RESUMEN Un método prometedor para el tratamiento de efluentes es el uso de macrófitas flotantes. El nitrógeno amoniacal es un componente típico de lixiviados de vertederos urbanos y su remoción es importante debido a su toxicidad para diversos organismos. Por eso, el estudio evaluó la supervivencia y el potencial de nitrificación de Typha domingensis flotando artificialmente en lixiviados de residuos sólidos domésticos. Las plantas fueron expuestas por 35 días a lixiviado (100, 75 y 50 %) y al agua de lluvia con N:P:K (control). El oxígeno disuelto (OD) fue medido periódicamente; el nitrógeno amoniacal, el nitrito y el nitrato fueron analizados antes y después de la exposición; al final del experimento, la tasa de supervivencia de las plantas fue calculada. Después de dos semanas, el OD aumentó dos veces en el control, tres veces en el lixiviado 50 %, cuatro veces en el lixiviado 75 % y ocho veces en el lixiviado 100 %. Al final del experimento, el nitrógeno amoniacal no fue más detectado, hubo una reducción significativa de nitrito y un aumento significativo de el nitrato en los tratamientos que contenían lixiviado. La supervivencia de las plantas fue mayor en aquellos individuos expuestos al lixiviado 100 % y se redujo en las menores concentraciones: 98 %, 94 %, 92 % y 86 %. El estudio demostró que la concentración de nitrógeno ammoniacal del lixiviado no fue tóxica para T. domingensis y que ésta fue eficiente en la remoción del compuesto del efluente, indicando que la especie puede ser utilizada flotando artificialmente para la remoción del contaminante de lixiviados de residuos sólidos domésticos en bajas concentraciones.

ABSTRACT A promising method for the treatment of effluents is the use of floating macrophytes. Ammoniacal nitrogen is a typical compound present in urban landfill leachates and its removal is important due its toxicity to several organisms. Therefore, the study evaluated Typha domingensis survival and nitrification potential artificially floating in domestic solid waste leachate. Plants were exposed for 35 days to leachate (100, 75 and 50 %) and to rainwater with N:P:K (control). Dissolved oxygen (DO) of the treatments was periodically measured, and ammoniacal nitrogen, nitrite and nitrate were analyzed before and after exposure. At the end of the experiment, plant survival rate was calculated. After two weeks, DO increased twice in the control, three times in 50 % leachate, four times in 75 % leachate, and eight times in 100 % leachate. At the end of the experiment, ammoniacal nitrogen was no longer detected, there was a significant reduction of nitrite, and a significant increase of nitrate in the treatments containing leachate. Plant survival was higher in those individuals exposed to 100 % leachate and decreased at lower leachate concentrations: 98 %, 94 %, 92 %, and 86 %. The study demonstrated that the ammoniacal nitrogen concentration of the leachate was not toxic to T. domingensis, and that it was efficient in the removal of this compound from the effluent, indicating that the species may be used artificially floating for the removal of this contaminant from domestic solid waste landfill leachate when in low concentrations.

Resumo As características das cascas de árvores hospedeiras podem influenciar na distribuição e no desenvolvimento de epífitos. Amostras de casca foram suspensas em 10 e 20 mL de água destilada por 1, 4 e 24 h, para avaliação de métodos para a determinação do pH. Cascas de Myrcia brasiliensis, M. glabra e Myrsine coriacea hospedeiras de Cattleya intermedia foram coletadas no fuste e na copa, para avaliação do pH, da capacidade de retenção de água e da espessura. A altura da parte aérea e o número de raízes de C. intermedia crescidas no fuste e na copa das três espécies de árvore foram avaliados. O volume de solvente e o tempo de suspensão da casca influenciaram nos valores de pH. O pH e a espessura da casca de Myrsine coriacea foram maiores do que nas espécies de Myrcia. A altura da parte aérea e o número de raízes dos indivíduos de C. intermedia foram superiores na copa. Os parâmetros de crescimento das plantas não se relacionaram com o pH e a espessura da casca. Os dados trazem informações para subsequentes estudos sobre epífitos vasculares que incluam novos parâmetros e maiores amostragens, visando a subsidiar propostas de conservação de espécies ameaçadas.

Abstract The traits of the bark of host trees can influence the distribution and development of epiphytes. Bark samples were suspended in 10 and 20 mL of distilled water for 1, 4 and 24 h, to evaluate pH determination methods. Bark samples of Myrcia brasiliensis, M. glabra and Myrsine coriacea, three host trees of Cattleya intermedia had bark samples collected from the trunk and the crown, for the evaluation of the pH, water retention capacity and thickness. Shoot height (cm) and number of roots of C. intermedia grown on trunk and into the crown of the three species were evaluated. Volume of solvent and suspension time of the bark samples influenced pH values. The pH and thickness of Myrsine coriacea barks were higher when compared to Myrcia species. Shoot height and number of roots of C. intermedia individuals were significantly higher in the crown. The growth parameters of the reintroduced plants were unrelated to pH and bark thickness. The data bring information for the application of further vascular epiphyte studies including new parameters and greater samplings, in order to support proposals of the conservation of endangered species.

A qualidade química e a genotoxicidade da água do arroio Luiz Rau em Novo Hamburgo (Rio Grande do Sul, Brasil) e a relação entre os danos genéticos observados em Tradescantia pallida var. purpurea e parâmetros químicos foram analisados. Amostras de água foram coletadas bimestralmente, entre setembro de 2012 e março de 2013 em dois sítios: próximo à nascente (S1) e próximo à foz (S2) do arroio. Ramos com botões florais foram expostos à água dos sítios e à água destilada (controle negativo). Frequências de micronúcleos (MCN) foram determinadas em células-mãe de pólen. Os seguintes parâmetros químicos: pH, sólidos dissolvidos totais, demanda bioquímica de oxigênio (DBO5), oxigênio dissolvido, fósforo total (PT) e os elementos-traço cádmio, chumbo, cobre, cromo total e zinco foram analisados. Em todas as amostragens, as frequências de MCN observadas nos botões florais expostos à água dos dois sítios foram significantemente superiores (S1: 2,48 a 3,38, S2: 3,24 a 5,19) às observadas nos respectivos controles negativos (1,33 a 1,62). O PT esteve acima do limite legal ao longo de todo o período monitorado e a DBO5 apresentou concentrações maiores àquelas estabelecidas pela legislação em dois meses no S1 e em três meses no S2. A análise de componentes principais mostrou relação entre a frequência de MCN, a DBO5 e o PT, indicando a influência negativa de poluentes presentes na água sobre a espécie bioindicadora e reforçando a importância de se considerar os fatores ambientais de forma integrada em programas de monitoramento de corpos hídricos.

This study assessed the chemical quality and genotoxicity of the water of the Luiz Rau Stream in Novo Hamburgo (Rio Grande do Sul, Brazil) and investigated the relationship between the genetic damage observed in Tradescantia pallida var. purpurea and the chemical parameters analyzed. Water samplings were collected bimonthly from September 2012 to March 2013 from two sites, near the headspring (S1) and near the mouth (S2). Cuttings with flower buds were exposed to water from the sites and distilled water (negative control). Micronuclei (MCN) frequencies were determined in pollen mother cells. The chemical parameters analyzed were pH, total dissolved solids, biochemical oxygen demand (DBO5), dissolved oxygen, total phosphorus (TP) and the trace elements cadmium, lead, copper, total chromium and zinc. In all samplings, the MCN frequencies observed in buds exposed to water from both sites were significantly higher (S1: 2.48 to 3.38, S2: 3.24 to 5.19) than those observed in the respective negative controls (1.33 to 1.62). The TP was above the legal limit throughout the monitored period and DBO5 presented concentrations higher than those established by legislation in two months at S1 and three months at S2. The principal component analysis showed a relationship between MCN frequency, DBO5 and TP, pointing to the negative influence of pollutants present in water on the bioindicator species and reinforcing the importance of considering the environmental factors in an integrated way in water-body monitoring programs.

Ramos de Tradescantia pallida var. purpurea contendo botões florais são utilizados em bioensaios para diagnosticar efeitos genotóxicos de água. Diferentes substâncias para adaptar e recuperar os ramos antes e após a exposição a amostras de água e diferentes tempos de exposição são relatados na literatura. O objetivo do estudo foi avaliar e comparar as frequências de micronúcleos (MCN) em T. pallida quando ramos com botões florais foram submetidos a diferentes condições metodológicas. O bioensaio foi então aplicado bimensalmente durante sete meses para avaliar o potencial genotóxico de um sítio do Rio dos Sinos em Campo Bom, Rio Grande do Sul, Brasil. As frequências de MCN em botões de ramos adaptados e recuperados em água destilada e em solução de Hoagland foram de 3,0 e 2,9, respectivamente, quando ramos foram expostos a água do rio e de 1,19 e 1,23 nos controles. Não foram observadas diferenças significantes entre frequências de MCN em ramos expostos por 8, 24 ou 32 h a água do rio (de 3,07 a 4,73) e nos controles (de 1,13 a 2,00) em todas as amostragens ao longo do ano. Adaptação e recuperação de ramos em água destilada ou solução de Hoagland e exposição durante diferentes tempos não influenciaram a resposta de T. pallida de formas diferentes, indicando que todas as condições testadas são viáveis para biomonitoramento de genotoxicidade de água. Amostras de água do Rio dos Sinos apresentaram genotoxicidade ao longo do período monitorado, evidenciada pelas frequências de MCN registradas, que foram significativamente superiores às dos controles.

Tradescantia pallida var. purpurea cuttings with flower buds are utilized in bioassays to diagnose genotoxic effects of water. The literature describes different substances used to adapt and recover the cuttings before and after exposure to water samples and also describes the effects of different exposure times. This study evaluated and compared the micronuclei (MCN) frequencies in T. pallida when cuttings with flower buds were submitted to different methodological conditions. The bioassay was then applied bimonthly during seven months to assess the genotoxic potential of a site located on the Sinos River in Campo Bom, Rio Grande do Sul, Brazil. Micronuclei frequencies in buds of cuttings adapted and recovered in distilled water and in Hoagland solution were 3.0 and 2.9, respectively, for cuttings exposed to river water, and 1.19 and 1.23 in controls. No significant differences among MCN frequencies were observed when cuttings were exposed for 8, 24 or 32 hours to river water (from 3.07 to 4.73) and in controls (from 1.13 to 2.00) in all samplings during a year. Adaptation and recovery of cuttings in distilled water or Hoagland solution and exposure for different times did not influence the response of T. pallida, indicating that all the conditions tested are viable for biomonitoring of water genotoxicity. Water samples from the Sinos River presented genotoxicity during the period monitored, evidenced by the MCN frequencies recorded which were significantly higher than the frequencies of the controls.
