Benzophenone Derivatives Showed Dual Anti-Inflammatory and Antiproliferative Activities by Inhibiting COX Enzymes and Promote Cyclin E Downregulation

Considering the promising antitumor effects of compounds with dual anti-inflammatory and antiproliferative activities, thus benzophenones analogs (2-7) were evaluated on in vivo anti-inflammatory assay and molecular docking analysis. Those with the best molecular docking results were in vitro evaluated on cyclooxygenase (COX) enzymes and tested regarding antiproliferative activity. All derivatives displayed in vivo anti-inflammatory activity. Among them, the substances 2’-hydroxy-4’-benzoylphenyl-β-D-glucopyranoside (4), 4-hydroxy-4’-methoxybenzophenone (5) and 4’-(4’’-methoxybenzoyl)phenyl-β-D-glucopyranoside (7) showed the best values of Glide Score in COX-2 docking evaluation and 4 and 5 selectively inhibited COX-2 and COX-1 in vitro enzymatic assay, respectively. Thus, 4 and 5 were tested against breast cancer (MCF-7, MDA MB-231, Hs578T) and non-small-cell-lung cancer (A549) cell lines. The estrogen-positive MCF-7 cell line was more responsive compared to other tested cell lines. They induced cell cycle arrest at G1/S transition in MCF-7 cell line once there was an increase in G0/G1 population with concomitant reduction of S population. The antiproliferative activity of these substances on MCF-7 was associated with their ability to inhibit cyclin E expression, a critical regulator of G1/S transition. Taken together, the data indicate that 4 and 5 have dual anti-inflammatory and antiproliferative activities and support further studies to evaluate their antitumor potential.

OBJECTIVES: To prospectively evaluate demographic, anthropometric and health-related quality of life (HRQoL) in pediatric patients with laboratory-confirmed coronavirus disease 2019 (COVID-19) METHODS: This was a longitudinal observational study of surviving pediatric post-COVID-19 patients (n=53) and pediatric subjects without laboratory-confirmed COVID-19 included as controls (n=52) was performed. RESULTS: The median duration between COVID-19 diagnosis (n=53) and follow-up was 4.4 months (0.8-10.7). Twenty-three of 53 (43%) patients reported at least one persistent symptom at the longitudinal follow-up visit and 12/53 (23%) had long COVID-19, with at least one symptom lasting for >12 weeks. The most frequently reported symptoms at the longitudinal follow-up visit were headache (19%), severe recurrent headache (9%), tiredness (9%), dyspnea (8%), and concentration difficulty (4%). At the longitudinal follow-up visit, the frequencies of anemia (11% versus 0%, p=0.030), lymphopenia (42% versus 18%, p=0.020), C-reactive protein level of >30 mg/L (35% versus 0%, p=0.0001), and D-dimer level of >1000 ng/mL (43% versus 6%, p=0.0004) significantly reduced compared with baseline values. Chest X-ray abnormalities (11% versus 2%, p=0.178) and cardiac alterations on echocardiogram (33% versus 22%, p=0.462) were similar at both visits. Comparison of characteristic data between patients with COVID-19 at the longitudinal follow-up visit and controls showed similar age (p=0.962), proportion of male sex (p=0.907), ethnicity (p=0.566), family minimum monthly wage (p=0.664), body mass index (p=0.601), and pediatric pre-existing chronic conditions (p=1.000). The Pediatric Quality of Live Inventory 4.0 scores, median physical score (69 [0-100] versus 81 [34-100], p=0.012), and school score (60 [15-100] versus 70 [15-95], p=0.028) were significantly lower in pediatric patients with COVID-19 at the longitudinal follow-up visit than in controls. CONCLUSIONS: Pediatric patients with COVID-19 showed a longitudinal impact on HRQoL parameters, particularly in physical/school domains, reinforcing the need for a prospective multidisciplinary approach for these patients. These data highlight the importance of closer monitoring of children and adolescents by the clinical team after COVID-19.

Resumo Apresenta-se uma síntese atualizada de informações sobre algas no Brasil objetivando refinar os dados reunidos até o presente, bem como avaliar os avanços sobre o conhecimento da diversidade de algas no país desde a publicação do Catálogo de Plantas e Fungos do Brasil. Os resultados de 2015 mostraram um acréscimo de 1.250 espécies (35.7%) a um total de 4.747 em relação a 2010. As classes mais diversas em número de espécies foram Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae e Euglenophyceae. Bacillariophyceae e Cyanophyceae tiveram o maior acréscimo de espécies no intervalo de cinco anos. A região Sudeste e Sul foram as mais diversas, porém, as regiões Nordeste com os estados do Piauí e Sergipe e Centro-Oeste com os estados de Mato Grosso, Goiás e Distrito Federal destacaram-se no cenário da biodiversidade nacional. Apesar da escassez de taxonomistas e da infraestrutura limitada, os resultados obtidos evidenciaram um avanço significativo no conhecimento da diversidade de algas no país nesse período de cinco anos, iniciando uma mudança quanto as diferenças geográficas regionais.
Abstract An updated synthesis of cyanobacteria and algae information is presented for Brazil aiming to refine the data gathered to date and evaluate the progress of the biodiversity knowledge about these organisms in the country since the publication of the Catálogo de Plantas e Fungos do Brasil. The results of 2015 showed an increase of 1,250 species (35.7%) when compared to 2010, reaching a total of 4,747 species. The most diverse classes in species number were the Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae and Euglenophyceae. Bacillariophyceae and Cyanophyceae had the highest increase in species number in the five-year interval. The Southeast and South regions were the most diverse, however, the Northeast, with the states of Piauí and Sergipe, and the Central-west region, with Mato Grosso, Goiás and Distrito Federal, also stood out in the national algal biodiversity scenario. Despite the shortage of taxonomists and limited infrastructure, the results showed a significant improvement in the knowledge regarding the diversity of cyanobacteria and algae in the country during the study period, starting to even out regional geographical differences caused by subsampling.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Estenose subglótica como manifestação clínica da granulomatose de Wegener em adolescentes: relato de caso e revisão de literatura

A Granulomatose de Wegener é uma patologia caracterizada por vasculite sistêmica e inflamação granulomatosa necrotizante que compromete o trato respiratório alto, pulmões e rins, cujas manifestações iniciais, na maioria das vezes, ocorre em maiores de 20 anos de idade. As queixas clínicas otorrinolaringológicas estão, com freqüência, presentes nas fases iniciais da doença, sendo rinite, sinusopatia de repetição e epistaxe as mais comuns. A estenose subglógica parece estar correlacionada com a Granulomatose de Wegener em adolescentes. Relatamos o caso de uma paciente portadora de Granulomatose de Wegener cujo início dos sintomas foi na infância, tendo evoluído com estenose laríngea durante o curso da doença.
Wegener's Granulomatosis is a well-characterized systemic vasculitis and necrotising granulomatous inflammation of the upper, lower respiratory tracts and the kidneys. The initial manifestations of the disease usually occur in patients over than twenty years old. The otolaryngologic symptoms like rhinorrhea, recurrent sinusitis and epistaxis are commonly present in early course of the disease. It seems that subglottic stenosis is correlated to Wegener's granumatosis in adolescents. We describe a case of a patient that developed subglottic stenosis associated with Wegener's granulomatosis in childhood during the course of the disease.

Fusobacterium nucleatum é um anaeróbio estrito e considerado membro da microbiota indígena da cavidade bucal humana. É comumente observado em diferentes infecções monomicrobianas e mistas em humanos e animais. O objetivo deste estudo foi avaliar o perfil plasmidial de F. nucleatum orais isolados de pacientes com doença periodontal, indivíduos sadios, e de macacos Cebus apella, e verificar a estabilidade plasmidial e a associação com genes de resistência à penicilina. Quarenta e cinco cepas de F. nucleatum orais isoladas de pacientes, 38 de indivíduos sadios e sete de macacos C. apella, foram examinadas. A extração plasmidial realizada detectou plasmídios em 26,7% das cepas humanas e em apenas uma cepa de C. apella. A maioria das cepas apresentou duas bandas plasmidiais variando de 4 a 16 Kb, e as digestões com endonucleases demonstraram serem essas bandas pertencentes a um único plasmídio, sendo classificados em três grupos segundo o tamanho. Cepas de indivíduos sadios não abrigaram plasmídios. O perfil plasmidial foi similar e estável nas cepas isoladas de humanos e de macacos. Duas cepas foram beta-lactamase-positivas, e não se observou hibridização de DNA plasmidial com a sonda do gene da beta-lactamase, sugerindo-se que a resistência à penicilina nestas cepas é cromossômica.
Fusobacterium nucleatum is a strict anaerobe and is indigenous of the human oral cavity. This organism is commonly recovered from different monomicrobial and mixed infections in humans and animals. In this study, the plasmid profile, the plasmid stability and the penicillin-resistance association in oral F. nucleatum isolated from periodontal patients, healthy subjects and Cebus apella monkeys were evaluated. Forty-five F. nucleatum strains from patients, 38 from healthy subjects and seven from C. apella were identified and analyzed. Plasmid extraction was performed in all the isolated strains. These elements were found in 26.7% strains from patients and one strain from C. apella. Strains from healthy subjects did not show any plasmid. Most of strains showed two plasmid bands ranging from 4 to 16 Kb, but digestions with endonucleases showed that they belonged to a single plasmid. The plasmid profile was similar and stable in human and monkey strains. Also, plasmids were classified into three groups according to size. Two strains were positive to beta-lactamase production and no plasmid DNA-hybridization with a beta-lactamase gene probe was observed, suggesting a chromosomal resistance.

A administração de insulina geralmente é realizada com seringa e agulha. Porém, a injeção é freqüentemente considerada dolorosa e traumática, causando problemas psicossociais em muitos pacientes com diabetes. Reconhece-se que as canetas de injeção de insulina são mais convenientes, causam menos dor à injeção e melhoram a qualidade de vida dos pacientes. Este estudo examinou a aceitabilidade e funcionalidade de uma nova caneta de administração de insulina reutilizável, Humapen®, em 94 pacientes diabéticos tipo 1 e 2, os quais usavam insulina por um tempo mediano de 7,8 anos com 2,7+3,0 aplicações por dia. Quarenta e dois porcento deles usava outras canetas, e os 58% restantes usava seringas. Os pacientes fizeram uso de Humapen® por 8 semanas, mantendo o esquema de insulinização prévio. Todos os pacientes completaram o estudo. Os resultados em pacientes em uso prévio de seringa ou canetas foram semelhantes. As características mais mencionadas como superiores em relação ao método de administração anterior foram a facilidade de ajustar/usar (89%), medição/correção/leitura da dose (88%) e troca do cartucho (80%). Noventa e oito porcento dos usuários prévios de seringa preferiu Humapen® à seringa, e cerca de 80% dos pacientes preferiu Humapen®, à caneta anterior. Nenhum paciente abandonou o uso da caneta e, ao final do estudo, 92% dos pacientes respondeu que continuaria usando Humapen®. Treze porcento mencionou alguma dificuldade com a caneta, e as razões mais freqüentes foram o tamanho da caneta e a adaptação inicial. Não houve relato de evento adverso ou problemas técnicos com a caneta. Os profissionais de saúde não referiram dificuldades na instrução da caneta ou na leitura/ ajuste de dose, e ao final do estudo, 100% deles referiu que continuaria recomendando Humapen®. O uso de caneta de insulina deve ser considerado em pacientes diabéticos, tanto naqueles com esquema de mistura fixa, de insulina basal intermediária, como naqueles em esquema de injeções múltiplas.
Traditionally, insulin has been injected using a syringe and a needle. However, injections are frequently considered painful and traumatic, causing psychological/social problems in many patients with diabetes. Insulin injection pens have been found to be more convenient, cause less pain and improve patients' quality of life. We studied the acceptance and functionality of a new reusable insulin pen, Humapen®, in 94 type 1 and type 2 diabetic patients. The patients used insulin for a median period of 7,8 years with 2.7+3.0 injections per day. 42% of them were in previous use of other pens, and the remaining was using syringes. The patients used Humapen® for 8 weeks, maintaining their previous insulin regimen. All patients completed the study. The results with previous users of syringes or pens were similar. Humapen® characteristics most frequently referred by the patients as superior in relation to the previous administration method, and the characteristics most mentioned were: easiness of adjustment/ use (89%), measurement/correction/reading of the dose (88%) and cartridge change (80%). 98% of the previous syringe users, preferred Humapen® to syringe and about 80% of the patients preferred Humapen® to the previous pen. No patient stopped using the pen, and at the end of the study, 92% of the patients answered that they would continue using Humapen®. 13% of the patients referred difficulty with the pen, and the most frequent reasons were the pen size and initial adaptation. There were no adverse events or technical problems with the pen. Health care professionals had no difficulties with the instructions of pen use or reading/adjustment of the dose, and at the end of the study 100% of them said they would continue recommending Humapen®. The use of an insulin pen should be considered in diabetic patients, in patients with fixed mixtures, basal intermediate insulin, and especially in multiple injections regimens.
