Abstract This study aimed to develop a protocol for the cryopreservation of Pseudoplatystoma corruscans semen. For this, mature males were hormonally induced with a single dose of carp pituitary extract (5 mg/kg body weight). Semen was collected and evaluated. Two cryoprotectants were tested to compose the diluents: dimethyl acetamide (DMA) and dimethyl sulfoxide (Me2SO), in two concentrations (8% and 10%), + 5.0% glucose + 10% egg yolk. The semen was diluted in a 1: 4 ratio (semen: extender), packed in 0.5 mL straws and frozen in a dry shipper container in liquid nitrogen vapors. After thawing, sperm kinetics, sperm morphology and DNA integrity of cryopreserved sperm were evaluated. Pseudoplatystoma corruscans males produced semen with sperm motility > 80%. After thawing, all treatments provided semen with total sperm motility > 40%, with no significant difference (P < 0.05) between them, as well as between the other sperm kinetic parameters evaluated. The treatments with DMA provided a smaller fragmentation of the DNA of the gametes. Sperm malformations were identified in both fresh and cryopreserved semen, with a slight increase in these malformations being identified in sperm from thawed P. corruscans semen samples.

Resumo Este estudo teve como objetivo desenvolver um protocolo para a criopreservação do sêmen de Pseudoplatystoma corruscans. Para tal, machos maduros foram induzidos hormonalmente com uma dose única de extrato de hipófise de carpa (5 mg/kg de peso vivo). O sêmen foi coletado e avaliado. Sendo testados para compor os diluentes, dois crioprotetores: dimetil acetamida (DMA) e dimetil sulfóxido (Me2SO), em duas concentrações (8% e 10%), + 5,0% glicose + 10% gema de ovo. O sêmen foi diluído na proporção 1: 4 (sêmen: extensor), embalado em palhetas de 0,5 mL e congelado em container dryshipper em vapores de nitrogênio líquido. Após o descongelamento, foram avaliados os aspectos cinéticos espermáticos, a morfologia espermática e a integridade do DNA dos espermatozoides criopreservados. Os machos de P. corruscans produziram sêmen com motilidade espermática > 80%. Todos os tratamentos proporcionaram após o descongelamento sêmen com motilidade espermática total > 40%, sem diferença significativa (P < 0,05) entre eles, como também entre os demais parâmetros cinéticos espermáticos avaliados. Os tratamentos com DMA proporcionaram uma menor fragmentação do DNA dos gametas. Malformações espermáticas foram identificadas, tanto no sêmen fresco, como no criopreservado, sendo identificado um aumento discreto dessas malformações nos espermatozoides das amostras de sêmen descongeladas de P. corruscans.

Abstract This study aimed to verify whether the inclusion of omega-3 polyunsaturated fatty acids (PUFAs-ô3) in the diet of Astyanax lacustris males improves seminal quality. Four hundred fish were arranged in 20 boxes of 180 L in the recirculation system. These were fed with isoprotein diets (32% CP), until apparent satiation, with four levels of marine fish oil rich in PUFAs-ô3 inclusion (In% = 0, 3, 6, and 9), for 105 days. After, males were hormonally induced to spermiation, and semen was collected after 226 ATUs. Parameters evaluated were osmolality, seminal volume and color, spermatic concentration, sperm morphology, membrane integrity and, sperm motility (total and progressive motility, velocity curvilinear, straight line and average path, linearity and rectilinearity coefficients, trajectory oscillation, head lateral displacement amplitude, and cross-beat frequency). Seminal volume was greater in the In-0 and In-3 groups. Inclusion of PUFAs-ô3 positively influenced the kinetic parameters, as treatment with 6% and 9% of inclusion fish oil resulted in higher values for most of these parameters and did not differ statistically from each other. Thus, it is concluded that the addition of PUFAs-ô3 to the feed of breeders significantly improved the seminal quality of A. lacustris males.

Resumo Este estudo teve como objetivo verificar se a inclusão de ácidos graxos poliinsaturados ômega-3 (PUFAs-ô3) na dieta de machos de Astyanax lacustris melhora a qualidade seminal. Quatrocentos peixes foram dispostos em 20 caixas de 180 L no sistema de recirculação. Estes foram alimentados com dietas isoproteicas (32% PB) até aparente saciedade, com quatro níveis de óleo de peixe marinho rico em inclusão de PUFAs-ô3 (In% = 0, 3, 6 e 9), durante 105 dias. Após, os machos foram induzidos hormonalmente à espermiação e o sêmen foi coletado após 226 UTAs. Os parâmetros avaliados foram osmolalidade volume e cor seminal concentração espermática morfologia espermática integridade de membrana e motilidade espermática (motilidade total e progressiva velocidade curvilínea linha reta e trajeto médio coeficientes de linearidade e retilinearidade, oscilação da trajetória, amplitude de deslocamento lateral da cabeça e frequência de batida cruzada). O volume seminal foi maior nos grupos In-0 e In-3. A inclusão de PUFAs-ô3 influenciou positivamente os parâmetros cinéticos, pois o tratamento com 6% e 9% de inclusão de óleo de peixe resultou em valores mais elevados para a maioria desses parâmetros e não diferiram estatisticamente entre si. Assim, conclui-se que a adição de PUFAs-ô3 na ração dos criadores melhorou significativamente a qualidade seminal dos machos de A. lacustris.

Resumo O objetivo deste estudo foi determinar as características do sêmen de Astyanax lacustris após indução hormonal e avaliar a sensibilidade dos espermatozoides da espécie a soluções crioprotetoras baseadas nos crioprotetores dimetilsulfóxido e metilglicol. Volume, cor, concentração espermática, motilidade total e aspectos do movimento espermático foram analisados usando o “Sistema Integrado de Análise de Sêmen (ISAS®CASA)”. Três extensores diferentes foram testados: A) glicose 5%+gema de ovo 10%, B) BTS® 5% e C) glicose 5% e dois crioprotetores permeáveis: dimetilsulfóxido (Me2SO) e metilglicol (MTG). O sêmen fresco de A. lacustris apresentou motilidade total 76,6±11,2%, duração da motilidade 33,0±2,2s, concentração de espermatozoides 7,22±3,2×109sptz/mL e osmolalidade seminal 219±0,03mOsm/kg-1. O teste de toxicidade apresentou maiores valores de motilidade total nas diluições MTG15%+A, Me2SO15%+B e Me2SO10%+C, e as diluições Me2SO10%+C e Me2SO15%+C apresentaram maiores valores de velocidade curvilínea, velocidade linear e velocidade média. O protocolo testado não foi eficaz em manter a viabilidade do sêmen de A. lacustris pós-congelamento, pois não foi observada motilidade em nenhuma das diluições. No entanto, o Ensaio Cometa demonstrou que as soluções crioprotetoras eram eficazes na proteção do material genético das células, pois os níveis de dano ao DNA eram baixos, sem diferença entre controle e Me2SO10%+A, MTG10%+C, Me2SO10%+B e Me2SO15%+B.

Abstract This study aimed to determine the semen characteristics of Astyanax lacustris after hormonal induction and to evaluate the sensitivity of the species sperm to cryoprotective solutions based on the cryoprotectants dimethyl sulfoxide and methyl glycol. Volume, color, sperm concentration, total motility and aspects of sperm movement were analyzed using “Integrated Semen Analysis System”. Three different extenders were tested: A) glucose 5%+egg yolk 10%, B) BTS®5% and C) glucose 5% and two permeable cryoprotectants: dimethyl sulfoxide (Me2SO) and methyl glycol (MTG). Fresh A. lacustris semen presented total motility of 76.6±11.2%, motility duration of 33.0±2.2s, sperm concentration of 7.22±3.2×109sptz/mL and seminal osmolality of 219±0.03mOsm/kg-1. The toxicity test showed the highest total motility values at the MTG15%+A, Me2SO15%+B and Me2SO10%+C dilutions, and the Me2SO10%+C and Me2SO15%+C dilutions presented the highest values for curvilinear velocity, linear velocity and average velocity. The tested protocol was not effective at maintaining the viability of A. lacustris semen after freezing because no motility was observed in any of the dilutions. However, the Comet Assay demonstrated that cryoprotectant solutions were effective in protecting the genetic material of cells, as DNA damage levels were low, with no difference between control and Me2SO10% + A, dilutions MTG10%+C, Me2SO10%+B and Me2SO15%+B.

RESUMO Este estudo avaliou os efeitos dos crioprotetores e das baixas temperaturas no desenvolvimento embrionário de Prochilodus lineatus, descrevendo suas principais alterações morfológicas. Nos testes de sensibilidade ao frio, as taxas de sobrevivência, no estágio de vinte somitos (20S), foram de 53,6% a 0ºC e 100% a 5ºC. Nos testes de toxicidade, os embriões foram expostos a uma série graduada dos crioprotetores: 1,2-Propanediol (PROP), dimetilsulfóxido (Me2SO4) e glicerol (GLY), terminando em uma combinação de alta osmolaridade. Não houve diferença significativa na sobrevivência dos embriões nas séries do PROP e Me2SO4 nos estágios de 6S e 20S. Nos protocolos de resfriamento, foram avaliados os efeitos da baixa temperatura associados as séries crioprotetoras. A 5ºC, o PROP apresentou taxas de sobrevivência acima de 75% no estádio de gástrula (G) e acima de 90% nos estádios 6S e 20S. Foram observadas altas taxas de anormalidades sendo as mais recorrentes: corpos pequenos, nadadeiras apresentando crescimento celular descontrolado, ruptura da membrana e retração vitelínica. Estes resultados demonstram a necessidade do uso das soluções crioprotetoras, mesmo não havendo nucleação de gelo e, em contrapartida, demonstra que as elevadas concentrações crioprotetoras promovem numerosas lesões morfológicas, comprometendo o desenvolvimento embrionário.

ABSTRACT This study evaluated the effect of the cryoprotectants and the low temperatures on the embryonic development of Prochilodus lineatus, describing their main morphological alterations. On chilling sensitivity test, the survival rates at the twenty somites stage (20S) were 53.6% at 0ºC, and 100% in 5ºC. To test toxicity, the embryos were exposed to a graded series of 1,2-Propanediol (PROP), dimethyl sulfoxide (Me2SO4) and glycerol (GLY), terminating in a solution of high osmolarity. There was no significant difference in the embryos survival of toxicity test between series of PROP and Me2SO4 in the 6S and 20S. In the cooling protocols, were evaluated the effects of low temperature associated with cryoprotectants. At 5ºC, PROP showed survival rates above 75% in the gastrula stage (G) and above 90% in the 6S and 20S stages. High rates of abnormalities were observed, and the most recurrent were: small bodies, fins presenting uncontrolled cell growth, membrane rupture, and retraction. These results demonstrate the need to use cryoprotectant solutions, even when there is no ice nucleation, and, on the other hand, shows that high cryoprotectant concentrations promote numerous morphological lesions, compromising normal embryonic development.

A descrição histológica da papila urogenital é uma importante ferramenta para a compreensão dos mecanismos reprodutivos em peixes, assim como um pré-requisito para a realização do transplante de células germinativas em peixes adultos, além de um bom indicador biológico de possíveis alterações ambientais. Foi realizada a investigação histológica da papila urogenital e seus ductos constituintes no lambari Astyanax altiparanae. Os ductos genital e urinário ocorrem separadamente ao longo de maior parte de sua extensão, entretanto, unem-se em um ducto simples antes de abrir para o meio externo. Nos machos esta abertura é assimétrica e parece ter dupla origem, sendo completamente envolvida por fibras musculares estriadas, enquanto nas fêmeas ela é simétrica e as fibras musculares não a envolve totalmente. O ducto espermático e oviducto sofrem alterações ao longo de sua extensão, principalmente na morfologia do epitélio que os envolve. No ducto espermático as células epiteliais passam de pavimentosas a colunares e cuboides, com possível atividade secretora, à medida que se aproxima dos testículos. No oviducto, anteriormente as células também são epiteliais pavimentosas, entretanto, próximo aos ovários, formam-se lamelas compostas por um epitélio pseudoestratificado composto por células cuboides e colunares. O ducto urinário é bastante similar em ambos os sexos apresentando células globosas, cuja descrição é conhecida em mamíferos, porém rara em peixes.

The histological description of the urogenital papilla is an important tool to comprehension of the reproductive mechanisms in fish, as well as a pre-requisite to germ cell transplantation in adult fish, besides to be a good biological indicator to environmental changes. Was performed the histological description of the urogenital papilla and its component ducts in the tetra Astyanax altiparanae. The genital and urinay ducts pass separately throughout most part of its extension, joining in a single duct before opening. In males this opening is asymmetric and seems to have double origin, being completely surrounded by striated muscle fibers, while in females it is symmetric and the muscle fibers does not surround it totally. Spermatic duct and oviduct undergo changes throughout their extension, mainly in the morphology of the surrounding epithelium. In the spermatic duct, squamous epithelial cells change to columnar and cuboid with possible secretory activity, close to testes. In the oviduct, anteriorly epithelial cells are also squamous, however, close to ovary there are lamellae composed by a pseudostratified epithelium with columnar and cuboid cells. The urinary duct is highly similar for both sexes presenting globoid cells, which description is known in mammals, however, rare in fish.

Cinquenta espécimes de Cichla piquiti foram colhidos do Rio Paraná, a jusante da Usina Hidrelétrica de Ilha Solteira, Brasil, na pesquisa de endohelmintos. Todos os peixes estavam parasitados por, pelo menos, uma espécie de helminto (prevalência total [P] = 100%). Foram encontrados oito taxa de parasitas: os nematóides Procamallanus (Procamallanus) peraccuratus no intestino; larvas de terceiro estádio dos anisaquídeos Contracaecum sp. e Hysterothylacium sp. na cavidade abdominal, mesentério e serosa do estômago e do intestino, e sobre o fígado e baço; metacercárias do trematódeo Austrodiplostomum compactum no olho e um indivíduo adulto de Genarchella genarchella no estômago; os cestóides Proteocephalus macrophallus, Proteocephalus microscopicus e Sciadocephalus megalodiscus no intestino. Larvas de Hysterothylacium sp. (P = 86%) e P. microscopicus (P = 74%) foram os parasitas mais prevalentes. Anisaquídeos apresentaram maior prevalência e abundância no período seco. Foi observada uma correlação negativa entre as abundâncias de Hysterothylacium sp. e P. microscopicus, sugerindo uma relação de antagonismo/competição entre esses parasitas. Cichla piquiti é descrito como novo hospedeiro para quatro espécies de parasitas. Estes novos registros aumentam significativamente a lista de parasitas de C. piquiti, contribuindo para o conhecimento da relação parasito-hospedeiro e distribuição geográfica desses helmintos.

Fifty specimens of Cichla piquiti were collected from the Paraná River downstream of the Ilha Solteira Hydroelectric Power Station in Brazil and surveyed for endohelminth parasites. All fish were parasitised by at least one helminth species (overall prevalence [P] = 100%). Eight parasite taxa were present: the nematode Procamallanus (Procamallanus) peraccuratus in the intestines; third-stage larvae of the anisakids Contracaecum sp. and Hysterothylacium sp. in the visceral cavity, mesentery and serosa of the stomach and intestines and on the liver and spleen; the trematodes Austrodiplostomum compactum in the eye (metacercariae) and Genarchella genarchella in the stomach; and the cestodes Proteocephalus macrophallus, Proteocephalus microscopicus, and Sciadocephalus megalodiscus in the intestines. Hysterothylacium sp. larvae (P = 86%) and P. microscopicus (P = 74%) were the most prevalent parasites. Anisakids were more prevalent and abundant in the dry season. A negative correlation between the abundances of Hysterothylacium sp. and P. microscopicus was observed, suggesting a competitive/antagonistic relationship between these parasites. Cichla piquiti represents a new host for four parasite species. These new records significantly increase the list of parasites of C. piquiti, contributing to the knowledge of the host-parasite relationship and the geographical distribution of these helminths.

O presente estudo descreve as fases de maturação testicular (associando o desenvolvimento das células germinativas e as alterações morfológicas sofridas pelo epitélio germinativo ao longo do ano), e a morfologia testicular do tucunaré amarelo Cichla kelberi, relacionando a outras espécies. Com este propósito, 78 indivíduos foram estudados de acordo com técnicas convencionais para microscopia de luz. Os testículos em C. kelberi foram classificados como lobular espermatogonial irrestrito, uma característica apomórfica encontrada nos grupos recentes dos teleósteos. Além disso, cinco fases de maturação testicular foram definidas para C. kelberi: Fase Preparatória; Desenvolvimento Inicial do epitélio germinativo; Desenvolvimento Intermediário do epitélio; Desenvolvimento Final do epitélio germinativo e Regressão. Classificações similares foram descritas para outras espécies, indicando que a classificação gonadal baseada nesta proposta, pode ser aplicada a muitas espécies de peixes. Entretanto, apesar desta similaridade, o ciclo reprodutivo testicular de C. kelberisegue um padrão diferente na fase de Regressão, na qual a reestruturação gonadal e a proliferação espermatogonial ocorrem ao mesmo tempo. Deste modo, os testículos em C. kelberinunca retornam à fase Preparatória para começar um novo ciclo reprodutivo nesta espécie. Este fato também explica a ausência de indivíduos totalmente esgotados após seu primeiro ciclo reprodutivo.

The present study describes the testicular maturation phases (associating the germ cells development and the morphological changes suffered by the germinal epithelium along the whole year), and the testicular morphology in the yellow peacock bass Cichla kelberi, relating it to other species. For this purpose, 78 specimens were studied according conventional techniques of light microscope. The testes in C. kelberi were classified as unrestricted spermatogonial lobular, an apomorphic characteristic in the recent groups of Teleost. Furthermore, were defined five testicular maturation phases: Preparatory phase; Early Germinal Epithelium Development; Mid Germinal Epithelium Development; Late Germinal Epithelium Development and; Regression. Similar classifications were described to other species indicating that the testicular classifications based on this propose, can be applied to lots of fishes. However, besides it similarity, the testicular reproductive cycle of C. kelberifollows a different pattern in the Regression phase, on which the gonadal restructuration and the spermatogonial proliferation gathers at the same time. So, the testes in C. kelberi never return to the Preparatory phase to start a new reproductive cycle, being this one present only at the first reproductive cycle in this species. This fact also explains the absence of individuals totally spent after their first reproductive cycle.

O desenvolvimento embrionário de peixes compreende eventos que ocorrem desde o ovo fertilizado à eclosão das larvas, podendo auxiliar na identificação dos ovos viáveis em estudos de produtividade e sobrevivência, como também nas pesquisas de cultivo desses animais. O objetivo do presente estudo foi caracterizar o desenvolvimento embrionário do Pimelodus maculatus (Siluriformes; Pimelodidae). A embriogênese foi característica de teleósteos, apresentando variações que difere de outras espécies como, tempo de desenvolvimento, tipo da blastocele, momento de segmentação dos somitos, entre outros. Seis estágios de desenvolvimento embrionário foram definidos: zigoto, clivagem, blástula, gástrula, organogênese (dividido em fases: segmentação inicial e segmentação final) e eclosão com período de incubação de 13 horas à 29ºC e de 17 horas à 25ºC. Os ovócitos extrusados apresentaram diâmetro médio de 812 µm antes da hidratação e após 1066 µm. Após a fertilização, apresentaram coloração amarelada e uma camada gelatinosa envolvendo o córion. O padrão de clivagens foi descrito como segue: 2; 4; 8 (4x2); 16 (4x4); 32 (4x8) e 64 (2x4x8) blastômeros até a fase de mórula (+64 células). Também foi possível observar nesta fase, o início da formação da camada sincicial do vitelo (CSV). Em seguida foram observados os estágios de blástula e gástrula. O final da gástrula caracterizou-se pela formação do tampão vitelino. A seguir, iniciou-se a diferenciação das regiões cefálica e caudal, o alongamento do embrião, estruturação das vesículas ópticas, de Kufper, óticas e de uma estrutura não identificada na literatura localizada na camada sincicial do vitelo. O final deste estágio é demarcado pelo desprendimento da cauda. A fase de segmentação final caracterizou-se pela cauda livre, presença de mais de 30 somitos, vesícula óptica e ótica, desenvolvimento do intestino posterior, pigmentação nas regiões cefálica e caudal do saco vitelino e crescimento do embrião. As larvas recém-eclodidas apresentaram esboço do trato digestório, olhos bem evidentes e pigmentados, boca fechada, presença das vesículas encefálicas e com comprimento médio de 3410 µm.

The fish embryonic development comprises the events between the egg fertilization up to larvae hatching, being useful for the identification of viable eggs in productivity and survival studies as well as in raising experiments of several species. The goal of the present study was to characterize the embryonic development of Pimelodus maculatus (Siluriformes; Pimelodidae). The embryogenesis was typical of teleosteans, but with differences in relation to other species such as duration of development, type of blastocoel, moment of somite segmentation among others. Six stages of embryonic development were defined: zygote, cleavage, blastula, gastrula, organogenesis (divided in phases: early segmentation and late segmentation) and hatching with a period of incubation equal to 13 hours at 29 ºC and 17 hours at 25 ºC. The extruded oocytes presented a mean diameter of 812 µm before and 1066 µm after hydration. When fertilized, they presented a yellowish coloration and a gelatinous layer surrounding the chorion. The cleavage pattern is described as: 2; 4; 8 (4x2); 16 (4x4); 32 (4x8) and 64 (2x4x8) blastomeres up to morula phase (+64 cells). It was also possible to observe at this phase, the beginning of the formation of the yolk syncyctial layer (YSL). Afterwards, the blastula and gastrula stages followed. The end of gastrula was characterized by the formation of the yolk plug. Subsequently, the differentiation between cephalic and caudal regions began, along with the embryo elongation, structuring of optic, Kupffer's and otic vesicles besides a previously unidentified structure in the yolk syncyctial layer. The end of this stage is typified by the tail detachment. The late segmentation phase was distinguished by a free tail, presence of more than 30 somites, optic and otic vesicles, development of posterior intestine, pigmentation of cephalic and caudal regions of yolk sac and embryo growth. The recently-hatched larvae presented a primordial digestive tract, quite evident and pigmented eyes, closed mouth, encephalic vesicles and a mean length of 3410 µm.

Visando o desenvolvimento das técnicas de reprodução em peixes, foi realizada a caracterização e a criopreservação do sêmen de Brycon cephalus. As características seminais observadas foram: sêmen de coloração leitosa quase transparente, com um volume médio de 4mL e concentração espermática de 9.617±1.630 x 10(6) espermatozóides/mm³. Os espermatozóides (comprimento = 31,288±4,47 µm) são do tipo "aquasperm", apresentando uma pequena cabeça arredondada (comprimento= 1,752±0,18 µm; largura = 1,752±0,17 µm) sem vesícula acrosomal, com um núcleo com cromatina altamente compactada, formando grumos grosseiros e um complexo centriolar localizado na fossa nuclear; uma peça intermediária (comprimento = 2,561±0,44 µm) que se estreita no sentido antero-posterior, um canal citoplasmático e um flagelo (comprimento = 29,521±4,37 µm). Os testes de fertilização com sêmen descongelado demonstraram um efeito significante (alfa = 0,05) no aumento da fertilidade quando utilizado o diluente tipo B, para ambos os tipos de palhetas, 0,5mL e 4,0mL. Não foi constatado efeito significativo (alfa = 0,05) entre a utilização de palhetas de 0,5mL ou de 4,0mL em relação à porcentagem de eclosão.

Aiming to improve fish reproduction techniques, the characterization and cryopreservation of semen of Brycon cephalus were performed. The seminal characteristics observed were: an almost transparent, milky semen with a mean volume of 4mL, and sperm concentration of 9.617±1.630 x 10(6) spermatozoa/mm³. Spermatozoa (length = 31.288 ±4.47µm) were of the aquasperm type and displayed a small, round head (length = 1.727 ±0.18 µm; width = 1.752 ±0.17µm) without acrosomal vesicle, nucleus with highly condensed chromatin forming coarse clots and centriolar complex located in the nuclear fossa; a midpiece (length = 2.561±0.44µm), narrowed rearward, with a cytoplasmic canal; and a flagellum (length = 29.521 ±4.37µm). Fertilization tests with thawed semen demonstrated a significant effect (alpha= 0.05) on the increase of thawed semen fertility with diluent type B in both 0.5mL and 4.0 mL straws. No significant effect (alpha= 0.05) on hatching rate was observed in both straw sizes used.

A criopreservação do sêmen de fêmeas masculinizadas de truta arco-íris tem como objetivo a racionalização do processo de produção de estoques 100% femininos. Para tal, foi coletado sêmen de machos normais (M) e de dois tipos de fêmeas genotípicas (R e G), masculinizadas pela administração oral de 17alfa-metiltestosterona. R foi obtido pela fertilização de ovócitos normais com sêmen de fêmeas masculinizadas enquanto G foi através de reprodução ginogenética. O sêmen foi diluído em uma solução crioprotetora (glicose 5,4 g, gema de ovo de galinha 10 ml, dimetil sulfóxido 10 ml, água destilada 80 ml) na razão de 1:3 (sêmen/diluidor), envasado em palhetas de 0,5 ml e congelado em um "container" tipo "seco" Cryopac CP-65, à temperatura de -180ºC. A descongelação foi feita em água a 70ºC por 3 segundos. As taxas de fertilização obtidas, não revelaram diferença estatística significativa (P<0.05) entre os três grupos de sêmen descongelados (M = 73,1±11,5%; R = 67,2±23,6%; G = 64±5,8%), indicando que a metodologia de congelação utilizada foi eficaz, tanto na criopreservação do sêmen das trutas normais como para o das masculinizadas.

Cryopreservation of semen from sex-reversed females of rainbow trout aims at rationalizing the production of stocks composed by 100% females. Semen from normal males (M) and two types of genotypic females (R and G), sex-reversed by the oral administration of 17alpha-methyltestosterone, were used. R was obtained by the fertilization of normal eggs with semen of sex-reversed females while G via gynogenetic reproduction. Semen was diluted in an extender solution (glucose 5,4 g, egg yolk 10 ml, dimetil sulfoxide 10 ml, water 80 ml) at 1:3 ratio (semen/extender), stored in straws of 0.5 ml and freezed in a dry container Cryopac CP-65, at -180ºC. Thawing was performed with water at 70ºC for 3 seconds. There were no significant fertilization rate differences (P>0.05) among thawed semen groups (M = 73.1±11.5%; R = 67.2±23.6%; G = 64±5.8%), confirming that the freezing methodology used was efficient to cryopreserve semen of all three trout groups.
