Abstract Adaptation of photoperiod-sensitive japonica rice varieties from long-day (LD) to short-day (SD) conditions is impeded by their extremely early flowering in response to photoperiod change, but the genetic factor underlying this is still elusive. We detected mutations in Hd1 in Zhenjing2400 through gene mapping and sequencing analysis. Genome resequencing of the varieties Zhenjing2400 and Jiahe218 identified single nucleotide polymorphisms (SNPs) in the other flowering-related genes Ehd1, SDG725, OsCOL15, DTH2, and DTH7. We constructed recombinant inbred lines (RILs) derived from a cross between Zhenjing2400 and Jiahe218, and selected homozygous lines with these six genes. We established that photoperiod insensitivity is caused by a defective Hd1 gene. In addition, the effect of Hd1 and Ehd1 on heading date was stronger than that of the other four genes. Resumo A aterosclerose é a causa mais comum de doença cardiovascular em todo o mundo, ela está associada a uma alta incidência de eventos clínicos. O acúmulo de evidências elucidou que os RNAs longos não codificantes (LncRNAs) são uma nova classe de transcritos com papéis críticos nos processos fisiopatológicos da aterosclerose. Nesta revisão, resumimos o progresso recente dos LncRNAs no desenvolvimento da aterosclerose. Descrevemos principalmente os diversos mecanismos regulatórios dos LncRNAs nos níveis transcricionais e pós-transcricionais. Este estudo pode fornecer informações úteis sobre os LncRNAs como alvos terapêuticos ou biomarcadores para o tratamento da aterosclerose.

Abstract Atherosclerosis is the most common cause of cardiovascular disease globally, associated with a high incidence of clinical events. Accumulating evidence has elucidated that long non-coding RNAs (lncRNAs) as a novel class of transcripts with critical roles in the pathophysiological processes of atherosclerosis. In this review, we summarize the recent progress of lncRNAs in the development of atherosclerosis. We mainly describe the diverse regulatory mechanisms of lncRNAs at the transcriptional and post-transcriptional levels. This study may provide helpful insights about lncRNAs as therapeutic targets or biomarkers for atherosclerosis treatment.

Abstract Acanthopanax gracilistylus W.W.Smith (AGS) is an traditional Chinese medicine and its leaves that have rich biological activities has been long used as a fresh vegetable in Chinese folk. There are two lupane-type triterpenoids, 3α,11α-dihydroxy-23-oxo-lup-20(29)-en-28-oic acid (1) and 3α,11α,23-trihydroxy-lup-20(29)-en-28-oic acid (2), were isolated as the active constituents from leaves of AGS. However, the anti-inflammatory effects and potential mechanisms of 1 and 2 on LPS-induced RAW264.7 Macrophages have not been evaluated. In this study, the results shown that compounds 1 and 2 reduced the levels of early pro-inflammatory cytokines TNF-α, IL-1β, the secretion of late pro-inflammatory cytokine HMGB1, as well as the activation of transcription factor NF-κB in RAW 264.7 macrophages induced by LPS. In conclusion, two lupane-type triterpenes (1 and 2) have anti-inflammatory activities in LPS-induced RAW264.7 cells through inhibiting expression of proinflammatory cytokines and NF‐κB activation and could be potentially used in treatment of inflammatory-related diseases in the future.

Abstract: A renewable tannin-based resin adhesive with enhanced bonding strength, good water resistance and long storage life has been prepared based on tannin, furfural and furfuryl alcohol coming from forest and agricultural products. Fourier transform infrared spectroscopy (FT-IR), Electrospray ionization mass spectroscopy (ESI-MS) and Differential scanning calorimetry (DSC) indicated that furfuryl alcohol and furfural can react with tannin under acidic conditions, with the involvement of -CH2-O- groups in the cross-linking of tannin-furfuryl alcohol-furfural resin (TFF) adhesive. The gel time of TFF was longer than that of tannin-furfural resin (TF), while the shear strength of TFF-bonded plywood suggested that the cured TFF adhesive acquired a performance superior to that of tannin (T) and TF adhesives. Furthermore, water resistance of TFF adhesive could be further enhanced with respect to T and TF adhesives through cross-linking with 3 % epoxy resin (EPR).

The predicted changes in precipitation patterns because of global change have profound effects on terrestrial ecosystems. In the present study, the principle and design details of a rainfall enrichment system (RAINES) for open field experiments in semi-arid and arid ecosystems are presented. The rainfall intensity, validity and uniformity of this experimental facility were also tested. During the period from 2008 to 2010, our data showed that the RAINES was able to simulate rainfall events with different rainfall sizes, frequencies and timing. The greatest advantage of the RAINES was its high uniformity in rainfall distribution over a relatively large experimental surface area (>65 m2), which was important for experimental studies of semi-arid and arid ecosystems where vegetation distribution is sparse. The rainfall validity of RAINES was steadily at 66% or higher as long as the hydraulic pressure exceeded 1.4 KPa and the wind speed was below 2.5 m s-1. Since the RAINES is light-weight, inexpensive and versatile enough to be used to simulate various rainfall events with needed properties in remote fields, it is able to provide reliable simulated rainfall in the field for studying possible responses of soil and vegetation processes to rainfall change in arid and semi-arid ecosystems. The application of the RAINES will improve our understanding on the relationship between water availability and ecosystem processes in arid and semi-arid ecosystems, which will provide useful knowledge for the protection, restoration and sustainable management of semi-arid and arid desert ecosystems worldwide.

Los cambios predichos en los modelos de precipitación como resultado del cambio global tienen efectos profundos en los ecosistemas terrestres. Un equipo que simule la lluvia es una herramienta de investigación efectiva para explorar los efectos de los cambios en los modelos de lluvia sin varias restricciones naturales. En el presente estudio, se presentan los principios y detalles de diseño de un equipo simulador de lluvia (RAINES) para estudios a campo en ecosistemas áridos y semiaridos. También se determinaron la intensidad de lluvia, validez y uniformidad del RAINES. Durante el perÃodo 2008 al 2010, nuestros datos mostraron que el RAINES fue capaz de simular eventos de lluvia con diferentes cantidades, frecuencias y momentos de lluvia. La mayor ventaja del RAINES fue su gran uniformidad en la distribución de la lluvia sobre una superficie experimental relativamente grande (>65 m2). Esto es importante para estudios experimentales en ecosistemas áridos y semiáridos donde la distribución de la vegetación es dispersa. La validez de la lluvia provista por el RAINES fue al menos de 66% siempre y cuando la presión hidráulica excedió los 1.4 KPa y la velocidad del viento fue menor que 2.5 m s-1. El RAINES es de bajo peso, no costoso y lo suficientemente versátil como para ser usado para simular varios eventos de lluvia bajo condiciones de campo distantes. Es capaz de proveer lluvia simulada en forma confiable en el campo para estudiar la respuesta de procesos en el suelo y la vegetación a cambios en la cantidad de lluvia en ecosistemas áridos y semiáridos. El uso del RAINES mejorará nuestro entendimiento en la relación entre la disponibilidad de agua y los procesos en ecosistemas áridos y semiáridos. También proveerá conocimiento útil para la protección, restauración y menejo sustentable de ecosistemas de desierto áridos y semiáridos a escala mundial.

Abstract There are several cases of liver abscesses caused by the ingestion of a foreign body, especially in the elderly. Fish bones or chicken bones are sharp foreign bodies that can migrate through the digestive tract to the liver parenchyma. We reported a 71-year-old man who presented to the emergency department with fever and epigastric pain. Computed tomography scan showed a liver abscess related to a long and sharp foreign body which is protruding from the left lobe of the liver. Systemic antibiotic treatment was initiated and later the foreign body was removed by laparoscopic surgery.

Resumen Existen numerosos casos publicados en la literatura que muestran abscesos hepáticos producidos por la ingestión de un cuerpo extraño, especialmente en ancianos. Los huesos de pollo, y con mayor frecuencia las espinas de pescado, pueden perforar el tubo digestivo y migrar hasta el parénquima hepático y originar un absceso. Reportamos el caso de un paciente de 71 años que acude a urgencias por dolor epigástrico y fiebre. Se realizó tomografía computarizada abdominal, que identificó un absceso hepático junto con un cuerpo extraño puntiforme que penetraba en el hígado. El paciente fue intervenido quirúrgicamente, realizando drenaje del absceso y retirada del cuerpo extraño mediante abordaje laparoscópico.

Abstract Aspergillus sp., Fusarium sp., and Ramularia sp. were endophytic fungi isolated from Rumex gmelini Turcz (RGT), all of these three strains could produce some similar bioactive secondary metabolites of their host. However the ability to produce active components degraded significantly after cultured these fungi alone for a long time, and were difficult to recover. In order to obtain more bioactive secondary metabolites, the co-culture of tissue culture seedlings of RGT and its endophytic fungi were established respectively, and RGT seedling was selected as producer. Among these fungi, Aspergillus sp. showed the most significant enhancement on bioactive components accumulation in RGT seedlings. When inoculated Aspergillus sp. spores into media of RGT seedlings that had taken root for 20 d, and made spore concentration in co-culture medium was 1 × 104 mL-1, after co-cultured for 12 d, the yield of chrysophaein, resveratrol, chrysophanol, emodin and physcion were 3.52-, 3.70-, 3.60-, 4.25-, 3.85-fold of the control group. The extreme value of musizin yield was 0.289 mg, which was not detected in the control groups. The results indicated that co-culture with endophytic fungi could significantly enhance bioactive secondary metabolites production of RGT seedlings.

Prostate cancer (PCa) is the second leading cause of cancer death in men. Irradiation is one of the available options for treatment of PCa, however, approximately 10–45% of PCa are resistant to irradiation. We aimed to explore the role of long non-coding RNA highly upregulated in liver cancer (HULC) in the sensitivity of PCa cells to irradiation. Survival rate, cell apoptosis, cycle, expressions of related proteins, and caspase-3 activity were assessed to explore the effects of HULC on sensitivity of PCa cells to irradiation. Expression of HULC in DU-145, PC3, LNCaP, and RWPE-1 cells was determined and the influence of HULC on DU-145 cells was explored. Then, PC3 cells aberrantly expressing HULC were implanted into NOD-SCID mice for tumor xenograft study. Changes of autophagy after aberrant expression of HULC in vivo and in vitro were tested. Furthermore, the interacted protein of HULC and involved signaling pathway were investigated. In PC3 and LNCaP cells under irradiation, survival rate and cell cycle were decreased and apoptosis was increased by HULC knockdown. HULC knockdown arrested PC3 cells at G0/G1 phase. DU-145 was sensitive to irradiation, and resistance to irradiation of DU-145 cells was enhanced by HULC overexpression. Moreover, HULC knockdown enhanced the sensitivity of PC3 xenografts to irradiation. HULC knockdown promoted autophagy through interaction with Beclin-1 and inhibition of mTOR, resulting in increased apoptosis. HULC knockdown improved sensitivity of PCa cells to irradiation both in vivo and in vitro. HULC suppressed Beclin-1 phosphorylation, thereby reduced autophagy, involving the mTOR pathway.

RESUMO Objetivo: Analisar o papel do receptor de hidrocarboneto arílico (AhR) nos linfócitos T CCR6+ CD4+ e CD4+ CD25+ no sangue periférico de pacientes com artrite reumatoide (AR). Métodos: Foi aplicada citometria de fluxo para determinar a proporção de células AhR positivas em linfócitos CCR6+ CD4+ e CD4+ CD25+ do sangue periférico e células mononucleares periféricas de cada indivíduo. Os níveis de expressão relativa de ácido ribonucleico mensageiro (do inglês ribonucleic acid, RNAm,) de AhR e RNAm de enzima de primeiro estágio essencial para o AhR (CYP1A1) foram testados por reação em cadeia de polimerase (do inglês polymerase chain reaction, PCR,) em tempo real. Resultados: A percentagem de células AhR positivas nas células mononucleares do sangue periférico foi maior no grupo com AR do que nos indivíduos saudáveis [(35,23 ± 10,71)% vs. (18,83 ± 7,32)%, (p < 0,01)]. Os níveis de expressão de AhR e CYP1A1 estavam aumentados em pacientes com AR quando comparados com os controles [(3,71 ± 1,63) vs. (2,00 ± 1,27), p = 0,002; (2,62 ± 2,08) vs. (0,62 ± 0,29), p < 0,01, respectivamente]. Em pacientes com AR, a percentagem de células AhR positivas nos linfócitos T CD4+ CD25+ foi significativamente inferior à dos controles [17,90 (6,10 ± 80,10)]% vs. (52,49 ± 19,18)%, p < 0,01]; em controles saudáveis, a percentagem de células AhR positivas nos linfócitos T CD4+ CD25+ foi significativamente mais elevada do que nos linfócitos T CCR6+ CD4+ e também foi significativamente maior do que nas células mononucleares do sangue periférico (do inglês peripheral blood mononuclear cells, PBMC,) [(52,49 ± 19,18)% vs. (23,18 ± 5,62)% vs. (18,06 ± 7,80)%, X 2 = 24,03, p < 0,01]; em pacientes com AR, a percentagem de células AHR positivas nos linfócitos T CCR6+ CD4+ era significativamente maior em comparação com os linfócitos T CD4+ CD25+ e PBMC (46,02 ± 14,68)% vs. [17,90 (6,10 ± 80.10)]% vs. (34,22 ± 10,33)%, X2 = 38,29, p < 0,01]; no entanto, não foi encontrada correlação estatisticamente significativa entre os dados clínicos e células AhR positivas em linfócitos T CCR6+ CD4+ e CD4+ CD25+. Conclusão: O Ahr pode participar do progresso patológico da AR ao controlar a diferenciação de linfócitos Th17 e Treg no sangue periférico.

ABSTRACT Objective: The goal of this study was to analyze the role of aryl hydrocarbon receptor in peripheral blood CCR6+CD4+ and CD4+CD25+T cells of patients with rheumatoid arthritis. Methods: Flow cytometry was applied to determine the proportion of AhR positive cells in CCR6+CD4+T, CD4+CD25+T and peripheral blood peripheral mononuclear cells from each subject. AhR mRNA and CYP1A1 mRNA relative expression levels were tested by real-time PCR. Results: The percentage of AhR positive cells in peripheral blood mononuclear cells was higher in RA group than that in healthy cases [(35.23 ± 10.71)% vs. (18.83 ± 7.32)%, p < 0.01]. The expression levels of AhR and CYP1A1 were both increased in patients with RA while compared to controls [(3.71 ± 1.63) vs. (2.00 ± 1.27), p = 0.002; (2.62 ± 2.08) vs. (0.62 ± 0.29), p < 0.01, respectively]. In RA patients, the percentage of AhR positive cells in CD4+CD25+T cells was significantly lower than that from controls [17.90 (6.10 ± 80.10)% vs. (52.49 ± 19.18)%, p < 0.01]; In healthy controls, the percentage of AhR positive cells in CD4+CD25+T cells was significantly higher than that in CCR6+CD4+T cells, and was also significantly higher than that in PBMCs [(52.49 ± 19.18)% vs. (23.18 ± 5.62)% vs. (18.06 ± 7.80)%, X 2 = 24.03, p < 0.01]; in RA patients, the percentage of AhR positive cells in CCR6+CD4+T cells was significantly increased than that in CD4+CD25+T cells and PBMCs [(46.02 ± 14.68)% vs. 17.90 (6.10 ± 80.10)% vs. (34.22 ± 10.33)%, X 2 = 38.29, p < 0.01]; Nevertheless, no statistically significant relationship was found between clinical data and AhR positive cells in CCR6+CD4+T and CD4+CD25+T cells. Conclusion: AhR may participate in the pathological progress of RA by controlling the differentiation of Th17 and Treg cells in peripheral blood.

ABSTRACT Graft-versus-host disease (GVHD) is a lethal complication of allogeneic haematopoietic stem cell transplantation which limits its application. Psoralen was utilized in the treatment of GVHD as a second-line method, which is also known as extracorporeal photochemotherapy (ECP). In the process of ECP, mononuclear cells must be isolated from the body in advance. Then a photosensitizer, 8-methoxypsoralen (8-MOP, 200 μg/L for the final concentration), would be added to the cell suspension before its exposure to ultraviolet A [UVA; 365 nm, 2J/cm2]. The disposed lymphocytes re-infused into the body account for 5% to 15% of the lymphocytes in the body. The process of ECP is complicated, expensive and very labour intensive, which limits its popularity. We hypothesized that psoralen should be activated by UVA, and should be kept in activation for a relatively long time before it gets in contact with mononuclear cells. This kind of psoralen is called previously activated psoralen (PAP), which may have the same effects on GVHD as ECP, but would be much easier and economical to work with.

RESUMEN La enfermedad de injerto-contra-huésped (EICH, o GVHD por las siglas en inglés) es una complicación letal del trasplante alogénico de células madre hematopoyéticas que limita su aplicación. El psoraleno se utilizó en el tratamiento de EICH como método de segunda línea, también conocido como fotoquimioterapia extracorpórea (F EC). En el proceso de FEC, las células mononucleares tienen que ser aisladas del cuerpo por adelantado. Entonces un fotosensibilizador, 8-methoxypsoraleno (8-MOP, 200 μg/L para la concentración final), se añadirá a la suspensión de células antes de su exposición a radiación ultravioleta A [UVA; 365 nm, 2J/cm2]. Los linfocitos eliminados re-infundidos en el cuerpo representan del 5% al 15% de los linfocitos en el cuerpo. El proceso de FEC es complicado, costoso, y requiere mucho trabajo, lo cual limita su popularidad. Partimos de la hipótesis de que el psoraleno debe ser activado por rayos UVA y debe mantenerse en activación por un tiempo relativamente largo, antes de que se ponga en contacto con las células mononucleares. Este tipo de psoraleno se denomina psoraleno activado previamente (PAP), el cual puede tener los mismos efectos sobre la EICH que la FEC, pero de manera mucho más fácil y económica.

The northern pike (Esox lucius L.), an important predatory freshwater species, is undergoing significant population decline. In this study, 18 novel polymorphic microsatellite loci were isolated and used for assessing genetic variation in the Chinese Ulungur and Hungarian Balaton populations of the species. The number of alleles ranged from 2 to 13, observed heterozygosity from 0.154 to 0.920 and expected heterozygosity from 0.145 to 0.921, thereby indicating the specific usefulness of these suites of markers for investigating genetic variability.

A large set of candidate nucleotide-binding site (NBS)-encoding genes related to disease resistance was identified in the sorghum (Sorghum bicolor) genome. These resistance (R) genes were characterized based on their structural diversity, physical chromosomal location and phylogenetic relationships. Based on their N-terminal motifs and leucine-rich repeats (LRR), 50 non-regular NBS genes and 224 regular NBS genes were identified in 274 candidate NBS genes. The regular NBS genes were classified into ten types: CNL, CN, CNLX, CNX, CNXL, CXN, NX, N, NL and NLX. The vast majority (97%) of NBS genes occurred in gene clusters, indicating extensive gene duplication in the evolution of S. bicolor NBS genes. Analysis of the S. bicolor NBS phylogenetic tree revealed two major clades. Most NBS genes were located at the distal tip of the long arms of the ten sorghum chromosomes, a pattern significantly different from rice and Arabidopsis, the NBS genes of which have a random chromosomal distribution.
