OBJETIVO: avaliar o efeito do uso prolongado de alta dose de tibolona na morfologia do endométrio em ratas castradas. MÉTODOS: foram utilizadas 15 ratas Wistar, fêmeas, com idade de oito semanas e peso médio de 250 g. Todas as ratas foram submetidas à ooforectomia bilateral e 30 dias depois foi coletada citologia vaginal, verificando-se o status de menopausa. As ratas foram divididas aleatoriamente em dois grupos: o tratado (n=9) recebeu via oral 1 mg tibolona/dia; o controle (n=6) recebeu apenas solução do veículo carboximetilcelulose. Após 20 semanas de tratamento, todos os animais foram sedados e sacrificados por deslocamento cervical. Os úteros foram retirados e fixados em formol 10% tamponado. Ambos os cornos uterinos foram clivados em três regiões (proximal, medial, distal) e processados para inclusão em parafina. Cortes histológicos corados com hematoxilina-eosina foram submetidos à análise morfológica e morfométrica. Foram avaliados os seguintes parâmetros: espessura do epitélio superficial endometrial, espessura do estroma endometrial, área endometrial, número absoluto de glândulas endometriais e número de glândulas/área endometrial. Os dados obtidos foram comparados mediante o teste t de Student. RESULTADOS: no Grupo Tibolona os úteros se apresentaram bem desenvolvidos e houve aumento significativo (p<0,01) de todos os parâmetros histomorfométricos. Por vezes, o epitélio cilíndrico tornava-se estratificado pavimentoso e recobria porções internas das glândulas bem como a cavidade endometrial. As ratas do Grupo Controle apresentaram útero atrofiado. Havia poucas glândulas de padrão tubular e escassa substância intercelular. As glândulas eram revestidas de epitélio cúbico que se estendia à cavidade endometrial. CONCLUSÕES: a tibolona em alta dose administrada por um longo período de tempo à ratas castradas tem efeito estrogênico, que pode ser dose-dependente, e causa proliferação no endométrio e alteração da diferenciação celular (metaplasia escamosa), porém não leva à hiperplasia.

PURPOSE: to evaluate the effect of long-term use of a high dose of tibolone on the morphology of the endometrium of castrated female rats. METHODS: fifteen female Wistar rats, aged eight weeks and weighting about 250 g were used. All the female rats were submitted to bilateral oophorectomy and 30 days afterwards, vaginal cytology was collected, to verify the menopause status. The female rats were randomly divided in two groups. The Treatment Group (n=9) received 1 mg of tibolone/day orally; the Control Group (n=6) received a solution of carboxymethylcellulose vehicle. After 20 weeks of treatment, all the animals were sedated and sacrificed by cervical dislocation. The uterus was removed and fixated in 10% buffer formaldehyde. Both uterine horns were divided in three regions (proximal, medial and distal) and processed to be included in paraffin. Histological sections, stained with hematoxylin-eosin were submitted to morphological and morphometrical analysis. The following parameters have been analyzed: thickness of the endometrial superficial epithelium, thickness of the endometrium stroma, endometrial area, absolute number of endometrial glands and number of glands/endometrial area. The data obtained were compared by the t-Student test. RESULTS: in the Tibolone Group, the uteri were well developed and there was a significant increase (p<0.01) of all the histomorphometric parameters. In some cases, the cylindrical epithelium became stratified, pavimentous and covered the internal portions of the glands, as well as of the endometrium cavity. Rats from the Control Group presented uterine atrophy. There were few tubular-like glands and scarce intercellular substance. Glands were covered by cubic epithelium which extended itself to the endometrial cavity. CONCLUSIONS: high doses of tibolone, given for long periods of time to castrated female rats, have an estrogenic effect which can be dose-dependent, causing proliferation in the endometrium and causing changes in the cell differentiation (squamous metaplasia), but do not lead to hyperplasia.

Parasympathetic dysfunction is an independent risk factor in patients with coronary artery disease; thus, cholinergic stimulation is a potential therapeutic measure that may be protective by acting on ventricular repolarization. The purpose of the present study was to determine the effects of pyridostigmine bromide (PYR), a reversible anticholinesterase agent, on the electrocardiographic variables, particularly QTc interval, in patients with stable coronary artery disease. In a randomized double-blind crossover placebo-controlled study, simultaneous 12-lead electrocardiographic tracings were obtained at rest from 10 patients with exercise-induced myocardial ischemia before and 2 h after the oral administration of 45 mg PYR or placebo. PYR increased the RR intervals (pre: 921 ± 27 ms vs post: 1127 ± 37 ms; P<0.01) and, in contrast with placebo, decreased the QTc interval (pre: 401 ± 3 ms vs post: 382 ± 3 ms; P<0.01). No other electrocardiographic variables were modified (PR segment, QT interval, QT and QTc dispersions). Cholinergic stimulation with PYR caused bradycardia and reduced the QTc interval without important side effects in patients with coronary disease. These effects, if confirmed in studies over longer periods of administration, may suggest a cardioprotection by cholinergic stimulation with PYR.

Os efeitos da desnutrição sobre o músculo «gastrccnemius plantaris» de ratos albinos, limitando-se à genitora a dieta em 6,7% de proteínas e aos seus produtos, em 3,2% após o desmame, foram observados em microscopia óptica e eletrônica e comparados com os controles. Os animais, em número de 40, foram distribuídos em grupo hipoproteico e normal, sendo a metade de cada grupo sacrificada aos 15 dias e a restante, após o desmame, aoa 30 dias. Observamos acentuada redução ponderai dos ratos com alimentação bipoproteica, chegando a cerca de 50% em relação aos normais. As fibras do músculo estudado, nos ratos desnutridos, apresentaram importante redução de seus diâmetros. As reações histoquímicas revelaram que tanto as fibras do tipo I como as do tipo II estavam comprometidas, ocorrendo maior redução nas últimas. Fibras do tipo II, de pequena espessura, com aspecto de fibra P (fetal) foram encontradas nos animais subnutridos de 15 dias, denotando retardo na maturação destas fibras. As avaliações em ultramicroscopia não mostraram alterações específicas dia ultra-estrutura muscular, revelando apenas intensa redução do calibre das fibras, quando comparadas às dos ratos normais. Concluímos pela hipoplasia e não atrofia do tecido muscular dos animais submetidos a desnutrição pré e pós-natal. O presente estudo, somado a outros nos quais foram analisados a ponta anterior da medula e o nervo periférico de ratos desnutridos, permite-nos considerar que na privação proteica ocorrem retardo no desenvolvimento e hipoplasia da unidade mootra. Conjecturamos a possibilidade de, na amiotrofía espinhal progressiva da infância (doença de Werdnig-Hoffmann), acontecer também hipoplasia das fibras do tipo II e não atrofia, aventando a hipótese de, nesta doença, haver alterações no metabolismo proteico.

The effects of undernutrition on the «gastrocnemius plantaris» muscle of young albino nats were observed with light and electronmicroscopy and were compared with controls. Pregnant rats received a diet containing 6.7% protein and the neonates had a 3.2% protein diet after weaning. A total number of 40 animals were distributed in two groups: one nypoproteic and one control. Half of the animals of each group were killed 15 days after birth and the remaining ones at 30 days. We could observe important reduction in the weight of the undernourished rats reaching about 50% when compared with the control animals. An important reduction in the diameter of muscle fibers was noted in undernourished rats. Histochemical reactions showed that not only the type I but also the type II fibers were involved, the latter being more severely reduced in size. Type II fibers os small diameter, resembling F (fetal) fiber were found in animals at 15 days indicating a delay in maturation. TJltrastructural evaluation of the muscle did not show specific changes except for a severe reduction in the caliber of fibers when compared with control animals. We concluded that there was hypoplasia and not atrophy of the muscular tissue in animals submitted to pre- and post-natal undernutrition. The present study, associated with others in which the spinal motor cells and peripheral nerves of undernourished rats were analysed, allow us to consider that with protein deprivation there is a delay in the development, a hypoplasia of the motor unit. We think that in infantile progressive spinal amyotrophy (Werdnig-Hoffmann disease) there is possibly hypoplasia and not atrophy of the type II fibers and we postulate that a metabolic proteic problem is involved in this disease.
