Resumo Os microRNAs (miRNAs) são reconhecidos como biomarcadores do diabetes mellitus tipo 2 (DM2), úteis para a compreensão do metabolismo da doença, e possuem grande potencial como alvos terapêuticos. O aumento da expressão de BDNF e IGF1 está altamente envolvido nos benefícios as vias de insulina e glicose, porém, são regulados negativamente em condições de resistência à insulina, enquanto seu aumento de expressão está correlacionado com a melhora do metabolismo da glicose e da insulina. Estudos sugerem a regulação desses genes por microRNA em vários contextos diferentes, proporcionando uma nova abordagem de investigação para compreender o metabolismo do DM2 e revelar potenciais alvos terapêuticos. No presente estudo, investigamos em diferentes modelos animais (humanos, ratos e camundongos) miRNAs que têm como alvo BDNF e IGF1 em tecido muscular esquelético com condições fisiológicas de DM2. As análises foram realizadas utilizando ferramentas de bioinformática e bancos de dados para predição de miRNA, homologia molecular, validação experimental de interações, expressão na condição fisiológica estudada e interação em rede. Os resultados mostraram três candidatos a miRNAs para IGF1 (miR-29a, miR-29b e miR-29c) e um para BDNF (miR-206). As avaliações experimentais e a busca pela expressão no músculo esquelético de indivíduos com DM2 confirmaram a interação prevista entre miRNA-mRNA para miR-29b e miR-206 através de modelos humanos, ratos e camundongos. Essa interação foi reafirmada em múltiplas análises de rede. Em conclusão, nossos resultados mostram a relação de regulação entre miR-29b e miR-206 com os genes investigados, em diversos tecidos, sugerindo um padrão de inibição. Contudo, esses dados mostram um grande número de possíveis processos fisiológicos de interação para perspectivas biotecnológicas.

Abstract microRNAs (miRNAs) are recognized as diabetes mellitus type 2 (T2DM) biomarkers useful for disease metabolism comprehension and have great potential as therapeutics targets. BDNF and IGF1 increased expression are highly involved in the benefits of insulin and glucose paths, however, they are down-regulated in insulin resistance conditions, while their expression increase is correlated to the improvement of glucose and insulin metabolism. Studies suggest the microRNA regulation of these genes in several different contexts, providing a novel investigation approach for comprehending T2DM metabolism and revealing potential therapeutic targets. In the present study, we investigate in different animal models (human, rat, and mouse) miRNAs that target BDNF and IGF1 in skeletal muscle tissue with T2DM physiological conditions. Bioinformatics tools and databases were used to miRNA prediction, molecular homology, experimental validation of interactions, expression in the studied physiological condition, and network interaction. The findings showed three miRNAs candidates for IGF1(miR-29a, miR-29b, and miR-29c) and one for BDNF (miR-206). The experimental evaluations and the search for the expression in skeletal muscle from T2DM subjects confirmed the predicted interaction between miRNA-mRNA for miR-29b and miR-206 through human, rat, and mouse models. This interaction was reaffirmed in multiple network analyses. In conclusion, our results show the regulation relationship between miR-29b and miR-206 with the investigated genes, in several tissues, suggesting an inhibition pattern. Nevertheless, these data show a large number of possible interaction physiological processes, for future biotechnological prospects.

Resumo Extratos de espécies do gênero Cordia têm sido relatados com potencial atividade biológica, como antioxidante, antimicrobiana, antiviral e antiparasitária. O objetivo deste estudo foi desenvolver microemulsões contendo extratos etanólicos de folhas de C. glabrata e avaliar sua estabilidade e atividades biológicas. As cinco microemulsões desenvolvidas apresentaram estabilidade físico-química e apresentaram comportamento newtoniano quando submetidas à análise reológica. Os valores de diâmetro dos glóbulos variaram entre 225,74 e 273,33 nm e o potencial zeta das formulações permaneceu entre -22,40 e -25,08. Todos os ácidos fenólicos do extrato, quantificados por CLAE, apresentaram consistência após serem microemulsionados. Os valores de EC50 para a atividade antioxidante pelo método de eliminação DPPH variaram entre 38,13 e 45,54 µgmL-1, e entre 34,46 e 39,60 mM para o método de eliminação ABTS+. A atividade viricida apresentou CV50 <0,195 µgmL-1 e índice de seletividade maior que 20. Os resultados da fotoproteção variaram entre 2,14 a 2,84. O estudo revelou microemulsões estáveis e potencialização do efeito antioxidante nos extratos microemulsionados.

Abstract Extracts of species from the Cordia genus have been reported with potential biological activities, such as antioxidant, antimicrobial, antiviral, and antiparasitic. The aim of this study was to develop microemulsions containing ethanolic extracts of C. glabrata leaves and to evaluate their stability and biological activities. The five developed microemulsions presented physicochemical stability and presented Newtonian behavior when submitted to rheological analysis. The diameter values of the globules ranged between 225.74 and 273.33 nm and the zeta potential of the formulations remained between -22.40 and -25.08. All phenolic acids of the extract, quantified by HPLC, showed consistency after being microemulsified. The EC50 values for the antioxidant activity by the DPPH scavenging method ranged between 38.13 and 45.54 µgmL-1, and between 34.46 and 39.60 mM for the ABTS+ scavenging method. The virucidal activity presented a CV50 <0.195 µgmL-1 and a selectivity index greater than 20. The photoprotection results ranged between 2.14 to 2.84. The study revealed stable microemulsions and potentiation of the antioxidant effect in the microemulsified extracts.

ABSTRACT Sweet potato is among the most important food crops in the world. Due to its high genetic diversity, this species has cultivars that can respond in different ways depending on mineral fertilization. Among the nutrients, phosphorus is related to physiological and biochemical processes that can influence the final quality of the product. This study aimed to assess the influence of P doses on the quality and bioactive compounds of sweet potato cultivars - Paraná, BRS Amélia, and BRS Cuia. The pH, soluble solids (SS), titratable acidity (TA), SS/TA ratio, soluble sugars, and the bioactive compounds - flavonoids, anthocyanins, and carotenoids - were evaluated. The P doses influenced sweet potato cultivars. The dose of 120 kg ha-1 promoted increments in the parameters pH, SS, TA, and SS/TA ratio for the cultivar BRS Cuia. The same dose promoted an increase in total soluble sugars for the cultivar BRS Amélia. P dose of 240 kg ha-1 promoted higher levels of flavonoids and anthocyanins in the cultivar Paraná, and there was an increase in carotenoid content at the dose of 60 kg ha-1.

RESUMO A batata-doce está entre as culturas alimentícias mais importantes do mundo. Por apresentar alta diversidade genética, essa espécie possui cultivares que podem responder de maneiras diferentes em função da adubação mineral. Dentre os nutrientes, o fósforo está relacionado com processos fisiológicos e bioquímicos que podem influenciar na qualidade final do produto. Este estudo teve como objetivo avaliar a influência de doses de P na qualidade e compostos bioativos de cultivares de batata-doce - Paraná, BRS Amélia e BRS Cuia. Foram avaliados o pH, sólidos solúveis (SS), acidez titulável (AT), relação SS/AT, açúcares solúveis e os compostos bioativos - flavonoides, antocianinas e carotenoides. As doses de P influenciaram as cultivares de batata-doce. A dose de 120 kg ha-1 promoveu incrementos nos parâmetros pH, SS, AT e relação SS/AT para a cultivar BRS Cuia. A mesma dose promoveu incrementos nos açúcares solúveis totais para a cultivar BRS Amélia. A dose de 240 kg ha-1 promoveu maiores teores de flavonoides e antocianinas na cultivar Paraná, e houve aumento no teor de carotenoides na dose de 60 kg ha-1.

ABSTRACT The objective of this work was to evaluate the agro-bioeconomic feasibility of immature cowpea and beet cultivar combinations in intercropping systems in a semi-arid environment. Experiments were carried out in two cropping years using a randomized block design with four replicates. The treatments consisted of four cowpea cultivars (‘BRS Tumucumaque,’ ‘BRS Cauamé,’ ‘BRS Guariba,’ and ‘BRS Itaim’) and two beet cultivars (‘Early Wonder’ and ‘Fortuna’). In each block of the experiments, plots in monocropping of the cultivars were sown as additional treatments to obtain the following agronomic and competition indexes and economic indicators: land equivalent ratio (LER), area time equivalent ratio (ATER), land use efficiency (LUE), beet equivalent production (BEP), competitive ratio (CR), crop aggressivity (A), gross income (GI), net income (NI), rate of return (RR), and corrected monetary advantage (CMA). The highest agro-bioeconomic efficiency of the tested intercropping systems was achieved in the combination of cowpea cultivar BRS Guariba and beet cultivar Fortuna. Beet behaved as the dominant crop, and cowpea as the dominated crop in all evaluated intercropping systems. The indexes and indicators demonstrated the complementarity and sustainability of the combination of cowpea cultivar BRS Guariba with beet cultivar Fortuna.

RESUMO O objetivo deste trabalho foi avaliar a viabilidade agrobioeconômica de combinações de cultivares de feijão-caupi imaturo e de beterraba em sistemas consorciados em ambiente semiárido. Os experimentos foram conduzidos em dois anos agrícolas, em delineamento de blocos casualizados com quatro repetições. Os tratamentos consistiram da combinação de quatro cultivares de feijão-caupi (‘BRS Tumucumaque,’ ‘BRS Cauamé,’ ‘BRS Guariba’ e ‘BRS Itaim’) e duas cultivares de beterraba (‘Early Wonder’ e ‘Fortuna’). Em cada bloco dos experimentos, parcelas em monocultivo das cultivares foram semeadas como tratamentos adicionais para obtenção dos seguintes índices agronômicos e de competição e indicadores econômicos: razão equivalente de terra (RET), razão equivalente de área no tempo (REAT), eficiência de uso da terra (EUT), produção equivalente de beterraba (PEB), razão competitiva (RC), indice de superação das culturas (ISC), renda bruta (RB), renda líquida (RL), taxa de retorno (TR) e vantagem monetária corrigida (VMC). A mais alta eficiência agrobioeconômica dos sistemas consorciados testados foi alcançada na combinação da cultivar de feijão-caupi BRS Guariba com a cultivar de beterraba Fortuna. A beterraba se comportou como a cultura dominante e o feijão-caupi como a cultura dominada em todos os consórcios avaliados. Os índices e indicadores determinados demonstraram a complementaridade e sustentabilidade da combinação da cultivar de feijão-caupi BRS Guariba com a cultivar de beterraba Fortuna.

Resumo Espécies do gênero Cordia apresentam atividades biológicas, como anti-inflamatória, analgésica, antioxidante, antiviral e antifúngica. Para a espécie Cordia glabrata (MART) A.DC., ainda não existem informações sobre seu perfil fitoquímico e possíveis atividades biológicas, deste modo, o presente estudo teve como objetivo avaliar este perfil em extratos etanólicos de folhas jovens, adultas e senescentes, bem como o potencial antioxidante, fotoprotetor, antimicrobiano e virucida. A análise fitoquímica foi realizada por CCD (Cromatografia em Camada Delgada), mostrando a presença de flavonóides, taninos e terpenos. Na avaliação por CLAE EM/EM (Cromatografia Líquida de Ultra Eficiência acoplada a Espectrometria de Massas) foi evidenciado a presença dos ácidos caféico (3,89 mgL-1), p-cumárico (6,13 mgL-1) e ferúlico (0,58 mgL-1), paralelamente, na CG/EM (Cromatografia Gasosa acoplada a Espectrometria de Massas) verificou-se maior quantidade dos ácidos graxos palmítico (51,17%), esteárico (20,34%), linoléico (9,62%) e mirístico (8,16%). Os radicais DPPH (2,2-Difenil-1-picrilhidrazil) e ABTS+ (2′-Azino-bis (ácido 3-etilbenzotiazolina-6-sulfônico)) foram utilizados para verificar o potencial antioxidante, observando-se uma atividade superior para o extrato da folha em sua fase fenológica adulta: 54,63 ± 1,06 µgmL-1 (DPPH) e 44,21 ± 1,69 mM (ABTS+). A potencial atividade fotoprotetora dos extratos foi determinada espectrofotometricamente e os valores in vitro de FPS (Fator de Proteção Solar) em folhas jovens e adultas (5,47 e 5,41 respectivamente) apresentaram valores próximos ao FPS mínimo de 6,0 exigido pela ANVISA (Agência Nacional de Vigilância Sanitária). Não foi observada atividade antimicrobiana para Staphylococcus aureus sendo a concentração inibitória mínima de 2000 μgmL-1, no entanto o ensaio anti-herpético contra o vírus Herpes simplex tipo 2 (HSV-2) mostrou uma potente atividade virucida nas concentrações testadas com um valor de CV50 <0,195 μgmL-1 e um Índice de Seletividade (IS = CC50 / CV50) maior que 448. Os resultados obtidos neste estudo sugerem que extratos de folhas de C. glabrata em seu estágio fenológico adulto apresentam potencial antioxidante, fotoprotetora e virucida, considerando os resultados de testes in vitro.

Abstract Species of the genus Cordia have shown biological activities, such as anti-inflammatory, analgesic, antioxidant, antiviral, and antifungal activities. The species Cordia glabrata (MART) A.DC. Has no information concerning its phytochemical profile and possible biological activities. Thus, this study aimed to evaluate this profile in ethanolic extracts of young, adult and senescent leaves, as well as their antioxidant, photoprotective, antimicrobial, and virucidal potentials. Phytochemical analysis was performed by TLC (thin-layer chromatography) and showed the presence of flavonoids, tannins, and terpenes. The evaluation by UPLC-MS/MS (Ultra performance liquid chromatography - tandem mass spectrometer) evidenced the presence of caffeic (3.89 mgL-1), p-cumaric (6.13 mgL-1), and ferulic (0.58 mgL-1) acids, whilst, in GC/MS (Gas chromatography–mass spectrometry) analysis there was a greater amount of palmitic (51.17%), stearic (20.34%), linoleic (9.62%), and miristic (8.16%) fatty acids. The DPPH (2,2-Diphenyl-1-picrylhydrazyl) and ABTS+ (2′-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)) radicals were used to verify the potential antioxidant activity, observing a better activity for the leaf extract in the adult phenological stage: 54.63 ± 1.06 µgmL-1 (DPPH) and 44.21 ± 1.69 mM (ABTS). The potential photoprotective activity of the extracts was determined by spectrophotometry and the in vitro values of SPF (Sun Protection Factor) in young and adult leaves (5.47 and 5.41, respectively) showed values close to the minimum SPF of 6.0 required by ANVISA (Brazilian Health Regulatory Agency). It was not observed an antimicrobial activity for Staphylococcus aureus with a minimum inhibitory concentration of 2000 μgmL-1, however the anti-herpetic assay against the Herpes simplex virus type 2 (HSV-2) showed a potent virucidal activity at the tested concentrations with CV50 value <0.195 μgmL-1 and a Selectivity Index (SI = CC50 / CV50) greater than 448. The results obtained in this study suggest that extracts of leaves of C. glabrata in their adult phenological stage have potential antioxidant, photoprotective and virucidal activity, considering in vitro test results.

Abstract In this work, a thermal plasma-based ablation test system was used to evaluate the ablative performance of the EPDM composite. The system produces a high enthalpy plasma jet generated by a plasma (DC) torch, operating at atmospheric pressure using compressed air as working gas, enabling the variation of the thermal flux concerned with the studied EPDM composites. The samples were characterized using Scanning Electron Microscopy (SEM), Energy Dispersive Spectroscopy (EDS), Fourier-Transform Infrared spectroscopy (FTIR), and Thermogravimetric Analysis (TGA) to investigate the morphology, mass-loss rate, the reaction layer (char formation), and chemical changes of the samples for each thermal flux. For a complete evaluation, the thermal fluxes were varied in 0.30, 0.45, 0.60, 0.75, and 0.90 MW/m2 and for each thermal flux, disk-shape samples remained exposed to the plasma jet for 10s. During the plasma jet exposure time, the temperatures of the surface and the back of the samples were collected to verify the formed char layer's insulator capacity and the samples' thermal diffusivity for each experimental condition. The mass loss is continuous under the thermal fluxes of 0.30 and 0.45 MW/m2, stabilizing at 60% until 0.75 MW/m2. The formed char layer begins to lose its protective capacity, evidenced by the size decrease (from 800 µm to 700 µm), due to the ablation process of the reaction layer from the thermal flux of 0.90 MW/m2.

Abstract Bacteria responsible for causing infections are common in hospital environments, water, soil, and food products. The infection risk is intensified by the absence of public sanitation, poor quality of life, and food scarcity. These external factors promote the dissemination of pathogens by direct contamination or biofilm formation. In this work, we identified bacterial isolates obtained from intensive care units in the southern region of Tocantins, Brazil. We compared matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) techniques and 16S ribosomal ribonucleic acid (rRNA) molecular analysis; we also performed phenotypic characterization. Fifty-six isolates characterized using morphotinctorial tests were classified as gram-positive (80.4%; n = 45) and gram-negative (19.6%; n = 11) and were resistant to several antibiotic classes; notably, we identified the blaOXA-23 resistance gene in the ILH10 isolate. Microbial identification using MALDI-TOF MS resulted in the identification of Sphingomonas paucimobilis and Bacillus circulans. 16S rRNA sequencing revealed four isolates belonging to the genera Bacillus and Acinetobacter. The similarity was superior to 99% for Acinetobacter schindleri in the Basic Local Alignment Search Tool (BLAST), grouped in the clade superior to 90%. Several strains isolated from intensive care units (ICU) were resistant to various antibiotic classes. These techniques allowed for the identification of several microorganisms of importance in public health, enabling improvements in human infection control and proving the quality of inputs, food, and water.

Resumo As bactérias responsáveis por causar infecções são comuns em ambientes hospitalares, água, solo e produtos alimentícios. O risco de infecção é intensificado pela ausência de saneamento público, má qualidade de vida e escassez de alimentos. Esses fatores externos promovem a disseminação de patógenos por contaminação direta ou formação de biofilme. Neste trabalho, identificamos isolados bacterianos obtidos de unidades de terapia intensiva na região sul do Tocantins, Brasil. Comparamos técnicas de espectrometria de massa de tempo de voo com ionização por dessorção a laser assistida por matriz (MALDI-TOF MS) e análise molecular de ácido ribonucleico ribossômico 16S (rRNA); também realizamos caracterização fenotípica. Cinquenta e seis isolados caracterizados por testes morfotintoriais foram classificados como gram-positivos (80,3%; n = 45) e gram-negativos (19,6%; n = 11) e foram resistentes a várias classes de antibióticos; notavelmente, identificamos o gene de resistência blaOXA-23 no isolado de ILH10. A identificação microbiana usando MALDI-TOF MS resultou na identificação de Sphingomonas paucimobilis e Bacillus circulans. O sequenciamento do 16S rRNA revelou quatro isolados pertencentes aos gêneros Bacillus e Acinetobacter. A similaridade foi superior a 99% para Acinetobacter schindleri no BLAST, agrupado no clado superior a 90%. Várias cepas isoladas de ICU foram resistentes a várias classes de antibióticos. Essas técnicas permitiram a identificação de diversos microrganismos de importância em saúde pública, possibilitando melhorias no controle de infecções humanas e comprovando a qualidade dos insumos, alimentos e água.

Abstract The objective of this study was to evaluate, through changes in the gills and livers of Sciades herzbergii, the environmental contamination to which estuarine organisms are exposed in two areas in São Marcos Bay - MA. Two collection areas located in São Marcos Bay were selected for this study: A1, an area close to the Port Complex, and A2, an area on Caranguejos Island (included in the Environmental Protection Area of Baixada Maranhense). Collections were carried out during rainy and dry periods. Sediments (for trace element analyses), surface water (for physico-chemical analyses), and specimens of S. herzbergii (for biometric measurements and identification of branchial and hepatic histopathology) were collected. Physico-chemical parameters (pH, dissolved oxygen, temperature and salinity) were within limits established by Brazilian legislation. Arsenic (in A1) and nickel (in A1 and A2) were above the legal standards in both periods. The highest percentage of histological alterations in the gills (aneurysms, lamellar fusion and detachment of the epithelium) occurred in the port area, in the rainy (93%) and dry (74%) periods. Liver alterations (melanomacrophage centers and necrosis) occurred only in specimens from the same area, in the rainy (41%) and dry (36%) periods. The highest histological indices of gill and liver changes were recorded in A1. This result was further supported by the total HI value of the lesions, which was higher in the port area compared to A2 (less impacted area), suggesting that the environmental conditions in that location are less favorable for the well-being of these organisms. Permanent environmental monitoring of the area is necessary to control environmental impacts efficiently.

Resumo O objetivo deste estudo foi avaliar, por meio de alterações nas brânquias e fígados de Sciades herzbergii, a contaminação ambiental a que os organismos estuarinos estão expostos em duas áreas da Baía de São Marcos - MA. Para este estudo foram selecionadas duas áreas de coleta localizadas na Baía de São Marcos: A1, área próxima ao Complexo Portuário, e A2, área na Ilha dos Caranguejos (incluída na Área de Proteção Ambiental da Baixada Maranhense). As coletas foram realizadas nos períodos chuvoso e seco. Foram coletados sedimentos (para análises de elementos traço), águas superficiais (para análises físico-químicas) e espécimes de S. herzbergii (para medidas biométricas e identificação de histopatologia branquial e hepática). Os parâmetros físico-químicos (pH, oxigênio dissolvido, temperatura e salinidade) estiveram dentro dos limites estabelecidos pela legislação brasileira. Arsênio (em A1) e níquel (em A1 e A2) estiveram acima dos padrões legais em ambos os períodos. A maior porcentagem de alterações histológicas nas brânquias (aneurismas, fusão lamelar e descolamento do epítélio) ocorreu na área portuária, nos períodos chuvoso (93%) e seco (74%). As alterações hepáticas (centros de melanomacrófagos e necrose) ocorreram apenas nos espécimes dessa mesma área, nos períodos chuvoso (41%) e seco (36%). Os maiores índices histológicos de alterações branquiais e hepáticas foram registrados em A1. Esse resultado foi ainda corroborado pelo valor de IH total das lesões, que foi maior na área portuária em comparação com A2 (área menos impactada), sugerindo que as condições ambientais naquele local são menos favoráveis ao bem-estar desses organismos. O monitoramento ambiental permanente da área é necessário para controlar os impactos ambientais de forma eficiente.

Abstract Ortho-eugenol is a synthetic derivative from eugenol, the major compound of clove essential oil, which has demonstrated antidepressant and antinociceptive effects in pioneering studies. Additionally, its effects appear to be dependent on the noradrenergic and dopaminergic systems. Depression and anxiety disorders are known to share a great overlap in their pathophysiology, and many drugs are effective in the treatment of both diseases. Furthermore, high levels of anxiety are related to working memory deficits and increased oxidative stress. Thus, in this study we investigated the effects of acute treatment of ortho-eugenol, at 50, 75 and 100 mg/kg, on anxiety, working memory and oxidative stress in male Swiss mice. Our results show that the 100 mg/kg dose increased the number of head-dips and reduced the latency in the hole-board test. The 50 mg/kg dose reduced malondialdehyde levels in the prefrontal cortex and the number of Y-maze entries compared to the MK-801-induced hyperlocomotion group. All doses reduced nitrite levels in the hippocampus. It was also possible to assess a statistical correlation between the reduction of oxidative stress and hyperlocomotion after the administration of ortho-eugenol. However, acute treatment was not able to prevent working memory deficits. Therefore, the present study shows that ortho-eugenol has an anxiolytic and antioxidant effect, and was able to prevent substance-induced hyperlocomotion. Our results contribute to the elucidation of the pharmacological profile of ortho-eugenol, as well as to direct further studies that seek to investigate its possible clinical applications.

Resumo Ortho-eugenol é um derivado sintético do eugenol, composto principal do óleo essencial de cravo, que demonstrou efeitos antidepressivos e antinociceptivos em estudos pioneiros. Além disso, seus efeitos parecem ser dependentes dos sistemas noradrenérgico e dopaminérgico. Sabe-se que os transtornos de depressão e ansiedade compartilham uma grande sobreposição em sua fisiopatologia, e muitas drogas são eficazes no tratamento de ambas as doenças. Além disso, altos níveis de ansiedade estão relacionados a déficits de memória de trabalho e aumento do estresse oxidativo. Assim, o presente estudo investigou os efeitos do tratamento agudo de orto-eugenol, nas doses de 50, 75 e 100 mg/kg, na ansiedade, memória de trabalho e estresse oxidativo em camundongos Swiss machos. Nossos resultados mostram que a dose de 100 mg/kg aumentou o número de mergulhos e reduziu a latência no teste da placa perfurada. A dose de 50 mg/kg reduziu os níveis de malondialdeído no córtex pré-frontal e o número de entradas no labirinto em Y em comparação com o grupo de hiperlocomoção induzida por MK-801. Todas as doses reduziram os níveis de nitrito no hipocampo. Também foi possível avaliar uma correlação estatística entre a redução do estresse oxidativo e a hiperlocomoção após a administração do orto-eugenol. No entanto, o tratamento agudo não foi capaz de prevenir os déficits de memória de trabalho. Portanto, o presente estudo mostra que o orto-eugenol tem efeito ansiolítico e antioxidante, sendo capaz de prevenir a hiperlocomoção induzida pela substância. Nossos resultados contribuem para a elucidação do perfil farmacológico do orto-eugenol, bem como para direcionar novos estudos que busquem investigar suas possíveis aplicações clínicas.

Abstract Fungi exhibit three adverse effects on human health: inflammatory, allergic and toxic effects, these implications affect mainly immunodepressed patients. The objective of this work was to analyze the fungal microbiota of the ambient air of an Intensive Care Unit. Three collections were carried out in an Intensive Care Unit in the city of Rio Branco, Acre, Western Amazon, Brazil from March to May 2017. 126 Petri dishes were exposed with the culture medium Agar Sabouraud with chloramphenicol and Agar Mycosel, considering the distribution of the 21 air conditioners, split residential model. The plates were incubated for seven days at room temperature and the growth of Colony Forming Units was observed. Colony counting and isolation for the morphological characterization of the granted fungi was performed. After quantification, the concentration of fungi per cubic meters of air (CFU.m-3) was settled. The third collection had a larger number of colony forming units with 48.6%. In the total of the analyzed samples, filamentous fungi (85.5%) and yeasts (14.5%) were isolated. Thirteen genera of fungi were identified, with the most frequent filaments being Cladosporium spp. 33.0%, Aspergillus spp. 30.4% and Penicillium spp. 19.6%, and yeasts Candida spp. 52.6%, Trichosporon spp. 36.9%. The colony-forming unit per cubic meter (CFU.m-3) did not shown any difference between the Cores in the same collection period, however in the 1st and 3rd collection, Core 1 had the highest average. The fungal microbiota of this Unit presented thirteen different genera potentially pathogenic, revealing the need for monitoring microorganisms and prevention actions.

Resumo Os fungos exibem três efeitos adversos para a saúde humana: inflamatórios, alérgicos e tóxicos, esses efeitos atingem principalmente pacientes imunodeprimidos. O objetivo do trabalho foi analisar a microbiota fúngica do ar ambiente em uma Unidade de Terapia Intensiva na cidade de Rio Branco, Acre, Amazônia Ocidental, Brasil. Foram realizadas 03 coletas em uma Unidade de Terapia Intensiva na cidade de Rio Branco, Acre, Amazônia Ocidental, Brasil no período de março a maio de 2017. Foram expostas 126 placas de Petri com os meios de cultura Ágar Sabouraud com cloranfenicol e Ágar Mycosel, considerando a distribuição dos 21 condicionadores de ar, modelo split residencial. As placas foram incubadas por sete dias em temperatura ambiente e observado o crescimento das Unidades Formadoras de Colônias. A contagem de colônias e o isolamento para a caracterização morfológica dos fungos isolados foi realizada. Após a quantificação, foi calculada a concentração de fungos por metros cúbicos de ar (UFC.m-3). A terceira coleta teve maior quantidade de unidades formadoras de colônias com 48,6%. No total das amostras analisadas, foram isolados fungos filamentosos (85,5%) e leveduriformes (14,5%). Foram identificados 13 gêneros de fungos, sendo os filamentosos mais frequentes Cladosporium spp. 33,0%, Aspergillus spp. 30,4% e Penicillium spp. 19,6% e os leveduriformes Candida spp. 52,6%, Trichosporon spp. 36,9%. A unidade formadora de colônia por metros cúbicos (UFC.m-3) não houve diferença entre os Núcleos no mesmo período de coleta, porém na 1ª e 3ª coleta, o Núcleo 1 apresentou maior média. A microbiota fúngica desta Unidade apresentou treze gêneros diferentes potencialmente patogênicos, mostra a necessidade de monitoramento dos microrganismos e ações de prevenção.

ABSTRACT: Several toxic diseases cause mortality in cattle in southern Rio Grande do Sul State, Brazil. The aim of this study was to carry out a comparative analysis to determine the evolution of the main intoxications that occurred in cattle in southern Rio Grande do Sul from 1979 to 1999 and from 2000 to 2020. The spatial distribution and trend of occurrence of these intoxications over the 42 years in southern Rio Grande do Sul were determined using data from the Regional Diagnostic Laboratory of the Faculty of Veterinary Medicine at the Federal University of Pelotas (LRD-UFPel), which will make it possible to predict their occurrence in the coming years. From January 1979 to December 1999, 3,753 bovine materials were received for diagnosis at LRD-UFPel, and 3,653 cases were received from 2000 to 2020. Of the total number of materials received, 394 were diagnosed as intoxications, with 140 cases from 1979 to 1999 and 252 cases from 2000 to 2020. Out of 140 cases diagnosed in the first 21 years of operation of the LRD-UFPel, 113 (80.7%) were poisoning by plants, 22 (15.7%) by fungi outbreaks, and five (3.57%) by chemical substances. From 2000 to 2020, of the 252 diagnosed outbreaks 224 (88.8%) were caused by toxic plants, 10 (3.96%) by fungi, 12 (4.76%) by chemical substances, and six (2.38%) by insects. In the temporal trend analysis, a significant linear trend was observed with values of p=0.03 and an annual percentage change (APC) of 2.5 for the increase in the number of diagnoses of poisoning in general over the 42 years of the study. When analyzing the temporal trend of poisoning by Senecio spp., there was an increase in the occurrence of the diagnosis, with different characteristics and inflection points over time, with an annual growth rate of 9% in diagnoses during the first 21 years. There was a decrease in the occurrence of poisoning by Solanum fastigiatum, Echium plantagineum, and Claviceps paspali. Furthermore, poisoning by Ramaria flavo-brunnescens and Baccharis coridifolia maintained a similar percentage throughout the study period. It is concluded that toxic diseases will likely remain important causes of cattle death in the region.

RESUMO: Na região sul do Rio Grande do Sul, dentre as principais enfermidades que causam mortalidade em bovinos estão diversas doenças tóxicas, destacando-se as intoxicações por plantas e micotoxicoses. O objetivo deste trabalho foi realizar um estudo comparativo para determinar a evolução das principais intoxicações que ocorreram em bovinos na região sul do Rio Grande do Sul de 1979 a 1999 e de 2000 a 2020, estabelecendo a distribuição espacial e a tendência de ocorrência dessas intoxicações nos 42 anos de atividades do Laboratório Regional de Diagnóstico da Faculdade de Veterinária da Universidade Federal de Pelotas (LRD-UFPel), possibilitando prever sua ocorrência nas próximas décadas. Foram recebidos no laboratório de janeiro de 1979 a dezembro de 1999 e de janeiro de 2000 a dezembro de 2020, 3.753 e 3.653 materiais de bovinos para diagnóstico, respectivamente. Do total de materiais recebidos, 392 foram diagnosticados como intoxicações, sendo 140 entre os anos de 1979 a 1999 e 252 casos de 2000 a 2020. Dos 140 casos diagnosticados nos primeiros 21 anos de funcionamento do LRD-UFPel, 113 (80,7%) foram intoxicações por plantas, 22 (15,7%) intoxicações por fungos e cinco (3.6%) intoxicações por substâncias químicas. Entre os anos de 2000 e 2020 dos 252 casos diagnosticados 224 (88,8%) foram por plantas tóxicas, 10 (3,9%) por fungos, 12 (4,8%) por substâncias químicas e seis (2,4%) foram por insetos. Na análise de tendência temporal foi observada uma tendência linear significativa com valores de p=0,003 e mudança percentual anual (APC) de 2,5 no crescimento do número de diagnósticos de intoxicações em geral ao longo de todo o período de funcionamento do LRD-UFPel. Quando analisada a tendência temporal da intoxicação por Senecio spp. houve aumento na ocorrência do diagnóstico, com características e pontos de inflexão diferentes ao longo do tempo, sendo observado crescimento anual de 9% nos diagnósticos durante os primeiros 21 anos. Conclui-se que houve queda na ocorrência das intoxicações por Solanum fastigiatum, Echium plantagineum e Claviceps paspali e que as intoxicações por Ramaria flavo-brunnescens e Baccharis coridifolia mantiveram um percentual semelhante em todo o período. Além disso, as doenças tóxicas provavelmente permanecerão como importantes causas de morte de bovinos na região apesar de bem conhecidas, devido em parte, às dificuldades em fazer o controle das mesmas.

Quinones and organochalcogens are classes of compounds with great biological applicability, such as antioxidant, anticancer, anti-Alzheimer, and antidepressant activities, among others. Thus, the combination of these two classes of compounds is important to obtain new hits with biological activities that are additive or synergistic. Several methodologies for the preparation of this class of hybrid compound have been widely described. Many of the prepared hybrid molecules have shown increased biological activities and, in some cases, to act as two distinct pharmacophores. In this review, methods for the preparation of selenium-quinones, tellurium-quinones and their biological applications are highlighted.

Abstract Previous studies have investigated the preparation of heterogeneous biocatalysts based on the immobilization of lipases on distinct types of supports. However, few works have investigated the influence of the textural properties of these supports on the immobilization parameters. Thus, the present article reports the preparation of copolymers based on divinylbenzene by aqueous suspension polymerizations, using different amounts of porogenic agents to prepare particles with distinct textural properties. The particles were used for immobilization of lipase from Candida antarctica fraction B and the performance of the biocatalysts was evaluated in hydrolysis reactions, using p-nitrophenyl laurate as substrate. The use of Sty/DVB particles resulted in higher immobilization yields (89.5% and 99.2%) and higher hydrolytic activities, when compared to the Sty/VBC/DVB particles. Particularly, the increase of the pore diameters of the particles resulted in higher immobilization yields. Also, the hydrolytic activities depended simultaneously on the average pore diameter, porosity, and specific area (the most influential variable) of the supports. Thus, it was observed that the distinct morphological features of the polymer support can exert significant and conflicting effects on the final biocatalyst performance, since the specific surface area is normally expected to decrease with the increase of the average pore size.

Abstract Objectives This analysis describes the protocol of a study with a case-cohort to design to prospectively evaluate the incidence of subclinical atherosclerosis and Cardiovascular Disease (CVD) in Chronic Inflammatory Disease (CID) participants compared to non-diseased ones. Methods A high-risk group for CID was defined based on data collected in all visits on self-reported medical diagnosis, use of medicines, and levels of high-sensitivity C-Reactive Protein >10 mg/L. The comparison group is the Aleatory Cohort Sample (ACS): a group with 10% of participants selected at baseline who represent the entire cohort. In both groups, specific biomarkers for DIC, markers of subclinical atherosclerosis, and CVD morbimortality will be tested using weighted Cox. Results The high-risk group (n = 2,949; aged 53.6 ± 9.2; 65.5% women) and the ACS (n=1543; 52.2±8.8; 54.1% women) were identified. Beyond being older and mostly women, participants in the high-risk group present low average income (29.1% vs. 24.8%, p < 0.0001), higher BMI (Kg/m2) (28.1 vs. 26.9, p < 0.0001), higher waist circumference (cm) (93.3 vs. 91, p < 0.0001), higher frequencies of hypertension (40.2% vs. 34.5%, p < 0.0001), diabetes (20.7% vs. 17%, p = 0.003) depression (5.8% vs. 3.9%, p = 0.007) and higher levels of GlycA a new inflammatory marker (p < 0.0001) compared to the ACS. Conclusions The high-risk group selected mostly women, older, lower-income/education, higher BMI, waist circumference, and of hypertension, diabetes, depression, and higher levels of GlycA when compared to the ACS. The strategy chosen to define the high-risk group seems adequate given that multiple sociodemographic and clinical characteristics are compatible with CID.
