In this study, nanostructured coatings of VN2/Fe3N/Fe4N are synthesized by Plasma Deposition (CCPD) using a vanadium cathodic cage and subject to cathodic and floating potential treatments. Microstructural and mechanical properties of coated SAE 1080 steel samples are investigated using X-ray Diffraction (XRD), Scanning Electron Microscopy (SEM), Vickers microhardness (HV), and surface roughness analysis. The coating produced under cathodic potential treatment exhibited a nitride-based microstructure (VN2, Fe3N, Fe4N), with surface morphology consisting of clusters of granular structures with varied particle sizes. The coating produced under floating potential predominantly exhibited the iron nitride phase (Fe3N) with a morphology composed of uniformly sized grains. Microhardness testing showed that both layers exhibited more excellent plastic deformation resistance than the substrate. Roughness measurements confirmed a more organized microstructure profile for the sample treated under floating potential. This study demonstrates that the CCPD-produced cathodic potential coating can be utilized in tribological applications. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. METHODS This is a descriptive and comparative study of raw data on deaths in the SIM, RC and 2010 Census databases, the latter held in Brazilian states and municipalities between August 2009 and July 2010. The percentage of municipalities was confirmed by the database showing the highest death count. The association between the source of the highest death count and socioeconomic indicators - the Índice de Privação Brasileiro (IBP – Brazilian Deprivation Index) and Índice de Desenvolvimento Humano Municipal (IHDM – Municipal Human Development Index) - was performed by bivariate choropleth and Moran Local Index of Spatial Association (LISA) cluster maps. RESULTS Confirmed that the SIM is the database with the highest number of deaths counted for all Brazilian macroregions, except the North, in which the highest coverage was from the 2010 Census. Based on the indicators proposed, in general, the Census showed a higher coverage of deaths than the SIM and the RC in the most deprived (highest IBP values) and less developed municipalities (lowest IDHM values) in the country. CONCLUSION The results highlight regional inequalities in how the databases chosen for this study cover death records, and the importance of maintaining the issue of mortality on the basic census questionnaire.

Growth in the international popularity of Brazil nuts oil (Bertholletia excelsa, Bonpl.) has demanded a closer look at quality aspects of the nuts. In this context, time-domain 1H nuclear magnetic resonance spectroscopy (TD-NMR) can be a useful tool for assessing the lipid portion of such matrices. In this work, a TD-NMR method was developed and validated to assess the oil content directly from Brazil nuts following the ISO 10565:1998 and DOQ-CGCRE-008. Full validation was addressed covering the working range from 0.67 to 25.00 g (coefficient of determination, R2= 0.9993). Limits of detection (LOD) and quantification (LOQ) were 0.20 and 0.67 g, respectively. The repeatability and intermediate precision assays met the guide requirements. Oil contents obtained in two laboratories (different equipment) were statistically similar (66.93 ± 4.69 and 70.13 ± 4.15%), highlighting the method’s robustness. The developed approach has the potential to be adopted as an industrial reference method for in situ monitoring of Brazil nuts oil contents.

Resumo Neste estudo, descrevemos um método para atingir uma população-alvo (dentistas atuando no Brasil) para se engajar em pesquisa do tipo enquete (survey) utilizando convites tradicionais por e-mail e campanhas de recrutamento criadas no Instagram. Este estudo aborda aspectos metodológicos e compara as pessoas entrevistadas alcançadas por diferentes métodos. Foi utilizado um questionário pré-testado e os(as) participantes foram recrutados(as) por 10 dias por meio de uma lista de endereços de e-mail e duas campanhas abertas orgânicas independentes no Instagram. Foram coletadas 3122 respostas: 509 participantes foram recrutados(as) por e-mail (taxa de resposta 2,1%) e 2613 pelas duas campanhas do Instagram (taxas de conversão 20,7% e 11,7%), respectivamente. As taxas de coleta de resposta/min nas primeiras 24h variaram entre 0,23 (e-mail) e 1,09 (primeira campanha). No total, 98,8% das respostas foram recebidas nas primeiras 48h para as diferentes estratégias de recrutamento. Houve diferenças significativas para todas as variáveis demográficas (p<0,001) entre recrutados(as) por e-mail e Instagram, com exceção de sexo (p=0,37). As pessoas recrutadas via Instagram eram um pouco mais velhas, tinham mais experiência profissional (anos na prática) e nível de pós-graduação superior às entrevistadas por e-mail. Além disso, a maioria dos entrevistados por e-mail e Instagram trabalhava no setor público e na prática privada, respectivamente. Embora ambas estratégias tenham sido capazes de coletar respostas de todas as regiões brasileiras, as respostas por e-mail foram ligeiramente melhor distribuídas nas cinco áreas territoriais em comparação ao Instagram. Este estudo fornece evidências de que o recrutamento de uma amostra de população diversificada e grande usando o Instagram é viável. No entanto, a combinação de e-mail e Instagram no recrutamento levou a uma população mais diversificada e melhores taxas de resposta.
Abstract In this study, we describe a method for reaching a target population (i.e., dentists practicing in Brazil) to engage in survey research using traditional e-mail invites and recruitment campaigns created on Instagram. This study addresses methodological aspects and compares respondents reached by different methods. A pre-tested questionnaire was used and participants were recruited for 10 days via a source list of email addresses and two discrete Instagram organic open campaigns. A total of 3,122 responses were collected: 509 participants were recruited by email (2.1% response rate) and 2,613 by the two Instagram campaigns (20.7% and 11.7% conversion rates), respectively. Response/min collection rates in the first 24 h ranged between 0.23 (email) and 1.09 (first campaign). In total, 98.8% of all responses were received in the first 48 h for the different recruitment strategies. There were significant differences for all demographic variables (p< 0.001) between email and Instagram respondents, except for sex (p=0.37). Instagram respondents were slightly older, had more professional experience (years in practice), and a higher graduate education level than email respondents. Moreover, most email and Instagram respondents worked in the public sector and private practice, respectively. Although both strategies could collect responses from all Brazilian regions, email responses were slightly better distributed across the five territorial areas compared to Instagram. This study provides evidence that survey recruitment of a diverse, large population sample using Instagram is feasible. However, combination of email and Instagram recruitment led to a more diverse population and improved response rates.

BACKGROUND Biomphalaria glabrata snails are widely distributed in schistosomiasis endemic areas like America and Caribe, displaying high susceptibility to infection by Schistosoma mansoni. After the availability of B. glabrata genome and transcriptome data, studies focusing on genetic markers and small non-coding RNAs have become more relevant. The small RNAs have been considered important through their ability to finely regulate the gene expression in several organisms, thus controlling the functions like cell growth, metabolism, and susceptibility/resistance to infection. OBJECTIVE The present study aims on identification and characterisation of the repertoire of small non-coding RNAs in B. glabrata (Bgl-small RNAs). METHODS By using small RNA sequencing, bioinformatics tools and quantitative reverse transcription polymerase chain reaction (RT-qPCR), we identified, characterised, and validated the presence of small RNAs in B. glabrata. FINDINGS 89 mature miRNAs were identified and five of them were classified as Mollusk-specific. When compared to model organisms, sequences of B. glabrata miRNAs showed a high degree of conservation. In addition, several target genes were predicted for all the mature miRNAs identified. Furthermore, piRNAs were identified in the genome of B. glabrata for the first time. The B. glabrata piRNAs showed strong conservation of uridine as first nucleotide at 5’ end, besides adenine at 10th position. Our results showed that B. glabrata has diverse repertoire of circulating ncRNAs, several which might be involved in mollusk susceptibility to infection, due to their potential roles in the regulation of S. mansoni development. MAIN CONCLUSIONS Further studies are necessary in order to confirm the role of the Bgl-small RNAs in the parasite/host relationship thus opening new perspectives on interference of small RNAs in the organism development and susceptibility to infection.

ABSTRACT: The objective of this study was to evaluate the yield and energy balance of four sorghum genotypes in irrigated and rainfed crops. The experiment was conducted in an irrigation and drainage unit at the Federal University of Grande Dourados (UFGD), in Dourados, Mato Grosso do Sul state (Brazil). The experimental design was a randomized complete block design with split plots (with and without irrigation), testing four genotypes (BRS 506, CV 007, CV 147, and EJ 7281) with four replicates (32 plots). Irrigation provided yield increase in all four genotypes. The respective yield increases were 85.89%, 71.82%, 64.28%, and 63.36% for genotypes EJ 7281, BRS 506, CV 147, and CV 007. The energy efficiency (produced/ used ratio) was on average 3.5 under irrigation, and 2.8 for rainfed crops. These results indicate the lack of competition between sorghum and sugarcane, being the first an alternative for off-season. Irrigation increased productivity, leading to an increase in yield and, consequently, in the amount of extracted energy. Yield gains in response to irrigation were more pronounced for genotype EJ 7281. There was a positive impact of irrigation on the energy balance of the four genotypes, increasing energy efficiency.

RESUMO Introdução: A hanseníase é uma doença infecciosa crônica causada pelo parasita intracelular Mycobacterium leprae. O diagnóstico é essencialmente clínico, com base em sintomas, exame da pele, nervos periféricos e história epidemiológica. Testes laboratoriais são realizados para complementar o resultado de diagnóstico clínico, ou mesmo servindo como método de confirmação. Objetivo: Investigar a positividade e a concordância da baciloscopia, do ensaio de imunoadsorvente ligado à enzima (ELISA) com o antígeno sintético ND-O-BSA, do ML Flow e da reação em cadeia da polimerase (PCR) para a detecção de Mycobacterium leprae em casos novos de hanseníase. Métodos: Foi realizada uma série de casos, avaliando uma amostra de conveniência de 39 novos casos de hanseníase e um grupo-controle de 18 contatos domiciliares em Belém (PA) e Igarapé-Açu (PA) a partir de março 2014 a setembro de 2015. Resultados: A concordância entre as combinações ELISA, ML Flow e PCR mostrou reprodutibilidade leve a ausente (Kappa ≤ 0,24). Os resultados mostraram maior sensibilidade no ensaio de PCR, com maior positividade em casos multibacilares. O teste ELISA mostrou baixa positividade, mesmo em casos multibacilares, resultando em nenhuma reação nos casos paucibacilares e contatos domiciliares. Conclusão: A alta sensibilidade da PCR diminui a concordância com outros testes.
ABSTRACT Introduction: Leprosy is a chronic infectious disease caused by the intracellular parasite Mycobacterium leprae. The diagnosis is essentially clinical, based on symptoms, skin exam, peripheral nerves and epidemiological history. Laboratory tests are carried out to complement the result of clinical diagnosis, or even serving as a confirmatory method. Objective: To investigate the positivity and agreement between skin smear, enzyme-linked immunosorbent assay (ELISA) with synthetic antigen ND-O-BSA, ML Flow test and polymerase chain reaction (PCR) for detection of Mycobacterium leprae in new cases of leprosy. Methods: We conducted a case series study assessing a convenience sample of 39 new cases of leprosy and a control group of 18 household contacts in Belém (PA) and in Igarapé-Açu (PA) from March 2014 to September 2015. Results: The agreement between ELISA, ML Flow and PCR tests combinations showed slight to absent reproducibility (Kappa ≤ 0.24). The results showed greater sensitivity in PCR assay, with higher positivity in multibacillary cases. The ELISA test showed low positivity, even in multibacillary cases, resulting in no reaction to paucibacillary cases and household contacts. Conclusion: The high sensitivity of PCR decreases the agreement with other tests.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Schistosoma mansoni in a low-prevalence area in Brazil: the importance of additional methods for the diagnosis of hard-to-detect individual carriers by low-cost immunological assays

Schistosomiasis diagnosis is based on the detection of eggs in the faeces, which is laborious and lacks sensitivity, especially for patients with a low parasite burden. Immunological assays for specific antibody detection are available, but they usually demonstrate low sensitivity and/or specificity. In this study, two simple immunological assays were evaluated for the detection of soluble Schistosoma mansoni adult worm preparation (SWAP) and egg-specific IgGs. These studies have not yet been evaluated for patients with low parasite burdens. Residents of an endemic area in Brazil donated sera and faecal samples for our study. The patients were initially diagnosed by a rigorous Kato-Katz analysis of 18 thick smears from four different stool samples. The ELISA-SWAP was successful for human diagnosis with 90% sensitivity and specificity, confirming the Kato-Katz diagnosis with nearly perfect agreement, as seen by the Kappa index (0.85). Although the ELISA-soluble S. mansoni egg antigen was 85% sensitive, it exhibited low specificity (80%; Kappa index: 0.75) and was more susceptible to cross-reactivity. We believe that immunological assays should be used in conjunction with Kato-Katz analysis as a supplementary tool for the diagnosis of schistosomiasis for patients with low infection burdens, which are usually hard to detect.

INTRODUÇÃO: A correlação entre o ensaio imunológico e o título de anticorpos serve como ferramenta para a determinação das diferentes fases da doença. MÉTODOS: Dois ensaios imunológicos simples para detecção de IgG específico para antígenos de vermes adultos e ovos do Schistosoma mansoni com amostras de soro murino foram padronizados e avaliados em nosso laboratório. Cinquenta camundongos negativos e positivos foram avaliados e os resultados obtidos por enzyme-linked immunosorbent assays (ELISA) foram comparados com o número de vermes adultos contados em tempos diferentes de infecção. RESULTADOS: Os dados mostraram que a ELISA com antígenos de vermes adultos (ELISA-SWAP) apresentou uma correlação satisfatória entre a absorbância obtida para os títulos de IgG e o número individual de vermes contados por perfusão do sistema porta hepático (R2=0,62). Adicionalmente, a ELISA-SWAP foi capaz de detectar diferencialmente amostras positivas com 30 e 60 dias de infecção (p=0,011 e 0,003, respectivamente), enquanto a ELISA com antígenos de ovos (ELISA-SEA) detectou amostras positivas com 140 dias de infecção (p=0,03). CONCLUSÕES: Estes dados mostram que o uso de antígenos diferentes em métodos imunológicos pode ser usado como ferramentas potenciais para a análise da evolução cronológica da infecção por S. mansoni na esquistossomose murina. Correlações com a esquistossomose humana devem ser discutidas.
INTRODUCTION: The correlation between the immunological assay and the antibody titer can offer a tool for the experimental analysis of different phases of the disease. METHODS: Two simple immunological assays for Schistosoma mansoni in mice sera samples based on specific IgG detection for worms soluble antigens and eggs soluble antigens were standardized and evaluated in our laboratory. Fifty mice were used in negative and positive groups and the results obtained by enzyme-linked immunosorbent assays (ELISA) assays were compared with the number of worms counted and the IgG titers at different times of infection. RESULTS: Data showed that ELISA using adult worm antigens (ELISA-SWAP) presented a satisfactory correlation between the absorbance value of IgG titers and the individual number of worms counted after perfusion technique (R²=0.62). In addition, ELISA-SWAP differentially detected positive samples with 30 and 60 days post infection (p=0.011 and 0.003, respectively), whereas ELISA using egg antigens (ELISA-SEA) detected samples after 140 days (p=0.03). CONCLUSIONS: These data show that the use of different antigens in immunological methods can be used as potential tools for the analysis of the chronological evolution of S. mansoni infection in murine schistosomiasis. Correlations with human schistosomiasis are discussed.

Do extrato metanólico das folhas de Cratylia mollis, foram isolados através de técnicas cromatográficas, (3S,5S,6S,9R)-3,6-di-hidróxi-5,6-diidro- β-ionol (1) e um novo nor-isoprenóide identificado como (4S*, 6S*)-4-but-1E-enil-4,6-diidróxi-3,5,5-trimetil-ciclo-hex-2-enona (2) além do 5a,8a-epidioxiergosta-6,22-dien-3-β-ol. As estruturas foram elucidadas por meio da análise dos dados de EM, IV, RMN ¹H e 13C.
From the methanolic extract of the leaves of Cratylia mollis were obtained by chromatographic techniques (3S,5S,6S,9R)-3,6-dihydroxy-5,6-dihydro- β-ionol (1), and a new bis-norisoprenoid named (4S*, 6S*)-4-but-1E-enyl-4,6-dihydroxy-3,5,5-trimethyl-cyclohex-2-enone (2) as well as 5a,8a-epidioxyergosta-6,22-dien-3-β-ol. The structures of the pure compounds were elucidated based on MS, ¹H and 13C NMR spectroscopic data analyses.

As formigas são componentes funcionais importantes em florestas tropicais devido aos papéis ecológicos que exercem, à grande biomassa e à riqueza de espécies. Embora a Mata Atlântica seja um dos ecossistemas mais bem estudados no Brasil, ainda faltam informações sobre a diversidade de formigas nos fragmentos florestais do Estado do Rio de Janeiro. A riqueza e composição da assembléia de formigas em floresta ombrófila de encosta na ilha da Marambaia (RJ) foi estudada através de um inventário estruturado em uma área de 0,6 ha. Armadilhas do tipo "pitfall" e coletas manuais foram empregadas na serapilheira e sobre a vegetação entre os meses de janeiro e julho de 2004. Um total de 29 gêneros e 82 espécies foi encontrado na amostragem. A abundância e a riqueza de espécies foram maiores nas amostras de março do que de julho. Já a eqüitatividade e diversidade de formigas nas amostras não foram influenciadas pela época da coleta. As amostras de formigas em galhos mortos adicionaram seis espécies à lista, acrescentando informações sobre a biologia das espécies. As amostras sobre plantas totalizaram 32 espécies de formigas, das quais 12 foram exclusivas, como as espécies de Pseudomyrmex e algumas de Crematogaster e Pachycondyla. Este estudo pretende contribuir para o desenvolvimento de prioridades conservacionistas em um dos ecossistemas mais ameaçados do mundo.
Ants are an important functional component in tropical forest due to their ecological roles, biomass and species diversity. Although the Atlantic Forest is one of the best studied ecosystems in Brazil, there is a lack of information about ant diversity in forest fragments of the state of Rio de Janeiro. The composition and richness of the ant fauna from atlantic slope rain-forest in Marambaia island-RJ were assessed by the structured inventory in an area of 0.6 ha. Pitfalls traps and hand collecting were used for sampling ants in the litter and on vegetation from January to July/2004. A total of 29 genera and 82 species were found in this survey. Ant abundance and richness were higher in March than in July samples. Eveness and diversity of ants in the samples were not affected by the time of the year. Collections of ants from dead twigs yielded an additional seven species and provided information about the ant biology. Sampling ants on plants resulted in 32 species but 12 species were not recorded in litter samples. Some species were recorded exclusively on vegetation, such as Pseudomyrmex, Crematogaster and Pachychondyla species. This study aims to contribute to the development of conservation priorities in one of the most threatened ecosystems in the world.
