Resumo O objetivo deste estudo foi verificar o desenvolvimento e reprodução do zoofitófago Podisus nigrispinus (Dallas) (Heteroptera: Pentatomidae) em couve, brócolis e repolho. Ninfas e adultos deste predador foram alimentados com lagartas de Plutella xylostella (L.) (Lepidoptera: Plutellidae) como presa e receberam folhas de couve, repolho ou brócolis. Durante o período ninfal, a duração do período e o consumo de presas foram semelhantes com as diferentes cultivares de brassicácea. Porém, a viabilidade ninfal foi maior para predadores com folhas de brócolis. O peso de ninfas de quinto instar e de fêmeas recém-emergidas e a razão sexual de P. nigrispinus foram semelhantes entre as cultivares de brassicáceas, enquanto que o peso de machos recém-emergidos foi maior com folhas de couve e brócolis. Folhas de brócolis proporcionaram maiores número de oviposições, ovos por postura e longevidade de machos e fêmeas de P. nigrispinus. Além disso, o consumo de lagartas de P. xylostella por adultos desse predador fora maiores com esta cultivar. A taxa líquida de reprodução (R0) e o tempo médio de geração (T) foram maiores para predadores com presa e folhas de brócolis. Podisus nigrispinus alimentados com lagartas de P. xylostella e folhas de brócolis apresentaram melhores parâmetros reprodutivos, podendo ser uma alternativa para a criação deste predador em laboratório.

Abstract The purpose of this study was to investigate the development and reproduction of the zoophytophagous predator Podisus nigrispinus (Dallas) (Heteroptera: Pentatomidae) fed kale, broccoli and cabbage affects its. Nymphs and adults of this predator were fed on larvae of Plutella xylostella (L.) (Lepidoptera: Plutellidae) as prey with kale, cabbage, or broccoli. In the nymph period, the duration and prey consumption were similar with all the Brassicacea cultivar. However, nymph viability was higher for predators with broccoli leaves. The mean weight of 5th-instar nymphs, newly emerged females and the sex ratio were similar among the Brassicacea cultivars, while newly emerged males were heavier with kale and broccoli leaves. The supply of broccoli leaves resulted in greater oviposition, higher number of eggs per egg mass and longer longevity of P. nigrispinus males and females. Furthermore, the consumption of P. xylostella larvae by adult predators was higher with these cultivars. The net reproductive rate (R0) and mean generation time (T) were highest for predators with prey and broccoli leaves. The reproductive parameters of P. nigrispinus were enhanced when fed on P. xylostella larvae with and broccoli leaves, which can be an alternative diet in laboratory rearing of this predator.

ABSTRACT Objectives To assess the evolution to permanent or transient conditions in children with positive neonatal TSH tests in Sergipe, Brazil, from 2004 to 2010. Subjects and methods Out of 193,794 screened newborns, 713 presented a neonatal TSH level higher than the local cutoff (5.2 µU/mL). From the confirmatory serum TSH values, the children were diagnosed with initial congenital hypothyroidism (CH) or suspect CH. From the evolution, they were classified as permanent CH, hyperthyrotropinemia, or transient TSH elevation. The mean incidence of each final condition was calculated for the total period of time. Results The initial diagnosis included 37 CH (18.1%) and 167 suspect CH (81.9%) cases. The final diagnosis included 46 cases of permanent CH (22.5%), 56 of hyperthyrotropinemia (27.5%), and 102 of transient TSH elevation (50.0%). Out of the 37 cases of initial CH, 23 (62.2%) had permanent CH, nine (24.3%) had hyperthyrotropinemia, and five (13.5%) had transient TSH elevation. Out of the 167 suspect CH cases, 23 (13.8%) had permanent CH, 47 (28.1%) had hyperthyrotropinemia and 97 (58.1%) had transient TSH elevation. The mean incidence after the follow up was 1:4,166 for permanent CH, 1:3,448 for hyperthyrotropinemia, and 1:1,887 for transient TSH elevation. Eighty-six percent of the children with an initial diagnosis of CH and 41.9% with suspect CH had a permanent condition (CH or hyperthyrotropinemia). Conclusions The follow-up of children with an initial diagnosis of CH or suspect CH is necessary to determine whether the disorder is permanent because predicting the evolution of the condition is difficult.

Ácidos galoilquínicos, substâncias que têm apresentado atividade leishmanicida, foram isolados do extrato de acetato de etila da planta Byrsonima coccolobifolia. Estes compostos fenólicos juntamente com o ácido gálico demonstraram ser uma nova classe de inibidores não-competitivos potentes em arginase (ARG) de Leishmania amazonensis (Ki variando de 0,10 a 0,68 µmol L-1). O ácido quínico não apresentou atividade inibitória significativa em ARG demonstrando que a unidade galoila tem características importantes que permitem a interação enzima-inibidor. A atividade inibitória significativa do ácido gálico frente à ARG pode ser uma indicação para o entendimento da resposta imune previamente observada em Leishmania donovani, uma vez que a atividade enzimática da arginase está associada à diminuição dos níveis de NO no processo de infecção por Leishmania.

Leishmanicidal galloylquinic acids were isolated from the ethyl acetate extract of Byrsonima coccolobifolia. These phenols and gallic acid showed to be a new class of potent noncompetitive inhibitors of arginase ARG (Ki ranging from 0.10 to 0.68 µmol L-1) from Leishmania amazonensis. Quinic acid did not exhibit significant inhibition of ARG, indicating that galloyl moiety has important features that allows the enzyme-inhibitor interactions. The significant inhibitory activity of gallic acid on ARG can be a clue to understand the immune response previously observed on L. donovani, since ARG activity is associated with the decrease of the levels of NO in Leishmania infection.

Kava é um fitoterápico ansiolítico usado no tratamento da insônia e da ansiedade. Alguns casos de hepatotoxicidade induzida pela kava foram relatados na literatura, levando à proibição do seu uso em muitos países. Clinicamente, o espectro dessas alterações variou de elevações transitórias das enzimas hepáticas, até à falência hepática fulminante e morte. Em alguns casos, realizou-se transplante hepático. Este artigo revisa a literatura atual sobre a hepatite tóxica provavelmente relacionada à kava, discute os possíveis mecanismos responsáveis pela hepatotoxicidade potencialmente grave e descreve alguns aspectos que devem ser considerados quando eventos adversos hepáticos pareçam ser relacionados à administração dessa substância. Conclui-se que a possível toxicidade hepática pela kava ainda deve ser investigada e que algumas medidas antes e durante o seu uso são importantes, dada a possibilidade de disfunção hepática grave.

Kava is an anxiolytic herbal medicine used in the treatment of sleep and anxiety disorders. Some cases of kava-induced hepatotoxicity have been reported in the literature leading to its banishment in most countries worldwide. Clinically, the spectrum ranged from transient elevations of liver enzyme levels to fulminant liver failure and death. Liver transplantation was performed in a few cases. This paper provides a review of the currently available literature on kava-related toxic hepatitis which may result from its use, discusses the possible mechanisms for the potentially severe hepatotoxicity and describes some features which must be considered when adverse liver effects seem to be associated to kava administration. In conclusion, the incidence of kava toxicity on the liver remains to be investigated; however, some concerns before or during kava use are important, due to the possibility of severe liver dysfunction.

A Serina Hidroximetiltransferase de Plasmodium falciparum nunca foi vista como alvo para a quimioterapia antimalarial, possivelmente devido a sua grande similaridade seqüencial com a enzima humana. Esta similaridade sugere que este parasita pode ser incapaz de mutar esta enzima para desenvolver resistência à quimioterapia. Neste trabalho, diferenças observadas no comportamento dinâmico dos sítios ativos da estrutura cristalográfica da enzima humana e um modelo, por homologia, da enzima do parasita, ambos complexados com glicina, como N-glicina-[3-hidróxi-2-metil-5-fosfonooximetil-piridin-4-il-metano], e ácido 5-formil-6-hidrofólico, foram utilizadas para o planejamento de inibidores seletivos desta enzima como novos potenciais antimalariais. Estes potenciais inibidores são derivados do ácido 5-formil-6-hidrofólico com diferentes cargas e comprimentos de cauda. Simulações por dinâmica molecular e estimativas das energias de interação dos compostos nos sítios ativos de ambas as enzimas mostraram que os compostos com carga líquida negativa e caudas curtas ou caudas longas e anfóteras, seriam potencialmente mais seletivos em relação à enzima do parasita.

Plasmodium falciparum Serine Hydroxymethyltransferase has never been used as target for antimalarial chemotherapy, possibly because its great sequence similarity with the human enzyme. This similarity suggests implies that P. falciparum may not be able to mutate this enzyme to develop resistance for chemotherapy. In this work, we have used differences on the dynamic behavior of the active sites of the crystallographic structure of the human enzyme and a homology model of parasite's enzyme, both in complex with glycine, as N-glycine-[3-hydroxy-2-methyl-5-phosphonooxymethyl-pyridin-4-yl-methane] and 5-formyl-6-hydrofolic acid, to design prototypes for selective inhibitors of this enzyme as new potential antimalarials. Those potential inhibitors are 5-formyl-6-hydrofolic acid derivatives with different charges and tail lengths. Molecular dynamics simulations and interaction energy estimates of the compounds within the active sites of both enzymes showed that compounds with a negative net charge and either shorter tails or longer amphoteric tails, would be more selective towards pfSHMT.
