ABSTRACT Introduction. CIGB-300 is an antitumor peptide that inhibits the CK2-mediated phosphorylation by direct targeting of substrates. This paper aims to describe breakthroughs about the CIGB-300 antineoplastic mechanism related to the inhibition of CK2-mediated phosphorylation and the synergism with anticancer drugs. Methods. CK2 phosphorylation assays were performed with catalytic CK2α subunit or the CK2 holoenzyme in presence or not of CIGB-300. CIGB-300/CK2 interaction was verified by pull-down experiments, in situ colocalization and phosphoproteomic analysis of CIGB-300-treated lung cancer cells were also performed. Synergism of the peptide with anticancer drugs was evaluated in vitro and for Cisplatin; it was also tested in vivo. Besides, comparative proteomics of CIGB-300 combined with Cisplatin was conducted. Results. CIGB-300 targeted the CK2α subunit and inhibited the enzymatic activity of the holoenzyme in different experimental settings. Likewise, phosphoproteomic and Western Blot analysis allowed for knowing the early CK2 inhibition profile elicited by CIGB-300 at 10 and 30 minutes of treatment. Moreover, CIGB-300 did synergize with chemotherapeutics and EGFR inhibitors, and molecular events that reportedly support such synergistic interactions were also known. Conclusions. CIGB-300 inhibits the CK2-mediated phosphorylation by using an alternative mechanism of direct interaction with the enzyme itself. Besides, CIGB-300 synergizes with chemotherapeutics and EGFR inhibitors by modulating different proteins related to drug resistance.

RESUMEN Introducción. El CIGB-300 es un péptido antitumoral inhibidor de la fosforilación de los sustratos de la proteína quinasa CK2. El presente trabajo persigue como objetivo fundamental exponer nuevos elementos vinculados al mecanismo antineoplásico del CIGB-300 relacionados con el modo de inhibición de dicha fosforilación y la sinergia del péptido con drogas anticáncer. Métodos. Se realizaron ensayos de fosforilación mediada por la subunidad catalítica CK2( o la holoenzima en presencia o no del CIGB-300. La interacción CIGB-300/CK2 se verificó mediante ensayos de pull-down, colocalización in situ y además se realizó un análisis fosfoproteómico en células de cáncer de pulmón tratadas con el péptido. El sinergismo del CIGB-300 y las drogas anticáncer se evaluó in vitro y en el caso del cisplatino también in vivo. Además, se realizó un estudio de proteómica comparativa de la combinación del CIGB-300 con el cisplatino. Resultados. El CIGB-300 interaccionó directamente con la subunidad CK2( e inhibió la actividad de la holoenzima en diferentes condiciones experimentales. Asimismo, el análisis de fosfoproteómica y Western blot permitió conocer el perfil de inhibición temprana de CK2 por el CIGB-300 a los 10 min y 30 min Adicionalmente, el CIGB-300 sinergizó con quimioterapéuticos e inhibidores del receptor del factor de crecimiento epidérmico (EGFR) y se conocieron los eventos moleculares que pueden sustentar la sinergia del péptido con las drogas anticáncer estudiadas. Como conclusiones, el CIGB-300 inhibe la fosforilación mediada por CK2 a través de un mecanismo alternativo de interacción directa con la enzima. Además, sinergiza con quimioterapéuticos e inhibidores del EGFR, modulando diferentes proteínas relacionadas con la resistencia a dichas drogas.

Abstract Background: Acute otitis media (AOM) is one of the most prevalent acute conditions in the pediatric population worldwide. This work aimed to elaborate a Clinical Practice Guideline with clinical recommendations systematically developed to assist decision-making of specialists, patients, caregivers, and public policymakers involved in managing patients with AOM in children. Methods: This document was developed by the College of Pediatric Otorhinolaryngology and Head, and Neck Surgery of Mexico (COPEME) in compliance with international standards. The SIGN quality of evidence classification was used. On behalf of the COPEME, the Guideline Development Group (GDG) was integrated, including otolaryngologists, infectologists, pediatricians, general practitioners, and methodologists with experience in systematic literature reviews and the development of clinical practice guidelines. Results: A consensus was reached on 18 clinical questions, covering what was previously established by the GDG in the scope document of the guidelines. Scientific evidence answering each of these clinical questions was identified and critically evaluated. The GDG agreed on the final wording of the clinical recommendations using the modified Delphi panel technique. Specialists and patient representatives conducted an external validation. Conclusions: This Clinical Practice Guideline presents clinical recommendations for the prevention, diagnosis, and management of AOM to assist shared decision-making among physicians, patients, and caregivers and improve the quality of clinical care.

Resumen Introducción: La otitis media aguda (OMA) es uno de los padecimientos agudos más prevalentes en la población pediátrica a escala global. El objetivo de este trabajo fue elaborar una guía de práctica clínica con recomendaciones para asistir la toma de decisiones de médicos especialistas, pacientes, cuidadores de pacientes y elaboradores de políticas públicas involucrados en el manejo de la OMA en niños. Métodos: El documento ha sido desarrollado por parte del Colegio de Otorrinolaringología y Cirugía de Cabeza y Cuello Pediátricas de México (COPEME) en cumplimiento con los estándares internacionales. Se empleó la clasificación de calidad de la evidencia de SIGN. En representación del COPEME, se integró el Grupo de Desarrollo de la Guía (GDG), que incluyó otorrinolaringólogos, infectólogos, pediatras, médicos generales y metodólogos con experiencia en revisiones sistemáticas de la literatura y el desarrollo de guías de práctica clínica. Resultados: Se consensuaron 18 preguntas clínicas que abarcaron lo establecido previamente por el GDG en el documento de alcances de la Guía. Se identificó la evidencia científica que responde a cada una de estas preguntas clínicas y se evaluó críticamente. El GDG acordó la redacción final de las recomendaciones clínicas mediante la técnica Delphi de panel. Se llevó a cabo una validación externa por colegas especialistas y representantes de pacientes. Conclusiones: En esta Guía de Práctica Clínica se presentan recomendaciones clínicas para la prevención, el diagnóstico y el manejo de la OMA, con el fin de asistir la toma de decisiones compartidas entre médicos, pacientes y cuidadores con la intención de contribuir a mejorar la calidad de la atención clínica.

Abstract: Fabaceae are an option to increase the availability of protein in the prairies, but species of the Fabaceae family contain phenolic compounds, which depending on their number and type, benefit or harm livestock production. The aim of the study was to determine the concentration of phenolic regrowvth compounds of different ages of Arachis pintoi, Stylosanthes guianensis, Clitoria ternatea and Pueraria phaseoloides. The study was carried out in an experimental field, INIFAP-Tabasco, Mexico, from March to August 2013, covering the dry and rainy seasons, and in four ages of regrowth after uniformity cutting: 21, 42, 63 and 84 d. The variables evaluated were the concentrations of five types of phenolic compounds: total polyphenols (PFT), no tannins (FNT), condensed tannins (TC) hydrolysable (TH) and total tannins (TT). Sixteen treatments were evaluated in a design of completely randomly divided plots with three replications, the species as main plot, and regrowth age as a minor plot. Comparisons between means were performed using the Tukey test (p≤0.05). We observed the interaction species x age of regrowvth in the five types of compounds. The highest concentrations of phenolic compounds (CF) were obtained in all species at 42 d of regrowth. Clitoria ternatea presents the highest concentrations in the months without rain (155.4, 40.1, 16.9, 98.4 and 115.3 g kg-1 MS of PFT, FNT, TC and TH) and rainy (121.7, 39.6, 20.1, 61.9 and 82.0 g kg-1 MS of PFT, FNT, TC and TH). In contrast, S. guianensis presented lower concentrations of CF. The TC concentration varied between species, in the months without rain from 4.62 (Stylo) to 20.21 (Kudzú) g kg-1 MS and in rainy months from 3.66 (Stylo) to 20.10 (Clitoria) g kg-1 MS. No species reached the toxic level of 60 g kg-1 MS of TC. Results showed that the concentration of CT varied among species and age of regrowth. Among species, the highest concentrations were observed in C. ternatea and the lowest in S. guianensis. The interaction species-regrowvth age determined the concentration of phenolic compounds.

Resumen: Las fabáceas son una opción para incrementar la disponibilidad de proteína en las praderas pero especies de la familia Fabaceae contienen compuestos fenólicos que, según su cantidad y tipo, benefician o perjudican la producción pecuaria. El objetivo del estudio fue determinar la concentración de compuestos fenólicos del rebrote de diferentes edades de Arachis pintoi, Stylosanthes guianensis, Clitoria ternatea y Pueraria phaseoloides. El estudio se realizó en un campo experimental del INIFAP-Tabasco, México, de marzo a agosto del año 2013, abarcando las épocas seca y lluviosa, y en cuatro edades del rebrote después del corte de uniformidad: 21, 42, 63 y 84 d. Las variables evaluadas fueron las concentraciones de cinco tipos de compuestos fenólicos: polifenoles totales (PFT), no taninos (FNT), taninos condensados (TC) hidrolizables (TH) y totales (TT). Dieciséis tratamientos se evaluaron en un diseño de parcelas divididas completamente al azar con tres repeticiones, la especie como parcela mayor, y la edad del rebrote como parcela menor. Las comparaciones entre medias se realizaron con la prueba de Tukey (p≤0.05). La interacción especie x edad del rebrote se observó en los cinco tipos de compuestos. Las concentraciones mayores de compuestos fenólicos (CF) se obtuvieron en todas las especies a los 42 d del rebrote. Clitoria ternatea presentó las concentraciones mayores en los meses sin lluvias (155.4, 40.1, 16.9, 98.4 y 115.3 g kg-1 MS de PFT, FNT, TC y TH) y en los lluviosos (121.7, 39.6, 20.1, 61.9 y 82.0 g kg-1 MS de PFT, FNT, TC y TH). En contraste, S. guianensis presentó las concentraciones menores de CF. La concentración de TC varío entre especies, en los meses sin lluvias de 4.62 (Sytlo) a 20.21 (Kudzú) g kg-1 MS y en meses lluviosos de 3.66 (Stylo) a 20.10 (Clitoria) g kg-1 MS. Ninguna especie alcanzó el nivel tóxico de 60 g kg-1 MS de TC. Los resultados mostraron que la concentración de CT varió entre las especie y con la edad de rebrote. Entre especies, las concentraciones mayores se observaron en C. ternatea y las menores en S. guianensis. La interacción especie por edad del rebrote determina la concentración de compuestos fenólicos.

Objectives: To describe the characteristics of patients admitted in hospitals with soft tissue infections, and analyse the variables whose died, in order to define risk groups. Method: retrospective analysis of medical reports of all patient admitted during 2002 year for soft tissue infections in public malacitans hospitals. We excluded the patient with soft tissue infections associated with burns, surgery, pressure ulcers, and orbit cellulitis. We analysed clinical, biochemical variables and indications for yields and imaging tests, so the empiric antibiotic treatment established and its correlations with practice guidelines. Results: We analysed 391 admissions of 374 patients. Cellulitis was the most frequent diagnosis (69.3%).We did imaging tests in 51.6%. In 94.3% of cases were treated with empirics antibiotics. The most prescribed drug was amoxiciline plus clavulanate (39%). 27 patients died, 40.7% of them for septic cause. All deceased patients had chronic diseases. The only biochemical parameters associated with mortality were serum proteins and albumina (55 ± 9 g/L vs. 63 ± 8 g/L; p = 0.0231) and (22 ± 7 g/L vs. 29 ± 7 g/L; p = 0.0125) respectively. Conclusions: Cellullitis are the most frequent soft tissue infections that requires admissions in hospitals. We overuse imaging test and don't follow the practice guidelines recommendations in antibiotic therapy. Primary soft issue infection's mortality is low and it's restricted to people with chronic illness, deep infections and bad nutritional status.

Objetivos: Describir las características de los pacientes hospitalizados con infecciones de piel y partes blandas (IPPB) y analizar las variables de los que fallecen durante el ingreso para definir grupos de riesgo. Método: Análisis retrospectivo de las historias clínicas de todos los pacientes que fueron dados de alta con IPPB durante el año 2002, en los hospitales públicos de la provincia de Málaga. Se excluyen infecciones secundarias a pie diabético, cirugía, quemados, úlceras por decúbito y la celulitis orbitaria. Se analizaron variables clínicas, bioquímicas, indicación y validez diagnóstica de pruebas microbiológicas y de imagen, así como el tratamiento antibiótico empírico iniciado y su correlación con las recomendaciones terapéuticas establecidas. Resultados: Analizamos 391 ingresos de 374 pacientes. La celulitis fue el diagnóstico más frecuente (69,3%). Se realizaron pruebas de imagen en el 51,6% de los casos. El antibiótico más empleado fue la amoxicilina con ácido clavulánico (39%). Fallecieron 27 (6,7%), el 40,7% por el proceso séptico en sí. Todos los fallecidos, tenían algún tipo de enfermedad crónica asociada. Las únicas variables analíticas que se asociaron de forma significativa a mayor mortalidad fueron las proteínas totales (55 ± 9 g/L vs. 63 ± 8 g/L; p = 0,0231) y los concentraciones séricas de albúmina (22 ± 7 g/L vs. 29 ± 7 g/L; p = 0,0125). Conclusiones: Las celulitis es la IPPB que mayor número de ingresos genera en nuestro medio. Apreciamos una sobreutilización de pruebas de imagen y una desviación sobre las recomendaciones terapéuticas y el tratamiento aplicado en nuestros hospitales. La mortalidad global observada es baja y afecta de manera exclusiva a pacientes con mal estado nutricional al ingreso, enfermedades crónicas o infecciones profundas.

Aurelia aurita is a scyphozoan, abundant in the Mexican Caribbean during summer. Although usually innocuous, there is evidence of it causing harm to humans. This work investigates the biological activities of crude and fractionated extracts of A. aurita. Live specimens were collected between July and September 1999 from the Mexican Caribbean. The tentacular margin was dissected immediately and frozen at -50ºC. A nematocyst suspension was prepared, discharged, and the supernatants lyophilized. Hemolytic assay was performed with lyophilized crude extract on bovine, sheep, and human red blood cells. Erythrocyte sensitivity to the toxin was ranked in descending order: human, sheep, and bovine. Toxic activity on Artemia nauplii was evaluated using the same crude extract for different exposure periods (3, 5, and 10 hours); only 48 and 72 hour old Artemia nauplii showed 50% mortality. Partial toxin purification was completed by sequential liquid chromatography using three gels (Sephadex G-200, DEAE Sephadex A-50, and Sephadex G-100). Intramuscular neuroactivity was detected in the crab Ocypode quadrata for two partially purified fractions. These fractions were found to have molecular weight components of 66 and 45 kDa, respectively.
