ABSTRACT Introduction. The behavior of COVID-19 is influenced by diverse social differentials that transcend demographic variables and include morbidity, the conditions of their homes, family dynamics, job performance, gender roles and stereotypes, care tasks, habits, and behaviors of life and consumption, among others. Methods. Statistical and demographic analysis and tools of metric studies of the information were used. Results and Discussion. The research showed the importance of incorporating into the analysis of the behavior of COVID-19 in Cuba the influence of social and epidemiological differentials determined by skin color, marital status, employment status, income levels, types of families, mobility, hygienic environmental conditions, access to health services, among others, which can place populations in greater or lesser conditions of vulnerability. Conclusions: The composition and dynamics of the population together with other social differentials are determinants of the behavior and expression of the pandemic. The crossing of epidemiological factors with the analysis of containment measures and the composition and structure of the affected population allows for the design of health policies that are more adjusted to the needs of the pandemic. Distance psychological counseling makes it possible to identify and deal with situations related to confinement and changes in life routines that affect the mental health of different groups of people.

RESUMEN Introducción. El comportamiento de la COVID-19 está influenciado por diferenciales sociales que trascienden las variables demográficas e incluyen la morbilidad, las condiciones de sus viviendas, las dinámicas familiares, el desempeño laboral, los roles y estereotipos de género, las tareas de cuidado, los hábitos y comportamientos de vida y consumo, entre otros. Métodos. Se empleó el análisis estadístico y demográfico y herramientas de los estudios métricos. Resultados y Discusión. La investigación mostró la importancia de incorporar a los análisis del comportamiento de la COVID-19 en Cuba la influencia de diferenciales sociales y epidemiológicos determinados por el color de la piel, el estado conyugal, la situación laboral, los niveles de ingreso, los tipos de familias, la movilidad, las condiciones higiénico ambientales, el acceso a los servicios de salud, entre otros, que pueden ubicar a las poblaciones en mayores o menores condiciones de vulnerabilidad. Conclusiones, la composición y la dinámica de la población junto a otros diferenciales sociales son determinantes del comportamiento de la pandemia. El cruzamiento de los factores epidemiológicos con el análisis de las medidas de contención y la composición y estructura de población afectada permite diseñar políticas sanitarias más ajustadas a las necesidades de la pandemia. La orientación psicológica a distancia permite identificar y atender situaciones vinculadas al confinamiento y los cambios en las rutinas de vida que afectan la salud mental de diferentes grupos de personas.

Abstract Polyamines are thought to be healers of stressinduced physiological damage in plants. It may be assumed that this healing function applies to coral as well, and that polyamines could be a marker of physiological state in these organisms. The levels of Putrescine, Spermidine and Spermine polyamines were measured in three different sites of coral reefs of the Central Mexican Pacific coasts. One of these sites, Playa Mora, exhibited a low tide which almost exposed the coral above the sea level. Tide levels and polyamines in coral samples were measured during February and October from 2012 to 2016. Surprisingly, the highest polyamines levels were measured during October, at a period when the highest tides and precipitations, were recorded with (42.91±1.40 µg/cm2 of Putrescine, 5.31±1.06 µg/cm2 of Spermidine and 12.10±0.75 µg/cm2 of Spermine). The lowest polyamines levels were observed during February, the month with the lowest tides and precipitations, with 1.35±0.16 µg/cm2 of Putrescine, 0.15±0.03 µg/cm2 of Spermidine and 0.07±0.02 µg/cm2 of Spermine. A relation between Spermidine and Spermine concentrations, and tide levels was established. It could be speculated that Polyamines levels are correlated with physiological stress levels of coral reef in these zones of the Central Mexican Pacific coast.

Resumen Se cree que las poliaminas son restauradores del daño fisiológico inducido por el estrés en las plantas. Se podría suponer que esta función benéfica de curación también se aplica al coral, y que las poliaminas podrían ser un marcador del estado fisiológico en estos organismos. Los niveles de las poliaminas Putrescina, Espermidina y Espermina se midieron en tres sitios diferentes de arrecifes de coral de la costa del Pacífico Central Mexicano. Uno de estos sitios, Playa Mora, exhibió una marea baja que casi expuso al coral sobre el nivel del agua. Los niveles de marea y las poliaminas en las muestras se midieron durante febrero y octubre del año 2012 a 2016. Sorprendentemente, los niveles más altos de poliaminas (42.91±1.40 μg/cm2 de Putrescina, 5.31±1.06 μg/cm2 de Espermidina, y 12.10±0.75 μg/cm2 de Espermina) se observaron durante octubre, una época en la que se registraron las mareas más altas y precipitación pluvial. En cambio, los niveles más bajos de poliaminas (1.35±0.16 μg/cm2 de Putrescina, 0.15±0.03 μg/cm2 de Espermidina, y 0.07±0.02 μg/cm2 de Espermina) se vieron durante febrero, el mes con las mareas y precipitación más bajas. Se estableció la asociación entre la concentración de Espermidina y Espermina y los niveles de marea. Se podría especular que los niveles de poliaminas se correlacionan con los niveles de estrés fisiológico del arrecife de coral en estas zonas de la costa del Pacífico Central Mexicano.

1- (fur-2-yl) -2-nitroprop-1-ene (UC-244) is a highly bioactive compound, synthesized in the Center for Bioactive Chemicals at the Central University of Las Villas from sugar cane production wastes. In this work, a preliminary photostability study was carried out as part of the chemical and pharmaceutical analysis prior to product formulation. The effect of sunlight (exposure or protection) on product stability - in terms of organoleptic properties, melting point and UC-244 content - was evaluated both in solid state and in 80:20 acetonitrile-water solutions. In order to determine the UC-244 content in all samples under study, a high-performance liquid chromatography (HPLC) procedure was validated. This chromatographic technique demonstrated to be specific and linear in the range from 5 mg/L to 40 mg/L, as well as precise and accurate under the conditions studied. Degradation of solid samples exposed to light was higher than 90 % in 7 days, whereas those samples protected from sunlight suffered a degradation of only a 25.5 % after 150 days. Liquid samples exposed to light suffered a degradation higher than 90 % in only 50 s, but in those protected from light, UC-244 content only decreased in about 5 % after 50 days. All degraded samples showed significant changes in their organoleptic properties and an increase in their melting point. Therefore, it can be concluded that any formulation developed with UC-244 should be adequately protected from light.

El 1-(fur-2-il)-2-nitroprop-1-eno (UC-244) es un compuesto de marcada acción biológica, sintetizado en el Centro de Bioactivos Químicos de la Universidad Central "Marta Abreu" de Las Villas, a partir de los desechos de la caña de azúcar. En el presente trabajo se realizó un estudio preliminar de estabilidad como parte del análisis químico-farmacéutico previo a la formulación del producto. Se analizó el comportamiento del producto en estado sólido y en disolución acetonitrilo-agua 80:20, al ser expuesto a la luz solar y protegiéndolo de la misma, para lo cual se tuvieron en cuenta las propiedades organolépticas, temperatura de fusión y cuantificación del compuesto. Para la cuantificación del producto durante el estudio se validó una técnica por Cromatografía Líquida de Alta Eficacia (CLAE). La técnica cromatográfica resultó ser específica, lineal en el intervalo de 5 mg/L-40 mg/L, así como precisa y veraz en las condiciones estudiadas. Las muestras sólidas expuestas a la luz se degradaron más del 90 % en 7 días y las que estuvieron protegidas de la misma solo un 25,5 % en 150 días, para las disoluciones expuestas a la luz, la degradación fue de más del 90 % en solo 50 s y las que estuvieron protegidas solo lo hicieron alrededor del 5 % en 50 días. Las muestras degradadas experimentaron cambios significativos en sus propiedades organolépticas y aumento en la temperatura de fusión. Lo anteriormente expuesto permite concluir que cualquier formulación que se desarrolle con el UC-244 debe estar debidamente protegida de la luz.

By the classic biological assays it has been possible to determine the use of HIV-1 strains co-receptor, which have been classified in R5, X4 or R5/X4, characteristic related to the NIS or IS phenotype and to the clinical course. By bio-information methods the amino acid changes in the region of V3 loop of the env using the co-receptor. In present paper are showed the results of a characterization study of eleven strains from HIV-1 infected subjects, five seropositive and six with a fast progression to AIDS. Authors compared the results of phenotype study conducted by two classic assays, use of co-receptors and ability to induce syncytia in MT2 and three bio-information methods, rule 11/25, point matrices in specific locations (PSSM) and the geno2pheno program. The five strains from asymptomatic seropositive patients coincided according all methods used as R5/NIS. Five of the six strains from fast progression patients were classified as R5/X4 and IS by biological assays; whereas two were classified as R5/X4 by Geno 2 Pheno, one by PSSM and all were pure X4 by the rule 11/25. The assays used in the study allowed us to characterize the isolated strains and to relate the phenotype to the infection course, thus, we considered that using biological assays or bio-information methods it is possible to conduct characterization studies, its usefulness depends on the pursued objectives.

Mediante ensayos biológicos clásicos se ha podido determinar el uso del correceptor por las cepas de VIH-1, las cuales se han clasificado en R5, X4 o R5/X4, características que guardan relación con el fenotipo NIS o IS y con la evolución clínica. Por métodos bioinformáticos se han relacionado cambios aminoacídicos en la región del lazo V3 del env con el uso del correceptor. En este trabajo se presentan los resultados del estudio de caracterización de once cepas provenientes de individuos infectados con VIH-1, cinco de ellos seropositivos asintomáticos y seis progresores rápidos al SIDA. Se compararon los resultados del estudio fenotípico realizado por dos ensayos biológicos clásicos, uso de correceptores y capacidad de inducir sincicios en MT2 y tres métodos bioinformáticos, regla 11/25, matrices de puntos en posiciones específicas (PSSM) y el programa geno2pheno. Las cinco cepas de seropositivos asintomáticos coincidieron por todos los métodos como R5/NIS. Cinco de las seis cepas de progresores rápidos se clasificaron R5/X4 e IS por los ensayos biológicos; mientras dos clasificaron R5/X4 por Geno 2 Pheno, una por PSSM y todas fueron X4 puras por la regla 11/25. Los ensayos utilizados en el estudio permitieron caracterizar las cepas aisladas y relacionar el fenotipo con la evolución de la infección, por lo que valoramos que por ensayos biológicos o métodos bioinformáticas se pueden hacer estudios de caracterización, su utilidad está en dependencia del objetivo que se persiga.

The use of ionizing irradiation has been useful for the genetic improvement of forest species. However, its use should be preceded by assays that determine the effect that irradiations have on the plant material. With this objective, an experiment was carried out on Abies religiosa (Kunth) Schltd. et Cham .from the Cofre de Perote, Veracruz, Mexico. We tested the effect of five gamma ray doses (2, 5, 10 and 20 Gy) and an unirradiated control on seed germination, seedling height and number of first leaves. Seedling germination and number of first leaves were severely affected by irradiation doses. Cluster analysis revealed that doses of 5 and 10 Gy, classified in group I, were beneficial for plant height. We suggest that future studies evaluate a wider dose range with doses below 2 Gy and up to 5 Gy.

La utilización de radiaciones ionizantes ha resultado de utilidad en los trabajos de mejoramiento genético de especies forestales. Sin embargo, su uso debe estar precedido por ensayos que permitan determinar el efecto que tiene sobre el material vegetal utilizado. Se realizó un experimento con el fin de evaluar el efecto de diversas dosis de radiaciones gamma (2, 5, 10, 15 y 20 Gy) comparado con un control sin irradiación, sobre el porcentaje de germinación de semillas de Abies religiosa (Kunth) Schltd. et Cham. procedentes del Cofre de Perote, Veracruz, México, así como sobre la variable altura y el número de hojas primarias de los propágulos germinados. Al analizar los datos y compararlos con el control se observó que las dosis produjeron un efecto severo en los caracteres evaluados. La germinación varió de 0 a 39, el control fue de 62 %. Las dosis de 5 Gy, clasificadas dentro del grupo I, por análisis de conglomerado, produjeron un efecto beneficioso en el carácter altura de la planta. No obstante, en estudios futuros que se desarrollen de esta especie, es recomendable evaluar el efecto de varias dosis que incluya valores por debajo de 2 Gy hasta 5 Gy.

BACKGROUND: viral quantification is essential to demonstrate biological product safety during the validation of the production process. OBJECTIVE: to evaluate the accuracy, linearity and detection limit of a titration method for the human immunodeficiency virus type 1. METHODS: the titration was performed by calculating the end-point dilution method, through the cytopathic effect assay and the determination of Ag p24 production by an enzyme immunoassay. Mean infective dose in cultured cells per milimeter was estimated by Reed and Muench´s method. RESULTS: the accuracy of the method was similar to that of the reference titration, the variation coefficient figures for accuracy (repeatability and intermediate accuracy) were lower than 5 % and 10 %, respectively, the linearity showed adequate correlation coefficient and the detection limit was determined in the 10-12 dilution CONCLUSIONS: the evaluated method was suitable for viral quantification

INTRODUCCIÓN: en la validación de los procesos de producción de productos biológicos, la cuantificación viral es esencial para demostrar su seguridad. OBJETIVO: en este trabajo se evaluó el método de titulación del virus de la inmunodeficiencia humana tipo 1, para ello se determinó la exactitud, precisión, linealidad y límite de detección. MÉTODOS: la titulación se realizó por el método del cálculo de la dilución punto final, mediante la visualización del efecto citopático y la determinación de la producción de antígeno p24 por medio de un ensayo inmunoenzimático, la dosis infectiva media en cultivo de células por mililitro se calculó según el método de Reed y Muench. RESULTADOS: la exactitud del método coincidió con el título de referencia, los valores del coeficiente de variación de la precisión (repetibilidad y precisión intermedia) fueron menores que 5 y 10 %, respectivamente; la linealidad mostró un adecuado coeficiente de correlación y el límite de detección se determinó en la dilución 10-12. CONCLUSIONES: el método evaluado demostró un adecuado desempeño para la cuantificación viral.

Objectives. To determine the prevalence of drug resistance and to analyze the subtyping in HIV-1 samples from Cuba. Methods. From an estimated total number of 1 950 HIV-1-infected persons in Cuba, a sample of 103 patients were studied, 76 of whom had received drug treatment for HIV and 27 who had not. The RNA plasma viral load was measured, and automated sequencing was used to assess resistance mutations to reverse transcriptase inhibitors (RTIs) and to protease inhibitors (PIs). Subtyping in the V3 region was performed using heteroduplex mobility assay (HMA). In order to corroborate the HMA results, sequencing of env (C2-V3-C3) was done with one-third of the samples in each of the subtype groups detected by HMA. Results. Out of the 103 samples, 81 of them (78.6%) were classified as subtype B, 19 (18.5%) as subtype A, and 3 (2.9%) as subtype C. The prevalence of resistance mutations was 26.2% to RTIs, none to PIs alone, and 3.9% to both categories of drugs. The prevalence of resistance to nucleoside RTIs (NRTIs) was 27.6% in treated patients and 7.4% in the untreated patients, and for nonnucleoside RTIs (NNRTIs) it was 5.3% and 0%, respectively. Among treated patients a low frequency (2.6%) of dual resistance to zidovudine (ZDV) plus lamivudine (3TC) and abacavir (ABC) was detected, and multidrug resistance to NRTIs was not found. In relation to PIs together with RTIs, the prevalence of resistance was 5.3% for treated patients and 0% for untreated patients. Conclusions. Even though Cuba is generally considered an area where subtype B is dominant, we detected a high proportion of non-B subtype viruses. The low prevalence of resistance mutations to RTIs and PIs reflects the delay in introducing these drugs to Cuba. Multidrug resistance to RTIs was not found, so, as of now, the use of these drugs continues to be an option for Cuban patients.

Objetivos. Determinar la prevalencia de la resistencia a los fármacos y analizar la presencia de mutaciones de resistencia genotípica en los subtipos B y no B del VIH-1 en Cuba. Métodos. Entre un total de 1 950 personas que se estima que están infectadas por el VIH-1 en Cuba, se estudió una muestra de 103 pacientes, de los cuales 76 habían recibido antirretrovíricos y 27 no. Se determinó la carga de ARN vírico en el plasma y se utilizó la secuenciación automatizada para detectar mutaciones de resistencia a los inhibidores de la transcriptasa inversa (ITI) y de la proteasa (IP). También se procedió a la subtipificación de la región V3 con prueba de movilidad de heteroduplex (HMA). Para confirmar los resultados de esta prueba se secuenció el gen env (C2-V3- C3) en un tercio de las muestras de cada uno de los subtipos detectados por HMA. Resultados. De las 103 muestras, 81 (78,6%) fueron clasificadas como pertenecientes al subtipo B, 19 (18,5%) al A y 3 (2,9%) al C. La prevalencia de mutaciones de resistencia fue del 26,2% para los ITI y 3,9% para los ITI más IP. Para los ITI nucleosídicos fue del 27,6% en los pacientes tratados y del 7,4% en los no tratados; para los ITI no nucleosídisos, las cifras correspondientes fueron del 5,3% y 0%, respectivamente. En los pacientes tratados se detectó una baja frecuencia (2,6%) de resistencia a la zidovudina más lamivudina y abacavir, y no se encontró multirresistencia a los ITI nucleosídicos. Para las combinaciones de IP e ITI, la prevalencia de la resistencia fue del 5,3% en los pacientes tratados y del 0% en los no tratados. Conclusiones. Aunque generalmente se considera que en Cuba predomina el sub-tipo B, en este estudio se detectó una alta proporción de virus del subtipo no B. La baja prevalencia de mutaciones de resistencia a los IP e ITI refleja la introducción más tardía de estos fármacos en el país. Como no se observó multirresistencia a los ITI, el empleo de estos fármacos sigue siendo una opción válida para los pacientes cubanos.

Protective effect of policosanol on atherosclerotic lesions in rabbits with exogenous hypercholesterolemia

Policosanol is a mixture of higher aliphatic alcohols purified from sugar cane wax, with cholesterol-lowering effects demonstrable in experimental models and in patients with type II hypercholesterolemia. The protective effects of policosanol on atherosclerotic lesions experimentally induced by lipofundin in rabbits and rats and spontaneously developed in stumptail monkeys have been described. The present study was conducted to determine whether policosanol administered orally to rabbits with exogenous hypercholesterolemia also protects against the development of atherosclerotic lesions. Male New Zealand rabbits weighing 1.5 to 2 kg were randomly divided into three experimental groups which received 25 or 200 mg/kg policosanol (N = 7) orally for 60 days with acacia gum as vehicle or acacia gum alone (control group, N = 9). All animals received a cholesterol-rich diet (0.5%) during the entire period. Control animals developed marked hypercholesterolemia, macroscopic lesions and arterial intimal thickening. Intima thickness was significantly less (32.5 ± 7 and 25.4 ± 4 µm) in hypercholesterolemic rabbits treated with policosanol than in controls (57.6 ± 9 µm). In most policosanol-treated animals, atherosclerotic lesions were not present, and in others, thickness of fatty streaks had less foam cell layers than in controls. We conclude that policosanol has a protective effect on the atherosclerotic lesions occurring in this experimental model.

Policosanol is a mixture of higher aliphatic primary alcohols isolated from sugar cane wax, whose main component is octacosanol. An inhibitory effect of policosanol on platelet aggregation and cerebral ischemia in animal models has been reported. Thus, the objective of the present study was to evaluate the effect of policosanol on cerebral ischemia induced by unilateral carotid ligation and bilateral clamping and recirculation in Mongolian gerbils. Policosanol (200 mg/kg) administered immediately after unilateral carotid ligation and at 12- or 24-h intervals for 48 h significantly inhibited mortality and clinical symptoms when compared with controls, whereas lower doses (100 mg/kg) were not effective. Control animals showed swelling (tissue vacuolization) and necrosis of neurons in all areas of the brain studied (frontal cortex, hippocampus, striatum and olfactory tubercle), showing a similar injury profile. In the group treated with 200 mg/kg policosanol swelling and necrosis were significantly reduced when compared with the control group. In another experimental model, comparison between groups showed that the brain water content of control gerbils (N = 15) was significantly higher after 15 min of clamping and 4 h of recirculation than in sham-operated animals (N = 13), whereas policosanol (200 mg/kg) (N = 19) significantly reduced the edema compared with the control group, with a cerebral water content identical to that of the sham-operated animals. cAMP levels in the brain of control-ligated Mongolian gerbils (N = 8) were significantly lower than those of sham-operated animals (N = 10). The policosanol-treated group (N = 10) showed significantly higher cAMP levels (2.68 pmol/g of tissue) than the positive control (1.91 pmol/g of tissue) and similar to those of non-ligated gerbils (2.97 pmol/g of tissue). In conclusion, our results show an anti-ischemic effect of policosanol administered after induction of cerebral ischemia, in two different experimental models in Mongolian gerbils, suggesting a possible therapeutic effect in cerebral vascular disorders.

The biological characteristics of 11 HIV-1 strains isolated from patients with a fast clinical evolution to AIDS were studied. The viral isolates were classified according to their replication kinetics and cell tropism. Taking into account these criteria, it was observed that 8 of the isolated strains (72,7 %) were of rapid high growth (RH) or slow low 3 (SL3) with preferential tropism to the lymphocytic stock, as it corresponds to AIDS patients. 3 (27,3 %) had characteristics of slow low 1 (SL1). The cytopatogenecity of the strains was studied in the MT4 celular line, and it was observed that most of them (72,7 %) were syncytium-inducing strains (SI), which allowed to prove the in vivo and in vitro relation of the biological properties. It was not so in 3 of the cultures (27,3 %) that behaved as non-syncytium inducers.

Se estudiaron las características biológicas de 11 cepas de VIH-1 aisladas de pacientes con rápida evolución clínica al sida. Los aislados virales se clasificaron según su cinética de replicación y tropimo celular, con estos criterios se observó que 8 de las cepas aisladas (72,7 %) resultaron de crecimiento rápido alto o lento bajo 3 y con tropismo preferencial a la estirpe linfocítica, como corresponde a los pacientes con sida, mientras 3 (27,3 %) tuvieron características de lenta baja 1. La citopatogenicidad de las cepas se estudió en la línea celular MT4 y se observó que la mayoría (72,7 %) resultó inductora de sincicios (IS), por lo que se comprobó relación in vivo e in vitro de las propiedades biológicas, no ocurrió así en 3 de los cultivos (27,3 %) que se comportaron como no inductores de sincicios.
