 28,2% e especificidade de 57,1%. Um total de 24/32 (75,0%) dos animais negativos ao TCC foi positivo no cultivo (confirmado por PCR), sendo considerados falso-negativos ao TCC. A manutenção destes animais falso-negativos nos rebanhos tem sérias implicações para o controle da enfermidade, já que os mesmos podem ser fonte de infecção. A adição de testes complementares poderia auxiliar na identificação destes animais, aumentando a cobertura diagnóstica.

ABSTRACT: According to the Brazilian National Program for the Control and Eradication of Animal Brucellosis and Tuberculosis (PNCEBT), the routine tests for the diagnosis of bovine tuberculosis in the country are the simple intradermal tuberculin test (SITT) of the Ministry of Agriculture, Livestock and Food Supply (MAPA), the caudal fold test and the comparative intradermal tuberculin test (CITT). The latter is also used as a confirmatory test. A group of 53 animals from three dairy herds in a focal area for bovine tuberculosis, that were submitted to depopulation in the state of Rio Grande do Sul, were submitted to the CITT. Tissues were cultured and the resulting colonies were confirmed by PCR and DNA sequencing. Among the 53 animals analyzed using the CITT, 32 (60.4%) were negative, 14 (26.4%) were positive and seven (13.2%) results were inconclusive. The CITT detected 11 of the 39 animals with culture-confirmed M. bovis infection as positive. Among the total of 14 uninfected animals based on cultures, the CBT detected eight as negative. Thus, the CITT demonstrated sensitivity of 28.2% and specificity of 57.1% for the population sampled. A total of 24/32 (75.0%) of the animals with negative CITT results were culture positive (confirmed by PCR) and were considered false negatives based on the CITT. The maintenance of these false-negative animals in herds has serious implications for the control of the disease, since they can be a source of infection. The addition of complementary tests could help identify such animals and increase the odds of diagnostic success.

Os cadinhos filtrantes (porosidade 1) originalmente usados na técnica in vitro semi-automática de produção de gases estabelecida no Laboratório de Produção de Gases da Escola de Veterinária da UFMG podem ocasionar interferências nos estudos de quantificação da degradação de alimentos, principalmente os ricos em amido, devido à obstrução dos poros da placa filtrante pelo material residual após fermentação. Este estudo objetivou avaliar a substituição dos cadinhos filtrantes por sacos F57 (Ankom(r)) visando a realização de experimentos in vitro para descrição da cinética de fermentação e degradação ruminal. Esta substituição causou a redução do peso/volume da amostra incubada e desta forma, interferiu na relação pressão (P) / volume (V) e consequentemente inviabilizou a utilização de equação previamente estabelecida por Mauricio et al. (2003). Desta forma, objetivou-se neste estudo estabelecer equação para estimar o volume de gases produzidos a partir de dados de P e V obtidos pela fermentação de sorgo forrageiro, incluindo partes da planta e planta inteira, incubados em sacos F57 (Ankom(r)). Foram quantificados 474 dados simultâneos de pressão e volume os quais variaram de 0 a 4.670 psi e os de volume entre 0 e 38mL. A equação relacionando P e V obtida foi: V (mL) = 0.747 (s.e. 0.0733) + 7.23 P (s.e. 0.1165) + 0.101 P2 (s.e. 0.0372), (R2 = 0.99). O desenvolvimento desta equação viabilizou a utilização dos sacos F57 (Ankom(r)) pela na técnica in vitro semi-automática de produção de gases e, portanto permitindo a quantificação dos resíduos da fermentação favorecendo a acurácia dos resultados.

Avaliaram-se suplementos nitrogenados contendo ou não monensina sódica, adicionados à silagem, sobre o consumo de matéria seca (CMS), a digestibilidade da MS (DMS) e a produção de vacas F1 Holandês x Zebu. Vacas (n=15) com 21 dias em lactação, produzindo 19,2kg/dia, foram distribuídas em um delineamento quadrado latino 5X5, com três vacas por tratamento (T). Os T foram: 1- concentrado durante as ordenhas e silagem de milho após as ordenhas (C.SM); 2- C.SM + nitromineral (C.SMNM); 3- C.SM + nitroproteico (C.SMNP), 4- C.SMNM + monensina (C.SMNM+MO) e 5- C.SMNP + monensina (C.SMNP+MO). Os suplementos NP e NM adicionados à SM, (C.SMNP e C.SMNM) aumentaram o CMS (P<0,05). A adição de monensina não alterou o CMS nem a DMS (P>0,05). A suplementação proteica não aumentou o acetato (P>0,05), mas aumentou o propionato e o butirato (P<0,05). A adição de MO ao T C.SMNP reduziu o propionato e o butirato (P<0,05) em relação ao T C.SMNP. A concentração de nitrogênio amoniacal aumentou (P<0,05) no T C.SMNM em relação ao T C.SM e não diferiu dos demais. As produções de leite nos T C.SMNP e C.SMNPMO foram maiores do que no T C.SM (P<0,05). Vacas do T C.SM foram mais eficientes produzindo leite do que as do T C.SMNM (P<0,05).

The objectives of this study were to evaluate consumption, ruminal parameters and milk production of F1 Holstein x Zebu milking cows fed nitrogenated supplements with or without monensin added to the silage. Cows with 21 days on milk and producing initially 19.2kg of milk/day were distributed in a 5X5 Latin square design with three replicates per treatment. The treatments (T) were: 1- concentrate fed during the milking time and corn silage fed after milking (C.SM); 2- C.SM + nitromineral(NM) (C.SMNM); 3- C.SM + nitroproteico (NP) (C.SMNP); 4- C.SMNM + monensin (C.SMNM+MO) e 5- C.SMNP + monensin (C.SMNP+MO). The addition of NP e NM to the T C.SM increased the DMI when compared to C.SM (P<0.05). The addition of monensin did not change the DMI (P>0,05). The protein supplementation did not alter acetate (P>0,05), but increased propionate and butyrate concentrations. (P<0.05) The N-NH3 concentration increased (P<0.05) only in T C.SMNM (P<0.05). The addition of monensin to T C.SMNP decreased propionate and butyrate concentrations (P<0.05) related to T C.SMNP. The milk production was greater in the C.SMNP treatments than T C.SM (P<0.05). Cows in T C.SM produced milk more efficiently than cows fed C.SMNM (P<0.05), but did not differ from the others.

Several forebrain and brainstem neurochemical circuitries interact with peripheral neural and humoral signals to collaboratively maintain both the volume and osmolality of extracellular fluids. Although much progress has been made over the past decades in the understanding of complex mechanisms underlying neuroendocrine control of hydromineral homeostasis, several issues still remain to be clarified. The use of techniques such as molecular biology, neuronal tracing, electrophysiology, immunohistochemistry, and microinfusions has significantly improved our ability to identify neuronal phenotypes and their signals, including those related to neuron-glia interactions. Accordingly, neurons have been shown to produce and release a large number of chemical mediators (neurotransmitters, neurohormones and neuromodulators) into the interstitial space, which include not only classic neurotransmitters, such as acetylcholine, amines (noradrenaline, serotonin) and amino acids (glutamate, GABA), but also gaseous (nitric oxide, carbon monoxide and hydrogen sulfide) and lipid-derived (endocannabinoids) mediators. This efferent response, initiated within the neuronal environment, recruits several peripheral effectors, such as hormones (glucocorticoids, angiotensin II, estrogen), which in turn modulate central nervous system responsiveness to systemic challenges. Therefore, in this review, we shall evaluate in an integrated manner the physiological control of body fluid homeostasis from the molecular aspects to the systemic and integrated responses.

The antioxidant and free radical scavenger properties of melatonin have been well described in the literature. In this study, our objective was to determine the protective effect of the pineal gland hormone against the DNA damage induced by cyclophosphamide (CP), an anti-tumor agent that is widely applied in clinical practice. DNA damage was induced in rats by a single intraperitoneal injection of CP (20 or 50 mg/kg). Animals received melatonin during the dark period for 15 days (1 mg/kg in the drinking water). Rat bone marrow cells were used for the determination of chromosomal aberrations and of formamidopyrimidine DNA glycosylase enzyme (Fpg)-sensitive sites by the comet technique and ofXpf mRNA expression by qRT-PCR. The number (mean ± SE) of chromosomal aberrations in pinealectomized (PINX) animals treated with melatonin and CP (2.50 ± 0.50/100 cells) was lower than that obtained for PINX animals injected with CP (12 ± 1.8/100 cells), thus showing a reduction of 85.8% in the number of chromosomal aberrations. This melatonin-mediated protection was also observed when oxidative lesions were analyzed by the Fpg-sensitive assay, both 24 and 48 h after CP administration. The expression of Xpf mRNA, which is involved in the DNA nucleotide excision repair machinery, was up-regulated by melatonin. The results indicate that melatonin is able to protect bone marrow cells by completely blocking CP-induced chromosome aberrations. Therefore, melatonin administration could be an alternative and effective treatment during chemotherapy.

The involvement of the hypothalamic-pituitary-adrenal axis in the control of body fluid homeostasis has been extensively investigated in the past few years. In the present study, we reviewed the recent results obtained using different approaches to investigate the effects of glucocorticoids on the mechanisms of oxytocin and vasopressin synthesis and secretion in response to acute and chronic plasma volume and osmolality changes. The data presented here suggest that glucocorticoids are not only involved in the mechanisms underlying the fast release but also in the transcriptional events that lead to decreased synthesis and secretion of these neuropeptides, particularly oxytocin, under diverse experimental conditions of altered fluid volume and tonicity. The endocannabinoid system, through its effects on glutamatergic neurotransmission within the hypothalamus and the nuclear factor κB-mediated transcriptional activity, seems to be also involved in the specific mechanisms by which glucocorticoids exert their central effects on neurohypophyseal hormone synthesis and secretion.

Determinaram-se a composição bromatológica do material original e das silagens de quatro genótipos de girassol (M742, MG4, PM92007 e VDH483) utilizados para produção de grãos, ensilados em diferentes idades de corte após o plantio e aos 90, 97, 104, 111, e 118 dias, e o melhor momento de colheita desses cultivares para ensilagem. Os teores de matéria seca variaram entre as idades e cultivares, sendo observados valores de 12,7% a 42,2% no material original e 11,1% e 40,2% na silagem. Os valores de proteína bruta variaram de 8,3% a 12,6% no material original e de 9,4% a 12,9% na silagem. Foi observada diminuição (P<0,05) nos teores de carboidratos com a ensilagem. Os teores de extrato etéreo do material original e das silagens variaram de 2,2 a 13,3% no material original e de 3,0 a 17,9% na ensilagem. Houve diminuição dos teores de hemicelulose com a ensilagem, sugerindo que essa fração foi utilizada com substrato adicional para fermentação. Os teores de ácido lático diminuíram com o estágio de maturação da planta. Houve diminuição nos valores de digestibilidade in vitro da matéria seca com o avanço do estádio de maturação da planta. O melhor momento de colheita para os genótipos M742, MG4 e VDH483 foi aos 104 dias de idade e para o genótipo PM92007, aos 111dias.

The bromatological composition of four sunflower genotypes (M742, MG4, PM92007 and VDH483) used for producing seed, ensiled at different harvest ages (90, 97, 104, 111 and 118 days) after sowing, was used to determine the best time to ensile. Differences among ages and cultivars were found for dry matter of the genotypes. The crude protein values varied from 8.3 to 12.6% for the original sample and from 9.4 to 12.9% for the silage. A decrease (P<0.05) in the soluble carbohydrate values for the ensiled genotypes was observed. The ether extract values varied from 2.2 to 13.3% for the original sample and from 3.0 to 17.9% for the silage. The values of hemicellulose and lignin decreased with the ensiling process, suggesting the fibrous fractions contributed, as additional substrate source, for fermentation. The lactic acid values decreased with the plant maturity stage. The dry matter in vitro digestibility decreased with the plant maturity. The best time to ensile, based on the quality of the sunflower genotype M742, MG4 and VDH483 silage, was at 104 days of age and for the genotype PM92007 silage was at 111 days of age.

Um delineamento inteiramente ao acaso, com três repetições, foi utilizado para avaliar os efeitos da adição de 0,5% de uréia, de 0,5% de carbonato de cálcio (CaCO3), de 0,5% de uréia mais 0,5% de CaCO3 e de inoculante bacteriano sobre o pH, teores de matéria seca (MS), proteína bruta (PB), fibra em detergente neutro (FDN), fibra em detergente ácido (FDA), hemicelulose, celulose, lignina, carboidratos solúveis, ácido lático e ácido acético sobre a digestibilidade in vitro da matéria seca (DIVMS) de silagens de quatro híbridos de sorgo (BR700, BR701, BR601 e AG2002). Os híbridos ensilados sem a adição de aditivos constituíram o grupo-controle. De cada genótipo foram coletadas três amostras do material original (MOR). Foram utilizados 60 silos de PVC com 10cm de diâmetro interno e 50cm de comprimento, sendo três por tratamento. A abertura dos silos ocorreu depois de 56 dias de fermentação. As silagens contendo CaCO3 e inoculante bacteriano apresentaram, em geral, características muito semelhantes às do controle. A adição de uréia e de uréia mais CaCO3 às silagens resultou em aumento (P<0,05) nos teores de PB. A adição conjunta de uréia e CaCO3 não propiciou aumento (P<0,05) superior ao obtido nas silagens contendo apenas uréia. Uréia pura, como aditivo, pode ser recomendada na ensilagem dos híbridos BR700, BR601 e AG2002.

A completely randomized design with three replicates was used to evaluate the addition effects of 0.5% urea, 0.5% calcium carbonate, 0.5% urea plus 0.5% calcium carbonate, and bacteria culture (according to producer recommendations) on quality of four hybrids sorghum silages (BR700, BR701, BR601 e AG2002). All hybrids ensiled with no additives were used as the control group. The pH and the contents of dry matter (DM), crude protein (CP), neutral detergent fiber (NDF), acid detergent fiber (ADF), hemicellulose, cellulose, lignin, soluble carbohydrates, lactic and acetic acids, and the in vitro dry matter digestibility (IVDMD) values were determined. Three samples of each original material (MOR) were collected and analyzed. Sixty "PVC" experimental silos with 10cm of internal diameter and 50cm of length, three by treatment, were used. The silos were open after 56 days of fermentation. The results of silages containing calcium carbonate or bacteria culture were, in general, similar to the control group (without additives). The addition of urea and urea plus calcium carbonate to silages improved their CP contents (P<0.05). The addition of urea plus calcium carbonate did not improve the CP contents of silages in relation to the addition of only urea. The addition of urea can be recommended for silages of BR700, BR601 and AG2002 hybrids.

DNA samples of six bovines obtained from three tissues (blood, semen and hair) were extracted using two different techniques. After the extraction procedures the samples were divided in six fractions. Three were stored at -20° C and three at 4° C. Every three months one sample of each tissue/extraction procedure was analyzed in spectrophotometer, to determine the quantity of the DNA and the extract was amplified using the primer RM 29. No differences in the DNA quantity or in the level of protein contamination among the three periods of analyses were observed. All the DNA extracted by quick extraction technique showed good amplification patterns during the nine months, meaning that this technique can be used in laboratory routine instead of the permanent extraction technique. The extract obtained from blood, using the permanent extraction technique, showed the higher quantity of DNA with the smaller index of protein contamination. The high quantity of protein contamination found in the semen samples preserved in egg yolk demanded modifications in both extraction techniques. After that the results were positive, showing good amplification patterns.

Quantificaram-se o consumo voluntário e a digestibilidade aparente da matéria seca, proteína bruta, energia bruta e balanço de nitrogênio das silagens de cinco genótipos de milho (HT01, HT47C, HT129, AG 5011 e BR 3123). Foram utilizados 15 carneiros alojados em gaiolas metabólicas para coleta total de fezes e urina. O delineamento experimental foi o inteiramente ao acaso com cinco tratamentos e seis repetições. Não foram observadas diferenças entre os genótipos quanto ao consumo e digestibilidade da MS, da EB e da PB (P>0,05). Os consumos de MS, EB e PB digestíveis e energia metabolizável também não foram diferentes entre os híbridos (P>0,05). Quanto às relações consumo de energia digestível/consumo de MS e consumo de energia metabolizável/consumo de MS, o genótipo AG5011 foi semelhante ao HT01 (P>0,05) e superior aos demais (P<0,05). Todos os tratamentos apresentaram balanço de nitrogênio positivo e não diferiram entre si (P>0,05). Todos os genótipos produziram silagens de bom valor nutritivo, entretanto o genótipo AG5011 apresentou maior eficiência na utilização da energia (P<0,05).

The voluntary intake and the apparent digestibility of dry matter, crude protein and crude energy and the nitrogen balance, of silages of five maize genotypes (HT01, HT47C, HT129, AG5011 and BR3123) were quantified. Fifteen sheep were stored in metabolic cages for total collection of feces and urine. A complete randomized design, with five treatments and six repetitions was used. The intake and digestibility of dry matter, crude protein and crude energy did not differ (P>0.05) among treatments. The intake of digestible dry matter, crude protein, crude energy and metabolic energy did not differ (P>0.05) among the genotypes. The ratios of digestible energy intake/dry matter intake and metabolic energy intake/dry matter intake of the genotype AG5011 silage was similar (P>0.05) to HT01, and higher than the other genotypes (P<0.05). All genotypes showed similar and positive nitrogen balance, and all of them produced silages of good nutritional value. Nevertheless, silage of AG5011 genotype showed higher efficiency of energy (P<0.05) utilization.
