Abstract Background: Real-world, primary data on the treatment of psoriasis are scarce, especially concerning the role of soluble biomarkers as outcome predictors. Objective: The authors evaluated the utility of Th1/Th17 serum cytokines along with clinical characteristics as predictors of drug survival in the treatment of psoriasis. Methods: The authors consecutively included participants with moderate to severe psoriasis who were followed up for 6 years. Baseline interferon-α, tumor necrosis factor-α, and inter-leukin (IL)-2, IL-4, IL-6, IL-10, and IL-17A were measured using a cytometric bead array; clinical data were assessed. The authors calculated hazard ratios (HRs) for drug survival using a Cox proportional hazards model. Results: The authors included 262 patients, most of whom used systemic immunosuppressants or biologics. In the multivariate model, poor quality of life measured by the Dermatology Life Quality Index (HR = 1.04; 95% CI 1.01–1.07; p = 0.012) and elevated baseline IL-6 (HR = 1.99; 95% CI 1.29–3.08; p = 0.002) were associated with treatment interruption. Study limitations: The main limitation of any cohort study is the presence of confounders that could not be detected in clinical evaluation. Conclusions: Poor quality of life and elevated baseline serum IL-6 level predicted treatment interruption in patients with moderate to severe psoriasis. Although IL-6 is not the most important mediator of the inflammatory pathway in the skin environment, it is an interesting biomarker candidate for predicting psoriasis treatment response. Resumen Objetivo Describir el proceso de desarrollo y validación de un software web de apoyo a la gestión de la atención prenatal. Métodos Investigación de producción tecnológica, realizada entre marzo y agosto de 2022, con base en principios y técnicas de la metodología Scrum. Las funcionalidades del software web, con acceso y funciones específicas para tres usuarios (administrador, gestor y visitante), se desarrollaron de forma colegiada entre desarrolladores, investigadores, gestores y profesionales de la salud, a partir de necesidades prácticas relacionadas con la gestión de la atención prenatal. Resultados El software web es una herramienta de apoyo a la gestión de la atención prenatal, con potencial para generar infográficos que indiquen complicaciones gestacionales y que permitan tomar decisiones estratégicas locorregionales de forma ágil y segura. ABSTRACT Sampling is an essential step in estimating a parameter: thus, cost and time associated to this step should be minimized. Sequential sampling is characterized by using samples of variable sizes given as a function of observations, and sequential sampling provides a smaller sample size than a fixed-size sample in most cases. In addition, the Bayesian decision theory can be incorporated into sequential sampling to perform parameter estimation because it allows the inclusion of a priori information about the parameter of interest, which optimizes the procedure. However, the great challenge to performing the Bayesian sequential estimation in establishing the stopping criteria. Most studies in this area investigate binomial distributions, while few analyze multinomial distributions. This study aimed to define the stopping criteria for the Bayesian sequential estimation of the parameters of multinomial distributions with conjugate Dirichlet priors. The proposed methodology was applied to a set of X-ray test data for quality control of maize seed lots. This test uses conventional sampling techniques in which a sample has a fixed size with 200 seeds. The influence of two priors on the stopping criteria was evaluated, one uniform and one conjugate, with hyperparameters based on reference information from the literature. Abstract Pathogenic DNA alterations in GJB2 are present in nearly half of non-syndromic hearing loss cases with autosomal recessive inheritance. The most frequent variant in GJB2 causing non-syndromic hearing loss is the frameshifting c.35del. GJB2 encodes Cx26, a protein of the connexin family that assembles hemichannels and gap junctions. The expression of paralogous proteins is believed to compensate for the loss of function of specific connexins. As Cx26 has been involved in cell differentiation in distinct tissues, we employed stem cells derived from human exfoliated deciduous teeth (SHEDs), homozygous for the c.35del variant, to assess GJB2 roles in stem cell differentiation and the relationship between its loss of function and the expression of paralogous genes. Primary SHED cultures from patients and control individuals were compared. SHEDs from patients had significantly less GJB2 mRNA and increased amount of GJA1 (Cx43), but not GJB6 (Cx30) or GJB3 (Cx31) mRNA. In addition, they presented higher induced differentiation to adipocytes and osteocytes but lower chondrocyte differentiation. Our results suggest that GJA1 increased expression may be involved in functional compensation for GJB2 loss of function in human stem cells, and it may explain changes in differentiation properties observed in SHEDs with and without the c.35del variant. Abstract Objective Kidney shortage for pediatric kidney transplantation (PKT) entails the need to use low-weight and age donors, despite the apprehension. The aim of this study was to analyze the pediatric deceased donor kidney transplantations (pDDKT) outcomes in the first year after the procedure, stratified by donor age. Method Retrospective cohort of pDDKTs carried out between January 2013, and January 2018, at a PKT reference hospital in Southern Brazil. Donors were divided into group 1 (≤ 6 years), and group 2 (> 6 years); the analysis of the outcomes was carried out in the same period. Results There were 143 pDDKTs; 51 (35.66%) in group 1; and 92 (64.34%) in group 2. In both groups there were 17 graft losses (11.8%), with vascular thrombosis as the main cause (group 1: 5; group 2: 4). Among the complications, renal artery stenosis (RAS) with indication for angioplasty and stenting was more frequent in group 1 (7.8%; group 2: 2.2%). The 1-year Renal Transplant Recipients' and graft survival did not show significant differences between the groups, (p= = 0.95). However, the Glomerular Filtration Rate analysis was higher in group 2, reaching, in the 12th month, 79.3 mL/min/1,73m2, compared to 69.7 mL/min/1,73m2 in group 1(p= = 0.033). Conclusions Small donors can be considered for pDDKTs, as long as there is an expert team to perform the transplantation. ABSTRACT This study aimed to identify factors associated with colonization by community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA) in adult patients admitted to a Brazilian hospital. This is a cross-sectional study, in which patients underwent a nasal swab and were asked about hygiene behavior, habits, and clinical history. Among the 702 patients, 180 (25.6%) had S. aureus and 21 (2.9%) MRSA. The factors associated with MRSA colonization were attending a gym (OR 4.71; 95% CI; 1.42 – 15.06), smoking habit in the last year (OR 2.37; 95% CI; 0.88 – 6.38), previous hospitalization (OR 2.18; CI 95%; 0.89 – 5.25), and shared personal hygiene items (OR 1.99; 95% CI; 0.71 – 5.55). At the time of admission, colonization by CA-MRSA isolates was higher than that found in the general population. This can be an important public health problem, already endemic in hospitals, whose factors such as those associated with habits (smoking cigarettes) and behaviors (team sports practice and activities in gyms) have been strongly highlighted. Resumo: A preocupação com o bem-estar dos outros faz parte do desenvolvimento normativo. Contudo, enquanto algumas crianças respondem à angústia dos outros com preocupação e ajuda, outras respondem com suspeita, hostilidade e indiferença. Embora a literatura em torno da empatia tenha aumentado nos últimos anos, ainda não há consenso no que respeita as suas associações com a pró-socialidade, com problemas internalizantes ou externalizantes. Numa amostra de 199 crianças (50,8% meninas) entre os 10 e os 15 anos (M=12.05; DP=0.98), foram reportados a empatia e os comportamentos sociais através do QACE - Questionnaire to Assess Affective and Cognitive Empathy (Zoll & Enz, 2010). e o SDQ - Strengths and Difficulties Questionnaire (Goodman, 1997), respetivamente. Os resultados indicam as raparigas como mais pró-sociais e empáticas, mas também com níveis mais elevados de problemas internalizantes, em comparação com os rapazes. A empatia afetiva, mas não cognitiva, foi relacionada com problemas internalizantes. A empatia cognitiva foi significativamente relacionada com o comportamento pró-social. Não foram encontradas relações significativas entre empatia e comportamentos externalizantes.

Abstract: Concern for others’ welfare is part of normative development. However, some children respond to others’ distress with concern and helpful approaches, while others respond with suspicion, hostility, indifference. Although the literature around empathy has increased over the years, there isn’t a consensus over its associations with prosociality and internalizing or externalizing problems. A sample of 199 children (50.8% girls) between 10 and 15 years (M=12.05; SD=0.98), reported on their empathy and social behaviours using the QACE - Questionnaire to Assess Affective and Cognitive Empathy (Zoll & Enz, 2010) and the SDQ - Strengths and Difficulties Questionnaire (Goodman, 1997) respectively. Our results indicate that girls were more prosocial and empathic, but also presented higher levels of internalizing problems, compared to boys. Affective but not cognitive empathy was related with internalizing problems. Cognitive empathy was significantly related with prosocial behaviour. No significant relations between empathy and externalizing behaviours were found.

ABSTRACT The herpes simplex virus type 1 (HSV-1) and type 2 (HSV-2) occur worldwide. Infections caused by these viruses have great public health importance due to the growing resistance to the first-choice drug, acyclovir, especially in immunosuppressed patients. Alkaloids derived from species of Annonaceae have been reported as antiviral agents against HSV and others viruses. Within this context, we evaluated the antiviral activity of the total alkaloid fraction (TAF) extracted from the branches of Fusaea longifolia (Aubl.) Saff. (Annonaceae), a species native to the Amazon region, against the HSV-1 and HSV-2 viruses. The antiviral activity was evaluated through the plate reduction assay and the mode of action was investigated by a set of other assays. The TAF was active against the HSV-2 strain 333 and against the HSV-1 strains KOS and 29R (acyclovir resistant), with selectivity index values (SI = 50% cytotoxic concentration/50% effective concentration) of 5, 4 and 3, respectively. In the preliminary study of the anti-HSV-2 mode of action, TAF showed viral inhibitory effects if added up to 12 h post-infection, had virucidal activity and did not present viral inhibition in pre-treatment. Our results showed that the TAF exhibited anti-HSV activity. Regarding HSV-2, TAF acted after the viral infection and had virucidal activity. A mass spectrometry analysis revealed the presence of nine alkaloids in the TAF that had previously been reported for Annonaceae, including liriodenine, lysicamine and isoboldine, which have been described as potential anti-HSV-1 agents. RESUMO Os vírus herpes simplex tipo 1 (HSV-1) e tipo 2 (HSV-2) têm ampla ocorrência global. As infecções causadas por esses vírus têm grande importância em saúde pública devido à crescente resistência ao fármaco de primeira linha, aciclovir, principalmente em pacientes imunossuprimidos. Alcaloides derivados de espécies de Annonaceae têm sido relatados como agentes antivirais contra o HSV e outros vírus. Neste contexto, nós avaliamos a atividade antiviral da fração alcaloide total (TAF) dos ramos de Fusaea longifolia (Aubl.) Saff. (Annonaceae), uma espécie nativa da região amazônica, contra os vírus HSV-1 e HSV-2. A atividade antiviral foi avaliada através do ensaio de redução em placa e o modo de ação foi investigado por um conjunto de ensaios. O TAF foi ativo contra a cepa HSV-2 333 e contra as cepas HSV-1 KOS e 29R (resistente ao aciclovir), com valores de índice de seletividade (IS = 50% concentração citotóxica/50% concentração efetiva) de 5, 4 e 3, respectivamente. No estudo preliminar do modo de ação da atividade anti-HSV-2, o TAF inibiu a replicação viral quando adicionados até 12 h pós-infecção, apresentou atividade virucida e não apresentou inibição viral no pré-tratamento. Nossos resultados mostraram que o TAF exibiu atividade anti-HSV. Em relação ao HSV-2, o TAF atuou após a infecção viral e apresentou atividade virucida. RESUMO: Com o objetivo de avaliar a ocorrência de depressão pós-parto e alguns fatores sociodemográficos associados, entre mulheres acompanhadas na Atenção Primária à Saúde, um questionário sociodemográfico e a Escala de Depressão Pós-parto de Edimburgo foram aplicados a 123 puérperas entre a segunda semana e o sexto mês após o parto. A frequência de depressão pós-parto na amostra foi determinada e sua associação com variáveis sociodemográficas foi avaliada por meio do teste do qui-quadrado. A provável depressão pós-parto foi identificada em 19,51% das participantes, e essa condição foi associada à faixa etária (36 a 44 anos), idade do bebê (4 ou 6 meses), estado civil (divorciado), baixa renda e não houve associação com a escolaridade materna. Assim, a depressão pós-parto precisa ser investigada na atenção primária à saúde, atentando para os aspectos sociodemográficos e individuais. Esses aspectos precisam ser valorizados pelo enfermeiro, que deve estabelecer um plano assistencial integral desde o pré-natal, para prevenir esse frequente transtorno puerperal.

RESUMEN: Con el objetivo de rastrear la depresión pos parto entre las mujeres em Atención Primaria de Salud, se aplicó un cuestionario sociodemográfico y la Escala de Depresión Postnatal de Edimburgo a 123 puérperas entre la segunda semana y el sexto mes después del parto. Se determinó la frecuencia de depresión pos parto en la muestra y se evaluó su asociación con variables sociodemográficas mediante la prueba de chi-cuadrado. Se identifico depresión posparto probable en el 19,51% de las participantes, y esta condición se asoció con el grupo de edad (36 a 44 años), edad del bebé (4 o 6 meses), estado civil (divorciada), bajos ingresos y no había asociación con la escolarización materna. Por lo tanto, la depresión del parto necesita ser investigada en la atención primaria de salud, prestando atención a los aspectos sociodemográficos e individuales. Estos aspectos necesitan ser valorados por las enfermeras, qui deben establecer un plan de cuidados integral desde el prenatal, para prevenir este frecuente trastorno puerperal.

ABSTRACT: Aiming to track postpartum depression among women followed in Primary Health Care, a sociodemographic questionnaire and the Edinburgh Postnatal Depression Scale were applied to 123 postpartum women between the second week and the sixth month after delivery. The frequency of postpartum depression in the sample was determined, and its association with sociodemographic variables was assessed using the chi-square test. Probable postpartum depression was identified in 19.51% of the participants, and this condition was associated with age group (36 to 44 years), age of the baby (4 or 6 months), marital status (divorced), low income and there was no association with maternal schooling. Thus, postpartum depression needs to be investigated in primary health care, paying attention to sociodemographic and individual aspects. These aspects need to be valued by nurses, who must establish a comprehensive care plan from prenatal care, to prevent this frequent puerperal disorder.

ABSTRACT To evaluate the effect of different iron dextran application programs on the performance, fecal score, and skin color of suckling piglets, as well as sow performance, 288 piglets from 24 sows were allocated to four treatments in six replications with 12 piglets per experimental unit, in a block design. The treatments were as follows: T200_000, T200_100, T200_200, and T100_100, whose respective values (in mg) corresponded to the applications of the first dose of iron dextran on the second day of life and of the second dose on the 10th day. Piglets on T200_000 and T200_200 showed the highest feed intake. In the period from 10 to 22 days, piglets receiving T100_100 exhibited the highest fecal score. On the 10th day of age, the lowest lightness (L*) value on the ear was obtained with T200_100, and the highest with T100_100. Treatments T200_000, T200_100, and T200_200 generated the highest red color (a*) intensity on the ear, whereas T100_100 provided the lowest hue and parameter b* values on the leg and snout. On the 22nd day of age, the highest b* and hue values of the ear and snout were found in the group fed T100_100. In conclusion, all evaluated programs were efficient in preventing iron deficiency anemia in newborn piglets. RESUMO Foram utilizados 288 leitões de 24 matrizes, distribuídos em delineamento em blocos com quatro tratamentos, seis repetições, com 12 leitões por unidade experimental com objetivo de avaliar diferentes programas de aplicação de ferro dextrano sobre desempenho, escore fecal e coloração de pele de leitões em aleitamento, assim como as repercussões sobre o desempenho das matrizes. Os tratamentos consistiram em: T200_000; T200_100; T200_200; e T100_100; nos quais os valores corresponderam, respectivamente (em mg), à aplicação da primeira dose de ferro dextrano no segundo dia de vida e à aplicação da segunda dose no 10º dia. Os leitões que foram submetidos a T200_000 e a T200_200 tiveram maior consumo de ração. No período de 10 a 22 dias, os leitões de T100_100 obtiveram maior escore fecal. Ao 10º dia de idade dos leitões, o menor valor de luminosidade (L*) da orelha foi em T200_100 e o maior valor em T100_100. A maior intensidade da cor vermelha (a*) na orelha foi obtida em T200_000, T200_100 e T200_200. Na tonalidade e no parâmetro b* do pernil e do focinho, T100_100 foi aquele que apresentou menor valor. No 22º dia de idade dos leitões, o maior valor de b* e da tonalidade da orelha e do focinho foi verificado em T100_100. Conclui-se que todos os programas avaliados foram eficientes na prevenção da anemia ferropriva em leitões neonatos. Abstract Objective To evaluate clinical characteristics and outcomes of COVID-19 patients infected with HIV, and to compare with a paired sample without HIV infection. Methods This is a substudy of a Brazilian multicentric cohort that comprised two periods (2020 and 2021). Data was obtained through the retrospective review of medical records. Primary outcomes were admission to the intensive care unit, invasive mechanical ventilation, and death. Patients with HIV and controls were matched for age, sex, number of comorbidities, and hospital of origin using the technique of propensity score matching (up to 4:1). They were compared using the Chi-Square or Fisher's Exact tests for categorical variables and the Wilcoxon for numerical variables. Results Throughout the study, 17,101 COVID-19 patients were hospitalized, and 130 (0.76%) of those were infected with HIV. The median age was 54 (IQR: 43.0;64.0) years in 2020 and 53 (IQR: 46.0;63.5) years in 2021, with a predominance of females in both periods. People Living with HIV (PLHIV) and their controls showed similar prevalence for admission to the ICU and invasive mechanical ventilation requirement in the two periods, with no significant differences. In 2020, in-hospital mortality was higher in the PLHIV compared to the controls (27.9% vs. 17.7%; p = 0.049), but there was no difference in mortality between groups in 2021 (25.0% vs. 25.1%; p > 0.999). Conclusions Our results reiterate that PLHIV were at higher risk of COVID-19 mortality in the early stages of the pandemic, however, this finding did not sustain in 2021, when the mortality rate is similar to the control group. Abstract Aims Although reduced life expectancy in Parkinson's Disease (PD) patients has been related to severe cardiac arrhythmias due to autonomic dysfunctions, its molecular mechanisms remain unclear. To investigate the role of cardiac β1-Adrenergic (β1AR) and A1-Adenosine (A1R) receptors in these dysfunctions, the pharmacological effects of stimulation of cardiac β1AR (isoproterenol, ISO), in the absence and presence of cardiac β1AR (atenolol, AT) or A1R (1,3-dipropyl-8-cyclopentyl xanthine, DPCPX) blockade, on the arrhythmias induced by Ischemia/Reperfusion (CIR) in an animal PD model were studied. Methods PD was produced by dopaminergic lesions (confirmed by immunohistochemistry analysis) caused by the injection of 6-hydroxydopamine (6-OHDA, 6 μg) in rat striatum. CIR was produced by a surgical interruption for 10 min followed by reestablishment of blood circulation in the descendent left coronary artery. On the incidence of CIR-Induced Ventricular Arrhythmias (VA), Atrioventricular Block (AVB), and Lethality (LET), evaluated by Electrocardiogram (ECG) analysis, the effects of intravenous treatment with ISO, AT and DPCPX (before CIR) were studied. Results VA, AVB and LET incidences were significantly higher in 6-OHDA (83%, 92%, 100%, respectively) than in control rats (58%, 67% and 67%, respectively). ISO treatment significantly reduced these incidences in 6-OHDA (33%, 33% and 42%, respectively) and control rats (25%, 25%, 33%, respectively), indicating that stimulation of cardiac β1AR induced cardioprotection. This response was prevented by pretreatment with AT and DPCPX, confirming the involvement of cardiac β1AR and A1R. Conclusion Pharmacological modulation of cardiac β1AR and A1R could be a potential therapeutic strategy to reduce severe arrhythmias and increase life expectancy in PD patients. Abstract Background The 6-OHDA nigro-striatal lesion model has already been related to disorders in the excitability and synchronicity of neural networks and variation in the expression of transmembrane proteins that control intra and extracellular ionic concentrations, such as cation-chloride cotransporters (NKCC1 and KCC2) and Na+/K+-ATPase and, also, to the glial proliferation after injury. All these non-synaptic mechanisms have already been related to neuronal injury and hyper-synchronism processes. Objective The main objective of this study is to verify whether mechanisms not directly related to synaptic neurotransmission could be involved in the modulation of nigrostriatal pathways. Methods Male Wistar rats, 3 months old, were submitted to a unilateral injection of 24 µg of 6-OHDA, in the striatum (n= 8). The animals in the Control group (n= 8) were submitted to the same protocol, with the replacement of 6-OHDA by 0.9% saline. The analysis by optical densitometry was performed to quantify the immunoreactivity intensity of GFAP, NKCC1, KCC2, Na+/K+-ATPase, TH and Cx36. Results The 6-OHDA induced lesions in the striatum, were not followed by changes in the expression cation-chloride cotransporters and Na+/K+-ATPase, but with astrocytic reactivity in the lesioned and adjacent regions of the nigrostriatal. Moreover, the dopaminergic degeneration caused by 6-OHDA is followed by changes in the expression of connexin-36. Conclusions The use of the GJ blockers directly along the nigrostriatal pathways to control PD motor symptoms is conjectured.
