ABSTRACT Plants grown under salinity are subject to many morphological and physiological disorders. In this context, practices that can enable the use of saline water are essential. The deleterious effects of salinity can be mitigated by using beneficial microorganisms, especially diazotrophic bacteria. In this context, this study aimed to evaluate the effect of bacterial inoculation and co-inoculation using Bradyrhizobium spp. and Azospirillum brasilense on the growth, nodulation and production of cowpea under salt stress. The experiment was conducted using a randomized block design, in a 4 × 5 factorial arrangement, corresponding to four nitrogen sources (N1 - without nitrogen and without inoculant; N2 - fertilization with mineral nitrogen and without inoculant; N3 - inoculation of Bradyrhizobium spp.; and N4 - co-inoculation of Bradyrhizobium spp. and Azospirillum brasilense) and five values of electrical conductivity of irrigation water - ECw (0.4, 1.9, 3.4, 4.9 and 6.4 dS m-1), with five replicates. Co-inoculation with Azospirillum brasilense and Bradyrhizobium spp. favors the growth, production and nodulation of cowpea plants up to ECw of 0.4 dS m-1. Increase in the electrical conductivity of irrigation water negatively affects the growth, production and nodulation of cowpea plants in Corujinha variety, regardless of the nitrogen source used.

Abstract Healthcare workers present an increased risk of contagion for the SARS-CoV-2 virus due to their labor exposure. Here, we describe the clinical, laboratory, and immunological characteristics of healthcare workers, before vaccine application, exposed to SARS-CoV-2-infected patients. We collected sociodemographic, clinical, and laboratory information from 50 professionals who worked during the COVID-19 pandemic at the Clinical Hospital of the Northwest in Brazil. The results showed that most workers are women, over 50 years old, and worked as nursing technicians. Approximately 56% of workers were positive for a previous infection by RT-PCR and/or anti-SARS-CoV-2-immunoglobulin tests. Increased levels of hematocrit, neutrophils, NK lymphocytes, and fibrinogen, were found in positive healthcare workers, suggesting a light inflammatory status. The immunological findings showed an increase in IL-17 production and a Th2/Th17/Th22 profile followed by high serology for anti-SARS-CoV-2 IgM and IgG. Those data reveal the importance of studies with healthcare workers to investigate if the continuous exposition to the virus may result in chronic activation of the immune system and/or pulmonary inflammation in this target group.

Abstract Health professionals working to mitigate the COVID-19 pandemic are one of the main risk groups for the disease, being prioritized for vaccination. Considering this, the aim of this study was to analyze the immune response of these professionals immunized with CoronaVac in the first and second doses. Blood samples were collected after the first and second doses of the vaccine (CoronaVac) and used to investigate hematological and biochemical parameters, analysis of immunoglobulin production, cytokines, and gene expression profile, as well as the identification of subsets of immune cells. Post-first dose immunological phenotypic memory (CD27+) profiles (T CD4+, TCD8+ and CD19+) showed a significant increase, as did Monocyte APCs (CD80+HLA-DR+) in relation to the second dose. The cytokines IL-2, IL-6 and IFN-° showed increased values in relation to the other analyzed cytokines. The Th2/Th17 profile in the second dose was characterized by gene expression analysis. The production of IgM and IgG after vaccination showed statistically significant values in the comparison between doses. CoronaVac showed activation of APCs monocytes, memory response of T and B lymphocytes, with immunoglobulins production. This set of responses is characterized by the Th2/Th17 immunological profile.

Abstract Shellfish gathering is an artisanal activity and a means of family subsistence for several coastal communities along the Brazilian coast and Anomalocardia brasiliana is one of the main exploited mollusk species. This paper aimed to obtain information about the temporal distribution, growth, yield, and gonadosomatic index of the bivalve mollusk A. brasiliana in a natural stock to provide subsidies for the sustainable extraction of this resource. The study was carried out in an area of the Acaraú River estuary, on Volta do Rio Beach, municipality of Acaraú, Ceará, Brazil. Samples were collected monthly for one year at spring low tide, using the random squares method. Water and sediment temperature and salinity were measured in situ. Sediment samples were collected for granulometric analysis. The specimens were individually quantified, measured, and weighed. A total of 8,507 A. brasiliana specimens were sampled, and the highest density recorded was 2,534 individuals/m2, whose longest shell length reached between 21 and 25 mm, while the mean meat yield was 12.42%. The means of biometric variables of individuals showed significant differences during the sample period. Frequency histograms evidenced the presence of adult and juvenile individuals, showing a continuous reproductive cycle in the studied natural stock. These results can assist the sustainable shellfish gathering activity on Volta do Rio Beach. We recommend that A. brasiliana is above 20 mm in length to be captured and the reduction in the capture effort from April to September, when the highest density of juveniles was observed, to guarantee the sustainability of this stock.

Abstract Background Cathepsin D (CatD) is a lysosomal proteolytic enzyme expressed in almost all tissues and organs. This protease is a multifunctional enzyme responsible for essential biological processes such as cell cycle regulation, differentiation, migration, tissue remodeling, neuronal growth, ovulation, and apoptosis. The overexpression and hypersecretion of CatD have been correlated with cancer aggressiveness and tumor progression, stimulating cancer cell proliferation, fibroblast growth, and angiogenesis. In addition, some studies report its participation in neurodegenerative diseases and inflammatory processes. In this regard, the search for new inhibitors from natural products could be an alternative against the harmful effects of this enzyme. Methods An investigation was carried out to analyze CatD interaction with snake venom toxins in an attempt to find inhibitory molecules. Interestingly, human CatD shows the ability to bind strongly to snake venom phospholipases A2 (svPLA2), forming a stable muti-enzymatic complex that maintains the catalytic activity of both CatD and PLA2. In addition, this complex remains active even under exposure to the specific inhibitor pepstatin A. Furthermore, the complex formation between CatD and svPLA2 was evidenced by surface plasmon resonance (SPR), two-dimensional electrophoresis, enzymatic assays, and extensive molecular docking and dynamics techniques. Conclusion The present study suggests the versatility of human CatD and svPLA2, showing that these enzymes can form a fully functional new enzymatic complex.

ABSTRACT: Salmonella spp. are one of the most important agents of foodborne disease in several countries, including Brazil. Poultry-derived products are the most common food products, including meat and eggs, involved in outbreaks of human salmonellosis. Salmonella has the capacity to form biofilms on both biotic and abiotic surfaces. The biofilm formation process depends on an interaction among bacterial cells, the attachment surface and environmental conditions. These structures favor bacterial survival in hostile environments, such as slaughterhouses and food processing plants. Biofilms are also a major problem for public health because breakage of these structures can cause the release of pathogenic microorganisms and, consequently, product contamination. The aim of this study was to determine the biofilm production capacity of Salmonella serotypes at four different temperatures of incubation. Salmonella strains belonging to 11 different serotypes, isolated from poultry or from food involved in salmonellosis outbreaks, were selected for this study. Biofilm formation was investigated under different temperature conditions (37°, 28°, 12° and 3°C) using a microtiter plate assay. The tested temperatures are important for the Salmonella life cycle and to the poultry-products process. A total of 92.2% of the analyzed strains were able to produce biofilm on at least one of the tested temperatures. In the testing, 71.6% of the strains produced biofilm at 37°C, 63% at 28°C, 52.3% at 12°C and 39.5% at 3°C, regardless of the serotype. The results indicate that there is a strong influence of temperature on biofilm production, especially for some serotypes, such as S. Enteritidis, S. Hadar and S. Heidelberg. The production of these structures is partially associated with serotype. There were also significant differences within strains of the same serotype, indicating that biofilm production capacity may be strain-dependent.

Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Plants experience various environmental stresses, but tolerance to these adverse conditions is a very complex phenomenon. The present research aimed to evaluate a set of genes involved in osmotic response, comparing soybean and medicago with the well-described Arabidopsis thaliana model plant. Based on 103 Arabidopsis proteins from 27 categories of osmotic stress response, comparative analyses against Genosoja and Medicago truncatula databases allowed the identification of 1,088 soybean and 1,210 Medicago sequences. The analysis showed a high number of sequences and high diversity, comprising genes from all categories in both organisms. Genes with unknown function were among the most representative, followed by transcription factors, ion transport proteins, water channel, plant defense, protein degradation, cellular structure, organization & biogenesis and senescence. An analysis of sequences with unknown function allowed the annotation of 174 soybean and 217 Medicago sequences, most of them concerning transcription factors. However, for about 30% of the sequences no function could be attributed using in silico procedures. The establishment of a gene set involved in osmotic stress responses in soybean and barrel medic will help to better understand the survival mechanisms for this type of stress condition in legumes.

The SBBrasil 2010 Project (SBB10) was designed as a nationwide oral health epidemiological survey within a health surveillance strategy. This article discusses methodological aspects of the SBB10 Project that can potentially help expand and develop knowledge in the health field. This was a nationwide survey with stratified multi-stage cluster sampling. The sample domains were 27 State capitals and 150 rural municipalities (counties) from the country's five major geographic regions. The sampling units were census tracts and households for the State capitals and municipalities, census tracts, and households for the rural areas. Thirty census tracts were selected in the State capitals and 30 municipalities in the countryside. The precision considered the demographic domains grouped by density of the overall population and the internal variability of oral health indices. The study evaluated dental caries, periodontal disease, malocclusion, fluorosis, tooth loss, and dental trauma in five age groups (5, 12, 15-19, 35-44, and 65-74 years).

BACKGROUND: Mechanisms underlying vascular abnormalities in obesity remain to be completely clarified. OBJECTIVE: L-arginine/nitric oxide pathway was evaluated on vascular response of high-fat diet-obese rats, focusing on endothelial and smooth muscle cells. METHODS: 30-day-old rats were divided in two groups: control (C) and obese (OB, high-fat diet for 30 weeks). After 30 weeks, body weight, adiposity index, blood pressure, and metabolic and endocrine profiles of the animals were recorded. Curves to noradrenaline were obtained in absence and presence of nitric oxide synthase inhibitor (L-NAME, 3x10-4M) on intact and denuded thoracic aorta from C and OB rats. RESULTS: Body weight, adiposity index, leptin and insulin levels were increased in OB, while blood pressure was unchanged. Obesity also produced glucose tolerance and insulin resistance. Reactivity to noradrenaline of intact aorta was similar in C and OB rats. L-NAME presence produced a similar increase in maximal responses, but a higher leftward shift of noradrenaline responses in intact aorta from C than in OB rats [EC50 (x10-7M): C = 1.84 (0.83-4.07), O = 2.49 (1.41-4.38); L-NAME presence C = 0.02 (0.01-0.04)*, O = 0.21 (0.11-0.40)*†,*p < 0.05 vs respective control, †p < 0.05 vs control plus L-NAME, n = 6-7]. None of the protocols altered the reactivity to noradrenaline of denuded aortas. CONCLUSION: High-fat diet-induced obesity promotes metabolic and vascular alterations. The vascular alteration involved an endothelial L-arginine/NO pathway improvement was probably correlated to diet-induced hyperinsulinemia and hyperleptinemia. The greater resistance to L-NAME effects in aorta of obese rats raises concerns about the lower cardiovascular vulnerability of obese individuals in the presence of associated pathologies that impair NO-system activity.

Duguetia furfuracea, Annonaceae, a pasture weed, is also used in folk medicine in several Brazilian states. Because of the significant difference in odor between two groups of this plant in a remnant patch of savanna in Campo Grande county, Mato Grosso do Sul, six samples were selected according to the odor intensity in the leaves (In-01, In-03, and In-05: pronounced odor; In-02, In-04, and In-06: weak odor or its absence). The leaves were collected and subjected to steam distillation for extraction of essential oils, which were analyzed by GC-MS. A good agreement was found between CG/MS results and olfactory evaluation of the samples: specimens exhibiting leaf scent had a high percentage of monoterpenes and some sesquiterpenes, as follows: In-01 (β-phellandrene, 42.2%; myrcene, 6.8%; α-phellandrene, 4.6%); In-03 (terpin-4-ol, 21.6%; sabinene, 17.3%; p-cymene, 5.6%); In-05 (sabinene, 25.1%; terpin-4-ol, 16.2%; p-cymene, 8.3%). Only sesquiterpenes were found in the specimens having weakly scented or scentless leaves (main constituent bicyclogermacrene: 21.4%, 24.0%, and 29.1%, respectively, for In-02, In-04 and In-06).

The object of this work was to evaluate the effectof essential oils from Long-pepper leaves Piper hispidinervum on the mycelial growth of Alternaria alternata and to analyze the influence of emulsifier Tween® 80 concentration in the control of this phytopathogen. The essential oil was obtained by steam stream distillation, using a modified Clevenger apparatus, and the chemical composition was analyzed by gas chromatography coupled to mass spectroscopy GC-MS and GC. For biological tests, the bioanalytical in vitro method was used, observing the growth or inhibition of such phytopathogen in BDA culture media, in the presence of different essential oil concentrations (0, 100, 250, 500, e 1000 mgL -1) under different Tween® 80 concentrations. Factorial scheme with four repetitions, in entirely randomized outline was adopted. The essential oil of Long-pepper inhibited the micellial growth of the A. alternata fungus at all of the concentrations tested. A 100% inhibition at the 1000 mgL -1 concentration was observed, and the percentage of emulsifier (Tween® 80) influenced the fungitoxic activity at the 250 mgL-1 and 500 mgL -1 concentrations of the essential oil.
