RESUMO O objetivo deste trabalho foi avaliar a utilização periódica de testes de diagnóstico mais sensíveis aliados às práticas de manejo, visando ao controle eficaz da artrite encefalite caprina (CAE). Foram realizadas oito coletas de sangue em matrizes e reprodutores. Da primeira à sétima análise, as coletas foram quadrimestrais, utilizando-se os testes de imunodifusão em gel de agarose (IDGA), ensaio imunoenzimático indireto (ELISA-i) e Western Blot (WB). A oitava coleta aconteceu seis meses após a sétima, utilizando-se o WB e a reação em cadeia de polimerase (PCR). A prevalência da CAE foi de 6,8%, 14,9% e 39,2% no IDGA, ELISA-i e WB, respectivamente. Na última análise, foram detectados 0,9% de animais positivos pelo WB e 10,8% pela PCR. Apesar de não erradicarem a CAE, as medidas adotadas, aliadas à utilização periódica dos testes sorológicos e à combinação com a PCR, foram importantes para reduzir significativamente os animais soropositivos no rebanho.

ABSTRACT The aim of this study was to evaluate the periodic use of more sensitive diagnostic tests associated to management practices for the effective control of caprine arthritis-encephalitis (CAE). We carried out eight blood samples in does and bucks. From the first to the seventh analysis, the samples were quarterly, using Agarose Gel Immunodiffusion (AGID), Enzyme linked immunosorbent assay (i-ELISA) and Western Blot (WB) tests. The eighth collection was made six months after the seventh, using the WB and Polymerase Chain Reaction (PCR). The prevalence of CAE was 6.8%, 14.9% and 39.2% in the AGID, i-ELISA and WB respectively. The last analysis detected 0.9% of animals positive by WB and 10.8% by PCR. Although they do not eradicate CAE, steps taken together with the periodic use of serological tests and the combination with PCR were important to significantly reduce positive animals in the herd.

O exame de rotina de sêmen, o qual avalia a concentração de espermatozoides, a motilidade e a morfologia, pode não identificar defeitos sutis na arquitetura da cromatina de espermatozoides. Os touros parecem ter cromatina estável com baixos níveis de fragmentação do DNA. No entanto, a natureza da fragmentação e o seu impacto sobre a fertilidade ainda não estão claros e não há relatos que caracterizam a organização do DNA e os danos nessa espécie com mais detalhes. A seleção genética intensiva e o uso da inseminação artificial e da produção in vitro de embriões, além do processo de criopreservação, podem contribuir para o dano da cromatina, e sabe-se a importância da integridade do DNA espermático para o sucesso dessas tecnologias. Amostras de sêmen de três ejaculados de um touro Nelore com altos níveis de alterações morfológicas (55,8±5,1%) foram selecionadas para realização de exames complementares. Os danos de acrossoma (76,9±8,9%) e das membranas plasmáticas (75,7±9,3%), bem como quebras de fita de DNA de espermatozoides (13,8±9,5) e deficiência de protamina (3,7±0,6) foram significativamente maiores em comparação aos valores avaliados no sêmen de cinco touros Nelore com normospermia (24,3±3,3%; 24,5±6,1%; 0,6±0,5%; 0,4±0,6% para acrossoma, membrana plasmática, quebras de DNA e deficiência de protamina, respectivamente) (p<0,05). Motilidade e porcentagem de espermatozoides com baixo potencial mitocondrial não diferiram estatisticamente. Essas avaliações mostram que análises de sêmen de rotina (neste caso, morfologia) podem apontar para a extensão e a complexidade dos danos na célula espermática, o que indica que a deficiência de protamina e os danos no DNA podem ocorrer simultaneamente a defeitos morfológicos. Tal ocorrência enfatiza a importância das análises de sêmen clássicas e dos testes complementares.

The routine semen evaluation assessing sperm concentration, motility and morphology, does not identify subtle defects in sperm chromatin architecture. Bulls appear to have stable chromatin, with low levels of DNA fragmentation. However, the nature of fragmentation and its impact on fertility remain unclear and there are no detailed reports characterizing the DNA organization and damage in this species. The intensive genetic selection, the use of artificial insemination and in vitro embryo production associated to the cryopreservation process can contribute to the chromatin damage and highlights the importance of sperm DNA integrity for the success of these technologies. Frozen-thawed semen samples from three ejaculates from a Nellore bull showed high levels of morphological sperm abnormalities (55.8±5.1%), and were selected for complementary tests. Damage of acrosomal (76.9±8.9%) and plasma membranes (75.7±9.3%) as well as sperm DNA strand breaks (13.8±9.5%) and protamination deficiency (3.7±0.6%) were significantly higher compared to the values measured in the semen of five Nellore bulls with normospermia (24.3±3.3%; 24.5±6.1%; 0.6±0.5%; 0.4±0.6% for acrosome, plasma membrane, DNA breaks and protamine deficiency, respectively) (P<0.05). Motility and percentage of spermatozoa with low mitochondrial potential showed no differences between groups. This study shows how routine semen analyses (in this case morphology) may point to the length and complexity of sperm cell damage emphasizing the importance of sperm function testing.

Objetivou-se avaliar as características morfológica e funcional do sêmen bovino congelado comparando-se a eficácia de dois diferentes diluidores. O ejaculado de quatro touros foi dividido em duas partes iguais, uma submetida ao diluidor Tris e gema de ovo (A) e outra ao diluidor à base de lecitina de soja (Andromed®) (B). No experimento I, cinco palhetas dos diluidores A e B de cada touro foram descongeladas e avaliadas quanto à motilidade, vigor, concentração, morfologia espermática e teste de termor-resistência lento. Foram feitas, ainda, avaliação da integridade de membranas, por meio da associação das sondas iodeto de propídio, isotiocionato de fluoresceína - Pisum sativum e carbocianina catiônica lipofílica, e avaliação funcional da membrana plasmática com teste hiposmótico. A avaliação da integridade da cromatina foi realizada pelo método de coloração com laranja de acridina. No experimento II, o sêmen com os diferentes diluidores foi utilizado na fecundação in vitro, sendo observadas taxas de clivagem e desenvolvimento embrionário in vitro. Em relação aos resultados obtidos, apenas a porcentagem de espermatozoides no sêmen congelado foi discretamente maior com o diluidor A, concluindo-se que o diluidor composto por lecitina de soja pode substituir o composto por Tris e gema de ovo, respeitando-se as variações individuais de cada touro utilizado no presente experimento.

The goal of this study was to evaluate the protective effect of egg yolk compared with a soybean lecithin-based extender on morphologic and functional characteristics of frozen bovine semen. The ejaculate of four bulls was divided into two equal parts, one diluted with egg-yolk-Tris extender (A) and the other with a soy-lecithin based extender (Andromed®) (B). In experiment I, five straws of extender A and B from each bull were thawed and assessed regarding motility (subjective and computerized analysis), vigor, concentration, sperm morphology and slow thermoresistance (STR), evaluation of membrane integrity through association of propidium iodite probes (PI), fluorescein isotiocianate - Pisum sativum (FITC-PSA) and lipophilic cationic carbocyanine (JC-1) and functional evaluation of the plasmatic membrane through Hyposmotic Swelling Test (HOST). An evaluation of chromatin integrity was performed with the acridine orange staining procedure. In experiment II, the frozen semen with different extenders was used for in vitro fertilization (IVF), analyzing cleavage rates and in vitro embryo development. No statistical difference between extenders was identified, suggesting that the soy lecithin extender may substitute egg-yolk-Tris extender, considering the individual variations of each bull used in this experiment.

The objective of this study was to evaluate the effect of metabolic syndrome (MetS) and its individual components on the renal function of patients with type 2 diabetes mellitus (DM). A cross-sectional study was performed in 842 type 2 DM patients. A clinical and laboratory evaluation, including estimated glomerular filtration rate (eGFR) calculated by the modification of diet in renal disease formula, was performed. MetS was defined according to National Cholesterol Education Program - Adult Treatment Panel III criteria. Mean patient age was 57.9 ± 10.1 years and 313 (37.2%) patients were males. MetS was detected in 662 (78.6%) patients. A progressive reduction in eGFR was observed as the number of individual MetS components increased (one: 98.2 ± 30.8; two: 92.9 ± 28.1; three: 84.0 ± 25.1; four: 83.8 ± 28.5, and five: 79.0 ± 23.0; P < 0.001). MetS increased the risk for low eGFR (<60 mL·min-1·1.73 (m²)-1) 2.82-fold (95%CI = 1.55-5.12, P < 0.001). Hypertension (OR = 2.2, 95%CI = 1.39-3.49, P = 0.001) and hypertriglyceridemia (OR = 1.62, 95%CI = 1.19-2.20, P = 0.002) were the individual components with the strongest associations with low eGFR. In conclusion, there is an association between MetS and the reduction of eGFR in patients with type 2 DM, with hypertension and hypertriglyceridemia being the most important contributors in this sample. Interventional studies should be conducted to determine if treatment of MetS can prevent renal failure in type 2 DM patients.

Diabetic retinopathy has been associated with cardiac autonomic dysfunction in both type 1 and type 2 diabetes mellitus (DM) patients. Heart rate (HR) changes during exercise testing indicate early alterations in autonomous tonus. The aim of the present study was to investigate the association of diabetic retinopathy with exercise-related HR changes. A cross-sectional study was performed on 72 type 2 and 40 type 1 DM patients. Autonomic dysfunction was assessed by exercise-related HR changes (Bruce protocol). The maximum HR increase, defined as the difference between the peak exercise rate and the resting rate at baseline, and HR recovery, defined as the reduction in HR from the peak exercise to the HR at 1, 2, and 4 min after the cessation of the exercise, were determined. In type 2 DM patients, lower maximum HR increase (OR = 1.62, 95%CI = 1.03-2.54; P = 0.036), lower HR recovery at 2 (OR = 2.04, 95%CI = 1.16-3.57; P = 0.012) and 4 min (OR = 2.67, 95%CI = 1.37-5.20; P = 0.004) were associated with diabetic retinopathy, adjusted for confounding factors. In type 1 DM, the absence of an increase in HR at intervals of 10 bpm each during exercise added 100% to the odds for diabetic retinopathy (OR = 2.01, 95%CI = 1.1-3.69; P = 0.02) when adjusted for DM duration, A1c test and diastolic blood pressure. In conclusion, early autonomic dysfunction was associated with diabetic retinopathy. The recognition of HR changes during exercise can be used to identify a high-risk group for diabetic retinopathy.

Os ácaros do gênero Brevipalpus colonizam um grande número de espécies frutíferas e ornamentais e têm sido associados a viroses de muitas plantas. A leprose dos citros é a virose de maior importância econômica transmitida por esses ácaros. Recentemente foi comprovado que algumas plantas ornamentais podem ser hospedeiras alternativas desse vírus. Os grandes volumes e a alta movimentação das plantas ornamentais fazem com que estas ajam como eficientes disseminadoras de pragas. Por essa razão, é necessário ampliar o conhecimento sobre pragas potenciais, de modo a subsidiar a implementação de medidas quarentenárias. Neste trabalho são relatadas as plantas ornamentais hospedeiras de ácaros Brevipalpus no Distrito Federal, assim como a ocorrência de sintomas típicos de viroses a elas transmitidas por Brevipalpus. Entre julho e setembro de 2005, foram realizadas cinco coletas em 14 localidades do DF. Foram amostradas folhas e ramos de 55 espécies de ornamentais. Pithecellobium avaremotemo Mart. é relatada, pela primeira vez, como hospedeira das espécies B. phoenicis (Geijskes), B. californicus Banks e B. obovatus Donnadieu. Adicionalmente, são relatadas sete novas espécies como hospedeiras de Brevipalpus na América do Sul. Novos hospedeiros são também listados para cada uma das espécies. Sintomas típicos de viroses transmitidas por Brevipalpus foram observados em Ligustrum sinense Lour., Pelargonium hortorum L.H. Bailey, Hibiscus rosa-sinensis L. e orquídeas (Dendrobium e Oncidium). Os resultados deste trabalho enfatizam o papel potencial de plantas ornamentais como veículo para disseminação de ácaros Brevipalpus.

Brevipalpus mites colonize a great number of fruit and ornamental plants. Mite species belonging to this genus have been associated with many plant viruses. Citrus leprosis (CiLV) is the most economically important virus transmitted by B. phoenicis mites. It has recently been shown that ornamental plant species can be alternative hosts of this virus. The high volume of trade and frequent movement of live ornamental plants make them efficient pest disseminators. Because of this, it is desirable to expand knowledge of potential pests aiming to guide the adoption of quarantine measures. This work reports ornamental plant hosts of Brevipalpus mites in the Distrito Federal (DF), as well the occurrence of symptoms consistent with Brevipalpus-borne plant viruses in these same hosts. Between July and September of 2005, five surveys were carried out in 14 localities within DF. Leaves and branches of fifty-five ornamental plant species were sampled. The species Pithecellobium avaremotemo Mart. is for the first time reported as a host for B. phoenicis (Geijskes), B. californicus Banks and B. obovatus Donnadieu species. Additionally, seven new species are reported as hosts for Brevipalpus within South America. New hosts are also listed for individual mite species. Typical symptoms of Brevipalpus-borne viruses were observed in Ligustrum sinense Lour., Pelargonium hortorum L.H. Bailey, Hibiscus rosa-sinensis L. and orchids (Dendrobium and Oncidium). The results of this work emphasize the potential role of the ornamental plants as vehicles for dissemination of Brevipalpus mites.

RESUMO O objetivo desse trabalho foi verificar a ação de marcas diferentes de estreptomicinas polivalentes para o tratamento da leptospirose bovina em dose única (25 mg de estreptomicina por kg de peso corpóreo). O trabalho foi realizado com 14 touros adultos sorologicamente reagentes para Leptospira interrogans sorovar Hardjo, com título mínimo de 800 e com cultivo positivo. Os animais foram separados em 2 grupos de acordo com a marca da estreptomicina utilizada no tratamento, grupo 1: estreptomicina A e grupo 2: estreptomicina B. Os bovinos controles não receberam nenhum tratamento. Foram obtidas amostras de sangue e urina dos animais tratados e controles nos dias -1, 0, 1, 2, 3, 5, 10, 15 e 25; considerando-se o dia do tratamento como dia 0. Na urina dos bovinos tratados e controle foi realizada a reação em cadeia da polimerase (PCR) e isolamento com inoculação em hamsters. Observou-se que a estreptomicina da marca A, em dose única de 25 mg/kg de peso corpóreo, conseguiu eliminar a leptospirúria no grupo de touros após 24h do tratamento. Já no grupo de touros tratados com a estreptomicina da marca B, foi constatado a leptospirúria entre 48h e 72h, após o tratamento. Em ambos os grupos tratados, a resposta sorológica apresentou uma variação da queda dos níveis dos títulos de anticorpos aglutinantes, sendo que embora a estreptomicina A tenha aparentemente apresentado um melhor desempenho quando comparada com as médias geométricas dos títulos do grupo tratado com a estreptomicina B as médias não diferiram entre si pelo Teste de Tukey (P > 0,05). Nos bovinos tratados a leptospirúria foi intermitente e a média geométrica dos títulos foi menor que a média geométrica dos títulos dos bovinos controles. A diferença do efeito da ação das diferentes marcas de estreptomicina está possivelmente na qualidade da matéria prima produzida pelos laboratórios.

ABSTRACT The objective of this work was evaluated different marks of streptomycin for the treatment of bovine leptospirosis, in an only one dose of 25 mg for kg of corporeal weight. Thirty serologically reactive bulls for Leptospira interrogans serovar Hardjo were utilized. The bulls had a minimum antibody titer of 800 and were positive in the isolation from urine samples. The animals had been separated in 2 groups according with the streptomycin mark used in the treatment, group 1: streptomycin A and group 2: streptomycin B. The bovine controls had received any application from streptomycin. Samples of blood and urine of treated and controls bulls were collected in days -1, 0, 1, 2, 3, 5, 10, 15 and 25; considering the day of the treatment as day 0. A research for the detection of leptospira genetic material was realized in the urine of all bulls by PCR. It was observed that the streptomycin A ended leptospiruria in 24h after of the treatment. In the group of bulls treated with the streptomycin B, the end of leptospiruria was evidenced in 48h and 72h, after the treatment. In both treated groups, the serological reply presented a variation in the fall of antibodies titers, and streptomycin A was better in comparative performance with geométric means of the antibodies titers of the group that received streptomycin B, although by the Tukey Test the difference was no significant (P > 0.05). In the bovine controls the leptospiruria was intermittent and the average of the antibody titers of the treated bovines was lower than the average of the bovine controls, during all the experiments. The difference in the effect of different brands of streptomycin is possibly due to the quality of the substances produced in the laboratories.
