Abstract The emergence of bacterial resistance to antimicrobials poses a significant health threat. To address this issue, exploring the fungal diversity in freshwater environments in the Amazon Forest has potential in the search for new antimicrobials. This study aimed to investigate the production of antibacterial metabolites by aquatic fungi from Amazon lakes, specifically Lake Juá and Lake Maicá (Brazil-PA). The fungal isolates were obtained from wood fragments submerged in these lakes, and the ethyl acetate extracts were evaluated for antibacterial activity against Staphylococcus aureus ATCC 25923, S. aureus (MRSA), ATCC 43300, Escherichia coli ATCC 25922, and E. coli (ESBL) NCTC 13353. Additionally, toxicity of the extracts (EtOAc with antimicrobial activity) against human fibroblasts MRC-5 was investigated. The study identified 40 fungal strains with antimicrobial screening, and the ethyl acetate extracts of Fluviatispora C34, Helicascus C18, Monodictys C15, and Fusarium solani LM6281 exhibited antibacterial activity. F. solani LM6281 showed the lowest minimum inhibitory concentration (MIC) of 50 µg/mL against S. aureus strains and MIC of 100 µg/mL against E. coli strains including ESBL. The cytotoxicity (IC50) of the extract (EtOAc) of F. solani LM6281 was 34.5 µg/mL. Preliminary studies of the TLC culture and RNM-H from the extract (EtOAc) of F. solani suggested the presence of substances from the class of terpenes, quinones, phenolics, and flavonoids. This study highlights the potential of submerged wood fungi in the Amazon region to produce antibacterial substances, thus identifying them as sources of novel bioactive compounds with potential use in the pharmaceutical industry and regional bioeconomy.

Resumo O surgimento de resistência bacteriana a antimicrobianos representa uma ameaça significativa à saúde. Para abordar esta questão, explorar a diversidade fúngica em ambientes de água doce na Floresta Amazônica tem potencial na busca de novos antimicrobianos. Este estudo teve como objetivo investigar a produção de metabólitos antibacterianos por fungos aquáticos de lagos amazônicos, especificamente os lagos Juá e Maicá (Brasil-PA). Os isolados fúngicos foram obtidos de fragmentos de madeira submersos nesses lagos, e os extratos de acetato de etila foram avaliados quanto à atividade antibacteriana contra Staphylococcus aureus ATCC 25923, S. aureus (MRSA), ATCC 43300, Escherichia coli ATCC 25922 e E. coli (ESBL) NCTC 13353. Além disso, a toxicidade dos extratos (EtOAc com atividade antimicrobiana) contra fibroblastos humanos MRC-5 foi investigada. O estudo identificou 40 cepas de fungos com triagem antimicrobiana, e os extratos de acetato de etila de Fluviatispora C34, Helicascus C18, Monodictys C15 e Fusarium solani LM6281 exibiram atividade antibacteriana. F. solani LM6281 apresentou a menor concentração inibitória mínima (CIM) de 50 µg/mL contra cepas de S. aureus e CIM de 100 µg/mL contra cepas de E. coli incluindo ESBL. A citotoxicidade (IC50) do extrato (EtOAc) de F. solani LM6281 foi de 34,5 µg/mL. Estudos preliminares da cultura TLC e RNM-H do extrato (EtOAc) de F. solani sugeriram a presença de substâncias da classe dos terpenos, quinonas, fenólicos e flavonoides. Este estudo destaca o potencial de fungos de madeira submersos na região amazônica para produzir substâncias antibacterianas, identificando-os como fontes de novos compostos bioativos com potencial de uso na indústria farmacêutica e na bioeconomia regional.

Resumo Plantas que produzem metabólitos secundários com atividade alelopática ou fitotóxica podem ser biotecnologicamente importantes, servindo como fontes de aleloquímicos e, assim, contribuindo para o setor agroindustrial. Vismia japurensis (Hypericaceae) é uma espécie amazônica que cresce em grupos, formando vismiais. Assim, o objetivo deste estudo foi identificar possíveis efeitos fitotóxicos de extratos hexânicos e metanólicos de folhas e ramos de Vismia japurensis in vivo e de plântulas cultivadas in vitro sobre Lactuca sativa. Além disso, folhas frescas e secas foram analisadas pelo método sanduíche, a fim de determinar sua capacidade de liberação de aleloquímicos. O extrato hexânico de plântulas in vitro reduziu a germinação em 10% e o extrato metanólico promoveu uma redução de 16% na velocidade de germinação. O crescimento radicular de Lactuca sativa foi inibido em 64,7% quando submetido ao extrato hexânico das folhas, em 39,3% sob influência do extrato hexânico dos galhos e em 96,09% para o extrato de hexânico das plântulas in vitro. Quando analisados por cromatografia em camada delgada e ressonância magnética nuclear de 1H, os extratos mostraram evidências de terpenos, antraquinonas e flavonoides, com maior intensidade de sinais na região aromática do extrato hexânico das plântulas in vitro. Assim, Vismia japurensis possui elevado potencial biotecnológico em termos de produção de substâncias de baixa polaridade com capacidade de interferência no desenvolvimento de plantas.

Abstract Plants that produce secondary metabolites with allelopathic activity or phytotoxicity can be biotechnologically important, serving as sources of allelochemicals, and thus contributing to the agroindustrial sector. Vismia japurensis (Hypericaceae) is an Amazonian species that grows in clumps called vismiais, from which most other plants are absent. Accordingly, the objective of this study was to identify possible phytotoxicity effects of hexane and methanol extracts of Vismia japurensis leaves and branches in vivo and from seedlings grown in vitro on Lactuca sativa. In addition, fresh and dry leaves were assayed by the sandwich method in order to determine their ability to release allelochemicals. The hexanic extract from in vitro seedlings reduced germination by 10%, while the methanol extract produced a 16% reduction in germination speed. Root growth of Lactuca sativa was inhibited by 64.7% when subjected to hexane leaf extract, by 39.3% under the influence of hexane branch extract, and by 96.09% for in vitro seedling hexanic extract. When analysed by thin layer chromatography and 1H nuclear magnetic resonance, extracts showed evidence of terpenes, anthraquinones and flavonoids, with greater intensity of signals in the aromatic region of in vitro seedling hexanic extract. Clearly, Vismia japurensis has a high biotechnological potential in terms of the production of substances of low polarity with capacity to interfere in plant development.

Resumen: La acción alelopática puede verificarse por la presencia de sustancias fitotóxicas, moléculas que están dirigidas al sector agroindustrial. El objetivo de este trabajo fue evaluar la composición química de los extractos metanólicos de Colletotrichum dianesei y Xylaria sp. y su potencial fitotóxico sobre Lactuca sativa. Los hongos se cultivaron en medio líquido Sabouraud suplementado con extracto de levadura al 0,2 %, en agitación. Los micelios se sometieron a extracción con disolventes en orden de polaridad creciente, pero en esta investigación se usaron los extractos metanólicos. La actividad alelopática de los mismos se evaluó a una concentración de 1000 µg/mL en cuadruplicado, evaluando: porcentaje y velocidad de germinación y crecimiento de plántulas de lechuga en condiciones de laboratorio. El extracto de C. dianesei redujo el porcentaje de la germinación en un 36 % y retrasó la velocidad de germinación en un 63,8 %, mientras que el extracto de Xylaria sp. no interfirió en el porcentaje de germinación, solamente retrasó en 43,9 % la velocidad de germinación. El crecimiento de las raíces se vio afectado negativamente en 30,5 % y 51,1 % bajo la influencia de los extractos de Xylaria sp. y C. dianesei; también hubo una reducción en la longitud de la parte aérea de 33,1 y 31,8 %, respectivamente. Las Cromatografías en capa fina mostraron la presencia de sustancias aromáticas y terpenos para ambos extractos, y alcaloides solo para el extracto de C. dianesei. Los espectros de rmn de 1H mostraron señales entre 6 y 8,5 ppm (sustancias aromáticas), siendo relativamente más intensas para Xylaria sp., señales entre 3 y 4 ppm (azúcares, mostrando los anómericos solo para Xylaria sp.) y grupos metilas entre 0,7 y 1,3 ppm, sugiriendo terpenos. Los resultados sugieren que las especies de hongos evaluadas tienen potencial fitotóxico y producen terpenos y sustancias aromáticas.

Abstract: The allelopathic action can be verified by the presence of phytotoxic substances, molecules that are directed to the agribusiness sector. The objective of this study was to evaluate the chemical composition of the methanolic extracts from Colletotrichum dianesei and Xylaria sp., and phytotoxic potential on Lactuca sativa. The fungi were grown in Sabouraud liquid medium supplemented with 0.2% yeast extract, under stirring. The mycelia were subjected to solvent extraction in order of increasing polarity, but methanol extracts were used in this investigation. The allelopathic activity was evaluated at a concentration of 1000 µg/mL in quadruplicate, evaluating: percentage and speed of germination, and growth of lettuce seedlings under laboratory conditions. C. dianesei extract reduced germination percentage by 36% and delayed germination speed by 63.8%, while Xylaria sp. didn't interfere with the germination percentage, it only delayed the germination rate by 43.9%. Root growth was negatively affected in 30.5% and 51.1% under the influence of extracts from Xylaria sp. and C. dianesei, there was also a reduction of 33.1 and 31.8% in the aerial part length, respectively. The thin layer chromatography showed the presence of aromatic substances and terpenes in both extracts, and alkaloids only in C. dianesei extract. The 1H nmr spectra showed signals between 6 and 8.5 ppm (aromatic substances), being relatively more intense for Xylaria sp., signals between 3 and 4 ppm (sugars, showing anomers only for Xylaria sp.) and methyl groups between 0.7 and 1.3 ppm, suggesting terpenes. The results suggest that the fungal species evaluated have phytotoxic potential, and produce terpenes and aromatic substances.

Resumo: A ação alelopática pode ser verificada pela presença de substâncias fitotóxicas, moléculas que são direcionadas ao setor agroindustrial. O objetivo deste trabalho foi avaliar a composição química dos extratos metanólicos de Colletotrichum dianesei e Xylaria sp. e seu potencial fitotóxico sobre Lactuca sativa. Os cogumelos foram cultivados em meio líquido Sabouraud suplementado com 0,2% de extrato de levedura, sob agitação. Os micélios foram submetidos à extração por solvente em ordem crescente de polaridade, mas apenas os extratos metanólicos foram utilizados nesta investigação. A atividade alelopática foi avaliada na concentração de 1000 μg/mL em quadruplicado, avaliando-se: porcentagem e velocidade de germinação e crescimento de mudas de alface em condições de laboratório. O extrato de C. dianesei reduziu a porcentagem de germinação em 36% e atrasou a velocidade de germinação em 63,8%, enquanto a Xylaria sp. não interferiu na porcentagem de germinação, apenas atrasou a velocidade de germinação em 43,9%. O crescimento radicular foi negativamente afetado em 30,5% e 51,1% sob a influência dos extratos de Xylaria sp. e C. dianesei; também houve redução no comprimento da parte aérea de 33,1 e 31,8%, respectivamente. As cromatografias em camada delgada mostraram a presença de substâncias aromáticas e terpenos para ambos os extratos, e alcaloides apenas para o extrato de C. dianesei. Os espectros de RMN de 1H mostraram sinais entre 6 e 8,5 ppm (substâncias aromáticas), sendo relativamente mais intensos para Xylaria sp., sinais entre 3 e 4 ppm (açúcares, mostrando os hidrogênios anoméricos apenas para Xylaria sp.) e grupos metila entre 0,7 e 1,3 ppm, sugerindo terpenos. Os resultados sugerem que as espécies fúngicas avaliadas possuem potencial fitotóxico e produzem terpenos e substâncias aromáticas.

The inorganic composition of the bark and leaf of a plant from the Amazon rainforest, Andira surinamensis, was determined using two non-destructive, multi-element techniques: X-ray fluorescence (XRF) and Rutherford backscattering spectrometry (RBS). XRF measurements were made using both a conventional X-ray source and synchrotron radiation. It was observed that although magnesium, aluminum, silicon, phosphorus, sulfur, chlorine, and potassium are present in higher concentrations in the leaf, calcium is about three times more concentrated in the bark. Manganese, iron, copper, zinc, strontium and barium were also detected, with barium showing a concentration above the minimum toxicity level for plants. Chemical speciation of sulfur, performed using the X-ray absorption near edge structure (XANES) technique, showed that sulfur is present in several oxidation states, with a much larger contribution from the inorganic sulfate in the leaves. The article evidences that the combined use of synchrotron radiation and non-destructive multi-element techniques allows for an efficient and accurate determination of the inorganic composition and chemical speciation in plants.

ABSTRACT Candida spp. is associated with almost 80% of all nosocomial fungal infections and is considered a major cause of blood stream infections. In humans, Cryptococcosis is a disease of the lungs caused by the fungi Cryptococcus gattii and Cryptococcus neoformans. It can be potentially fatal, especially in immune-compromised patients. In a search for antifungal drugs, Deguelia duckeana extracts were assayed against these two fungi and also against Candida albicans, which causes candidiasis. Hexane branches and CH2Cl2 root extracts as well as the substances 4-hydroxylonchocarpine, 3,5,4′-trimethoxy-4-prenylstilbene and 3′,4′-methylenedioxy-7-methoxyflavone were assayed to determine the minimal inhibitory concentration. Phytochemical study of CH2Cl2 root and hexane branch extracts from D. duckeana A.M.G. Azevedo, Fabaceae, resulted in the isolation and characterization of nine phenolic compounds: 4-hydroxyderricine, 4-hydroxylonchocarpine, 3′,4′,7-trimethoxy-flavonol, 5,4′-dihydroxy-isolonchocarpine, 4-hydroxyderricidine, derricidine, 3,5,4′-trimethoxy-stilbene, 3′,4′,7-trimethoxyflavone and yangambin. The only active extract was a CH2Cl2 root showing minimal inhibitory concentration 800 µg/ml against C. gattii, and the investigation of compounds obtained from this extract showed that 4-hydroxylonchocarpine was active against all three fungi (C. neoformans, C. gattii and C. albicans). These results suggest that D. duckeana extracts have potential therapeutic value for the treatment of pathogenic fungi.

RESUMORubiaceae é uma família botânica com grande potencial químico e biológico Neste trabalho realizou-se a prospecção fitoquímica e avaliou-se as atividades citotóxica, antioxidante e antifúngica dos extratos diclorometânicos e metanólicos das folhas de Duroia saccifera, Ferdinandusa goudotiana, F. hirsuta, F. paraensis, Ferdinandusa sp., Palicourea corymbifera e P. guianensis. A avaliação da citotoxicidade foi realizada empregando-se o ensaio de toxicidade sobre Artemia salina. A atividade antifúngica foi determinada pelo método de difusão em ágar utilizando-se os fungos: Epidermophyton floccosum, Microsporum canis 32905, M. gipseum 29/00, Trichophyton mentagrophytes ATCC 9533/03, T. rubrum ATCC 28189, T. tonsurans 21/97, Cladosporium cladosporioides, C. sphaerospermum, FusariumU. 662/06, Scytalidium U. 661/06, Candida albicans ATCC 3632 e C. albicans U. 5/99. A atividade antioxidante foi determinada através dos ensaios de redução do radical livre 1,1-difenil-2-picrilhidrazila (DPPH) e de descoloração do cátion radical ácido-6-sulfônico-2,2-azinobis-3-etilbenzotiazolina (ABTS). Para o ensaio de toxicidade sobre A. salina observou-se maior toxicidade no extrato metanólico de F. goudotiana, que se mostrou tóxico até a concentração de 5 µg mL-1. A presença de princípios antifúngicos foi observada em F. hirsuta e F. paraensiscontra quatro dos fungos testados. Na avaliação da atividade antioxidante os extratos metanólicos mostraram maior atividade que os extratos diclorometânicos, sendo coincidente o resultado obtido através de ambos os métodos (redução do DPPH e descoloração do ABTS). Os resultados criam possibilidades para futuras investigações relacionadas à estrutura e atividade dos componentes de cada extrato ativo.

ABSTRACTRubiaceae is a plant family with great chemical and biological potential. In this work a phytochemical prospection was performed and it was assessed the cytotoxic, antioxidant and antifungal activities of dichloromethanic and methanolic extracts of the leaves of Duroia saccifera, Ferdinandusagoudotiana, F. hirsuta, F. paraensis, Ferdinandusa sp., Palicourea corymbifera and P. guianensis. The cytotoxicity evaluation was carried out using the test toxicity on Artemia salina. The antifungal activity was determined by agar diffusion method using fungi: Epidermophyton floccosum, Microsporum canis 32905, M. gipseum 29/00, Trichophyton mentagrophytes ATCC 9533/03, T.rubrum ATCC 28189, T. tonsurans 21/97 , Cladosporium cladosporioides, C. sphaerospermum, FusariumU. 662/06, Scytalidium U. 661/06, Candida albicans ATCC 3632 and C. albicans U. 5/99. The antioxidant activity was determined by testing the amount of free radical 1,1-diphenyl-2-picrylhydrazyl (DPPH) and the discoloration of the radical cation 6-sulfonic acid-2 ,2-azinobis-3-ethylbenzothiazoline (ABTS). For the toxicity test on A. salina greater toxicity in the methanol extract of F. goudotiana was observed, which proved to be toxic up to a concentration of 5 µg mL-1. The presence of antifungal principles was observed in the F. hirsuta and F. paraensis against four tested fungi. In the evaluation of the antioxidant activity, the methanol extracts showed greater activity than the dichloromethanic ones, being similar the obtained result through both methods (reduction of DDPH and ABTS decolorization) . The results create possibilities for future researches related to the structure and activity of the components of each active extract.
