Lipid-core nanocapsules (LNCs) are core-shell structures, in which the core is an organogel formed by a solid lipid dispersed in a triglyceride, and the shell is a polymer. Those nanocapsules encapsulate poorly-water soluble drugs by dispersion in the core and/or interaction with the polymer. The core and the shell act as diffusional barriers differenting the LNC from previously developed polymeric nanocapsules containing an oily core and a polymer shell, which unique diffusional barrier is the shell. In this review, we discuss the supramolecular structure, the physico-chemical characterization, including sizing, surface potential, fluorescent labeling strategies, as well as the interfacial reactions in water to coat and functionalise the LNC surface. Different organometallic complexes have been synthesized to decorate the nanocapsules envisaging the treatment of atherosclerosis, cancer, mucopolysaccharidosis I, and hypertension. We also describe the main applications of the nanoformulations in the pharmaceutical, medical, and veterinary fields, including in vitro and in vivo evaluations of toxicity and efficacy. In this work we present a simple model for the kinetics of agglomeration and aggregation of colloidal particles. We consider that particles agglomerate rapidly and endothermically forming oligomers. These oligomers can, in turn, aggregate irreversibly, in a process that leads to the destabilization of the colloidal system. As these two processes have very different relative energy activations, they occur in different time-scales: the first step is faster and reaches a state of quasi‑equilibrium. Because of this, the enthalpy change during the agglomeration can be experimentally determined through the variable temperature multiple light scattering (VTMLS) method. Interestingly, this value is related to the relative kinetic stability of the system and can be used to evaluate the stability of new colloidal compositions. Our results are in qualitative agreement with experimental data of low concentration colloidal dispersions consisted of polymer particles and/or surfactant-coated particles.

In spite of different methods reported in the literature to determine olanzapine in biological fluids, all of them used high volumes of plasma. Therefore, the purpose of this paper was to develop an LC-MS/MS method using small plasma volume (0.1 mL) to apply in a preclinical pharmacokinetic investigation. The method was linear over the concentration ranges of 10 - 1000 ng mL-1. Extraction recoveries, stability, and validation parameters were evaluated. Results were within the acceptable limits of international guidelines. A significant decrease in clearance led to a significant 2.26-times increase in AUC0 - 6h of olanzapine-loaded lipid-core nanocapsules compared with free-olanzapine.

Several studies have described the benefits of nanoscience and nanotechnology (N&N) in different sectors such as agriculture, energy, environmental preservation, and public health. The rapid evolution of N&N can be shown through a panoramic analysis of scientific papers and patents. In the area of public health, it is estimated that the global market for nanotechnology products will expand to 160 billion U.S. dollars in 2015. The Brazilian government has also strengthened its innovative potential in N&N through economic subsidies, as observed for other countries. This review is focused on the current landscape of N&N in a therapeutic context, highlighting the development of nanotech-products produced with biocompatible and biodegradable materials that are already commercially available or under investigation. Most studies under investigation are focused on the development of nanotechnology-based formulations intended for treatment of cancer, inflammatory, cardiovascular, and neurological diseases. Although there are several advantages of N&N in healthcare, many challenges have to be conquered to increase the availability of nanotechnology products in toxicological, preclinical and clinical studies, scale-up, regulatory, and private investments.

Polymeric nanoparticle systems (nanocapsules and nanospheres) present potential applications as drug delivery systems. Nevertheless, their full applications have not been exploited due to their limited shelf life when stored in aqueous medium. Drying polymeric nanoparticles using spray-drying represents a promising platform to improve the physicochemical stability of formulations and/or to control the release of hydrophilic and lipophilic drugs. This article presents a brief overview of the most recent and ongoing research in the use of spray-drying process to prepare and/or to dry polymeric nanoparticles formulations intended for drug administration.

This paper was devoted to studying the influence of coating material (nanocapsules or nanospheres), drug model (diclofenac, acid or salt) and method of preparation on the morphological characteristics of nanoparticle-coated microparticles. The cores of microparticles were obtained by spray drying or evaporation and the coating was applied by spray drying. SEM analyses showed nanostructures coating the surface of nanocapsule-coated microparticles and a rugged surface for nanosphere-coated microparticles. The decrease in their surface areas was controlled by the nanoparticulated system, which was not dependent on microparticle size. Optical microscopy and X-ray analyses indicated that acid diclofenac crystals were present in formulations prepared with the acid as well as in the nanocapsule-coated microparticles prepared with the salt. The control of coating is dependent on the use of nanocapsules or nanospheres and independent of either the characteristics of the drug or the method of preparing the core.
