ABSTRACT. Black spot (Asperisporium caricae) is one of the main foliar fungal diseases of papaya crops. This disease acts directly on leaves and fruits causing leaf area reduction and fruit deterioration. The quantification of diseases is a fundamental part of the disease management and control process; therefore, a scale is required to help quantify black spot disease. The objective of this work was to propose a standardized methodology to quantify black spot severity in papaya leaves. A scale was developed considering the maximum and minimum values of the disease in the field that included eight levels of severity: 0.1, 0.3, 0.6, 1.0, 2.3, 5.0, 10.0, and 20.0%. Without the aid of a scale the disease is often overestimated, with absolute errors of approximately 75%. When the scale was used, 100% of the evaluators showed improved accuracy and precision, and absolute error was reduced to the 10% range. The scale also provided good repeatability and high reproducibility. The use of the scale provided an improvement in the R2 values, with mean values of 93 and 92 in the second and third evaluations, respectively, demonstrating that the scale is useful for different aspects of the pathosystem of A. caricae, such as for determining the efficiency of fungicides, characterization of varietal resistance, construction of the disease progression curve, and estimation of damage.

ABSTRACT Despite the advantages in production, mechanization may expose workers to high noise levels in the work environment, which is considered one of the main causes of work-related hearing loss. In this sense, this study aimed to analyze the spatial variability of noise generated by a self-propelled coffee harvester in an open area to define safe zones for operators and workers involved in coffee harvesting activities. The noise source used was an Electron Auto TDI self-propelled coffee harvester (model MWM D229-4), with a cabin manufactured in 2012 and a 67-hp 4-cylinder engine, working at 1200-rpm rotation. The noise level was measured by a digital decibel meter at points distributed within a regular 2.5 x 2.5 m sampling mesh (32.5 x 35.0 m area) surrounding the harvester in operation, which was configured according to the regulatory standard. Noise level spatial dependence was analyzed through geostatistics, characterizing structure and magnitude, and mapping spatial variability. Results showed that noise levels were above the limit established by relevant legislation (i.e., 85 dB), both for operators and employees at a distance of about 5.5 m from the generating source.

RESUMO Objetivo: Descrever as práticas de ressuscitação volêmica em unidades de terapia intensiva brasileiras e compará-las com as de outros países participantes do estudo Fluid-TRIPS. Métodos: Este foi um estudo observacional transversal, prospectivo e internacional, de uma amostra de conveniência de unidades de terapia intensiva de 27 países (inclusive o Brasil), com utilização da base de dados Fluid-TRIPS compilada em 2014. Descrevemos os padrões de ressuscitação volêmica utilizados no Brasil em comparação com os de outros países e identificamos os fatores associados com a escolha dos fluidos. Resultados: No dia do estudo, foram incluídos 3.214 pacientes do Brasil e 3.493 pacientes de outros países, dos quais, respectivamente, 16,1% e 26,8% (p < 0,001) receberam fluidos. A principal indicação para ressuscitação volêmica foi comprometimento da perfusão e/ou baixo débito cardíaco (Brasil 71,7% versus outros países 56,4%; p < 0,001). No Brasil, a percentagem de pacientes que receberam soluções cristaloides foi mais elevada (97,7% versus 76,8%; p < 0,001), e solução de cloreto de sódio a 0,9% foi o cristaloide mais comumente utilizado (62,5% versus 27,1%; p < 0,001). A análise multivariada sugeriu que os níveis de albumina se associaram com o uso tanto de cristaloides quanto de coloides, enquanto o tipo de prescritor dos fluidos se associou apenas com o uso de cristaloides. Conclusão: Nossos resultados sugerem que cristaloides são usados mais frequentemente do que coloides para ressuscitação no Brasil, e essa discrepância, em termos de frequências, é mais elevada do que em outros países. A solução de cloreto de sódio 0,9% foi o cristaloide mais frequentemente prescrito. Os níveis de albumina sérica e o tipo de prescritor de fluidos foram os fatores associados com a escolha de cristaloides ou coloides para a prescrição de fluidos.
Abstract Objective: To describe fluid resuscitation practices in Brazilian intensive care units and to compare them with those of other countries participating in the Fluid-TRIPS. Methods: This was a prospective, international, cross-sectional, observational study in a convenience sample of intensive care units in 27 countries (including Brazil) using the Fluid-TRIPS database compiled in 2014. We described the patterns of fluid resuscitation use in Brazil compared with those in other countries and identified the factors associated with fluid choice. Results: On the study day, 3,214 patients in Brazil and 3,493 patients in other countries were included, of whom 16.1% and 26.8% (p < 0.001) received fluids, respectively. The main indication for fluid resuscitation was impaired perfusion and/or low cardiac output (Brazil: 71.7% versus other countries: 56.4%, p < 0.001). In Brazil, the percentage of patients receiving crystalloid solutions was higher (97.7% versus 76.8%, p < 0.001), and 0.9% sodium chloride was the most commonly used crystalloid (62.5% versus 27.1%, p < 0.001). The multivariable analysis suggested that the albumin levels were associated with the use of both crystalloids and colloids, whereas the type of fluid prescriber was associated with crystalloid use only. Conclusion: Our results suggest that crystalloids are more frequently used than colloids for fluid resuscitation in Brazil, and this discrepancy in frequencies is higher than that in other countries. Sodium chloride (0.9%) was the crystalloid most commonly prescribed. Serum albumin levels and the type of fluid prescriber were the factors associated with the choice of crystalloids or colloids for fluid resuscitation.

Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

ABSTRACT Leaf-cutting ants are the main pests in forest plantations. The most commonly used chemical control for the ants is toxic bait. However, the active ingredients in these baits have been restricted by forest certification organizations, justifying the search for new active compounds to control these insects. Thus, this work aimed to evaluate the formicidal activity of alkaloid fractions from the roots, stems, branches and leaves of Psychotria hastisepala and Psychotria leiocarpa (Rubiaceae) against Atta sexdens Forel, 1908 (Hymenoptera: Formicidae). The alkaloid fractions were obtained from the crude methanolic extracts of P. hastisepala and P. leiocarpa by the acid-base extraction method employing liquid-liquid partitions. The fractions, previously solubilized in propanone, were applied topically to the pronotum of worker ants. Bioassays were conducted in a completely randomized design, with six replicates of each treatment and 10 ants per replicate. Insect survival was assessed daily for 21 days. The median lethal time was estimated by Weibull models. The root fractions from P. hastisepala presented the highest toxicity, followed by those from the branches, stems and leaves. Among fractions from P. hastisepala roots, the most toxic were a fraction that apparently is rich in lipophilic compounds and nonbasic alkaloids and a fraction that is likely rich in quaternary benzophenanthridine alkaloids, protopine alkaloids and tertiary bases. Psychotria leiocarpa fractions were not toxic to this insect. Psychotria hastisepala is potentially useful for the development of new products for A. sexdens control.

Objective: To compare results of positron emission tomography (PET) with carbon-11-labeled Pittsburgh compound B (11C-PIB) obtained with cerebellar or global brain uptake for voxel intensity normalization, describe the cortical sites with highest tracer uptake in subjects with mild Alzheimer’s disease (AD), and explore possible group differences in 11C-PIB binding to white matter. Methods: 11C-PIB PET scans were acquired from subjects with AD (n=17) and healthy elderly controls (n=19). Voxel-based analysis was performed with statistical parametric mapping (SPM). Results: Cerebellar normalization showed higher 11C-PIB uptake in the AD group relative to controls throughout the cerebral cortex, involving the lateral temporal, orbitofrontal, and superior parietal cortices. With global uptake normalization, greatest cortical binding was detected in the orbitofrontal cortex; decreased 11C-PIB uptake in white matter was found in the posterior hippocampal region, corpus callosum, pons, and internal capsule. Conclusion: The present case-control voxelwise 11C-PIB PET comparison highlighted the regional distribution of amyloid deposition in the cerebral cortex of mildly demented AD patients. Tracer uptake was highest in the orbitofrontal cortex. Decreased 11C-PIB uptake in white-matter regions in this patient population may be a marker of white-matter damage in AD.

ABSTRACT Using biological inputs to improve the efficiency of nitrogen fertilizers represents an alternative for the cultivation of grasses in tropical regions. Azospirillum brasilense is a species of plant growth promoting bacteria widely studied and used in inoculants. Thus, this study aimed to evaluate the performance of Mombasa grass ( Megathyrsus maximus ) in association with A. brasilense and nitrogen (N) fertilization. The study was conducted under field conditions in Araguaína-Tocantins State, between December 2017 and May 2018. The treatments were arranged in randomized blocks, in a 5 × 2 factorial arrangement, with five doses of N fertilization (0, 25, 50, 75, and 100 kg ha-1) combined with two inoculation treatments (inoculated and non inoculated), in four replicates. For the number of tillers and root production, the inoculation efficiency varied as a function of the supplied N doses. However, the percentage of leaf N was higher for inoculated plants regardless of the application of nitrogen. In the absence of nitrogen fertilization, it was possible to increase forage production by up to 36 % with inoculation.

Abstract INTRODUCTION In recent decades, the prevalence of carbapenem-resistant Acinetobacter isolates has increased, and the production of oxacillinase (OXA)-type carbapenemases is the main mechanism underlying resistance. We evaluated OXA production from 114 Acinetobacter isolates collected between March and December 2013 from different clinical specimens of patients in two hospitals (Hospital 1 [n = 61] and Hospital 2 [n = 53]) located in Niterói, Rio de Janeiro, Brazil. We also evaluated the genetic diversity of OXA-producing isolates. METHODS All the isolates were identified through the automated system Vitek II and matrix-assisted laser desorption ionization-time of flight mass spectrometry MALDI-TOF MS as belonging to the A. baumannii-A. calcoaceticuscomplex. Antimicrobial susceptibility profiles were verified through agar diffusion tests. The presence of OXA-encoding genes was confirmed by PCR. The genetic diversity of isolates positive for carbapenemase production was analyzed through pulsed-field gel electrophoresis. RESULTS There was a high rate of resistance to carbapenems in the isolates (imipenem: 96%; meropenem: 92%) from both hospitals. Moreover, a high percentage (95.6%) of OXA-23-positive isolates was observed for both hospitals, indicating that this was the main mechanism of carbapenem-resistance among the studied population. In addition, most isolates (96.5%) were positive for bla OXA-51. A high genetic diversity and a few major genotypes were found among the OXA-23-positive isolates analyzed. Only intra-hospital dissemination was observed. CONCLUSIONS The elevated dissemination of bla OXA-23-like observed among Acinetobacter isolates from both the studied hospitals highlights the need for continuous epidemiological surveillance in these institutions.

Resumen Los efectos de extractos de las plantas Actinostemon concolor, Geonoma shottiana, Palicourea rigida y Rudgea viburnoides sobre Spodoptera frugiperda fueron evaluados en este estudio. Los extratos se incorporaron en la dieta artificial (1 mg mL-1 de la dieta) y se ofrecieron a larvas de S. frugiperda. El extracto de A. concolor causó 36,1 % de mortalidad. Se detectaron cambios ultraestructurales en los huevos de adultos alimentados en sus estados larvales con extractos de P. rigida y A. concolor. Se observó reducción de la actividad de la tripsina de S. frugiperda después del tratamiento con el extracto de A. concolor. Al cuantificar los fenoles totales en A. concolor, G. shottiana, P. rigida y R. viburnoides, se encontró que A. concolor fue la especie con mayor contenido. El residuo de metanol/agua extracción, que se obtuvo del extracto crudo de A. concolor, contenía compuestos fenólicos que causan cambios en la biología de S. frugiperda similar a los generados por el extracto crudo. En conclusión, A. concolor produce metabolitos secundarios con actividad insecticida sobre S. frugiperda.
Abstract The effects of plant extracts from Actinostemon concolor, Geonoma schottiana, Palicourea rigida and Rudgea viburnoides on Spodoptera frugiperda were evaluated in this study. The extracts were incorporated into an artificial diet (1 mg mL-1 of diet) and offered to S. frugiperda caterpillars. The extract of A. concolor caused 36.1 % mortality in S. frugiperda. Ultrastructural changes were detected in the eggs of adults when the caterpillars were fed with P. rigida and A. concolor extracts. A reduction in trypsin activity was observed after treatment with A. concolor extract. Total phenolics were quantified for A. concolor, G. schottiana, P. rigida and R. viburnoides, with A. concolor presenting the highest content. The residue from methanol/water extraction, which was obtained from the crude extract of A. concolor, contained the phenolic compounds that caused changes in S. frugiperda biology similar to those caused by the crude extract. In conclusion, A. concolor produces secondary metabolites that have insecticidal activity on S. frugiperda.

To contribute to the development of new products to control the coffee leaf miner (Leucoptera coffeella), which is a major pest for coffee plants, this work aimed to select plant species able to produce substances active against this insect and to identify the active substances in the selected plant and their protein targets in the insect. Among the extracts of nineteen plant species, only that from leaves of Merremia tomentosa (Choisy) Hall. f. (Convolvulaceae) reduced the oviposition of L. coffeella on leaves of coffee plants. This extract was submitted to successive fractionation steps to achieve the isolation and identification of two active substances, ursolic acid (UA) and cis-tiliroside (CT). An in silico study showed that UA inhibits glycogen phosphorylases (GP) by binding to their allosteric site, while CT probably inhibits xanthine dehydrogenases (XT). As both GP and XT appear to be essential enzymes for insects, these results suggest that, during evolution, L. coffeella learned to identify plants producing inhibitors of these enzymes to avoid oviposition on such plants. Thus, both UA and CT have potential to be used as lead compounds in the development of new products for the control of the coffee leaf miner.

Lupeol, a triterpene frequently found in Asteraceae plant species, showed moderate to low activity in different strains of Plasmodium falciparum, the most virulent malaria etiological agents. In this work, lupeol was isolated from Parahancornia fasciculata, a plant that is used to treat malaria in the Amazonia region. In the search of more activity lupeol derivatives, five new 1,2,3-triazole hybrid molecules were synthetized by copper-catalyzed azide-alkyne cycloaddition. The antiplasmodial activity of the semi-synthetic compounds were evaluated by the lactate dehydrogenase assay; the lupeol propargyl ether was the only one to disclosing increased activity (half maximal inhibitory concentration-IC50-62.0 ± 1.92 µmol L-1) in relation to lupeol (IC50 117.00 µmol L-1). Therefore, this work revealed a new class of interesting lupeol derivatives that can be obtained by linking electron donors to the hydroxy group at C-3.

Resumo Objetivos Avaliar incidência de tuberculose e triagem para tuberculose latente em brasileiros com artrite reumatoide em uso de agentes biológicos na prática clinica. Pacientes e métodos Estudo de coorte com dados do Registro Brasileiro de Monitoração de Terapias Biológicas (BiobadaBrasil), de 01/2009 a 05/2013, abrangeu 1.552 tratamentos, 415 somente com drogas modificadoras do curso da doença (MMCDs) sintéticas, 942 MMCDs sintéticas em associação com anti-TNF (etanercepte, infliximabe, adalimumabe) e 195 MMCDs sintéticas em associação com outros biológicos (abatacepte, rituximabe e tocilizumabe). Avaliaram-se ocorrência de tuberculose, tempo de exposição às drogas e triagem para TB. Análise estatística: teste t não pareado e teste de Fisher bicaudal; p < 0,05. Resultados O tempo de exposição dos controles foi de 981 pacientes-ano, do grupo de anti-TNF foi de 1.744 pacientes-ano (adalimumabe = 676, infliximabe = 547 e etanercepte = 521 pacientes-ano) e o de outros biológicos de 336 pacientes-ano. A incidência de TB foi de 1,01/1.000 pacientes-ano nos controles e de 2,87 pacientes-ano nos usuários de anti-TNF (adalimumabe = 4,43/1.000 pacientes-ano; etanercepte = 1,92/1.000 pacientes-ano e infliximabe = 1,82/1.000 pacientes-ano). Não houve casos de tuberculose no grupo de outros biológicos. O tempo médio de exposição até a ocorrência de tuberculose foi de 27(11) meses para o grupo anti-TNF. Conclusões A incidência de tuberculose foi maior nos usuários de MMCDs sintéticas e anti-TNF do que nos usuários de MMCDs sintéticas e de MMCDs sintéticas e biológicos não anti-TNF, e também mais tardia, sugerindo infecção durante o tratamento, e não falha na triagem.
Abstract Objectives To assess the incidence of tuberculosis and to screen for latent tuberculosis infection among Brazilians with rheumatoid arthritis using biologics in clinical practice. The occurrence of tuberculosis and the drug exposure time were assessed, and screening for tuberculosis was performed. Statistical analysis: Unpaired t-test and Fisher's two-tailed test; p < 0.05. Results The exposure times were 981 patient-years in the controls, 1744 patient-years in the anti-TNF group (adalimumab = 676, infliximab = 547 and etanercept = 521 patient-years) and 336 patient-years in the other biologics group. The incidence rates of tuberculosis were 1.01/1000 patient-years in the controls and 2.87 patient-years among anti-TNF users (adalimumab = 4.43/1000 patient-years; etanercept = 1.92/1000 patient-years and infliximab = 1.82/1000 patient-years). No cases of tuberculosis occurred in the other biologics group. The mean drug exposure time until the occurrence of tuberculosis was 27(11) months for the anti-TNF group. Conclusions The incidence of tuberculosis was higher among users of synthetic DMARDs and anti-TNF than among users of synthetic DMARDs and synthetic DMARDs and non-anti-TNF biologics and also occurred later, suggesting infection during treatment and no screening failure.

O objetivo deste trabalho foi selecionar genótipos de capim-elefante com base na produção de matéria seca, utilizando a metodologia de modelos mistos (REML/BLUP). O experimento foi conduzido na Pesagro, RJ, Brasil. Foram obtidos 25 híbridos em esquema de dialelo parcial, 10 genitores e a cultivar Capim Cana D’África. Foram avaliadas as características largura da folha (LL), comprimento da folha (CF), diâmetro do colmo (DC) e número de perfilhos (NP) em seis semanas a partir da 5ª semana após o plantio. Na última semana foram avaliadas a porcentagem (%MS) e produção de matéria seca (PMS). Os componentes de variância e coeficientes de repetibilidade foram estimados pelo método da máxima verossimilhança restrita (REML) e a predição de valores genotípicos pela melhor predição linear não viciada (BLUP). Para análise das variáveis foi utilizado o software Selegen-REML/BLUP. Observou-se grande influência da variância ambiental na variância fenotípica para as características LL, CF, DC e NP. Foram obtidos valores de repetibilidade de média magnitude para a LL, CF e DC. Os genótipos que apresentaram potencial para maior produção de biomassa foram H16, H17, G, P1, H4, P4, H25 e H22.
The objective of this work was to select elephant grass genotypes based on dry matter production, using the mixed model methodology (REML/BLUP). The experiment was conducted at Pesagro, RJ, Brazil. Twenty five hybrids were obtained in partial diallel scheme, 10 parents and the cultivar Capim Cana D'Africa. We evaluated the characteristics sheet width (LL), leaf length (CF), stem diameter (DC) and number of tillers (NP) in six weeks from the fifth week after planting. The percentage (% MS) and dry matter production (PMS) was evaluated in the last week. The components of variance and repeatability coefficients were estimated by restricted maximum likelihood (REML) and the prediction of phenotypic and genotypic values for the best linear unbiased prediction (BLUP). The Selegen-REML/BLUP software was used for analysis of the variables. There was great influence of environmental variance in phenotypic variance for traits, CF, DC and NP. Average magnitude of repeatability values were obtained for the LL, CF and DC. The genotypes that presented potential for greater production of biomass were H16, H17, G, P1, H4, P4, H25 and H22.
