ABSTRACT Background: The aim of this research was to investigate the efficiency of methods to overcome primary dormancy and biochemical processes associated with germination of Erythrina falcata Benth seeds. Seeds were submitted to dormancy overcoming treatments and the water uptake pattern was analysed. We then evaluated the activity of the antioxidant enzymes SOD, CAT, and APX, and quantified lipid peroxidation levels, hydrogen peroxide content and total protein content. The experiments were performed in a completely randomized design and the statistical analysis used was Scott-Knott test at 5% probability. Results: Mechanical scarification with sandpaper leads to approximately 94% of germination in E. falcata, while control non-scarified seeds show only approximately 37% of germination. Treatments with hot water led to high mortality in E. falcata seeds. Rapid water absorption was observed in the first 18 hours of imbition when the seeds were scarified with sandpaper, and radicle emergence was observed after 36 hours. During imbibition there was an increase of antioxidant enzyme activity and a decrease in lipid peroxidation and H2O2 production, suggesting an efficient mechanism for regulating imbibition damage. A decrease in the total protein content was observed during germination of E. falcata seeds. Conclusions: Mechanical scarification is an efficient method for breaking dormancy of E. falcata seeds. During germination of E. falcata seeds there is a reduction in the production of H2O2 and lipid peroxidation, and an increase in the activity of antioxidant enzymes. The total protein content decreased along the germination time.

ABSTRACT Genipa americana L. is a forest species of high socioeconomic potential. However, predatory extractivism actions threaten its existence, making it necessary to adopt conservationist practices. G. americana seeds show sensitivity to desiccation and cooling, making it unfeasible for conservation by conventional methods. Thus, cryopreservation is an promising alternative for the long-term conservation of species that produce unorthodox seeds, such as the genipap. In this sense, the objective was to cryopreservation of seeds of G. americana and to evaluate the effects of desiccation and freezing on germination and establishment of seedlings. Initially, seeds were dehydrated in silica gel 0, 16, 18, 20, 22 and 24 h, and then were cryopreserved in liquid nitrogen (LN) (-196 °C) during 24 h. After thawing, the viability and germination were analyzed. Dehydrated and non-cryopreserved seeds were also analyzed. The silica gel desiccation caused a reduction in viability and germination of the seeds of G. americana. The initial seed water content was so high (47%) that storage in LN (+LN) without prior dehydration treatment resulted in seed mortality. It was verified that, the dehydration in silica gel for the minimum time 20 h (corresponding to 14% water content) provides greater freezing tolerance, allowing the successful cryopreservation. Silica gel dehydration followed by immersion in LN was shown to be highly efficient for cryopreservation of seeds of G. americana, besides germinatio after thawing, high survival rates (100%) were obtained, with growth and normal establishment of the seedlings after acclimatization.

Resumo Strelitzia ou ave-do-paraíso, é uma importante planta ornamental tropical de difícil propagação devido aos problemas de dormência das sementes e ao reduzido número de mudas obtidas nas divisões das plantas. Com o maior conhecimento de sua estrutura, é possível desenvolver estratégias para melhorar o processo de propagação dessa espécie, uma vez que as informações sobre estes aspectos são limitadas. Assim, o objetivo foi caracterizar sementes e embriões de Strelitzia reginae. A caracterização das sementes foi realizada por meio de análise de imagens em equipamento GroundEye, Raio-X e análises anatômicas e bioquímicas. Adicionalmente, foram obtidas as seguintes características biométricas das sementes: área de 0,33 cm2, diâmetro máximo de 0,736 cm, diâmetro lateral máximo de 0,59 cm, diâmetro mínimo de 0,58 cm e perímetro de 2,47 cm. As sementes de S. reginae podem ser consideradas amiláceas por conterem 15% de amido. Pela análise anatômica do tegumento, observou-se a presença de uma exotesta (ex) e uma camada parenquimática posterior que foi dividida em mesotesta (m) e endotesta (en). A caracterização de sementes e embriões de estrelícia apresentou observações relevantes para a taxonomia e fisiologia desta espécie. As sementes são aleuro-amiláceas e apresentam área média de 0,33 cm2, com diâmetro lateral máximo de 0,59 cm. Como resultado de estudos anatômicos, foi possível identificar o tegumento responsável pela dormência.

Abstract Strelitzia or bird-of-paradise, is an important tropical ornamental plant that is difficult to propagate because of seed dormancy issues and the low number of seedlings obtained from plant divisions. With greater knowledge of its structure, it is possible to develop strategies to improve the propagation process for this species, since information on this subject is currently limited. Thus, the objective was to carry out an anatomical, morphological and biochemical characterization of Strelitzia reginae seeds and embryos. The characterization of the seeds was performed through image analysis using GroundEye equipment, X-rays, and anatomical and biochemical analyses. Additionally, the following biometric characteristics of the seeds were obtained: an area of 0.33 cm2, a maximum diameter of 0.736 cm, a maximum lateral diameter of 0.59 cm, a minimum diameter of 0.58 cm, and a perimeter of 2.47 cm. The seeds of S. reginae can be considered starchy since they contain 15% starch. By integument anatomical analysis, we observed the presence of an exotesta (ex) and a posterior parenchymatic layer that was divided into a mesotest (m) and endotest (en). The characterization of Strelitzia reginae seeds and embryos showed relevant observations for the taxonomy and physiology of this species. The seeds are aleurostarches and present an area of 0.33 cm2 on average, with maximum lateral diameter of 0.59 cm. As result of anatomical studies, it was possible to determine coat responsible for integument dormancy.

Resumo: O girassol (Helianthus annuus L.) é uma espécie oleaginosa relevante utilizada na alimentação humana e animal e, em múltiplas aplicações industriais. No entanto, as oleaginosas precisam lidar com a perda de vigor quando utilizadas como propágulos, devido à perda da qualidade fisiológica da semente (viabilidade e vigor) causada por eventos de deterioração. Avaliamos no presente estudo os efeitos de técnicas de condicionamento de sementes (água, ácido ascórbico e nitroprussiato de sódio) no vigor, germinação, enzimas antioxidantes e integridade de membrana de sementes de girassol híbrida HELIO 251 armazenadas. Os parâmetros de germinação mostraram que as sementes não condicionadas (controle) atrasaram a germinação em quase 72 h enquanto todas as sementes condicionadas germinaram 100% em 24 h. O extravasamento de eletrólitos e os níveis de malondialdeído foram maiores nas sementes não condicionadas, indicando maior grau de dano à membrana. Um aumento na atividade da catalase e ascorbato peroxidase também foi observado após 8 h de embebição de sementes condicionadas em comparação com sementes não condicionadas. Em relação à superóxido dismutase, não houve diferenças significativas entre os tratamentos após 8 h de embebição, enquanto a maior atividade foi após 24 h de embebição em sementes não condicionadas. Portanto, pode-se concluir que o priming com ácido ascórbico ou nitroprussiato de sódio apresentou melhor desempenho na germinação de sementes de girassol armazenadas, provavelmente decorrente do menor acúmulo de espécies reativas de oxigênio e consequente redução do dano oxidativo devido a um sistema enzimático antioxidante eficiente.

Abstract: Sunflower (Helianthus annuus L.) is a relevant oilseed species used as feed in human and animal nutrition and in multiple industrial applications. However, oilseeds need to deal with the loss of vigor when used as a propagule, due to loss of seed physiological quality (viability and vigor) caused by deterioration events. We have evaluated in the present study the effects of seeds priming techniques (water, ascorbic acid, and sodium nitroprusside) on vigor, germination, antioxidant enzymes and membrane integrity of stored sunflower hybrid HELIO 251 seeds. Germination parameters showed that non-primed seeds (control) delayed germination in almost 72 h while all primed seeds germinated 100% in 24 h. Electrolyte leakage and malondialdehyde levels were higher in non-primed seeds indicating higher degree of membrane damage. An increase in the activity of catalase and ascorbate peroxidase was also observed after 8 h of imbibition of primed seeds compared to non-primed seeds. Regarding superoxide dismutase, there was no significant differences between treatments after 8 h of imbibition, whereas the highest activity was after 24 h of imbibition in non-primed seeds. Therefore, it can be concluded priming with ascorbic acid or sodium nitroprusside exhibited better performance in germination of stored sunflower seeds, probably resulting from lower accumulation of reactive oxygen species and consequent reduced oxidative damage due to an efficient antioxidant enzyme system.

RESUMO. As sementes da cultura do gergelim contém cerca de 50% de teor de óleo, sendo uma das principais razões que estimulam o seu cultivo. No entanto, informações sobre metodologias para avaliação da qualidade de sementes dessa cultura são escassas. Dessa forma, objetivou-se adequar as metodologias dos testes de envelhecimento acelerado para sementes de gergelim e investigar alterações no comportamento de isoenzimas das sementes submetidas ao teste. Foram utilizados quatro cultivares de sementes de gergelim da safra 2014/2015. Foi feita a caracterização fisiológica das sementes e a análise eletroforética das isoenzimas superóxido dismutase, esterase, catalase, álcool desidrogenase, malato desidrogenase e isocitrato liase. Para o teste de envelhecimento acelerado, as sementes foram submetidas ao método tradicional e com solução saturada de NaCl, pelos períodos de envelhecimento de 0, 24, 48, 72 e 96 horas. Concluiu-se que é possível avaliar o vigor de sementes de gergelim pelo teste de envelhecimento acelerado, pelo método tradicional por 72 horas e com solução saturada de NaCl a 45ºC por 48 horas. Quando associado à atividade das isoenzimas EST, CAT, ADH e ICL foi possível visualizar que houve variação significativa na intensidade da expressão das bandas conforme avança o processo deterioração das sementes.

ABSTRACT. The seeds of a sesame crop contain approximately 50% oil content, one of the main reasons for their cultivation. However, information regarding methodologies for the assessment of quality seeds of this culture are scarce. Therefore, the objective of this study was to adapt the methods of accelerated ageing tests for sesame seeds and investigate changes in the behaviour of isoenzymes. The physiological characterization of the seeds and electrophoretic analysis of the isoenzymes superoxide dismutase, esterase, catalase, alcohol dehydrogenase, malate dehydrogenase and isocitrate lyase were also performed. For the accelerated ageing test, the seeds were subjected to the traditional method, with a saturated NaCl solution, at ageing times of 0, 24, 48, 72, and 96 hours. It was found that it is possible to evaluate the effect of accelerated ageing tests on sesame seeds by using the traditional method for 72 hours and saturated NaCl solution at 45°C for 48 hours. When associated with the activity of isozymes EST, CAT, ADH, and ICL, it was possible to observe that there was a significant variation in the intensity of expression of the bands as the seed deterioration process progressed.

RESUMO A germinação in vitro de sementes é uma alternativa eficaz para obtenção de explantes com rapidez e qualidade sanitária. No entanto, sementes de jenipapeiro apresentam germinação lenta e desuniforme. Para tanto, fatores internos e externos às sementes que interferem diretamente no processo, devem ser identificados, visando à adaptação de melhores técnicas para obtenção de mudas. Neste sentido, objetivou-se otimizar a germinação in vitro de sementes de Genipa americana L. por meio da avaliação de diferentes fatores (qualidade de luz, tratamento com GA3, pré-embebição em água destilada, diferentes composições no meio de cultivo e estratificação no escuro). Verificou-se que a germinação das sementes foi indiferente a luz, no entanto, os melhores resultadados foram obtidos sob condições de escuro contínuo; Não houve efeito da aplicação de GA3 exógeno; A pré-embebição em água destilada durante 48h contribui para obtenção de melhores taxas de germinação; E a redução de sais do meio MS, bem como o pré-tratamento de estratificação no escuro maximizam o potencial germinativo das sementes. Portanto, as condições ótimas para germinação in vitro de sementes de G. americana L. requer a pré-embebição em água destilada por 48 horas e a inoculação em meio de cultivo constituído de 1/2 MS + 15 g L-1 de sacarose, com estratificação no escuro por 16 dias, seguida pela transferência a salas de crescimento com iluminação fornecida por lâmpadas fluorescentes branca.

ABSTRACT The in vitro seed germination is an effective alternative for quickly obtaining explants with sanitary quality. However, jenipapo seeds present slow and uneven germination. Therefore, internal and external factors to seed which directly interfere in the process, they must be identified, in order to adapt better techniques to obtain seedlings. In this sense, this work aimed to optimize the in vitro germination of Genipa americana L. seeds by evaluating different factors (light quality, GA3 treatment, pre-soaking in distilled water, growing media and stratification in the dark). It was found that the seed germination of G. americana was indifferent to light, however, the best results were obtained under conditions of continuous darkness; There was no effect of the application of exogenous GA3; The pre-soaking in distilled water for 48 h contributes to obtaining better germination rates; And the reduction in MS medium salts, and laminating the pretreatment in the dark maximizes the germination potential of seeds.Therefore, the optimal conditions for in vitro germination of G. americana L. seeds requires pre-soaking in distilled water for 48 hours and inoculation into culture media consisting of 1/2 MS + 15 g L-1 sucrose, with stratification in the dark for 16 days, followed by the transfer to growth chambers with lighting provided by white fluorescent lamps.

Objetivou-se, com essa pesquisa, realizar a classificação quanto a tolerância à dessecação e a capacidade de armazenamento de sementes de cinco espécies florestais de matas ciliares da bacia do Alto Rio Grande, no Estado de Minas Gerias. Sementes de Casearia sylvestris Swart (Salicaceae), Qualea grandiflora Mart. (Vochysiaceae), Guarea kunthiana A. Juss. (Meliaceae), Eremanthus incanus Less. (Asteraceae), Protium heptaphyllum March. (Burseraceae) foram coletadas e transportadas ao Laboratório de Sementes Florestais, aonde foram beneficiadas e submetidas a dois tipos de secagem, rápida e lenta. Em seguida, foram armazenadas em freezer ou em câmara fria e avaliadas quanto à viabilidade. Após a classificação fisiológica, verificou-se que sementes de C. sylvestris e E. incanus são ortodoxas. Em contra-partida, sementes de G. kunthiana e P. heptaphyllum foram classificadas como recalcitrantes e Q. grandiflora como intermediárias, pois não toleram a secagem a baixos conteúdos de água.

Seeds of five forest species were classified according to their physiological storage behavior. Seeds of Casearia sylvestris Swart (Salicaceae), Qualea grandiflora Mart. (Vochysiaceae), Guarea kunthiana A. Juss. (Meliaceae), Eremanthus incanus Less. (Asteraceae), Protium heptaphyllum March. (Burseraceae) were collected and taken to the laboratory, where they were processed and submitted to both rapid and slow drying, storage and assayed for viability. After physiological classification regarding storage behavior, it was observed that seeds of C. sylvestris and E. incanus presented orthodox behavior. Seeds of G. kunthiana and P. heptaphyllum were classified as recalcitrant and Q. grandiflora as an intermediate, which did not tolerate low moisture content.

A maturação das síliquas de nabo forrageiro ocorre de maneira desuniforme, o que dificulta a definição do momento ideal de colheita para produção de sementes de qualidade superior. Além disso, o alto conteúdo lipídico da semente pode dificultar sua conservação. Para avaliar o efeito do estádio de maturação na colheita sobre a qualidade fisiológica e a composição química de sementes de nabo forrageiro armazenadas sob diferentes condições, as síliquas foram colhidas em três estádios de maturação identificados pelas colorações, verde, bege e marrom. As sementes provenientes dos diferentes estádios de colheita das síliquas foram armazenadas em condições ambientais e câmara fria e seca (10 ºC e 46% de UR) e avaliadas quanto à sua qualidade fisiológica, sanitária e composição química antes de armazenadas e aos 3, 6 e 9 meses de armazenamento. A cor das síliquas não é indicativo ideal para definição do momento de colheita das sementes de nabo forrageiro pela variação de maturidade e qualidade fisiológica das sementes dentro das síliquas. O armazenamento das sementes de nabo forrageiro em câmara fria preserva a qualidade fisiológica conforme avaliação do teste de emergência. Não foram observadas alterações consistentes na composição centesimal em síliquas de nabo forrageiro colhidas em diferentes estádios de maturação relacionados à sua alteração de cor.

The oil radish siliques maturation occurs in a non-uniform way, which makes it difficult to define the ideal harvesting time in order to produce high quality seeds. In addition, the high lipid content of the seed may hamper its preservation. To evaluate the effect of the maturation stage at harvesting on the physiological quality and chemical composition of the oil radish seeds stored under different conditions, the siliques were harvested at three stages, according to three colors: green, beige and brown. Seeds from different siliques harvesting stages were stored at ambient conditions and in a cold chamber under dry conditions (10 ºC and 46% RH) and evaluated as to their physiological and health quality and chemical composition before being stored and after 3, 6 and 9 months of storage. The siliques color is not the ideal indicator to define the oil radish seeds harvesting time due to the variation of the maturity and physiological quality of oil radish seeds within the siliques. The oil radish seeds storage in cold chamber preserves the physiological quality as assessed by the emergence test. There were no consistent changes in the centesimal composition of oil radish siliques harvested at different stages of maturation in relation to their color alteration.

Herbicidas que apresentam atividade residual no solo podem se tornar problema para os cultivos agrícolas, pela possibilidade de intoxicar culturas sucessoras. No caso dos hormonais, há grande dificuldade na determinação de resíduos pela técnica do bioensaio, em função da variação na resposta em crescimento, apresentada pela espécie indicadora. Neste trabalho, objetivou-se determinar espécies e variáveis sensíveis aos herbicidas 2,4 D e picloram, para indicação de resíduos no solo. Para isso, realizaram-se dois bioensaios, sendo que cada um, constituiu-se de 60 tratamentos, em esquema fatorial, compostos pela combinação de três espécies vegetais (Lycopersicon esculentum, Cucumis sativus e Beta vulgaris), dois substratos (areia e amostra de solo) e dez subdoses do herbicida (0; 3; 6; 12; 22,5; 47; 94; 187,5; 375 e 750 g ha-1) e (0; 3; 6; 9; 18; 30; 60; 123; 246 e 501 g ha-1) para o 2,4-D e picloram, respectivamente. O delineamento experimental utilizado para ambos os ensaios foi em blocos casualizados, com quatro repetições. O aumento das doses dos herbicidas provocou reduções nas variáveis altura de planta, matéria seca e, aumento da intoxicação visual das espécies avaliadas. A intoxicação visual mostrou-se a variável mais adequada para determinação de baixos níveis de resíduos de 2,4 D e picloram no solo. B. vulgaris foi a espécie com maior sensibilidade ao 2,4 D enquanto que L. esculentum foi a espécie mais sensível ao picloram.

Herbicides that present soil residual activity may become a problem in agricultural crops due to the possibility of crops successors intoxication. It is hard to detect residues of hormonal herbicides using the bioassay technique, due to the variation in the growth response of the indicator plant. The objective of this work was to select plants and variables susceptible to the herbicides 2,4-D and picloram for soil residue indication..To make it possible, two bioassays were performed,, where each of them consisted of 60 treatments in a factorial scheme with the combination of three plant species (Lycopersicon esculentum, Cucumis sativus and Beta vulgaris), two substrates (sand and soil sample) and 10 subdoses of herbicide (0, 3, 6, 12, 22.5, 47, 94, 187.5, 375 and 750 g ha-1) and (0, 3, 6, 9, 18, 30, 60, 123; 246 and 501 g ha-1) for 2,4-D and picloram, respectively. The experimental design for both trials was randomized blocks with four replications. Increasing doses of the herbicide caused reductions in plant height, total dry matter and visual intoxication of the assessed species. The visual intoxication was the most suitable variable for the determination of low residues levels of D 2.4 and picloram in soil. B. vulgaris was the most sensitive species to 2,4 D and L. esculentum was the most sensitive species to picloram,

A mangabeira destaca-se por possuir um grande potencial como planta frutífera. Suas sementes apresentam recalcitrância, dificultando sua propagação, o que torna evidente a necessidade da obtenção de mudas por via assexuada. Neste contexto, a cultura de tecidos apresenta-se como uma alternativa a ser utilizada. Entre os fatores que afetam a morfogênese in vitro, as citocininas merecem destaque, pois influenciam na diferenciação de gemas e no crescimento das brotações. Assim sendo, avaliou-se, neste trabalho, a influência de três diferentes fontes de citocinina (6-benzilaminopurina BAP, cinetina CIN e thidiazuron TDZ) na indução de brotações in vitro de mangabeira, bem como o efeito residual dessa classe de reguladores no enraizamento e na multiplicação em subcultivos sucessivos. Os resultados mostraram que, entre as citocininas testadas, a 6-benzilaminopurina foi a que induziu maior número de brotações (1,98), gemas (19,22) e folhas (18,86) por explante, tendo promovido a formação de brotos de maior comprimento (4,55 cm). As brotações oriundas de meio WPM basal apresentaram maior facilidade de enraizamento. A 6-benzilaminopurina foi responsável pela maior taxa de multiplicação (9,61) de brotações de mangabeira. Os subcultivos sucessivos diminuíram a capacidade de multiplicação de explantes caulinares da espécie.

The mangabeira stands out for having a strong potential for fruit production. Its seeds present recalcitrance, making its propagation difficult, which makes much clear the need to obtain seedlings through asexual methods. In this context, the tissue culture presents as an alternative to be used. Among the factors that affect in vitro morphogenesis, the cytokinins should be highlighted as it influences the differentiation of buds and shoot growth. In this context the influence of three different sources of cytokinins (6-benzylaminopurine BAP, kinetin CIN and thidiazuron TDZ) was evaluated on the induction of in vitro mangabeira shoots, as well as the residual effect of this type of regulator on rooting and multiplication in successive subcultures. The results showed that among the tested cytokinins, the 6-benzylaminopurine was the one that induced a higher number of shoots (1.98), buds (19.22) and leaves (18.86) per explant, promoting the formation of shoots with higher length (4.55 cm). The shoots from basal WPM medium rooted easier. The 6-benzylaminopurine was responsible for the highest rate of multiplication (9.61) of mangabeira shoots. Sucessive subcultures decreased the capacity of shoot multiplication.

Estudos de ecofisiologia sob condições semicontroladas em viveiros e casas de vegetação são essenciais para o uso de espécies nativas em programas de recuperação de áreas degradadas e para o plantio comercial. Talisia subalbens (Mart) Radlk, cascudo, é uma espécie frutífera, nativa do Cerrado em vias de extinção. O comportamento ecofisiológico dessa espécie foi avaliado em viveiro sob differentes condições de sombreamento (pleno sol, 30%, 50% e 70%), sendo avaliados o crescimento inicial, a alocação de biomassa, as trocas gasosas e os teores de clorofilas. Plantas sob pleno sol apresentam maior acúmulo de massa seca total, parte aérea e sistema radicular. Para a relação raiz/parte aérea, não houve diferença significativa entre os tratamentos. Mudas cultivadas a pleno sol e a 30% de sombreamento apresentaram maiores valores de altura, diâmetro do colo e área foliar. Não foram observadas diferenças na condutância estomática e fotossíntese entre os níveis de sombreamento. Os teores de clorofila a, b e total foram superiores em plantas a 70% de sombreamento. Níveis mais elevados de radiação são fundamentais para o desenvolvimento inicial de mudas de T. subalbens.

Ecophysiological studies under semi-controlled conditions in nurseries and greenhouses are essential to enable the use of native species to recover degraded areas and for commercial planting. Talisia subalbens (Mart) Radlk, 'cascudo', is a native fruiting species of the Cerrado on the verge of extinction. The ecophysiological performance of this species was evaluated in nursery conditions under different levels of shading (full sunshine, 30%, 50% and 70%). Initial growth, biomass allocation, gas exchange and chlorophyll content of the plants were analyzed. Full sunshine cultivated plants showed a higher accumulation of total, shoot, and root dry biomass. There was no significant difference in the root/shoot ratio among the treatments. Seedlings cultivated under full sunshine and 30% shading showed higher values for height, basal diameter, and leaf area. Differences in stomata conductance and photosynthesis rate were not observed among the different shading levels. Plants cultivated under 70% of shading had higher contents of chlorophyll a, b, and total. During the initial phase with higher levels of radiation were fundamental for the development of T. subalbens seedlings.

A Talisia subalbens (Mart.) Radlk. (Sapindaceae) é uma espécie arbustiva apontada como endêmica do Mato Grosso. Objetivou-se neste estudo avaliar os efeitos da temperatura e condições de secagem sobre a germinação de sementes de T. subalbens. No primeiro experimento avaliou-se a germinação das sementes em diferentes regimes térmicos: 15ºC-25ºC e 25ºC-35ºC (alternadas), 25ºC e 35ºC (constante), e no segundo experimento, as sementes foram submetidas à diferentes condições de secagem: rápida (estufa à 35ºC) e lenta (temperatura ambiente à 28±2ºC). Os testes de germinação foram realizados em câmaras do tipo B.O.D., sob fotoperíodo de 12 horas, utilizando-se vermiculita como substrato. O delineamento foi o inteiramente casualizado, com quatro repetições de 25 sementes cada amostra. Os valores médios de porcentagem de germinação e índice de velocidade de emergência (IVE) foram comparados pelo teste de Tukey (5%). As sementes germinadas a 25ºC, 35ºC e 25ºC-35ºC apresentaram maiores porcentagens de germinação, não diferindo entre si, contudo, maior vigor foi observado à 35ºC, sendo esta considerada a temperatura ótima de germinação da espécie. Os menores valores de germinação e IVE ocorreram a 15ºC-25ºC. O grau de umidade inicial das sementes foi de 48%. A velocidade de secagem das sementes afeta negativamente a porcentagem de germinação, sendo o teor crítico de grau de umidade de 25%, e a secagem rápida mais favorável para a espécie.

Talisia subalbens (Mart.) Radlk. (Sapindaceae), a shrubby species, is considered endemic to Mato Grosso. The aim of this study was to evaluate the responses of its seeds to factors such as the optimum germination temperature and the effect of different drying conditions on the physiological quality of the seeds. In the first experiment, seed germination under different temperatures was evaluated: 15ºC-25ºC and 25ºC-35ºC (alternate), 25ºC and 35ºC (constant). In the second experiment, the seeds were submitted to different drying conditions: fast (oven at 35ºC) and slow (room temperature at 28±2ºC). The germination tests were performed in a B.O.D-type chamber, under a photoperiod of 12 hours, using vermiculite as the substrate. The experimental design was the completely randomized with four replications of 25 seeds for each sample, and the average values of germination at the different temperatures compared by the Tukey test (5%). Seeds germinated at 25ºC, 35ºC, and 25ºC-35ºC presented higher percentages of germination, not differing from each other. However, greater vigor was found at 35ºC, this being considered the optimum temperature. The lowest values of germination and germination index (GI) occurred at 15ºC-25ºC. The initial moisture of the seeds was 48%. The drying of the seeds negatively affected the germination, with the critic moisture content of 25% and the fast drying being more favorable to the germination.

A qualidade do pão de queijo está diretamente ligada à matéria-prima utilizada, a preparação da massa, ao congelamento e ao assamento, sendo que falhas nestes processos podem resultar em um produto de baixa aceitação no mercado, tanto nacional quanto internacional. O presente trabalho foi desenvolvido no Departamento de Ciência dos Alimentos da Universidade Federal de Lavras, Minas Gerais e objetivou acompanhar as características físico-químicas da massa e do pão de queijo, durante um período de 120 dias de congelamento. Observou-se para a massa modelada de pão de queijo um menor valor de umidade na massa fresca; os teores de cinzas, extrato etéreo e pH decresceram com o congelamento; já o conteúdo de acidez titulável aumentou durante o período. Com relação aos pães de queijo, verificou-se que os valores de umidade e cinzas não apresentaram variação durante o período; o teor de pH apresentou variações durante todo o congelamento, com menores valores ao final de 120 dias; o conteúdo lipídico apresentou decréscimo em todo o período e a acidez titulável decresceu até próximo aos 60 dias, com posterior aumento até o final do congelamento. O armazenamento congelado também levou a um aumento na força de compressão, ou seja, a uma maior firmeza dos pães de queijo.

The quality of cheese bread is directly linked to the raw material used, the preparation of the dough, freezing, and baking. Fail in these processes may result in a product of low acceptance in the national and international market. The present research was carried out at the Food Science Department of the Universidade Federal de Lavras, Minas Gerais with the aim to accompany the physical and chemical characteristics of the dough and cheese bread, during a period of 120 days of freezing. The modeled dough of cheese bread presented lower moisture content value in the fresh dough; the ash content, ethereal extract and pH decreased with freezing however, the acidity increased during this period. In relation to the cheese bread it was verified that the moisture content and ash values did not present variation during the period; the pH presented variations during the freezing, with smaller values at the end of 120 days; the lipid content decreased with the freezing, and the acidity decreased until approximately the 60th day, with subsequent increase until the end of the freezing. The frozen storage also led to an increase in the compression force, that is, a larger firmness of the cheese breads.

A mangabeira (Hancornia speciosa Gomes) destaca-se por possuir um grande potencial como planta frutífera e produtora de borracha. As dificuldades encontradas no seu processo de propagação por meio de sementes, devido, principalmente, à baixa taxa de germinação e à recalcitrância, valorizam a busca por soluções alternativas para a produção de mudas dessa espécie, de maneira rápida e eficiente. Objetivou-se, neste trabalho, realizar o estudo da germinação de sementes de mangabeira em condições in vitro, tendo como precedente a obtenção de explantes, para posterior utilização no cultivo in vitro. Neste estudo foram avaliados os efeitos de diferentes meios de cultura, concentrações de sacarose e GA3 e de três níveis de pH na germinação da mangabeira. Frutos maduros foram coletados, passaram por processo de beneficiamento e tiveram suas sementes retiradas e utilizadas como explantes. Maior porcentagem de germinação de sementes de mangabeira in vitro foi obtida com a utilização dos meios de cultura WPM e MS/2, suplementados com 15,0 g L-1 de sacarose, 0,2 mg L-1 de GA3 e com pH corrigido para 5,8.

The Hancornia speciosa Gomes species presents potential for fruit and rubber production. Propagation is difficult primarily due, to a reduced seed germination and occurrence of recalcitrant seeds that stimulate the search of rapid and efficient propagation alternatives. In this context, the aim of this work was to study in vitro seed germination conditions in order to produce explants to be used on in vitro culture. The effect of different culture media, sucrose and GA3 concentrations and three pH levels were evaluated. Seeds were extracted from mature fruits after being harvested and processed. Higher in vitro germination was obtained using WPM and MS/2 media supplemented with 15.0 g L-1, 0.2 mg L-1 GA3 and pH adjusted to 5.8.
