Nonalcoholic fatty liver disease in women with polycystic ovary syndrome: associated factors and noninvasive fibrosis staging in a single Brazilian center

ABSTRACT Objective Polycystic ovary syndrome (PCOS) is a recognized risk factor for nonalcoholic fatty liver disease (NAFLD). The aims of this study were to investigate the prevalence and factors associated with NAFLD in women with PCOS and evaluate noninvasive indices of hepatic fibrosis in patients with PCOS and NAFLD. Subjects and methods Patients with PCOS (n = 87) and women without PCOS (n = 40; controls) were included. NAFLD was diagnosed by abdominal ultrasonography after exclusion of alcohol consumption and viral or autoimmune liver disease. Anthropometric, clinical and metabolic variables, homeostasis model assessment of insulin resistance (HOMA-IR) index, lipid accumulation product (LAP), FIB-4 index, NAFLD score, and transient elastography (TE; FibroScan) were obtained in subsets of patients with PCOS and NAFLD. Results A total of 87 patients with PCOS were included (mean age: 34.4 ± 5.7 years, mean body mass index [BMI]: 34.7 ± 4.7 kg/m 2 ). NAFLD was present in 67 (77.0%) patients with PCOS versus 21 of 40 (52.5%) controls (p = 0.005). Women with PCOS and liver steatosis, compared with their NAFLD-free counterparts, had higher values of BMI, waist circumference, triglycerides, total cholesterol, alanine and aspartate aminotransferases, and γ-glutamyltransferase, along with higher frequencies of obesity, metabolic syndrome, and insulin resistance. NAFLD was independently associated with waist circumference, serum triglycerides, and alanine aminotransferase levels. The FIB-4 index was not compatible with advanced fibrosis in any of the evaluated patients, while NAFLD score and TE were compatible with advanced liver fibrosis in 1 of 26 (3.8%) and 3 of 25 (12%) patients, respectively. Conclusion Women with PCOS had a high risk of NAFLD, and a combination of both was associated with central obesity, dyslipidemia, insulin resistance, and metabolic syndrome. Noninvasive methods suggested low rates of severe hepatic fibrosis in Brazilian women with PCOS. Arch Endocrinol Metab. 2020;64(3):235-42

Abstract In this study, phenotypic methods presented >80% agreement with the molecular identification of 59 Candida parapsilosis complex. Growth at 15% NaCl or pH 7.0 significantly reduced cfu-counts of Candida orthopsilosis, suggesting these conditions may support the development of phenotypic methods for the differentiation of the cryptic species of C. parapsilosis complex.

Nectar robbing in Collaea cipoensis (Fabaceae), an endemic shrub of the Brazilian rupestrian grasslands

Abstract: Nectar robbery is common in flowering plants with tubular corollas and can affect plant reproductive success. Our study characterized the interaction between potential pollinators and nectar robbers, and assessed the effects on flower abortion in a population of the restricted endemic Collaea cipoensis (Fabaceae) at Serra do Cipó, Brazil. We conducted observations of floral visitors to identify potential pollinators from nectar robbers and described visitor behavior. We also analyzed the frequency of robbery and its relationship with flower visitation. Trigona spinipes and Apis mellifera were identified as the main nectar robbers and two hummingbird species Colibri serrirostris and Eupetomena macroura, as potential pollinators. Overall, 570 sampled flowers presented robber damage while 716 were intact. Intact flowers received more visits than flowers with damage. Mean flower abortion ratio did not vary between damaged and intact flowers. Accordingly, a survey of developed fruits indicated that 86% of fruits came from intact flowers. Our results suggest that the damage by nectar robbers in C. cipoensis flowers could affect the reproductive success of this species, interfering with fruit production.
Resumen: El robo de néctar es común en plantas con flores de corolas tubulares y puede afectar su éxito reproductivo. Se caracteriza la interacción entre robadores de néctar y polinizadores, así como sus consecuencias en la aborción de flores en una especie endémica, Collaea cipoensis (Fabaceae) en la Serra do Cipó, Brasil. Se realizaron observaciones de visitantes florales para identificar a los polinizadores, así como a los robadores de néctar basados en su conducta. Se analizó la frecuencia de robo y su relación con los visitantes florales. Trigona spinipes y Apis mellifera fueron los principales ladrones de néctar y 2 especies de colibrí Colibri serrirostris y Eupetomena macroura, como polinizadores potenciales. Se presentó daño en 570 flores y 716 estuvieron intactas. Las flores sin daño recibieron más visitas vs. las flores dañadas. El promedio de flores abortadas no difirió entre flores dañadas vs. intactas. El 86% de los frutos proviene de flores intactas. Los resultados sugieren que el robo de néctar en C. cipoensis podría afectar negativamente el éxito reproductivo de esta especie interfiriendo con la producción de frutos.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The Port of Salvador (12°58'S, 38°30'W) receives cargo ships from different regions such as southeast Asia, North Atlantic, Mediterranean, Africa, North and South America. Thus, the presence of this port enhances the probability of new species arriving and establishing in Todos os Santos Bay (TSB), in which the port is located. Ascidians are sessile filter-feeding invertebrates with short lived larvae and thus are good indicators of bioinvasion. We surveyed the ascidian fauna on three different occasions: August 1999, June 2004 and December 2007. Nineteen species were identified belonging to the following families: Ascidiidae (Phallusia nigra, Ascidia cf. multitentaculata, A. nordestina, A. papillata, A. scalariforme, A. cf. tapuni, A. tenue), Corellidae (Rhodosoma turcicum), Pyuridae (Microcosmus anchylodeirus, M. exasperatus, M. helleri, Pyura vittata, Herdmania pallida), and Styelidae (Polycarpa cf. reviviscens, P. spongiabilis, P. tumida, Polycarpa sp., Styela canopus, Cnemidocarpa irene). Only A. nordestina, A. papillata, A. scalariforme and P. spongiabilis are possibly native to this region, while P. tumida and C. irene were classified as introduced. Microcosmus anchylodeirus, H. pallida, P. vittata, M. exasperatus, M. helleri, S. canopus, A. cf. multitentaculata, A. tenue and P. nigra were classified as cryptogenic. Most are widely distributed in all oceans and their native geographic distribution is unknown, while A. cf. multitentaculata and A. tenue occur only in the Atlantic, with disjunct distributions. Polycarpa cf. reviviscens, P. tumida and M. anchylodeirus are registered for the first time on the coast of Brazil. Colonial ascidians from this collection are still being studied. The large number of cryptogenic and introduced species indicates the necessity of monitoring TSB for expansion of these populations and the need for the establishment of control programs.

Substratos artificiais em regiões de portos e marinas geralmente abrigam muitas espécies introduzidas e sua colonização constitui o primeiro passo no estabelecimento de bioinvasores. O grau de movimentação do substrato influencia a comunidade incrustante e o conhecimento da assembléia de espécies associada a cada situação é crucial como ferramenta de manejo. Neste trabalho, reportamos a estrutura de comunidades de seis meses desenvolvidas em substratos de granito na baía de Paranaguá. Foram encontradas 12 espécies na condição flutuante (profundidade constante) e 15 na condição fixa (profundidade variável), mas o número médio de espécies por placa não foi diferente nos tratamentos. A comunidade das placas flutuantes foi dominada pelo bivalve nativo Mytella charruana (66.1 ± 5.5% de cobertura), enquanto as placas fixas foram dominadas pelos cirripédios Fistulobalanus citerosum (49.8 ± 3.5%) e Amphibalanus reticulatus (33.9 ± 3.7%), este último introduzido na região. Outras espécies introduzidas encontradas foram Garveia franciscana, em apenas uma placa fixa, e Megabalanus coccopoma também em uma placa fixa e em metade das placas flutuantes, mas sempre com baixa cobertura (< 2%). Em conclusão, placas fixas foram mais suscetíveis às espécies introduzidas, uma delas ocorrendo em alta abundância, o que sugere que este tipo de substrato deveria ser priorizado em ações de controle e manejo de bioinvasão. Também hipotetizamos que o bivalve nativo M. charruana é o competidor dominante por espaço na condição flutuante, reduzindo a susceptibilidade deste substrato à bioinvasão.
Artificial substrates in and near ports and marinas commonly have many non-indigenous species and are the first stepping stone for the establishment of bioinvasors. Substrate movement influences fouling communities and so understanding of how species assemblages are related to specific substrate conditions is crucial as a management tool. Here we describe the species assemblage of the community after six months of development on granite plates in Paranaguá Bay. Species richness was similar in the two treatments, with 12 species on floating (constant depth) plates and 15 on stationary (variable depth) plates. However, species composition differed, with the community on floating plates being dominated by the native bivalve Mytella charruana (66.1 ± 5.5% cover) and that on stationary plates dominated by the barnacles Fistulobalanus citerosum (49.8 ± 3.5% cover) and the introduced Amphibalanus reticulatus (33.9 ± 3.7% cover). Other introduced species were Garveia franciscana, on one stationary plate, and Megabalanus coccopoma also on one stationary plate and not very abundant on half of the floating plates (< 2%). Thus, stationary plates were more susceptible to introduced species that may become very abundant, suggesting that this type of substrate should be a priority in management for bioinvasion control. We also hypothesize that the native bivalve M. charruana is the dominant competitor for space on floating substrates, thereby reducing the invasiveness of that type of substrate.

The investigation of extracts from six species of marine invertebrates yielded one new and several known natural products. Isoptilocaulin from the sponge Monanchora aff. arbuscula displayed antimicrobial activity at 1.3 mg/mL against an oxacillin-resistant strain of Staphylococcus aureus. Five inactive known dibromotyrosine derivatives, 2 6, were isolated from a new species of marine sponge, Aplysina sp. The sponges Petromica ciocalyptoides and Topsentia ophiraphidites yielded the known halistanol sulfate A (7) as an inhibitor of the antileishmanial target adenosine phosphoribosyl transferase. The ascidian Didemnum ligulum yielded asterubin (10) and the new N,N-dimethyl-O-methylethanolamine (11). The octocoral Carijoa riisei yielded the known 18-acetoxypregna-1,4,20-trien-3-one (12), which displayed cytotoxic activity against the cancer cell lines SF295, MDA-MB435, HCT8 and HL60.

No presente estudo apresentamos resultados da triagem biológica realizada com 349 extratos obtidos de esponjas marinhas, ascídias, briozoários e octocorais do Brasil, em testes contra 16 linhagens de bactérias comuns e resistentes à antibióticos, uma levedura (Candida albicans), Mycobacterium tuberculosis H37Rv, três linhagens de células tumorais MCF-7 (mama), B16 (melanoma murínico) e HCT8 (cólon), e de inibição da enzima adenina fosforribosil transferase de Leishmania tarentolae (L-APRT). Menos de 15% dos extratos de esponja marinhas apresentaram atividade antibacteriana, contra linhagens resistentes ou não a antibióticos. Quase 40% dos extratos de esponjas marinhas apresentaram atividade antimicobacteriana contra Mycobacterium tuberculosis H37Rv. Foi observada citotoxicidade para 18% dos extratos de esponjas marinhas. Finalmente, menos de 3% dos extratos de esponjas apresentaram atividade inibitória da enzima L-APRT. Menos de 10% dos extratos de ascídias apresentaram atividade antibacteriana. Mais de 25% dos extratos de ascídias apresentaram atividade contra M. tuberculosis e as três linhagens de células tumorais. Somente dois extratos obtidos da ascídia Polysyncraton sp. coletada em duas diferentes épocas (1995 e 1997) apresentaram atividade contra L-APRT. Menos de 2% dos extratos de briozoários e octocorais apresentaram atividade antibacteriana, mas uma alta percentagem de extratos destes animais apresentaram atividades citotóxica (11% e 30%, respectivamente) e antimicobacteriana (60%). O extrato de somente uma espécie de briozoário, Bugula sp., apresentou atividade inibitória da enzima L-APRT. A análise taxonômica de algumas espécies de invertebrados que forneceram alguns dos extratos mais ativos, indicou a ocorrência de atividade biológica em espécies pertencentes a grupos taxonômicos que já foram anteriormente investigados do ponto de vista químico. Estes incluem esponjas marinhas pertencentes aos gêneros Aaptos, Aplysina, Callyspongia, Haliclona, Niphates, Cliona, Darwinella, Dysidea, Ircinia, Monanchora e Mycale, ascídias dos gêneros Didemnum, Aplidium, Botrylloides, Clavelina, Polysyncraton e Symplegma, o briozoário Bugula sp. e octocorais dos gêneros Carijoa e Lophogorgia. A subseqüente investigação química de alguns dos extratos ativos levou ao isolamento de vários metabólitos secundários biologicamente ativos. Os resultados obtidos estão de acordo com resultados obtidos em programas de triagem de atividade biológica de extratos brutos de invertebrados marinhos anteriormente reportados, que apresentaram altas percentagens de extratos bioativos oriundos de Porifera, Ascidiacea, Cnidária e Bryozoa.
Herein we present the results of a screening with 349 crude extracts of Brazilian marine sponges, ascidians, bryozoans and octocorals, against 16 strains of susceptible and antibiotic-resistant bacteria, one yeast (Candida albicans), Mycobacterium tuberculosis H37Rv, three cancer cell lines MCF-7 (breast), B16 (murine melanoma ) and HCT8 (colon), and Leishmania tarentolae adenine phosphoribosyl transferase (L-APRT) enzyme. Less than 15% of marine sponge crude extracts displayed antibacterial activity, both against susceptible and antibiotic-resistant bacteria. Up to 40% of marine sponge crude extracts displayed antimycobacterial activity against M. tuberculosis H37Rv. Cytotoxicity was observed for 18% of marine sponge crude extracts. Finally, less than 3% of sponge extracts inhibited L-APRT. Less than 10% of ascidian crude extracts displayed antibacterial activity. More than 25% of ascidian crude extracts were active against M. tuberculosis and the three cancer cell lines. Only two crude extracts from the ascidian Polysyncraton sp. collected in different seasons (1995 and 1997) displayed activity against L-APRT. Less than 2% of bryozoan and octocoral crude extracts presented antibacterial activity, but a high percentage of crude extracts from bryozoan and octororal displayed cytotoxic (11% and 30%, respectively) and antimycobacterial (60%) activities. The extract of only one species of bryozoan, Bugula sp., presented inhibitory activity against L-APRT. Overall, the crude extracts of marine invertebrates herein investigated presented a high level of cytotoxic and antimycobacterial activities, a lower level of antibacterial activity and only a small number of crude extracts inhibited L-APRT. Taxonomic analysis of some of the more potently active crude extracts showed the occurrence of biological activity in taxa that have been previously chemically investigated. These include marine sponges belonging to genera Aaptos, Aplysina, Callyspongia, Haliclona, Niphates, Cliona, Darwinella, Dysidea, Ircinia, Monanchora and Mycale, ascidians of the genera Didemnum, Aplidium, Botrylloides, Clavelina, Polysyncraton and Symplegma, the bryozoan Bugula sp. and octocorals of the genera Carijoa and Lophogorgia. The subsequent chemical investigation of some of the active extracts led to the isolation of several new biologically active secondary metabolites. Our results are in agreement with previous screening programs carried out abroad, that showed a high percentage of bioactive extracts from Porifera, Ascidiacea, Cnidaria and Bryozoa.

Hemodynamic parameters and neurogenic pulmonary edema following spinal cord injury: an experimental model

Edema pulmonar neurogênico é complicação séria e aumenta o risco de vida em pacientes com várias lesões do sistema nervoso central. Apresentamos uma experiência com 58 ratos Wistar machos e adultos. Foram formados dois grupos: controle (n=4) e experimental (n=54). O grupo experimental sofreu trauma raquimedular torácico médio com o cateter-balão de Fogarty contendo 20µL de salina por 5, 15, 30 ou 60 segundos de compressão. Os ratos foram anestesiados com pentobarbital sódico (p.s.), 60 mg/Kg intraperitoneal (i.p.). Foi investigada a relação entre a lesão medular e o tempo de compressão. A evolução neurológica foi quantificada e apresentada com 4, 24 e 48 horas da compressão para caracterizar a graduação da lesão nos diferentes grupos. A pior evolução ocorreu com 60 segundos de compressão. Seis animais morreram subitamente com edema pulmonar. Vinte ratos foram randomicamente distribuídos em um dos seguintes grupos: controle (1, n=4, anestesiados com p.s. i.p., 60 mg/Kg, mas sem compressão) e experimental (2, n=7, anestesiados com xilasina 10 mg/Kg e ketamina 75 mg/Kg) e (3, n=9, anestesiados com p.s. i.p., 60 mg/Kg). O índice pulmonar (100 x peso pulmonar / peso corporal) foi 0,395 ± 0,018 no grupo controle, 0,499 ± 0,060 no grupo 2, e 0,639 ± 0,14 no grupo 3. O exame histológico da medula espinhal mostrou rupturas no parênquima e hemorragia aguda. Comparando-se o índice pulmonar com o índice morfométrico através da microscopia óptica, evidenciou-se que ocorreu edema intra-alveolar relevante para índices acima de 0,55. A presente experiência sugere que o edema pulmonar induzido pela compressão medular é de natureza neurogênica e que o tipo de anestesia utilizada no experimento poderia ter participação na gênesis do edema pulmonar.
Neurogenic pulmonary edema is a serious and always life-threatening complication following several lesions of the central nervous system. We report an experiment with 58 Wistar-Hanover adult male rats. Two groups were formed: control (n=4) and experimental (n=54). The experimental group sustained acute midthoracic spinal cord injury by Fogarty’s balloon-compression technique containing 20µL of saline for 5, 15, 30 or 60 seconds. The rats were anesthetized by intraperitoneal (i.p.) sodium pentobarbital (s.p.) 60 mg/Kg. The quantitative neurological outcome was presented at 4, 24 and 48 hours from compression to characterize the injury graduation in different groups. Poor outcome occurred with 60 seconds of compression. Six animals died suddenly with pulmonary edema. Using the procedure to investigate the pulmonary edema during 60 seconds of compression, followed by decompression and time-course of 60 seconds, 20 rats were randomly asigned to one of the following groups: control (1, n=4, anesthetized by i.p. s.p., 60 mg/Kg but without compression) and experimental (2, n=7, anesthetized by i.p. xylazine 10 mg/Kg and ketamine 75 mg/Kg) and (3, n=9, anesthetized by i.p. s.p., 60 mg/Kg). The pulmonary index (100 x wet lung weight / body weight) was 0.395 ± 0.018 in control group, rose to 0.499 ± 0.060 in group 2, and was 0.639 ± 0.14 in group 3. Histologic examination of the spinal cord showed parenchymal ruptures and acute hemorrhage. Comparison of the pulmonary index with morphometric evaluation of edema fluid-filled alveoli by light microscopy showed that relevant intra-alveolar edema occurred only for index values above 0.55. The results suggest that the pulmonary edema induced by spinal compression is of neurogenic nature and that the type of anesthesia used might be important for the genesis of lung edema.

Two new marine metabolites, 3Z, 6Z, 9Z-dodecatrien-1-ol (1) from the ascidian Botrylloides giganteum and 4H-pyran-2ol acetate from the sponge Ircinia felix (4) are herein reported. The known bromotyrosine compounds, 2-(3,5-dibromo-4-methoxyphenyl)-N,N,N-dimethylethanammonium (2) and 2,6-dibromo-4-(2-(trimethylammonium)ethyl)phenol (3), have been isolated from the sponge Verongula gigantea. Serotonin (5) is reported for the first time from the sponge Cliona delitrix, and tambjamines A (15) and D (16) isolated as their respective salts from the nudibranch Tambja eliora. Only tambjamine D presented cytotoxicity against CEM (IC50 12.2 µg/mL) and HL60 (IC50 13.2 µg/mL) human leukemya cells, MCF-7 breast cancer cells (IC50 13.2 µg/mL), colon HCT-8 cancer cells (IC50 10.1 µg/mL) and murine melanoma B16 cancer cells (IC50 6.7 µg/mL).

Only rarely do myxomas originate from the mitral valve. This is the report of a 49-year-old woman presenting with congestive heart failure. The diagnosis of an intracardiac tumor involving the anterior cuspid of the mitral valve was made by transesophageal echocardiography. The patient underwent surgery for tumor resection and plasty of the valve was made with reconstruction and preservation of the valve. The diagnosis of myxoma was confirmed by histology. This is the 23rd case of myxoma of the mitral valve reported in the literature.
