Abstract Capsicum is represented by peppers and sweet peppers and comprises a group with remarkable genetic variability. Different cultivated Capsicum peppers of Brazil were evaluated by using CMA3 and DAPI specific fluorochromes. There was high polymorphism of highly GC-rich CMA heterochromatic bands among the analyzed species, ranging from six (BAGC 114; C. annuum) to 26 blocks (BAGC 81; C. baccatum). Heterochromatin percentage ranged from 3.14% (BAGC 114; C. annuum) to 8.72% (BAGC 81; C. baccatum), corroborating the variation in the number of heterochromatic bands, particularly those distributed in the terminal and subterminal regions of the chromosomes. The information reported in this paper supports the cytogenetic characterization of the domesticated peppers accessions belonging to the Capsicum Germplasm Active Bank of the Federal University of Piauí (BAGC-UFPI). Moreover, the present data helped to better understand the karyotype features of peppers and provide additional information that could contribute to the improvement and maintenance of Capsicum genetic breeding programs.

Abstract We evaluated domesticated Capsicum pepper species from Brazil using classical and molecular cytogenetic techniques, and describe here for the first time their immunostaining patterns using anti-H4K5ac and anti-H3S10ph antibodies. All accessions showed 2n = 24 chromosomes with metacentric and submetacentric morphologies. CMA3 patterns ranged from four terminal bands in most accessions to 18 variable CMA++/DAPI- and CMA+/DAPI0 bands. Anti-H4K5ac signals were detected in the decondensed terminal euchromatin of most chromosome arms, indicating gene-rich open chromatin regions. Anti-H3S10ph signals were restricted to pericentromeric regions and were associated with sister chromatid cohesions and/or the condensation of all mitotic chromosomes. These karyological analyses constitute valuable diagnostic tools for identifying cultivated peppers, will be important to Capsicum breeding programs and will provide additional cytogenetic features that can be used for the conservation of capsicum genetic resources.

Abstract This study evaluated the action at the cellular level of long life whole milk, full type of six renowned companies operating in the Brazilian market, as well as in other South American countries. The evaluation was performed using root meristem cells of Allium cepa L., at exposure times 24 and 48 hours, directly in milk products marketed. The results indicated that all the milk samples reduced root meristem cell proliferation, proving, in this study, a significant cytotoxic effect. Still, exposure to milk resulted in a significant frequency of mitotic spindle changes in meristem cells, characterizing these foods as genotoxic and mutagenic under the study conditions. It can be concluded that the long life milk samples caused significant genetic instability to cells of the examined tissue. The results obtained for cytotoxic, mutagenic and genotoxic action of these long life milks are of great relevance because, to date, there are no published toxicity studies on such foods and food additives present in the composition.

Resumo Objetivos Avaliar o perfil dos trabalhadores rurais do município de Picos, no Estado do Piauí e suas práticas e atitudes quanto à utilização de agrotóxicos. Métodos A coleta dos dados (n=159) foi realizada em Picos com levantamento de dados socioeconômicos, utilização de equipamentos de proteção individual (EPIs), armazenamento e destino das embalagens de agrotóxicos, toxicidade e risco ambiental. Resultados A maioria dos agricultores era do sexo feminino (63%), não concluiu o ensino fundamental (55,6%), tinha entre 31 e 50 anos (55,3%), com renda de até um salário mínimo (66%) e possuía água encanada (84,3%). Quanto ao uso, 64,1% (102) utilizavam agrotóxicos, principalmente na agricultura (86,3%). Eles obtiveram informações sobre essas substâncias com vizinhos (44,2%), liam o rótulo para utilizar o produto (64,8%), usavam agroquímicos devido à ação rápida (81,4%), compravam em casas agropecuárias (87,4%), sem receituário agronômico (92%) e armazenavam embalagens dentro da própria casa (33,6%). Mais da metade não usava EPIs (56,8%), embora considerassem os agrotóxicos prejudiciais à saúde humana (94,1%) e ao meio ambiente (80,4%), além de 15% (24 pessoas) terem sentido algum sintoma de envenenamento. Conclusões Os entrevistados, em sua maioria, usavam agrotóxicos inadequadamente, tinham baixa escolaridade e conheciam sobre os riscos individuais e coletivos aos quais estavam expostos, mas não usavam EPIs.

Abstract Objectives To evaluate rural workers’ profile at Picos (Piauí, Brazil) and their practices and attitudes regarding the use of pesticides. Methods Data collection (n=159) was performed in Picos (Piauí, Brazil) to describe socioeconomic information, use of personal protective equipment (PPE), storage and disposal of pesticide packaging, toxicity and environmental risk. Results Most farmers were women (63%), with incomplete primary education (55.6%), age between 31-55 years-old (55.3%), income of up to a 1 minimum wage (66%) and with access to city water (84.3%). In relation to the use, 64.1% (102) use pesticides, especially in agriculture (86.3%). They receive information about pesticides with neighbors (44.2%), read label to use the product (64.8%), use pesticides due to their quick action (81.4%), purchase in agricultural houses (87.4%) without agronomic prescription (92%) and store packages indoors (33.6%). More than half do not use PPE (56.8%), though they consider pesticides damaging products to the human health (94.1%) and environment (80.4%) and 15% (24 persons) reported some poisoning symptoms. Conclusions The majority of the farmers use pesticides incorrectly, has low educational status and know about the individual and collective risks to which they are exposed but do not use PPE.

ABSTRACT The objective of this study was to evaluate the antiproliferative, cytotoxic and genotoxic potential of salty liquid synthetic flavorings of Butter, Cheddar Cheese and Onion. The antiproliferative potential (2.9-1500 µg/mL) was assessed by MTT assay after 72h using the human tumor lines SF-295 (glioblastoma), OVCAR-8 (ovarian), HCT-116 (colon) and HL-60 (promyelocytic leukemia) and primary cultures of murine Sarcoma 180 (S180) and peripheral blood mononuclear cells (PBMC). Allium cepa bulbs were exposed to growing respective doses (1 mL and 2 mL). Only Butter and Cheddar flavorings revealed cytotoxic activity on cancer cells, with IC50 values ranging from 125.4 µg/mL (Cheddar - HCT-116) to 402.6 µg/mL (Butter - OVCAR-8). Butter flavoring was the most cytotoxic on PBMC (136.3 µg/mL) and increased cell division rate in relation to the mitotic index but did not cause cellular aberrations. Onion and Cheddar flavorings reduced the mitotic index after 24h and 48h exposure, but only Onion flavoring resulted in cellular aberrations and mitotic spindle abnormalities, such as anaphase and telophase bridges, micronucleated cells, conchicine-metaphases and amplifications. So, Butter, Onion and/or Cheddar flavorings caused significant changes in the division of meristematic cells of A. cepa and presented cytotoxic action even on decontrolled proliferating human tumor cells.

Abstract Powdered juices are widely consumed by the population especially because of their convenient preparation, availability in various fruit flavors and low cost when compared to other industrialized beverages. They have complex formulation, consisting of several classes of food additives. However, there are no scientific studies on the toxicity of these foods. Thus, this study evaluated the toxicity at the cellular level of industrialized powdered juices of orange and guava flavors of three different food companies. This analysis was made using root meristem cells of Allium cepa L., at the exposure times of 24 and 48 hours, and two concentrations, 30 g/1000 mL, considered ideal for consumption according to the label of the products, and 30 g/500 mL. Both flavors of juices, of the three companies, in both concentrations and the two exposure times promoted significant antiproliferative effect to root meristem cells and caused a statistically significant number of mitotic spindle changes and micronuclei in cells of the test system used. Therefore, under the studied conditions, all the samples of juice powder exhibited cytotoxic and genotoxic potential.

RESUMOCasearia sylvestris Swartz é uma planta medicinal amplamente distribuída no Brasil. Ela possui atividade anti-inflamatória, antiulcerogênica e antitumoral e é popularmente utilizada para tratar envenenamentos ofídicos, ferimentos, diarreia, gripes e resfriados. Suas folhas são ricas em diterpenos clerodânicos tricíclicos oxigenados, particularmente casearinas. Aqui, nós avaliamos as atividades antioxidantes de um fração com casearinas (FC) isolada de C. sylvestrise as alterações histológicas no sistema nervoso central e fígados de camundongos Mus musculus. Primeiramente, estudos in vitro(0,9, 1,8, 3,6, 5,4 e 7,2 μg/mL) revelaram valores de CE50 de 3,7, 6,4 e 0,16 µg/mL para nitrito, radical hidroxila e níveis de TBARS, respectivamente. Em seguida, a FC (2,5, 5, 10 e 25 mg/kg/dia) foi administrada intraperitonealmente por 7 dias consecutivos em camundongos Swiss. Os níveis de nitrito no hipocampo (26,2, 27,3, 30,2 e 26,6 µM) e no corpo estriado (26,3, 25,4, 34,3 e 27,5 µM) aumentaram em todos os animais tratados com a FC (P < 0.05). As menores doses reduziram os níveis de glutationa reduzida, de catalase e de TBARS no hipocampo e no corpo estriado. Com exceção da redução na formação de TBARS, a FC demonstrou atividade antioxidante apenas in vitro. Os animais exibiram alterações histológicas que sugerem neurotoxicidade e hepatotoxicidade, indicando a necessidade de cuidados em relação ao consumo de preparações medicinais à base de Casearia sylvestris.

ABSTRACTCasearia sylvestris Swartz is a medicinal plant widely distributed in Brazil. It has anti-inflammatory, antiulcer and antitumor activities and is popularly used to treat snakebites, wounds, diarrhea, flu and chest colds. Its leaves are rich in oxygenated tricyclic cis-clerodane diterpenes, particulary casearins. Herein, we evaluated the antioxidant activities of a fraction with casearins (FC) isolated from C. sylvestrisand histological changes on the central nervous system and livers of Mus musculus mice. Firstly, in vitro studies (0.9, 1.8, 3.6, 5.4 and 7.2 μg/mL) revealed EC50 values of 3.7, 6.4 and 0.16 µg/mL for nitrite, hydroxyl radical and TBARS levels, respectively. Secondly, FC (2.5, 5, 10 and 25 mg/kg/day) was intraperitoneally administered to Swiss mice for 7 consecutive days. Nitrite levels in the hippocampus (26.2, 27.3, 30.2 and 26.6 µM) and striatum (26.3, 25.4, 34.3 and 27.5 µM) increased in all treated animals (P < 0.05). Lower doses dropped reduced glutathione, catalase and TBARS levels in the hippocampus and striatum. With the exception of this reduction in TBARS formation, FC displayed only in vitro antioxidant activity. Animals exhibited histological alterations suggestive of neurotoxicity and hepatotoxicity, indicating the need for precaution regarding the consumption of medicinal formulations based on Casearia sylvestris.

Este trabalho teve por objetivo caracterizar numérica e morfologicamente os cariótipos de acessos de Capsicum annum, Capsicum chinense, Capsicum frutencens e Capsicum baccatum pertencentes à coleção do Banco ativo de Germoplasma de Capsicum sp. da Universidade Federal do Piauí (BGC-UFPI). Estas espécies têm grande importância econômica em todo mundo, e a caracterização citogenética fornece informações importantes para estudos de taxonomia bem como para programas de melhoramento genético do gênero na qual estão inseridas. Os cariótipos foram obtidos pelo método de esmagamento e coloração com Giemsa de células dos meristemas das radículas dos acessos estudados. A partir dos resultados verificou-se número cromossômico 2n = 2x = 24 para todas as quatro espécies. Foi observado polimorfismo cromossômico para o acesso BGC 37 da espécie C. frutencens, que apresentou 12 pares de cromossomos metacêntricos, diferindo dos outros que apresentaram 11 pares de cromossomos metacêntricos e 1 par de cromossomo sub-metacêntrico. Visualizou-se nos acessos BGC 01 e BGC 37 desta espécie constrições secundárias nos homólogos 1 e 12, e 6 e 11, respectivamente. Os cariótipos das espécies estudadas foram assimétricos entre si. Os resultados obtidos neste estudo corroboram com a grande diversidade genética descrita na literatura para o gênero Capsicum.

This study aimed to determine the chromosome number and the karyotype of Capsicum annuum, Capsicum chinense, Capsicum frutencens and Capsicum baccatum accessions in the active Capsicum sp. genebank at the Federal University of Piauí (BGC-UFPI). These species have great economic importance throughout the world, and their cytogenetic characterization can inform taxonomy and lead to improvement in the genus. Karyotypes were obtained from the rootlet meristems of the studied accessions using the squash method and Giemsa staining. The chromosome number 2n=2x=24 was verified for each of the four species. Chromosomal polymorphisms were observed for the C. frutencens accession BGC 37, which presented 12 pairs of metacentric chromosomes instead of 11 pairs of metacentric chromosomes and 1 submetacentric chromosome pair present in other accessions. In the accessions BGC 01 and BGC 37, secondary constrictions were observed in the 1 and 12 as well as in the 6 and 11 homologs, respectively. The karyotypes of the studied species were asymmetrical among themselves. The results obtained in this study confirm the high genetic diversity previously described in the literature for this genus.

Os flavonóides são um grupo de substâncias naturais com estrutura fenólica variável, e portanto, de considerável interesse científico e terapêutico. Assim, teve-se por objetivo realizar uma revisão bibliográfica, de forma extensa porém objetiva, sobre as principais ações terapêuticas e a toxicidade das principais classes de flavonóides. A partir desta revisão da literatura, verificou-se, em ensaios pré-clínicos, que estes compostos tem o potencial de modificar a biossíntese de eicosanoides (resposta anti-prostanoide e antiinflamatoria); de proteger o colesterol-LDL da oxidacão (inibindo formação de placa aterosclerotica); de prevenir a agregacão plaquetária tendo efeitos anti-trombóticos; de promover o efeito anti-hipertensivo e anti-isquêmico; de regenerar antioxidantes primários, como a vitamina C, no organismo; de ter efeito antipromocionais na carcinogênese de alguns tipos de câncer; de amenizar os sintomas da menopausa; de aumentar a lipólise; de desacelerar o processo degenerativo em bainhas de mielina; de aumentar a secreção de insulina; de aumentar a expressão de genes responsáveis pela produção de proteínas sinápticas; de estimular os linfócitos B a produzirem anticorpos e de possuir atividade leishmanicida. Quanto a toxicidade dos flavonóides, estudos demostram que em altas doses e em utilização crônica, estes compostos desencadeiam reações alérgicas em humanos; modificam as membranas de hepatócitos causando necrose e morte celular em ratos; causam mutagenicidade e clastogenicidade em medula óssea de camundongos; interferem de forma significativa no funcionamento da tireóide e inibe a ação de citocromos P450 causando alterações drásticas no funcionamento do fígado de roedores. Dessa forma, pode-se verificar que os flavonóides tem uma grande capacidade terapêutica quando bem utilizado, devendo-se sempre levar em consideração doses e tempo de exposição aos organismos aos quais são administrados. Essas considerações são importantes por servirão de base para o estabelecimento de doses seguras e eficazes de utilização dos flavonóides para a população.

Flavonoids are a group of natural substances with variable phenolic structure and therefore, considerable scientific interest and therapeutic. Thus, the objective was to conduct a literature review of but objective extensively on major therapeutic actions and on the toxicity of flavonoids class. In this literature review, it was found that, in preclinical studies, these compounds have the potential to modify the biosynthesis of eicosanoids; protect LDL cholesterol from oxidation; prevent platelet aggregation with antithrombotic effects; promote the antihypertensive and anti-ischemic effect; regenerate primary antioxidants like vitamin C in the body; not to promote some carcinogenic effects on certain types of cancer; alleviate menopausal symptoms; increase lipolysis; slow down degenerative process in myelin sheaths; increase insulin secretion; increase the expression of genes responsible for the production of synaptic proteins; stimulate lymphocytes B to produces antibodies and have leishmanicydal activity. About the toxicity of flavonoids, studies show that, at high doses and in chronic use, these compounds trigger allergic reactions in humans; modify the membranes of hepatocytes, causing necrosis and cell death, in rats; cause mutagenicity and clastogenicity in mouse bone marrow, interfere significantly on thyroid function and inhibit the action of cytochrome P450, causing drastic changes in the function of the liver in rodents. Thus, it can be seen that the flavonoids have a wide therapeutic capacity when properly used, taking into account doses and exposure times to whom they are administered. These considerations are important because they set the bases for the establishment of safe and effective doses of flavonoids on the population.

This study aimed to evaluate the cytotoxic and genotoxic potential of food flavorings (Strawberry, Condensed Milk and Chocolate) on Allium cepa meristematic root cells, with exposure times of 24 and 48 hours. Cytotoxic and mutagenic potential were evaluated separately at doses of 0.2, 0.4 and 0.6 ml and in combination, in which for each dose, the same dose of one other flavoring was combined. The results were analyzed by the Chi-square test (p <0.05). The Strawberry flavor in both exposure times and the three studied doses, the Condensed Milk at 0.6 ml in the 48 hour exposure time, the Chocolate flavor at 0.4 ml, exposure time of 48 hours, and at 0.6 ml, in both exposure times and all treatments with combined doses, significantly reduced the cell division rate, proving to be cytotoxic. No treatment resulted in a significant number of cellular aberrations in A. cepa cells, therefore, the flavorings, under the conditions studied, were non- mutagenic.

O objetivo deste trabalho foi avaliar o potencial ornamental de dez acessos de pimenta Capsicum spp. procedentes da Coleção de Germoplasma da Universidade Federal do Piauí, Brasil. O experimento foi conduzido em casa de vegetação, no Departamento de Fitotecnia, do Centro de Ciências Agrárias, no município de Teresina, Piauí, obedecendo ao delineamento experimental inteiramente casualizado, com três repetições. Na caracterização morfoagronômica dos acessos, conforme orientação do Bioversity International, utilizaram-se os seguintes descritores qualitativos e quantitativos para o gênero Capsicum: forma da folha, hábito de crescimento da planta, antocianina no nó da planta, posição da flor, cor da corola, cor do fruto no estado intermediário e maduro, forma do fruto, forma do ápice do fruto, textura da epiderme do fruto, comprimento do fruto, comprimento do pedicelo, persistência do fruto maduro e altura da planta. Dentre os dez acessos avaliados, sete foram considerados com potencial ornamental: CGC-06 (C. chinense), CGC-07 (C. chinense), CGC-11 (C. annuum var. glabriusculum), CGC-23 (C. chinense), CGC-36 (C. annuum var. glabriusculum), CGC-59 (C. annuum var. glabriusculum) e CGC-67 (C. annuum var. glabriusculum). O CGC 11 apresentou estética mais harmoniosa, com flores eretas, as quais são de fácil percepção; frutos eretos e vistosos e folhas de formato lanceolado.

The objective of this research was to evaluate the potential for ornamental use of ten Capsicum spp. accessions from Germplasm Collection of Federal University of Piauí, Brazil The experiment was carried out in a greenhouse at the Department of Plant Science, Centre of Agricultural Sciences, in Teresina City, Piauí State, according to the completely randomized design, with three replications. In the morphoagronomic characterization of pepper accessions, according to Bioversity International, the following qualitative and quantitative descriptors for the Capsicum genus were used: leaf shape, plant growth habit, anthocyanin node of the plant, flower position, color of the corolla, fruit color and mature in the intermediate state, fruit shape, fruit shape of apex of the texture of the fruit skin , fruit length, length of pedicel, persistence of ripe fruit and plant height. Among the ten accessions, seven were considered with ornamental potential: CGC-06 (C. chinense), CGC-07 (C. chinense), CGC-11 (C. annuum var. glabriusculum), CGC-23 (C. chinense), CGC-36 (C. annuum var. glabriusculum), CGC-59 (C. annuum var. glabriusculum) and CGC-67 (C. annuum var. glabriusculum). The CGC 11 was the more harmonious aesthetic accession, by presenting erect flowers , which are easy to read; erect and showy fruits and lanceolate leaves format.

Este trabalho teve por objetivo avaliar a ação dos aditivos alimentares urucum e cúrcuma em células meristemáticas de pontas de raízes de Allium cepa nas doses de 5 e 10 g, nos tempos de exposição de 24 e 48 horas. Utilizou-se para cada dose um grupo de cinco bulbos de cebolas, que primeiramente foram enraizados em água destilada, e em seguida transferidos para as suas respectivas doses. Analisaram-se células em todo ciclo celular, totalizando 5.000 para cada controle e tempo de exposição. Os dados obtidos foram submetidos à análise estatística do Qui-quadrado (p<0,05). A partir dos resultados observou-se que as doses do urucum e do cúrcuma tiveram efeito antiproliferativo significativo sobre o ciclo celular deste sistema-teste. Também foi verificado número significativo de aberrações celulares nos dois tempos de exposição avaliados de todas as doses estudadas. Portanto, nas condições analisadas, o urucum e o cúrcuma mostraram-se citotóxicos e genotóxicos.

This study aimed to evaluate the effect of food additives annatto and turmeric tip cells of Allium cepa roots in doses of 5 or 10 g in exposure times of 24 and 48 hours. A group of five onion bulbs was used for each dose. Each dose was first embedded in distilled water and then transferred to their respective doses. Cells were analyzed throughout the cell cycle, totaling 5000 for each control and exposure time. The obtained data were subjected to statistical analysis Chi-square (p <.05). From the results it was observed that the doses of turmeric and annatto had significant antiproliferative effect on the cell cycle of this test system. They also found a significant number of cellular aberrations in the two exposure times evaluated all doses studied. Therefore, under the conditions studied, annatto and turmeric proved cytotoxic and genotoxic.

Este estudo teve por objetivo avaliar a ação de extratos aquosos das folhas secas de Phyllanthus niruri L. (popular quebra-pedras) sobre as células meristemáticas de raízes de Allium cepa L. em quatro concentrações, 0,02 (concentração usual), 0,04, 0,06 e 0,08mg/mL, nos tempos de exposição de 24 e 48 horas. Para cada concentração utilizou-se um grupo de cinco bulbos de cebolas, que primeiramente foram enraizados em água destilada, e em seguida transferidos para as suas respectivas concentrações. As radículas foram coletadas e fixadas em ácido acético (3:1) por 24 horas. As lâminas foram preparadas pela técnica de esmagamento e coradas com orceína acética a 2%. Analisaram-se células em todo ciclo celular, totalizando 5.000 para cada controle e tempo de exposição. Os índices mitóticos calculados foram submetidos à análise estatística do Qui-quadrado (p<0,05). A partir dos resultados obtidos observou-se que as quatro concentrações testadas, tiveram efeito antiproliferativo significativo sobre o ciclo celular deste sistema-teste. Também se verificou a presença de aberrações celulares como metáfases colchicínicas, pontes anáfásicas e telofásicas, e micronúcleos nos dois tempos de exposição avaliados de todas as concentrações. Portanto, nas condições analisadas, as concentrações de extratos aquosos de folhas secas de P. niruri mostraram-se citotóxicas e genotóxicas.

This study aimed to evaluate the effects of aqueous extracts of dried Phyllanthus niruri L. (stonebreaker) leaves on Allium cepa L. root meristem cells at four concentrations, 0.02 (usual concentration), 0.04, 0.06 and 0.08mg/mL and exposure times of 24 and 48 hours. For each concentration we used a group of five onion bulbs that were first embedded in distilled water and then transferred to their respective concentrations. The radicles were collected and fixed in acetic acid (3:1) for 24 hours. The slides were prepared by the crushing technique and stained with 2% acetic orcein. Cells were analyzed throughout the cell cycle, totaling 5000 for each control and exposure time. The calculated mitotic indices were subjected to the Chi-squared statistical analysis (p<0.05). From the results obtained it was observed that all four concentrations tested had significant antiproliferative effect on the cell cycle of this test system. We also found the presence of cellular aberrations such as colchicined metaphases, anaphasic and telophasic bridges, and micronuclei in the two exposure times for all concentrations evaluated. Therefore, under the conditions studied the concentrations of aqueous extracts of leaves of P. niruri showed to be cytotoxic and genotoxic.
