Abstract The extraction of native orchids from natural habitats is relevant for the reduction of populations in the Cerrado biome, making it necessary to establish practices aiming their production both for reintroduction and commercialization. The objective here is to evaluate light sources and sealing systems on the in vitro and ex vitro growth of Schomburgkia crispa. Two flask sealing systems were tested: conventional (CSS) and with gas exchange (SSGE), and eight light sources: FL1-100% white LED, FL2-100% blue LED, FL3-100% red LED, FL4-50% white + 25% red + 25% blue LED, FL5-50% red + 50% blue LED, FL6-25% red + 75% blue LED, FL7-75% red + 25% blue LED, and FL8- with fluorescent lamp, with five replications in each treatment. A completely randomized design was adopted with a 2x8 factorial scheme (vial sealing system x light sources). After 120 days of cultivation in vitro and 180 days ex vitro, the plants were evaluated as for number of leaves, roots and shoots, plant height, pseudobulb diameter, length of the largest root, largest leaf, and fresh mass. For the in vitro growth, the use of SSGE together with the light sources blue and red favors the cultivation of S. crispa. For the ex vitro growth, the cultivation in vitro in SSGE together with FL4 affects the acclimatization of plants.

Resumo A extração de orquídeas nativas dos habitats naturais constitui um fator relevante para a diminuição das populações do bioma Cerrado, tornando-se necessário estabelecimento de práticas visando sua produção tanto para reintrodução quanto para comercialização. Objetivou-se avaliar as fontes de luz e o sistema de vedação no crescimento in vitro e ex vitro de Shomburgkia crispa. Foram testados dois sistemas de vedação dos frascos: convencional (CSS) ou com trocas gasosas (SSGE) e oito fontes de luz: FL1 - LED 100% branca; FL2- LED 100% azul; FL3- LED 100% vermelha; FL4- LED 50% branca + 25% vermelha + 25%azul; FL5- LED 50% vermelha + 50% azul; FL6- LED 25% vermelha + 75% azul; FL7- LED 75% vermelha + 25% azul e FL8- fluorescente branca, com cinco repetições, em cada tratamento. Foi adotado um delineamento inteiramente casualizado em esquema fatorial 2x8 (sistema de vedação dos frascos x fontes de luz). Após 120 dias de cultivo in vitro e 180 dias ex vitro, as plantas foram avaliadas quanto ao número de folhas, raízes e brotos, altura da planta, diâmetro do pseudobulbo, comprimento da maior raiz e da maior folha e massa fresca. Para o crescimento in vitro, a utilização do SSGE em conjunto com fontes de luz azul e vermelho, favoreceu o cultivo de S. crispa. Para o crescimento ex vitro, o cultivo in vitro em SSGE em conjunto com FL4, influenciou a aclimatização das plantas.

Abstract One of the obstacles for the production of Orchidaceae plants is the seed propagation. Thus, in in vitro cultivation, luminosity and the type of sealing of the bottles are factors that influence germination and initial development of orchids. Therefore, the objective was to evaluate the light condition, the type of sealing of the bottles and the evaluation time in germination and initial in vitro development of Dendrobium nobile Lindl. After sowing, cultures were subjected to two irradiance conditions according to the sealing of the flasks: 7.0 µmol m-2 s-1 (threaded cap) and 19.0 µmol m-2 s-1 (plastic film) and two light conditions: white fluorescent lamp and red fluorescent lamp. At 30, 60 and 90 days, the percentage of germination, survival, of protocorms in stage 1, stage 2, stage 3 and stage were evaluated 4. DIC was used, in a sub-divided plot scheme for 30 and 60 days after sowing and in a 2 x 2 factorial scheme at 90 days. The highest germination percentage was observed when using white fluorescent lamp with a plastic film cover (70.33%) and at 30 days (70.13%). The highest survival percentage (100%) was observed when using plastic film cover and white fluorescent lamp at 90 days. The greatest development of propagules, reaching stage 4 was verified in the red fluorescent lamp with threaded cap (9.55%).

Resumo Um dos obstáculos para produção de plantas de Orchidaceae é a propagação por sementes. Dessa forma, no cultivo in vitro a luminosidade e o tipo de vedação dos frascos são fatores que influenciam a germinação e o desenvolvimento inicial de orquídeas. Sendo assim, objetivou-se avaliar a condição de luz, o tipo de vedação dos frascos e o tempo de avaliação na germinação e desenvolvimento inicial in vitro de Dendrobium nobile Lindl. Após semeadura, as culturas foram submetidas à duas condições de irradiâncias conforme a vedação dos frascos: 7,0 µmol m-2 s-1 (tampa rosqueável) e 19,0 µmol m-2 s-1 (filme plástico) e duas condições de luz: lâmpada fluorescente branca e lâmpada fluorescente vermelha. Aos 30, 60 e 90 dias foi avaliado a porcentagem de germinação, de sobrevivência, de protocormos em estádio 1, estádio 2, estádio 3 e estádio 4. Foi utilizado DIC, em esquema de parcelas sub-subdivididas para 30 e 60 dias após semeadura e em esquema fatorial 2 x 2 aos 90 dias. A maior porcentagem de germinação foi observada quando se utilizou lâmpada fluorescente branca com tampa de filme plástico (70,33%) e aos 30 dias (70,13%). A maior porcentagem de sobrevivência (100%) foi observada quando se utilizou tampa de filme plástico e lâmpada fluorescente branca aos 90 dias. O maior desenvolvimento de propágulos, chegando ao estádio 4 foi verificado na lâmpada fluorescente vermelha com tampa rosqueável (9,55%).

Resumo As orquídeas são um dos grupos de plantas com maior potencial de comercialização, por apresentarem características florais muito valorizadas no setor das flores e plantas ornamentais. Entretanto, as espécies mais comercializadas são exóticas e as nativas ainda são pouco exploradas quanto ao seu potencial ornamental. Assim, objetivou-se com este trabalho descrever aspectos morfológicos vegetativos e florais de Schomburgkia crispa Lindl., visando avaliar seu potencial ornamental. Para a avaliação do potencial ornamental da espécie, foram escolhidos sete indivíduos, dos quais os caracteres quantitativos foram avaliados em todas as plantas, totalizando cinquenta pseudobulbos e oitenta e oito flores. Durante a floração foi escolhida uma planta mais representativa para a realização de avaliação visual dos aspectos morfológicos da planta por meio de fotografias. Para retratar os aspectos morfológicos das flores foi elaborada uma ficha de diagnose floral, a partir de fotos das flores frescas, mantendo o máximo de detalhes morfológicos. Ao final do período experimental foram calculados as médias e o desvio padrão para cada um dos caracteres avaliados. A espécie S. crispa apresentou, em média, plantas com 46,17 cm de comprimento, 31,50 cm de largura e 88 cm de diâmetro. Quanto aos caracteres de florescimento, a espécie, de modo geral, apresentou 16,50 flores por planta, sendo 14,67 por haste floral e 4,66 flores abertas por dia, com durabilidade média de 32 dias. Sendo assim foi possível concluir que a espécie S. crispa apresenta elevado potencial ornamental para utilização na horticultura ornamental.

Abstract Orchids are one of the groups of plants with the greatest commercialization potential. They have floral characteristics highly valued in the sector of flowers and ornamental plants. However, exotics species are the most commercialized. The native species are still little explored for their ornamental potential. Thus, this work describes the vegetative and floral morphological characteristics of Schomburgkia crispa Lindl. aiming at evaluating its ornamental potential. For such, seven individuals were chosen. The quantitative characters were evaluated in all plants, totaling fifty pseudobulbs and eighty-eight flowers. During flowering, the most representative plant was chosen for the visual evaluation of its morphological features through photographs. A floral diagnosis sheet based on photos of fresh flowers was prepared by keeping the maximum level of morphological details. At the end of the experimental period, averages and standard deviations were calculated for each character evaluated. The species S. crispa presented, on average, plants 46.17 cm long, 31.50 cm wide, and with 88 cm of diameter. For flowering characters, the species presented in general 16.50 flowers per plant, 14.67 per flower stem, and 4.66 flowers opened per day, with an average durability of 32 days. Thus, we conclude that the species S. crispa has a great ornamental potential for use in ornamental horticulture.

RESUMO: A luz é um dos fatores que influenciam a germinação e o estabelecimento inicial no cultivo in vitro de orquídeas. Assim, objetivou-se avaliar o efeito de diferentes fontes de luz na germinação e no estabelecimento inicial in vitro de Schomburgkia crispa Lindl. Após semeadura em ambiente asséptico, as culturas foram acondicionadas em viveiro telado (luz natural) ou em sala de crescimento nas seguintes fontes luminosas: LEDs amarelo 3.000 K, branco 6.500 K [1], branco 6.500 K [2] ou lâmpada fluorescente branca 6.500 K (controle). Aos 45 e 90 dias após a semeadura foi avaliada a porcentagem de germinação e o estabelecimento inicial dos propágulos. A luz não influenciou a germinação das sementes de S. crispa, mas a utilização do LED 3.000 K proporcionou estabelecimento inicial de S. crispa em menor período e com menor mortalidade das plântulas, quando comparado com as demais fontes de luz utilizadas. A fonte de luz LED 3.000 K pode ser indicada como um substituto potencial para as lâmpadas fluorescentes e LEDs 6.500 K, utilizadas em salas de crescimento em laboratórios de cultivo in vitro.

ABSTRACT: Light is one of the factors that influence the germination and initial establishment of orchids under in vitro cultivation. This study evaluated the effect of different light sources on these stages in in vitro cultivation of Schomburgkia crispa Lindl. After sowing in an aseptic environment, we stored the cultures in a screened greenhouse (natural light) or in a growth room with the following light sources: 3,000 K yellow LED; 6,500 K white LED [1]; 6,500 K white LED [2]; or 6,500 K white fluorescent lamp (control). We assessed germination percentage and initial seedling establishment at 45 and 90 days after sowing. Light did not influence the germination of S. crispa. However, the use of 3,000 K LED provided a faster initial establishment of S. crispa when compared to the other light sources, also presenting lower seedling mortality. Thus, the light source 3,000 K LED is a potential substitute for the 6,500 K fluorescent lamps and LEDs used in growth rooms in in vitro culture laboratories.

RESUMO Alguns fatores podem influenciar no cultivo in vitro de orquídeas nativas, destacando-se o meio de cultura, o sistema de vedação dos frascos e a luz. Objetivou-se avaliar a adição de polpa e casca de banana na formulação dos meios de cultura, sistema de vedação e irradiância, no crescimento in vitro de Cattleya nobilior Rchb. f. Foi utilizado delineamento inteiramente casualizado, em esquema fatorial com cinco meios de cultura [Murashige & Skoog suplementado ou não com polpa ou casca de banana (100 ou 200g L-1)], duas condições de luz [lâmpadas LED 3.000 K (43 µmol m-2 s-1 ou 86 µmol m-2 s-1)] e dois sistemas de vedação (ventilação natural e convencional), com seis repetições de um frasco cada. Foram avaliados a altura da planta, diâmetro do maior pseudobulbo, comprimento da maior raiz e da maior folha, massa fresca e número de folhas, raízes e brotos. Tanto a utilização de polpa quanto de casca de banana como aditivos na formulação dos meios de cultura promoveram o crescimento de C. nobilior. O sistema de ventilação natural promoveu incremento na altura de planta, diâmetro de pseudobulbo e comprimento de folha, enquanto o sistema convencional promoveu perfilhamento.

ABSTRACT Some factors may influence the in vitro culture of native orchids, especially the culture medium, sealing system and light condition. This study aimed to evaluate the addition of banana pulp and peel in the formulation of culture media, sealing system and irradiance, in the in vitro growth of Cattleya nobilior Rchb. f. A completely randomized design was used in a factorial scheme with five culture media [Murashige & Skoog supplemented or not with banana pulp or peel (100 or 200g L-1)], two light conditions [3,000 K LED lamps (43 µmol m-2 s-1 or 86 µmol m-2 s-1)] and two sealing systems (natural and conventional ventilation), with six replicates of one vial each. The plant height, diameter of the largest pseudobulb, length of the largest root and of the largest leaf, fresh weight and number of leaves, roots and shoots were evaluated. The use of both the banana pulp and peel as additives in the culture medium formulation promoted the C. nobilior growth. The natural ventilation system increased the plant height, pseudobulb diameter and leaf length, while the conventional system promoted tillering.

RESUMO Um fator essencial para a formação de um banco de sementes de orquídeas nativas é a identificação da viabilidade das sementes armazenadas. Objetivou-se determinar uma metodologia para a otimização do teste de tetrazólio, na identificação da viabilidade de sementes armazenadas de duas espécies medicinais de Orchidaceae nativas do Cerrado. Sementes de Miltonia flavescens Lindl. e Schomburgkia crispa Lindl. foram submetidas a três pré-condicionamentos: sem embebição (controle), solução de sacarose ou água destilada. Em seguida foram submetidas a solução de tetrazólio, com três tipos de condicionamento: estufa, banho maria ou temperatura ambiente. As sementes de M. flavescens apresentaram germinação de 86,91 % e, quando submetidas ao pré-condicionamento com sacarose + condicionamento em estufa, mostraram maior porcentagem de sementes viáveis (62,32 %). Já para S. crispa, a germinação foi de 97,78 % e observou-se maior porcentagem de sementes viáveis no tratamento controle + temperatura ambiente (89,49 %). Esses resultados sugerem que, em Orchidaceae, protocolos específicos devem ser utilizados na condução do teste de tetrazólio e, ainda, a realização do teste de maneira amostral deve ser utilizada apenas para a indicação da viabilidade das sementes.

ABSTRACT An essential factor for the formation of a native orchid seed bank is the identification of the viability of stored seeds. This study aimed to determine a methodology for optimizing the tetrazolium test, in the identification of the viability of stored seeds of two Orchidaceae medicinal species native to the Brazilian Savanna. Seeds of Miltonia flavescens Lindl. and Schomburgkia crispa Lindl. were submitted to three pre-conditioning conditions: no soaking (control), sucrose solution or distilled water. The seeds were then submitted to a tetrazolium solution, with three types of conditioning: oven, water bath or ambient temperature. The M. flavescens seeds showed a germination of 86.91 %, with a higher percentage of viable seeds (62.32 %) when submitted to pre-conditioning with sucrose + oven conditioning. For S. crispa, the germination was 97.78 %, with a higher percentage of viable seeds for the control treatment + ambient temperature (89.49 %). These results suggest that specific protocols should be used to conduct the tetrazolium test in Orchidaceae. Moreover, when performed on a sample basis, the tetrazolium test should only be used to indicate the seed viability.

Resumo Devido ao extrativismo, Orchidaceae apresenta elevada vulnerabilidade, necessitando de medidas para a conservação de suas espécies. Assim, objetivou-se determinar um protocolo para germinação e estabelecimento inicial de Cattleya nobilior, Cattleya lundii e Brassavola tuberculata quanto à desinfestação de sementes e a utilização de meios de cultura. Sementes foram desinfestadas em solução de NaClO a 0,8%, por 5 min ou 15 min, recebendo ou não tríplice lavagem. Em seguida, foram semeadas nos meios MS, MS 1/2, K ou VW. Posteriormente, foram transferidas para sala de crescimento com temperatura e fotoperíodo controlados. A porcentagem de germinação e o estabelecimento inicial das espécies estudadas foram avaliados 45 dias após a semeadura. Para todas as espécies, as sementes desinfestadas por 5 min e submetidas à tríplice lavagem apresentaram maiores %G. Dessa forma, recomenda-se para a germinação in vitro e desenvolvimento inicial de protocormos das espécies C. nobilior e B. tuberculata, a utilização do tempo de desinfestação de 5 min, seguido da tríplice lavagem e semeadura em meio MS 1/2 (%G = 100 e 72,8%, propágulos em estágios 3 e 2, respectivamente). Para C. lundii, a semeadura deve ser realizada em meio KC, com a desinfestação por 5 min e tríplice lavagem das sementes (%G = 95,8% e propágulos em estágio 2).

Abstract Due to the extractivism Orchidaceae presents high vulnerability, needing measures for the conservation of its species. The aim of this study was to determine a protocol for germination and initial protocorm development of Cattleya nobilior, Cattleya lundii and Brassavola tuberculata for seed disinfestation and the use of culture media. Seeds were disinfested in 0.8% NaClO solution by 5 min or 15 min, receiving or not triple-wash. They were then sowing in MS, 1/2 MS, K or VW culture media. Subsequently, they were transferred to growth room with controlled temperature and photoperiod. The percentage of germination and initial development of the species studied were evaluated 45 days after sowing. For all species, the seeds disinfested by 5 min and submitted to the triple-wash showed higher germination percentage. Thus, is recommended the use of the 5 min disinfestation time, followed by the practice of triple-wash and sowing in 1/2 MS medium, for the germination and in vitro initial establishment of C. nobilior and B. tuberculata (%G = 100 and 72.8% and seedlings in stages 3 and 2, respectively). For C. lundii, sowing should be done in KC medium, with 5 min disinfestation and triple-wash (%G = 95.8% and seedlings in stage 2).

Abstract The situation of native orchids from natural environments becomes more vulnerable each day, especially those not protected by conservation units. Thus, studies on the propagation of species, especially on in vitro germination and initial establishment, are necessary for maintaining species in their habitat and reintroduction in restoration areas. This study verifies the influence of the nutritional composition of the culture media on in vitro germination and initial establishment of Miltonia flavescens. Seeds from natural pollination were sown in four different culture media and put in a growing room under controlled photoperiods and temperature. The germination was evaluated 30 days after sowing, and the initial growth of the seedlings at 120 and 180 days after sowing. The results indicated the Murashige and Skoog medium, complete and with half of the salt concentrations, promoted a higher germination percentage and were more effective in the initial development of M. flavescens seedlings.
