ABSTRACT: We evaluated the compatibility between two nitrogen–fixing Bradyrhizobium inoculant strains and phosphate–solubilizing fungal strains and the effect of co–inoculation of these bacterial and fungal strains on cowpea growth under different N and P conditions. First, the compatibility between Bradyrhizobium strains UFLA03–84 and INPA03–11B and fungi Haematonectria ipomoeae FSA381, Eleutherascus lectardii FSA257a, Pochonia chlamydosporia var. catenulata FSA109, and Acremonium polychromum FSA115 was tested in both solid and liquid media. Cowpea growth and nodulation promotion under two mineral N doses and two P conditions (a low dose of soluble P plus a high dose of Ca3(PO4)2 and another condition with a high dose of soluble P) were tested with two N2 fixing Bradyrhizobium strains co–inoculated with each of the P–solubilizing fungal strains FSA109, FSA115, and FSA381. There was compatibility between each fungal strain and the two Bradyrhizobium strains, except for FSA257a with either of the bacterial strains in liquid medium. When both mineral N and P were limiting, plants were able to grow and accumulate N and P based on biological N2 fixation and solubilization of calcium phosphate in the same amount as the mineral N and soluble phosphate. Even when both nutrients were fully available, the type of co–inoculation promoted plant growth and nutrient accumulation. The responses varied in accordance with the co–inoculated strains, the N source, and the P source, reflecting the enormous complexity of the biological interactions between plants and microorganisms, and the nutrient conditions provided by the environment.

A queima da saia, causada por Rhizoctonia solani AG 1-IB, é uma importante doença da cultura da alface no Brasil, para a qual o controle biológico com Trichoderma não foi desenvolvido até o momento no país. O presente estudo foi realizado com o objetivo de selecionar isolados de Trichoderma para serem usados para o controle da queima da saia em alface. Quarenta e seis isolados de Trichoderma obtidos com iscas contendo micélio do patógeno foram avaliados em experimentos conduzidos in vitro e in vivo, os quais foram realizados em casa de vegetação, em duas fases.Em laboratório os isolados foram avaliados com relação às capacidades de parasitismo e de produção de substâncias metabólicas tóxicas, com capacidade de inibir o crescimento micelial do patógeno. Na primeira fase dos experimentos in vivo, foram avaliados o número e a massa seca de plântulas de alface da cultivar White Boston. Na segunda fase, 12 isolados eficientes na primeira fase e que também apresentaram rápido crescimento e esporulação abundante em laboratório, foram avaliados com relação à sua capacidade de controlar a queima da saia em dois experimentos repetidos, e também tiveram suas espécies identificadas. A maioria dos isolados de Trichoderma spp. (76%) apresentou elevada capacidade de parasitismo e 50% dos isolados produzim metabólitos tóxicos capazes de inibir 60-100% do crescimento micelial de R. solani AG1-1-B. Vinte e quatro isolados promoveram aumento do número e 23 na massa seca de plântulas de alface inoculadas com o patógeno na primeira fase dos experimentos in vivo. Nos dois experimentos da segunda fase dois isolados de T. virens, IBLF 04 e IBLF 50, reduziram a severidade da queima da saia e promoveram o aumento do número e da massa seca das plântulas de alface inoculadas com R. solani AG 1-IB. Esses isolados haviam apresentado elevada capacidade de parasitismo e de produção de substâncias metabólicas tóxicas, indicando que as fases in vitro e in vivo empregadas no presente estudo foram eficientes para a seleção dos antagonistas a serem utilizados para o controle da queima da saia em alface.

Bottom rot, caused by Rhizoctonia solani AG 1-IB, is an important disease affecting lettuce in Brazil, where its biological control with Trichoderma was not developed yet. The present study was carried out with the aim of selecting Trichoderma isolates to be used in the control of lettuce bottom rot. Forty-six Trichoderma isolates, obtained with baits containing mycelia of the pathogen, were evaluated in experiments carried out in vitro and in vivo in a greenhouse in two steps. In the laboratory, the isolates were evaluated for their capabilities of parasitizing and producing toxic metabolic substances that could inhibit the pathogen mycelial growth. In the first step of the in vivo experiments, the number and the dry weight of lettuce seedlings of the cultivar White Boston were evaluated. In the second step, 12 isolates that were efficient in the first step and showed rapid growth and abundant sporulation in the laboratory were tested for their capability of controlling bottom rot in two repeated experiments, and had their species identified. The majority of the isolates of Trichoderma spp. (76%) showed high capacity for parasitism and 50% of them produced toxic metabolites capable of inhibiting 60-100% of R. solani AG1-IB mycelial growth. Twenty-four isolates increased the number and 23 isolates increased the dry weight of lettuce seedlings inoculated with the pathogen in the first step of the in vivo experiments.In both experiments of the second step, two isolates of T. virens, IBLF 04 and IBLF 50, reduced the severity of bottom rot and increased the number and the dry weight of lettuce seedlings inoculated with R. solani AG1-IB. These isolates had shown a high capacity for parasitism and production of toxic metabolic substances, indicating that the in vitro and in vivo steps employed in the present study were efficient in selecting antagonists to be used for the control of lettuce bottom rot.

Total de 22 fungos endofíticos isolados de café (Coffea arabica L.) foi cultivado in vitro e seus extratos testados. A triagem foi conduzida pelo método de difusão em agar contra bactérias Gram-positiva, Gram-negativa e uma levedura. O isolado mais efetivo foi Alternaria alternata e, subsequentemente, seu extrato foi analisado. O conteúdo de fenólicos totais do extrato bruto foi de 3,44 μg EAG/mg de extrato. Para os testes de atividade antimicrobiana, a concentração inibitória mínima (CIM) e concentração bactericida e fungicida mínima (CBM e CFM) contra Staphylococcus aureus, Escherichia coli e Candida albicans foram determinadas. Resultados da CIM variaram entre 50-100 μg/mL para S. aureus e 400-800 μg/mL para E. coli. O extrato bruto não apresentou atividade nas concentrações testadas para C. albicans. Foram analisadas as atividades antioxidantes do extrato bruto. Sua habilidade para seqüestrar radicais DPPH e a atividade antioxidante pela oxidação do sistema β-caroteno/ácido linoléico não foram significativas. Além disso, a atividade antitumoral foi estudada pelo teste do MTT. À diluição de 400 μg/mL, o extrato apresentou atividade de aproximadamente 50% sobre as células HeLa in vitro. Os resultados indicam que fungos endófitos poderiam ser uma fonte promissora de compostos bioativos necessitando de estudos futuros.

A total of 22 endophytic fungi isolated from coffee (Coffea arabica L.) were cultivated in vitro and their crude extracts tested. The screening was carried out using the agar diffusion method against Staphylococcus aureus, Escherichia coli and Candida albicans. The most effective isolate was Alternaria alternata, and subsequently, its extract was assayed. The total phenolic content was 3.44 μg GAE/mg of the crude extract. For the antibacterial and antifungal activity assays, minimum inhibitory concentrations (MIC) and minimum bactericidal and fungicidal concentrations (MBC and MFC) were determined. The ranges of MIC values were 50-100 μg/mL for S. aureus and 400-800 μg/mL for E. coli. The extract did not show activity in the tested concentrations for C. albicans. The fungal crude extract was assayed for antioxidant activities. Its ability to scavenge DPPH radicals and antioxidant activity by β-carotene/linoleic acid system oxidation was not significant. In addition, antitumor activity was studied using the MTT assay. At a dilution of 400 μg/mL, the extract displayed a cytotoxic activity of approximately 50% towards HeLa cells in vitro. The results indicate that endophytic fungi could be a promising source of bioactive compounds and warrant further study.

A obtenção de protoplastos é uma ferramenta importante para a transformação genética de fungos. Neste trabalho foi estudada a influência de fatores como idade micelial, tipo e concentração de enzimas e estabilizadores osmóticos na produção de protoplastos de Aspergillus ochraceus. Os melhores resultados de produção de protoplastos foram obtidos utilizando-se NH4Cl 0,8mol L-1 como estabilizador osmótico, micélio com 24h de crescimento e a combinação de Lysing Enzymes e Meicelase ambas, a 20mg mL-1. Entretanto, bons resultados foram também obtidos com a utilização apenas de Lysing Enzymes.

Production of protoplasts is an important tool for genetic transformation of fungi. A protocol for protoplasts production in Aspergillus ochraceus was developed, evaluating culture aging of mycelium, different commercial enzymes and osmotic stabilizers. The best results were obtained with NH4Cl 0.8mol L-1 as osmotic stabilizer, mycelial age of 24 hours and Lysing Enzymes (20mg mL-1) plus Meicelase (20mg mL-1) as lytic enzymes. Good results were also obtained with Lysing Enzymes alone.

A presença de fungos em associação natural com frutos do cafeeiro é considerada um fator importante influenciando a qualidade do café. A influência negativa de algumas espécies de Aspergillus é conhecida, comprometendo inclusive a segurança do produto. Os relatos de fungos influenciando positivamente a qualidade se resumem à ocorrência de Cladosporium sp. associados a grãos que originaram cafés de boa qualidade, porém informações exatas sobre a espécie e a sua dinâmica no campo são escassas. Objetivando caracterizar a espécie associada ao cafeeiro e sua dinâmica de colonização, 18 isolados de Cladosporium foram caracterizados e identificados. A dinâmica de colonização do fungo nas comunidades externa e interna do fruto do cafeeiro foi estudada ao longo do período de desenvolvimento do fruto. A espécie associada ao cafeeiro foi identificada como Cladosporium cladosporioides (Fresen.) de Vries. A dinâmica do fungo é característica de um fungo saprófita encontrado em intensidade máxima quando os frutos estão nos estágios de cereja.

The natural occurrence of fungi in coffee fruits is considered an important factor influencing the quality of coffee. The negative effect of some Aspergillus species in coffee, which can also affect safety of the product, is well known. Otherwise, there are reports describing the positive influence of fungi in coffee quality, but they are limited to the occurrence of Cladosporium sp. in fruits, and its correlation with a product of good quality. Indeed, the exact information about the species involved and dynamics of colonization are not available. The objective of this work was to identify and characterize the species of Cladosporium, detected in coffee fruits, and study the dynamics of colonization of the fruits during the maturation process. The species found on coffee fruits was identified as Cladosporium cladosporioides (Fresen.) de Vries. and the dynamics of colonization showed the typical pattern of a saprophytic fungus, with its highest incidence when the fruits were in complete maturation stage.

Situada no sul de Minas Gerais, principal região cafeeira do país, a Clínica Fitossanitária da Universidade Federal de Lavras tem auxiliado os produtores, dando suporte na diagnose de doenças de origem biótica e abiótica, de modo a gerar subsídios para minimizar as perdas na produção. Nos últimos doze anos, foram analisadas 378 amostras de café. Em 78,6% das amostras, foram detectados fungos, destacando-se Colletotrichum sp. (29%), Rhizoctonia solani (18%), Cercospora coffeicola (13%), Phoma sp. (13%) e Fusarium sp (11%). Ocorreram ainda em números representativos casos de fitotoxidez, deficiência de nutrientes e problemas no sistema radicular. Devido à localização da Clínica, a maior parte das amostras foi proveniente de localidades na região sul do Estado (63%), seguido do Triângulo Mineiro (12%) e Zona da Mata (10%).

The Phytossanitary Clinic of the Federal University of Lavras, situated in the south of the State of Minas Gerais has given support to coffee producers providing diagnoses of biotic and abiotic diseases, to generate subsidies to minimize losses in production of this crop. During the last twelve years, 378 samples have been analyzed. Fungi were detected in 78,6% of the samples. The major fungi found were Colletotrichum sp. (29%), Rhizoctonia solani (18%), Cercospora coffeicola (13%), Phoma sp. (13%), e Fusarium sp (11%). Other incidences were caused by phytotoxity, mineral deficiency and bad formation of the roots. Due to the location of the Phytossanitary Clinic, most of the samples came from locations of South of Minas Gerais (63%), followed by Triângulo Mineiro (12%) and Zona da Mata (10%).
