Occurrence of Salmonella spp. in fecal samples from foals with and without diarrhea in the state of São Paulo: microbiological diagnosis, antimicrobial susceptibility profile, and molecular detection

ABSTRACT: The present study investigated Salmonella spp. in the feces of 200 foals up to one year of age (100 with clinical signs of diarrhea and 100 without clinical signs of diarrhea). Bacteriological culture, serotyping, antimicrobial susceptibility, and real-time PCR (qPCR SYBR® Green or a TaqMan®) for detecting the invA gene (with and without a selective pre-enrichment step in tetrathionate broth) were performed. Bacterial culture revealed 15% (n=30) of positive animals (21 animals with diarrhea and nine without diarrhea). Among the 30 isolates, 13 different serovars were identified: S. Infantis, S. Minnesota, S. I.4,5,12:i:-; S. Anatum, S. Cerro, S. Oranienburg, S. Braenderup, S. Give, S. Newport, S. IIIb 61:c:z35, S. 109:-:1.5, S. I.4.12:d:-, S. I.6.8:-:-. Multidrug resistance was found in 43.33% (n=13) of the isolates, with one isolate obtained from animals without diarrhea and 12 isolates from animals with diarrhea. All qPCR techniques used in the study classified more samples as positive for Salmonella spp. than the bacterial culture of feces. In addition, all qPCR techniques detected more positive animals in the diarrhea group than in the diarrhea-free group. The results confirm the utility of the qPCR method without the pre-enrichment step in tetrathionate as a rapid test for Salmonella spp. in carrier animals. In animals with clinical signs of diarrhea, it can be combined with bacterial culture (antimicrobial susceptibility testing and serotyping). The isolation of Salmonella spp. in nine animals without diarrhea confirms the importance of asymptomatic carrier animals in the epidemiology of the disease. The multidrug resistance observed highlights the importance of rational antimicrobial use in horses and adopting biosecurity protocols that are efficacious in controlling the spread of infections between animals and zoonotic transmission in farms.
RESUMO: O presente estudo investigou a ocorrência de Salmonella spp. em fezes de 200 potros com até um ano de idade (100 com sinais clínicos de diarreia e 100 sem sinais clínicos de diarreia), utilizando as técnicas de cultivo bacteriológico e PCR em tempo real (qPCR) pelos métodos de corante fluorescente (SYBR® Green) e sonda específica (Taqman®) para a detecção do gene invA com e sem etapa de pré-enriquecimento seletivo em caldo de tetrationato. O cultivo bacteriológico revelou 15% (n=30) de animais positivos (21 animais com diarreia e nove animais sem diarreia). Dentre esses 30 isolados, 13 sorovares diferentes foram identificados: S. Infantis, S. Minnesota, S. I.4,5,12:i:-; S. Anatum, S. Cerro, S. Oranienburg, S. Braenderup, S. Give, S. Newport, S. IIIb 61:c:z35, S. 109:-:1.5, S. I.4.12:d:-, S. I.6.8:-:-. Multirresistência foi constatada em 43,33% (n=13) dos isolados, sendo um isolado obtido de animal sem diarreia e 12 isolados de animais com diarreia. Todas as técnicas de qPCR empregadas no estudo apresentaram maior número de amostras classificadas como positivas para Salmonella spp. comparadas ao cultivo bacteriológico de fezes. Adicionalmente, em todas as técnicas de qPCR houve maior número de animais detectados como positivos no grupo de animais com diarreia em relação aos animais sem diarreia. Os resultados confirmaram a utilidade do método qPCR sem a etapa de pré-enriquecimento em tetrationato, como um teste rápido para detecção de Salmonella spp. em animais portadores ou em animais com sinais clínicos de diarreia. O cultivo bacteriológico deve ser associado para a realização do teste de sensibilidade aos antimicrobianos e sorotipificação. O isolamento de Salmonella spp. em nove animais sem diarreia, confirma a importância dos animais portadores assintomáticos na epidemiologia da doença. A multirresistência observada evidencia a importância do uso racional de antimicrobianos em equinos e a importância da adoção de protocolos de biossegurança que sejam eficazes para controlar a disseminação de infecções entre animais e a transmissão zoonótica nas fazendas.

Stabilization of β-Galactosidase Encapsulated in Pectin-Alginate Hydrogel and Hydrolysis of Whey Lactose and Whole Milk

The operational conditions of β-galactosidase encapsulated in pectin-alginate hydrogel were investigated as well as its catalytic activity in the hydrolysis of lactose from whey and whole milk. β-Galactosidase encapsulated in pectin-alginate hydrogel showed the best results to yields and diffusional effect and pH range. Thermostability showed considerable income gain for half-life (1.7-fold), activation energy of denaturation (31.6-fold), activation of denaturation (217.3-fold at 40 ºC; 266.8-fold at 50 ºC; 345.6-fold at 60 ºC), and entropy of activation of denaturation (-42.0 J mol 1 K-1). Reusability of encapsulated β-galactosidase was observed in 8-cycles for the milk whey lactose hydrolysis (51.9%) and 7-cycles for whole milk (55.6%), and the lactose hydrolysis was observed in 13% in milk whey and 10.3% in whole milk after 10 cycles. These results revealed an industrial application potential of β-galactosidase encapsulated in pectin-alginate hydrogel for lactose hydrolysis of milk whey and whole milk processes.

Abstract Cultivation of species of the genus Eucalyptus is important for the Brazilian economy, with 6.97 million hectares planted. Glycaspis brimblecombei Moore (Hemiptera: Aphalaridae), detected in Brazil in 2003, has dispersed and now damages Eucalyptus crops in all regions of this country. The location and identification of entomopathogenic fungi isolates may increase the options for integrated pest management. The objective of this research was to evaluate the pathogenicity of Cordyceps cateniannulata and Cordyceps javanica isolates to G. brimblecombei. Ten nymphs of G. brimblecombei, with or without lerps, were placed per Eucalyptus leaf cut with one of its edges on hydroretentive gel inside Petri dishes. The fungi isolates were suspended in a solution of Tween 80 (0.1%) at the concentration of 1.0 x 108 conidia mL-1 and sprayed on the G. brimblecombei nymphs. The mortality of this insect was evaluated daily for seven days, and the dead individuals were transferred to humid chambers. The conidia viability of the isolates was greater than 93%. The mortality of G. brimblecombei nymphs, seven days after the application of the fungi, was 100%. This is the first report of the pathogenicity of C. cateniannulata and C. javanica isolates, occurring naturally in the field, to G. brimblecombei.

The population of small towns and riverine communities in Amazonia are extremely vulnerable to COVID-19 due to the absence of basic health care infrastructure in the region. The objective of this study was to (1) evaluate the effect of social distancing measures in the dissemination of COVID-19 in small towns, and (2) to estimate the impact of reducing contact between rural and urban populations on the contamination of riverine communities of Central Amazon. Our results indicate that social distancing can significantly reduce the speed of dissemination of COVID-19 in the population of small towns. However, we also observed that even in towns with intense actions to combat COVID-19, social distancing is below the ideal 70% isolation mark. Regarding the contamination of riverine communities, we observed that the reduction in the number of visits and duration of stay, as well as the interruption of visits during the five weeks with larger probability of contamination has major impact on delaying the arrival of COVID-19 in these vulnerable populations. We recommend that, given the low effectiveness of social distancing measures and the rapid contamination of urban populations, each municipality should evaluate implementing more restrictive measures such as a full or partial lockdown. Our results also suggest that three measures can be effective in delaying the arrival of COVID-19 in riverine communities of Amazonia: (1) the reduction in the number of visits that each community makes to a town, (2) the reduction in duration of each visit, and (3) avoiding visits during the five weeks with the largest number of infected people in towns. It is imperative that implementation of any of the restrictive measures suggested in this publication be accompanied by a vast communication campaign as well as social assistance actions for the rural population and poor families of small towns to guarantee their basic needs for survival.
As populações da Amazônia são extremamente vulneráveis à COVID-19 devido ao intenso fluxo entre as localidades rurais e as sedes municipais e à falta de infraestrutura de saúde básica adequada na região. O objetivo deste estudo foi (1) avaliar o efeito da medida de distanciamento social sobre a disseminação da COVID-19 em sedes municipais e (2) estimar o impacto da redução do contato entre a população rural e urbana sobre a contaminação de localidades rurais da Amazônia Central, região do Médio Solimões. Nossos resultados indicam que o distanciamento social pode reduzir significativamente a velocidade de contaminação da população em sedes municipais. No entanto, mesmo em sedes municipais com atividade intensa no combate à COVID-19, o distanciamento social está muito abaixo do ideal de 70%. Observamos também que a redução do número de visitas e do tempo de permanência durante cada visita, além da interrupção do fluxo durante as cinco semanas com maior probabilidade infecção têm impacto marcante sobre a velocidade de chegada da COVID-19 nas localidades rurais. Recomendamos que, dada a baixa efetividade de medidas de distanciamento social e o rápido contágio nas sedes municipais, cada município avalie a implementação de lockdown. Nossos resultados sugerem também que três medidas podem ser eficientes em atrasar a chegada da COVID-19 às localidades rurais: (1) a redução no número de visitas que cada uma faz a um centro urbano, (2) a redução do tempo de permanência durante cada visita e (3) evitar qualquer ida às sedes municipais durante as cinco semanas com maior número de pessoas infectadas. Destacamos que todas as medidas restritivas sugeridas precisam ser acompanhadas por uma ampla campanha de conscientização e ações de assistência à população rural e às famílias mais carentes das sedes municipais para garantir acesso a itens básicos para sua sobrevivência.

Abstract Abstract: The dispersion capacity is fundamental to establish a biological control program with parasitoids. This information is used to determine the efficiency and the number of release points. Thus, the objective of this work was to determine the dispersion and to estimate the number of release points of Trichogramma pretiosum Riley, 1879 (Hymenoptera: Trichogrammatidae), in sweet corn, cucumber and cabbage in the Ceará State. The experiments were carried out in areas of maize with four leaf pairs (V4) and eight leaves pair (V8), stacked and cabbage. Unviable eggs of an alternative host were distributed in concentric circles of radius 2.5; 5.0; 9.0 and 12.0 m. Mean dispersal distance in the V4 stage maize was 4.7 m with a dispersion area of 48.6 m2, parasitism index of 18.4%, requiring 206 points/ha. In the V8 stage maize, the mean distance was 5.9 m, dispersion area of 60.3 m², mean parasitism index of 22.7% and 166 release points/ha. For the cucumber culture the mean distance was 6.0 m, dispersion area 62.2 m², mean parasitism index of 21.1% and 161 release points/ha. For cabbage the mean distance was 5.6 m, dispersion area of 56.8 m², mean parasitism index of 22.1% and 176 release points/ha.

RESUMO Os efeitos da compactação do solo no crescimento e produtividade das culturas têm sido muito estudados nos últimos anos, porém, as respostas fisiológicas das culturas à compactação não têm recebido a devida atenção. Objetivou-se avaliar o efeito da compactação do solo nas trocas gasosas e na eficiência fotoquímica de genótipos de fava (Phaseolus lunatus L.) de diferentes hábitos de crescimento. O delineamento experimental utilizado foi em blocos casualizados no arranjo fatorial 3 × 4, sendo três genótipos de fava (Branca pequena, Orelha de vó e Roxinha) e quatro níveis de compactação (densidades de solo de 1,1; 1,3; 1,5 e 1,7 g cm-3), com quatro repetições. Aos 38 dias após a semeadura, analisaram-se as seguintes variáveis: taxa fotossintética (A), transpiração foliar (E), condutância estomática (gs), concentração interna de CO2 (Ci), eficiência instantânea no uso da água (EUA), eficiência intrínseca no uso da água (EIUA), eficiência instantânea de carboxilação (EiC) e eficiência fotoquímica (Fo, Fm, Fv e Fv/Fm). Os dados foram submetidos à análise de variância a 5% de probabilidade pelo teste F. Os genótipos apresentaram redução na taxa fotossintética com aumento da compactação do solo. Houve efeito da compactação do solo na eficiência fotoquímica apenas para o genótipo ‘Orelha de vó’, com ajustes do modelo linear para Fm e Fv, e quadrático para relação Fv/Fm. O genótipo ‘Orelha de vó’ atingiu a maior taxa de E e gs nas densidades de solo 1,24 e 1,29 g cm-3, respectivamente. Em relação a taxa fotossintética, o genótipo ‘Roxinha’ é mais eficiente que o genótipo ‘Branca pequena’ na densidade do solo de 1,7 g cm-3.
ABSTRACT The effects of soil compaction on crop growth and productivity have been well studied in recent years, however, studies on the physiological responses of crops to compaction are scarce. The objective of this study was to evaluate the effect of soil compaction on gas exchange, and photochemical efficiency of lima bean (Phaseolus lunatus L.) genotypes of different growth habits. The experimental design was a randomized block in a 3×4 factorial arrangement, with three lima bean genotypes (Branca-Pequena, Orelha-de-Vó and Roxinha) and four compaction levels (soil densities of 1.1, 1.3, 1.5 and 1.7 g cm-3), with four replications. The following variables were evaluated at 38 days after sowing: photosynthetic rate (A), leaf transpiration (E), stomatal conductance (gs), internal CO2 concentration (Ci), instantaneous water use efficiency (WUE), intrinsic water use efficiency (iWUE), instantaneous carboxylation efficiency (iCE) and photochemical efficiency (Fo, Fm, Fv and Fv/Fm). The data were subjected to analysis of variance at 5% probability by the F test. The genotypes showed a reduction in the photosynthetic rate with increasing soil compaction. The soil compaction affected the photochemical efficiency of the genotype Orelha-de-Vó, with the Fm and Fv fitting to the linear model, and the Fv/Fm fitting to the quadratic model. The genotype Orelha-de-Vó had the highest rate of E and gs at the soil densities of 1.24 and 1.29 g cm-3, respectively. Regarding the photosynthetic rate, the genotype Roxinha is more efficient than Branca-Pequena at the soil density of 1.7 g cm -3.

The inhibitory ability of an oxazinoquinoxaline derivative (OAQX) against the corrosion of AISI 1018 mild steel induced by aqueous 0.6 mol L-1 NaCl solution is herein evaluated. Linear polarization resistance studies showed the inhibitory efficiency of OAQX varying from 62.75% to 75.93% with OAQX concentration ranged from 0.259 x 10-4 mol L-1 to 3.243 x 10-4 mol L-1. Aiming at to investigate the Tafel curves on the OAQX electrochemical behavior at saline (0.6 mol L-1 NaCl) aqueous media the linear Tafel segments of anodic and cathodic curves were extrapolated to the intersection point which afforded corrosion potential (Ecorr) and corrosion current density (icorr) data. By applying Tafel approach the efficiency of OAQX ranges from 55.30% to 87.60%. In both analysis Langmuir isotherm lead to optimized adsorption parameter. The adsorption mechanism of OAQX is proposed from FTIR experiments and quantum calculations. The theoretical employed method is hybrid B3LYP combined with 6-311++G(d,p). Theoretical results showed OAQX as a promising inhibitor that form a stable protective metal-ligand film on metal surfaces, and differs from several heterocyclic compounds due to its solubility in small amount of DMSO (0.627 x 10-4 mol L-1) which is resistant to a saline aqueous media (0.6 mol L-1 NaCl).

Resumo Introdução: o câncer de mama é a segunda causa de morte no mundo, sendo 90% dessas mortes decorrentes de metástases. A metaloprotease de matriz 2 (MMP-2) possui atividade de gelatinase capaz de degradar o principal constituinte do microambiente tumoral, o colágeno do tipo IV. Há duas proteínas bem estabelecidas utilizadas como marcadores na prática clínica para o câncer de mama, os receptores de estrógeno (RE) e de progesterona (RP). Embora a presença desses receptores tenha sido associada a um melhor prognóstico, a perda delas pode ocorrer durante a progressão do tumor, com subsequente resistência à terapia hormonal. Objetivo: analisar a correlação entre as proteínas MMP-2, RE e RP por imuno-histoquímica e estabelecer o processo metastático em tumores de mama primários. Método: amostras de tumor de mama (n=44) foram analisadas por imuno-histoquímica para MMP-2, receptor de estrógeno e progesterona. Resultados: observou-se que 90% das pacientes que tinham metástases e morreram apresentaram coloração positiva para a MMP-2 (p=0,0082 para ambos). Usando a análise de Kaplan-Meier, verificou-se que as pacientes RE negativas, também positivas para MMP-2, apresentaram sobrevida livre de doença (SLD) e sobrevida global (SG) (p=0,012 e p=0,005, respectivamente) piores quando comparadas às pacientes MMP-2 negativas. Resultados semelhantes foram encontrados em pacientes RP negativas para SLD (p=0,077) e SG (p=0,038). Conclusão: embora o número de amostras avaliadas tenha sido baixo (n=44), esses dados iniciais permitem inferir que a MMP-2 em combinação com os marcadores já bem estabelecidos poderia ajudar na previsão do surgimento de metástase e morte em pacientes com câncer de mama.
Summary Introduction: Breast cancer is the most cause of death, and approximately 90% of these deaths are due to metastases. Matrix metalloproteinase-2 (MMP-2) gelatinase activity is able to degrade a major constituent of the tumor microenvironment, type IV collagen. Two well-established proteins used as markers in clinical practice for breast cancer are the receptors for estrogen (ER) and progesterone (PR). Although the presence of these receptors has been associated with a better prognosis, loss of these proteins can occur during tumor progression, with subsequent resistance to hormone therapy. Objective: To study the correlation among MMP-2, ER, and PR, as well as the establishment of the metastatic process in primary breast tumors. Method: Breast cancer samples (n=44) were analyzed by immunohistochemistry for MMP-2, ER, and PR. Results: We observed that 90% of patients who had metastases and died showed positive staining for MMP-2 (p=0.0082 for both). Using Kaplan-Meier analysis, we found that negative ER patients who were also positive for MMP-2 had even worse disease-free survival (DFS) and overall survival (OS) (p= 0.012 and p=0.005, respectively). Similar results were found in PR-negative patients for DFS (a trend p=0.077) and OS (p=0.038). Conclusion: Regardless of our small sample size (n=44), the data obtained strongly suggest that MMP-2 in combination with already well-established markers could help to predict the emergence of metastases and death in patients with breast cancer.

RESUMO O objetivo deste trabalho foi avaliar o efeito da aplicação de silicato de cálcio e de cinza de casca de arroz (CCA) na incidência de fungos associados a manchas em sementes de arroz irrigado. Plantas de arroz foram submetidas à aplicação de silicato de cálcio ou CCA nas doses de 0, 51, 153, 256 e 357 kg ha-1de silício (Si). Dois experimentos foram conduzidos, sendo um na safra 2007/2008 e outro na safra 2008/2009 e, posteriormente, amostras de sementes foram analisadas em laboratório. Foram realizadas avaliações do Índice de Escurecimento de Sementes (IES), da concentração de Si no pericarpo das sementes e a determinação da diversidade dos fungos presentes nas sementes. Não houve efeito das duas fontes de Si empregadas, nas doses utilizadas nos dois experimentos no IES e na concentração de Si. Os fungos fitopatogênicos encontrados em ambos experimentos foram Alternaria padwickiii, Bipolarisoryzae, Botrytis cinerea, Curvularia lunata, Fusarium semitectum, F. solani, Microdochium oryzae, Nigrospora oryzae, Phoma sorghina e Pyriculariaoryzae. A incidência destes fungos não foi afetada pela aplicação das fontes de Si nas doses utilizadas.
ABSTRACT The aim of this study was to evaluate the effect of calcium silicate and rice husk ash (CCA) application on the incidence of fungi associated with spots in flooded rice seeds. Rice plants were subjected to application of calcium silicate or CCA at the levels of 0, 51, 153, 256 and 357 kg ha-1 of silicon (Si). Two experiments were performed, one in the 2007/2008 crop season and another one in the 2008/2009 season; subsequently, seed samples were analyzed in laboratory. Evaluations were performed for Seed Browning Index (IES), Si concentration in the pericarp of the seeds and determination of the diversity of seedborne fungi. There was no effect of the two employed Si sources at the used levels in both experiments, for IES and Si concentration. The phytopathogenic fungi found in both experiments were Alternaria padwickiii, Bipolaris oryzae, Botrytis cinerea, Curvularia lunata, Fusarium semitectum, F. solani, Microdochium oryzae, Nigrospora oryzae, Phoma sorghina and Pyricularia oryzae. The incidence of these fungi was not affected by the application of Si sources at the used levels.

RESUMO Plutella xylostella L. é um dos principais agentes de danos as brássicas causando impactos negativos às culturas. A utilização de extratos botânicos é relatada na literatura para proteção de plantas, no entanto, para as espécies analisadas no presente trabalho ainda não foi relatado. Examinamos o efeito de extratos aquosos e metanólicos de quatro espécies de plantas: Schinus terebinthifolius Raddi (pimenta-rosa), Annona coriacea Mart. (araticum), Duguetia furfuracea (A. ST.-Hil.) Benth. & Hook. (pindaúva-do-campo) e Trichilia silvatica C. DC. (catiguá-branco) de ocorrência no estado de Mato Grosso do Sul, e que, até o momento não foram avaliadas quanto à preferência alimentar por larvas de 3º ínstar de P. xylostella. Pretendemos responder as seguintes questões: (1) as espécies vegetais analisadas são fagodeterrentes? (2) qual o tipo de extrato que produz a menor preferência alimentar? Para responder a essas questões discos de couve foram tratados com extratos aquosos armazenados em refrigerador nos períodos de 0, 7, 14 e 21 dias e os extratos metanólicos nas concentrações de 0,5 mg/mL, 1,0 mg/mL, 2,0 mg/mL. Os extratos aquoso e metanólico de T. silvatica apresentaram os menores valores de preferência alimentar 0,113 e 0,06, respectivamente, em relação aos outros extratos.
ABSTRACT Plutella xylostella L. is one of the main agents to cause damages to plants of Brassica genus, provoking negative impacts in cultures. The use of botanical extracts in plants protection has been related in literature, however, their use in the species analyzed in this study is not yet reported. We assessed the effect of aqueous and methanolic extracts of the species: Schinus terebinthifolius Raddi (Pink Pepper), Annona coriacea Mart. (Araticum), Duguetia furfuracea (A. St.-Hil.) Benth. & Hook. (Pindaúva do campo) and Trichilia silvatica C. DC. (Catiguá-branco), occuring in the state of Mato Grosso do Sul and whose feeding preference of P. xylostella larvae of 3rd instar. We intend to answer the following questions: (1) Are the plant species analyzed fagodeterrentes? (2) what type of extract produces the least food preferrence? To answer these questions, we treated cabbage disks with aqueous extracts stored in a refrigerator in periods of 0, 7, 14 and 21 days and the methanolic extracts were treated at concentrations of 0.5 mg/mL, 1.0 mg/mL, 2.0mg/mL. The aqueous and methanolic extracts of T. silvatica presented the lowest values of feeding preference, 0.113 and 0.06, respectively, compared to other extracts.

RESUMO A contaminação por chumbo (Pb) é uma preocupação crescente em todo o mundo uma vez que os ambientes aquáticos são sujeitos sobretudo a inúmeras fontes de poluição. Este trabalho teve, como objetivo, avaliar a tolerância de E. grandiflorus ao Pb e sua capacidade de acumular e alocar o poluente e a relação dessas características com a anatomia radicular. Foram montados experimentos com plantas de E. grandiflorus expostas às seguintes concentrações de Pb em solução nutritiva: [0; 0,75; 1,5; 3,0 e 9,0 μM de Pb(NO3)2]. As plantas foram mantidas nessas condições, durante 60 dias. Avaliaram-se, ao final do experimento, a concentração de chumbo na biomassa das plantas e as modificações na anatomia radicular. As plantas de E. grandiflorus acumularam 113,74 mg Pb kg-1 de matéria seca com translocação reduzida para a parte aérea. As modificações no xilema radicular demonstraram aumento na capacidade de condução de água e nutrientes nas plantas expostas ao Pb. Constatou-se espessamento da exoderme e do córtex radicular; é provável que essas modificações estejam relacionadas com a maior alocação de Pb nas raízes. As plantas de Echinodorus grandiflorus são capazes de absorver e acumular Pb principalmente nas raízes; esta absorção é, por outro lado, modulada pela anatomia radicular.
ABSTRACT Lead (Pb) contamination is a growing concern worldwide and water environment is particularly subjected to many sources of pollution. This study aimed to evaluate the tolerance of Pb in Echinodorus grandiflorus and its capacity to absorb and accumulate this pollutant, as well as the relationship of these characteristics with root anatomy. Experiments were conducted using E. grandiflorus plants exposed to the following Pb levels in nutrient solution: [0, 0.75, 1.5, 3.0, and 9.0 μM of Pb(NO3)2]. Plants were maintained in these conditions for 60 days. At the end of the experiment Pb levels in plant biomass as well as the modifications in the root anatomy were evaluated. E. grandiflorus accumulated 113.74 mg Pb kg-1 of dry matter and reduced translocation to shoots. Xylem modifications in roots showed increased water and nutrient conduction capacity in plants exposed to Pb. Exodermis and root cortex thickening were detected and may be related to lead allocation in roots. Echinodorus grandiflorus plants are able to absorb and accumulate Pb mainly in roots and this absorption is modulated by root anatomy.

Considerando a importância econômica de Crambe abyssinica, o presente estudo é o primeiro relato das abelhas ocorrentes na espécie representando contribuição para a Ciência, pois permite adequar métodos alternativos de controle para outros insetos daninhos com menor impacto aos polinizadores. O objetivo do presente trabalho foi responder as seguintes questões: (1) qual é a espécie polinizadora potencial em C. abyssinica? (2) como as condições ambientais interferem na flutuação populacional dos polinizadores? Os insetos foram amostrados semanalmente, com rede entomológica, das 8 às 16 horas durante cinco semanas de florescimento. O delineamento experimental foi inteiramente casualizado em esquema fatorial 5 x 5 (horário x semanas de florescimento). Havendo significância na análise de variância, os dados foram ajustados por equações de regressão à 5% de probabilidade e para as variáveis ambientais por correlação de Pearson. Foram observados Apis mellifera, Geotrigona mombuca, Exomalopsis (Exomalopsis) fulvofasciata, Plebeia sp. e Dialictus sp. Observou-se que o polinizador potencial é A. melliferaapresentando pico máximo de visitas na II e IV semana de florescimento as 12:00 e 8:00 horas respectivamente e G. mombucanas semanas III e IV as 12:00 e 10:00 horas respectivamente. Os fatores ambientais como temperatura, umidade relativa e velocidade do vento interferem de forma diferenciada na atividade das abelhas.
Despite the economic importance of Crambe abyssinica, the present study is the first report on bees that occur with this species, and could aid in developing alternative methods for controlling insect pests without seriously impacting pollinators. The present study examined the following questions: (1) Which species are potential pollinators of C. abyssinica? (2) How do environmental conditions influence pollinator visitation fluctuations? Insects were sampled on a weekly basis between 08:00 and 16:00 during five weeks of flowering. When the results of analyses of variance were significant, the data was adjusted using regression equations at a 5% level of probability; the environmental variables were examined using Pearson's correlation coefficient. Apis mellifera, Geotrigona mombuca, Exomalopsis (Exomalopsis) fulvofasciata, Plebeia sp. and Dialictus sp. were observed visiting C. abyssinica flowers. A. melliferaand G. mombucawere observed to be potential pollinators, with the former demonstrating visitation peaks during flowering weeks II and IV at 12:00 and 08:00, respectively and the latter visiting during weeks III and IV at 12:00 and 10:00, respectively. Environmental factors such as temperature, relative humidity, and wind speed have different effect on the activity of bees.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Lead tolerance of water hyacinth (Eichhornia crassipes Mart. - Pontederiaceae) as defined by anatomical and physiological traits

Este estudo teve como objetivo verificar a tolerância do aguapé ao chumbo e verificar as modificações anatômicas e fisiológicas decorrentes. As plantas de aguapé foram cultivadas em solução nutritiva com cinco diferentes concentrações de chumbo sendo: 0,00; 0,50; 1,00; 2,00 e 4,00 mg L–1 por um período de 20 dias. Foram avaliadas a fotossíntese, transpiração, condutância estomática e a razão Ci/Ca aos 15 dias decorridos do início do experimento. Ao final do experimento, as modificações anatômicas nas raízes e folhas além da atividade das enzimas do sistema antioxidante foi avaliada. A fotossíntese e a razão Ci/Ca aumentaram em todos os tratamentos com chumbo. A análise da anatomia foliar não demonstrou nenhuma evidência de efeitos tóxicos mas demonstrou modificações nos estômatos e na espessura do parênquima esponjoso na presença de chumbo. De forma semelhante, a anatomia das raízes não demonstrou nenhum efeito tóxico mas o xilema e floema, bem como as barreiras apoplásticas demonstraram modificações favoráveis. Todas as enzimas do sistema antioxidante demonstraram aumento na atividade nas folhas e algumas modificações nas raízes em função da presença de chumbo. Portanto, a tolerância ao chumbo do aguapé está relacionada com modificações anatômicas e fisiológicas como o aumento da fotossíntese e modificações anatômica que aumentam a capacidade de captação de CO2 e a condutividade hidráulica.
This study aimed at verifying the lead tolerance of water hyacinth and at looking at consequent anatomical and physiological modifications. Water hyacinth plants were grown on nutrient solutions with five different lead concentrations: 0.00, 0.50, 1.00, 2.00 and 4.00 mg L–1 by 20 days. Photosynthesis, transpiration, stomatal conductance and the Ci/Ca rate were measured at the end of 15 days of experiment. At the end of the experiment, the anatomical modifications in the roots and leaves, and the activity of antioxidant system enzymes, were evaluated. Photosynthetic and Ci/Ca rates were both increased under all lead treatments. Leaf anatomy did not exhibit any evidence of toxicity effects, but showed modifications of the stomata and in the thickness of the palisade and spongy parenchyma in the presence of lead. Likewise, root anatomy did not exhibit any toxicity effects, but the xylem and phloem exhibited favorable modifications as well as increased apoplastic barriers. All antioxidant system enzymes exhibited increased activity in the leaves, and some modifications in roots, in the presence of lead. It is likely, therefore, that water hyacinth tolerance to lead is related to anatomical and physiological modifications such as increased photosynthesis and enhanced anatomical capacity for CO2 assimilation and water conductance.

Infection process of Fusarium oxysporum f. sp. phaseoli on resistant, intermediate and susceptible bean cultivars

The objective of this study was to understand the infection process of Fusarium oxysporum f. sp. phaseoli (Fop) in bean cultivars classified as resistant (Manteigão Fosco 11), intermediate (VP8) and susceptible (Meia Noite). Plants of the three cultivars were inoculated at 10 days after emergence with a suspension of 1×10(6) conidia of Fop per mL. At 43 days after the inoculation, stem segments were observed with a scanner electronic microscope. The cultivars Manteigão Fosco 11 and VP8 presented an occluding material in the xylem vessels, which may have restricted tissue colonization by Fop. The resistance of bean cultivars to Fop seemed also to be explained by structural differences in the xylem tissue.
