Resumo Este estudo objetivou avaliar os efeitos da temperatura e motilidade embrionária sobre o desenvolvimento esquelético de jacaré-do-pantanal (Caiman yacare). Os ovos foram incubados com 90% de umidade e empregou-se a temperatura de 29°C por 45 dias. Após, para a incubação do Grupo I a temperatura continuou em 29°C, mas associou-se à injeção de 4-aminopiridina (29°C-4AP, n = 15) aplicada nos dias 46, 47, 48 e 49, do Grupo II permaneceu em 29°C (n = 14) e do Grupo III elevou-se para 33°C (n = 14). A movimentação foi mensurada através do monitor digital Egg Buddy® nos dias 30, 35, 42, 49, 56 e 60 dias. Aos 60 dias, os embriões foram eutanasiados e coletadas amostras embrionárias. Na análise estatística não foram observadas diferenças entre os grupos para o fator temperatura sobre a motilidade embrionária no desenvolvimento esquelético. Em contraste, a motilidade evidenciou diferença estatística no dia 49 para o Grupo I (P < 0,001) e apresentou maiores proporções de nariz e mão. Esses dados demonstraram pela primeira vez que o aumento na motilidade, induzidos farmacologicamente resultam em divergências fenotípicas na proporção de segmentos anatômicos durante a ontogenia pré-natal, podendo alterar efetivamente a adaptação dos animais em ambientes específicos.

Abstract This study aimed to evaluate whether skeletal development of the Pantanal Caiman (Caiman yacare) is similarly influenced by temperature variation and controlled increases in embryo motility. All eggs were incubated at 90% humidity and 29 °C for the first 45 days. Thereafter, the incubation temperature was either maintained at 29 °C and embryos were treated with 4-aminopyridine (4-AP) on days 46, 47, 48, and 49 (Group I, 29 °C 4-AP, n = 15); maintained at 29 °C (n = 14; Group II); or at 33 °C (n = 14, Group III). Embryonic movement was measured using an Egg Buddy® digital monitor on days 30, 35, 42, 49, 56, and 60, at which point embryos were euthanized and samples were collected for analysis. No differences were observed between groups with varying incubation temperatures. In contrast, embryonic motility was greater in embryos treated with 4-AP (P < 0.001) on day 49, and this was associated with higher proportions of snout-vent and hand lengths. This study demonstrates for the first time that pharmacologically induced increases in embryo motility result in phenotypic changes to the proportion of elements during prenatal ontogeny, thereby effectively altering the adaptation of the species to specific environments.

Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development.

Abstract A complete genome of the first anellovirus infecting the wild felid Leopardus pardalis (ocelot) and a partial genome were assembled and annotated through high-throughput sequencing protocols followed by Sanger sequencing validation. The full-length virus obtained comprises 2,003 bp, while the partial genome comprises 1,224 bp. Phylogenetic analysis grouped these two sequences in two distinct clusters related to previously described Felidae anelloviruses. The ORF1 of the partial genome was identified as a new species provisionally called Torque teno ocelot virus, with 53.6% identity with its sister lineage. The complete genome was inferred as a new representative of the Torque teno felid virus 3 species, with 73.28% identity to the closest reference. This study expands known virus diversity and the host span of anelloviruses.

Abstract The presence of fungi in healthcare settings, including hemodialysis units, represents a significant risk for immunocompromised patients. This study aimed to investigate the occurrence of fungi in the air and water of a hemodialysis unit located in a tertiary public hospital in Maceió, Alagoas, Brazil. Over a period of three consecutive months, monthly air samples were collected and analyzed using the spontaneous sedimentation technique on Petri dishes containing Sabouraud Dextrose Agar (SDA). Simultaneously, water samples (100 mL) were collected from four specific water distribution points and subjected plating on SDA. Fungi were phenotypically identified based on their macroscopic and microscopic characteristics. In total, 498 colony-forming units (CFUs) of fungi were isolated, with 86 CFUs originating from the air and 412 CFUs from the water. Regarding the water samples, a higher concentration of fungal CFUs was observed in the potable water from the supply network (229 CFUs). Unexpectedly, 23 CFUs were identified in the reverse osmosis samples and 11 CFUs in the storage tank, which are post-treatment points where the presence of microorganisms is not desired. The fungus Cladosporium spp. was the most prevalent in both air and water samples, followed by Penicillium spp. in the air and Rhodotorula spp. in the water. These findings underscore the need to implement effective control and monitoring measures for fungi in the hemodialysis unit to ensure patient safety.

Resumo A presença de fungos em ambientes de saúde, incluindo unidades de hemodiálise, representa um risco significativo para pacientes imunocomprometidos. Este estudo teve como objetivo investigar a ocorrência de fungos presentes no ar e na água de uma unidade de hemodiálise localizada em um hospital público terciário na cidade de Maceió, Alagoas, Brasil. Ao longo de três meses consecutivos, amostras mensais de ar foram coletadas e analisadas por meio da técnica de sedimentação espontânea em placas de Petri contendo Ágar Sabouraud Dextrose (ASD). Paralelamente, amostras de água (100 mL) foram coletadas em quatro pontos específicos de distribuição de água e submetidas ao plaqueamento por esgotamento em meio ASD. Os fungos foram identificados fenotipicamente por meio de associação de suas características macroscópicas e microscópicas. No total, foram isoladas 498 unidades formadoras de colônias (UFC) de fungos, sendo 86 UFC provenientes do ar e 412 UFC da água. Em relação à água, observou-se uma maior concentração de UFC de fungos na água potável proveniente da rede de abastecimento (229 UFC). Inesperadamente, foram identificados 23 UFC nas amostras da osmose reversa e 11 UFC no reservatório de armazenamento, pontos de pós-tratamento nos quais a presença de microrganismos não é desejada. O fungo Cladosporium spp. foi o mais prevalente tanto no ar quanto na água, seguido por Penicillium spp. no ar e Rhodotorula spp. na água. Esses achados enfatizam a necessidade de implementar medidas eficazes de controle e monitoramento de fungos na unidade de hemodiálise, visando garantir a segurança dos pacientes.

Considering the promising antitumor effects of compounds with dual anti-inflammatory and antiproliferative activities, thus benzophenones analogs (2-7) were evaluated on in vivo anti-inflammatory assay and molecular docking analysis. Those with the best molecular docking results were in vitro evaluated on cyclooxygenase (COX) enzymes and tested regarding antiproliferative activity. All derivatives displayed in vivo anti-inflammatory activity. Among them, the substances 2’-hydroxy-4’-benzoylphenyl-β-D-glucopyranoside (4), 4-hydroxy-4’-methoxybenzophenone (5) and 4’-(4’’-methoxybenzoyl)phenyl-β-D-glucopyranoside (7) showed the best values of Glide Score in COX-2 docking evaluation and 4 and 5 selectively inhibited COX-2 and COX-1 in vitro enzymatic assay, respectively. Thus, 4 and 5 were tested against breast cancer (MCF-7, MDA MB-231, Hs578T) and non-small-cell-lung cancer (A549) cell lines. The estrogen-positive MCF-7 cell line was more responsive compared to other tested cell lines. They induced cell cycle arrest at G1/S transition in MCF-7 cell line once there was an increase in G0/G1 population with concomitant reduction of S population. The antiproliferative activity of these substances on MCF-7 was associated with their ability to inhibit cyclin E expression, a critical regulator of G1/S transition. Taken together, the data indicate that 4 and 5 have dual anti-inflammatory and antiproliferative activities and support further studies to evaluate their antitumor potential.

Abstract Flow accelerated corrosion (FAC) is a steel degradation that occurs in heat recovery steam generators in the power industry. The mechanism of this corrosion comprises an electrochemical dissolution of the semi-protective magnetite layer (Fe3O4) that is formed within the pipes employed in the boilers. The FAC is influenced by different factors such as fluid velocity, pH and dissolved oxygen concentration. In this context, Rotating Cage tests were used to evaluate the influence of pH and dissolved oxygen content on FAC of A210, P11 and P22 steels. General corrosion was lower for P11 and P22 steels, an effect consistent with the presence of chromium in their compositions. General corrosion was most critical at pH 8.5. The corrosion intensity decreased for the three steels when the dissolved oxygen concentration increased, at 2 m/s. This behavior is coherent considering the most effective precipitation of Fe2O3 inside the pores of the Fe3O4 layer in a more oxidizing environment. For 3 m/s, the corrosion intensity increased for both materials when oxygen increased, showing the shear stress effect. These behaviors were more significant at pH 8.5. About pitting corrosion, the analyzes show localized attack in the majority of coupons, however with very low number of pits.

Resumo O interesse por espécies vegetais com ação antiviral tem crescido exponencialmente e algumas tem sido relatadas como possuídoras de propriedades anti-HIV. Essa pesquisa tem como objetivo realizar o fracionamento fitoquímico biodirecionado pela atividade antirretroviral das cascas do caule da espécie Lafoensia pacari. Trata-se de um estudo experimental in vitro e a metodologia envolve preparo do material vegetal, obtenção do extrato etanólico, fracionamento, purificação, identificação e quantificação das frações, extração ácido-base, ressonância magnética nuclear, teste de inibição da TR do HIV-1 e estudos de docking molecular. A partir do fracionamento biodirecionado pela atividade antirretroviral verificou-se uma maior atividade nas subfrações acetanólica. Com destaque para a subfração acetanólica neutros com 60,98% de inibição de TR e o ácido elágico com 88,61% de inibição de TR e ausência de citotoxicidade. Verificou-se com o teste de citotoxicidade em linhagem de macrófagos que a fração clorofórmica foi mais tóxica que a fração acetanólica. A análise do espectro J-resolvido na região aromática apresentou um simpleto em 7.48 e 6.93 ppm que foram identificados como ácido elágico e ácido gálico respectivamente. A enzima 5TIQ obteve melhor parâmetro de afinidade com o ligante ácido elágico que foi confirmado pelos espectros HSQC-1H-13C. O ácido gálico mostrou-se também favorável a formar interação com a enzima 5TIQ, sendo confirmado através do espectro HSQC-1H-13C. Através da avaliação do PreADMET verificou-se que o ácido elágico é uma molécula promissora pela sua atividade de inibição da TR e parâmetros farmacocinéticos e de toxicidade.

Abstract Interest in antiviral plant species has grown exponentially and some have been reported to have anti-HIV properties. This research aims to perform the bio-guided phytochemical fractionation by antiretroviral activity of Lafoensia pacari stem barks. This in vitro experimental study involved the preparation of plant material, obtention of ethanolic extract, fractionation, purification, identification and quantification of fractions, acid-base extraction, nuclear magnetic resonance, HIV-1 RT inhibition test and molecular docking studies. From the bio-guided fractionation by the antiretroviral activity there was a higher activity in the acetanolic subfractions, highlighting the acetate subfraction – neutrals with 60.98% of RT inhibition and ellagic acid with 88.61% of RT inhibition and absence of cytotoxicity. The macrophage lineage cytotoxicity assay showed that the chloroform fraction was more toxic than the acetate fraction. The analysis of the J-resolved spectrum in the aromatic region showed a singlet at 7.48 and 6.93 ppm which was identified as ellagic acid and gallic acid, respectively. The 5TIQ enzyme obtained better affinity parameter with the ellagic acid ligand, which was confirmed by the HSQC-1H-13C spectra. Gallic acid was also favorable to form interaction with the 5TIQ enzyme, being confirmed through the HSQC-1H-13C spectrum. From the PreADMET evaluation it was found that ellagic acid is a promising molecule for its RT inhibition activity and pharmacokinetic and toxicity parameters.

Resumo Objetivo: Este artigo analisa a capacidade absortiva e as ações de inovação de empresas incubadas na Incubadora de Agronegócios das Cooperativas, Organizações Comunitárias, Associações e Assentamentos Rurais do Semiárido da Paraíba (Iacoc). Originalidade/valor: O valor desta pesquisa é a validação da literatura e efetividade do processo de fomento de incubadoras aos empreendimentos incubados, cuja transferência de recursos/conhecimento pode gerar inovação. Apresentamos a relação teórica entre a capacidade absortiva e as ações de inovação no contexto de empreendimentos incubados, inspirando práticas de gestão para inovações de diversos tipos, inclusive em contextos específicos, como o do agronegócio. Design/metodologia/abordagem: Realizamos um estudo multicasos qualitativo, cujas análises se concentram em seis empreendimentos incubados na Iacoc. Foram utilizadas pesquisa documental e entrevistas semiestruturadas com os principais gestores, sendo os dados processados pelo software ATLAS.ti. Posterior análise de conteúdo enquadrou os dados em quatro categorias centrais: capacidade absortiva realizada aliada às inovações de produto, de marketing, de processo e organizacional. Resultados: Os resultados mostram que os empreendimentos analisados apresentaram avanços a partir da capacidade absortiva realizada, com a geração de inovações em gestão e planejamento, gestão de pessoas, direcionamento de técnicas de produção, em características de concepção de produtos e acesso a mercados, entre outros. Defendemos que a associação teórica do campo da capacidade absortiva e a inovação no contexto de empreendimentos incubados podem desencadear ganhos substanciais ao campo temático e aos atores de ecossistemas de inovação: captação de conhecimento pelas empresas, ações de inovação, transformação dos envolvidos nas iniciativas e impacto socioeconômico.

Abstract Purpose: This article analyzes companies’ absorptive capacity and innovation actions incubated in the Agribusiness Incubator of Cooperatives, Community Organizations, Associations, and Rural Settlements of the Semiarid Region of Paraíba (Iacoc). Originality/value: The value of this research is the validation of the literature and the process’ effectiveness of fostering incubators with incubated enterprises, whose transfer of resources/knowledge can generate innovation. We present the theoretical relationship between absorptive capacity and innovation actions in the context of incubated enterprises, inspiring management practices for different types of innovation, including in specific contexts, such as agribusiness. Design/methodology/approach: We carried out a qualitative multicase study whose analysis focused on six projects incubated at Iacoc. Documentary research and semi-structured interviews with the leading managers were used, and the ATLAS.ti software processed the data. Subsequent content analysis framed the data into four main categories: absorptive capacity combined with the product, marketing, process, and organizational innovations. Findings: Results show that the analyzed projects presented advancements from the absorptive capacity realized, with the generation of innovations in management and planning, people management, the direction of production techniques, product design characteristics, and access to markets. We argue that the theoretical association of the field of absorptive capacity and innovation in the context of incubated enterprises can trigger substantial gains for the thematic area and the actors of innovation ecosystems: the capture of knowledge by companies, innovation actions, the transformation of those involved in the initiatives and socioeconomic impact.

RESUMO Este trabalho propõem o uso da Matemática Intervalar em conjunto com o Método dos Mínimos Quadrados e a linguagem Python para obter a melhor solução aproximada para sistemas intervalares obtidos de experimentos físicos. Este método de resolução será aplicado para obter a melhor função aproximada que ajusta um conjunto de dados oriundos de um experimento físico, no qual um carro se desloca com aceleração constante sob um trilho de ar inclinado. Ao fazer esse tipo de abordagem usando a Matemática Intervalar, busca-se inferir como as incertezas provenientes do experimento, assim como os erros gerados pelas representações e operações numéricas em computadores, interferem no resultado obtido. Para isso, se fez necessário a utilização da biblioteca Python for Extended Scientific Computing (Python- XSC), a qual é baseada na estrutura da aritmética intervalar e fornece funções para a resolução de sistemas lineares intervalares. A aplicação do estudo feito se mostrou bastante eficiente e de fácil utilização, o que motiva sua utilização.

ABSTRACT In this work, we used the Interval Mathematics, Method of Least Squares and Python language to obtain the best approximate solution for interval systems obtained from physical experiments. This resolution method will be applied to obtain the best approximate function that fits a data set from a physical experiment, in which a car was travels with constant acceleration under an inclined air trail. When doing this type of approach using Interval Mathematics, we seek to infer how the uncertainties arising from the experiment, as well as the errors generated by the representations and operations on computer, interfere with the obtained result. For this, it was necessary to use the Python for Extended Scientific Computing (Python-XSC) library, which is based on the interval arithmetic structure and provides functions for the solving of interval linear systems. The application of the study was very efficient and easy to use, which motivates its use.

Abstract Eutrophic estuaries receive organic matter (OM) inputs from multiple sources. This study evaluated the distribution and origin of sedimentary OM in an eutrophic estuary (Pina Sound, NE Brazil). Thirteen samples were collected in the sublittoral in addition to major local sources. Biochemical (chlorophyll - Chl), elemental [(C/N)a and C/S ratios] and isotopic (δ 15N and δ 13C) analyses were carried out for characterizing OM and redox conditions. The SIAR mixing model was used to quantify contribution from main sources. At Pina Sound, distribution of OM is associated with mud, reflecting the hydrodynamics control on deposition of suspended particles. Microphytobenthic production is limited ([Chl a] < 1000 µg/g organic carbon) in the sublittoral where the Chl degradation products prevail (mean [Pheopigments] = 2643 ± 958 µg/g organic carbon). Anoxic conditions (C/S ratio ≈ 2) are typically observed in sediments of deeper portions of Pina Sound. Such sediments receive high organic loads and are subject to poor water renewal. According to SIAR mixing model, sedimentary OM of Pina Sound is composed of, on average: 50% phytoplankton, 24% sewage and 26% C3 plants. This reflects fertilization of Pina Sound with high loads of untreated sewage. Pina Sound has a great potential to retain sewage-derived OM.

ABSTRACT Objective: Feeding restriction in rats alters the oscillators in suprachiasmatic, paraventricular, and arcuate nuclei, hypothalamic areas involved in food intake. In the present study, using the same animals and experimental protocol, we aimed to analyze if food restriction could reset clock genes ( Clock, Bmal1 ) and genes involved in lipid metabolism ( Pgc1a, Pparg, Ucp2 ) through nutrient-sensing pathways ( Sirt1, Ampk, Nampt ) in peripheral tissues. Materials and methods: Rats were grouped according to food access: Control group (CG, food ad libitum ), Restricted night-fed (RF-n, food access during 2 h at night), Restricted day-fed (RF-d, food access during 2 h in the daytime), and Day-fed (DF, food access during 12 h in the daytime). After 21 days, rats were decapitated at ZT3 (0900-1000 h), ZT11 (1700-1800 h), or ZT17 (2300-2400 h). Blood, liver, brown (BAT) and peri-epididymal (PAT) adipose tissues were collected. Plasma corticosterone and gene expression were evaluated by radioimmunoassay and qPCR, respectively. Results: In the liver, the expression pattern of Clock and Bmal1 shifted when food access was dissociated from rat nocturnal activity; this phenomenon was attenuated in adipose tissues. Daytime feeding also inverted the profile of energy-sensing and lipid metabolism-related genes in the liver, whereas calorie restriction induced a pre-feeding increased expression of these genes. In adipose tissues, Sirt1 expression was modified by daytime feeding and calorie restriction, with concomitant expression of Pgc1a , Pparg , and Ucp2 but not Ampk and Nampt . Conclusion: Feeding restriction reset clock genes and genes involved in lipid metabolism through nutrient-sensing-related genes in rat liver, brown, and peri-epididymal adipose tissues.

In this work, it is described the design of twenty-four heterocyclic amine-azachalcones compounds through molecular hybridization of chalcone scaffold and fragments of isoniazid, fluoroquinolones, and linezolid with antituberculosis potential. The new compounds were synthesized via Claisen-Schmidt condensation, providing yields of 36-95%. Fifteen compounds showed antituberculosis activity against Mycobacterium tuberculosis H37Rv strain. Two amine-azachalcones 15 and 17 showed relevant biological activity with minimum inhibitory concentration (MIC) values of 6.62 and 4.85 µM, respectively. Compound 12 showed the best profile of antitubercular activity with MIC = 9.54 µM and selectivity index (SI) = 9.33. It was found that morpholine group is important to increase potency of antimycobacterial activity but also to add some toxicity to the chalcone molecular framework. The results described herein would be a guide in the designing of novel and optimized antitubercular derivatives based on the chalcone scaffold.

This study reports the preparation of nanostructured cerium-doped titanium dioxide photocatalysts containing different cerium concentrations by a modified sol-gel route involving the use of acetylacetone. The nanocrystalline powders had their structural, morphological, and optical properties characterized by X-ray diffraction, vibrational and Raman spectroscopy, thermogravimetric analysis, X-ray photoelectron spectroscopy, Brunauer, Emmett and Teller (BET) surface area analysis, scanning electron microscopy combined with energy dispersive X-ray spectroscopy, total reflection X-ray fluorescence, and diffuse reflectance spectroscopy. Cerium incorporation reduced the bandgap energy of the solids to values that allowed them to absorb visible light. Cerium presence improved the textural properties that are crucial for the catalytic process. The photocatalytic activities were evaluated through photodegradation experiments of methyl orange dye solution. The synthesis with acetylacetone as stabilizer provided nanostructured materials with higher Ce3+/Ce4+ ratio, which enhanced the photocatalytic activity. Smaller quantities of the doped photocatalysts afforded higher degradation rates than the rates reported in the literature.

ABSTRACT The agricultural sector is considered an important source of greenhouse gases, in which the coffee crop has an important contribution, participating in the dynamics of soil C and CO2 emissions. In this way, the aim of this study was to analyze and quantify CO2 emissions in different phenological phases in soil cultivated with conilon coffee (Coffea canephora Pierre ex A. Froehner), and compare it with CO2 flow in forest and pasture area. The experiment was conducted in the Experimental Area of the Federal Institute of Espírito Santo, Southeastern Brazil, in an experimental plot of 0.5 ha cultivated with conilon Vitória coffee clones. The quantification of soil CO2 flux was performed between July 2016 and June 2017, this period comprising all the seasons of the year, using in two rounds of ratings (day and night), with five replicates each. For comparison purposes, another measurement of the CO2 stream was conducted in June 2017 with an evaluation period of 24 h. The highest CO2 emission occurred during the harvest phases of the fruit (206.7 mg CO2 m-2 h-1). The lowest daily CO2 flow records were observed in the rest and flowering phases with values of 82.1 and 83.6 CO2 mg m-2 h-1, respectively.
